嗜热与嗜常温微生物的蛋白质氨基酸组成比较   总被引:11,自引:0,他引:11  
嗜热微生物的嗜热特性与其蛋白质的高度热稳定性紧密相关。为了探索嗜热蛋白质的热稳定机制,比较嗜热和嗜常温微生物的蛋白质在氨基酸组成上的差别,收集110对分别来自嗜热和嗜常温微生物的同源蛋白质序列,比较两组蛋白质各种氨基酸含量以及疏水性氨基酸组成、疏水性指数和荷电氨基酸组成的差别,结果两者在多种氨基酸含量上存在微小但统计学上显著的差别,嗜热蛋白质比嗜常温蛋白质具有较高的平均疏水性和荷电氨基酸组成。对两组蛋白质的“脂肪族氨基酸指数”进行分析,证明嗜热蛋白质之所以具有较高的脂肪族氨基酸指数是由于其亮氨酸含量较高,与影响该指数的其它几种氨基酸无关;从而认为该指数的意义值得怀疑。通过对大量同源嗜热蛋白质和嗜常温蛋白质氨基酸组成的比较,能够揭示一些有关蛋白质热稳定性的普遍规律。  相似文献

HVA and 5-HIAA in human cerebrospinal fluid were quantitatively determined by both fluorometry and mass fragmentography. The homovanillic acid values obtained by fluorometry were significantly lower than those obtained by mass fragmentography (P < 0.05). The correlation coefficient between values for HVA obtained by the two methods was high, r= 0.90. For 5-HIAA the concentrations obtained by the two methods were not significantly different while the correlation coefficient was lower, r= 0.55.  相似文献

Abstract— The abilities of AOAA and EOS to modify the utilisation of radioactively labelled glucose, acetate, glutamine and GABA were studied in isolated rat retina. AOAA inhibited the activities of GAD and GABA-T, while EOS inhibited GABA-T but not GAD. AOAA lowered the free amino acid contents of incubated retinae and suppressed the outflow of amino acids into the incubation medium, while EOS had no effect on either parameter. AOAA strongly inhibited the incorporation of 14C from labelled glucose, acetate and glutamine into GABA, and also suppressed the labelling of glutamate, aspartate and glutamine. These effects were qualitatively similar but quantitatively smaller with EOS. Both compounds markedly decreased the syntheses of aspartate and glutamate from exogenous GABA, while the passage of carbon from GABA to glutamine was much less affected. It is suggested that AOAA and EOS may act predominantly on neurones. It appears that inhibition of GABA-T alone does not cause a profound disturbance of the metabolism of other amino acids. Other metabolic inhibitors such as ouabain, malonate and fluoroacetate did not greatly affect the metabolism of GABA in rat retina.  相似文献

When dry decotyledonized embryos of Raphanus are supplied withwater, a brief period of water absorption (phase A) is followedby a period of no fresh weight increase (phase B) which lastsfor 8 hr at 30°. In this period, embryos become ready toadvance into the period of fresh weight increase (phase C). When embryos were exposed to various concentrations of thiouracilor actinomycin D solution from 0 hr of water supply, increasesin fresh weight and in RNA content measured at 13 hr were inhibitedin parallel with each other. Chloramphenicol and puromycin inhibitedthe fresh weight increase without affecting the RNA increase.When embryos were exposed to thiouracil or puromycin for 2,4 and 6 hr, beginning at 0 hr of water supply, the start ofphase C delayed 2, 4 and 6 hr, respectively. When these drugswere given after phase B had progressed at least for 2 hr, thedelay of the start of phase C was shorter than the period ofthe drug treatment. If given at the end of phase B, thiouraciland actinomycin D inhibited the incorporation of 14C-uracilbut not the fresh weight increase, while chloramphenicol andpuromycin inhibited the latter without inhibiting the former. During phase B, protein content per dry weight of embryo didnot increase, but the rate of 14C-leucine incorporation increasedremarkably to reach the level in phase C. Incorporation of labeledleucine was inhibited if embryos were subjected to thiouracilor actinomycin D action during phase B, but not if the drugswere given when phase B had been completed. Puromycin and chloramphenicolinhibited the incorporation whenever they were given. The increase in respiratory activity during phase B was inhibitedrelatively little by the above mentioned four drugs. In conclusion syntheses of RNA and protein seem to be essentialfor the progress of phases B and C, protein synthesis havinga more direct effect. (Received September 17, 1965; )  相似文献

