A simple T-shaped plasmonic nanostructure composed of two perpendicular coupled nanorods is proposed to produce strong Fano resonances. By the near-field coupling between the “bright” dipole and “dark” quadrupole plasmons of the nanorods, a deep Fano dip is formed in the extinction spectrum, which can be well fitted by the Fano interference model. The effects of the geometry parameters including nanorod length, coupling gap size, and coupling location to the Fano resonances are analyzed in detail, and a very high refractive index sensitivity is achieved by the Fano resonance. Also by adjusting the incident polarization direction, double Fano resonances can be formed by the interferences of the dipole, quadrupole, and hexapole plasmons. The proposed nanorod dimer structure is agile, and a trimer which supports double Fano resonances can be easily formed by introducing a third perpendicular coupled nanorod. The proposed T-shaped nanorod dimer structure may have applications in the fields of biological sensing and plasmon-induced transparency.  相似文献

对不同葡萄糖浓度下光滑球拟酵母分批发酵生产丙酮酸的动力学模型分析发现, 葡萄糖浓度是影响光滑球拟酵母发酵生产丙酮酸过程功能的关键因素。在发酵初始阶段, 低浓度葡萄糖可维持较高的菌体比生长速率; 对数生长中前期, 葡萄糖快速进料使菌体浓度接近最大值, 并实现碳流从菌体生长转向丙酮酸积累; 对数生长后期葡萄糖浓度控制在33.4 g/L以维持高丙酮酸对葡萄糖产率系数 (0.71 g/g)。采用奇异控制的葡萄糖流加方式, 在7 L发酵罐上控制不同发酵阶段葡萄糖浓度处于最佳水平以强化光滑球拟酵母过程功能, 丙酮酸产量 (83.1 g/L)、产率 (0.621 g/g)、生产强度[1.00 g/(L·h)]与分批发酵对比, 分别提高了21.3%、21.6%和29.9%。  相似文献

In this paper, we probed surface-enhanced Raman scattering (SERS) and surface-enhanced fluorescence (SEF) from probe molecule Rhodamine 6G (R6G) on self-standing Au nanorod array substrates made using a combination of anodization and potentiostatic electrodeposition. The initial substrates were embedded within a porous alumina template (AAO). By controlling the thickness of the AAO matrix, SEF and SERS were observed exhibiting an inverse relationship. SERS and SEF showed a non-linear response to the removal of AAO matrix due to an inhomogeneous plasmon activity across the nanorod which was supported by FDTD calculations. We showed that by optimizing the level of AAO thickness, we could obtain either maximized SERS, SEF or simultaneously observe both SERS and SEF together.  相似文献

We present the design, synthesis and characterization of new functionalized fluorescent optical switches for rapid, all-visible light-mediated manipulation of fluorescence signals from labelled structures within living cells, and as probes for high-contrast optical lock-in detection (OLID) imaging microscopy. A triazole-substituted BIPS (TzBIPS) is identified from a rational synthetic design strategy that undergoes robust, rapid and reversible, visible light-driven transitions between a colorless spiro- (SP) and a far-red absorbing merocyanine (MC) state within living cells. The excited MC-state of TzBIPS may also decay to the MC-ground state emitting near infra-red fluorescence, which is used as a sensitive and quantitative read-out of the state of the optical switch in living cells. The SP to MC transition for a membrane-targeted TzBIPS probe (C12-TzBIPS) is triggered at 405 nm at an energy level compatible with studies in living cells, while the action spectrum of the reverse transition (MC to SP) has a maximum at 650 nm. The SP to MC transition is complete within the 790 ns pixel dwell time of the confocal microscope, while a single cycle of optical switching between the SP and MC states in a region of interest is complete within 8 ms (125 Hz) within living cells, the fastest rate attained for any optical switch probe in a biological sample. This property can be exploited for real-time correction of background signals in living cells. A reactive form of TzBIPS is linked to secondary antibodies and used, in conjunction with an enhanced scope-based analysis of the modulated MC-fluorescence in immuno-stained cells, for high-contrast immunofluorescence microscopic analysis of the actin cytoskeleton.  相似文献

