Simultaneous intracellular recordings were made from interneurons and from closer or opener mandibular motor neurons in the isolated suboesophageal ganglion of the larva of Manduca sexta. This article describes various morphologically and physiologically distinguishable premotor spiking interneurons which make direct excitatory connections with the motor neurons. In addition, two presumptive non-spiking interneurons make excitatory and inhibitory connections respectively with opener motor neurons. Both classes of interneurons receive excitatory and inhibitory sensory inputs from the mouthparts. Their circuitry and functions are discussed.Abbreviations

We have tested the effect of a known insect neuromodulator, octopamine, on flight initiation in the cockroach. Using minimally dissected animals, we found that octopamine lowered the threshold for windevoked initiation of flight when applied to either of two major synaptic sites in the flight circuitry: 1) the last abdominal ganglion, where wind-sensitive neurons from the cerci excite dorsal giant interneurons, or 2) the metathoracic ganglion, where the dorsal giant interneurons activate interneurons and motoneurons which are involved in producing the rhythmic flight motor pattern in the flight muscles (Fig. 2).Correlated with this change in flight initiation threshold, we found that octopamine applied to the last abdominal ganglion increased the number of action potentials produced by individual dorsal giant interneurons when recruiting the cereal wind-sensitive neurons with wind puffs (Figs. 3, 4, 5) or with extracellular stimulation of their axons (Fig. 6). Octopamine increases the excitability of the giant interneurons (Figs. 7, 8). Also, when we stimulated individual dorsal giant interneurons intracellularly, the number of action potentials needed to initiate flight was reduced when octopamine was applied to the metathoracic ganglion (Fig. 9).Abbreviations

Parallel processing of proprioceptive information in the terminal abdominal ganglion of the crayfish

The processing of proprioceptive information from the exopodite-endopodite chordotonal organ in the tailfan of the crayfish Procambarus clarkii (Girard) is described. The chordotonal organ monitors relative movements of the exopodite about the endopodite. Displacement of the chordotonal strand elicits a burst of sensory spikes in root 3 of the terminal ganglion which are followed at a short and constant latency by excitatory postsynaptic potentials in interneurones. The afferents make excitatory monosynaptic connections with spiking and nonspiking local interneurones and intersegmental interneurones. No direct connections with motor neurones were found.Individual afferents make divergent patterns of connection onto different classes of interneurone. In turn, interneurones receive convergent inputs from some, but not all, chordotonal afferents. Ascending and spiking local interneurones receive inputs from afferents with velocity thresholds from 2–400°/s, while nonspiking interneurones receive inputs only from afferents with high velocity thresholds (200–400°/s).The reflex effects of chordotonal organ stimulation upon a number of uropod motor neurones are weak. Repetitive stimulation of the chordotonal organ at 850°/s produces a small reduction in the firing frequency of the reductor motor neurone. Summary We describe the structures and physiological properties of thirteen kinds of local interneurons in the swimmeret system of the crayfish,Pacifastacus leniusculus. Eight are unilateral, with processes confined to one side of the midline (Figs. 1, 2); five are bilateral, with processes on both sides of the ganglion (Fig. 6). All have most of their branches in the lateral neuropils. All of the unilateral local interneurons were nonspiking; two of the bilateral interneurons generate action potentials. Three kinds of unilateral interneurons could reset the bursting rhythm or could initiate bursting in quiescent nerve cords. Four others drove tonic firing of motor neurons. Four kinds of bilateral interneurons were premotor, and could affect the period and phase of both pattern generators in their ganglion. One unilateral and one bilateral interneuron were sensory interneurons. At least one bilateral interneuron received input from both pattern generators.Different premotor local interneurons function either in pattern generation, or in hemisegmental coordination of groups of motor neurons, or in bilateral synchronization of the ganglionic pairs of local pattern-generators for the swimmerets.Abbreviations

