Chloroplast envelopes were isolated from chloroplasts purifiedfrom Spinacea oleracea L. (C3), Panicum miliaceum L. (NAD-malicenzyme-type C1), Digitaria sanguinalis (L.) Scop. (NADP-malicenzyme-type C4), Kalanchoe daigremontiana Hamet et Perrier (constitutiveCAM), and from Mesembryanthemum crystallinum L. (inducible CAM)performing either C3 photosynthesis or Crassulacean acid metabolism(CAM). For each species, methods were developed to isolate chloroplastenvelopes free of thylakoid contamination. The polypeptidesof ribulose bisphosphate (RuBP) carboxylase which has been consistentlyreported in envelope preparations of spinach were not foundin envelope preparations of C4 mesophyll chloroplasts. Silverstaining of envelope polypeptides resolved electrophoreticallyon sodium dodecylsulfate polyacrylamide gradient slab gels produceda more complex profile than did Coomassie staining which haspreviously been used with C3 envelope preparations, even thoughsilver reacted poorly with polypeptides corresponding to thesubunits of RuBP carboxylase. All of the plants examined possesseda major polypeptide of 27 to 29 kilodaltons (kD) which was previouslysuggested to be the phosphate translocator in spinach. WithC3 M. crystallinum, the 29 kD polypeptide stained most intensely.After induction of CAM, a 32 kD polypeptide also stained intensely,giving a profile similar to that obtained with the constitutiveCAM species. A 32 kD polypeptide was also prominent in C4 envelopepreparations, suggesting that a 32 kD polypeptide may be a translocatorprotein which is required in Crassulacean acid metabolism andC4 photosynthesis, but not in C3 photosynthesis. The photosynthetic carbon fixation pathways and levels of carbon-fixing enzymes of four dominant submersed macrophytes of Lawrence Lake, southern Michigan, were investigated during the main growth season (May to November). All four species (Scirpus subterminalis Torr., Najas flexilis (Willd.) Rostk. and Schmidt, Potamogeton praelongus Wulf., and Myriophyllum heterophyllum Michx.) were C3 plants based on their patterns of 14C pulse-chase incorporation. High levels of phosphoenolpyruvate carboxylase were also found in these species. These levels, as well as the ribulose 1,5-biphosphate carboxylase/phosphoenolpyruvate carboxylase ratio of the leaves, varied throughout the growing season and exhibited highest values in July. No shift in carbon fixation pathways, however, could be detected from July to October. The possible functions of phosphoenolypyruvate carboxylase in these plants, as well as the significance of C3 metabolism in submersed plants of temperate lakes, are delineated.  相似文献

Activities of carbonic anhydrase (CA), phosphoenolpyruvate carboxylase(PEPCase) and ribulose 1,5-bisphosphate carboxylase (RuBPCase)were compared in air- and 5% CO2adapted and adapting cells ofChlamydomonas segnis during the cell cycle in continuously illuminatedsynchronous cultures. Air-adapted cells exhibited considerablylower PEPCase activities, but higher RuBPCase and CA activitiesthan 5% CO2-adapted cells. Most (75 to 88%) of the CA activityin air-adapted cells appeared to be located in the periplasmicspace. Transferring 5% CO2-adapted cells to air scarcely influencedRuBPCase activity, but led to 85% decrease in the activity ofPEPCase and to 400% increase in that of CA. In such air-adaptingcells, more than half (54 to 70%) of the CA activity was intracellular.The proposal that PEPCase and RuBPCase in addition to CA maybe involved in the regulation of inorganic carbon uptake byair-adapted and adapting cells under CO2-limiting conditionsis discussed. RuBP carboxylase-oxygenase protein in three C3 species (Nicotianatabacum L., Solanum tuberosum L., Triticum aestivum L.) andthree C4 species (Panicum miliaceum L., Panicum texanum Buckl.,Zea mays L.) was quantitatively determined by sucrose densitygradient centrifugation and by immunochemical assay using antibodyraised to crystallized tobacco leaf RuBP carboxylase-oxygenase.The C3 species had 3- to 6-fold higher concentrations of RuBPcarboxylase-oxygenase than the C4 species when expressed oneither a chlorophyll or a leaf area basis. The C3 species alsoallocated a higher fraction of their total soluble protein tothis enzyme (from 25 to 60% for the C3 species compared to 8to 23% for the C4 species). There was no RuBP carboxylase-oxygenaseprotein or activity in the C4 mesophyll cells, while the enzymeconstituted from 20 to 40% of the total soluble protein in theC4 bundle sheath cells. A close correlation (r = +0·91)was found between catalytic activity and level of the enzymeprotein in the species examined.  相似文献

