Relations between neurons of the reticular and specific relay nuclei of the thalamus were studied in cats immobilized with tubocurarine. Under the influence of stimulation of the reticular nucleus (RN) unit activity in the thalamic relay nuclei was found to be considerably modulated. Cases of the appearance of IPSPs (possibly of monosynaptic nature), evoked by stimulation of RN, in neurons of the ventroposterolateral nucleus (VPLN) and lateral geniculate body (LGB) are described. During simultaneous recording of unit activity in RN and VPLN or LGB by means of two electrodes interaction of several types was found: inhibition of discharges of VPLN or LGB neurons accompanied by excitation of RN neurons: alternation of excitation-inhibition in neuron pairs in RN and VPLN or RN and LGB during low-frequency afferent or cortical stimulation (in this case excitation of RN neurons is associated with inhibition of VPLN or LGB neurons), and strengthening of the discharge of VPLN or LGB neurons during excitation of RN neurons. The possibility of the existence both of direct monosynaptic inhibition of activity of VPLN or LGB relay neurons under the influence of excitation of RN neurons and of their inhibition by activation of hypothetical interneurons of the relay nuclei themselves is accepted.I. Synaptic responses (postsynaptic potentials and action potentials) were evoked in mesencephalic decerebellated cats by stimulating pontine bulbar locomotor and inhibitory sites (LS and IS, respectively) with a current of not more than 20 µA in "medial" and "lateral" neurons of the medulla. Some neurons even produced a response to presentation of single (actually low — 2–5 Hz — frequency) stimuli. The remaining cells responded to stimulation at a steady rate of 30–60 Hz only. Both groups of medial neurons were more receptive to input from LS. Lateral neurons responding to even single stimuli reacted more commonly to input from LS and those responding to steady stimulation only to input from IS. Many neurons with background activity (whether lateral or medial) produced no stimulus-bound response, but rhythmic stimulation either intensified or inhibited such activity. This response occurs most commonly with LS stimulation. Responses of lumbar interneurons located in the most lateral regions of Rexed's laminae IV–VII to stimulation of the medial longitudinal bundle and gigantocellular reticular nucleus of medullary pyramids, red nucleus, and peripheral nerves were investigated in cats anesthetized with pentobarbital. Stimulation of the reticulospinal fibers evoked monosynaptic excitation of many interneurons specialized for transmitting activity of the lateral descending systems, but not of peripheral afferents. Convergence of excitatory influences of all three descending systems (cortico-, rubro-, and reticulospinal) was observed on some cells of this group. In addition, monosynaptic "reticular" E PSPs appeared in interneurons transmitting activity of group Ia muscle fibers and in some interneurons of the flexor reflex afferent system. Stimulation of reticulospinal fibers evoked IPSPs in some neurons of this last group. Neurons not exposed to reticulofugal influences (both specialized neurons and interneurons of segmental reflex arcs) were located chiefly in the dorsal zones of the region studied. Experiments on immobilized unanesthetized cats showed that hypothalamic stimulation effectively modified spontaneous unit activity and activity evoked by photic stimulation in the superior colliculus. Long-latency responses, often with a tonic type of formation, were predominant. Meanwhile, definite differences were found in the character of influences from different regions of the hypothalamus. Stimulation of the anterior hypothalamic region and lateral hypothalamus led more frequently to inhibition of spontaneous activity, often expressed as the development of initial inhibition, especially during stimulation of the lateral hypothalamus. Definite modulation of spontaneous activity of cyclic type also developed. Influences from these structures on activity evoked by photic stimulation were chiefly facilitatory and modulating in character. Stimulation of the ventromedial nucleus could produce inhibitory and facilitatory effects equally on activity of tectal neurons, with a tendency for the frequency of manifestation of facilitation to increase when a series of stimuli was used. Inward rectifying potassium (KIR) currents in medium spiny (MS) neurons of nucleus accumbens inactivate significantly in ~40% of the neurons but not in the rest, which may lead to differences in input processing by these two groups. Using