Changes in the concentrations of endogenous free, conjugated and bound polyamine were determined in petals of two different species of rose, viz. Rosa damascena and Rosa bourboniana, from small bud (stage 1) till full bloom (stage 8). High free putrescine and spermidine concentrations were associated with early stages of flower development and then decreased in R. damascena. At full bloom, the concentration of free putrescine was higher than rest of the polyamines measured. A steady increase in conjugated putrescine, spermidine and spermine was observed during entire period of flower development with predominance of conjugated putrescine at full bloom. In R. damascena the bound spermine was higher than rest of the polyamines during full bloom. In R. bourboniana, during the early stages of flower development, similar situation was observed, however, at full bloom, free spermidine concentration was higher than rest of the polyamines. In this species, the concentration of conjugated and bound spermine was higher than rest of the polyamines during full bloom. Polyamine concentrations were generally lower in the petals of R. bourboniana than R. damascena which may be due to genotypic differences. Changes in endogenous abscisic acid (ABA) and phenols were determined in petals of two diverse species of rose, viz., Rosa damascena Mill and Rosa bourboniana Desport during flower development. A progressive increase in free ABA was observed during flower development till full bloom in both the species with higher content of free ABA in Rosa damascena. While bound ABA level increased in Rosa damascena till stage 6, in Rosa bourboniana it continued to increase till full bloom. Acidic phenols increased slowly in both the species till stage 4, but sharply afterwards and no significant differences were noticed during full bloom period. Bound phenols content was higher in Rosa damascena during full bloom period. The significance of these changes in relation to flowering in the two diverse species of rose is discussed.  相似文献

Polyamine contents in xylem (root) and phloem (leaf) exudates in two diverse species of rose, viz. Rosa damascena Mill and Rosa bourboniana Desport, were analyzed before, during, and after flowering in the main flowering season, that is, April–May. Only free putrescine (Put) was detected in the xylem and phloem exudates at these time points, and it was high during the peak flowering period. In phloem, Put content was significantly higher in R. bourboniana than in R. damascena at all three stages; whereas in the xylem exudate it was relatively higher in R. damascena at the peak flowering period. A spray of α-difluoromethylornithine (DFMO), an irreversible inhibitor of the putrescine biosynthetic inhibitor ornithine decarboxylase (ODC), markedly decreased the flowering. This effect was reversed by application of Put alone or in combination with DFMO. 在东海赤潮爆发区域运用围隔生态系实验方法,研究了不同营养盐条件下围隔生态系内多胺浓度变化。结果表明:2010年选用东海原甲藻赤潮爆发处海水,东海原甲藻是各围隔生态系内主要优势种,没有种群演替现象发生。两种营养盐添加方式下各围隔内精胺浓度维持较高水平,都呈现先波折下降后波折上升的趋势,与东海原甲藻的生长变化正好相反;各围隔内腐胺浓度水平较高,变化起伏较大,其中有两个实验组腐胺整体变化趋势与东海原甲藻生长趋势类似;所有围隔内亚精胺浓度最低,波动较小。2011年取用中肋骨条藻赤潮爆发处海水,所有围隔生态系内优势种都发生了从中肋骨条藻到东海原甲藻的演替。各围隔生态系内腐胺浓度最高,在中肋骨条藻生长初期腐胺浓度下降,随着中肋骨条藻的生长有所上升,实验后期随着东海原甲藻的生长又整体呈现出下降趋势;各实验组精胺浓度较低,在中肋骨条藻消亡东海原甲藻出现的种群演替期间,都呈现出较大波动;各围隔内亚精胺浓度较低,在整个种群演替过程中没有明显的变化。围隔生态系中补充营养盐,通过对浮游植物生长的影响,间接影响围隔生态系内的多胺变化。  相似文献

To understand the factors that induce floral senescence in Hibiscus syriacus L., we have investigated the effects of various chemical agents on flower senescence at two different flowering stages, before and after full bloom, as well as the relationship between flower longevity and endogenous ethylene production before full bloom. Treatments with ethylene, 1-aminocyclopropane-1-carboxylic acid (ACC), and ethephon enhanced floral senescence, while aminoethoxyvinylglycine (AVG) promoted flower longevity regardless of treatment timing. Although ethanol slightly extended flower longevity, abscisic acid (ABA), nitric oxide, boric acid and sucrose, which have been reported to affect flower longevity or senescence, had no effect on H. syriacus floral senescence. The polyamine spermine (SPM), methylglyoxal-bis(guanylhydrazone) (MGBG), an inhibitor of SPM biosynthesis, and cycloheximide (CHI) accelerated flower senescence when applied before full bloom, but had no effect when applied after full bloom. SPM, MGBG and CHI treatments resulted in enhanced ethylene production during flower opening, and the promotion of flower senescence is mediated by ethylene production prior to full bloom. Furthermore, endogenous ethylene, spontaneously produced before blooming, was closely associated with floral senescence. These results suggest that ethylene production during flower opening plays a key role in determining the timing of Hibiscus flower senescence.  相似文献