The activity of the enzyme ribulose bisphosphate carboxylase(RuBPCase) was estimated after rapidly extracting it from intactwheat leaves pretreated under different light and CO2 levels.No HCO3 was added to the extraction buffer since it isshown to inhibit RuBPCase. The activity increased as light intensityor CO2 concentration during pretreatment was increased. Enzymeactivity increased as temperature during pretreatment was decreased.Light activation did not affect the affinity of RuBPCase forCO2. A Km of 30 µM CO2 under air level O2 was determined.CO2, light and temperature are three main limiting factors ofphotosynthesis. It seems that the activity of RuBPCase is regulatedby these factors according to the requirements for CO2 fixation.  相似文献

The extent to which photorespiration occurs in CrassulaceanAcid Metabolism (CAM) plants has received limited attention.No comparative studies of C3 and CAM development have been made.To address this problem, activities of several photorespiratoryenzymes were measured in a facultative CAM plant—Mesembryanthemumcrystallinum L.—during induction of CAM by water stress(NaCl-treatment). Salt-treatment over a 22 d period produceda progressive change in metabolism from C3 to CAM. This wasconfirmed by (I) changes in gas exchange from C3 fixation tothe characteristic CAM pattern of nocturnal CO2 uptake; (2)increases in did acid fluctuation and (3) a 30-fold increasein phosphoenol pyruvate (PEP) carboxylase activity. In contrast,no significant changes were observed in the activities of glycollateoxidase (GO), NAD-dependent-hydroxypyruvate reductase (HPR),glutamine synthetase (GS) or glutamate dehydrogenase (GDH) whenplants were induced into CAM. Ion exchange chromatography onDEAE Sephacel detected only one GS isoenzyme (GS2, chloroplastic)at all stages of CAM induction, Western blot analysis, however,detected an additional, although minor, band of GS1 (cytosolic),in C3 plants, which disappeared following CAM induction. Ourresults show that, after development of CAM, these plants stillretain the capacity to photorespire. This may be an essentialrequirement of CAM plants growing in habitats with variablerainfall. When water availability is high, stomata may openin the light allowing ribulose bisphosphate carboxylase oxygenase(Rubisco) activity and photorespiration to occur. Effects of external nitrogen supply on quantity and activityof ribulose 1,5-bisphosphate carboxylase (RuBPCase) in mulberryleaves (Morus alba L. cv. Shin-ichinose) were examined. PhotosyntheticCO2 fixation and the contents of chlorophyll and phosphoruswere increased by the increased N supply. RuBPCase-protein concentrationand its proportion to total soluble-protein or to total-N rosecorrespondingly as the N supply increased. In species of Clusia, switching from C3-photosynthesis (C3-PS)to crassulacean acid metabolism (CAM) may be a means of optimizingwater use, plant carbon balance and photon utilization duringperiods of stress. We ask whether, in perennial species of Clusia,the switch from CAM back to C3-PS is also of ecophysiologicalsignificance. Our objective was to investigate the performanceof C. minor L. during a short-term shift from CAM to C3-PS.During the transition from CAM to C3-PS, nocturnal malate andcitrate accumulation decreased whereas CO2uptake increased duringthe daytime. However, after 7 d, marked nocturnal accumulationof citrate and 24 h CO2uptake occurred. Inflorescence leaves of citrus (Citrus sinensis L. Osbeck) accumulatecarbohydrate reserves at the beginning of the fruit set period.This effect is mimicked by exogenous GA3 applications in deflorateinflorescences. The effect of the hormone on two processes thatcould result in carbohydrate accumulation, i.e. increased ribulosebisphosphate carboxylase (RuBPCase) activity and changes inthe import/export balance of assimilates in the sprouts, wasstudied. Neither RuBPCase activity nor respiration rates were affectedby the hormonal treatment. On the other hand, blocking of thephloem connections with the rest of the plant induced an increasein carbohydrate contents in the leaves that was proportionallylower in GA3-treated sprouts than in control deflorate inflorescences.This increase was minimal in leaves from inflorescences at laterstages of fruit development. This was in spite of a lower fruitgrowth rate that, in turn, brought about a decrease in totaldry weight of the sprout. In contrast, total dry weight washigher in girdled deflorate inflorescences, whether or not theywere treated with the hormone. The presence of the fruit induceda