a 189-compartment computational model of the MS neuron, we investigate the influence of this property using injected current as well as spatiotemporally distributed synaptic inputs. Our study demonstrates that KIR current inactivation facilitates depolarization, firing frequency and firing onset in these neurons. These effects may be attributed to the higher input resistance of the cell as well as a more depolarized resting/down-state potential induced by the inactivation of this current. In view of the reports that dendritic intracellular calcium levels depend closely on burst strength and spike onset time, our findings suggest that inactivation of KIR currents may offer a means of modulating both excitability and synaptic plasticity in MS neurons.  相似文献

The input/output relationship in primary visual cortex neurons is influenced by the history of the preceding activity. To understand the impact that membrane potential trajectory and firing pattern has on the activation of slow conductances in cortical neurons we compared the afterpotentials that followed responses to different stimuli evoking similar numbers of action potentials. In particular, we compared afterpotentials following the intracellular injection of either square or sinusoidal currents lasting 20 seconds. Both stimuli were intracellular surrogates of different neuronal responses to prolonged visual stimulation. Recordings from 99 neurons in slices of visual cortex revealed that for stimuli evoking an equivalent number of spikes, sinusoidal current injection activated a slow afterhyperpolarization of significantly larger amplitude (8.5±3.3 mV) and duration (33±17 s) than that evoked by a square pulse (6.4±3.7 mV, 28±17 s; p<0.05). Spike frequency adaptation had a faster time course and was larger during plateau (square pulse) than during intermittent (sinusoidal) depolarizations. Similar results were obtained in 17 neurons intracellularly recorded from the visual cortex in vivo. The differences in the afterpotentials evoked with both protocols were abolished by removing calcium from the extracellular medium or by application of the L-type calcium channel blocker nifedipine, suggesting that the activation of a calcium-dependent current is at the base of this afterpotential difference. These findings suggest that not only the spikes, but the membrane potential values and firing patterns evoked by a particular stimulation protocol determine the responses to any subsequent incoming input in a time window that spans for tens of seconds to even minutes.  相似文献

In response to stimulation of the posterior lateral nucleus in unanesthetized cats immobilized with D-tubocurarine an evoked potential consisting of three components with a latent period of 3–5 msec appeared in area 5b of the suprasylvian gyrus. All three components were reversed at about the same depth in the cortex (1500–1600 µ). Reversal of the potential shows that it is generated in that area by neurons evidently located in deeper layers of the cortex and is not conducted to it physically from other regions. Responses of 53 spontaneously active neurons in the same area of the cortex to stimulation of the posterior lateral nucleus were investigated. A characteristic feature of these reponses was that inhibition occurred nearly all of them. In 22 neurons the responses began with inhibition, which lasted from 30 to 400 msec. In 30 neurons inhibition appeared immediately after excitation while one neuron responded by excitation alone. The latent periods of the excitatory responses varied from 3 to 28 msec. The short latent period of the evoked potentials and of some single units responses (3–6 msec) confirms morphological evidence of direct connections between the posterior lateral nucleus and area 5b of the suprasylvian gyrus. Repetitive stimulation of that nucleus led to strengthening of both excitation and inhibition. Influences of the posterior lateral nucleus were opposite to those of the specific nuclei: the posterior ventrolateral nucleus and the lateral and medial geniculate bodies. Stimulation of the nonspecific reticular nucleus, however, evoked discharges from neurons like those produced by stimulation of the posterior lateral nucleus.A. A. We investigated the pre- and postsynaptic effects of pancreatic polypeptide (PP) on identified pancreas-projecting neurons of the rat dorsal motor nucleus of the vagus in thin brain stem slices. Perfusion with PP induced a TTX- and apamin-sensitive, concentration-dependent outward (22% of neurons) or inward current (21% of neurons) that was accompanied by a decrease in input resistance; PP was also found to affect