A salt tolerant alfalfaMedicago sativa L. cell line (HG2-N1) has been selected for growth in 171 mM NaCl. The salt tolerance characteristic is stable and is retained after growth in absence of salt selection for two months.In vitro translation was used to compare mRNA composition from the salt tolerant HG2-N1 and parent salt sensitive HG2 cell lines grown in the presence and absence of 171 mM NaCl. The results suggest that the mRNA composition differs between HG2-N1 and HG2 in a number of RNA species. The salt tolerant HG2-N1 shows both increases and decreases in specific polypeptides as compared to HG2. Many of the enhanced polypeptide bands from mRNA in the salt tolerant HG2-N1 variant appear to be constitutively expressed, since they can be detected from HG2-N1 cells grown in presence and absence of NaCl, but the expression of a few bands may depend on the presence of added NaCl. Most enhanced polypeptides, which are detected from mRNA in the salt tolerant variant HG2-N1 (grown on NaCl) are different from polypeptide bands enhanced in the salt sensitive HG2 line as a result of 24 hour salt stress. Similar results were obtained from two dimensional analysis ofin vivo labeled polypeptides. At least one isolated cDNA clone shows selective expression of mRNA in salt tolerant cells grown in NaCl. These results indicate that adaptive mechanisms for salt tolerance may differ in some aspects from acute stress mechanisms.  相似文献

The effect of NaCl, KCl and LiCl on the growth and morphogeneis of tissue cultures originating from immature embryos of four wheat (Triticum aestivum L.) and one triticale (Triticosecale)varieties was investigated. The morphogenetic pathway to plant regeneration in Chinese Spring wheat was determined as incomplete somatic embryogenesis because the differentiation and subsequent germination of the shoot apices happened in the early phase of embryo development. Culture medium supplemented by NaCl suppressed the differentiation of shoot apices resulting in the development of more typical somatic embryoids. Forty mM concentrations of both NaCl or KCl increased the formation of somatic embryos in Chinese Spring. Arthur and GK Kincso wheat varieties while Lasko triticale regenerated well without the addition. The salts inhibited plantlet formation from somatic embryoids so the salts supplement should be omitted. Tissue culture responses to three levels of NaCl (0, 85mM and 170 mM) were evaluated in several Medicago species including: M. dzhawakhetica, M. marina, M. rhodopea, M. rupestris, M. sativa (alfalfa) and M. suffruticosa. The whole plant responses of the same genotypes were evaluated in half-strength Hoagland's solution containing 0, 51.5, and 103 mM NaCl. One or more genotypes of M. dzhawakhetica, M. rhodopea, M. rupestris, and M. sativa exhibited in vitro NaCl tolerance at 85 mM. In addition, one genotype each of M. dzhawakhetica, M. rhodopea, and M. sativa was tolerant of 170 mM NaCl. However, all of the genotypes that demonstrated NaCl tolerance in vitro were NaCl sensitive at the whole plant level. Conversely, M. marina the only species exhibiting whole plant NaCl tolerance, had the most NaCl sensitive genotypes at the in vitro level. Although an in vitro NaCl tolerance mechanism which confers whole plant NaCl tolerance was not observed, a potential NaCl tolerance germplasm source, M. marina, was identified.  相似文献

In vitro-grown cells of Sesuvium portulacastrum L., an important ‘salt accumulator’ mangrove associate, were incubated on a medium containing different levels of salt, including 0, 100, 200, or 400 mM NaCl, in order to evaluate biochemical, physiological, and growth responses. A significant decrease in callus growth, water status, and cell membrane damage was observed under salt stress. Osmotic adjustment was revealed by the accumulation of inorganic ions, such as sodium (Na+), and organic osmolytes (proline, glycine betaine, and total soluble sugars) in NaCl-treated calli compared to control. However, accretion of osmolytes and inorganic ions did not support growth of calli under NaCl stress conditions. The observed reduced growth rate in calli subjected to stress, up to 200 mM NaCl, was coupled with lower catalase and ascorbate peroxidase activities and with a significantly higher superoxide dismutase activity. These findings suggested that S. portulacastrum cell cultures exhibited higher osmotic adjustment to salt stress.  相似文献