thickening of the stems of intact inflorescences that wasalso seen in ungirdled GA2-treated sprouts. This increase indry weight per unit length, however, had a different chemicalbasis depending upon whether it was caused by the fruit or thehormone. From these data, the possible causes for the enhancement ofcarbohydrate accumulation in inflorescence leaves, and the involvementof gibberellin in the effect of fruit on the sprouts is discussed. C4 cereals (Zea maya L. and Sorghum bicolor L. Moench) and C3cereals (Triticum aestivum L. and Hordeum vulgare L) were grownin nutrient solutions with constant, interrupted, or absentpotassium supply. The lack of potassium retarded shoot growthand depressed the chlorophyll accumulation in all species ina similar way. After the renewal of potassium, the differencesin the compensation for growth retardation were not correlatedwith the photosynthetic system, but with the recovery of chlorophyllaccumulation in younger leaves. As important for the compensationof shoot growth retardation was a slower senescence of old leavescompared to plants with a constant potassium supply. This wasshown by the chlorophyll content and PEP carboxylase activity.In contrast to C3 cereals, the C4 cereals did not react withhigher chlorophyll contents to the same extent after the renewalof the postassium supply. The PEP carboxylase activity, however,was immediately raised higher than in control leaves. Chlorophylland PEP carboxylase activity increased simultaneously only inless aged leaves.  相似文献

The Km and Vmax of ribulose-1,5-bisphosphate carboxylase (RuBPCase)in selenium absorbing plants (Astragalus flavus Barn., Astragalusrafaelensis Barn. and Stanleya pinnata Bril.) were similar toRuBPCase from tomato (Lycopersicon esculentum L. var. tropic).The pH optima for RuBPCase activity was 8.0 for L. esculentumand A. flavus and 7.0 for A. rafaelensis and S. pinnata. TheActivation Energy (E) values for the enzymes were as follows:A.flavus (21.37), S.pinnata (19.85), A. rafaelensis (19.12)and L. escudentum (18.58). The energy of activation was higherfor the desert plants as compared to the tomato. The Arrheniusplot curves were linear to 50?C far the desert plants as comparedto 45?C for tomato. Enzyme kinetics of RuBPCase from halophytic plants (Salicorniapacifica Stand., var. utahensis (Tidestrom) Munz. and Salicorniarubra Nels.) indicated the enzyme was at least as sensitiveto NaCl concentrations as the enzyme from tomato. The introduction and repression of CAM in Sedurn telephiunmL, a temperate succulent, was investigated in watered, progressivelydrouglited and rewatered plants in growth chambers. Measurementswere made of water vapour and CO2 exchange, titratable acidity(TA) and xylem sap tension. Effects of photoperiod were alsostudied. CAM was induced by drought under long or short days,although when watered no CAM activity was expressed. C3-CAM intermediate plants were used for the investigation ofwater supply. Those which had received water and those drought-stressedboth displayed a similar nocturnal increase in TA, with a day-nightmaximum (H+) of 69 µmol g–1 fr. wt. The wateredplants took up CO2 at a maximum rate of 2?2 µmol m–2s–1 only in the light period, while the droughted plantsshowed a maximum nocturnal CO2 uptake rate of 0?69 µmolm–2 s–1. Subsequently, as CAM was repressed, thewatered S. telephiwn displayed little variation in TA, withconstant levels at 42 µmol g–1 fr. wt. (day 10).After 10 d of drought stress, the CAM characteristics of S.telephiurn were aLso affected, with reduced net CO2 uptake andH+. The transition between C3 and CAM in S. telephium can be describedas a progression in terms of the proportion of respiratory CO2which is recycled and refixed at night as malic acid, in comparisonwith net CO2 uptake. Recycling increased from 20% (day 1) to44% (day 10) as a result of the drought stress and was highin both the CAM-C3 stage (no net CO2 uptake at night) and alsoin the drought-stressed CAM stage (reduced net CO2 uptake atnight). The complete C3-CAM transition occurred in less than8 d, and the stages could be characterized by xylem sap tensionmeasurements: CAM = 0?50 MPa C3-CAM = 0?36 MPa C3 = 0?29 MPa. Immunolocalization of the bundle sheath-specific enzyme, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCase), and of the mesophyll-specific enzyme, phosphoenolpyruvate carboxylase (PEPCase), was used to follow development of the C4 pattern of photosynthetic enzyme expression during leaf growth in Atriplex rosea. The leaf tissue used for this characterization was also used in a parallel ultrastructural