Summary It is demonstrated that sequential resonance assignment of the backbone 1H and 15N resonances of proteins can be obtained without recourse to the backbone amide protons, an approach which should be useful for assignment of regions with rapidly exchanging backbone amide protons and for proteins rich in proline residues. The method relies on the combined use of two 2D experiments, HA(CA)N and HA(CACO)N or their 3D analogs, which correlate 1H with the intraresidue 15N and with the 15N resonance of the next residue. The experiments are preferably conducted in D2O, where very high resolution in the 15N dimension can be achieved by using 2H decoupling. The approach is demonstrated for a sample of human ubiquitin, uniformly enriched in 13C and 15N. Complete backbone and 13C/1H resonance assignments are presented.  相似文献

Removing agricultural cellulosic residues from fields for the production of ‘second generation biofuels'has the potential to profoundly alter C and N cycling in soil, increasing the risk of soil organic matter depletion and favoring soil–atmosphere gaseous exchanges. However, these negative impacts could potentially be offset by amending the soil with the solid by‐product which is generated during bioethanol production. In a 100 days laboratory study, we investigated the fate of C and N after soil amendment with doubly labeled (13C, 15N) wheat residue (WR) and the corresponding bioethanol by‐product (i.e. nonfermentable wheat residue NFWR) with and without extra N addition. Substituting WR with the corresponding amount of recovered bioethanol by‐product partially compensated the C losses of full crop residue removal. When the equivalent amount of C was added as WR and NFWR, NFWR‐derived C was found in significantly higher proportion in macroaggregates in soil (17.0 vs. 8.9%) after 100 days. Addition of both WR and NFWR reduced soil organic C (SOC) mineralization, i.e. it caused a negative priming effect in soil. However, this pattern was reversed when extra N was added. Both WR and NFWR increased the proportion of soil water‐stable macroaggregates from 16% (in control) to 20–24% (in the different treatments). The results suggest that the more recalcitrant compounds derived from bioethanol production may stabilize more strongly and persist within the protected fractions of SOM pools. Our study demonstrates that NFWR, compared with WR application, neither increased N2O emissions nor had a negative impact on aggregate formation in the midterm. This demonstrates that NFWR has potential for replenishing SOC stocks.  相似文献

Epidemiological studies corroborate a correlation between pesticide use and Parkinson's disease (PD). Thiocarbamate and dithiocarbamate pesticides are widely used and produce neurotoxicity in the peripheral nervous system. Recent evidence from rodent studies suggests that these compounds also cause dopaminergic (DAergic) dysfunction and altered protein processing, two hallmarks of PD. However, DAergic neurotoxicity has yet to be documented. We assessed DAergic dysfunction in Caenorhabditis elegans (C. elegans) to investigate the ability of thiocarbamate pesticides to induce DAergic neurodegeneration. Acute treatment with either S‐ethyl N,N‐dipropylthiocarbamate (EPTC), molinate, or a common reactive intermediate of dithiocarbamate and thiocarbamate metabolism, S‐methyl‐N,N‐diethylthiocarbamate (MeDETC), to gradual loss of DAergic cell morphology and structure over the course of 6 days in worms expressing green fluorescent protein (GFP) under a DAergic cell specific promoter. HPLC analysis revealed decreased DA content in the worms immediately following exposure to MeDETC, EPTC, and molinate. In addition, worms treated with the three test compounds showed a drastic loss of DAergic‐dependent behavior over a time course similar to changes in DAergic cell morphology. Alterations in the DAergic system were specific, as loss of cell structure and neurotransmitter content was not observed in cholinergic, glutamatergic, or GABAergic systems. Overall, our data suggest that thiocarbamate pesticides promote neurodegeneration and DAergic cell dysfunction in C. elegans, and may be an environmental risk factor for PD.  相似文献

