Rats treated continually and chronically with trifluoperazine (ca 3 mg/kg/day) for six months initially developed mild catalepsy and an inhibition of spontaneous locomotor activity; both effects disappeared by three months. An initial increase in dopamine turnover (as measured by levels of homovanillic acid and dihydroxyphenylacetic acid) also disappeared by three months. Apomorphine-induced stereotypy was completely inhibited in drug-treated animals at two weeks, but progressively returned to normal after three months of drug intake. An exaggerated response to apomorphine developed in animals after six months of drug administration. Inhibition of striatal dopamine-stimulated adenylate cyclase found during the first month of drug intake was reversed at three months, a trend exaggerated after continuous drug administration for six months. Specific striatal 3H-spiperone binding affinity decreased acutely, but was increased after six months drug intake; no change in number of receptor sites occurred.These changes suggest that at least striatal dopamine receptors may become “supersensitive” during chronic neuroleptic treatment.  相似文献

Rats administered chronic neuroleptics for 6–7 weeks-- haloperidol (2.5 mg/rat or 1 mg/kg), clozapine (25 mg/kg), or thioridazine (20 mg/kg)--after termination of chronic drug treatment exhibited greater apomorphine-induced stereotyped behavior than their saline controls. Rats treated with thioridazine or clozapine, but not haloperidol, also showed increases in locomotor activity during withdrawal. These findings indicate that behavioral supersensitivity may develop after chronic clozapine treatment as well as after chronic haloperidol.  相似文献

Rats treated acutely with i.p. fenfluramine showed a profound inhibition of emptying of a chow meal from the stomach. This inhibition was attenuated by previous chronic treatment with fenfluramine. Tolerance to the gastric slowing occured with as few as 2–4 prior injections, a time course which closely matches tolerance to the anorectic effects of the drug. Cerebroventricular injections of fenfluramine and norfenfluramine in anorectic doses also inhibited gastric emptying.  相似文献

Rats treated chronically with haloperidol show evidence of supersensitive dopamine receptors by increased stereotypy when challenged with apomorphine. When such animals are treated acutely with choline chloride at the time of challenge, no changes in stereotypy were observed. Chronic treatment, either during or after induction of supersensitivity, mitigated stereotypy after challenge. This model of dopaminergic supersensitivity is pertinent to the development of tardive dyskinesia in man after treatment with neuroleptic drugs. Our results with choline chloride in the animal model are consistent with their therapeutic action in tardive dyskinesia.  相似文献

Rats were treated for one year with either trifluoperazine dihydrochloride (2.5–3.5 mg/kg/day) or thioridazine dihydrochloride (30–40 mg/kg/day) when prolactin levels were measured in comparison to animals treated acutely with a single oral bolus of the same drugs in approximately the same dose. Serum prolactin levels at the end of the year of neuroleptic treatment with either drug remained elevated compared to those in control animals, and the elevation was no different from that obtained by administration of an equivalent acute single oral bolus. In contrast, the inhibition of apomorphine-induced stereotypy produced by the acute administration of either drug disappeared during chronic treatment, to be replaced after a year's neuroleptic administration by a supersensitive response. Similarly, the increase in dopamine turnover produced by acute neuroleptic administration, evidenced by raised striatal 3, 4-dihydroxy-phenylacetic acid (DOPAC) levels, also disappeared at the end of a year's treatment, when specific binding of 3H-spiperone to striatal homogenates indicated an increased number of dopamine receptors. The disappearance of evidence of blockade of striatal dopamine receptors, which appeared to become supersensitive during a year's chronic treatment with either trifluoperazine or thioridazine, contrasts with the persistence of the effect of these drugs on serum prolactin levels.  相似文献

Benznidazole (bz) is the active component of the antichagasic drug Rochagan. Tests were carried out to detect the induction of chromosomal aberrations and micronuclei in rodent bone marrow cells and peripheral blood cells, respectively. Rats were exposed to acute treatment with Rochagan by gavage at total doses of 150, 300, 1500, 2000 and 3000 mg bz/kg body weight and killed at different times. In the chronic treatments, healthy and chagasic Balb/c mice were treated with Rochagan by gavage at a dose of 100 mg bz/kg/day for 10 and 25 days. No significant increase in frequency of chromosomal aberrations in bone marrow cells or of micronuclei in peripheral blood cells was detected in the animals acutely or chronically exposed to Rochagan in vivo.  相似文献