study, so that the temporal coordination of developmental changes in enzyme expression and cell structure could be monitored. Bundle sheath-specific accumulation of RuBPCase occurs early, at the time that bundle sheath tissue is delimited from the ground meristem, and follows the order of vein initiation. PEPCase proteins were detected 2–4 days after the first appearance of RuBPCase. PEPCase accumulation is restricted to ground meristem cells that are in direct contact with bundle sheath tissue and that will become C4 mesophyll; PEPCase was never found in more distant ground tissue. This pattern suggests that, while bundle sheath-specific accumulation of RuBPCase coincides with formation of the appropriate precursor cells, PEPCase expression is delayed until mesophyll tissue reaches a critical developmental stage. Cell-specific expression of both photosynthetic enzymes occurs well before the striking anatomical divergence of bundle sheath and mesophyll tissues, suggesting that biochemical compartmentation might serve as a developmental signal for subsequent structural differentiation.  相似文献

The effects of pruning shoot tops on the synthesis and degradationof ribulose 1,5–Wsphosphate carboxylase (RuBPCase) inleaves on remaining shoots were investigated in mulberry trees.Leucine labelled with 14C was fed to leaf discs from field-grownmulberry trees and 14C incorporation into RuBPCase was examined.Proportion of 14C in RuBPCase to leucine–14C absorbedby leaf discs was remarkably lowered by top-pruning, thoughoccasionally a slight increase was observed soon after pruning.Yet RuBPCase content in leaves on top-pruned shoots became progressivelyhigher than that in leaves on intact shoots. Changes in 14Cin Ru1BPCase in leaves of mulberry saplings previously fed 14CO2were followed. Following 14CO2 feeding, the attainment of themaximal level of 14C in RuBPCase was retarded by top-pruning.The highest level of 14C in RuBPCase was maintained in leaveson top-pruned shoots but decreased in leaves on intact shoots.Specific radioactivity in RuBPCase continued to increase inleaves on top-pruned shoots even after attaining a maximum levelin the control leaves. These facts suggest that the increasein RuBPCase by top-pruning results from a cessation of its degradationfor the remobilization of nitrogen for newly developing leaveson shoot tops. Photosynthesis is known to occur in rice panicles, but littlehas been reported about the photosynthetic or biochemical characteristicsof such panicles. The estimated gross amount of photo-syntheticallyassimilated CO2 in a panicle is 30% of that in a flag leaf.This result and the good light-intercepting characteristicsof the panicle in the canopy suggest that photosynthesis inthe panicle may contribute significantly to grain filling. Therice panicle is composed of spikelets and of rachis-branchesincluding rachis which have estimated gross rates of photosynthesisduring the 30-day period after anthesis of 130 to 180 and 50to 100 µmol CO2.(mg Chl)–1.h–1, respectively.The corresponding rate for the flag leaf is 180 to 230 µmolCO2.(mg Chl).h. On the basis of Chl, spikeletshave a high photosynthetic capability which is similar to thatof the flag leaf. The activities of ribulose-l,5-bisphosphate carboxylase (RuBPCase),phosphoenolpyruvate carboxylase (PEPCase), and pyruvate.Pi dikinase(PPDK) in spikelets were 129, 220, and 87 µmol.(mg Chl).h,respectively. The activities of PEPCase and PPDK in spikeletswere considerably higher than those in the flag leaf or rachis-branches.Oxygen-insensitive photosynthesis was found only in spikelets.The Km of NaHCO3 for photosynthesis by slices of spikelets inan aqueous solution (0.6 mM) was considerably lower than thatfor slices of flag leaf (4.2 mM). All these results indicatethat spikelets have different photosynthetic characteristicsfrom those of the flag leaf and rachis-branches. Enzymes of the C4, C3 pathway and photorespiration have beenanalyzed for P. hians and P. milioides, which have chlorenchymatousbundle sheath cells in the leaves. On whole leaf extracts thelevels of PEP carboxylase are relatively low compared to C4species, RuDP carboxylase is typical of C3 species, and enzymesof photorespiratory metabolism appear somewhat intermediatebetween C3 and C4. Substantial levels of PEP carboxylase, RuDPcarboxylase, and photorespiratory enzymes were found in bothmesophyll and bundle sheath cells. Low levels of C4-acid decarboxylatingenzymes may limit the capacity for C4 photosynthesis in P. hiansand P. milioides. Panicum hians and Panicum milioides were found to have characteristicsintermediate