Bird mitochondrial gene order: insight from 3 warbler mitochondrial genomes   总被引:1,自引:0,他引:1  
Two main gene orders exist in birds: the ancestral gene order and the remnant control region (CR) 2 gene order. These gene orders differ by the presence of 1 or 2 copies of the CR, respectively. Among songbirds, Oscines were thought to follow the ancestral gene order, with the exception of the lyrebird and Phylloscopus warblers. Here, we determined the complete mitochondrial genome sequence of 3 non-Phylloscopus warblers species and found that the blackcap (Sylvia atricapilla) and the reed warbler (Acrocephalus scirpaceus) have 2 almost identical copies of the CR, whereas the eastern orphean warbler (Sylvia crassirostris) follows the remnant CR 2 gene order. Our results contradict previous studies suggesting that Acrocephalus and most sylvioid warblers exhibit the ancestral gene order. We were able to trace this contradiction to a misidentification of gene order from polymerase chain reaction length determination. We thus suggest that passerine gene order evolution needs to be revised.  相似文献

A 40 kDa chitinase from Shewanella inventionis HE3 was purified (ChiA-Si40) and characterized. Using fermentor with an optimized medium for 48 h at 37 °C, enzyme activity was enhanced by 10-times compared to those using shaking-flask-culture. Purified chitinase is a homogenous monomer with molecular mass of 40 kDa. Its N-terminal residues revealed significant identity with glycoside hydrolase family 18 (GH18) chitinases from Gammaproteobacteria. Using colloidal chitin as a substrate, its finest activity was accomplished at pH 4 and a temperature of 70 °C. Its catalytic efficiency (kcat/Km) was superior to that of some bacterial GH18 chitinases and commercial enzyme, Chitodextrinase®. For scale-up and with regards to the improvement of ChiA-Si40 with PEG 6000 storage stability (6 months), the atomizing process was more pronounced than that of lyophilizing. Bio-assay of ChiA-Si40 against grain weevil Sitophilus granarius, indicates that it had an efficient insecticidal effect. About 10–100 % mortality rates were obtained 1-h after insect came in contact with ChiA-Si40. Histological study clearly demonstrated that luxury larval mid-gut, peritrophic-membrane, and epithelial-cells have been affected considerably after ChiA-Si40 treatment. These properties make ChiA-Si40 a potential bio-insecticidal agent for the biological control of S. granarius that is popular among insect pests of stored grains in Algeria.  相似文献

番木瓜果实内生细菌MGP3 菌株的鉴定及拮抗作用   总被引:1,自引:0,他引:1  
【目的】从番木瓜果皮内筛选具有较强拮抗活性的内生细菌防治番木瓜采后炭疽病和疫霉病,以减少果实采后病害带来的损失。【方法】采用稀释分离和平板抑菌圈法进行内生细菌的分离筛选,结合形态特征、生理生化特性及16S rDNA部分序列同源性分析对菌株进行鉴定,菌株经利福平诱抗处理后田间接种到果树树干上,测定内生菌的定殖动态,采用采前和采后生防试验测定菌株对番木瓜炭疽病和疫霉病的生防效果。【结果】从番木瓜果皮中分离筛选到一株具有拮抗活性的内生细菌MGP3,对10种病原菌有较强的拮抗作用,鉴定该细菌为铜绿假单胞菌(Pseudomonas aeruginosa,登录号JF708186),MGP3可进入番木瓜叶片、叶柄和果皮中定殖。MGP3对采后番木瓜炭疽病和疫霉病的防治效果分别达到50%和71%;除苗期外,采前4个不同时期经MGP3菌液处理可以显著降低采收后果实炭疽菌的潜伏侵染率和炭疽病的病情指数。【结论】番木瓜内生拮抗细菌MGP3具有潜在的生防应用价值。  相似文献

The γ phase Li3VO4 which possesses higher ionic conductivity is more preferable for lithium ion batteries, but it is only stable at high temperature and would convert to low temperature β phase spontaneously when cooling down. Here, the phase control of Li3VO4 to stabilize its γ phase in room temperature is successfully mediated by introducing proper Si‐doping, and for the first time the electrochemical performances of γ‐Li3VO4 is investigated. It is found that pure γ‐Li3VO4 can be obtained in a doping ratio of x = 0.05–0.15 in Li3+xV1?xSixO4 with addition of excess Li source in synthesis design. The doping mechanism and the energy changes are investigated in detail by using the first principle calculations, it reveals that an interstitial Li+ is formed with doping of Si4+ in Li3VO4 to balance the system charge. When served as an anode, the Si‐doped γ‐Li3VO4 shows much smoothed Li+ insertion/extraction and enhanced cycle stability with only a single pair of redox peaks, which behaves much different with the complex multicouples of redox peaks in typical β‐Li3VO4. These changes in electrochemical performances implies that γ‐Li3VO4 can effectively accommodate Li+ in an easier and more facile way and relieved structure stress during the charge/discharge process.  相似文献