Summary Mutant first instar cockroaches (Periplaneta americana) with supernumerary filiform hair sensilla on their cerci were used to study the effects of cell body position on axonal morphology and synaptic connections. The wild-type cercus has two hairs, one lateral (L) and the other medial (M), each with an underlying sensory neuron. Silver-intensified cobalt fills show that the supernumerary lateral neuron (SIN) in the mutant has the same shape of arborization as L, and electrophysiological recording shows that it forms synaptic connections with the same subset of giant interneurons (GIs) as L in the terminal ganglion: GI3 and GI6. The supernumerary medial neuron (SuM) has the same axonal morphology as M and synapses with the same GIs as does M: ipsilateral GIs 1 and 2 and contralateral GIs 1, 2, 3, 5 and 6. In 0.1% of approximately 8000 animals screened, a supernumerary hair arose on the cereal midline (C hair). The C neuron sends its axon to the CNS in the same branch of the cereal nerve as the L and SIN, and has a similar arborization. However, the C neuron forms synapses with the same GIs as do M and SuM. Electron microscopy of horseradish peroxidase-injected neurons was used to confirm that the C afferent forms a monosynaptic connection to GI2. It was concluded that the position of the sensory neuron cell body does control its axonal morphology and synaptic connectivity, but that these characteristics are produced by independent mechanisms.Abbreviations

Summary About 60 pairs of ascending interneurons are present in the terminal ganglion of the crayfish Procambarus clarkii (Girard). Some of these interneurons have been impaled intracellularly, characterized physiologically, and then labeled with horseradish peroxidase (HRP) to examine the distribution and ultrastructure of synapses. A close relationship between ultrastructure and physiological properties has been found between two types of interneurons, which either have a pre-motor effect upon motor neurons or have no such effect. In one interneuron with a pre-motor effect (6D2), input and output synapses are intermingled on thicker branches, whereas only input synapses are found on small diameter branches. Only input synapses have been observed on the branches in another interneuron with-out a pre-motor effect (6B1). No differences in branch morphology are found in these two interneurons. Interneuron 6D2 contains large numbers of small round agranular vesicles, but the same type of synaptic vesicles is rarely seen in interneuron 6B1, which has no output synapses. Our results indicate a good correlation between the synaptic distribution and pre-motor effects of interneurons in the terminal ganglion.Abbreviations

Summary Computer reconstruction of the antennal lobe of Drosophila melanogaster has revealed a total of 35 glomeruli, of which 30 are located in the periphery of the lobe and 5 in its center. Several prominent glomeruli are recognizable by their location, size, and shape; others are identifiable only by their positions relative to prominent glomeruli. No obvious sexual dimorphism of the glomerular architecture was observed. Golgi impregnations revealed: (1) Five of the glomeruli are exclusive targets for ipsilateral antennal input, whereas all others receive afferents from both antennae. Unilateral amputation of the third antennal segment led to a loss of about 1000 fibers in the antennal commissure. Hence, about 5/6 of the approximately 1200 antennal afferents per side have a process that extends into the contralateral lobe. (2) Afferents from maxillary palps (most likely from basiconic sensilla) project into both ipsi-and contralateral antennal lobes, yet their target glomeruli are apparently not the same as those of antennal basiconic sensilla. (3) Afferents in the antennal lobe may also stem from pharyngeal sensilla. (4) The most prominent types of interneurons with arborizations in the antennal lobe are: (i) local interneurons ramifying in the entire lobe, (ii) unilateral relay interneurons that extend from single glomeruli into the calyx and the lateral protocerebrum (LPR), (iii) unilateral interneurons that connect several glomeruli with the LPR only, (iv) bilateral interneurons that link a small number of glomeruli in both antennal lobes with the calyx and LPR, (v) giant bilateral interneurons characterized by extensive ramifications in both antennal lobes and the posterior brain and a cell body situated in the midline of the suboesophageal ganglion, and (vi) a unilateral interneuron with extensive arborization in one antennal lobe and the posterior brain and a process that extends into the thorax. These structural results are discussed in the context of the available functional and behavioral data.Abbreviations

The cockroachPeriplaneta americana responds to wind puffs by turning away, both on the ground and when flying. While on the ground, the ventral giant interneurons (ventrals) encode the wind direction and specify turn direction, whereas while flying the dorsal giant interneurons (dorsals) appear to do so. We report here on responses of these cells to controlled wind stimuli of different directions. Using improved methods of wind stimulation and of positioning the animal revealed important principles of organization not previously observed.All six cells of largest axonal diameter on each side respond preferentially to ipsilateral winds. One of these cells, previously thought to respond non-directionally (giant interneuron 2), was found to have a restricted directional response (Fig. 3). The organization of directional coding among the ventral giant interneurons is nearly identical to that among the dorsals (Fig. 2). Each group contains, on each side, one cell that responds primarily to wind from the ipsilateral front, another primarily in the ipsilateral rear, and a third responding more broadly to ipsilateral front and rear.These results are discussed in terms of the mechanisms of directional localization by the assembly of giant interneurons.Abbreviations

