Growth and symbiotic performance of soybean (Glycine max (L.)Merrill) cv. Bragg and three of its induced nodulation mutants(nod49, non-nodulating; ntsl 116, intermediate supernodulator;nts1007, extreme supernodulator) were compared throughout developmentunder different nitrogen regimes (0, 2, 5 and 10 mol nitratem–3). Nitrogen fixation was assessed using 15N-isotopedilution and xylem sap analysis for ureide content. Both techniquesconfirmed a complete lack of N2 fixation activity in nod49.Plant reliance on nitrogen fixation by the other genotypes wasdependent on the nitrate regime and the developmental stage.The ntsl007 and ntsl 116 mutants fixed more nitrogen than theparent cultivar in the presence of 10 mol m–3 nitratein the nutrient solution, but higher input of symbioticallyderived nitrogen was still insufficient to offset the amountof nitrogen removed in the harvested seed. However, the mutantsutilized less nitrate for growth than Bragg. Comparison of estimatesof N2 fixation derived from the 15N-dilution technique withthose based on relative ureide content of xylem sap indicatedthat the latter offered a simple and reliable procedure forevaluating the symbiotic performance of supernodulating plants. Three methods were used to study N2 fixation and effects ofwater deficit on N2 fixation: C2H2 reduction assay (ARA), 15Ndilution technique and accumulated N content. In addition, 15Ndilution was calculated both in a traditional way and in a modifiedway, which takes into consideration N and 15N content for theplants before the experiment started. The three methods wereapplied on the following Rhizobium-symbioses: Acacia albidaDel (Faidherbia albida (Del) A. Chev.) and Leucaena leucocephala(Lam) de Wit., and the Frankia-symbiosis Casuarina equisetifoliaL. The plants wereabout 4-months-old when they were harvested. Nitrogen derived from N2 fixation in control plants of Acaciaalbida was 54·2 mg as measured with ARA, while it was28·5 mg as measured with the 15N dilution technique,compared to 30·7 mg calculated as accumulated N. In comparison,L. leucocephala fixed 41·6 mg N (ARA), 53·5 mgN(15N dilution technique) and 56·3 mg N (accumulatedN). The Frankia-symbiosis had fixed 27·4 mg N as measuredby ARA, 8·1 mg N as measured by 15N dilution techniqueand 12·3 mg N as accumulated N. There were no differencesbetween the estimates based ontraditional and modified waysof calculating 15N dilution. The immediate effect of water deficit treatment on N2 fixationwas continuously measured inall species with ARA, which startedto decrease approximately 10 d after the initiation of the treatment,and declined to less than 5% of the initial level after 21–28d. The decrease in the amount of N derived from N2 fixation wasstudied in L. leucocephala during the period of treatment. Therewas a 26% decrease in amount of N derived from N2 fixation asresult of water deficit (as measured with ARA), while the decreasewas 23% when measured withboth the 15N dilution method and asaccumulated N. The three different methods for measuring N2 fixation and effectsof water deficit on N2 fixation are discussed. A technique used for hydroponics was adapted to measure instantaneousroot water uptake from the soil for a leaf succulent CAM species,Agave deserti. Comparisons were made to previously modelledwater fluxes for A. deserti and to Encelia farinosa, a non-succulentC3species. Net CO2uptake and transpiration forA. deserti underwell-watered conditions occurred primarily at night whereasroot water uptake was relatively constant over 24 h. Leaf thicknessdecreased when transpiration commenced and then increased whenrecharge from the stem and soil occurred, consistent with previousmodels. A drought of 90 d eliminated net CO2uptake and transpirationand reduced the water content of leaves by 62%. Rewetting theentire root system for 7 d led to a full recovery of leaf waterstorage but only 56% of maximal net CO2uptake. Root water uptakewas maximal immediately after rewetting, which replenished rootwater content, and decreased to a steady rate by 14 d. Whenonly the distal 50% of the root system was rewetted, the timefor net CO2uptake and leaf water storage to recover increased,but by 30 d gas exchange and leaf water storage were similarto 100% rewetting. Rewetting 10 or 20% of the root system resultedin much less water uptake; these plants did not recover leafwater storage or gas exchange by 30 d after rewetting. Water deficit is a very serious constraint on N2 fixation rates and grain yield of soybean (Glycine max Merr.). Ureides are transported from the nodules and they accumulate in the leaves during soil drying. This accumulation appears responsible for a feedback mechanism on nitrogen fixation, and it is hypothesized to result from a decreased ureide degradation in the leaf. One enzyme involved in the ureide degradation, allantoate amidohydrolase, is manganese (Mn) dependent. As Mn deficiency can occur in soils where soybean is grown, this deficiency may aggravate soybean sensitivity to water deficit. In situ ureide breakdown was measured by incubating soybean leaves in a 5 mol m ? 3 allantoic acid solution for 9 h before sampling leaf discs in which remnant ureide was measured over time. In situ ureide breakdown was dramatically decreased in leaves from plants grown without Mn. At the plant level, allantoic acid application in the nutrient solution of hydroponically grown soybean resulted in a higher accumulation of ureide in leaves and lower acetylene reduction activity (ARA) by plants grown with 0 mol m ? 3 Mn than those grown with 6·6 mol m ? 3 Mn. Those plants grown with 6·6 mol m ? 3 Mn in comparison with those grown with 52·8 mol m ? 3 Mn had, in turn, higher accumulated ureide and lower ARA. To determine if Mn level also influenced N2 fixation sensitivity to water deficit, a dry‐down experiment was carried out by slowly dehydrating plants that were grown in soil under four different Mn nutritions. Plants receiving no Mn had the lowest leaf Mn concentration, 11·9 mg kg ? 1, and had N2 fixation more sensitive to water deficit than plants treated with Mn in which leaf Mn concentration was in the range of 21–33 mg kg ? 1. The highest Mn treatments increased leaf Mn concentration to 37·5 mg kg ? 1 and above but did not delay the decline of ARA with soil drying, although these plants showed a significant increase in ARA under well‐watered conditions.  相似文献

Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Acetylene reduction activity (ARA) has been widely used to estimatecomparative rates of nitrogen fixation in soybean [Glycine max(L.) Merr.]. To use the non-destructive closed-container ARAassay to study plant-mediated regulation of N2 fixation, itwas first necessary to determine the validity of the ARA procedure.Thus, well-nodulated, hydroponically-grown soybean plants wereindividually assayed for ARA twice weekly for 5 weeks. Plantswere subsequently analysed for total nitrogen by the Kjeldahlprocedure and the values obtained were compared with the N-accumulationvalues estimated by ARA. For the 168 plants examined, ARA measurementsunderestimated N2 fixation by approximately 50%. However, N2fixation was underestimated by approximately 80% in plants witha very low ARA whereas N2 fixation was underestimated by only20% in plants with a high ARA. Because of the accuracy and reliabilityof the Kjeldahl measurements, these results show that nodulatedsoybean plants can exhibit different efficiencies in acetylenereduction. Thus, vigorous, well-nodulated, hydroponically-grownsoybean plants readily control nodule efficiency and the N fixationprocess.  相似文献

The combined effects of carbon dioxide (CO2) enrichment and water deficits on nodulation and N2 fixation were analysed in soybean [Glycine max (L.) Merr.]. Two short-term experiments were conducted in greenhouses with plants subjected to soil drying, while exposed to CO2 atmospheres of either 360 or 700 μmol CO2 mol–1. Under drought-stressed conditions, elevated [CO2] resulted in a delay in the decrease in N2 fixation rates associated with drying of the soil used in these experiments. The elevated [CO2] also allowed the plants under drought to sustain significant increases in nodule number and mass relative to those under ambient [CO2]. The total non-structural carbohydrate (TNC) concentration was lower in the shoots of the plants exposed to drought; however, plants under elevated CO2 had much higher TNC levels than those under ambient CO2. For both [CO2] treatments, drought stress induced a substantial accumulation of TNC in the nodules that paralleled N2 fixation decline, which indicates that nodule activity under drought may not be carbon limited. Under drought stress, ureide concentration increased in all plant tissues. However, exposure to elevated [CO2] resulted in substantially less drought-induced ureide accumulation in leaf and petiole tissues. A strong negative correlation was found between ureide accumulation and TNC levels in the leaves. This relationship, together with the large effect of elevated [CO2] on the decrease of ureide accumulation in the leaves, indicated the importance of ureide breakdown in the response of N2 fixation to drought and of feedback inhibition by ureides on nodule activity. It is concluded that an important effect of CO2 enrichment on soybean under drought conditions is an enhancement of photoassimilation, an increased partitioning of carbon to nodules and a decrease of leaf ureide levels, which is associated with sustained nodule growth and N2 rates under soil water deficits. We suggest that future [CO2] increases are likely to benefit soybean production by increasing the drought tolerance of N2 fixation.  相似文献