Recent advances in understanding the molecular pathogenesis of polycystic kidney diseases (PKD) are being translated into promising treatments. Currently, a response to therapy in preclinical animal models of PKD can only be evaluated after several weeks of treatment. The availability of biomarkers for rapid efficacy assessment would greatly facilitate the drug development process. Here we applied SELDI-TOF technology to establish serum and urinary biomarker signatures associated with a rapid therapeutic response to cyclin dependent kinase (CDK) inhibitor roscovitine in the jck mouse model of PKD. A set of 74 serum and 56 urinary markers was identified in the group receiving chronic treatment over 5 weeks. This set was further screened for early efficacy biomarkers in acutely (3-5 days) treated animals with mild (26 days of age) and advanced disease (50 days of age). A third group with intermediate disease (33 days of age) received a single injection to monitor rapid changes in protein profiles within 4, 24 or 48 hours after drug administration. Multifactorial comparative analysis of the acutely treated groups identified a set of 20 urinary and 21 serum efficacy biomarkers. This biomarker signature provides a necessary tool for further assessment of CDK inhibitors as therapeutic agents for PKD.  相似文献

Onconase (Onc), is a novel amphibian cytotoxic ribonuclease with antitumor activity, and is currently in a confirmatory phase III clinical trial for the treatment of malignant mesothelioma. It was recently reported that Rana pipiens oocytes contain still another ribonuclease, named Amphinase (Amph). Amph shows 38 – 40 % amino acid sequence identity with Onc; presents as four variants varying between themselves from 87 to 99 % in amino acid sequence identity and has a molecular mass ~ 13,000. In the present study we describe the effects of Amph on growth of several tumor cell lines. All four variants demonstrated cytostatic and cytotoxic activity against human promyelocytic HL-60-, Jurkat T-cell- and U-937 monocytic leukemia cells. The pattern of Amph activity to certain extent resembled that of Onc. Thus, cell proliferation was suppressed at 0.5 – 10.0 µg/ml (40 – 80 nM) Amph concentration with distinct accumulation of cells in G1 phase of the cell cycle. In addition, the cells were undergoing apoptosis, which manifested by DNA fragmentation (presence of “sub-G1” cells, TUNEL-positivity), caspases and serine proteases activation as well as activation of transglutaminase. The cytotostatic and cytotoxic effects of Amph required its ribonuclease activity: the enzymatically inactive Amph-2 having histidine at the active site alkylated was ineffective. The effectiveness and cell cycle specificity was generally similar for all four Amph variants and at the equimolar concentrations was somewhat more pronounced than that of Onc. The observed cytostatic and cytotoxic activity of Amph against tumor cell lines suggests that similar to Onc this cytotoxic ribonuclease may have antitumor activity and find an application in clinical oncology.  相似文献

The long-term effects of methamphetamine (15 mg/kg, s.c.) on neostriatal enzyme activity were measured in rats treated every 6 hours for 24 hours and subsequently monitored for periods up to 30 days following administration of the drug. Tyrosine hydroxylase was maximally decreased after the first day of drug treatment but the depression persisted throughout a 30-day period. Within 15 hours after methamphetamine treatment was initiated, trytophan hydroxylase was dramatically depressed to 17% of control and was still maximally depressed 7.5 days after the last dose of the drug. In contrast, neostriatal choline acetyltransferase and glutamate decarboxylase activities were not altered acutely nor chronically by methamphetamine administration. It would appear that the toxic effects of methamphetamine reported previously cannot be attributed to a generalized destruction of the neostriatum but are demonstrable only in selected biogenic amine nerve terminals.  相似文献

Male albino rats were administered daily with haloperidol, clozapine or L-DOPA and sacrificed 18 hours after the last dose of the drug. Acutely haloperidol (5mg/kg, i.p.) greatly lowered nigral GABA levels whereas after 167 daily doses the nigral GABA levels were not significantly different from controls, but were significantly increased as compared with the acutely treated animals. In contrast, acute L-DOPA (2 × 100mg, p.o.) greatly raised nigral GABA levels whereas after chronic L-DOPA (167 days) nigral GABA levels were not significantly different from controls and were significantly lower as compared with the animals receiving the acute treatment. Clozapine (20 mg/kg, i.p. either acutely or chronically) did not have as marked an effect on nigral GABA levels as did haloperidol. Of these various drug regimens only chronic L-DOPA significantly affected nigral GAD activity, producing a moderate decrease.  相似文献