Polyamines (PAs) retarded the senescence of leaf discs of two diverse speciesof rose viz., Rosa bourboniana andRosa damascena, while polyamine biosynthetic inhibitorsdifluoromethylornithine (DFMO), difluoromethylarginine (DFMA), methylglyoxal-bis(guanylhydrazone) (MGBG) and abscisic acid (ABA) promoted senescence. Sperminewas significantly the most effective polyamine in retarding senescence inR. bourboniana while MGBG and DFMA were more prominent inaccelerating senescence in R. damascena and R.bourboniana respectively. Protein and RNA content were significantlyhigher in polyamine treated leaf discs compared to those treated with polyaminebiosynthetic inhibitors and ABA. Total and reducing sugars decreased under alltreatments while the starch content increased significantly only in polyaminetreated leaf discs. Peroxidase and cellulase activities were retarded bypolyamine treatments and accelerated by polyamine biosynthetic inhibitors andABA. The role of PAs is discussed in relation to senescence.  相似文献

Kinetin at 10 mg l–1 increased the number of flowers produced on Rosa damascena plants while GA3 inhibited flowering. In the leaves of non-flowering plants GA-like activity was high while specific cytokinin activity (fraction-II) was significantly higher in flowering plants. The effect of the exogenous application of polyamines on the flowering induction of the short-day plant Pharbtis nil was investigated. Putrescine, spermidine and spermine applied on the cotyledons of 4-day seedlings had no significant effect on the flowering of this plant under conditions of full induction caused by a 16-hour-long inductive night. Under the conditions of partial induction caused by a 13-hour-long subinductive night, polyamines inhibit or stimulate flowering, depending on the time of application. Also, inhibitors of the biosynthesis of polyamines influenced the flowering process. Analysis of endogenous polyamines revealed significant fluctuations in their content in cotyledons during an inductive night, as well as under continuous light conditions. Particularly large changes occurred in spermidine and spermine levels. The putrescine level in induced seedlings was lower than in non-induced ones. However, induced seedlings contained a higher level of spermine and spermidine. The highest spermidine and spermine levels were observed at the 8th h of the night, although the total concentration of spermine during photoinduction was always 2–3 times lower than that of spermidine. A break in the inductive night, leading to a complete inhibition of flowering, had caused significant changes in the polyamine level by the end of the night. The results suggest that the flowering induction of Pharbitis nil took place at a low putrescine level and increased spermidine and spermine levels.  相似文献

Summary The promotive effect of ethylene inhibitors (Els), i.e. AgNO3 and aminoethoxyvinylglycine (AVG) on de novo shoot regeneration from cultured cotyledonary explants of Brassica campestris ssp. pekinensis cv. Shantung in relation to polyamines (PAs) was investigated. The endogenous levels of free putrescine and spermidine in the explant decreased sharply after 1–3 days of culture, whereas endogenous spermine increased, irrespective of the absence or presence of Els. AgNO3 at 30 M did not affect endogenous PAs during two weeks of culture. In contrast, explants grown on medium containing 5 M AVG produced higher levels of free putrescine and spermine which increased rapidly after three days and reached a peak at 10 days. An exogenous application of 5 mM putrescine also resulted in a similar surge of endogenous free spermine of the explant. More strikingly, shoot regeneration from explants grown in the presence of 1–20 mM putrescine, 0.1–2.5 mM spermidine, or 0.1–1 mM spermine was enhanced after three weeks of culture. However, exogenous PAs generally did not affect ethylene production, and endogenous levels of 1-aminocyclopropane-1-carboxylate (ACC) synthase activity and ACC of the explant. Summary The ability of suspension-cultured rose (Rosa damascena Mill. cv Gloire de Guilan) cells to reduce ferricyanide is decreased by 50% during an overnight incubation in a low-nutrient (1 mM CaCl2, 0.1 mM KCl) solution. This loss is not observed when nitrate and/or glutamate is added to the low-nutrient medium, but it occurs in medium containing all the components needed for normal growth except nitrate plus glutamate. Thus, the cells possess both constitutive and inducible enzymes for the reduction of ferricyanide, and nitrate or glutamate is both necessary and sufficient to stimulate the production of the inducible enzyme.  相似文献