Nitrogen-fixing activity in two nodulated African legumes, Bambaragroundnut (Vigna subterraneaL.) and Kersting's bean (MacrotylomageocarpumL.), was assessed in the presence of nitrate (NO3-)ions in the rooting medium. Nitrogenase activity was unimpairedby the supply of 5 mol m-3NO3to both species. The feed back control mechanism proposed to explain the inhibitionof N2 fixation by N was investigated using Vicia faba cv. Fiord.Plants were grown under controlled conditions without mineralN in coarse river sand. Asparagine was supplied to plants activelyfixing N2 by absorption through cut roots and via a wick ordirect injection into the stem just above the bottom leaf. Responsesin N2 fixation were measured by acetylene reduction (AR). Feedingplants with [14C]-labelled asparagine showed that the amidewas taken up when exogenously applied. Asparagine (10 mM) suppliedby the above procedures resulted in a 50-70% inhibition of ARby 48 h. Glutamine produced a similar effect. The cut root methodallowed higher levels of these amides to be supplied but theinhibition observed with 10 mM asparagine was only increasedslightly with higher levels of the amide. The antibiotic Securopenprevented bacterial contamination of root solutions of asparagineand glutamine and had no effect on nodule activity. Quantification of N2 fixation by pigeonpea (Cajanus cajan (L.)Millsp.) in the field has proved difficult using techniquessuch as 15N isotope dilution, acetylene reduction and N difference.We report experiments to develop the ureide assay of N2 fixationbased on extraction and analysis of xylem exudate. Plants ofpigeonpea cv. Quantum, inoculated with effective Rhizobium spp.CB756, were grown in a temperature-controlled glasshouse inlarge pots filled with a sand: vermiculite mixture, in waterculture and in a slightly acidic, red-brown earth in replicatedfield plots. Xylem exudate was collected as bleeding sap fromboth nodulated and unnodulated roots, and from detached nodules.Exudate was extracted also from detached shoots and stems ofpigeonpea using a mild vacuum (60–70 kN m–2). Largedifferences in the composition of N solutes exported from rootsof N2-dependent and nitrate-dependent plants suggested thatshifts in plant dependence on N2 fixation may be reflected byconcomitant changes in N solutes. Thus, nodulated plants weresupplied throughout growth with either N-free nutrients or nutrientssupplemented with 1, 2, 5, 5, 10, or 20 mol m–3 15. Plants were harvested at regular intervals fordry matter and vacuum-extracted exudate. The relative abundanceof ureides ([ureide-N/ureide-N + nitrate-N + -amino-N] ? 100)in the exudate was highly correlated with the proportion ofplant N (calculated using a 15N isotope dilution technique)derived from N2 fixation. Two distinct phases of plant growthwere recognized and standard curves were prepared for each.The relationship between proportional dependence of plants onN2 and xylem relative ureides was unaffected by mineral-N source,i.e. nitrate or ammonium. This result is discussed in relationto interpretation of material from field-grown plants. The effectsof plant genotype, strain of rhizobia, section of stem extracted,removal of leaves, time delay between shoot detachment and extraction,and diurnal characteristics were examined in order to identifypotential sources of error and to optimize sampling procedures. The variations of leaf carbohydrate concentration, carbon isotopediscrimination () of leaf soluble carbohydrate, gas-exchangeand growth during a soil drying cycle under 350 and 700 µmolmol-1 CO2 concentrations ([CO2]) inQuercus robur seedlings wereanalysed. In well-watered conditions, a doubling of [CO2] causedan increase of CO2 assimilation rate (A) ( +47%) and a decreaseof stomatal conductance for water vapour (g) (–25%),anddoubled the intrinsic water-use efficiency (A/g). The valuesof A were not affected by elevated [CO2] which was consistentwith the 2-fold increase of A/g. Elevated [CO2 also significantlyincreased sucrose and starch leaf concentrations as well asaerial growth and plant dry weight. The stimulating effect ofCO2 enrichment on A and A/g was maintained in moderate droughtconditions, but disappeared in the most severe drought conditions.Drought induced an increase of hexose concentrations in both[CO2], but this effect was more pronounced under elevated [CO2],which may contribute to increase osmoregulation. From the onsetof drought, starch was depleted in both [CO2]. Carbon isotopediscrimination decreased in response to drought, which correspondedto an increase in A/g according to the two-step model of isotopicdiscrimination. In contrast, the A/g values derived from instantaneousleaf gas-exchange measurements decreased along the drying cycle.The discrepancy observed between the two independent estimatesof water-use efficiency is discussed in terms of time-scaleintegration. The results obtained with the isotopic approachusing soluble carbohydrate suggest a predominant stomatal limitationof CO2 assimilation in response to drought. The role of water relations and abscisic acid (ABA) in the responsesto drought were studied in a mediterranean forage crop, Trifoliumsubterraneum L. under field conditions. Soil and plant waterstatus, leaf gas exchange parameters, and xylem sap ABA contentwere determined at different times during a long-term soil dryingepisode in irrigated and droughted plants. The diurnal time-coursesof these parameters were also measured at the end of a droughtperiod. In response to soil drying stomatal conductance (g) was reducedearly to 50% that of irrigated plants before any substantialchange in water potential was detected. A close logarithmicregression between photosynthesis rate (A) and g was present.For the first weeks of drought the decline in A was less pronouncedthan in g, thus increasing water use efficiency. Stomatal conductanceduring diurnal time-courses showed no consistent relationshipswith respect to etther ABA or leaf water potential. Throughoutthe experimental period dependence of g on leaf water statuswas evident from the tight correlation (r2=0.88, P<0.01)achieved between stomatal conductance and midday water potential,but the correlation was also high when comparing g with respectto ABA content in xylem sap (r=0.83, P<0.001). However, thestomata from drought acclimated plants were apparently moresensitive to xylem ABA content. For similar xylem ABA concentrationsstomatal conductance was significantly higher in irrigated thanin waterstressed plants. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. Aims

