1. Chloroquine accumulation in rat liver after a single and repeated drug administration and lysosomal changes resembling some symptoms of lysosomal storage diseases were observed. 2. Repeated chloroquine treatment of rats resulted in increased activity of liver lysosomal enzymes acid phosphatase and beta-galactosidase and a significant enhancement of the activities of cathepsin D and cysteine proteinases were found. 3. No changes in the activity of liver macrophages (as assessed by the colloidal carbon clearance test) or in fluid-phase endocytosis of the marker 125I-polyvinyl-pyrrolidone by hepatocytes in vivo were found.  相似文献

A developing Friend's viral leukemia is accompanied by a 2,0--2,5-fold increase in the activity of lysosomal DNAse (DNAse II) in mouse liver as compared to normal. The increase in activity is observed on the 10--12th day after inoculation of virus-containing material and reaches its maximum on the 20th post-inoculation day. The increase in DNAse II activity is due to activation of the lysosomal system of Kupffer's and endothelial cells of the liver. The activity of mitochondrial DNAse (DNAse I) in the livers of leukemic mice showed no deviations from the normal level. A possible role of DNAse II in the protective response of the organism is discussed.  相似文献

Electrostimulation of hypothalamus results in an increase of RNA synthesis of the AU-type in liver cells of intact and adrenalectomized rats. No such effect is observed in the animals with denervated liver. An increase in RNA synthesis is accompanied by an increase in the activities of RNA-polymerases II and III and histone hydrolase and a decrease in the activities of RNAse and DNAse. The mechanisms of the effect of the nervous system on the activities of the nucleic acid metabolism enzymes are discussed.  相似文献

The activities of adenosine 3':5'-monophosphate (cyclic AMP)-dependent protein kinase may be partially controlled by a ubiquitous acidic heat-stable protein which inhibits the phosphotransferase reaction by interaction with the catalytic subunit of protein kinase (Walsh, D.A. et al. (1971), J. Biol. Chem. 246, 1977-1985). Since reported purification of this inhibitor involved subjecting tissue extracts to denaturing conditions, its existence under physiological conditions remained uncertain. A protein inhibitor, molecular weight 22,500, has been isolated from bovine myocardium by methods that do not include exposure to extreme heat or acid precipitation. The activity of this acidic protein is destroyed by exposure to trypsin and is unaffected by treatment with neuraminidase, RNAse or DNAse.  相似文献

Some of the biochemical indices relevant to the "free radical theory" of aging have been assessed in mice subjected to chronic low-dose whole-body irradiation. Radiation exposure results in enhanced accumulation of the lipofuscins in brain, heart, and intestine. In these animals, the degree of lipoperoxidation in liver was greatly increased, as were the free activities of acid phosphatase and cathespin, indicating damage to lysosomal membranes. The activity of SOD in brain and liver 20,000g post-mitochondrial supernatants was lower in the irradiated mice. All these changes arising from chronic whole-body irradiation are similar to those observed during aging and are effectively prevented by dietary supplementation with BHT. These observations lend considerable support to the "free radical theory" of aging.  相似文献

Phenobarbital was given to male rats as a single injection and as repetitive injections for 7 days. The effects of treatment on the lysosomal hydrolases acid phosphatase, cathepsin D, and aryl sulfatase were analyzed at different intervals ranging from 1 to 15 days after seven injections, and from 1 to 48 h after a single injection. In both cases, microsomal protein and NADPH-cytochrome c reductase were measured to ensure proper induction. After a single injection, a slight decrease in hydrolytic activities was observed. Repetitive administration of phenobarbital gave rise to a marked decrease of lysosomal enzyme activities 1 day after cessation of treatment. This decrease was followed by a continuous increase in activity up to day 3 and 4. One or 2 weeks after treatment, enzyme activities declined to control values. The increase in activity of lysosomal hydrolytic enzymes was correlated with the onset of induced autophagy of endoplasmic reticulum membranes described as occurring in liver upon cessation of phenobarbital exposure. It is concluded that phenobarbital treatment per se decreases lysosomal enzyme activities, whereas the induced autophagy following cessation of exposure is associated with enhanced levels of lysosomal hydrolases in rat liver.  相似文献

Growth of rabbit skin epithelial cells on a collagen gel is markedly enhanced by a soluble factor synthesized in a chemically defined minimal growth medium by fibroblasts of dermal origin. In the presence of this factor, the incorporation of thymidine and phosphate into skin epithelial cells is stimulated, and survival time of skin epithelial cells is prolonged. The growth factor is small (mol.wt < 1 000 D) and is not sensitive to lyophilization, heating, repeated freezing and thawing or the action of degradative enzymes RNAse, DNAse, trypsin and pronase. The chemical properties of the growth factor are not those expected of a free amino acid, peptide, non-polar lipid or nucleic acid.  相似文献