The influence of polyamines, polyamine inhibitors and ethylene inhibitors were tested in Coffea canephora for in vitro morphogenetic response and caffeine biosynthesis. Coffea canephora produced non-embryogenic and embryogenic calli. Somatic embryos were produced only from the embryogenic callus. Endogenous polyamine pools were estimated in these tissues. Somatic embryos were subjected to secondary embryogenesis under the influence of putrescine, silver nitrate and specific inhibitors of polyamine biosynthesis. Estimation of endogenous total polyamines revealed that embryogenic callus contained 11-fold more spermine and 3.3-fold higher spermidine when compared to non-embryogenic callus. Incorporation of polyamines resulted in 58% explant response for embryogenesis when compared to control with 42% response. Incorporation of silver nitrate resulted in 65% response for embryogenesis. Incorporation of polyamine biosynthetic pathway inhibitors DFMO and DFMA resulted in 83% reduction in embryogenic response with concomitant increase in caffeine levels by two-fold as compared to control. These results have clearly demonstrated that polyamines play a crucial role in embryogenesis and caffeine biosynthesis.  相似文献

To elucidate the role of the plant hormones—abscisic acid (ABA) and ethylene during flower senescence in roses, experiments were conducted on two cultivars of cut-roses (Rosa hybrida L.), ‘Grandgala’ and ‘First Red,’ obtained from a commercial grower. An apparent similarity was observed during flower senescence and accumulation of endogenous ABA in petal tissue. Several fold increase in ABA concentration was observed during the later stages of senescence which was found to be associated with a drastic reduction of flower water potential and water uptake. During the later stages of senescence (S5–S6) higher ABA concentration coincides with the elevated concentration of ethylene production. ABA and ethylene both stimulate senescence and are suggested to interact during flower senescence under water limitations.  相似文献

Diurnal vertical profile sampling of the water column, during a fish killing bloom of the raphidophycean alga Heterosigma akashiwo, revealed a phytoplankton population otherwise composed almost entirely of a variety of dinoflagellates. Of these Glenodinium danicum, Dinophysis acuta, Polykrikos schwartzii, Ceratium furca and Gyrodinium spirale were predominant. The distribution of the major species within the phytoplankton were documented and evidence of synchronous vertical migration of H. akashiwo, G. danicum and P. schwartzii was observed. Extracts of shellfish obtained during the bloom and tested by mouse bioassay showed no PSP toxicity but a marginal degree of DSP toxicity. During a subsequent one year phytoplankton monitoring programme another potentially noxious species (Chaetoceros convolutus) appeared and the seasonal reoccurrence of species present during the bloom (e.g. H. akashiwo) was observed. Important year to year differences in the summer phytoplankton (diatom versus flagellate dominated populations) were apparent and analysis of climate data showed that these differences related to different weather conditions prevailing during the two summer periods sampled. The data suggest the fish killing bloom was giving a chance to develop by a prolonged period of warm, calm weather (during which several heavy rainfall events occurred) leading to stable hydrographic conditions (i.e. stratification) and an increase in the retention time of water within the bay.  相似文献

The production of transgenic broccoli (Brassica oleracea) with increased shelf-life using an Agrobacterium rhizogenes-mediated co-transformation protocol is reported. An Agrobacterium rhizogenes Ri vector, pRi1855:GFP was constructed to allow expression of the green fluorescent protein to identify insertion of Ri TL-DNA into plant cells. The Brassica oleracea ACC synthase 1 and ACC oxidase 1 and 2 cDNAs in sense and antisense orientations were co-transformed into GDDH33, a doubled haploid calabrese-broccoli cultivar. Transformation efficiency was 3.26%, producing 150 transgenic root lines, of which 18 were regenerated into mature plants. The floral buds from T0 broccoli heads were assayed for post-harvest production of ethylene and chlorophyll levels. Buds from T0 lines transformed with ACC oxidase 1 and 2 constructs produced significantly less post-harvest ethylene at 20 °C than the untransformed plants and chlorophyll loss was significantly reduced over a 96 h post-harvest period. The T0 plants transformed with sense and antisense ACC synthase 1 had a significantly reduced 24 h post-harvest ethylene peak and delayed chlorophyll loss. A positive correlation between post-harvest bud ethylene production and chlorophyll loss was described by a regression. This demonstrates that the shelf-life of a very perishable vegetable may be increased up to 2 days at 20 °C by reducing post-harvest ethylene production.  相似文献