The meaning of viral blips in human immunodeficiency virus type 1 (HIV-1)-infected patients treated with seemingly effective highly active antiretroviral therapy (HAART) is still controversial and under investigation. Blips might represent low-level ongoing viral replication in the presence of drug or simply release of virions from the latent reservoir. Patients treated early during HIV-1 infection are more likely to have a lower total body viral burden, a homogenous viral population, and preserved HIV-1-specific immune responses. Consequently, viral blips may be less frequent in them than in patients treated during chronic infection. To test this hypothesis, we compared the occurrence of viral blips in 76 acutely infected patients (primary HIV infection [PHI] group) who started therapy within 6 months of the onset of symptoms with that in 47 patients who started HAART therapy during chronic infection (chronic HIV infection [CHI] group). Viral blip frequency was approximately twofold higher in CHI patients (0.122 +/- 0.12/viral load [VL] sample, mean +/- standard deviation) than in PHI patients (0.066 +/- 0.09/VL sample). However, in both groups, viral blip frequency did not increase with longer periods of observation. Also, no difference in viral blip frequency was observed between treatment subgroups, and the occurrence of a blip was not associated with a recent change in CD4(+) T-cell count. Finally, in PHI patients the VL set point was a significant predictor of blip frequency during treatment.  相似文献

Amphiphysin (Amph) is a src homology 3 domain-containing protein that has been implicated in synaptic vesicle endocytosis as a result of its interaction with dynamin. In a screen for novel members of the amphiphysin family, we identified Amph2, an isoform 49% identical to the previously characterized Amph1 protein. The subcellular distribution of this isoform parallels Amph1, both being enriched in nerve terminals. Like Amph1, a role in endocytosis at the nerve terminal is supported by the rapid dephosphorylation of Amph2 on depolarization. Importantly, the two isoforms can be coimmunoprecipitated from the brain as an equimolar complex, suggesting that the two isoforms act in concert. As determined by cross-linking of brain extracts, the Amph1–Amph2 complex is a 220- to 250-kDa heterodimer. COS cells transfected with either Amph1 or Amph2 show greatly reduced transferrin uptake, but coexpression of the two proteins rescues this defect, supporting a role for the heterodimer in clathrin-mediated endocytosis. Although the src homology 3 domains of both isoforms interact with dynamin, the heterodimer can associate with multiple dynamin molecules in vitro and activates dynamin’s GTPase activity. We propose that it is an amphiphysin heterodimer that drives the recruitment of dynamin to clathrin-coated pits in endocytosing nerve terminals.  相似文献

Rats treated with the trypanocidal drug suramin, a potent inhibitor of several lysosomal enzymes, develop a storage disorder which mimics the features of mucopolysaccharidosis (Constantopoulos et al. 1983). In this paper we have examined the reversibility of the biochemical and pathological changes induced in the liver of the suramin-treated rat. Rats were injected with a single intravenous dose of suramin (250 mg/kg) and allowed to survive for periods of up to 6 months. The liver was examined for suramin content, pathological changes, biochemical storage of glycosaminoglycans (GAGs) and for the blockade of the relevant hydrolytic enzymes. GAG storage in the liver peaked at approximately 14 days after administration of suramin when there was a five-fold increase in the GAG content. Thereafter GAGs decreased in parallel with the fall of suramin concentrations so that within 6 months the content had returned to control levels. The activity of most of the enzymes tested had also returned to control levels within 6 months. The pathological changes which are evident in the liver 1-2 weeks after administration of the drug had diminished considerably within 6 months. These results indicate that significant reversibility of both the biochemical and pathological changes induced by suramin occurs and they support the suitability of the suramin treated rat as a model to assess the value of therapeutic treatments of mucopolysaccharidosis.  相似文献

