GABAergic brain system is an important link in the pathological circuits of rodent audiogenic epilepsy (AE). The number of GABAergic neurons in the inferior colliculi of KM strain rats (AE prone) was not different from that of the control non-prone strain. At the same time, the glutamate decarboxylase mRNA expression (the key enzyme of GABA synthesis) was 5 times higher than in control. The data evidence that the GABAergic system does play an important role in AE proneness.  相似文献

Anxiety (Anx) and depression (Dp) levels were evaluated in rats of 4 lines: 2 of them (KM and “4”) exhibited audiogenic seizures (AS), and 2 (Wistar and “0”) had no AS. In KM rats (with AS), Anx and Dp levels were higher than in Wistars (without AS), while in “4” and “0” rats with the related genetic background but contrasting in AS severity, Anx and Dp indices were not different. Fluoxetine treatment exerted antidepressant effect in all rat lines irrespective of its effect on AS. Thus, phenotypic expression of AS is not directly associated with the mechanisms of Anx and Dp development.  相似文献

Krushinsky-Molodkina rats (KM strain) with genetically determined seizure susceptibility (clonic and tonic seizures in response to the sound of an electric bell, Krushinsky, 1960) were tested in two versions of Morris water maze and compared with normal albino rats (Sprague-Dawley and Wistar). The tests revealed a learning deficit in KM rats. They showed slow acquisition in both the spatial version of the test and the version with the platform, less efficient strategy of searching for target platform, and high scores of floating and thigmotaxis. However, males of KM rats (not females) did not differ significantly from Wistar strain in the probe trial in the spatial variant of the Morris test. No preference for searching for the platform at the place of its previous localization was observed in KM females. Together with our previous findings of the low scores in Revecz-Krushinsky test and data of other authors (Batuev et al., 1983) concerning a working memory deficit in the radial maze, the results suggest the of complex cognitive deficit combined with possible increased stress reactivity in KM rats.  相似文献

目的 :研究饮用加银水对模拟失重大鼠肠道微生态及骨钙代谢的影响。方法 :30只 SD雄性大鼠随机分为地面对照组 ( GC)、模拟失重对照组 ( SC)和模拟失重饮用 0 .2 0 mg/ L 加银水组 ( SS) ,实验期 2 1d。第 7、14和 2 1天用选择性培养基分别对肠球菌、肠杆菌、双歧杆菌、乳杆菌及类杆菌定量测定 ,并测定饲料钙表观吸收率 ;第 2 1天取右侧股骨测骨密度和骨钙含量。结果 :骨密度和骨钙含量 SC组显著低于 GC组 ,SS组也低于 SC组 ;SC组和 GC组比较 ,SC组出现厌氧菌群的减少和需氧菌群的增加及饲料钙表观吸收率的下降 ,且这种变化出现较早 ,SS组和 SC组比较 ,SS组出现以厌氧菌群中 G 菌的减少和需氧菌群G-菌的增加为主的菌群失调及饲料钙表观吸收率的下降 ,且这种变化出现较晚。结论 :饮用 0 .2 0 mg/ L 加银水可以加重模拟失重大鼠肠道微生态的失调和骨钙代谢的紊乱 ,但需要较长的作用时间 ,且其微生态的失调以 G 细菌减少为主的厌氧菌群减少型  相似文献

Objective: The suckling period is one potentially “critical” period during which nutritional intake may permanently “program” metabolism to promote increased adult body weight and insulin resistance in later life. This study determined whether fructose introduced during the suckling period altered body weight and induced changes in fatty acid transport leading to insulin resistance in adulthood in rats. Methods and Procedures: Pups were randomly assigned to one of four diets: suckle controls (SCs), rat milk substitute formula (Rat Milk Substitute), fructose‐containing formula (Fructose), or galactose‐containing formula (Galactose). Starting at weaning, all pups received the same diet; at 8 weeks of age, half of the SC rats began ingesting a diet containing 65% kcal fructose (SC‐Fructose). This continued until animals were 12 weeks old and the study ended. Results: At weeks 8, 10, and 11, the Fructose group weighed more than SC and SC‐Fructose groups (P < 0.05). At weeks 8 and 10 of age, the Fructose group had significantly higher insulin concentrations vs. rats in the SC‐Fructose group. 3H‐Palmitate transport into vesicles from hind limb skeletal muscle was higher in Fructose vs. SC rats (P < 0.05). CD36 expression was increased in the sarcolemma but not in whole tissue homogenates from skeletal muscle from Fructose rats (P < 0.05) suggesting a redistribution of this protein associated with fatty acid uptake across the plasma membrane. This change in subcellular localization of CD36 is associated with insulin resistance in muscle. Discussion: Consuming fructose during suckling may result in lifelong changes in body weight, insulin secretion, and fatty acid transport involving CD36 in muscle and ultimately promote insulin resistance.  相似文献

