Twenty enzyme loci were examined in the diploid species ofTriticum andAegilops for allelic variation by starch gel electrophoresis. SectionSitopsis, including the five species,Ae. speltoides, Ae. lingissima, Ae. sharonensis, Ae. bicornis andAe. searsii form a close subgroup withAe. speltoides slightly removed from the others.T. monococcum s. lat., was found to be closest to the species of theSitopsis group.Ae. comosa, Ae. umbellulata andAe. uniaristata form a second subgroup withAe. caudata most closely related to these species.Ae. squarrosa appears almost equally related to all of the species, showing no special affinity for any one species group. Nineteen out of twenty loci examined were polymorphic with a mean of 6.7 alleles per locus. Species could be, for most loci, characterized by the presence of predominant alleles. A conspicious genetic characteristic ofTriticum-Aegilops is the sharing of these predominant alleles between species. Within species variation is characterized by a diffuse distribution of secondary alleles.  相似文献

Parasitism ofLygus lineolaris (Palisot de Beauvois) eggs by three species ofMymaridae, Anaphes iole Girault,Erythmelus miridiphagus Dozier andPolynema pratensiphagum Walley and one species ofScelionidae, Telenomus sp., was investigated at Ste-Anne-de-Bellevue, Québec. The maximum level of field parasitism ofL. lineolaris eggs by individual species was 15.4, 53.8, 70.0 and 16,7%, respectively. Parasitoids were retrieved from eggs ofL. lineolaris inserted in stems and branches ofAmaranthus retroflexus L.,Chenopodium album L.,Coronilla varia L.,Rumex obtusifolius L. andSolanum tuberosum L. Amaranthus retroflexus andS. tuberosum sustained a large population ofL. lineolaris and egg parasitism was high enough to consider it as a potential control factor.   相似文献

Genetic divergence in four taxa of three species ofSaurida, S. undosquamis south (S) and north (N) types,S. wanieso andS. elongata caught from the East China Sea, Sea of Hyuga and Tosa Bay, was studied based on allelic frequencies at 23 genetic loci surveyed electrophoretically. Fixed allele substitution was observed at eight loci between the S and N types ofS. undosquamis and their genetic distance was 0.5582, within a range of differentiation at the species level. The S type ofS. undosquamis was found to inhabit the Sea of Hyuga and off Cape Ashizuri along the Kuroshio Current, in addition to the East China Sea. The low level of genetic variation found for this type was discussed in relation to its restricted habitat at the edge of the continental shelf. These factors, along with some morphological characters, indicate that the two types ofS. undosquamis should be recognized as distinct species.  相似文献

The genetic frequencies of 9 isozyme loci have been estimated in 23 samples of 4 species ofSecale by means of starch gel electrophoresis. The populations ofS. silvestre andS. vavilovii were monomorphic and uniform within each species, those ofS. montanum andS. cereale were polymorphic for most of the isozyme loci. On the basis of isozyme patterns as well as allelic and genotypic frequencies of isozyme loci,S. silvestre can be distinguished fromS. vavilovii, and both fromS. cereale andS. montanum; but there is no clear differentiation between the two latter species. Clusters constructed from genetic distances separateS. silvestre andS. vavilovii, whereasS. cereale andS. montanum were grouped together. The isozymatic data presented here, along with cytogenetic and life habit data, agree with the generally admitted existence of 4 species inSecale, and support the relationships suggested byKhush & Stebbins (1961).  相似文献

The genetic variation between two allopatric populations ofBulnesia retama and that ofB. schickendantzii andB. foliosa was investigated. The Peruvian population ofB. retama showed low values of P (0.117) and He (0.057) compared to those in the Argentine population with P = 0.59 and He = 0.269;B. schickendantzii showed P = 0.54 andB. foliosa P = 0.17. Genetic identity between the latter was 0.836 and between the allopatric populations ofB. retama was 0.89; the Peruvian population had reduced allelic variation per locus (A = 1.176) in comparison to the Argentine population (A = 1.955). The values of A, P, He andWright's fixation indices suggest that the Peruvian population could have originated from a single or very few migrants from southern latitudes (founder effect).  相似文献