We investigated the role of retrograde signals in the regulation of short-term synaptic depression and facilitation by characterizing the form of plasticity expressed at novel synapses on four giant interneurons in the cricket cercal sensory system. We induced the formation of novel synapses by transplanting a mesothoracic leg and its associated sensory neurons to the cricket terminal abdominal segment. Axons of ectopic leg sensory neurons regenerated and innervated the host terminal abdominal ganglion forming monosynaptic connections with the medial giant interneuron (MGI), lateral giant interneuron (LGI), and interneurons 7-1a and 9-2a. The plasticity expressed by these synapses was characterized by stimulating a sensory neuron with pairs of stimuli at various frequencies or with trains of 10 stimuli delivered at 100 Hz and measuring the change in excitatory postsynaptic potential amplitude recorded in the postsynaptic neuron. Novel synapses of a leg tactile hair on 7-1a depressed, as did control synapses of cercal sensory neurons on this interneuron. Novel synapses of leg campaniform sensilla (CS) sensory neurons on MGI, like MGI's control synapses, always facilitated. The form of plasticity expressed by novel synapses is thus consistent with that observed at control synapses. Leg CS synapses with 9-2a also facilitated; however, the plasticity expressed by these sensory neurons is dependent on the identity of the postsynaptic cell since the synapses these same sensory neurons formed with LGI always depressed. The neural pathways underlying the processing of signals from locust (Schistocerca gregaria) ovipositor hairs by different classes of interneurones are investigated.Spikes in the sensory neurones from these hairs evoke chemically-mediated, unitary EPSPs with a short and constant latency in six identified non-giant projection interneurones with cell bodies in the terminal abdominal ganglion. Five of these interneurones receive direct inputs from the valves ipsilateral to their neuropilar branches, whereas the other receives direct inputs from valves on both sides. The sensory neurone from a single hair makes divergent connections with several interneurones and those from different hairs make convergent connections with a given interneurone. The amplitude of the EPSPs evoked depends on the position of a hair along the proximal-distal axis of the valve, with sensory neurones from more distal hairs generating larger amplitude EPSPs.Deflection of hairs also excites three of the four giant projection interneurones through polysynaptic pathways and some local interneurones in the terminal abdominal ganglion through monosynaptic connections. Summary A new sensory system, in the abdomen of the cricketAcheta domesticus, is described. It consists of hair-like receptors, which we have called bristles, distributed on the cercus and abdomen. The sensory neurons, innervating bristles of a wide variety of shapes and sizes, project to a common area of the terminal abdominal ganglion. This region is distinct from the area called the cereal glomerulus which receives input from other receptor types.Three interneurons, whose dendrites are located exclusively in the projection area of bristle sensory neurons, are then described. These interneurons respond to tactile stimuli of the cercus and abdomen, but not to infrasound or to body orientation as do previously described interneurons. Based on the anatomical segregation of the afferents and interneurons, as well as the functional distinction from previously described cereal sensory systems, it is becoming clear that the cereal system is a multimodal sensory system. 相似文献

The non-spiking neurons 151 are present as bilateral pairs in each midbody ganglion of the leech nervous system and they are electrically coupled to several motorneurons. Intracellular recordings were used to investigate how these neurons process input from the mechanosensory P neurons in isolated ganglia. Induction of spike trains (15 Hz) in single P cells evoked responses that combined depolarizing and hyperpolarizing phases in cells 151. The phasic depolarizations, transmitted through spiking interneurons, reversed at around -20 mV. The hyperpolarization had two components, both reversing at around -65 mV, and which were inhibited by strychnine (10 micromol l(-1)). The faster component was transmitted through spiking interneurons and the slower component through a direct P-151 interaction. Short trains (<400 ms) of P cell spikes (15 Hz) evoked the phasic depolarizations superimposed on the hyperpolarization, while long spike trains (>500 ms) produced a succession of depolarizations that masked the hyperpolarizing phase. The amplitude and duration of the hyperpolarization reached their maximum at the initial spikes in a train, while the depolarizations persisted throughout the duration of the stimulus train. Both phases of the response were relatively unaffected by the spike frequency (5-25 Hz). The non-spiking neurons 151 processed the sensory signals in the temporal rather than in the amplitude domain. 相似文献

