 Golgi apparatus of both plant and animal cells are characterized by an extensive system of approximately 30 nm diameter peripheral tubules. The total surface area of the tubules and associated fenestrae is thought to be approximately equivalent to that of the flattened portions of cisternae. The tubules may extend for considerable distances from the stacks. The tubules are continuous with the peripheral edges of the stacked cisternae, but the way they interconnect differs across the stack. In plant cells, for example, tubules associated with the near-cis and mid cisternae often begin to anastomose close to the peripheral edges of the stacked cisternae, whereas the tubules of the trans cisternae are less likely to anastomose and are more likely to be directly continuous with the peripheral edges of the stacked cisternae. Additionally, the tubules may blend gradually into fenestrae that surround some of the stack cisternae. Because of the large surface area occupied by tubules and fenestrae, it is reasonable to suppose that these components of the Golgi apparatus play a significant role in Golgi apparatus function. Tubules clearly interconnect closely adjacent stacks of the Golgi apparatus and may represent a communication channel to synchronize stack function within the cell. A feasible hypothesis is that tubules may be a potentially static component of the Golgi apparatus in contrast to the stacked cisternal plates which may turn over continuously. The coated buds associated with tubules may represent the means whereby adjacent Golgi apparatus stacks exchange carbohydrate-processing enzymes or where resident Golgi apparatus proteins are introduced into and out of the stack during membrane flow differentiation. The limited gradation of tubules from cis to medial to trans offers additional possibilities for functional specialization of Golgi apparatus in keeping with the hypothesis that tubules are repositories of resident Golgi apparatus proteins protected from turnover during the flow differentiation of the flattened saccules of the Golgi apparatus stack. An apparatus was made for automatic replenishment of water lost by evapotranspiration in pot experiments. The system can handle 80 pots. The upper and lower weight limit for each pot, and the weighing frequency can be set. In this way, constant soil moisture levels as well as fluctuations can be arranged. The apparatus can be programmed for continuous cycling, thus minimizing position effects within an experiment. Cumulative daily water use per pot or transpiration per plant is recorded on tape and printed. Special applications, e.g. controlled nutrient dosage and determination of transpiration coefficients are discussed.  相似文献

Complete room temperature fluorescence emission spectra of green and etiolated leaves (Raphanus sativus L., cv. Saxa Treib, Hordeum vulgare L., cv. Villa) are continuously recorded up to 4 min after onset of excitation. In green leaves two emission bands appear, whereas in etiolated leaves only one band is observed. In both cases the emission intensity increases with time, the high-energy band of green leaves decreasing more rapidly than the low-energy band. This phenomenon can be interpreted in terms of energy transfer. During the observation time of the fluorescence induction kinetic no shift of the emission peaks is found within the accuracy of the apparatus (±2nm).  相似文献

Investigations on non-native organisms have become an important task of modern ecology throughout the world. The major objective of this study was to identify the characteristics and habitats of non-native plant species in Korean cities in order to derive conclusions on the success of biological invaders. The city of Chonju in southwest Korea was selected as a representative investigation area. All wild-growing vascular plants were recorded in the city on a total of 106 sample plots located in representative land-use types and within various distances of the city center. Frequency, origin, life forms, and plant families of non-native species were analyzed. Additionally, the percentage of non-native species along a gradient from the city center to the outskirts was investigated. We found that most common non-native species of the flora of Chonju are: (i) native to the Northern hemisphere; (ii) belong to the annuals; (iii) belong to the plant families of the Compositae or Leguminosae; and (iv) they perform best on sites that are continuously disturbed by human impact. In conclusion, non-native species play a significant role in enhancing biological diversity in urban areas.  相似文献

