The Atlantic cod (Gadus morhua) is an important natural resource for northern societies and is now also considered to be a promising candidate for aquaculture. In recent years, much effort has been directed towards the development of genomic tools, and genome initiatives for Atlantic cod have been established. Despite the growing attention devoted to the Atlantic cod genomics, basic aspects of its genome structure and organization remain unknown. Thus, the present work aims to study cytogenetic features of the Atlantic cod as a contribution to the knowledge of this species' genome. The Atlantic cod displays a diploid number of 46 chromosomes, with a karyotypic formula 16 m/sm + 30 st/t. Conventional karyotyping was improved by chromosomal mapping of two classes of repetitive sequences. 18S rDNA clusters were assigned to pairs 2 and 4; small amounts of 18S rDNA clusters were occasionally detected on pair 5. These findings could not be related to the geographical origin of the specimens, but were consistent with the variability of these repeated genes in fish in general. 5S ribosomal gene clusters, apparently corresponding to a single 5S rDNA class, were detected on twelve chromosomes (pairs 11, 12, 14, 17, 20 and 21). The present update of the existing but meagre information on the karyotype of Atlantic cod, plus the first physical mapping of repetitive genes in this species herein, opens the way for an integrated approach that combines genetic and physical mapping with the assembly of the genome of this commercially important species. 相似文献

The influence of feeding behaviour on growth of Atlantic cod (Gadus morhua,Linnaeus, 1758) in the North Sea 总被引:1,自引:0,他引:1 下载免费PDF全文
The objective of this study was to resolve key mechanisms driving individual growth patterns of Atlantic cod (Gadus morhua). Growth dynamics were analysed by linking growth patterns with stomach content composition and environmental temperature. Samples were collected in August/September of the years 2009, 2010 and 2011 in the north‐eastern part of the central North Sea. Prey selection was assessed by identification of individual prey items in the stomach content to species. Ten feeding groups were identified consisting of individuals with one prey type dominating their stomach contents (≥75% by mass), of which six were used for growth analyses: “Sandeel”, “Clupeids”, “Norway pout”, “Flatfishes”, “Crustaceans” and “Brittle stars”. For each group, growth patterns were estimated based on measures of otolith growth increments. The stomach contents showed that cod as a species are opportunistic in their prey selection, but at the same time indicated that the total, broad feeding niche width of the population is dominated by individual diet specialization and that many individuals temporally show a preference for a particular prey type. The contribution of invertebrates and particularly crustaceans decreased with increasing cod size, whereas that of fish and predominantly herring increased. Prey type had a significant effect on growth, while temperature had no effect. Slowest growth was observed in the cod group preying on sandeel, while cod preying on Norway pout showed the fastest growth. No significant difference was observed between groups preying on brittle stars, crustaceans, flatfishes and herring. Growth in the year before capture did however not differ between any of these groups. Across sampling years, growth chronology patterns were similar but not significantly influenced by temperature. 相似文献

Identified were suitable dosages of tetracycline hydrochloride (TET) in three single treatments and three combined treatments of TET with 2 mg/kg strontium chloride (STR) in wild western Baltic cod (Gadus morhua), in terms of (a) obtainable mark qualities (visibility of fluorescent bands), (b) growth assessment, and (c) induced mortality rates. Isotonic NaCl solution was injected in a control group (25 cod per treatment). The results provide the basis for imperative age validation studies of Baltic Sea cod. Cod originating from pound nets near Fehmarn Island were kept in swimming net cages at the harbor of Warnemünde for 1.5 months. Mean initial total length was 28(±3) cm (salinity: 13, water temperature: 13 to 8°C). Overall average growth of surviving cod was 0.8 mm/day. In single TET treatments, lowest mortality rates and best mark quality were observed for TET concentrations of 100 compared to 50 and 25 mg/kg wet mass. Mortality rates of the 100 mg/kg treatment group were remarkably lower than in the control group emphasizing the antibiotic effect of TET. By contrast, the double treatment in the TET‐STR groups resulted in a binding interaction between both markers in the fish body causing either the antibiotic potency being inhibited or TET and STR forming a non‐beneficial chelate (increased mortality), and decreased incorporation of TET in the otolith (reduced visibility of TET bands). Consequently, TET (short‐term marker) and STR (long‐term marker) should not be injected together. Our results demonstrate that the binding interactions between these substances known from homoiotherm animals also apply for poikilotherms such as fish. 相似文献