Nonspiking local interneurons in the terminal abdominal ganglion of the crayfish Procambarus clarkii receive inhibitory inputs from mainly glutamatergic spiking local interneurons and GABAergic nonspiking interneurons. In this study, the inhibitory responses of nonspiking interneurons to local application of glutamate and GABA into the neuropil were compared. Glutamate and GABA injection mediated the hyperpolarization of the nonspiking interneurons with an increase in membrane conductance. The glutamate-mediated membrane hyperpolarization was reversed by injection of 1 or 2 nA hyperpolarizing current. By contrast, more than 3 nA hyperpolarizing current was frequently necessary to reverse the GABA-mediated hyperpolarization. Bath application of a chloride channel blocker, 50 microM picrotoxin (PTX), reduced the glutamate-mediated hyperpolarization, but had no effect on the GABA-mediated hyperpolarization. The GABA-mediated hyperpolarization was not consistently affected by bath application of low chloride solution. These results suggest that the glutamate-mediated inhibition was related to the gating of a Cl(-) conductance, while the GABA-mediated inhibition was not. Electrical stimulation of sensory afferents innervating the exopodite elicited ipsps in uropod opener motor neurons. These sensory-evoked ipsps were also PTX-insensitive, suggesting GABAergic nonspiking interneurons could be the predominant premotor elements in organizing the uropod motor control system. 相似文献

Intracellular recordings of mesothoracic common inhibitory neurons (CI1, CI2 and CI3) were made while tactile hairs of the middle legs of locusts (Locusta migratoria) were mechanically stimulated. Generally the three common inhibitory neurons were excited by stimulation of tactile hairs on the ventral and dorsal surface of femur and tibia. The response pattern of all three CI neurons was similar suggesting that they work as a functional unit. Touching hairs on the dorsal surface of tibia and tarsus in some cases led to inhibition of CIs. The connection between sensory cells of tactile hairs and common inhibitory neurons is polysynaptic.To identify interneurons which mediate afferent signals, simultaneous intracellular recordings from CIs and interneurons were made. Different spiking interneurons were identified which made excitatory or inhibitory monosynaptic connections with CIs. Interneurons with inhibitory input to CIs belonged to the ventral midline group of spiking local interneurons. Behavioral and electrophysiological results indicate that reflex movements of the leg are accompanied by activity of CI neurons. Further it appears that CI activity is inhibited when reflex movements of the leg are actively suppressed by the animal.Abbreviations CI

The central projections of sensory neurones innervating a strand chordotonal organ (CO) in the tailfan of the crayfish, Procambarus clarkii (Girard) have been investigated. The CO monitors movement of the exopodite of the tailfan relative to the endopodite. Intracellular recording and staining were used to characterise the response of the sensory neurones to applied stretches of the chordotonal organ and to reveal their morphology. Two gross morphological types of afferents were found: those that terminated in the terminal (6th) abdominal ganglion on the side ipsilateral to the sensory receptor, and those that had branches in the terminal ganglion and an intersegmental axon that ascended rostrally. Afferents responded to position, velocity and direction of imposed CO displacement. Afferents with particular physiological properties had similar morphologies in different crayfish. Irrespective of their directional responses, afferents had central projection areas dependent upon their velocity thresholds. Many afferents responded only during movement of the CO, and those with the lowest velocity thresholds (2°/s) had branches that projected most anteriorly, while those with progressively higher velocity thresholds (up to 200°/s) projected progressively more posteriorly. Afferents that responded to low velocity ramp movements and spiked tonically projected to more posterior areas of the ganglion than those that responded only to movements.Abbreviations A6SCI

Summary The transplantation of appendages from one place to another on the body of crickets (Acheta domesticus) has been used to study the similarities and differences between the sensory systems of various ganglia. Mesothoracic legs have been transplanted to the abdomen in place of a cercus and cerci have been transplanted to thoracic leg stumps. After the ectopic sensory neurons had time to regenerate into the CNS, they were stained and their axonal arborizations examined. The results, which were concerned primarily with bristle receptors, revealed that bristle afferents on ectopic cerci arborized in ventral neuropil (the ventralmost association center) and leg afferents arborized in a ventral anterior region of the terminal abdominal ganglion. The results support the idea that each ganglion contains only a few distinct regions of neuropil (probably three), each receiving separate subsets of the afferent projection.The ectopic cerci were also shown to excite interneurons in the thoracic ganglia whose dendrites were located in the most ventral neuropil. These neurons normally respond to thoracic bristle afferents. Thus, the segregation of afferent axons has a correlate in the interneurons they excite. 相似文献