to those of C3 and C4 species with respect to CO2compensation point, percentage inhibition of photosynthesisby O2 at various O2/CO2 solubility ratios, and water use efficiency.C4 species have a higher carboxylation efficiency than eitherthe intermediate or C3 species. During photosynthesis, evenunder 2.5% O2, C4 species have a higher affinity for intercellularCO2 (Km 1.6 µM) apparently due to the initial carboxylationthrough PEP carboxylase. Under low O2 the intermediate and C3species had a similar affinity for intercellular CO2 duringphotosynthesis (Km 5–7 µM) consistent with carboxylationof atmospheric CO2 through RuDP carboxylase. There were considerablevariation in photosynthesis/unit leaf area at saturating CO2levels in the species examined which in part is due to differencesin RuDP carboxylase /unit leaf area. The highest rates of photosynthesis/unitleaf area under CO2-saturating conditions were with the C3 specieswhich had a correspondingly high level of RuDP carboxylase/unitleaf area. A comparison of carbon metabolism in the constitutive crassulacean acid metabolism (CAM) plant Kalanchoë daigremontiana Hamet et Perr. and the C3-CAM intermediate Clusia minor L. was undertaken under controlled environmental conditions where plants experience gradual changes in light intensity, temperature and humidity at the start and end of the photoperiod. The magnitude of CAM activity was manipulated by maintaining plants in ambient air or by enclosing leaves overnight in an atmosphere of N2 to suppress C4 carboxylation. Measurements of diel changes in carbonisotope discrimination and organic acid content were used to quantify the activities of C3 and C4 carboxylases in vivo and to indicate the extent to which the activities of phosphoenolpyruvate carboxylase (PEPCase), ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco) and decarboxylation processes overlap at the start and end of the photoperiod. These measurements in vivo were compared with measurements in vitro of changes in the diel sensitivity of PEPCase to malate inhibition. The results demonstrate fundamental differences in the down-regulation of PEPCase during the day in the two species. While PEPCase is inactivated within the first 30 min of the photoperiod in K. daigremontiana, the enzyme is active for 4 h at the start and 3 h at the end of the photoperiod in C. minor. Enclosing leaves in N2 overnight resulted in a two-to threefold increase in PEPCase-mediated CO2 uptake during Phase II of CAM in both species. However, futile cycling of CO2 between malate synthesis and decarboxylation does not occur during Phase II in either species. In terms of overall carbon balance, C4 carboxylation accounted for ≈ 20% of net daytime assimilation in both species under control conditions, increasing to 30–34% after a night in N2. Although N2-treated leaves of K. daigremontiana took up 25% more CO2 than control leaves during the day this was insufficient to compensate for the loss of CO2 taken up by CAM the previous night. In contrast, in N2-treated leaves of C. minor, the twofold increase in daytime PEPCase activity and the increase in net CO2 uptake by Rubisco during Phase III compensated for the inhibition of C4 carboxylation at night in terms of diel carbon balance.  相似文献

Growth and production of the temperate C4 species Cyperus longusL. was measured throughout a growing season in an establishedplot in Eastern Ireland. The maximum standing live biomass reachedwas 2·5 kg m–2. Estimates of unit leaf rate (ULR)and leaf area index (LAI) were made. The product of these quantitiesgave the crop growth rate (CGR) each week. C. longus was foundto maintain high values of LAI throughout the summer, with amaximum value of about 13 in early August. CGR reached a peakin early July. The optimum LAI was 11·6. Temperaturesat five levels in the plant canopy, and the amount of solarradiation intercepted by the canopy were measured continuouslyduring the summer. The mean daily rate of leaf extension waspositively correlated with the mean daily air temperature abovethe canopy but the temperature coefficient of the process waslow compared with other temperate species. The percentage ofsolar radiation intercepted by the canopy increased rapidlyin early summer, and canopy closure had occurred by mid-June.Rates of net photosynthesis were measured on young and old leafmaterial in situ at the time of peak LAI. In young leaves themaximum rates of net photosynthesis were higher than those publishedfor a range of temperate C3 species, but similar to those foundin another temperate C4 species, Spartina townsendii.