Using a Western blot method it was been shown that a aontent of beta-CaMKII subunit was decreased on 40 % in hippocampus of Krushinskii-Molodkina rats (rats, genetically prone to audiogenic seizures) in comparison with normal Wistar rats. Additionally, we have investigate the temporal modifications of alpha/beta-CaMKII subunits in dependent from number audiogenic fits (n = 1 or n = 5). The level of beta-CaMKII in hippocampus of naive KM rats and KM rats in 24 h after single audiogenic seizure was not different, but it was increased on 56 % through 72 h after convulsion fit. In contrast, both 5-fold seizures and 20-fold seizures (audiogenic kindling) reduced the beta-CaMKII levels in comparison with naive group. The levels of alpha-subunit protein kinase in hippocampus these experimental groups of animals were not different. We suggest a hypothesis, that modifications alpha/beta ratio can be viewed as specific "homeostatic mechanism", which provide for neuronal function in hippocampus, thereby changing the rules governing synaptic plasticity in dependence from preliminary neuronal activity.  相似文献

Randomly selected adult, male, Sprague-Dawley rats exhibit a range of behaviors in an open field. Exploration without defecation or urination is interpreted as stable behavior. On the basis of their open field behavior we selected the five most “emotional” and five most “stable” rats from two separate groups of thirty rats. Norepinephrine (NE), dopamine (DA), and serotonin (5HT) levels were determined in brains from these ten “emotional” and ten “stable” rats. The NE levels of “emotional” rats were elevated about 60 ng/g relative to the “stable” rats. There was no difference in DA levels, but there appeared to be a trend toward elevation of 5HT levels in the “emotional” rats. These findings directly support the hypothesis that elevated central nervous system norepinephrine levels may reflect a factor which contributes to emotionality in the rat, and suggest that brain norepinephrine levels may be a biochemical mechanism which influences performance as seen with the commonly used open field behavioral test of emotionality.  相似文献

The remote effects of neonatal (on the 3d-to-9th postnatal days) ketamine injections (10 and 50 mg/kg in 20 microliters of distilled water, s.c.) were analyzed in adult Wistar, WAG/Rij, and KM (a strain with high audiogenic sensitivity) rats. Both ketamine and water injections increased pain sensitivity in adult rats. Neonatally injected water increased the mean score of seizures in Wistar and WAG/Rij, whereas ketamine water solution injected in the dose of 50 mg/kg did not change the seizure intensity (as compared to the intact control). Consequently, ketamine significantly reduced the mean score of the audiogenic seizure fit without change in its latency. In highly sensitive KM rats the neonatally injected ketamine (50 mg/kg) significantly shortened the mean latency of the fit onset, and fit stages developed faster. Thus, the neonatal ketamine injection increased the audiogenic seizure susceptibility of brain structures in KM rats.  相似文献

Intestinal inflammation, caused by impaired intestinal homeostasis, is a serious condition in both animals and humans. The use of conventional extracted soybean meal (SBM) in diets for Atlantic salmon and several other fish species is known to induce enteropathy in the distal intestine, a condition often referred to as SBM induced enteropathy (SBMIE). In the present study, we investigated the potential of different microbial ingredients to alleviate SBMIE in Atlantic salmon, as a model of feed-induced inflammation. The dietary treatments consisted of a negative control based on fish meal (FM), a positive control based on 20% SBM, and four experimental diets combining 20% SBM with either one of the three yeasts Candida utilis (CU), Kluyveromyces marxianus (KM), Saccharomyces cerevisiae (SC) or the microalgae Chlorella vulgaris (CV). Histopathological examination of the distal intestine showed that all fish fed the SC or SBM diets developed characteristic signs of SBMIE, while those fed the FM, CV or CU diets showed a healthy intestine. Fish fed the KM diet showed intermediate signs of SBMIE. Corroborating results were obtained when measuring the relative length of PCNA positive cells in the crypts of the distal intestine. Gene set enrichment analysis revealed decreased expression of amino acid, fat and drug metabolism pathways as well as increased expression of the pathways for NOD-like receptor signalling and chemokine signalling in both the SC and SBM groups while CV and CU were similar to FM and KM was intermediate. Gene expression of antimicrobial peptides was reduced in the groups showing SBMIE. The characterisation of microbial communities using PCR-DGGE showed a relative increased abundance of Firmicutes bacteria in fish fed the SC or SBM diets. Overall, our results show that both CU and CV were highly effective to counteract SBMIE, while KM had less effect and SC had no functional effects.  相似文献