In acidifying lakes, pH decreases abruptly in response to acid precipitation events. We tested the hypothesis that, in comparison to a circumneutral lake, phytoplankton photosynthesis in an acidifying lake is less sensitive to a rapid decrease in pH (acid pH shock). Phytoplankton in Plastic Lake, which is undergoing acidification, was characterized by a predominance of Pyrrophyta, and phytoplankton photosynthesis decreased to a lesser extent in response to an acid pH shock than the photosynthesis of populations from St. Nora Lake, a circumneutral lake located nearby, in which Pyrrophyta were not abundant. Rates of phytoplankton photosynthesis in acid pH shock experiments were significantly correlated with hydrogen ion but not with dissolved inorganic carbon (DIC) concentrations. Depression of photosynthesis following an acid pH shock occurred in axenic cultures of Chlorella pyrenoidosa Chick but was not observed in axenic cultures of the acidophilic alga Chlorella saccharophila (Krug.) Nadson or in three species isolated from Plastic Lake. However, the three isolates were not acidophilic during growth. We conclude that phytoplankton in acidifying lakes consists predominantly of species which are tolerant to acid pH for short periods (hours) but cannot grow at these pHs.  相似文献

RESPIRATION AND ORGANIC ACID METABOLISM IN POTASSIUM DEFICIENT RICE PLANT   总被引:1,自引:0,他引:1  
Rice seedlings were grown in nutrient solutions containing 5,10, and 100 ppm of potassium (as K2O) for about one month. Respiratoryrate and mineral, organic acid and amino acid contents of theleaves of those seedlings were determined. At an earlier stageof growth, potassium deficiency induced a marked increase inrespiration. The amounts of cellular constituents were variabledependent on extent of potassium deficiency as well as age ofthe plant. The relationship between the respiration and organicand amino acid contents was discussed in terms of metabolicactivities for these compounds. Acetic and succinic acid contentsin the leaves of the lowest supply of potassium (K5 leaves)were smaller than those of the other leaves (K10 and K100 leaves)at the earlier step of growth. The highest rate of respirationwas observed in K5 leaves. The distinctive changes were observedin glutamine and asparagine contents by potassium treatment. (Received April 21, 1966; )  相似文献

The aim of this study was to test whether abiotic and biotic factors may affect allelopathic properties. Therefore, we investigated how solar radiation and bacteria influence allelopathic effects of the plant‐derived, polyphenolic tannic acid (TA) on microalgae. Using a block design, lake water samples with and without TA were exposed to solar radiation or kept in darkness with or without bacteria for 3 weeks. Dissolved organic carbon (DOC), specific size fractions of DOC analyzed by chromatography–organic carbon detection (LC‐OCD), and concentrations of total phenolic compounds (TPC) were measured to follow the fate of TA in lake water with natural DOC exposed to photolytic and microbial degradation. DOC and TPC decreased in dark‐incubated lake water with TA and bacteria indicating microbial degradation. In contrast, exposure to solar radiation of lake water with TA and bacteria did not decrease DOC. Chromatographic analyses documented an accumulation of DOC mean size fraction designated as humic substances (HS) in sunlit water samples with TA. The recalcitrance of the humic fraction indicates that photolytic degradation may contribute to a DOC less available for bacterial degradation. Subsequent growth tests with Desmodesmus armatus (Chodat) E. Hegewald showed low but reproducible difference in algal growth with lower algal growth rate cultured in photolytically and microbially degraded TA in lake water than cultured in respective dark treatments. This finding highlights the importance of photolytic processes and microbial degradation influencing allelopathic effects and may explain the high potential of allelochemicals for structuring the phytoplankton community composition in naturally illuminated surface waters.  相似文献