Studies were conducted to examine changes in soil (Ψs) and plant water status during summer in a 16-year old Quercus suber plantation in southern Portugal. Continuous measurements were conducted between May 2003 and August 2004, while discontinuous measurements were conducted on a monthly basis between May and September 2003 and repeated between March and September 2004. Intensive measurements were conducted on five trees with mean height and DBH of 5.3 m and 11.6 cm, respectively, growing at close proximity to each other. Weather conditions and soil water potential (Ψs) at the rhizosphere of each of the trees measured at 0.3 and 1 m soil depth were continuously monitored. Predawn (Ψpd) and midday (Ψmd) leaf water potentials were determined every month. Soil and plant samples were also collected in June and September from different locations within the study site for δ18O isotope composition analysis. Pressure–volume (pv) curves were constructed from plant shoots at different times during the vegetative period to determine osmotic potential at full saturation (Π100), water potential at turgor loss point (Ψtlp), relative water content at turgor loss point (R*tlp) and bulk modulus of elasticity (ε). Significant P < 0.05 decline in Ψs occurred between May and September, the lowest value recorded being –2.0 MPa. Decline in soil moisture affected tree water status, but decline in leaf water potential varied significantly (P < 0.05) among the trees. At the end of summer drought, lowest Ψpd measured was –1.7 MPa while the highest measured during this time was –0.8 MPa. Differences among trees were attributed to differences in rooting depth, as shown by regression analysis of 18O isotopes. Radial stem growth ceased when Ψs within the upper 0.3 m depth approached –1.5 MPa. The upper soil layers contributed approximately 33% of the total tree water requirement, between spring and mid summer when drought was experienced by trees. Deep soil layers however, supplied most of the water required during drought and no growth was recorded during this time. Stressed trees increased solute concentration of their tissues by a Magnitude of 0.7 MPa while bulk tissue elastic modulus increased by about 17 MPa. The study emphasizes the significance of roots as determinants of tree productivity and survival in the Mediterranean ecosystems.  相似文献

植物资源普查是掌握植物物种及数量变化的基础,对研究植物的地理分布和生物多样性具有重要意义。通过对2017年—2018年期间福建省本土植物调查过程中采集的植物标本进行整理和鉴定,发现了福建省被子植物5个新记录种,即台湾附地菜(Trigonotis formosana)、浙江琴柱草(Salvia nipponica subsp. zhejiangensis)、笔龙胆(Gentiana zollingeri)、大头茶(Polyspora axillaris)和钝颖落芒草(Piptatherum kuoi),均为福建省新记录种,大头茶属(Polyspora)和落芒草属(Piptatherum)为福建省新记录属。这些新记录植物的发现,丰富了福建省原生植物资源,为植物区系研究提供了新的资料。  相似文献

The point quadrat method can be used for determination of vegetational change in meadows of great species diversity. An appropriate sampling technique is described comprehending an apparatus of high rigidity and a shelter to keep off wind and rain. Sample size related to sampling time expense affects the number of species recorded and methodical error which is empirically determined by repetitive sampling. A setup of fixed points leads to higher accuracy than random sampling. When methodical error is quantitatively known, significant vegetational change can be detected by sampling at successive times.For fluctuation studies of plant populations in meadows the point quadrat technique should be preferred to visual estimates of plant cover because of its higher accuracy.Abbreviations f= frequency - s= standard deviation - Sr= variation coefficient - DWLS= distance weighted least squares  相似文献

枫香(Liquidambar formosana)因其叶片入秋后逐渐变红而极具观赏价值,是优良的景观生态树种。为了解枫香叶片结构变化与叶色的关系,该文通过连续监测枫香叶片变红过程中组织结构、光合特性及色素含量的变化,分析叶片结构与其光合特性和色素的关系。结果表明:(1)叶片变色过程中,表皮细胞均为椭圆形,紧密排列,未观察到明显的细胞变异,表面未附着绒毛和蜡质,且上表皮细胞与栅栏组织细胞间排列紧密,未出现较大的气室。(2)随着叶片逐渐变红,叶片结构变化显著,其中叶片、上表皮、栅栏组织和海绵组织厚度及气孔开度均逐渐减小,而气孔器长和宽、单个气孔器面积则逐渐增大。(3)随着叶片结构的变化,其叶绿素含量逐渐减少,致使净光合速率逐渐减小,在出现光破坏时,叶片通过在栅栏组织细胞液泡内合成花色苷来自我保护,而大量的花色苷致使叶片表面呈现红色。综上认为,叶绿素含量降低,花色素苷大量积累是导致枫香叶片变红的直接原因,而枫香叶色变红则是其一系列生理结构特征综合作用的结果。  相似文献

