Structural changes in positively and negatively supercoiled DNA   总被引:1,自引:0,他引:1  
The effect of superhelical constraint on the structure of covalently closed circular DNA (cccDNA; pBR322) with positive and negative writhe (superturn) has been investigated as a function of decreasing and increasing specific linking difference (mean superhelical density sigma). At low and moderate negative superhelical densities sigma, the overall average structure is maintained in an unwound B-form slightly modified. The overwound cccDNAs with positive writhe differ from those with negative writhe by an absence of cruciform structure. At high negative densities of supercoiling different changes involving the reversal of twist handedness are shown to lead to the formation of DNA segments in a conformation identical to the left-handed component of form V DNA.  相似文献

BackgroundChanges in endometrial cancer incidence rates after the precipitous decline in menopausal hormone therapy (MHT) use in 2002 have not been evaluated.MethodsUsing data from the Surveillance, Epidemiology, and End Results Program from 1992 to 2009 (SEER 13), we identified 63 428 incident endometrial cancer cases among women ages 20–74. We compared annual percent change (APC) in endometrial cancer incidence rates from 1992 to 2002 to rates from 2003 to 2009.ResultsIn contrast to the constant endometrial cancer rate pattern observed from 1992 to 2002 (APC 0.0%), rates increased after 2002 in women 50–74 years old (2.5%; PAPC comparison < 0.01). Endometrial cancer incidence increased over the entire time period among women ages 20–49 (1992–2002: 1.1%; 2003–2009: 2.1%; PAPC comparison = 0.21). Post-2002 increases in incidence among women ages 50–74 were specific to Type I endometrial tumors (1992–2002: ?0.6%; 2003–2009: 1.6%; PAPC comparison < 0.01).DiscussionThe increase in endometrial cancer incidence rates after 2002 may be related to the widespread decrease in estrogen plus progestin MHT use, which has been reported to lower endometrial cancer risk in overweight and obese women.  相似文献

The interaction between women's hormonal condition and subjective, physiological, and behavioral indices of desire or arousal remains only partially explored, in spite of frequent reports from women about problems with a lack of sexual desire. The present study recruited premenopausal women at two sites, one in the United States and the other in the Netherlands, and incorporated various measures of acute changes in sexual desire and arousal. A sample of 46 women who met criteria for Hypoactive Sexual Desire Disorder (HSDD) was compared to 47 women who experienced no sexual problems (SF). Half of each group used oral contraceptives (OCs). The specific goal was to investigate whether there is a relationship between women's hormone levels and their genital and subjective sexual responsiveness. Background demographics and health variables, including oral contraceptive (OC) use, were recorded and hormones (total testosterone (T), free testosterone (FT), SHBG, and estradiol) were analyzed along with vaginal pulse amplitude and self-report measures of desire and arousal in response to sexual fantasy, visual sexual stimuli, and photos of men's faces. Self-reported arousal and desire were lower in the HSDD than the SF group, but only for women who were not using oral contraceptives. Relationships between hormones and sexual function differed depending on whether a woman was HSDD or not. In line with prior literature, FT was positively associated with physiological and subjective sexual arousal in the SF group. The HSDD women demonstrated the opposite pattern, in that FT was negatively associated with subjective sexual responsiveness. The findings suggest a possible alternative relationship between hormones and sexual responsiveness in women with HSDD who have characteristics similar to those in the present study.  相似文献

Structure fluctuations and conformational changes accompany all biological processes involving macromolecules. The paper presents a classification of protein residues based on the normalized equilibrium fluctuations of the residue centers of mass in proteins and a statistical analysis of conformation changes in the side-chains upon binding. Normal mode analysis and an elastic network model were applied to a set of protein complexes to calculate the residue fluctuations and develop the residue classification. Comparison with a classification based on normalized B-factors suggests that the B-factors may underestimate protein flexibility in solvent. Our classification shows that protein loops and disordered fragments are enriched with highly fluctuating residues and depleted with weakly fluctuating residues. Strategies for engineering thermostable proteins are discussed. To calculate the dihedral angles distribution functions, the configuration space was divided into cells by a cubic grid. The effect of protein association on the distribution functions depends on the amino acid type and a grid step in the dihedral angles space. The changes in the dihedral angles increase from the near-backbone dihedral angle to the most distant one, for most residues. On average, one fifth of the interface residues change the rotamer state upon binding, whereas the rest of the interface residues undergo local readjustments within the same rotamer.  相似文献