Growth and muscle cellularity of diploid and triploid Atlantic cod (Gadus morhua Linnaeus, 1758) larvae 下载免费PDF全文
The aim of this study was to compare somatic growth and muscle fibre development in diploid and triploid siblings of Atlantic cod (Gadus morhua Linnaeus, 1758) during the larval stage. Newly hatched larvae were transferred into 200‐L tanks, three tanks per ploidy group (70 larvae L?1, continuous light, gradually increasing seawater temperature 7–11°C and flow rates 50–117 L h?1). Larvae were fed rotifers from 2 to 22 days post hatch (dph), Artemia 19–31 dph and weaned onto a microparticulate diet from 26 dph until the end of the experiment. Measurements of growth (dry weight, standard length) and muscle cellullarity were taken at intervals between 1 and 44 dph. Ploidy groups showed a similar performance throughout the trial, although a marked stagnation in growth was observed for triploids during the weaning from Artemia onto dry feed. Overall, diploid and triploid cod larvae showed a similar development in muscle fibre growth pattern during the experimental period. For both groups, the total number of fast muscle fibres showed a 10‐fold increase (from 384 to 3462), whereas the diameter of fast fibre increased from 8.9 to 13.3 μm (mean number from all treatments). Thus, a temporary but significant effect of triploidy on fast muscle fibre growth pattern was observed in 19 dph larvae in terms of fibre size and number, with triploids showing larger mean fast fibre diameter (11.62 ± 0.63 vs. 10.05 ± 0.34) and a lower number of fibres with a diameter <5 μm than their diploid siblings. Thus, this was found to be related to larvae size and to the differences in total fast fibre cross sectional areas rather than to ploidy status. Overall, our results suggest possible deficiencies in nutrients’ digestion and absorption of triploid cod larvae particularly during the transitional period from live food to inert diets. 相似文献

In 2014, the International Council for the Exploration of the Sea (ICES) was unexpectedly unable to provide an analytical assessment of eastern Baltic cod stock; factors such as data issues, assessment methodology, and the ecological situation of cod were indicated as the reasons for this failure. Some evidence suggests that the natural mortality (M) of cod could increase substantially in forthcoming years and that the selectivity could change. In this paper, age‐structured and stock‐production assessment models were applied to simulate the dynamics of cod stock; in the models, both constant and increasing natural mortalities were permitted. In the age‐structured model, the effects of selectivity related to the cod size on the cod assessment were also analysed. In addition, stock with characteristics similar to Baltic cod stock and increasing natural mortality was generated and assessed with the age‐structured model using both constant and increasing M. It was shown that models with increasing natural mortality of cod in recent years perform much better than models with constant natural mortality in terms of the distribution of residuals and retrospective patterns. The models with size‐dependent selectivity did not perform better than other standard assessments. The assessment of generated stock (where natural mortality was increasing) with constant natural mortality in the assessment model showed a poor distribution of residuals and strong retrospective patterns, similar to the ICES assessment with constant M. The conducted simulations strongly suggest that the main reason for the poor recent cod assessment is the increase in natural mortality, which is not considered in the assessment methodology. 相似文献

Acoustic tags and receivers were used to investigate the spatial ecology of coastal Atlantic cod Gadus morhua (n = 32, mean fork length: 50 cm, range: 33–80 cm) on the Norwegian Skagerrak coast in 2012. Monthly home ranges (HR), swimming activity and depth use varied considerably among individuals and through the months of June, July and August. HR sizes for the period ranged from 0·25 to 5·20 km2 (mean = 2·30 km2). Two thirds of the tagged G. morhua were infected with black spot disease Cryptocotyle lingua parasites; these fish had larger HRs and occupied deeper water compared with non‐infected fish. The infected fish also tended to be more active in terms of horizontal swimming. From an ecological and evolutionary perspective, any environmental change that modifies G. morhua behaviour may therefore also alter the parasite load of the population, and its conservation and fishery status. 相似文献