Summary Direct evidence for monosynaptic connections between filiform hair sensory axons and giant interneurons (GIs) in the first instar cockroach, Periplaneta americana, was obtained using intracellular recording and HRP injection followed by electron microscopy. GIs 1–6 all receive monosynaptic input from at least one filiform afferent axon. GI1, GI2 and GI5 receive input only from the medial (M) axon, while GI3, GI4 and GI6 receive input from both M and lateral (L) axons. The dendrites of GI3 and GI6 which are contralateral to the cell bodies receive input from both axons whereas the smaller ipsilateral dendritic fields have synapses only from the L axon. GI5 has M axon input only onto its contralateral dendrites. In 50% of preparations GI7 receives weak input from the ipsilateral L axon. There is no obvious relationship between the morphology of the giant interneurons and the pattern of input they receive from the filiform afferents.Abbreviations

The morphology and physiology of the large adapting unit (LAU: Fig. 1) and of the small tonic unit (STU: Fig. 2) ofTeleogryllus correspond well to those of the ascending neuron 2 (AN2) and the ascending neuron 1 (AN1) ofGryllus (Figs. 1, 2), respectively.A summary of the ascending auditory interneurons described by various authors in 5 species of crickets is presented in order to establish common identities.Physiological evidence for direct connections between auditory afferents and the ascending auditory interneurons AN1 (STU) and AN2 (LAU) is presented. Simultaneous intracellular recordings from receptors and interneurons in response to sound as well as the activity of auditory interneurons upon electrical stimulation of the tympanal nerve reveal short and constant latencies of receptor-evoked synaptic activity in AN1 (STU) and AN2 (LAU).Abbreviations

The role of non-spiking local interneurones in the synaptic interactions between abdominal extension-evoking descending interneurones
and uropod motor neurones in the terminal abdominal ganglion of the crayfish Procambarus clarkii (Girard) was investigated electrophysiologically. Continuous electrical stimulation of the lateral region of the 3rd-4th
abdominal connective that included abdominal extension evoking interneurones excited the opener motor neurones and inhibited
the closer, reductor motor neurone. Spikes from a single descending interneurone evoked consistent and short latency (0.8–0.9
ms) excitatory postsynaptic potentials (e.p.s.ps) in the opener motor neurones, and evoked rather long-latency (1.5–2.7 ms)
inhibitory postsynaptic potentials (i.p.s.ps) in the reductor motor neurone. Many non-spiking interneurones also received
depolarizing p.s.ps (0.8–2.5 ms in latency) that were usually faster than i.p.s.ps of the reductor motor neurone if both neurones
were recorded sequentially in the same preparation. Non-spiking interneurones received convergent inputs from several descending
interneurones and made inverting connection with the reductor motor neurone. Elimination of descending inputs to a particular
non-spiking interneurone could reduce the inhibitory response of the reductor motor neurone. These observations strongly suggested
that descending inhibitory inputs to the closer, reductor motor neurone were mediated by non-spiking interneurones. Furthermore,
some non-spiking interneurones made output connections with the opener motor neurones. The disynaptic pathway through non-spiking
interneurones is significant to control and modulate the opening pattern of the uropod during abdominal extension.

Manduca sexta molts several times as a larva (caterpillar) before becoming a pupa and then an adult moth. Each molt culminates in ecdysis behavior, during which the old cuticle is shed. Prior to each larval ecdysis, the old cuticle is loosened by pre-ecdysis behavior, which includes rhythmic, synchronous compressions of the abdomen. A previous study indicated that motor neuron activity during pre-ecdysis compression behavior is driven by an ascending neural pathway from the terminal abdominal ganglion. The present study describes a pair of interneurons, designated IN-402, that are located in the terminal ganglion and belong to the ascending pathway. Each IN-402 is synchronously active with pre-ecdysis compression motor bursts, and bilaterally excites compression motor neurons throughout the abdominal nerve cord via apparently monosynaptic connections. The pair of IN-402s appears to be the sole source of rhythmic synaptic drive to the motor neurons during the pre-ecdysis compression motor pattern. These interneurons play a key role in the production of larval pre-ecdysis behavior, and are candidates for contributing to the developmental weakening of pre-ecdysis behavior at pupation.Abbreviations
