The changes taking place during fruit development in the concentration of the 3 polyamine fractions, i.e. free, perchloric acid-soluble conjugates and perchloric acid-insoluble bound polyamines, were analyzed in tomato fruits ( Lycopersicon esculentum Mill, cv. F121) induced to set by either pollination or auxin application. Before the onset of cell division, total polyamines were 50% higher in auxin-treated fruits than in pollinated ones, most of the polyamines being found as perchloric acid-soluble conjugates in both fruit set treatments. At the onset the level of polyamines in both fruit types was 100 times higher than during cell expansion or ripening. The highest polyamine found during cell division was perchloric acid-soluble conjugated spermidine in both fruit set treatments. After cell division, polyamine concentration was similar in both fruit set treatments. The concentration of polyamines in the jelly was similar in pollinated and auxin-induced fruits during cell expansion but not during ripening. At the onset of ripening there was an increase of one order of magnitude in the concentration of perchloric acid-insoluble bound putrescine in the jelly of pollinated fruits. Polyamines were more than 5-fold higher in unpollinated ovaries than in fruits induced to set by either pollination or auxins. It is suggested that pollinated and parthenocarpic fruits differ in their polyamine metabolism during the initial stages of development, but not after cell division. It is also suggested that polyamines undergo rapid turnover during cell division. Perchloric acid-insoluble bound putrescine might play a role in seed formation in tomatoes.  相似文献

The possible involvement of polyamines during strawberry ( Fragaria × ananassa Duch.) fruit development was investigated. Putrescine, spermidine, and spermine were identified in strawberry receptacles and achenes at all stages of development. Total (free) polyamine levels decreased from a maximum of 485 nmol g−1 fresh weight at pollination to a minimum of 55 nmol g−1 fresh weight in ripe receptacles. Total polyamine concentrations during corresponding stages of development were consistently higher in achenes than in receptacles, and ranged from 891 to 203 nmol g−1 fresh weight. Removal of achenes from the surface of developing receptacles 10 days after pollination reduced receptacle growth, and re-initiation of growth by application of 1 m M α-naphtaleneacetic acid (α-NAA) was accompanied by a rapid increase in polyamine concentrations 24 h after treatment. Polyamine content per receptacle increased >3-fold in normally developing receptacles and in de-achened, auxin-treated receptacles 10 days after removal of achenes, but did not increase during this period in de-achened receptacles not treated with exogenous auxin. α-NAA increased growth and polyamine levels to a greater extent than the structurally related, but less effective auxin, β-NAA. Polyamine concentrations in receptacles with intact achenes remained similar to those of auxin depleted (de-achened) receptacles, implying that the concentration of these compounds may not be limiting following achene removal.  相似文献

The aim of the study was to examine the response of pear (Pyrus communis L.) trees to soil and foliar applications of boron (B). The experiment was carried out during 2000–2001 in a commercial orchard in Central Poland on mature `Conference' pear trees grafted on Pyrus communis var. caucasica seedlings planted at a spacing of 4 × 2.5 m on a sandy loam soil with a low hot water-extractable B status. Annually, foliar sprays with B were applied. (i) before full bloom (at green and white bud stage, and when 1–5% of flowers was at full bloom), (ii) after flowering (at petal fall, and 7 and 14 days after the end of flowering), or (iii) postharvest in fall (approximately 6 weeks before leaf fall). Spray treatments involved application of B at a rate of 0.2 kg ha–1 in spring or 0.8 kg ha–1 in fall. Additionally, other trees were supplied with soil-applied B at the bud break stage at a rate of 2 kg ha–1. Trees untreated with B served as the control. The results revealed that foliar applications of B before full bloom or after harvest increased fruit set and fruit yield. Tree vigor, mean fruit weight, firmness, soluble solids concentration and titratable acidity of fruits at harvest were not affected by B treatments. Foliar B sprays before full bloom or after harvest increased B concentrations in flowers, and both leaves and fruitlets at 40 days after flowering. Only the foliar treatments after flowering and soil fertilization with B increased the content of this microelement in fruit and leaves at 80 and 120 days after full bloom. Foliar B application before full bloom or after harvest increased calcium (Ca) in fruitlets at 40 days after full bloom, in fruit, and in leaves at 80 and 120 days after full bloom. Nitrogen (N), phosphorus (P), potassium (K), and magnesium (Mg) in plant tissues were not affected by B fertilization. After storage, and also after the ripening period, fruits from the trees sprayed with B before full bloom or after harvest had higher firmness and titratable acidity than those from the control trees. After the ripening period, fruits from the trees sprayed with B before full bloom or after harvest had lower membrane permeability and were less sensitive to internal browning than the control fruits. These findings indicate that prebloom and postharvest B sprays are successful in increasing pear tree yielding and in improving fruit storability under the conditions of low B availability in the soil.  相似文献