A study was conducted in 2000 and 2003, following the collapse of the commercial fishery in 1990, to compare metazoan parasites of Atlantic cod Gadus morhua, captured off coastal Labrador, with samples taken in 1980 and 1986. Fish were captured by otter trawl offshore in the North Atlantic Fish Organisation subarea 2J. Parasites were removed from the digestive tract, stained, identified and compared between the different groups. Both the prevalence and mean abundance of trematodes, larval nematodes and E. gadi were significantly lower in fish taken in 2000 and 2003 than in 1980. While mean values of trematodes and nematodes declined in 1986, those of Echinorhynchus gadi remained unchanged in 1986 and 1990. Four-year-old cod sampled in 1990 harboured significantly fewer E. gadi than older age groups. The most commonly occurring trematodes included Podocotyle reflexa, Lepidapedon elongatum, Derogenes varicus and Hemiurus levinseni while the larval nematode, Anisakis sp. was predominant. Comparison of offshore samples taken in 2000 and 2003 with others taken in previous years suggests an overall decline of parasites coincident with a change in climatic conditions, the absence of a major food source, namely capelin Mallotus villosus, of cod and ultimately the decline of the Labrador population. 相似文献

The effects of the putative neurotransmitters acetylcholine, adrenaline, adenosine, ATP, bombesin, 5-hydroxytryptamine, met-enkephalin, neurotensin, somatostatin, substance P and VIP have been investigated in the perfused intestine of the cod, Gadus morhua. The presence and distribution of the different types of nerves was investigated with immunohistochemistry and Falck-Hillarp fluorescence histochemistry. A spontaneous rhythmic activity of the perfused preparations usually occurred within a few minutes from the start of the experiment. This activity was diminished or abolished by addition of atropine, methysergide or tetrodotoxin to the perfusion fluid. Acetylcholine, 5-hydroxytryptamine or substance P caused a contraction of the intestinal wall. The response to acetylcholine was blocked by atropine but not by tetrodotoxin, while the response to 5-hydroxytryptamine was blocked by methysergide and usually also by tetrodotoxin. This indicates that the effect of acetylcholine is direct on the muscle cells, while the effect of 5-hydroxytryptamine may be at least partly via a second neuron. All adrenergic agonists (adrenaline, isoprenaline and phenylephrine) had a dominating inhibitory effect on the intestine. Experiments with antagonists showed that the inhibition is due to stimulation of both alpha-adrenoceptors and beta-adrenoceptors. ATP, adenosine and somatostatin also caused a relaxation of the intestinal wall, often followed by a contraction. Met-enkephalin produced variable responses, either a relaxation, a contraction or both. Bombesin caused a weak inhibition, if anything. The spectral sensitivity of the cod was determined under both dark adapted and light adapted conditions in the laboratory. Cod were trained by cardiac conditioning to detect a difference in radiance between an image of spots and the background radiance of a screen. Thresholds for this response were measured for a range of different wavelengths, and expressed as quantum adjusted values. Electroretino‐graphic studies were also performed on the eyes of cod, and spectral sensitivity curves prepared. Under dark adapted conditions both the behavioural and e.r.g. derived curves showed greatest sensitivity in the blue/green at 490 nm, matching the absorption curve for rhodopsin. A secondary peak in the behaviourally derived curve in the green/yellow at 550 nm indicated that a population of yellow cones may be implicated with the rods in scotopic vision. Under light adapted conditions the behavioural curves showed a shift to the blue, perhaps indicating an adaption to the high red content of the illuminating source. The e.r.g. curve showed greatest sensitivity to blue/green, as in the scotopic experiments but with an enhanced response at 550 nm, indicating greater cone activity. It is suggested that there is complex interaction between rods and cones in the cod retina, both types of receptor being active over a wide range of light intensities. 相似文献

enzyme activity in different parts of the digestive tract was shown to be dependent on the phase of the digestive process.
High concentrations of the chitin degradation product N-acetyl-D-glucosamine were determined in the stomach and in the intestinal
contents. Based on the chitin concentration in the food organisms and the individual food uptake, the amount of chitin consumed
by each fish could be calculated. Only up to 9% of the ingested chitin was recovered from the intestinal contents of the fish
at any given time after feeding (6, 12, 24 and 48 h). In addition, only 2.4% of the chitin consumed with the food could be
recovered in the collected faeces of the fish. The 4 cod killed 48 h after feeding had completely emptied their stomach. Chitin