We aimed to assess the feasibility of enhancing the intestinal development of weaned rats using glucagon‐like peptide‐2 (GLP‐2)‐expressing Saccharomyces cerevisiae (S. cerevisiae). GLP‐2‐expressing S. cerevisiae (GLP2‐SC) was generated using a recombinant approach. The diet of weaned rats was supplemented with the GLP2‐SC strain. The average daily gain (ADG), the intestinal morphology and the activities of the digestive enzymes in the jejunum were tested to assess the influence of the GLP2‐SC strain on intestinal development. The proliferation of rat enterocytes was also assessed in vitro. The study revealed that the ADG of the weaned rats that received GLP2‐SC was significantly greater than that of the controls fed a basal diet (Control) and S. cerevisiae harbouring an empty vector (EV‐SC) (P < 0.05) but was equivalent to that of positive control rats fed recombinant human GLP‐2 (rh‐GLP2) (P > 0.05). Furthermore, GLP2‐SC significantly increased villous height (P < 0.01) and digestive enzyme activity (P < 0.05) in the jejunum. Immunohistochemistry analysis further affirmed that enterocyte proliferation was stimulated in rats fed the GLP2‐SC strain, as indicated by the greater number of enterocytes stained with proliferative cell nuclear antigen (P < 0.05). In vitro, the proliferation of rat enterocytes was also stimulated by GLP‐2 expressed by the GLP2‐SC strain (P < 0.01). Herein, the combination of the GLP‐2 approach and probiotic delivery constitute a possible dietary supplement for animals after weaning.  相似文献

Since 2008, Drosophila suzukii, the spotted wing drosophila, has colonized most of the American and European continents, being able to adapt to very different climates. In Italy, this pest has been captured from northern Alpine to southern Mediterranean regions where it can infest a broad range of fruits, including wine grapes. Studies have shown that oviposition levels and developmental rates of D. suzukii on wine grapes are lower than on other berries, although recent observations indicate that grapes may become a suitable host plant in particular conditions. Here, we report, for the first time, the results of a series of no‐choice oviposition experiments using berries of five table grape varieties sampled in the provinces of Taranto and Bari (Apulia region, south‐eastern Italy) during 2013 from both organic and conventional farming systems. The sugar content (SC) and skin hardness of each sample were analysed to assess the influence of these physiological parameters on the susceptibility of table grapes to D. suzukii infestation. A negative correlation was found between the number of eggs laid and berry skin penetration force, whereas there was a positive one between the number of eggs and Brix values, as well as sampling date. In organic grapes, SC and skin hardness of two varieties (“Crimson” and “Scarlotta”) were measured, respectively, higher and lower than in conventional grapes, thus making them more susceptible to pest infestation. The study hence shows that in laboratory conditions D. suzukii is able to heavily infest and develop on table grapes and that susceptibility to its infestation significantly depends on both variety and farming system.  相似文献

Overproduction of desired metabolites usually sacrifices cell growth. Here we report that quorum sensing (QS) can be exploited to coordinate cell growth and lactic acid production in Escherichia coli. We engineered two QS strains: one strain overexpressing acyl-homoserine lactone (AHL) synthesis genes (“ON”), the other strain overexpressing both AHL synthesis and degradation gene (aiiA) (“ON to semi-OFF”). To clarify the impact of the QS system on lactic acid production, D-lactate dehydrogenase gene ldhA was deleted from the E. coli genome, and Enhanced Green Fluorescence Protein (eGFP) was used as the reporter. Compared to the “ON” strain, the “ON to semi-OFF” strain showed delayed log growth and decreased egfp expression at stationary phase. When egfp was replaced by ldhA for lactic acid production, compared to the wild-type strain, the “ON to semi-OFF” strain demonstrated 231.9% and 117.3% increase in D-lactic acid titer and space-time yield, respectively, while the “ON” strain demonstrated 83.6%, 31%, and 36% increase in growth rate, maximum OD600, and glucose consumption rate, respectively. Quantitative real-time PCR revealed that both ldhA and the genes for phosphotransferase system were up-regulated in ldhA-overexpressing “ON” strain compared to the strain only harboring QS system. Moreover, the “ON” strain showed considerable increase in glucose consumption after a short lag phase. Compared to the reference strain harboring only ldhA gene in vector, both the “ON” and “ON to semi-OFF” strains demonstrated synchronization between cell growth and D-lactic acid production. Collectively, QS can be leveraged to coordinate microbial growth and product formation.  相似文献