the amplitude of the action potential afterhyperpolarization. The remaining 57% of neurons were unaffected. PP induced a concentration-dependent inhibition in amplitude of excitatory (n = 22 of 30 neurons) and inhibitory (n = 13 of 17 neurons) postsynaptic currents evoked by electrical stimulation of the adjacent nucleus of the solitary tract, with an estimated EC(50) of 30 nM for both. The inhibition was accompanied by an alteration in the paired pulse ratio, suggesting a presynaptic site of action. PP also decreased the frequency, but not amplitude, of spontaneous excitatory (n = 6 of 11 neurons) and inhibitory currents (n = 7 of 9 neurons). In five neurons, chemical stimulation of the area postrema (AP) induced a TTX-sensitive inward (n = 3) or biphasic (outward and inward) current (n = 2). Superfusion with PP reversibly reduced the amplitude of these chemically stimulated currents. Regardless of the PP-induced effect, the vast majority of responsive neurons had a multipolar somata morphology with dendrites projecting to areas other than the fourth ventricle or the central canal. These results suggest that pancreas-projecting rat dorsal motor nucleus of the vagus neurons are heterogeneous with respect to their response to PP, which may underlie functional differences in the vagal modulation of pancreatic functions.  相似文献

A small modification to a voltage-clamp set-up for studying isolated neurons, and the use of simple hippocampal slices allowed stable recording of excitatory postsynaptic currents (EPSCs) that were evoked by stimulating the Shaffer's collaterals of individual CA1 pyramidal neurons. With the developed method EPSCs and focal extracellular potentials could be recorded simultaneously. It was confirmed that the EPSC consists of two components that are mediated via N-methyl-D-aspartate (NMDA)- and non-NMDA-receptors. The effects of different blockers of these receptors on the postsynaptic current were investigated, as were the effects of adenosine, which, depending on its concentration, could either depress or potentiate the synaptic transmission.A. A brain stem slice preparation from adult and neonatal (less than or equal to 12 days old) rats and intracellular recordings were used to examine the cellular properties of neurons within the hypoglossal (HYP) nucleus. Resting membrane potential (Vm) for adult hypoglossal neurons was -80 +/- 2 (SE) mV. Rheobase was 2.1 +/- 0.4 nA, and input resistance (RN) was 20.8 +/- 1.5 M omega and decreased during the hyperpolarizing period ("sag"). Compared with adult HYP cells, newborn HYP neurons had significantly lower resting potentials (Vm = -73 +/- 2 mV), lower rheobase (0.7 +/- 0.2 nA), and higher RN (27.6 +/- 3.9 M omega). Single action potentials, elicited by short depolarizing-current pulses, were followed by a slow afterhyperpolarization in adult [6.4 +/- 0.3 mV, time constant (tc) 31.0 +/- 1.2 ms] and newborn cells (7.4 +/- 0.2 mV, tc 37.2 +/- 8.2 ms). Prolonged outward current (2 s) produced little spike frequency adaptation in either adult or newborn neurons. Onset of spike activity was not delayed by hyperpolarizing pulses preceding depolarizations. In addition, pharmacological experiments showed that HYP neurons have a tetrodotoxin-sensitive Na+ current and a delayed and an inward rectifier current but no major Ca2+ current. We conclude the following. 1) Electrophysiological membrane properties mature postnatally in HYP neurons; some of these developmental changes can be ascribed to an increase in soma size and dendritic outgrowth but others cannot. 2) Adult HYP neurons, compared with other brain stem neurons (i.e., vagal cells or cells in the nucleus tractus solitarius), are not endowed with major Ca2+ currents or K+ currents such as the A current and the Ca2(+)-activated K+ current.  相似文献

Synaptic processes in red nucleus neurons evoked by stimulation of different parts of the substantia nigra and nucleus interpositus of the cerebellum were investigated by an intracellular recording method in acute experiments on cats. Stimulation of this sort was shown to induce mono- and polysynaptic activation of rubrospinal neurons. Monosynaptic cerebellar and nigral excitatory influences were found to be very similar. These influences were shown to converge on the same rubrospinal neurons. The functional significance of inputs from the substantia nigra to the red nucleus for movement performance is discussed.L. A. During heteromodal extraneous stimulation (ES), a large part of responding neurons of the thalamic reticular nucleus (RN) exhibit selectivity by responding with excitation to the stimulation of only one type of