The effect of L-arginine on the pattern of lipids and lipoproteins in normal and diabetic rats was studied. Three groups of 48 rats were studied during 12 days and compared with a control group (Group I, n = 5). Group I consisted of normal rats not treated with L-arginine. Group II. Normal rats treated with 10 mM L-arginine (i.p.). Group III. Diabetic rats (alloxan 120 mg/kg, i.p.) not treated (diabetic control). Group IV. Diabetic rats treated with 10 mM L-arginine (i.p.). The rats of each group were divided in subgroups of four each. Rats were anesthetized and blood was taken from aorta to determine glucose, triglycerides, cholesterol, total lipids, and low (LDL) and high density lipoproteins (HDL) and their corresponding apoproteins (Apo A-I and Apo B-100). We observed that the alloxan concentration used in this study reproduces the clinical manifestations of disease including hyperglycemia (from 132.5 +/- 7.6 to 544.3 +/- 16.9 mg/dL) in 96 h. As a consequence the levels of triglycerides, cholesterol, total lipids, and LDL and its apoprotein Apo B-100 were increased, whereas HDL and its apoprotein Apo A-I were diminished. The L-arginine injection tends to normalize the glycemia from 24 h; similarly, hyperlipidemia (triglycerides from 924.7 +/- 220.1 to 68.5 +/- 8.4 mg/dL, cholesterol from 107.7 +/- 0.6 to 64.5 +/- 4.2 mg/dL, LDL from 24.2 +/- 2.5 to 8.0 +/- 2.9 mg/dL) was also diminished. These results suggest that the beneficial effect of L-arginine administration on serum glucose values and lipid levels in diabetic rats can be mediated by polyamine formation, although the effect of L-arginine on insulin release as observed by other authors is not discarded.  相似文献

Rats treated chronically with caffeine become tolerant to caffeine-induced stimulation of locomotor activity and cross-tolerant to theophylline. This study was performed to determine if the cross-tolerance between these two methylxanthine drugs is symmetrical. Symmetrical cross-tolerance produced by two different drugs implies a common underlying mechanism of action. Separate groups of rats were given scheduled access to drinking bottles containing either drug-free tap water or 1.0 mg/ml theophylline solution. Daily theophylline intake averaged 59 mg/kg. Dose-effect curves were determined in both control and theophylline-treated groups for 5 drugs: the methylxanthines theophylline and caffeine, a nonxanthine psychomotor stimulant, d-amphetamine, and the adenosine analogs R(-)-N6-2-(phenylisopropyl)adenosine and 5'-(N-ethyl)carboxamidoadenosine. All drugs were injected i.p. and locomotor activity was measured for 30 min beginning 35 min later. Rats that were maintained chronically on theophylline were completely tolerant to the locomotor activity stimulant effects of acutely administered theophylline and cross-tolerant to caffeine-induced stimulation of locomotor activity. In contrast, both control and treated groups were fully responsive to the stimulant effects of d-amphetamine. Low doses of the adenosine analogs produced stimulation of locomotor activity in both groups of rats. Higher doses produced a dose-dependent depression of locomotor activity in control rats; curves for the theophylline-treated rats were shifted to the right of the control curves. Thus, adenosine antagonist activity of theophylline remained evident at a time of complete tolerance to the stimulant effect of the drug on locomotor activity.  相似文献

A cross sectional study was carried out in 200 normal pregnant women between 8-40th weeks of gestation, 25 women during delivery and 25 women 6 weeks after delivery. Plasma and lipoprotein lipids were measured using standard procedures. Apolipoprotein A (Apo A) and Apolipoprotein B (Apo B), were measured by electroimmunoassay. Plasma levels of Apo A were elevated in pregnant women but the elevations were not significant until 17-20 weeks of gestation. Apo A during pregnancy was significantly correlated (p less than 0.001) with high density lipoprotein cholesterol (HDL-C). The level of Apo B increased progressively during pregnancy and it was significantly correlated (p less than 0.001) with total cholesterol (TC), plasma triglycerides (TG) and phospholipids (PL). Apo A and Apo B levels returned to non pregnant values within the puerperium, whereas TC, TG and PL remained significantly elevated above controls (p less than 0.01) 6 weeks post partum.  相似文献

摘要 目的:探讨游离三碘甲腺原氨酸(FT3)/游离甲状腺素(FT4)比值、载脂蛋白B (Apo B)/载脂蛋白A1(Apo A1)比值与急性缺血性脑卒中(AIS)患者临床疗效和出院后短期预后的关系。方法:选取2020年2月至2022年2月安徽省第二人民医院收治的102例AIS患者,所有患者入院后接受血运重建、抗血小板等治疗,根据临床疗效将患者分为有效组(76例)和无效组(26例)。治疗后检测所有AIS患者血清FT3、FT4、Apo B、Apo A1水平,计算FT3/FT4比值、Apo B/Apo A1比值,所有AIS患者出院后随访3个月,根据AIS患者随访期间的预后情况将其分为预后不良组(31例)和预后良好组(71例)。比较不同组间血清FT3、FT4、Apo B、Apo A1水平以及FT3/FT4比值、Apo B/Apo A1比值差异,分析影响AIS患者出院后短期预后的因素。结果:无效组血清FT3水平、FT3/FT4比值低于有效组(P<0.05),血清Apo B水平、Apo B/Apo A1比值高于有效组(P<0.05)。预后不良组血清FT3水平、FT3/FT4比值低于预后良好组(P<0.05),血清Apo B水平、Apo B/Apo A1比值高于预后良好组(P<0.05)。美国国立卫生院神经功能缺损评分(NIHSS评分)高、Apo B/ApoA1比值增高是AIS患者出院后短期预后不良的危险因素(P<0.05),FT3/FT4比值增高是其保护因素(P<0.05)。结论:AIS临床治疗无效和预后不良患者FT3/ FT4比值降低,Apo B/ApoA1比值增高,低FT3/ FT4比值和高Apo B/ApoA1比值与AIS患者出院后短期预后不良有关。  相似文献