Atlantic cod, Gadus morhua, respond to starvation first by mobilising hepatic lipids, then muscle and hepatic glycogen and finally muscle proteins. The dual role of proteins as functional elements and energetic reserves should lead to a temporal hierarchy of mobilisation where the nature of a function dictates its conservation during starvation. We examined (1) whether lysosomal and anti-oxidant enzymes in liver and white muscle are spared during prolonged starvation, (2) whether the responses of these enzymes in muscle vary longitudinally. Hepatic contents of lysosomal proteases decreased with starvation, whereas those of catalase (CAT) increased and lysosomal enzymes of carbohydrate metabolism and glutathione S-transferase (GST) did not change. In white muscle, starvation decreased the specific activity of lysosomal enzymes of carbohydrate degradation and doubled that of cathepsin D (CaD). The activity of anti-oxidant enzymes and acid phosphatase in muscle was unchanged with starvation. In white muscle neither lysosomal enzymes nor anti-oxidant enzymes varied significantly with sampling position. In cod muscle, antioxidant enzymes, CaD and acid phosphatase are spared during a period of starvation that decreases lysosomal enzymes of carbohydrate metabolism and decreases glycolytic enzyme activities. In cod liver, the anti-oxidant enzymes, CAT and GST, were also spared during starvation.  相似文献

—A longitudinal study of the maturation of mouse cerebral lysosomal enzymes has been completed. Activity of the enzymes, acid phosphatase (I.U.B. 3.1.3.2), β-glucuronidase (I.U.B. 3.2.1.31) and β-acetylglucosaminidase (I.U.B. 3.2.1.30) was assayed spectrofluorimetrically on portions of supernatant from 0.25 M sucrose homogenates spun at 6 x 103 -min. Activities were obtained in native (free) and Triton X-100 activated samples (total). The neonatal period was characterized by relatively low free and high total acid phosphatase activities. An abrupt rise in free activity occurred during the period 10–20 days. Discontinuous anion exchange DEAE cellulose chromatography (0.01 m -tris–maleate, pH 6.3) with elution by ascending molarities of NaCl of the Triton X-100 activated supernatant revealed three major peaks in the adult. A fourth peak, designated as fraction II (‘maturation fraction’) occurred only during the neonatal period, a time also characterized by increased specific activity of fraction I, with no change in fraction IV. The chromatographic fractions were further characterized by optimal pH, ascorbate, fluoride, Cu2+ and Fe2+ ions. The maturation profiles of total, β-glucuronidase and total, β-acetylglucosaminidase differed from each other, and from that of total acid phosphatase. Comparable differences existed in the profiles of the free activities, and the ratio of free:total activity differed for each enzyme at any selected time especially during the neonatal period. These findings are are discussed with reference to the maturation of isoenzyme fractions with age, and suggest that the changes in structure-linked organization of individual lysosomal hydrolases are functions of heterogeneity in enzyme complement of individual lysosomes.  相似文献

The objective of the present study was to evaluate the effects of microcystin-LR (MCLR) on the activity of membrane enzymes from intestinal mucosa. In addition, serum chemistry and peroxidative status of both serum and intestinal homogenate were evaluated after treatment with MCLR. Wistar rats were treated with intraperitoneal injection of either 100 microg pure MCLR/Kg body weight or saline solution. A significant increase in liver weight and altered serum enzyme activities were found in MCLR-treated rats, indicating damage to the liver in these rats, as previously suggested. A higher specific activity of sucrase (1.5-fold) was observed after the administration of MCLR, whereas other intestinal apical membrane enzymes, such as lactase, maltase and alkaline phosphatase were not modified by the treatment. The specific activities of acid phosphatase and succinate dehydrogenase, markers for lysosomal and mitochondrial membranes, respectively, were also increased (32% and 60%, respectively) in treated rats. The analysis of lipid peroxidation showed that the peroxidative status was increased in both serum and intestinal mucosa from MCLR-treated rats, reflecting an excess production of oxygen free radicals induced by this cyanobacterial toxin. In conclusion, this study shows that acute exposure to MCLR affects the intestinal physiology by modifying the intestinal peroxidation status as well as the activity of membrane enzymes.  相似文献