Temporal trends of N2O fluxes across the soil–atmosphere interface were determined using continuous flux chamber measurements over an entire growing season of a subsurface aerating macrophyte (Phalaris arundinacea) in a nonmanaged Danish wetland. Observed N2O fluxes were linked to changes in subsurface N2O and O2 concentrations, water level (WL), light intensity as well as mineral‐N availability. Weekly concentration profiles showed that seasonal variations in N2O concentrations were directly linked to the position of the WL and O2 availability at the capillary fringe above the WL. N2O flux measurements showed surprisingly high temporal variability with marked changes in fluxes and shifts in flux directions from net source to net sink within hours associated with changing light conditions. Systematic diurnal shifts between net N2O emission during day time and deposition during night time were observed when max subsurface N2O concentrations were located below the root zone. Correlation (P < 0.001) between diurnal variations in O2 concentrations and incoming photosynthetically active radiation highlighted the importance of plant‐driven subsoil aeration of the root zone and the associated controls on coupled nitrification/denitrification. Therefore, P. arundinacea played an important role in facilitating N2O transport from the root zone to the atmosphere, and exclusion of the aboveground biomass in flux chamber measurements may lead to significant underestimations on net ecosystem N2O emissions. Complex interactions between seasonal changes in O2 and mineral‐N availability following near‐surface WL fluctuations in combination with plant‐mediated gas transport by P. arundinacea controlled the subsurface N2O concentrations and gas transport mechanisms responsible for N2O fluxes across the soil–atmosphere interface. Results demonstrate the necessity for addressing this high temporal variability and potential plant transport of N2O in future studies of net N2O exchange across the soil–atmosphere interface.  相似文献

Because N is frequently the most limiting mineral macronutrient for plants in terrestrial ecosystems, modulating N input may have ecological consequences through trophic levels. Thus, in agro‐ecosystems, the success of natural enemies may depend not only from their herbivorous hosts but also from the host plant whose qualities may be modulated by N input. We manipulated foliar N concentrations by providing to Camelina sativa plants three different nitrogen rates (control, optimal, and excessive). We examined how the altered host‐plant nutritional quality influenced the performances of two aphid species, the generalist green peach aphid, Myzus persicae, and the specialist cabbage aphid, Brevicoryne brassicae, and their common parasitoid Diaeretiella rapae. Both N inputs led to increased N concentrations in the plants but induced contrasted concentrations within aphid bodies depending on the species. Compared to the control, plant biomass increased when receiving the optimal N treatment but decreased under the excessive treatment. Performances of M. persicae improved under the optimal treatment compared to the control and excessive treatments whereas B. brassicae parameters declined following the excessive N treatment. In no‐choice trials, emergence rates of D. rapae developing in M. persicae were higher on both optimum and excessive N treatments, whereas they remained stable whatever the treatment when developing in B. brassicae. Size of emerging D. rapae females was positively affected by the treatment only when it developed in M. persicae on the excessive N treatment. This work showed that contrary to an optimal N treatment, when N was delivered in excess, plant suitability was reduced and consequently affected negatively aphid parameters. Surprisingly, these negative effects resulted in no or positive consequences on parasitoid parameters, suggesting a buffered effect at the third trophic level. Host N content, host suitability, and dietary specialization appear to be major factors explaining the functioning of our studied system.  相似文献

用本实验室构建的重组鸡痘病毒rFPV-H5HA-IL18、rFPV-H5HA-H7HA-IL18和rFPV-H5HA,经翼蹼免疫1日龄SPF鸡和7日龄商品Leghorn蛋鸡,同时以H5亚型AIV全病毒灭活疫苗作为对照.免疫后测定HI抗体效价、淋巴细胞转化指标、重组疫苗对增重的影响、免疫后的攻毒保护效力、免疫后的抑制排毒情况.免疫后不同时间分别测定特异性抗体和淋巴细胞刺激指数.试验结果表明,3株重组鸡痘病毒株均能诱导鸡体产生血凝抑制抗体(HI);共表达鸡IL-18的rFPV-H5HA-IL18和rFPV H5HA-H7HA-IL18诱导商品蛋鸡的细胞免疫水平明显高于非共表达鸡IL-18的rFPV-H5HA.重组鸡痘疫苗免疫SPF和商品蛋鸡后第21d进行攻毒实验,rFPV-H5HA-IL18和rFPV-H5HA-H7HA-IL18免疫攻毒保护率达10/10,rFPV-H5HA免疫攻毒保护率达9/10,与常规疫苗相当.免疫的商品蛋鸡于攻毒后7d采集泄殖腔棉试子样品,检测排毒情况.结果表明,rFPV-H5HA-IL18、rFPV-H5HA-H7HA-IL18免疫组在攻毒后第7d无排毒,其抑制免疫鸡排毒效果优于常规疫苗和单独表达HA的rFPV-H5HA重组鸡痘病毒.rFPV-H5HA-IL18和rFPV-H5HA-H7HA-IL18免疫组鸡,在14日龄时的体重明显高于rFPV-H5HA免疫组和常规疫苗对照免疫组,表明共表达的鸡IL-18能降低鸡痘病毒载体对雏鸡增重的影响.  相似文献