The polyamine synthesis inhibitor difluoromethylornithine (DFMO) inhibits growth and cell division in tomato fruits (Lycopersicon esculentum Mill. c.v. F 121) when applied after pollination by the split-stem technique. In napthaleneacetic acid (NAA)-induced fruits, however, growth is not inhibited by DFMO. We analyzed the effect of DFMO on cell division in auxin induced tomato fruits, and on the concentration of free, soluble conjugated and insoluble bound polyamines in pollinated and NAA-induced fruits, five days after pollination in the presence and absence of DFMO. Cell number was not significantly reduced by DFMO in NAA-induced fruits five days after fruit set. Free spermine, soluble conjugated and insoluble bound polyamines were higher in fruits treated with DFMO than in those treated with water. They were also higher in DFMO-treated pollinated fruits than in NAA-induced ones. Changes in the concentrations of endogenous free, conjugated and bound polyamine were determined in petals of two different species of rose, viz. Rosa damascena and Rosa bourboniana, from small bud (stage 1) till full bloom (stage 8). High free putrescine and spermidine concentrations were associated with early stages of flower development and then decreased in R. damascena. At full bloom, the concentration of free putrescine was higher than rest of the polyamines measured. A steady increase in conjugated putrescine, spermidine and spermine was observed during entire period of flower development with predominance of conjugated putrescine at full bloom. In R. damascena the bound spermine was higher than rest of the polyamines during full bloom. In R. bourboniana, during the early stages of flower development, similar situation was observed, however, at full bloom, free spermidine concentration was higher than rest of the polyamines. In this species, the concentration of conjugated and bound spermine was higher than rest of the polyamines during full bloom. Polyamine concentrations were generally lower in the petals of R. bourboniana than R. damascena which may be due to genotypic differences. Changes in the concentrations of endogenous polyamines and ethylene were determined in two diverse species of rose viz. Rosa damascena and Rosa bourboniana during post-harvest periods. At full bloom, the concentrations of free putrescine was significantly higher than rest of the polyamines, i.e. spermine and spermidine in both the species. The concentrations of all the polyamines decreased during subsequent periods upto 48 h after full bloom. Similar situation was also observed in conjugated fraction but in bound fraction, during full bloom, the concentration of spermine was higher than rest of the polyamines. In both the species, ethylene showed higher levels during full bloom with maximum in R. damascena, which increased, during post-harvest periods. The possible significance of polyamines, ethylene and their interactions is discussed during post-harvest periods in flowers.  相似文献

Pringlea antiscorbutica R. Br., an endemic crucifer from the Kerguelen Archipelago in the subantarctic, has been previously shown to be unable to acclimatize to 25°C when transferred after several months cultivation under cold conditions. Furthermore, the polyamine composition was greatly modified in such high-temperature-treated plants. The development of seedlings of this species was investigated under a regime mimicking the subantarctic summer thermoperiod (5/10°C night/day) and a regime with high temperatures (22/25°C night/day). In parallel, the associated changes in polyamine composition that occurred during the first 6 days of seedling life were determined. Marked acceleration of seedling growth and intense cotyledon greening were observed at day 4 in 5/10°C-grown seedlings but not in 22/25°C-grown seedlings. Seedlings grown at high temperature accumulated agmatine and putrescine, whereas cold-cultivated seedlings maintained high levels of spermidine. Cold-cultivated seedlings accumulated the uncommon long-chain polyamines norspermidine and homospermidine. These seedlings also accumulated free 1,3-diaminopropane, cadaverine, N1-acetylspermidine, N1-acetylspermine and bound polyamines, whereas seedlings under high temperature accumulated N1-acetylputrescine. Aromatic amine metabolism also appeared to be very responsive to temperature: seedlings under a cold regime accumulated free dopamine and bound phenylethylamine and tyramine, whereas seedlings grown at high temperature accumulated free tyramine. The possible relationships between the observed amine patterns and seedling growth under low and high temperature are discussed.  相似文献