sensory input. To visual ES, 12 of 32 tested neurons responded; and 4 of 21 tested neurons responded to auditory ES. Response of neurons to ES diminished during the process of habituation to these stimuli; and after habituation was completed, the number of neurons responsive to the ES also diminished. Use of ES led to disappearance of high-frequency, grouped discharges in the responses of the RN neurons. Initial responses to ES and to the following conditioning stimulus (CS) appeared during external inhibition of the conditioned reflex (CR), but the later components of impulse responses that ordinarily accompany realized CR were suppressed in nearly one half of the studied neurons. The reactions of 164 neurons of the orbitofrontal cortex (OFC) to stimulation of the mediodorsal nucleus of the thalamus (MD), the amygdaloid complex, and various sections of the hypothalamus, were investigated in acute experiments on cats. Stimulation of the MD led to the development in OFC neurons of reactions with a short (sometimes less than 6 msec) and stable latent period. Similar reactions were observed upon stimulation of the lateral amygdaloid nuclei. Stimulation of the basal and central nuclei of the amygdala evoked synchronization of the discharges in OFC neurons. Stable responses of OFC neurons developed from nuclei of the hypothalamus only in the lateral region. Stimulation of the other nuclei of the hypothalamus was accompanied by irregular responses or synchronization of the discharges. A study was done on base activity and changes in base activity (BA) of neurons in periaqueductal gray matter (PAG) during stimulation of monoaminergic structures of the brainstem: the nucleus raphe magnus (NRM), the locus coeruleus (LC), and the substantia nigra (SN), in rats anesthetized with hexenal (200 mg/kg). Three types of PAG neurons that differed in BA structure were identified. NRM, LC and SN stimulation changed BA only in type III neurons. Stimulation of these structures evoked an increase in BA frequency in 11.0–14.5%, and inhibition in 31.0–47.5% of type III neurons. Simultaneous stimulation of two structures led to a marked drop in intensity of effects. A depressing effect on BA was always detected if stimulation of one of the structures suppressed BA. Stimulation of two structures, with one being the NRM, was most effective. The role of PAG in the organization of the brain-stem component of the antinociceptive mechanism is discussed.A. A. In cultured pyramidal neurons of the rat brain cortex, we recorded (in the whole-cell configuration) postsynaptic currents (PSC) evoked by direct electrical microstimulation of an axon of the interneuron adjacent to the pyramidal cell. Application of 5 M bicuculline rapidly, entirely, and reversibly blocked these currents. Linear changes in the holding potential on the membrane of the postsynaptic cell resulted in linear changes in the amplitude of averaged currents. The currents underwent reversion when the holding potential was –16 mV, which was close to the reversal potential for Cl- ions at their respective concentrations in the extra- and intracellular solutions. We conclude that the recorded currents are inhibitory PSC (IPSC) mediated by GABA release. The amplitudes of the recorded currents varied from a measurable minimum (8 pA) to more than 150 pA at a holding potential on the postsynaptic cell membrane of –80 mV. Times to peak of the high- and low-amplitude currents showed no significant differences, being about 6.4 msec on average. Decays of the current could be satisfactorily approximated by a monoexponential function with a mean time constant of 17 msec. The time constants of IPSC decay were distributed accordingly to the Gaussian law. In some cases, the amplitude distributions of IPSC were unimodal ((with a rightward asymmetry), but in most cases they were clearly polymodal. The amplitude distribution can be described by the sum of several Gaussian distributions; the distance between modes of the Gaussians was 25 ± 6 pA, on average. The obtained estimates of the amplitude of monoquantal GABA-induced IPSC in neurons of the brain cortex allow us to conclude that in various CNS regions the dimension of the vesicles in GABA-ergic synapses formed by inhibitory interneurons is identical.  相似文献