目的:利用Bac-to-Bac Baculovirus Expression System表达重组HA蛋白,Western blot及IFA方法鉴定其表达。方法:采用PCR方法扩增A/California/04/2009(H1N1)HA基因,将其克隆到pFastBacHT A载体上,重组质粒pFastBacHT-HA经双酶切及测序鉴定正确后,转化阳性重组载体进入E.coli DH10Bac感受态细胞中,通过Bluo-gal蓝白斑筛选、PCR鉴定获得重组转座子rBacmid-HA。从重组转座子中提取rBacmid-HA质粒DNA转染sf 9昆虫细胞,制备重组杆状病毒。重组杆状病毒感染sf 9细胞表达重组蛋白,Western blot及IFA鉴定重组蛋白表达情况。结论:成功构建了甲型H1N1流感病毒HA基因的昆虫杆状病毒表达载体,该表达载体转染昆虫细胞后制备的重组杆状病毒病毒滴度较高,重组杆状病毒表达的重组蛋白经Western blot 及IFA 鉴定后具有良好的免疫反应原性。  相似文献

本研究自行设计合成两对特异性引物,通过RT-PCR扩增出1株鸽源H5N1亚型禽流感病毒血凝素(HA)和神经氨酸酶(NA)两个基因的cDNA片段,将它们成功克隆于pMD18-T载体上,然后进行序列测定。结果表明,HA基因全长1707bp,编码568个氨基酸, HA基因有7个糖基化位点,在裂解位点附近有连续6个碱性氨基酸(R-R-R-K-K-R)的插入,具有高致病性毒株的分子特征。受体结合位点的氨基酸分别为YWIHELY,左侧壁氨基酸为SGVSSA,右侧壁为NGQSGR;NA基因全长1350bp,编码446个氨基酸,NA基因有3个糖基化位点。  相似文献

目的从人源化噬菌体抗体库(human single fold scFv libraries I+J)中筛选到能高亲和性、特异结合人禽流感病毒H5N1的单链抗体,为建立H5N1快速筛查试剂和人源化治疗单抗奠定基础。方法以H5N1病毒的血凝素(hemagglutitin,HA)蛋白和核蛋白(nucleoprotein,NP)为目的蛋白,对上述单抗噬菌体文库以亲和性为原理进行筛选,经过3轮筛选富集后,随机挑选了96个噬菌体克隆扩增培养,ELISA法挑选能特异性、高亲和性结合目的蛋白的噬菌体克隆,并换用HB2151宿主菌对阳性单链抗体克隆进行可溶性表达,ELISA法鉴定可溶性单链抗体的结合活性,PCR扩增阳性克隆的轻、重链基因片段,并对阳性单链抗体分子测序和序列分析。结果经过3轮筛选,分别从96个噬菌体克隆中挑选到了两株能特异结合NP蛋白、3株能特异结合HA蛋白的单链抗体,PCR扩增都得到了长为300、302和935bp的轻链、重链和轻链-连接片段-重链的基因片段,测序结果分析发现上述5条单链抗体片段在轻链的47、49、50、51、53、54、56、96、97、98和99位的氨基酸组成不同,而特异结合NP蛋白的单链在重链区域氨基酸组成完全相同,而特异结合HA蛋白的单链在重链的44、47、85、86、87、88和89位氨基酸组成不同。结论从噬菌体抗体库中筛选到的特异结合HA和NP蛋白的单链抗体片段,可为进一步研发H5N1快速筛选试剂和人源性治疗抗体奠定基础,也可为鉴定HA和NP蛋白中的抗原决定簇提供结构信息。  相似文献

Earth's atmospheric composition has changed significantly over geologic time. Many redox active atmospheric constituents have left evidence of their presence, while inert constituents such as dinitrogen gas (N2) are more elusive. In this study, we examine two potential biological indicators of atmospheric N2: the morphological and isotopic signatures of heterocystous cyanobacteria. Biological nitrogen fixation constitutes the primary source of fixed nitrogen to the global biosphere and is catalyzed by the oxygen‐sensitive enzyme nitrogenase. To protect this enzyme, some filamentous cyanobacteria restrict nitrogen fixation to microoxic cells (heterocysts) while carrying out oxygenic photosynthesis in vegetative cells. Heterocysts terminally differentiate in a pattern that is maintained as the filaments grow, and nitrogen fixation imparts a measurable isotope effect, creating two biosignatures that have previously been interrogated under modern N2 partial pressure (pN2) conditions. Here, we examine the effect of variable pN2 on these biosignatures for two species of the filamentous cyanobacterium Anabaena. We provide the first in vivo estimate of the intrinsic isotope fractionation factor of Mo‐nitrogenase (εfix = ?2.71 ± 0.09‰) and show that, with decreasing pN2, the net nitrogen isotope fractionation decreases for both species, while the heterocyst spacing decreases for Anabaena cylindrica and remains unchanged for Anabaena variabilis. These results are consistent with the nitrogen fixation mechanisms available in the two species. Application of these quantifiable effects to the geologic record may lead to new paleobarometric measurements for pN2, ultimately contributing to a better understanding of Earth's atmospheric evolution.  相似文献