Transcutaneous PCO2 (PtcCO2), which is linearly related to arterial PCO2, was continuously recorded in healthy, adult, normal volunteers for 8-h periods. Recording this variable with the apparatus employed permits measurement of changes in the level of ventilation while subjects are freely ambulant and unencumbered by invasive and flow-resistive respiratory apparatus. The time series obtained exhibited marked periodicities. The frequencies and amplitudes varied between subjects. Peak-to-peak variation was 10-20% of mean values. There was no apparent association between fluctuations in PCO2 and activity other than formal exercise. Visual inspection of the time series and preliminary statistical analysis of digitally converted data suggested that the time-dependent changes of PtcCO2 were normally distributed. However, more rigorous statistical examination revealed that in no case was PtcCO2 actually normally distributed.  相似文献

A device, which automatically and continuously keeps moving plants in fixed positions, has been constructed. The apparatus is based on optical detection of any movements of the plant organ under study. The plant organ is kept in the desired position by means of wires, and the force necessary to apply to the wires to achieve this is recorded. The force reflects any tendency of the plant organ to move. The system is controlled by an Apple II computer. The device can work in one or two dimensions and record compensation forces in a wide range. In the experiments mentioned below, the magnitude of the forces necessary to keep the plants in a fixed position was of the order of 10-4N.
Circadian leaf movements of Oxalis regnellii Mig. were studied in the device. The leaf rhythm continued, although the leaflets were clamped and the light input on the leaf therefore constant. Circumnutation of hypocotyls of Helianthus annus L. cv. Californicus were drastically reduced in amplitude when the hypocotyls were kept in vertical position by the wires. Since the gravitropic input signal to this system was zeroed by the equipment, the results demonstrated that in the absence of gravitropic inputs the circumnutation reactions drastically diminish. This confirmed that circumnutations of these hypocotyls are influenced by gravity. Finally, the apparatus was used to study phototropic reactions: By clamping phototropically stimulated coleoptiles of Avena sativa L. cv. Seger no gravitational stimulations were involved and the phototropic reaction without interference from gravity could be studied.  相似文献

Abstract Phytophthora cinnamomi (cinnamon fungus) is a pathogenic soil fungus that infects plant communities along the southeastern coast of Australia and the southwestern corner of Western Australia. Infection of native plant communities with P. cinnamomi regularly leads to dramatic changes in both the structural and the floristic characteristics of these communities. This study aimed to assess the effect of P. cinnamomi induced changes in plant community attributes upon the abundance and diversity of invertebrates in an area of open forest in the Brisbane Ranges, Victoria. Pitfall trapping was conducted continuously for 1 week within each season over a 3 year period. Invertebrates were sorted to order level, and abundances were compared between times (season and year) and infection status for taxa with normalized distributions (Coleoptera, Collembola, Dermaptera and ants). Non-parametric comparisons were made for other groups (Aranae, Blattodea, Diptera, Hemiptera, Orthoptera, ant ‘morpho-species’ and unidentified larvae) to assess differences on the basis of infection status. Significant differences on the basis of infection were uncommon and, where identified, elevated abundances were more commonly observed at sites infected by P. cinnamomi. Consistent temporal effects (season and year) were observed in normalized data sets. Abundances both within individual taxa and from pooled counts were generally weakly associated with ground-level habitat features. Overall, the impact of P. cinnamomi on vegetative structure and floristics was not reflected in different abundances of ground-dwelling invertebrates.  相似文献