The objectives of the present study were to develop microsatellite markers for the wild strawberry, Fragaria virginiana, to evaluate segregation patterns of microsatellite alleles in this octoploid species, and assess genetic variability at microsatellite loci in a wild population. A genomic library was screened for microsatellite repeats and several PCR primers were designed and tested. We also tested the use of heterologous primers and found that F. virginiana primers amplified products in cultivated strawberry, Fragaria × ananassa Duch. and Fragaria chiloensis. Similarly, microsatellite loci developed from cultivated strawberry also successfully amplified F. virginiana loci. We investigated four microsatellite loci in detail, three developed from F. virginiana and one from cultivated strawberry. A survey of 100 individuals from a population of F. virginiana in Pennsylvania demonstrated high heterozygosities (He or gene diversity ranged from 0.80 to 0.88 per locus) and allelic diversity (12–17 alleles per locus), but individual plants had no more than two alleles per locus. Segregation patterns in parents and progeny of two controlled crosses at these four loci were consistent with disomic Mendelian inheritance. Together these findings suggest that the genome of F. virginiana is "highly diploidized" and at least a subset of microsatellite loci can be treated as codominant, diploid markers. Significant heterozygote deficiencies were found at three of the four loci for hermaphroditic individuals but for only one locus among females in this gynodioecious species.Communicated by J. The genetic relationships ofAechmea mexicana, A. lueddemanniana, A. macvaughii andA. tuitensis were investigated using starch gel electrophoresis. Eight enzyme systems encoded by ten putative gene loci were resolved in seventeen populations.Nei's (1978) genetic distances were obtained from allelic frequencies and used with UPGMA algorithm. Results indicate that some populations belonging to different species display genetic similarities closer to each other than to some conspecific populations. Our results do not support the proposed genusUrsulaea (Read & Baensch 1994), sinceA. tuitensis was closer toA. lueddemanniana andA. mexicana than toA. macvaughii.  相似文献

The intentional introduction of red king crab, Paralithodes camtschatica (Tilesius, 1815) in the Barents Sea represent one of a few successful cases and one that now supports a commercial fishery. Introductions of alien species into new environments are often associated with genetic bottlenecks, which cause a reduction in the genetic variation, and this could be important for the spreading potential of the species in the Atlantic Ocean. Red king crab samples collected in the Varangerfjord located on the Barents Sea (northern Norway) were compared with reference crab samples collected from the Bering Sea and Kamchatka regions in the Pacific Ocean. All samples were screened for eleven microsatellite loci, based on the development of species-specific primers. The observed number of alleles per locus was similar, and no reduction in genetic variation, including gene diversity and allelic richness, was detected between the Varangerfjord sample and the reference sample from Okhotsk Sea near Kamchatka, indicating no genetic bottlenecking at least for the microsatellite loci investigated. The same results were found in comparison with the sample from Bering Sea. The level of genetic differentiation among the samples, measured as overall F ST across all loci, was relatively low (0.0238) with a range of 0.0035–0.1000 for the various loci investigated. The largest pairwise F ST values were found between the Bering Sea and Varangerfjord/Barents Sea samples, with a value of 0.0194 across all loci tested. The lowest value (0.0101) was found between the Varangerfjord and Kamchatka samples. Genetic differentiation based on exact tests on allele frequencies revealed highly significant differences between all pairwise comparisons. The high level of genetic variation found in the Varangerfjord/Barents Sea sample could be of significance with respect to further spreading of the species to other regions in the North Atlantic Ocean.  相似文献

The kuruma prawn, Marsupenaeus japonicus, is a commercially important benthic marine crustacean in East Asia. Understanding the species’ population structures will be very important for its proper stock assessment and management strategy. Herein, 13 polymorphic microsatellite loci originating from expressed sequence tag libraries of M. japonicus were isolated and characterized. Number of alleles per locus ranged from two to 15, observed and expected heterozygosities ranged from 0.250 to 0.906 and from 0.310 to 0.932, respectively. The adequate level of variability within the population renders these microsatellite loci ideal markers for population genetic analysis of M. japonicus.  相似文献