从GenBank上获得194株不同来源的H5N1亚型禽流感病毒HA基因核酸序列,利用MEGA3分析了HA基N核酸序列碱基突变的特点。并通过比较序列同源性,构建NJ系统进化树,探讨了不同来源的H5N1病毒的系统进化关系。序列分析结果表明,被研究的194条H5N1亚型禽流感病毒HA基因核酸序列,碱基长度大约在1700bp左右,共发现了757个可变位点,其中Parsimony—informative sites有537个,Singleton sites有220个;病毒的变异速率很快,平均变异率为3.23%;病毒的序列变异具有显著的地区特点和时间特点;同时,全球化的贸易以及候鸟的迁徙在传播病毒过程中起一定作用。  相似文献

为了构建更为安全有效能同时抵抗高致病性H5亚型和低致病忡H9亚型禽流行性感冒(禽流感)病毒的基因工程疫苗,将H5和H9亚型禽流感病毒分离株的血凝素(HA)基因,分别由鸡痘病毒早晚期启动子PS和PE/L调控其转求,定向插入鸡痘病毒转移载体p11s中,获得H5A和H9A基因分别处于PS及PE/L启动子转录调控下的重组转移载体p11SH5H9。以FuGene^TM6转染法将p11SH5H9转染至已感染鸡痘病毒282E4疫苗株(wt-FPV)的鸡胚成纤维细胞(CEF)中。p11SH5H9与wt—FPV基因组DNA之间的同源重组产生了重组鸡痘病毒rFPV11SH5H9。通过在含X-gal的营养琼脂上连续挑选蓝色病毒蚀斑获得并纯化rFPV-11SH5H9。以间接免疫荧光法试验证实,纯化的rFPV-11SH5H9感染的CEF能同时表达H5A和H9A。初步的动物试验表明,用10^5PFU的rFPV-11SH5H9免疫无特定病原体(SPF)鸡,免疫后血凝抑制(HI)抗体监测阳性率均为100%(8/8);该重组病毒能显著抑制H9亚型AIV滴鼻、点眼后7日龄SPF鸡从气管和泄殖腔排毒,同时也能抵抗H5亚型AIV肌肉注射后对7日龄SPF鸡致死性攻击,保护率均为100%,显示出一定的应用前景。  相似文献

Avian influenza A H7N7/NL/219/03 virus creates a serious pandemic threat to human health because it can transmit directly from domestic poultry to humans and from human to human. Our previous vaccine study reported that mice when immunized intranasally (i.n) with live Bac-HA were protected from lethal H7N7/NL/219/03 challenge, whereas incomplete protection was obtained when administered subcutaneously (s.c) due to the fact that H7N7 is a poor inducer of neutralizing antibodies. Interestingly, our recent vaccine studies reported that mice when vaccinated subcutaneously with Bac-HA (H7N9) was protected against both H7N9 (A/Sh2/2013) and H7N7 virus challenge. HA1 region of both H7N7 and H7N9 viruses are differ at 15 amino acid positions. Among those, we selected three amino acid positions (T143, T198 and I211) in HA1 region of H7N7. These amino acids are located within or near the receptor binding site. Following the selection, we substituted the amino acid at these three positions with amino acids found on H7N9HA wild-type. In this study, we evaluate the impact of amino acid substitutions in the H7N7 HA-protein on the immunogenicity. We generated six mutant constructs from wild-type influenza H7N7HA cDNA by site directed mutagenesis, and individually expressed mutant HA protein on the surface of baculovirus (Bac-HAm) and compared their protective efficacy of the vaccines with Bac-H7N7HA wild-type (Bac-HA) by lethal H7N7 viral challenge in a mouse model. We found that mice immunized subcutaneously with Bac-HAm constructs T143A or T198A-I211V or I211V-T143A serum showed significantly higher hemagglutination inhibition and neutralization titer against H7N7 and H7N9 viruses when compared to Bac-HA vaccinated mice groups. We also observed low level of lung viral titer, negligible weight loss and complete protection against lethal H7N7 viral challenge. Our results indicated that amino acid substitution at position 143 or 211 improve immunogenicity of H7N7HA vaccine against H7N7/NL/219/03 virus.  相似文献