Brefeldin A (BFA) is a useful tool for studying protein trafficking and identifying organelles in the plant secretory and endocytic pathways. At low concentrations (5–10 μg ml?1), BFA caused both the Golgi apparatus and trans‐Golgi network (TGN), an early endosome (EE) equivalent in plant cells, to form visible aggregates in transgenic tobacco BY‐2 cells. Here we show that these BFA‐induced aggregates from the Golgi apparatus and TGN are morphologically and functionally distinct in plant cells. Confocal immunofluorescent and immunogold electron microscope (EM) studies demonstrated that BFA‐induced Golgi‐ and TGN‐derived aggregates are physically distinct from each other. In addition, the internalized endosomal marker FM4‐64 co‐localized with the TGN‐derived aggregates but not with the Golgi aggregates. In the presence of the endocytosis inhibitor tyrphostin A23, which acts in a dose‐ and time‐dependent manner, SCAMP1 (secretory carrier membrane protein 1) and FM4‐64 are mostly excluded from the SYP61‐positive BFA‐induced TGN aggregates, indicating that homotypic fusion of the TGN rather than de novo endocytic trafficking is important for the formation of TGN/EE‐derived BFA‐induced aggregates. As the TGN also serves as an EE, continuously receiving materials from the plasma membrane, our data support the notion that the secretory Golgi organelle is distinct from the endocytic TGN/EE in terms of its response to BFA treatment in plant cells. Thus, the Golgi and TGN are probably functionally distinct organelles in plants.  相似文献

Golgins are a family of coiled-coil proteins that are associated with the Golgi apparatus. They are necessary for tethering events in membrane fusion and may act as structural support for Golgi cisternae. Here we report on the identification of an Arabidopsis golgin which is a homologue of CASP, a known transmembrane mammalian and yeast golgin. Similar to its homologues, the plant CASP contains a long N-terminal coiled-coil region protruding into the cytosol and a C-terminal transmembrane domain with amino acid residues which are highly conserved across species. Through fluorescent protein tagging experiments, we show that plant CASP localizes at the plant Golgi apparatus and that the C-terminus of this protein is sufficient for its localization, as has been shown for its mammalian counterpart. In addition, we demonstrate that the plant CASP is able to localize at the mammalian Golgi apparatus. However, mutagenesis of a conserved tyrosine in the transmembrane domain revealed that it is necessary for ER export and Golgi localization of the Arabidopsis CASP in mammalian cells, but is not required for its correct localization in plant cells. These data suggest that mammalian and plant cells have different mechanisms for concentrating CASP in the Golgi apparatus.†These authors have contributed equally to the work  相似文献

Summary A study was made of the effect of soil moisture tension (pF 0–3), regarding the soil moisture status and soil oxygen status as interrelated, on the growth and cation uptake byRaphanus sativus using a tension plate apparatus. A similar study was made usingBrassica sinensis in pots.Results suggested that the tension-plate apparatus would be useful in such studies, whereas the lack of adequate soil moisture control in the pot experiment made this type of method unsuitable.Increasing soil moisture tension resulted in an increase in the cation concentrations of both soil solution and plant material. The dilution and valency effect in the soil solution was demonstrated.The growth response to soil moisture tension was a reflection of changes in moisture status which in turn affected the soil oxygen tension.The value of logK–1/2log [Ca+Mg] in the plant was constant over the soil moisture tension range studied and was a reflection of the characteristic potassium intensity status of the soil solution, defined by pK–1/2p (Ca+Mg) which was also constant. The concentration ratio, K/Ca+Mg, in the plant varied with moisture tension in the same way as the ratio varied in the soil solution and was therefore a reflection of both the soil solution composition and soil moisture tension.The plant reflected the composition of the soil solution and it was concluded that both the adsorption and contact-exchange theories played no significant role in plant ion uptake.  相似文献

Real-time monitoring of gene expression by a bioluminescence reporter gene is a powerful method for large-scale, detailed analysis of gene expression in living cells and large-scale screening of mutants. We have developed a portable, compact, integrated automatic bioluminescence-monitoring apparatus that can continuously monitor 960 individual plant seedlings or micro-organism colonies under uniform light conditions at temperatures up to 50 °C. The apparatus gave reproducible and reliable results for both bioluminescence photon counts and period length of bioluminescence rhythms of Arabidopsis reporter strain. Using the apparatus, we measured bioluminescence rhythms in the thermophilic cyanobacterium Thermosynechococcus at temperature up to 43 °C. We also monitored the expression of the flowering regulator gene CONSTANS in Arabidopsis as bioluminescence in high time resolution under different photoperiodic conditions. The high-throughput bioluminescence-monitoring apparatus developed here is a powerful tool for real-time monitoring of gene expression and gene function.  相似文献