Salt stress perturbs a multitude of physiological processes such as photosynthesis and growth. To understand the biochemical changes associated with physiological and cellular adaptations to salinity, two lettuce varieties (Verte and Romaine) were grown in a hydroponics culture system supplemented with 0, 100 or 200 mM NaCl. Verte displayed better growth under 100 mM NaCl compared to Romaine, but both genotypes registered relatively similar reductions in growth under 200 mM NaCl treatment. Both varieties showed differences in net photosynthetic activity in the absence of salt and 8 days after salt treatment. These differences diminished subsequently under prolonged salt stress (14 days). Verte showed enhanced leaf proline and restricted total cations especially Na+, lesser malondialdehyde (MDA) formation and lignification in the roots under 100 mM NaCl salinity. Membrane damage estimated by electrolyte leakage increased with elevated salt concentrations in roots of both varieties, but Verte had significantly lower electrolyte leakage relative to Romaine under 100 mM NaCl. Moreover, Verte also accumulated greater levels of carotenoids under increasing NaCl concentrations compared to Romaine. Taken together, these findings suggest that the greater tolerance of Verte to 100 mM NaCl is related to the more restricted accumulation of total cations and toxic Na+ in the roots and enhanced levels of antioxidative metabolites in root and leaf tissue.  相似文献

Papaver somniferum L. (opium poppy) cells were elicited with a Botrytis sp. homogenate and cultured by a semi-continuous process. Elicitation induced synthesis of sanguinarine and dihydrosanguinarine. Significant release of both alkaloids into the culture medium occurred. Medium exchange at 2-day intervals enabled product recovery from spent medium and maintained culture viability. Culture growth was not inhibited by elicitor treatment necessitating sub-culture prior to re-elicitation. As glycerol was suggested as an osmotic agent in the salt tolerantDebaryomyces hansenii the concentrations of total, intracellular, and extracellular glycerol produced by this yeast was followed during growth in 4 mM, 0.68 M, and 2.7 M NaCl media. The total amount of glycerol was not directly proportional to biomass production but to the cultural salinity with maximum concentrations just prior to or at the beginning of the stationary phase. In all cultures the cells lost some glycerol to the media, at 2.7 M NaCl the extracellular glycerol even amounted maximally to 80% of the total. A distinct maximum of intracellular glycerol, related to dry weight or cell number, appeared during the log phase at all NaCl concentrations. As the intracellular calculated glycerol concentrations amounted to 0.2 M, 0.8 M, and 2.6 M in late log phase cells at 4 mM, 0.68 M, and 2.7 M NaCl, respectively, whereas the corresponding analysed values for the glycerol concentrations of the media were 0.7 mM, 2.5 mM, and 3.0 mM, glycerol contributes to the osmotic balance of the cells.During the course of growth all cultures showed a decreasing heat production related to cell substance produced, most pronounced at 2.7 M NaCl. At 2.7 M NaCl the total heat production amounted to-1690 kJ per mole glucose consumed in contrast to-1200 and-1130 kJ at 4 mM and 0.68 M NaCl, respectively. TheY m -values were of an inverse order, being 129, 120, and 93 at 4 mM, 0.68 M, and 2.7 M NaCl, respectively.  相似文献

The present study was carried out to compare the effect of NaCl on growth, cell membrane damage, and antioxidant defences in the halophyte Crithmum maritimum L. (sea fennel). Physiological and biochemical changes were investigated under control (0 mM NaCl) and saline conditions (100 and 300 mM NaCl). Biomass and growth of roots were more sensitive to NaCl than leaves. Roots were distinguished from leaves by increased electrolyte leakage and high malondialdehyde (MDA) concentration. Superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activities, ascorbic acid (AA) and glutathione (GSH) concentrations were lower in the roots than in the leaves of control plants. The different activity patterns of antioxidant enzymes in response to 100 and 300 mM NaCl indicated that leaves and roots reacted differently to salt stress. Leaf CAT, APX and glutathione reductase (GR) activities were lowest at 300 mM NaCl, but they were unaffected by 100 mM NaCl. Only SOD activity was reduced in the latter treatment. Root SOD activity was significantly decreased in response to 300 mM NaCl and root APX activity was significantly higher in plants treated with 100 and 300 mM compared to the controls. The other activities in roots were insensitive to salt. The concentration of AA decreased in leaves at 100 and 300 mM NaCl, and in roots at 300 mM NaCl, when compared to control plants. The concentrations of GSH in NaCl-treated leaves and roots were not significantly different from the controls. In both organs, AA and GSH were predominating in the total pool in ascorbic acid and glutathione, under control or saline conditions.  相似文献