Eight polymorphic microsatellite loci were isolated and characterized for the Cardinal fish (Apogon imberbis), a coastal-reef fish endemic to the Mediterranean Sea. Characterization of 30 Cardinal fish individuals form the western Mediterranean showed moderate to high allelic diversity ranging from 6 to 19 alleles per locus. Two loci showed significant departures from Hardy-Weinberg equilibrium presumably due to null alleles. No evidence of linkage disequilibrium was found for any locus pairwise comparasions. This microsatellite set could be useful for any basic population genetic studies of this species.  相似文献

We collected eggs and larvae of the Japanese temperate bass, Lateolabrax japonicus, and present horizontal and temporal changes of distribution relative to development and growth during the species pelagic life history in Ariake Bay. Sampling was conducted from the inner to central region (11 sampling stations) of Ariake Bay using a plankton net (80 cm diameter, 0.5-mm mesh) from November 2000 to February 2001. Both eggs and larvae were collected most abundantly in mid-December. The CPUE of eggs in the surface layer was higher than the middle layer, which is in contrast to that at the larval stage. Most eggs were collected around the central and western regions of the bay. The distribution of eggs shifted vertically to the middle layer with development. Yolk-sac larvae were collected in the central region of the bay, and preflexion and flexion larvae were more abundantly collected in the inner region of the bay. The body length of larvae around the inner bay was larger than in the central region. The pelagic life history can be summarized as follows: eggs are distributed around the central region of the bay and eggs and larvae expand their distribution to the inner and shallower waters with growth. We conclude that the shift of vertical distribution in pelagic stages and the hydrographic features of the middle layer form one of the mechanisms enabling the inshore migration of L. japonicus.  相似文献

Atlantic salmon,Salmo salar L., from four European locations show allelic variation at one of three triose-phosphate isomerase (TPI) loci (TPI-3*) when separated on horizontal starch gel electrophoresis, using either eye or liver extracts. Two common alleles (*100 and*103) and one rare allele (*97) segregate atTPI-3* with unambiguous typing being possible by observing the interlocus heterodimers. Family studies demonstrate thatTPI-3* 100 and*103 are of autosomal location and are inherited in a Mendelian fashion.TPI-3* variation can also be typed in adipose fin tissue, allowing nondestructive tissue sampling. Three loci are also active in brown trout,Salmo trutta, with two individuals being homozygous forTPI-3*, as are a small number ofS. salar from eastern Canada. The presence of this additional variable allozyme locus inS. salar is important, since genetic studies in that species have been limited by the low level of allozyme variability detectable.  相似文献

Four different populations segregating for one of the two closely linked (possibly allelic) tomato disease resistance genes to the fungusCladosporium fulvum,Cf-4 andCf-9, were generated and analysed for recombination frequencies between theCf-genes and restriction fragment length polymorphism (RFLP) loci. The population consisting of F2 progeny from the interspecific crossLycopersicon esculentum carryingCf-9 ×L. pennellii was identified as the most useful for RFLP mapping of theCf-4/9 locus and an RFLP map around this locus was constructed mainly using this population. The two closest markers identified were CP46, 2.6 cM distal, and a group of 11 markers including TG236, 3.7 cM proximal toCf-4/9. A polymerase chain reaction (PCR)-based procedure for the rapid identification of recombination events between these two markers was developed. The regions of foreign DNA introgression surroundingCf-4 andCf-9 in near-isogenic lines were delimited.  相似文献

The genusCrepidiastrum is distributed in East Asia and includes 7 species. In the Bonin Islands, three species ofCrepidiastrum occur, and all of them are endemic to the islands. For detecting the origin and speciation of these endemic species, electrophoretic studies have been done in three endemic species of the Bonin Islands as well as in the remaining four species ofCrepidiastrum, andYoungia denticulata which is considered to be closely related toCrepidiastrum. A total of 386 individuals were sampled from 14 populations. As a result, 17 loci of 10 enzyme systems were resolved and gene frequencies for each population were calculated. The genetic variability was low in island species, as reported in some oceanic island plants. Four groups were recognized in the dendrogram generated by the UPGMA method. The Bonin endemics were clustered together, suggesting a monophyletic origin.C. ameristophyllum andC. linguaefolium were found to be genetically very similar, and this may suggest recent and rapid speciation within the islands.  相似文献