It has been shown that modification of microtubule (MT) ultrastructure are accompanied by functional changes in microtubule-associated protein MAP2 in the hippocampus of Krushinsky--Molodkina rats (KM), which are prone to autogenic seizures. The morphogenetic analysis revealed that contrary to Wistar rats, which are insensitive to sound stimulation, in KM the middle length of microtubule fragments in the apical dendrites of pyramidal neurons in CA3 hippocampal area was reduced. Using immunoblot and autoradiography methods, we found that the level of MAP2 and the rate of its cAMP = and Ca(2+)-calmodulin-dependent phosphorylation were increased in hippocampus of KM, in comparison with Wistar rats. Daily repeated sound stimulation for 20 days (audiogenic kindling) induced a further decrease in length of MT fragments, and an increase of their density in the proximal part of apical dendrites of KM. Moreover, audiogenic kindling induced additional increase in MAP2 phosphorylation state, but did not change the level of MAP2 in KM hippocampus. We suppose that the obtained alteration of MAP2 phosphorylation state exerted influence on kinetic parameters of microtubule assembly, serving as part of genetically determined predisposition of KM to audiogenic epilepsy.  相似文献

巨龙竹( Dendrocalamus sinicus Chia et J. L. Sun)是云南特有的珍稀木本丛生竹,其秆形分为通直型和弯曲型两种变型。为了揭示巨龙竹不同秆形的组织结构特征,本文通过定点观测0 ~ 49 d的巨龙竹笋和幼竹生长发育状况,并采用石蜡切片技术对笋期0 ~ 45 d内样品的组织结构进行比较解剖学研究。结果显示:(1)巨龙竹在笋-幼竹(0 ~ 49 d)发育期,秆高生长呈“慢-快”的趋势,21 ~ 35 d时弯曲型茎秆开始显现,易于辨别;(2)5 ~ 30 d时,弯曲型茎秆中维管束的发育早于通直型;对比弯曲型茎秆内外两侧维管束,内侧维管束导管内径较小,但纤维鞘中的纤维细胞层数更多;(3)弯曲型茎秆中薄壁细胞的分化早于通直型,20 d后弯曲型茎秆中的薄壁细胞出现明显的可被番红-固绿染色的细胞核,并呈有规律的排列;弯曲型茎秆内侧薄壁细胞稍小于外侧薄壁细胞,但内侧被染色细胞核的薄壁细胞多于外侧。研究结果表明巨龙竹弯曲型茎秆性状在笋期第21 ~ 35 d内即可通过茎秆形态判别,弯曲型茎秆中维管束的发育以及薄壁细胞分化均早于通直型茎秆,同一时期弯曲型茎秆内侧细胞分裂较外侧旺盛、维管束木质化程度更高。  相似文献

KM+, a lectin purified from Artocarpus integrifolia seeds, is an attractant for neutrophils, and has properties similar to fMLP, IL-8 and MNCF. The endogenous lectin MNCF, inhibits carrageenan-induced neutrophil migration when intravenously administered in rats. In an attempt to mimic the activity of MNCF with KM+, we determined the effect of intravenous (iv) injection of KM+ (5 g) on neutrophil migration to the peritoneal cavity of Wistar rats induced by KM+ (50 g, intraperitoneal, ip), fMLP (5 ng, ip) and carrageenan (300 g, ip). Initially we evaluated the effect of the time interval between intravenous and intraperitoneal administration of KM+. The intervals ranged from 20 to 120 min and progressively stronger inhibition was observed with increasing time intervals up to a maximum of 60 min, with effect decreasing thereafter. With injections at the optimum interval of 60 min, we observed that KM+ inhibited KM+- and carrageenan-induced neutrophil migration by 72%, and fMLP-induced migration by 56%. White cell counts for Wistar rats that only received KM+iv, performed at 0 to 120 min intervals after injection, revealed early neutropenia lasting 60 min, followed by a marked increase in circulating neutrophils that reached a maximum of twice the initial levels within 90 min and after 120 min returned to levels near to that observed before intravenous administration of KM+. These results indicate that when KM+ is present in the intravascular space, it produces an inhibitory effect on neutrophil migration similar to that caused by the intravenous administration of other chemoattractants, regardless of whether they act through a mechanism independent of carbohydrate recognition, as does IL-8, or are dependent on carbohydrate recognition, like MNCF.  相似文献