Ile-de-France ewes had high plasma progesterone concentrations during early summer-late winter. Gentile-di-Puglia ewes had high progesterone values during the winter-spring-summer period but during autumn progesterone values were very low and oestrous behaviour was not displayed. The comparison with Ile-de-France ewes indicates that a phase shift occurs in the annual ovarian activity in ewes of the Gentile-di-Puglia breed.  相似文献

Uterine cytosol and nuclear receptors were measured at various time intervals after one dose or two consecutive doses of progesterone and the receptor values compared with the course of induction of uteroglobin, a progesterone-regulated uterine protein. The data of this study suggest (i) that progesterone action involves receptor consumption and (ii) that in the rabbit uterus the biologic response is not a direct function of available cytosol or nuclear receptor sites.  相似文献

This paper illustrates how cultural logics of desire are being transformed in the context of the global economy refashioning intimate lives. Exploring the experiences of Euro‐American female professionals in Jakarta, it suggests that they become uncomfortably visible as ‘white bodies’; their desirability appears compromised, especially given Orientalist discourses which valorise Asian women's bodies. At the same time, women's position as well‐paid employees generates a contradictory logic of desire: the ‘ego‐boost’ they experience at work may intensify their demands on the masculinity and enlightened views of potential partners, thus rendering Indonesian men, with their perceived bodily effeminacy and ‘traditional values’ unattractive. As one response to the lack of desirability, some women engage in a moral discourse that casts Indonesian women whom they consider ‘bargirls’, as well as the Euro‐American men they attract, as morally deficient. The paper thus provides an alternative perspective on reconfigurations of desire in the context of global gendered mobility.  相似文献

However, the underlying cause of this relationship is often unknown because multiple intrinsic and extrinsic factors can affect the relationship, and these have confounded previous attempts infer causation. The influence of different estrogen and/or progesterone treatments on concentrations of A and B forms of progesterone receptor (PR-A and PR-B) in the different cell types of chick oviduct was studied. A semiquantitative immunohistochemical assay for cellular PR concentrations was developed using a computer-assisted image analysis system. The staining intensity of nuclear PR in the basal layer of epithelial cells, glandular, smooth muscle and mesothelial cells was analysed separately using two monoclonal antibodies, PR6 and PR22. The measured concentrations of PR varied between different cell types and from cell to cell. A significant decrease in PR concentration, as noted by a decrease in staining intensity, was observed in all cell types studied 2 or 6 h after a single injection of progesterone with or without simultaneous estrogen administration. The decrease was also verified with immunoblotting and an immunoenzymometric assay (IEMA) for chicken PR. After down-regulation the concentration of PR recovered to the control level within 48 h after progesterone or estrogen administration. Estrogen administration alone was observed to cause changes in the concentration of PR-A only, having little or no effect on PR-B concentration depending on the cell type studied.

These findings indicate that estrogen and progesterone cause cell-specific changes not only to the total concentration of PR but also to the cellular ratio of PR-A and PR-B.  相似文献

Summary Male rats kept on a standard diet were treated either with progesterone or testosterone by a single intramuscular injection of preparations which are slowly absorbed and metabolized. The rats were anaesthetized 24 h after application of the hormones, perfused with glutaraldehyde, and the parathyroid glands prepared for electron microscopy. Morphometric analysis revealed that both progesterone and testosterone provoked (1) an increment in nuclear and cell volume and a concomitant increment in cell surface area, and (2) an increment in surface area of rough endoplasmic reticulum by 42% and 49%, and of the Golgi complex by 85% and 63%, respectively. Previously, we had found that oestradiol treatment led to a similar response in parathyroid cells. The conclusion is thus drawn that male and female sex hormones induce membrane synthesis resulting in an enhanced capacity for parathyroid hormone secretion since RER and Golgi complex are concerned with this secretion. It is considered probable that sex hormones have the ability fundamentally to modulate secretory activity in parathyroid cells.  相似文献