Common bean is a major source of protein for many people worldwide. However, the crop is often subjected to drought conditions and its advantage in undertaking symbiotic nitrogen fixation can be severely decreased. The primary objective of this study was to compare the resistance of nitrogen fixation of 12 selected genotypes to soil drying.

Methods

Twelve common bean genotypes of diverse genetic background were compared. Plants were grown in pots and subjected to soil drying over about 2 weeks. Nitrogen fixation was measured daily using a flow-through acetylene reduction technique. The plants were exposed to acetylene for only a short time period allowing repeated measures. The acetylene reduction rate of plants on drying soil was normalized against the rates measured for well-watered plants.

Results

Substantial variability was identified among genotypes in the threshold soil water content at which nitrogen fixation was observed to decrease. Genotypes SER 16, SXB 412, NCB 226, and Calima were found to have the greatest delay in their decrease in nitrogen fixation rates based on soil water content. These four genotypes expressed substantial tolerance of nitrogen fixation to soil drying. These experiments also resulted in data on the threshold soil water contents at which transpiration rates decreased. A decrease in transpiration rates at high soil water contents is potentially advantageous since it allows soil water conservation for use as the severity of the drought increases. There was a general trend of those genotypes with sustained nitrogen fixation rates to low soil water contents also expressing decreased transpiration rates at high soil water contents.