Polyamine oxidase (PAO, EC 1.5.3.3) activity and polyamine content in the cell wall and soluble fractions obtained from embryos, endosperms and shoots and roots of etiolated or green seedlings of maize ( Zea mays L. cv. WF9) during the first 7 days of germination were investigated. Polyamine content was also determined in the trichloroacetic acid-soluble (free polyamines) and trichloroacetic acid insoluble (bound polyamines) fraction obtained from the same tissues. PAO activity, determined by the radiometric method based on the recovery of the labelled reaction product 1-pyrroline, was mostly localized in the cell wall fraction. The activity was very low in embryos and endosperms and present in traces in roots. In etiolated shoots PAO activity increased sharply, while in green shoots it was low and increased slowly. No polyamines were found in the cell wall fraction and only putrescine was detected in the soluble fraction, with the exception of the embryo, where spermidine and spermine were also present. In the TCA-soluble fraction of embryos, putrescine increased during imbibition, while spermidine and spermine decreased; in the endosperm no relevant changes in polyamines occurred. In the same fraction of green and etiolated seedlings, putrescine increased, giving a peak at days 3–5, while spermidine decreased to very low levels. The amount of bound polyamines was 1–4% of the free ones. The pattern of PAO activity seems to be unrelated to endogenous free polyamine content, which is the same in shoots and roots of etiolated and green seedlings. Enzyme activity, very low in ungerminated seeds, increased continuously during the progression of germination, especially in etiolated shoots, indicating a possible involvement in cell wall formation.  相似文献

This is the first report correlating levels of polyamines and its fractions with differentiation in Dictyostelium discoideum. Temporal changes in endogenous levels of free, conjugated and bound putrescine, spermidine and spermine were analysed at critical stages of morphogenesis in this organism. No spermine was found at any given stage and putrescine was the most abundant polyamine. There was a sharp increase in the levels of both free (and total) and conjugated forms of putrescine and spermidine at the slug stage as compared to the growth phase. The levels of putrescine and spermidine were found to be higher in isolated prespore cells as compared to the prestalk cells. Remarkably, the levels of polyamine decreased at the early culminant stage. Data suggest that a moderate level of polyamines is needed for growth but it is important to have high levels of polyamines at the time of differentiation.  相似文献

Six peach and six nectarine cultivars were evaluated for the phenolic content in their pulp and peel tissues. Chlorogenic acid, catechin, epicatechin, rutin and cyanidin-3-glucoside were detected as the main phenolic compounds of ripened fruits. The concentration was always higher in peel tissue, with average values ranging from 1 to 8 mg g−1 dry weight (DW) depending on cultivar. Of the tested varieties, the white-flesh nectarine 'Silver Rome' emerged as the cultivar with the highest amount of total phenolics. Phenolic compounds were also profiled during fruit growth and ripening in the yellow nectarine cv. 'Stark Red Gold', which showed a decreasing concentration during fruit development in both peel and pulp tissues. Average amounts of total phenolics were approximately 25 mg g−1 DW 60 days after full bloom and decreased to 3 mg g−1 DW at ripening in pulp tissue. Differences among peel and pulp composition show the different dietetic and antioxidant potential of fruits consumed unpeeled and peeled.  相似文献

对不同倍性罗汉果果实的生长动态、甙类含量动态及其变化规律的研究。结果表明:多倍体与二倍体果实的生长和甙类动态变化规律基本一致。果实的生长动态均可分为3个时期:迅速生长期(1~20 d)、缓慢生长期(20~30 d)、停止生长期(30~90 d),80 d时果实的形态达到最大值,4x>2x>3x,多倍体表现出巨大性。而甙类含量的动态随日龄的增长均表现为:苦味的甙ⅡE转化为甙Ⅲ,两者依次出现和消失;最终都转化为高甜度的甙Ⅴ,即成熟罗汉果以甙Ⅴ为主;80 d时甙Ⅴ的含量达到最大,4x>3x>2x:多倍体的甜甙含量显著提高,特别是三倍体还具有无籽的特性。  相似文献

We have previously reported that aseptically cultured mesophyll protoplasts of Vigna divide rapidly and regenerate into complete plants, while mesophyll protoplasts of Avena divide only sporadically and senesce rapidly after isolation. We measured polyamine titers in such cultures of Vigna and Avena, to study possible correlations between polyamines and cellular behavior. We also deliberately altered polyamine titer by the use of selective inhibitors of polyamine biosynthesis, noting the effects on internal polyamine titer, cell division activity and regenerative events.In Vigna cultures, levels of free and bound putrescine and spermidine increased dramatically as cell division and differentiation progressed. The increase in bound polyamines was largest in embryoid-forming callus tissue while free polyamine titer was highest in root-forming callus. In Avena cultures, the levels of total polyamines decreased as the protoplast senesced. The presence of the inhibitors -difluoromethyl-arginine (specific inhibitor of arginine decarboxylase) and dicyclohexylamine (inhibitor of spermidine synthase) reduced cell division and organogenesis in Vigna cultures. Addition of low concentration of polyamines to such cultures containing inhibitors or removal of inhibitors from the culture medium restored the progress of growth and differentiation with concomitant increase in polyamine levels.  相似文献