Two cod stocks (western Baltic cod, WBC, and eastern Baltic cod, EBC) are managed in the Baltic Sea which is characterized by different main spawning areas and different main spawning periods. In this study we analyse the spatial and temporal occurrence of spawning individuals of both cod stocks in the main spawning grounds of the Baltic Sea based on eight microsatellite loci. Our results suggest that EBC (Gadus morhua callarias) has formed currently temporally stable, substantially homogeneous population not only in the Bornholm Sea (ICES SD: 25) but also in the Arkona Sea (ICES SD: 24). The presented analyses proved that EBC (G. m. callarias) can temporarily also spawn in the Belt Sea. 相似文献

Using histological and biochemical methods, ovaries of the Barents Sea cod Gadus morhua were studied in different physiological states (immature fish, normal maturation, omission of spawning). Among the fish missing spawning, two categories are identified: without signs of occyte maturation and with massive resorption of maturing oocytes. The maximal activity of lysosomal and cytoplasmic proteolytic enzymes was revealed in the fish ovaries with histological signs of massive oocyte resorption. In the ovaries of maturing fish the highest concentrations of water-soluble proteins was found. The molecular-mass protein composition and the fraction responsible for the cytosol proteinase activity were determined by electrophoresis and high-performance liquid chromatography (HPLC). The obtained results allow some suggestions to be made about mechanisms of degradation of cellular proteins and functions of proteinases in various physiological states of the cod gonads. 相似文献

In a preliminary study, 121 individually tagged juvenile Atlantic cod (Gadus morhua) were classified according to their haemoglobin genotypes into four groups, i.e., two main haemoglobin genotypes [Hb-I(1/2), Hb-I(2/2)] and two sub-types [Hb-I(1/2b), Hb-I(2/2b)], and reared for 3 months at 10 degrees C, 13 degrees C and T-step (fish reared at 16 degrees C and then subsequently moved to 13 and later to 10 degrees C). Overall growth rates across temperatures were 10% and 19% higher in the Hb-I(2/2b), Hb-I(1/2b) sub-types compared to corresponding Hb-I(2/2) and Hb-I(1/2) main types, respectively. Individual growth rate trajectories varied between the genotypes at all temperatures studied. Our study indicates that under certain environmental conditions higher growth in the two sub-types compared to the main genotypes could be expected. This may indicate difference in other physiological characters not studied here, but seen in previous studies, i.e., oxygen affinity and competitive performance. 相似文献

Aspects of the reproduction of reared cod, Gadus morhua L., with special emphasis on the females, were studied under laboratory conditions. The fecundity and condition factor were 2–5 and 1–5 times, respectively, that of wild cod. A total of 18 spawning females were kept in separate tanks/ chambers, each with one or two males. Seven of the 18 females were classified as stressed, based upon behaviour, irregular spawning intervals and low fertilization rates of the eggs. The reared cod were found to spawn 17–19 batches. The number of eggs liberated in each batch normally followed a smooth, dome-shaped curve with time. The fertilization rate was normally 100%. Egg size decreased from first to last batch and the egg dry weight decreased by about 20–30%. The reared cod showed the same egg diameter to dry weight relation as wild cod. Egg diameter of first batch and maternal fish length were significantly positively correlated. The mean spawning interval for the female and the mean water temperature during its spawning were negatively correlated. The reared cod spawned in both the night and the day for about 50–60 days. 相似文献

Individual behaviour of Atlantic cod Gadus morhua in the presence of hypoxic water was measured in situ in the vertically stratified Bornholm Basin of the Baltic Sea. Considering all recaptured individuals, the use of hypoxic habitat was comparable to data derived by traditional survey data, but some G. morhua had migrated towards the centre of the c. 100 m deep basin and spent about a third of their time at oxygen saturation <50%, possibly to forage on zoobenthos. Maximal residence time per visit in such hypoxic water was limited to a few hours, allowing for the digestion of consumed prey items in waters with sufficient dissolved oxygen. 相似文献