Berberine production in cell suspension cultures of Thalictrum minus L. var. hypoleucum Miq. was significantly enhanced by administration of spermidine, whereas other polyamines such as cadaverine, putrescine and spermine were ineffective. Polyamines are essential compounds for growth and development in plants. An attempt has been made to find out the endogenous polyamine profiles in various parts and during the ontogeny of fruit formation of two commercially important Coffea species viz., arabica and canephora. Putrescine (Put), spermine (Spm) and spermidine (Spd) are the predominant polyamines during the ontogeny of fruit and their level increased with the advancement of fruit development. However, in the initial stages of flower and fruit development Spm levels were found to be decreased. Elevated levels of major polyamines Put, Spd, and Spm were observed in zygotic embryos than in somatic embryos. Along with this cadavarine (Cad) and other biogenic amines viz., tyramine (Tyr) and tryptamine (Try) were also found during the ontogeny of fruit in C. canephora. In this study the enodogenous polyamine profiles in coffee tissues and beans have been addressed.  相似文献

Changes in the species composition, photosynthesis, calcification and size-fractionated carbon metabolism by natural phytoplankton assemblages were monitored in three mesocosms under different nutrient conditions during May 1993. In the 3 enclosures, the decline of the diatom-dominated assemblages was followed by the development of a bloom of the coccolithoporid Emiliania huxleyi. Highest growth of E. huxleyi was observed in the mesocosm with a high N : P ratio, suggesting this species is a good competitor at low phosphate concentrations. The transition from diatom- to E. huxleyi-dominated assemblages brought about a sharp reduction of the phytoplankton standing stock and carbon-specific photosynthetic rate. The relative contribution of the smaller size fraction to total photosynthesis increased as the succession progressed. Calcification rate and E. huxleyi cell-specified calcite production were highest during the early stages of development of the E. huxleyi bloom. Distinct changes in the patterns of 14C allocation into biomolecules were noticed during the diatom-E. huxleyi succession. The diatom-dominated assemblage showed high relative 14C incorporation into low molecular weight metabolites (LMWM), whereas proteins and, specially, lipids accounted for the largest proportion of carbon incorporation in the E. huxleyi bloom. The patterns of photoassimilated carbon metabolism proved to be strongly dependent on cellular size, as protein relative synthesis was significantly higher in the smaller than in the larger size fraction, irrespective of the nutrient regime and the successional stage. These results are discussed in relation to the ecological and physiological features of small phytoplankton.  相似文献

Metabolomics is the comprehensive and simultaneous identification and quantification of metabolites in living cells. The term metabolome is used to describe the observable chemical profile or fingerprint of the metabolites in a whole tissue. Although being a new approach to study natural compounds, metabolomics uses traditional analytical techniques, including extraction methods, which can be followed by nuclear magnetic resonance (NMR) spectroscopy and multivariate data analysis. Although metabolomics has been successfully applied to quality control issues, the examples of its use for chemosystematics are few. Thus, the analysis of four taxa of Rosa x damascena (R. damascena Mill., R. damascenasemperflorens, R. damascenatrigintipetala and R. duchesse of Portland) was carried out by NMR spectroscopy as a tool for their classification. A principal component analysis of the 1H NMR spectra, based on the metabolites found in organic and aqueous fractions, showed a clear similarity of the samples. In particular, the major contributions from the aqueous fraction, preliminarily considered as a biomarker of R. x damascena group, are the flavonoids kaempferol and quercetin, glycosilated with glucose and rhamnose units. Our analysis demonstrated a close chemotaxonomic correlation among the four taxa, making this method a reliable tool for chemosystematics.  相似文献

Variations in polyamines, proteins and RNA during in vivo gametogenesis and in vitro androgenesis in Datura innoxia and in Nicotiana tabacum were studied. Spermidine was the major polyamine during gametogenesis in both species. Marked differences in proteins, RNA and polyamines were evident during meiosis and at the first haploid mitosis. In Nicotiana an unknown amine (X60) appears at the beginning of the first haploid mitosis. At the same time a rapid increase in the concentrations of RNA and proteins is observed. In Datura, at the time of the first haploid mitosis there is large increase in amine and RNA levels followed by an arginine peak. During androgenesis, putrescine and spermidine were the major polyamines in both species. In Nicotiana during androgenesis an unknown amine (X81) was observed together with putrescine and spermidine. This unknown compound peaks during the developmental stages of embryogenesis. In Datura androgenic induction was marked by an arginine peak followed by an increase in the putrescine and spermidine levels associated with maximum RNA. These biochemical events are tentatively correlated with structural changes during pollen development. The significance of these results is discussed in relation to the role of polyamines during gametogenesis and androgenesis.  相似文献