The extent of the electrical transmission (ET) between different nonsymmetrical neurons in the segmental ganglion of the abdominal nerve chain of the leechHirudo medicinalis L. was investigated. ET was demonstrated by simultaneous recording of the intracellular potentials in two neurons when bursts of a polarizing current were passed alternately into each of them. At least 16 different pairs of nonsymmetrical identifiable neurons with ET which, as a rule, was symmetrical with a coefficient of attenuation of the order of 10 in the direction of greater effectiveness, were found. In different pairs of nonsymmetrical identifiable neurons, the ET of hyperpolarizing currents as a rule is more effective from neurons with a higher frequency of background activity. The presence of ET was also shown between many nonsymmetrical unidentifiable neurons of the ganglion. The assymetrical ET in some of the combinations of neurons investigated did not have rectifying properties. The direction of the ET was retained in them for any polarity of the polarizing current. In some experiments hyperpolarization of one of the neurons of a pair evoked depolarization and an increase in the frequency of the impulse activity of the other, while depolarization had the opposite effect ("negative" ET). Correlations between densities of various types of inward currents in the somatic membrane of dorsal root ganglion neurons were studied in three different rat age groups: 5–9 days, 45 days, and 90 days. A linear relationship was found in neurons with "slow" tetrodotoxin-sensitive sodium current between the densities of high-threshold calcium current and "slow" sodium current (Bravias-Pearson's correlation coefficient: r=0.84 and 0.70 for n1=16 and n2=28, respectively). No such correlation was observed in neurons with low-threshold calcium inward current. Cells with only two types of channel — "fast" sodium and high-threshold calcium — present in their somatic membrane manifested an inverse correlation (r=–0.48, where n4=95) between the densities of transmembrane currents passing through these channels. No inverse relationship was observed in the density of "fast" sodium and high-threshold calcium currents in neurons with tetradotoxinresistant "slow" sodium and/or low threshold calcium channels.A. Dispersed neurons from embryonic chicken sympathetic ganglia were innervated in vitro by explants of spinal cord containing the autonomic preganglionic nucleus or somatic motor nucleus. The maturation of postsynaptic acetylcholine (ACh) sensitivity and synaptic activity was evaluated from ACh and synaptically evoked currents in voltage-clamped neurons at several stages of innervation. All innervated cells are more sensitive to ACh than uninnervated neurons regardless of the source of cholinergic input. Similarly, medium conditioned by either dorsal or ventral explants mimics innervation by enhancing neuronal ACh sensitivity. This increase is due to changes in the rate of appearance of ACh receptors on the cell surface. There are also several changes in the nature of synaptic transmission with development in vitro, including an increased frequency of synaptic events and the appearance of larger amplitude synaptic currents. In addition, the mean amplitude of the unit synaptic current mode increases, as predicted from the observed changes in postsynaptic sensitivity. Although spontaneous synaptic current amplitude histograms with multimodal distributions are seen at all stages of development, histograms from early synapses are typically unimodal. Changes in the synaptic currents and ACh sensitivity between 1 and 4 days of innervation were paralleled by an increase in the number of synaptic events that evoked suprathreshold activity in the postsynaptic neurons. The early pre- and postsynaptic differentiation described here for interneuronal synapses formed in vitro may be responsible for increased efficacy of synaptic transmission during development in vivo.  相似文献

During the action of an extracellular polarizing current on neurons of the rabbit visual cortex electrical stimulation was applied to various hypothalamic nuclei (preoptic region, anterior hypothalamic region, lateral hypothalamus, mammillary bodies, and posterior hypothalamic nucleus). Hypothalamic stimulation was found to reduce the mean discharge frequency of most visual cortical neurons tested under conditions of anodal polarization, when the initial level of activity is considerably increased, than to a decrease in activity under conditions of cathodal polarization, when the initial level of activity is considerably reduced. The same tendency toward restoration of the initial (spontaneous) level of unit activity after hypothalamic stimulation was discovered when this level was shifted as a result of stimulation by regular flashes. The greatest effect was observed during stimulation of the preoptic region of the hypothalamus. Stimulation of the posterior hypothalamic nucleus was least effective in this respect.A. A.