This, however, does not engage all fibres of a muscle at a given time after denervation. The present study was carried out in order to determine if both type I (slow) and type II (fast) muscle fibres can react to denervation by increased endocytotic and lysosomal activities. Uptake of horseradish peroxidase as a marker for endocytosis was studied in conjunction with acid phosphatase staining for lysosomal activity in type I and type II fibres of the denervated mouse hemidiaphragm. Fibre typing was performed using a monoclonal antibody against fast skeletal myosin and by adenosine triphosphatase staining. The results show that increased endocytosis and lysosomal activation occur in both type I and type II fibres after denervation.  相似文献

Acid phosphatase activity, ATP and cyclic nucleotide levels were studied in primary cardiomyocyte cultures of newborn rats under hypoxic conditions. It was shown that incubation of cardiac cells in Eagle's medium supplemented with 10% calf serum during hypoxia (5% O2, 5% CO2, 90% N2) caused an hour later a decrease in lysosomal acid phosphatase activity, whereas free enzyme activity increased. The exposure of cells in Hanks salt solution to one hour hypoxia resulted in a 3-fold decrease of ATP level and further increase in free acid phosphatase activity. It was found that cAMP and cGMP levels do not depend on the presence of hypoxia. The mechanisms involved in the regulation of lysosomal membrane state are discussed.  相似文献

Non-latent (free) activities of two lysosomal enzymes (acid phosphatase and beta-glucuronidase) and urea production were measured in purified rat liver parenchymal cells incubated in the presence and absence of insulin. Non-latent enzyme activity was measured by including 0.25M sucrose in the assay mixtures to provide osmotic protection to the lysosomes. Total enzyme activity was estimated with Triton X-100 in the homogenates. Insulin was found to inhibit ureogenesis and to reduce non-latent lysosomal enzyme activity in the hepatocytes in vitro. Our data support the idea that insulin inhibits autophagy in rat liver parenchymal cells. Such an effect of insulin may also explain the inhibitory action of insulin on urea production in the rat liver.  相似文献

Summary The in-vivo uptake of exogenously applied horseradish peroxidase and the activities of the lysosomal enzymes acid phosphatase and cathepsin D were studied histochemically and/or biochemically in innervated and 2–14 day-denervated tibialis anterior muscles of the mouse. The biochemically determined uptake of horseradish peroxidase showed a large increase already 4 days after denervation. The activities of the lysosomal enzymes increased in a more gradual fashion, and only cathepsin D showed an increase in activity when expressed as total activity per muscle. Histochemically horseradish peroxidase was found to be localized in muscle fibres in characteristic spindle-shaped segments after denervation. The main increase in the number of such segments per transverse section of the muscle occurred between 3 and 6 days after denervation. In serial sections these segments frequently showed positive staining also for acid phosphatase.It is concluded that exogenously applied horseradish peroxidase is taken up into the lysosomal system, which after denervation becomes organized into characteristic spindle-shaped segments in the muscle fibres. The endocytic activity of muscle fibres increases early after denervation. This is followed by a more gradual increase in activity of lysosomal enzymes and finally by an organization of the lysosomal system into characteristic spindle-shaped segments. The results are compatible with the working hypothesis that increased endocytosis may initiate lysosomal activation in denervated skeletal muscle.  相似文献

Young adult male rats were fasted for 3 days, then fed a glucose-rich diet, ad libitum. At the end of the fasting period, the specific activity of liver glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase was decreased to 60% of control (nonfasted) levels. After 24 to 72 h of refeeding, the specific activity of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase increased seven- and twofold, respectively. During the fasting period, the liver lysosome fragility increased, as judged by increased release of bound acid phosphatase and β-N-acetylglucosammidase activity during standard homogenization. Three hours after feeding a carbohydrate-rich diet, a further increase in liver lysosomal fragility was observed that returned to control values prior to the induction of the dehydrogenases. Similarly, the susceptibility of liver lysosomes from fasted rats to increased fragility by the intraperitoneal injection of glucose or galactose was also observed. Prior starvation was not a requisite for labilization of lysosomal membranes by injected glucose, but induction of the pentose phosphate shunt dehydrogenase was not observed.In a group of 6-week old male rats fed a commercial pellet diet throughout, the injection of insulin caused no change in liver lysosomal fragility, though hypoglycemia resulted. Similar animals made diabetic by treatment with Streptozotocin and diabetic rats given insulin, showed no change in liver lysosmal fragility based on the percentage of free to total activities of β-N-acetylglucosaminidase, β-glucuronidase, β-galactosidase, and Cathespin D. However, when adult female rats were fasted for 24 h, then injected with sufficient insulin to produce hypoglycemia, liver lysosomal fragility, based on the release of β-N-acetylglucosaminidase during homogenization, increased nearly threefold. These studies demonstrate that stimulated lysosomal fragility can be initiated by refeeding fasted animals a carbohydrate-rich diet, by intraperitoneal injections of fasted rats with glucose or galactose, or by administering insulin alone to fasted rats. However, hyperglycemia induced by diabetogenic doses of Streptozotocin, or hypoglycemia induced in well-fed animals by insulin injection failed to elicit an enhanced liver lysosomal fragility. Whether induction of the enzymes of lipogenesis by rat liver is dependent upon a prior lysosomal membrane labilization remains to be determined.  相似文献