In order to characterise the effect of ectomycorrhiza on Na+-responses of the salt-sensitive poplar hybrid Populus × canescens, growth and stress responses of Paxillus involutus (strain MAJ) were tested in liquid cultures in the presence of 20 to 500 mM NaCl, and the effects of mycorrhization on mineral nutrient accumulation and oxidative stress were characterised in mycorrhizal and non-mycorrhizal poplar seedlings exposed to 150 mM NaCl. Paxillus involutus was salt tolerant, showing biomass increases in media containing up to 500 mM NaCl after 4 weeks growth. Mycorrhizal mantle formation on poplar roots was not affected by 150 mM NaCl. Whole plant performance was positively affected by the fungus because total biomass was greater and leaves accumulated less Na+ than non-mycorrhizal plants. Energy dispersive X-ray microanalysis using transmission electron microscopy analysis of the influence of mycorrhization on the subcellular localisation of Na+ and Cl in roots showed that the hyphal mantle did not diminish salt accumulation in root cell walls, indicating that mycorrhization did not provide a physical barrier against excess salinity. In the absence of salt stress, mycorrhizal poplar roots contained higher Na+ and Cl concentrations than non-mycorrhizal poplar roots. Paxillus involutus hyphae produced H2O2 in the mantle but not in the Hartig net or in pure culture. Salt exposure resulted in H2O2 formation in cortical cells of both non-mycorrhizal and mycorrhizal poplar and stimulated peroxidase but not superoxide dismutase activities. This shows that mature ectomycorrhiza was unable to suppress salt-induced oxidative stress. Element analyses suggest that improved performance of mycorrhizal poplar under salt stress may result from diminished xylem loading of Na+ and increased supply with K+.  相似文献

Growth rate and salt accumulation were investigated in experiments on Halimione portulacoides with seven sodium chloride treatments, in water culture. The growth of Halimione was found to be stimulated by moderate, 85–170 mM NaCl, levels of salinity, but increasingly depressed by salinities from 410–690 mM NaCl, which is comparable to salinities in salt marshes during the growing season. Using the same technique, growth rate, chloride and nitrogen uptake experiments at four different sodium chloride and nitrate treatment levels were conducted, in order to study the effect of nitrogen and salt. At 8 mM NaCl in the growth medium growth was depressed at 16.2 mM nitrate treatment levels. At 137 mM, 410 mM and 684 mM of NaCl growth was stimulated by increasing levels of nitrogen. The results of these experiments are discussed in relation to the nitrogen and salt conditions prevailing in Halimione portulacoides salt-marsh communities.  相似文献

In pigmented cells of Vitis vinifera suspension cultures, best accumulation of anthocyanins was obtained when nitrate concentration was reduced from 25 mM to 6.25 mM and when sucrose concentration was increased from 88 mM to 132 mM. Under such conditions growth was greatly decreased. However, cell viability was maintained. The increases in anthocyanins in pigmented cells were due largely to increases in peonidin — glucoside. The high sucrose and the low nitrate concentrations can be one of the important culture factors in controlling of anthocyanin production by cell cultures.  相似文献

NaCl (0 to 274 mM) was added to the culture media of normal and habituated (auxin and cytokinin independent) sugarbeet calli and its effect on growth (estimated by the increase of dry and organic matters), water content and osmotic potential was tested. Growth of normal callus was stimulated by 68 mM NaCl after a lag period of two weeks. This callus tolerated up to 137 mM NaCl without growth reduction and maintained its hydric status by readjustment of its osmotic potential in 24 h. NaCl quantities under 34 mM stimulated growth of the habituated callus from the 3rd day on; higher NaCl concentrations (68 to 274 mM) inhibited growth or were lethal. NaCl sensitivity of this habituated callus was not due to its inability to adjust its osmotic potential: this adjustment occurred from the 4th h of culture whatever the media. From the 3rd day on, however, this callus presented a water deficit which depended on NaCl concentration. It is suggested that the lowering of osmotic potential corresponds to an important water loss in relation to changes in membrane permeability. This study finally shows that mechanisms of salt tolerance may have developed at the cellular level. Lower growth and lower salt tolerance of the habituated callus need further investigation in relation to cell structure and hormone autonomy.   相似文献