The Ras oncogene transmits signals, which regulate various cellular processes including cell motility, differentiation, growth and death. Since Ras signalling is abnormally activated in more than 30% of human cancers, Ras and its downstream signalling pathways are considered good targets for therapeutic interference. Ras is post‐translationally modified by the addition of a farnesyl group, which permits its attachment to the plasma membrane. Exploiting this knowledge, a synthetic Ras inhibitor, S‐trans, trans‐farnesylthiosalicylic acid (FTS; Salirasib), was developed. FTS resembles the farnesylcysteine group of Ras, and acts as an effective Ras antagonist. In the present review, the effect of FTS in combination with various other drugs, as tested in vitro and in vivo, and its therapeutic potential are discussed. As reviewed, FTS cooperates with diverse therapeutic agents, which significantly improves treatment outcome. Therefore, combinations of FTS with other agents have a potential to serve as anti‐cancer or anti‐inflammatory therapies.  相似文献

Multi-fluorescence recovery after photobleaching is a microscopy technique used to measure the diffusion coefficient (or analogous transport parameters) of macromolecules, and can be applied to both in vitro and in vivo biological systems. Multi-fluorescence recovery after photobleaching is performed by photobleaching a region of interest within a fluorescent sample using an intense laser flash, then attenuating the beam and monitoring the fluorescence as still-fluorescent molecules from outside the region of interest diffuse in to replace the photobleached molecules. We will begin our demonstration by aligning the laser beam through the Pockels Cell (laser modulator) and along the optical path through the laser scan box and objective lens to the sample. For simplicity, we will use a sample of aqueous fluorescent dye. We will then determine the proper experimental parameters for our sample including, monitor and bleaching powers, bleach duration, bin widths (for photon counting), and fluorescence recovery time. Next, we will describe the procedure for taking recovery curves, a process that can be largely automated via LabVIEW (National Instruments, Austin, TX) for enhanced throughput. Finally, the diffusion coefficient is determined by fitting the recovery data to the appropriate mathematical model using a least-squares fitting algorithm, readily programmable using software such as MATLAB (The Mathworks, Natick, MA).Download video file.(77M, mp4)  相似文献

A neural network processing scheme is proposed which utilizes a self-organizing Kohonen feature map as the front end to a feedforward classifier network. The results of a series of benchmarking studies based upon artificial statistical pattern recognition tasks indicate that the proposed architecture performs significantly better than conventional feedforward classifier networks when the decision regions are disjoint. This is attributed to the fact that the self-organization process allows internal units in the succeeding classifier network to be sensitive to a specific set of features in the input space at the outset of training.  相似文献

Abstract: Obtaining reliable results from life-cycle assessment studies is often quite difficult because life-cycle inventory (LCI) data are usually erroneous, incomplete, and even physically meaningless. The real data must satisfy the laws of thermodynamics, so the quality of LCI data may be enhanced by adjusting them to satisfy these laws. This is not a new idea, but a formal thermodynamically sound and statistically rigorous approach for accomplishing this task is not yet available. This article proposes such an approach based on methods for data rectification developed in process systems engineering. This approach exploits redundancy in the available data and models and solves a constrained optimization problem to remove random errors and estimate some missing values. The quality of the results and presence of gross errors are determined by statistical tests on the constraints and measurements. The accuracy of the rectified data is strongly dependent on the accuracy and completeness of the available models, which should capture information such as the life-cycle network, stream compositions, and reactions. Such models are often not provided in LCI databases, so the proposed approach tackles many new challenges that are not encountered in process data rectification. An iterative approach is developed that relies on increasingly detailed information about the life-cycle processes from the user. A comprehensive application of the method to the chlor-alkali inventory being compiled by the National Renewable Energy Laboratory demonstrates the benefits and challenges of this approach.  相似文献

Abstract

The fluorescent 2-aminopurine probe (2-AP) incorporated into the loop of 23-mer RNA hairpin of HIV-1 genome dimerization initiation site (DIS) was used for discrimination of specific and unspecific binding of paromomycin and spermine to the kissing loop dimer (KD) formed in solution. While both ligands stabilized the KD RNA structure, only paromomycin binding resulted in significant increase of 2-AP fluorescence. These observations suggest that the 2-AP fluorescent RNA construct might be useful for selecting ligands specifically binding the HIV-1 kissing loop RNA dimer.  相似文献

Coatline A ( 1 ) and α‐epi‐coatline A ( 4 ) co‐occur in the trunk extract of Andira coriacea. Inspection of their chiroptical properties led to intriguing results. After a careful examination of the experimental data used for the previously reported absolute configuration of these compounds, some uncertainties were identified. A combined theoretical approach including conformational analyses and calculation of electronic circular dichroism (ECD) spectra, in addition with experimental data obtained for schoepfin A ( 5 ) and the new schoepfin D ( 6 ) isolated from Senna quinquangulata, allowed the revision of the absolute configuration of coatlines A ( 1 ) and B ( 2 ).