Male Sprague-Dawley rats were pair-fed a liquid diet containing 36% of calories as ethanol for at least 31 days. Mitochondria were isolated from the livers and assayed for state 3, state 4 and uncoupled respiration at all three coupling sites. Assay conditions were established that maximized state 3 respiration with each substrate while maintaining a high respiratory control ratio. In mitochondria from ethanol-fed animals, state 3 respiratory rates were decreased at all three coupling sites. The decreased state 3 rate observed at site III was still significantly higher than the state 3 rates observed at site II in mitochondria from either ethanol-fed or control animals. Moreover, the maximal (FCCP-uncoupled) rates with succinate and -ketoglutarate were the same in mitochondria from ethanol-fed and control animals, whereas with glutamate-malate as substrate it was lowered 23% by chronic ethanol consumption. To investigate the role of cytochrome oxidase in modulating the respiratory rate with site I and site II substrates, the effects of cyanide on state 3 and FCCP-uncoupled respiration were determined. When the mitochondria were uncoupled there was no decrease in the rate of succinate oxidation until the rates of ascorbate and succinate oxidation became equivalent. Conversely, parallel inhibition of ascorbate, succinate and glutamate-malate state 3 respiratory rates were observed at all concentrations (1–50 μM) of cyanide utilized. These observations suggest strongly that in coupled mitochondria ethanol-elicited decreases in cytochrome oxidase activity depress the state 3 respiratory rates with site I and II substrates.  相似文献

To study differences in the development of immunity, leukocytes from cord blood are often compared with those from adult peripheral blood. Western blot analysis is a common method for detecting proteins. In this study, we investigated the reliability of using different housekeeping proteins (β-actin, β-tubulin, and glyceraldehyde-3-phosphate dehydrogenase [GAPDH]) as internal controls for different leukocyte subpopulations from infants, children, and adults. Our results showed that the expression levels of β-actin and β-tubulin were much lower in cord blood leukocytes than in adult leukocytes, and this expression pattern persisted in children up to 3 years old. Further study revealed that the β-actin expression level in newborns was especially lower in CD14-positive monocytes. However, cord blood and adult peripheral blood monocytes had similar expression levels of β-actin messenger RNA (mRNA). Further experiments showed that posttranslational regulation was responsible for the low β-actin expression level in neonatal monocytes. Thus, researchers should carefully assess the appropriate use of housekeeping gene-encoded proteins as internal standards to normalize samples for comparisons of different leukocyte populations from subjects of different ages. In this study, we determined that GAPDH was a more reliable internal control than others in Western blot analysis for comparing the development of immunity among infants, children, and adults.  相似文献

In sub-confluent cultures of Balb/c-3T3 cells, pinocytosis rates were increased after exposure to specific growth factors (serum; platelet-derived growth factor, PDGF; epidermal growth factor, EGF). Conversely, as cells became growth-inhibited with increasing culture density, there was a corresponding decline in pinocytosis rate per cell. In order to test whether density-inhibition of pinocytosis was influenced either by the growth cycle or by cell contact independently of growth, cells were induced into a quiescent state at a range of subconfluent and confluent densities. Under such conditions, cell density did not significantly inhibit pinocytosis rate. When confluent quiescent cultures in 2.5% serum were exposed to 10% serum, the resulting round of DNA synthesis was accompanied by enhanced pinocytosis per cell, even though the cells were incontact with one another. Furthermore, in a SV40-viral transformed 3T3 cell line, both the growth fraction and the pinocytosis rate per cell remained unchanged over a wide range of culture densities. These studies indicate that density-dependent inhibition of pinocytosis in 3T3 cells appears to be secondary to growth-inhibition rather than to any direct physical effects of cell–cell contact.  相似文献

The optimal conditions for identification of mevalonic acid as the product of 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase are described, as well as the effect of different buffer constituents on the enzyme activity. Under the chosen assay conditions, reductase activity from neonatal chick liver increased with the incubation time up to 60 min and was proportional to the amounts of protein added in a range of 0.1-0.5 mg. The specific activity was maximal in brain and liver and lower in intestine of 6-day-old chicks. Thermostability of hepatic reductase was studied. When microsomal preparations were maintained at 4 degrees C, reductase activity remained unchanged for 6 hr and decreased afterwards. Addition of 50 mM KF to the homogenization medium had no effect on the reductase activity. Similarly, preincubation of microsomal preparations with 105,000 g supernatants in the presence or absence of KF did not significantly increase the reductase activity. These results suggest that HMG-CoA reductase was isolated from neonatal chick in the fully activated form.  相似文献