Larvae and juveniles ofLateolabrax japonicus andL. latus occurred from January to May 1986 in the shallow waters of the Shimanto estuary.L. japonicus markedly outnumberedL. latus. Distinct ecological differences were recognized in habitats and food habits between the two species:L. japonicus mainly inhabited eelgrass beds composed ofZostera nana, whileL. latus appeared evenly in both eelgrass beds and non-eelgrass habitats; the former fed on copepods and cladocerans, while the latter fed on copepods and fish larvae. From these habitat and food habit analyses, estuaries were considered to be important as a main habitat forL. japonicus, but not forL. latus. The fact that ecological differences have occurred during the early life stages was inferred to be one of the possible keys to speculate on the speciation of the two species.  相似文献

Fifty fungal types were isolated from the indoor atmosphere of saw mills by exposing Petri plates containing Czapek-dox Agar, Potato-dextrose Agar and Sabouraud Agar media for 5 min. The fungal flora of the outdoor surroundings was also studied for comparison. Species ofAspergilli dominated in the saw mills, being represented by 16 species including one ascosporic form. Other fungi were species ofCladosporium, Alternaria, Curvularia, Penicillium, Fusarium, etc. Variations in the fungal population in different months were also observed. Japanese temperate bass Lateolabrax japonicus juveniles recruit to the surf zone and grow by feeding on commonly occurring coastal copepods. However, little is known about diel and tidal patterns in their migration and feeding habits. We sampled wild juveniles during the neap and spring tides, over periods of 24 h, with small seine nets in the sand flat of the eastern part of Ariake Bay, Kyushu, western Japan. In both the neap and spring tides, abundance of juveniles significantly increased during the daytime, being highest around the time of high tide. The relative gut fullness indices of juveniles drastically increased in the morning (0700–0900) and during the flood tide in the daytime, while major prey copepod (Paracalanus spp.) density in the ambient water was relatively constant. We summarized that L. japonicus juveniles would migrate to the surf zone after sunrise to feed on copepods, and then emigrate from the surf zone after sunset. The migratory behavior of L. japonicus juveniles would be influenced by light (daytime) and feeding activity influenced by both light (morning) and tidal condition (flood tide). The intertidal region of the tidal flat was recognized to be one of the important habitats for L. japonicus during their early life history.  相似文献

Previous studies on allozyme variation in five populations of the grasshopperDichroplus elongatus along a geographical gradient in Argentina revealed a significant degree of population structuring and a significant association between one of the loci (Aat-1) and latitude. As this could not be entirely explained by historical factors, the possible adaptive significance of this locus or loci in linkage disequilibrium was investigated in the present study, with an emphasis on the role of environmental variables correlated with latitude. The present paper reports a study of the geographical organization of allozyme diversity over a wider range of the distribution ofD. elongatus. The relation of allelic frequencies with geographic climatic variables was analysed. We have found (i) that different loci (Aat-1 andPep-1) covary significantly with different variables, (ii) discordance between genetic and geographical distances, (iii) copious gene flow that would mask allelic frequency differences due solely to genetic drift, (iv) and temporal stability of the gradient found. The results suggest that causes other than drift and migration may explain the observed directional patterns of variation inD. elongatus.  相似文献

Two parthenogenic families of unisexual species of Caucasian rock lizards of genusLacerta, L. armeniaca andL. unisexualis, were analyzed by DNA fingerprinting. Inheritance of M13 minisatellite and of (GACA) n , (GATA) n , and (TCC) n microsatellite loci in the first generation of the lizards was studied. M13, (GACA) n , and (TCC) n loci in the families ofL. armeniaca were strictly inherited, as well as M13 and (GACA) n loci in the families ofL. unisexualis: each DNA fragment in the fingerprint patterns of progeny could be detected in the maternal pattern. However, when a (TCC)50 microsatellite probe was applied in the study ofL. unisexualis families, specific DNA fragments with altered mobility were revealed in the progeny patterns, and the frequency of such events was rather high. It might be hypothesized that some of the (TCC) n loci inL. unisexualis genome are highly mutable. Hence, the family analysis allowed us to demonstrate experimentally the presence of genetically unstable loci in genomes of parthenogenic species of vertebrates. The nature and mechanism of the instability of these loci in parthenogenesis remain obscure.  相似文献