Global climate change has been shown to cause variable shifts in phenology in a variety of animals and unexpected outcomes across food chains are to be found. Here we examined how rising annual spring temperatures affected the interactions between seed masting, cavity nesting birds and dormice using long-term data from Eastern Czech Republic. We have shown that climate change was associated with unequal shifts in the phenology of two cavity-breeding groups: dormice and birds. Rising spring temperatures have progressively advanced the termination of hibernation for the edible dormouse Glis glis , a common bird predator, leading to an increasing overlap in the use of nesting boxes between dormice and birds. In contrast, only the collared flycatcher Ficedula albicollis , of the four cavity-nesting bird species, advanced its breeding dates in response to rising temperatures. At the same time, favourable weather conditions, coupled with good seed masting years, have been associated with a substantial rise in dormice numbers. Concurrent with the increasing dormice abundance, the number of bird nests destroyed significantly increased in three out of four bird species. We showed that while there was a significant change in the date that the dormice emerged from hibernation during the course of the study, it did not significantly contribute to predation levels when controlling for their abundance and timing of breeding in birds. We found that the increasing dormice abundance was the main factor causing high brood losses in birds, while the timing of breeding in birds had a variable effect between bird species. This study illustrates how changes in climate might affect organisms at various trophic levels with often unexpected outcomes. Limited evidence from other study organisms suggests that species most at risk are those at different trophic levels that do not shift at the same rate or in the same direction as their food resources, predators or competitors.  相似文献

To assess the roles of endogenous estrogen (E2) and progesterone (P4) in regulating exercise carbohydrate use, we used pharmacological suppression and replacement to create three distinct hormonal environments: baseline (B), with E2 and P4 low; estrogen only (E), with E2 high and P4 low; and estrogen/progesterone (E + P), with E2 and P4 high. Blood glucose uptake (R(d)), total carbohydrate oxidation (CHO(ox)), and estimated muscle glycogen utilization (EMGU) were assessed during 60 min of submaximal exercise by use of stable isotope dilution and indirect calorimetry in eight eumenorrheic women. Compared with B (1.26 +/- 0.04 g/min) and E + P (1.27 +/- 0.04 g/min), CHO(ox) was lower with E (1.05 +/- 0.02 g/min). Glucose R(d) tended to be lower with E and E + P relative to B. EMGU was 25% lower with E than with B or E + P. Plasma free fatty acids (FFA) were inversely related to EMGU (r(2) = 0.49). The data suggest that estrogen lowers CHO(ox) by reducing EMGU and glucose R(d). Progesterone increases EMGU but not glucose R(d). The opposing actions of E(2) and P(4) on EMGU may be mediated by their impact on FFA availability or vice versa.  相似文献

Aerobic capacity and body composition were measured at 3 time points over a 1-year period in 26 Division 1A women soccer players from Texas A&M University, in order to determine whether there were seasonal changes in these parameters. Subjects were tested in December, immediately following a 4-month competitive season; in April, following 15 weeks of strength and conditioning; and immediately prior to the start of the regular season in August, following a 12-week summer strength and conditioning program. A periodized strength and conditioning program design was incorporated in order to optimize anaerobic and oxidative capacity immediately prior to the regular competitive season. Significant differences in VO2max were measured between August (49.24 +/- 4.38 ml x kg(-1) x min(-1)) and December (44.87 +/- 4.61 ml x kg(-1) x min(-1)). No significant changes in aerobic capacity were found between April (47.43 +/- 4.01 ml x kg(-1) x min(-1)) and August (49.64 +/- 5.25 ml x kg(-1) x min(-1)). Significant increases in body fat were measured between August (15.71 +/- 2.92%) and December (18.78 +/- 2.79%), before and after the competitive season, respectively. No significant changes in body fat were found between April (16.24 +/- 2.95%) and August (15.71 +/- 2.92%). The results of this study suggest that decreases in muscle mass over the course of a regular competitive season contribute to decreases in aerobic capacity in collegiate women soccer players. Although it is unknown whether this decrease in muscle mass is the result of inadequate training or a normal adaptation to the physiological demands imposed by soccer, the results of the current study suggest that resistance training volume should be maintained during the competitive season, in order to maintain preseason levels of muscle mass.  相似文献

Summary Water potential () measurements of Atriplex canescens at the base of the Red Desert near Tipton in Wyoming, revealed a range between-15.5 to-45.1 bars. Minimum values coincided with the lowest air and soil temperatures, maximum with the greatest atmospheric evaporative demand. Change in exceeded 12 bars h-1 during periods of rapidly moving storm systems. Changes in appeared to be independent of plant size, age, sex, and the spatial location of plants. Chemical analyses revealed that xylem sap was up to three times more concentrated at high than at low . It was observed that the flow rate of sap was greater at lower than at higher and that the increase in water movement accounted for the dilution of the baseline concentration of sap solutes. Together, Ca, Mg, K and Na contributed 58% of the mean osmolality of the xylem sap; the dominant ions, however, were K and Cl. We suggest that the ability of the species to respond rapidly to changing atmospheric conditions affords it a distinct advantage in a harsh environment.  相似文献