The polyamines (putrescine, spermidine, and spermine) are synthesized by almost all organisms and are universally required for normal growth. Ornithine decarboxylase (ODC), an initial enzyme of polyamine synthesis, is one of the most highly regulated enzymes of eucaryotic organisms. Unusual mechanisms have evolved to control ODC, including rapid, polyamine-mediated turnover of the enzyme and control of the synthetic rate of the protein without change of its mRNA level. The high amplitude of regulation and the rapid variation in the level of the protein led biochemists to infer that polyamines had special cellular roles and that cells maintained polyamine concentrations within narrow limits. This view was sustained in part because of our continuing uncertainty about the actual biochemical roles of polyamines. In this article, we challenge the view that ODC regulation is related to precise adjustment of polyamine levels. In no organism does ODC display allosteric feedback inhibition, and in three types of organism, bacteria, fungi, and mammals, the size of polyamine pools may vary radically without having a profound effect on growth. We suggest that the apparent stability of polyamine pools in unstressed cells is due to their being largely bound to cellular polyanions. We further speculate that allosteric feedback inhibition, if it existed, would be inappropriately responsive to changes in the small, freely diffusible polyamine pool. Instead, mechanisms that control the amount of the ODC protein have appeared in most organisms, and even these are triggered inappropriately by variation of the binding of polyamines to ionic binding sites. In fact, feedback inhibition of ODC might be maladaptive during hypoosmotic stress or at the onset of growth, when organisms appear to require rapid increases in the size of their cellular polyamine pools.  相似文献

Zuili plum (Prunus salicina L.) trees usually set fruit poorly, although they produce high quality fruit. To elucidate the causes of the poor fruit set, pollen tube growth into pistils and fruit set percentage were investigated after cross-, self- and open-pollination. Ovule development in Zuili pistils was also investigated. Pollen tube penetration into the ovules via the obturator and micropyle was best when Zuili pistils were pollinated by cv. Black Amber (P. domestica) pollen grains, although cross-pollinations with Hongxinli and Miili (P. salicina) pollen were more effective than self- and open-pollination. The fruit set percentage was also highest in pistils pollinated with Black Amber pollen grains. Morphological observation of Zuili pistils revealed that the trees produce "double pistils", developing two ovaries from a basal pistil, at a rate as high as 28%. In such abnormal pistils, most ovules were lacking an embryo sac or were entirely degenerated. The percentage of normally developed ovules was 24.3% and 8.9% in normal and double pistils, respectively. From these results, we conclude that the main causes of poor fruit set of Zuili plums are a lack of effective cross-pollination and the production of high percentages of double pistils in which normally developed ovules are scarcely formed.  相似文献

研究了宁夏枸杞不同发育时期果实和种子形态的变化特征及种子内胚的变化.结果表明:宁夏枸杞果实的生长发育曲线为花后8d以前为其第一次快速生长期,花后8~24 d为缓慢生长期,花后24~34 d是第二次快速生长期,属于典型的双“S”型.宁夏枸杞种子的生长曲线既不属于单“S”型,也不属于双“S”型,表现为果实的第一次快速生长期同样也是种子的快速生长期,但种子完成的生长比例快于果实完成的生长比例,此期种子内的胚乳生长快;当果实进入缓慢生长期,种子也表现出缓慢生长的特性,且种子长度和宽度的增加速率均显著低于果实第一次快速生长期种子的生长速率,此期种子主要进行胚的分化;在果实的第二次快速生长期,果实体积和重量迅速增加,而种子的长度和宽度增加很少,此期种子内仅胚进一步增大,从而反映出宁夏枸杞果实的发育与种子发育有一定的相关性.  相似文献

The polyamine profiles of the toxic dinoflagellate Alexandriumminutum during different growth stages were measured by high-performanceliquid chromatography. Both free and conjugated polyamines werefound, including putrescine, cadaverine, spermine, spermidineand norspermidine. During the growth cycle of A. minutum T1,the levels of norspermidine and putrescine in the free polyamineswere the highest after 3 days culture. Putrescine and cadaverinewere the major components in the conjugated polyamines. Theamount of conjugated amines was higher than that of free aminesduring the exponential phase of A. minutum T1.  相似文献