Abstract

In vitro culture of rooted and unrooted olive microshoots, established from seed lines of free-pollinated “Frantoio” and “Moraiolo” cultivars, were evaluated for NaCl tolerance. The aim was to use growth and physiological parameters in order to identify salt-adapted genotypes. Leaf tissue elemental distribution of Na, Cl and K was also investigated in unrooted plantlets by cryo-scanning electron microscopy and energy-dispersive X-ray microanalysis. Both in unrooted and rooted plantlets, increased concentrations of NaCl reduced shoot growth, whereas plant survival was not affected. However, no significant interactions between line and NaCl concentration were found. Elemental distribution showed that Moraiolo J and Frantoio Z accumulated more Na and Cl inside leaves, and that these elements followed a tissue-dependent pattern. Rooting capacity was reduced at the higher levels of NaCl. Significant interactions between seed line and salt treatment were found. Seed lines showed different abilities to develop roots at different salt levels. In particular, Frantoio Z showed a significant and different behaviour relative to the other seed lines at 50 mM NaCl with regard to both length and dry weight of roots. The results obtained suggest that rooting parameters are the most useful tools in the evaluation and screening of salt-tolerant olive genotypes through in vitro shoot culture.  相似文献

The halotolerant cyanobacterium Anabaena sp was grown under NaCl concentration of 0, 170 and 515 mM and physiological and proteomic analysis was performed. At 515 mM NaCl the cyanobacterium showed reduced photosynthetic activities and significant increase in soluble sugar content, proline and SOD activity. On the other hand Anabaena sp grown at 170 mM NaCl showed optimal growth, photosynthetic activities and comparatively low soluble sugar content, proline accumulation and SOD activity. The intracellular Na+ content of the cells increased both at 170 and 515 mM NaCl. In contrast, the K+ content of the cyanobacterium Anabaena sp remained stable in response to growth at identical concentration of NaCl. While cells grown at 170 mM NaCl showed highest intracellular K+/Na+ ratio, salinity level of 515 mM NaCl resulted in reduced ratio of K+/Na+. Proteomic analysis revealed 50 salt-responsive proteins in the cyanobacterium Anabaena sp under salt treatment compared with control. Ten protein spots were subjected to MALDI-TOF–MS/MS analysis and the identified proteins are involved in photosynthesis, protein folding, cell organization and energy metabolism. Differential expression of proteins related to photosynthesis, energy metabolism was observed in Anabaena sp grown at 170 mM NaCl. At 170 mM NaCl increased expression of photosynthesis related proteins and effective osmotic adjustment through increased antioxidant enzymes and modulation of intracellular ions contributed to better salinity tolerance and optimal growth. On the contrary, increased intracellular Na+ content coupled with down regulation of photosynthetic and energy related proteins resulted in reduced growth at 515 mM NaCl. Therefore reduced growth at 515 mM NaCl could be due to accumulation of Na+ ions and requirement to maintain higher organic osmolytes and antioxidants which is energy intensive. The results thus show that the basis of salt tolerance is different when the halotolerant cyanobacterium Anabaena sp is grown under low and high salinity levels.  相似文献

Production of the faba bean in semi-arid and coastal areas maybe limited by the salt sensitivity of faba bean symbiosis. Accordingly,this study was done to analyse the effects of salt on the effectivesymbiosis of faba bean (Vicia faba L. var. minor cultivar Alborea)and salt-tolerant Rhizobium leguminosarum biovar. viciae strainGRA19. After 4 weeks of growth, the nutrient solutions weresupplemented with 50, 75 and 100 mM NaCl for 21 d. Plants wereharvested four times at weekly intervals, beginning at 4 weeks.Vicia faba tolerated low (50 mM NaCl) but not higher levels(75 and 100 mM NaCl) of salt stress. Salinity affected shootgrowth more than root growth. At the end of the culture, thetotal nitrogen content in the shoot was affected more than plantgrowth; conversely, in the root, growth was influenced morethan total nitrogen content. In nodules, nitrogen fixation (acetylenereduction activity) was more sensitive to salinity than ammoniumassimilation (glutamine synthetase and glutamate synthase). Summary Salicornia fruticosa was collected from a salt marsh on the Mediterranean sea coast in Libya. Growth and gas exchange of this C3 species were monitered in plants pretreated at various NaCl concentrations (0, 171, 342, 513 and 855 mM). Maximum growth was at 171 mM NaCl under cool growth conditions (20/10° C) and at 342 mM NaCl under warm growth conditions (30/15° C) with minimum growth at 0 mM NaCl (control). Net photosynthesis (Pn) was greatest in plants grown in 171 mM NaCl with plants grown at 513 and 855 mM having lowest rates. Maximum Pn was at 20–25° C shoot temperatures with statistically significant reductions at 30° C in control plants while salt treated plants showed such reductions at 35° C. Salt treatments increased dark respiration over the control at 171 and 342 mM but reduced it at higher concentrations. Photorespiration was reduced by salt treatment and increased by increasing shoot temperature. Greatest transpiration was in 171 mM NaCl treated plants and increasing shoot temperature increased transpiration in all treatments. Stomatal resistance to CO2 influx was influenced only moderately by temperature while increasing salinity resulted in increased stomatal resistance. In general both temperature and salinity increased the mesophyll resistance to CO2 influx. The species seems adapted to the warm saline habitat along the Mediterranean sea coast, at least partially, by its ability to maintain relatively high Pn at moderate NaCl concentrations over a broad range of shoot temperatures.  相似文献