Isolated non-parenchymal cells from rat liver were separated by centrifugal elutriation into two fractions consisting of structurally intact Kupffer and endothelial cells with purities of 91 and 95%, respectively. Purified Kupffer and endothelial cells showed nearly equal specific activities for the lysosomal enzyme acid phosphatase, whereas the specific activity of cathepsin D was about 3 times higher in Kupffer cells. It was calculated that a significant amount of the cathepsin D activity in the liver is present in the Kupffer cells.  相似文献

Summary Pollen DNAse of Chlorophytum comosum was demonstrated cytochemically by coupling the breakdown of DNA (due to the endogenous DNAse) with acid phosphatase supplied to the reaction mixture. Precipitation of the free phosphate groups (from acid phosphatase reaction on the freed nucleotides) as lead phosphate results in a electron dense deposit, visible with the electron microscope. Enzyme activity using this technique was localized in vesicles along the intine (principally along the aperture region) and around the generative cell.  相似文献

Various lysosomal acid hydrolases from tissues of Niemann-Pick mice, a mutant strain of C57BL/KsJ mice (spm/spm), were examined and compared to those from control mice. Activities of beta-hexosaminidase, beta-galactosidase, acid phosphatase, and cathepsin L were elevated in the liver and spleen of the affected mice, whereas no significant changes in beta-glucosidase and acid alpha-glucosidase were observed. Alpha-Mannosidase and neutral alpha-glucosidase activities were rather decreased in the affected mouse liver. The level of beta-hexosaminidase in the Niemann-Pick mice was raised sixfold in the liver and two- to threefold in the spleen and brain, whereas its total activity was decreased in the kidney. Sixty to ninety percent of total activity of lysosomal hydrolases was solubilized with 0.1% Triton X-100 in control mice, but most of the beta-hexosaminidase activity of the Niemann-Pick mice remained associated with the membrane fraction of liver lysosomes. The beta-hexosaminidase of the Niemann-Pick mice was appreciably stable when heated at 55 degrees C, while hydrolases of the affected mice and all of the enzymes tested in control mice were heat labile. The relative content of two beta-hexosaminidase fractions separated by DEAE-cellulose column chromatography was 8% for beta-hexosaminidase I and 92% for beta-hexosaminidase II in the case of the control mouse liver. The isozyme pattern of hexosaminidases in Niemann-Pick mice was similar to that of control enzymes. However, the beta-hexosaminidase II accumulated in Niemann-Pick mouse liver was different from that of the control in optimum pH, Km values and thermostability.  相似文献

The effect of ischemia on the stability, i.e. the permeability of the lysosomal membrane of rat liver has been studied using quantitative histochemical analysis of acid phosphatase activity. Ischemia in vitro was performed for 0-240 min at 37 degrees C and ischemia in vivo for 60 min was followed by 1, 5, 24 and 48 h of reperfusion. Acid phosphatase activity was demonstrated in cryostat sections using naphthol AS-BI phosphoric acid as substrate and polyvinyl alcohol was added to the incubation medium to counteract diffusion phenomena. Ischemia in vitro up to 240 min did not affect the localization nor the total activity of acid phosphatase activity. After 60-min ischemia in vivo followed by 1-h reperfusion distinct areas showed decreased acid phosphatase activity. A further decrease in activity was observed after 5 h reperfusion. Final reaction product generated by acid phosphatase activity was rather diffusely distributed in border zones between normal and damaged tissue after 24 and 48 h of reperfusion following 60 min ischemia in vivo. It is concluded that not ischemia itself but rather reperfusion affects the stability of the lysosomal membrane due to the occurrence of oxygen-derived free radicals and/or imbalanced Ca2+ concentration. Restoration of the blood flow causes leakage of acid phosphatase from the lysosomes into the cytoplasm of liver parenchymal cells and from there to the blood.  相似文献