Cerebroside analogues were synthesized from DL-threo-and DL-erythro-3-phenylserines by the following sequence of reactions: estirification, N-acylation, reduction with sodium bis (2-methoxyethoxy)-aluminum hydride (SMEAH), and condensation with acetobromoglucose followed by deacetylation. Heat-induced conformational changes in lipoxygenase 3 were characterized by differential scanning calorimetry. The positions of the observed transitions were sensitive to the composition of the buffer. In particular, lipoxygenase 3 heated in carbonate buffer at pH 8.0 formed large soluble aggregates. Variable-temperature circular dichroism revealed that the formation of the aggregates was not accompanied by the unfolding of the C-terminal domain, which is composed primarily of alpha-helix. The aggregates were investigated using size exclusion chromatography, native polyacrylamide gel electrophoresis, dynamic light scattering, and electron microscopy. The data were consistent with the formation of roughly spherical particles with an average hydrodynamic radius of 26 nm and an approximate composite molecular weight of 10,000,000 Da. To account for the formation of soluble aggregates from lipoxygenase 3, we propose that hydrophobic amino acid residues are exposed by unfolding of the N-terminal beta-barrel domain of the protein resulting in the formation of protein micelles with a hydrophilic surface composed of the C-terminal domains.  相似文献

Galectin-3 is a galactose-/lactose-binding protein (M(r) approximately 30,000), identified as a required factor in the splicing of pre-mRNA. Immunofluorescence staining revealed that galectin-3 distributes differentially between the nucleus and the cytoplasm, depending on the proliferative state of the cells under analysis. Using digitonin-permeabilized mouse 3T3 fibroblasts, we provide evidence that galectin-3 is rapidly and selectively exported from the nucleus. Although both phosphorylated and nonphosphorylated isoforms of galectin-3 are found in the nuclear fraction, only phosphorylated galectin-3 is identified in the exported fraction, implying that phosphorylation is important for the nuclear export of the protein. The rate of galectin-3 export is temperature dependent and is decreased by the addition of wheat germ agglutinin. More strikingly, galectin-3 export can be inhibited by the addition of leptomycin B, a drug that disrupts the interaction between the leucine-rich nuclear export signal and its receptor, CRM1 (chromosome maintenance region 1). Indeed, a putative leucine-rich nuclear export signal can be found in residues 241-249 of the murine galectin-3 sequence. Finally, gel filtration of the exported material showed that galectin-3 can be found in at least two high molecular weight complexes (approximately 650 and approximately 60 kDa), both of which can be disrupted by lactose.  相似文献

Abstract There are several potential health or nutritional benefits possible from some species of lactic acid bacteria. Among these are: improved nutritional value of food, control of intestinal infections, improved digestion of lactose, control of some types of cancer, and control of serum cholesterol levels. Some potential benefits may result from growth and action of the bacteria during the manufacture of cultured foods. Some may result from growth and action of certain species of the lactic acid bacteria in the intestinal tract following ingestion of foods containing them. In selecting a culture to produce a specific benefit it is necessary to consider not only the wide variation among species of the lactic acid bacteria but also that among strains within a given species. With the possible exception of improving lactose utilization by persons who are lactose maldigestors, no specific health or nutritional claims can yet be made for the lactic acid bacteria.  相似文献

Biocontrol agents have not been previously associated with yak dung. We describe here the isolation of strain QM3 from Qinghai yak dung in China. In vitro antagonistic assay showed that this strain could suppress the growth of various plant pathogens effectively, especially the pathogen Alternaria solani. Based on phenotypic, physiological, biochemical and phylogenetic (16S rDNA) studies, strain QM3 could be classified as a strain of Bacillus subtilis. Both OD660 and inhibition effect of eight different kinds of culture media presented strongly significant difference with 0.006 (P < 0.01) and 0.003 (P < 0.01), respectively. Culture media YPF-Ca were selected from eight different kinds of culture media according to OD660 and inhibitory effect. Different thermal treatments on the crude supernatant showed the antibiotic activity presented no significant (P = 0.54, P > 0.05) with ANOVA at the 5% level when temperature was no higher than 100°C. These results were similarly in agreement with the reported thermo-resistance of iturin antibiotics produced by Bacillus. This is the first report on the isolation of Bacillus subtilis from the yak dung.  相似文献