 Sodium salt sensitivity of common oak (Quercus robur L.) was evaluated in hydroponic culture using INRA-Morizet solution. Addition of NaCl to the nutrient solution reduced only length and weight of roots and first flush stems. In contrast, the second flush was properly expanded even in the presence of 40 mM of NaCl in culture medium. Both leaf number and leaf area were not affected by increasing salt concentration in medium culture while this increase induced significant leaf damage especially in first flush leaves. Stem starch storage was reduced only at 40 mM NaCl treatment. Common oak seedlings seemed to be able to better compartmentalize sodium than chloride when the NaCl concentration increased in the medium culture. Chloride presented a lower uptake than sodium. Sodium was preferentially accumulated in roots and this accumulation occurred at the expense of potassium uptake. The decrease of ATP content in leaves of common oak seedlings submitted to NaCl treatments could indicate that it was used for sodium exclusion out of the leaves, especially in second flush leaves. A hydroponic experiment was conducted to elucidate the difference in growth and cell ultrastructure between Tibetan wild and cultivated barley genotypes under moderate (150 mM NaCl) and high (300 mM NaCl) salt stress. The growth of three barley genotypes was reduced significantly under salt stress, but the wild barley XZ16 (tolerant) was less affected relative to cultivated barley Yerong (moderate tolerant) and Gairdner (sensitive). Meanwhile, XZ16 had lower Na+ and higher K+ concentrations in leaves than other two genotypes. In terms of photosynthetic and chlorophyll fluorescence parameters, salt stress reduced maximal photochemical efficiency (F v/F m), net photosynthetic rate (Pn), stomatal conductance (Gs), and intracellular CO2 concentration (Ci). XZ16 showed relatively smaller reduction in comparison with the two cultivated barley genotypes. The observation of transmission electron microscopy found that fundamental cell ultrastructure changes happened in both leaves and roots of all barley genotypes under salt NaCl stress, with chloroplasts being most changed. Moreover, obvious difference could be detected among the three genotypes in the damage of cell ultrastructure under salt stress, with XZ16 and Gairdner being least and most affected, respectively. It may be concluded that high salt tolerance in XZ16 is attributed to less Na+ accumulation and K+ reduction in leaves, more slight damage in cell ultrastructure, which in turn caused less influence on chloroplast function and photosynthesis.  相似文献

Growth, photosynthesis, and Na+, K+, and Ca2+ distributions were investigated in 2-year-old hydroponically cultured Populus alba L. cuttings exposed to salt stresses (0, 0.85, 8.5, 17, and 85 mM NaCl in experiment 1 and 0, 50, 100, 150, and 200 mM in experiment 2) for 4 weeks in 2/5 Hoagland solution. Salt did not markedly inhibit height growth and diameter increment in 150 and 100 mM NaCl, respectively. The 85 mM NaCl treatment increased the dry weights of roots and total dry weight of plants, while 150 mM NaCl significantly reduced the dry weights of leaves, stems, and total plant weight. The decline in the photosynthetic rate lagged 2 weeks behind that of stomatal conductance in the 50 and 100 mM salt solutions. Different ions exhibited different distributions in different parts of the plant. Most Na+ ions were excluded and/or compartmentalized in roots at low and moderate salt stress (≤50 mM). K+ content in leaves increased with the increase in the salt concentration in the growth solutions.  相似文献