Conclusions

This study identified genetic variation among common bean genotypes in their response of nitrogen fixation and transpiration to soil drying. Five genotypes (SER 16, SXB 412, NCB 226, Calima, and SEA 5) expressed the desired traits for water-limited conditions, which might be exploited in breeding efforts.  相似文献

Transpiration, water uptake by the roots and CO2 exchange oftwo leaf succulents, Senecio medleywoodii (Asteraceae) and Aloejucunda (Asphodeliaceae), were monitored simultaneously andcontinuously with a gas exchange cuvette combined with an apparatusto quantify water uptake (= waterbudget- meter). Measurements,which are primarily valid for plants with a sufficient watersupply, were made with the same plant for up to 29 consecutivedays. Ambient air temperature varied between 17 and 35 C witha constant dewpoint temperature of 13C of the ambient air anda 12 h photoperiod at 400–500mol m–2s–1 photonirradiance. The net water flux (Jw(net)=water uptake–transpiration)and the water balance (Jw(net) integrated for a timespan) werecalculated. Various tests were made to determine the accuracyof the measurements made with this rather complex equipment.In most cases the errors for transpiration and uptake rateswere much lower than 8% determined under the conditions of drastically(about 10 K per 30 min) increased or decreased ambient air temperatures.The experimental set-up proved to be a most valuable tool todetermine and analyse interactions between transpiration andwater uptake, changes in plant water status and the bufferingof negative Jwnet). Increasing the temperature of ambient air resulted, for bothspecies investigated, in a quick and considerably enhanced transpiration,but there was only a minor impact on water uptake. Water lossexceeding uptake was buffered by internal water reserves whichwere refilled within about 1 d after the plant was relievedof heat and drought stress caused by a period of high ambientair temperatures and high water vapour saturation deficits ofthe air. Repeated simulation of such stress periods showed thatthe absolute values of transpiration and the water uptake for24 h can vary, but the diurnal course of the values showed thesame pattern if the environmental conditions were identical.Such standardized diurnal transpiration and water uptake curvescould be very useful for the validation of mathematical modelsused to describe plant water relations. Established, nodulated white clover plants were transferredto eight tanks of a flowing culture apparatus with solutiontemperatures of 5, 11, 17, and 25 ?C (two tanks per temperature).Shoot temperature and light environment were common to all plants.After 7 d, (10 mmol m–3) was continuouslysupplied to one tank at each temperature while in the remainingfour tanks (one at each temperature) the plants were completelydependent on nodule N2-fixation. Plants were randomly selected at intervals during the following14 d period in order to measure root and nodule respirationand acetylene reduction activity (ARA) in a flow-through systemset at the adapted root temperature. Additional plants wereassayed for in vitro nitrate reductase activity in leaves, roots,and nodules. Apparent nitrogenase activity (ARA) and respiration associatedwith it were each markedly affected by temperature in two ways;(1) Activity per unit weight of nodule was reduced at lowertemperatures; (2) Development of the plant, and thus also nodulemass, was restricted at lower temperatures which, in turn, restrictedtotal nodule activity per plant. The presence of nitrate significantly reduced ARA of nodules,particularly at higher temperatures. However, significant discrepancieswere found when N2-fixation rates, estimated from the acetylenereduction assay, were compared with N2-fixation rates calculatedfrom curves fitted to N accumulation data (minus the rate of uptake in the case of nitrate-treated plants). Carbon use efficiency (CO2 respired per C2H4 produced) was notsignificantly affected by temperature or the presence of nitrate. Nitrate reductase activity (NRA) developed in all plant partsat the three highest temperatures, but not at 5 ?C. We calculatethat leaf NRA may account for 82, 75, and 68% of total nitratereduction at 11, 17, and 25 ?C respectively. Aster kantoensis Kitam., an endangered plant species of thefamily Compositae, is a local endemic to the gravelly floodplainsof a few rivers in central Japan. The successful growth of A.kantoensis is mainly restricted to sparsely vegetated siteswhere, due to lack of continuous vegetation, high radiant energyinput results in stressful conditions with excessive light andheat. To reveal the ecophysiological characteristics which enablethe species to cope with such environmental stresses, we measuredleaf temperature, shoot architecture and photosynthetic andtranspirational responses together