A mutant of strain 69–1113a of Neurospora crassa, which shows periodic growth upon both complete and minimal media, was named “clock,” and some of the morphological and genetic differences between this mutant and the “patch” and “wild” strains were investigated. In contrast to the uniform growth of “wild,” the “clock” mutant produces a series of bands formed by cymelike aggregations of hyphae which become progressively more dense and finally mark the end (front) of a growth band. A new growth band is formed by a number of hyphae which grow out as in “wild” strains and dichotomize and form new cymes which again become progressively more dense and finally form a new front. It is shown that “clock” continues its rhythmic growth when cultured in continuous darkness. Some “wild” strains were induced to grow periodically on appropriate media. A medium containing equal quantities of sorbose and sucrose caused strain 65–811A (a “wild”) to produce the “patch” type growth. Random isolation, as well as ordered isolation, of ascospores following a cross between “wild” and “clock” show a 1: 1 segregation indicating that “clock” differs from “wild” by a single gene.  相似文献

The addition of aerobic exercise (AE) to a resistance exercise (RE) program (concurrent exercise, CE) can interfere with maximum muscle fiber growth achieved with RE. Further, CE appears to markedly affect the growth of myosin heavy chain (MHC) I, but not MHC IIa fibers. The mechanism responsible for this "interference" is unclear. Satellite cell (SC) responsiveness to exercise appears to influence muscle adaptation but has not yet been examined following acute concurrent exercise. Thus, we assessed the fiber-type-specific SC response to RE, AE, and CE exercise. Eight college-aged males completed the following two exercise trials: the RE trial, which consisted of unilateral leg extensions and presses (4 sets ≥ 10 repetitions: 75% 1 repetition maximum, RM); and the AE/CE trial, which included an identical RE protocol with the opposite leg, immediately followed by subjects cycling for 90 min (60% W(max)). Muscle biopsies were obtained from the vastus lateralis before and 4 days after each session. Samples were cross-sectioned, stained with antibodies against NCAM, Ki-67, and MHC I, counterstained with DAPI, and analyzed for SC density (SC per fiber), SC activation, and fiber type. SC density increased to a greater extent following RE (38 ± 10%), compared with CE (-6 ± 8%). Similarly, MHC I muscle fiber SC density displayed a greater increase following RE (46 ± 14%), compared with AE (-7 ± 17%) and CE (-8 ± 8%). Our data indicate that the SC response to RE is blunted when immediately followed by AE, at least in MHC I muscle fibers, and possibly MHC II fibers. This suggests that the physiological environment evoked by AE might attenuate the eventual addition of myonuclei important for maximum muscle fiber growth and consequent force-producing capacity.  相似文献

Soybean mosaic virus (SMV) is one of the most broadly distributed soybean (Glycine max (L.) Merr.) diseases and causes severe yield loss and seed quality deficiency. Multiple studies have proved that a single dominant gene can confer resistance to several SMV strains. Plant introduction (PI) 96983 has been reported to contain SMV resistance genes (e.g., Rsv1 and Rsc14) on chromosome 13. The objective of this study was to delineate the genetics of resistance to SMV in PI 96983 and determine whether one gene can control resistance to more than one Chinese SMV strain. In this study, PI 96983 was identified as resistant and Nannong 1138-2 was identified as susceptible to four SMV strains SC3, SC6, SC7, and SC17. Genetic maps based on 783 F2 individuals from the cross of PI 96983 × Nannong 1138-2 showed that the gene(s) conferring resistance to SC3, SC6, and SC17 were between SSR markers BARCSOYSSR_13_1114 and BARCSOYSSR_13_1136, whereas SC7 was between markers BARCSOYSSR_13_1140 and BARCSOYSSR_13_1185. The physical map based on 58 recombinant lines confirmed these results. The resistance gene for SC7 was positioned between BARCSOYSSR_13_1140 and BARCSOYSSR_13_1155, while the resistance gene(s) for SC3, SC6, and SC17 were between BARCSOYSSR_13_1128 and BARCSOYSSR_13_1136. We concluded that, there were two dominant resistance genes flanking Rsv1 or one of them at the reported genomic location of Rsv1. One of them (designated as “Rsc-pm”) conditions resistance for SC3, SC6, and SC17 and another (designated as “Rsc-ps”) confers resistance for SC7. The two tightly linked genes identified in this study would be helpful to cloning of resistance genes and breeding of multiple resistances soybean cultivars to SMV through marker-assisted selection (MAS).  相似文献