Rats were treated chronically with 20 mg/kg/day cocaine (by intraperitoneal injection) for 16 days, followed by 7 days of cocaine wash-out. On the next day, rats were challenged with an acute dose of cocaine administered by one of two routes (systemic or intracranial), and extracellular dopamine (DA) in the nucleus accumbens (Acb) was measured by in vivo microdialysis. Rats acutely challenged systemically with 20 mg/kg cocaine showed enhanced Acb extracellular DA levels (compared to control rats that had not previously received chronic cocaine). However, rats acutely challenged with intracranial cocaine by perfusion of 10−5 M cocaine directly into the Acb did not. It is suggested that both the development and triggering of cocaine sensitization, as manifested by enhanced Acb DA content to subsequent acute cocaine challenge, may involve more than just neural mechanisms occurring locally within the Acb.  相似文献

Rats with systemic carnitine deficiency induced by treatment with trimethylhydraziniumpropionate (THP) develop liver steatosis. This study aims to investigate the mechanisms leading to steatosis in THP-induced carnitine deficiency. Rats were treated with THP (20 mg/100 g) for 3 or 6 weeks and were studied after starvation for 24 h. Rats treated with THP had reduced in vivo palmitate metabolism and developed mixed liver steatosis at both time points. The hepatic carnitine pool was reduced in THP-treated rats by 65% to 75% at both time points. Liver mitochondria from THP-treated rats had increased oxidative metabolism of various substrates and of beta-oxidation at 3 weeks, but reduced activities at 6 weeks of THP treatment. Ketogenesis was not affected. The hepatic content of CoA was increased by 23% at 3 weeks and by 40% at 6 weeks in THP treated rats. The cytosolic content of long-chain acyl-CoAs was increased and the mitochondrial content decreased in hepatocytes of THP treated rats, compatible with decreased activity of carnitine palmitoyltransferase I in vivo. THP-treated rats showed hepatic peroxisomal proliferation and increased plasma VLDL triglyceride and phospholipid concentrations at both time points. A reduction in the hepatic carnitine pool is the principle mechanism leading to impaired hepatic fatty acid metabolism and liver steatosis in THP-treated rats. Cytosolic accumulation of long-chain acyl-CoAs is associated with increased plasma VLDL triglyceride, phospholipid concentrations, and peroxisomal proliferation.  相似文献

The enterochromaffin-like (ECL) cells play an important role in the regulation of gastric acid secretion. They respond to gastrin by a prompt increase in histamine secretion, an effect which is mediated by the CCK-(B)/gastrin receptor acting through the IP(3)/DAG pathway. In the rat, long-term treatment with acid secretion inhibitors induces hypergastrinaemia which, in turn, results in ECL cell hypertrophy and hyperplasia. The aim of the present study was to evaluate various functional parameters in acutely isolated rat ECL cells, following long-term hypergastrinaemia in vivo. Rats were treated with vehicle or a supramaximal daily dose of omeprazole for more than 10 weeks to ensure ECL cell hyperplasia. ECL cells were isolated from vehicle-treated animals and 24, 72 and 120 h after the last dose of omeprazole. The functional activity of the acutely isolated ECL cells was determined by measuring gastrin-and forskolin-induced histamine secretion. Changes in cytosolic free calcium upon gastrin stimulation were monitored by digital video imaging. ECL cells successively regained their ability to respond to gastrin following long-term hypergastrinaemia, reaching close to vehicle-treated levels 120 h after the last dose of omeprazole. In the rat, the response pattern of the ECL cells appears to normalise in parallel with the normalisation of plasma gastrin levels.  相似文献