with the microclimate ofthe natural habitat. Even under sunny summer conditions, theleaf temperature of A. kantoensis was much lower (35–39°C)than the soil surface temperature (max. 60°C). The relationshipbetween leaf position (height from the ground) and leaf temperatureshowed that the caulescent rosette form of A. kantoensis helpsavoid leaf overheating. Moreover, in situ gas exchange measurementsrevealed that the high transpirational capacity (as high as10 mmol H2O m-2s-1) was effective in controlling leaf temperature,as long as the soil water supply was not severely limited. Sinceit has effective mechanisms to avoid the multiple stresses indigenousto its gravelly floodplain habitat, A. kantoensis can maintaina high photosynthetic rate (up to 30 µmol CO2m-2s-1) withoutany midday depression under sunny summer conditions. The hypothesis that soil water potential (s) is better correlatedto heliotropic leaf orientation, photosaturated photosyntheticCO2 assimilation and stomatal conductance during periods oflimited water availability than is bulk leaf water potential(1) was examined in greenhouse-grown soybean (Glycine max) plants,submitted to a progressive drought. Paired plants were exposedto either 1000 or 100 µmol m–2 s–1 photonflux densities (PFD) for 45–60 mins. The higher irradianceinduced short-term decreases in 1, due to increased transpiration,while l in the plant exposed to low PFD did not decrease. Thesechanges in 1 occurred independently of changes in soil waterstatus. Concurrent to the light treatments, a single attachedleaf from each of the two plants was isolated from the restof the plant by shading, and the pulvinus of its terminal leafletwas exposed to a perpendicular PFD of 500 µmol m–2S–1. Leaf movement of this leaflet was recorded in responseto this light, until a stable leaflet angle was achieved. Valuesof s and l (before and after light treatment), and photosaturatedrates of photosynthesis and stomatal conductance, were thenmeasured on these leaves. Leaflet angle and gas exchange werebetter correlated with s (r2 = 0.50, 0.50 and 0.57 for angle,photosynthesis and conductance, respectively) than with l especiallywhen l was the result of short-term, high-light induced changesin leaf water status (r2 = 0.36, 0.32 and 0.49, for the sameparameters). Leaflet angle was also correlated with stomatalconductance (r2 = 0.61) and photosynthetic rate (r2 = 0.60),suggesting a close association between leaf orientation, leafmetabolism and soil water availability. The influence of hydrogenase in Bradyrizobum-Phaseoleae symbioseswas studied ex-planta and in-planra in soybean (Glycine max)and cowpea (Vigna unguiculata). The hydrogenase was activatedby the addition of hydrogen in the incubation gas phase whichmodified the response of nitrogenase activity of Hup+ (hydrogenuptake positive) symbiosis to the external oxygen partial pressure.For bacteroids the hydrogenase expression increased nitrogenaseactivity at supraoptimal pO2, acting possibly as a respiratoryprotection of nitrogenase. However, at suboptimal pO2, nitrogenaseactivity of Hup+ bacteroids decreased with hydrogen, a phenomenonattributed to the lower efficiency of ATP synthesis from hydrogenthan from carbon substrates oxidation. For undisturbed nodules,the hydrogenase expression in soybean increased the optimalpO2 for ARA (COP), from 35.3 to 40.3 kPa O2, and the ARA atsupraoptimal pO2; at suboptimal PO2 there was a negative effectof hydrogenase on ARA, although this inhibition was less thanon bacteroids and was not detected if plants were grown at 15°C rather than 20 °C root temperature. No H2 effectwas detected on cowpea nodules. The results on soybean nodulesare consistent with the concept that symbiotic nitrogen fixationis oxygen-limited and that hydrogenase activity has no beneficialeffect on nitrogen fixation in O2 limitation. The paper describes the effect of soil moisture content andair humidity on CO2 exchange (PN), CO2 diffusion resistance(Cr) and transpiration (E) in four varieties of japonica rice(Oryza sativa L.). A decrease in soil moisture content reducedthe rate of photosynthesis to a varying degree in the varieties.Reduction in photosynthesis was attributed to increase in Cr.The effect of low soil moisture on photosynthesis and CO2 diffusionwas further intensified by decrease in air humidity. By maintaininga high humidity in the air around the leaves however, the effectof soil moisture deficiency was reduced considerably, exceptin Rikuto Norin 21 which was very sensitive to soil-moisturedeficiency alone. Dryness of the air enhanced the transpirationrate, although the increase was relatively less in the plantsfacing a simultaneous water crisis at the root surface. In plantsgrowing under flooded conditions, a decrease in air humiditycaused a slight depression in PN despite the simultaneous decreasein Cr.