Summary The present observations concern changes in the cortical structure of the ciliateDileptus prior to and during conjugation. The results can be summarized as follows: (1) Mating cells join each other in heteropolar configuration, i.e., the distal part of the oral apparatus of one partner faces the proximal part of the oral apparatus of another partner. (2) The structural changes prior to conjugation occur in the area of the oral apparatus. (3) The bonding area is situated within the oral apparatus, while the oral ciliature that encircles the oral apparatus remains outside the bonding area. (4) The fusion area is formed within the bonding area as a newly formed zone, without cortical organelles typically associated with the cell membrane. Two aspects of the formation of cell-to-cell union inDileptus are discussed: firstly, the heteropolar configuration of mating cells and its possible bearing upon an ability to form double cells when partners separation is inhibited. Secondly, the bonding area ofDileptus is compared to bonding areas of peniculine and hymenostome ciliates. A striking likeness in structural changes was found within the bonding areas of all three ciliates, in spite of the fact that the changes in question are localized within the oral apparatus ofDileptus, while inParamecium andTetrahymena the changed territory is situated between the oral apparatus and the anterior pole of the cell.  相似文献

ABSTRACT. The construction of a continuously recording respirometer is described. The oxygen consumption patterns of male tsetse flies recorded with this apparatus generally match their patterns of locomotor activity previously reported from actograph data and direct observation. Several experiments have highlighted the complexity of stress interactions involved in plant response. The impact in field conditions of combined environmental constraints on the mechanisms involved in plant photosynthetic response, however, remains understudied. In a long‐term field study performed in a managed grassland, we investigated the photosynthetic apparatus response of the perennial ryegrass (Lolium perenne L.) to environmental constraints and its ability to recover and acclimatize. Frequent field measurements of chlorophyll a fluorescence (ChlF) were made in order to determine the photosynthetic performance response of a population of L. perenne. Strong midday declines in the maximum quantum yield of primary photochemistry (FVFM) were observed in summer, when a combination of heat and high light intensity increased photosynthetic inhibition. During this period, increase in photosystem I (PSI) activity efficiency was also recorded, suggesting an increase in the photochemical pathway for de‐excitation in summer. Strong climatic events (e.g. heat waves) were shown to reduce electron transport between photosystem II (PSII) and PSI. This reduction might have preserved the PSI from photo‐oxidation. Periods of low soil moisture and high levels of sun irradiance increased PSII sensitivity to heat stress, suggesting increased susceptibility to combined environmental constraints. Despite the multiple inhibitions of photosynthetic functionality in summer, the L. perenne population showed increased PSII tolerance to environmental stresses in August. This might have been a response to earlier environmental constraints. It could also be linked to the selection and/or emergence of well‐adapted individuals.  相似文献

Net photosynthesis and dark respiration of a natural Chara tomentosa community were continuously recorded. There is a close correlation between net rate of photosynthesis and variations in the PhAR flux under water. The net photosynthesis fluence Px correlates with the PhAR fluence Ix, but the quotient Px/Ix varies seasonally, apparently as a consequence of variations in chlorophyll content following long-range changes in radiation fluence. No influence of temperature, salinity or pH on net photosynthesis was detected, but water temperature stimulates dark respiration and consequently diminishes net primary production.  相似文献

The effects of ventricular geometry, muscle mass, muscle elasticity and external pressures on the pressure-volume and muscle stiffness-stress relations have been quantitated on the basis of a theoretical model. Data taken from patients before and after interventions with nitroprusside and angiotensin were applied to the model in order to explain the possible causes for the marked shifts in the pressure-volume relations. The results indicate that (a) ventricular geometry does not markedly alter the pressure-volume and stiffness-stress relations unless there is a drastic change from a spherical shape to an ellipsoidal shape orvice versa, (b) increases in muscle mass and muscle elasticity of the order of 30% result in significant alterations in the P-V relations but are not the cause for the parallel shifts unless accompanied by substantial external pressures, (c) the parallel shifts in the pressure-volume relations can be accounted for entirely by the presence of external pressures without changes in muscle mass or muscle elasticity. Thus manipulation of right ventricular pressures or pericardial pressures by drug interventions may be useful in the treatment of left heart disease and the presence of such pressures must be considered in the analysis of ventricular function curves.  相似文献
