Abstract Reciprocal crosses were performed with Glossina swynnertoni and Glossina morsitans morsitans and with G. swynnertoni and Glossina morsitans centralis , using strains that carried marker genes in all three linkage groups. Glossina swynnertoni males can inseminate, but not fertilize, G.m.morsitans; all other crosses produced some fertile females. Hybridization did not cause sex ratio distortion among F{ flies. Most F and backcross females were fertile, but all F, males were sterile. Sterility among backcross males was also high (99% in BXj, 85% in Bx2, and about 50% in Bx3 to Bx5). Chromosome transmission by hybrid females usually conformed to Mendelian expectations, but genetic recombination was lower than observed in G.m.morsitans. The reduction in fertility among backcross females was not associated with heterozygosity in any linkage group. Sterility among hybrid and backcross males was associated with heterozygosity of sex chromosomes and probably autosomes. The results support the systematic placement of G.swynnertoni closer to G.mxentralis than to G.m.morsitans.  相似文献

SYNOPSIS. Culture procyclic forms of Trypanosoma brucei rhodesiense and Trypanosoma congolense were fed to Glossina morsitans morsitans through artificial membranes. A very high percentage of the flies so fed produced established midgut infections, a proportion of which went on to develop into mature metacyclic trypanosomes capable of infecting mammalian hosts. The method offers a safe, clean way of infecting tsetse flies with African trypanosomes which reduces the need for trypanosome-infected animals in the laboratory.  相似文献

ABSTRACT. Post-feed buzzing in Glossina morsitans morsitans Westw. causes a rise in thoracic temperature relative to the length of the buzz. As lift is proportional to the square of wing-beat frequency, which increases with temperature up to 32°C, buzzing results in an increase in the lift which the fly can produce. Heat generated by buzzing, in combination with the heat received from the host at the time of feeding, may well allow the fly to maximize lift generated in the immediate post-feeding period. Buzzing flies excrete excess water from the meal more rapidly than non-buzzing flies. It is argued that this is due to a rise in abdominal temperature. Maximized lift in the immediate post-feeding period and the rapid elimination of water from the very large blood meals taken by these flies may be expected to have strong selective advantages for the flies.  相似文献

wo laboratory strains of Glossina morsitans centralis originating from different fly-belts (one from Singida, in Tanzania, and the other from Mumbwa, in Zambia) were compared with respect to vectorial competence for pathogenic Trypanosoma species, genetic variation and inter-colony fertility. The vectorial competence of G.m.centralis of Tanzanian origin for Trypanosoma vivax and T.congolense is similar to, whereas for T.brucei brucei it is lower than the colony of Zambian origin. Nevertheless, these two laboratory strains of G.m.centralis showed levels of susceptibility to the three pathogenic Trypanosoma species which were much greater than previously observed in laboratory colonies of other Glossina species. Electrophoresis of fifteen enzymes revealed that the two colonies differ significantly in allele frequencies at only three loci that are relatively close together on one of the autosomes. Hybridization experiments revealed that G.m.centralis from the two fly-belts are consubspecific.  相似文献

Abstract .In a single generation of selection, two lines of Glossina morsitans centralis were established that differed significantly in susceptibility to Trypanosoma congolense clone IL 1180. Reciprocal crosses demonstrated that susceptibility was a maternally inherited trait. Differences between the lines, to all phases of the trypanosome infection, were maintained for eight generations, whereas differences in susceptibility to midgut infections were maintained for twenty-eight generations. Thereafter, the lines did not differ in susceptibility to Trypanosoma congolense IL 1180. Susceptibility to infections with Trypanosoma congolense IL 1180 was only a weak predictor of susceptibility to T. congolense clones IL 13-E3 and K60/1, as well as clone T. brucei brucei STIB 247-L. However, the susceptible and refractory lines displayed these phenotypes when tested with Trypanosoma vivax, indicating that the factors that affect susceptibility to trypanosomes are expressed both within and outside the midgut.  相似文献

ABSTRACT. The effects of age and hunger on the responses of male Glossina morsitans morsitans Westwood and G.pallidipes Austen to freeze-killed female decoys, were examined in the laboratory. In both species, activity, estimated as the total number of interactions between males and decoys, increased with both age and hunger. Interactions were divided into short-stay (<60 s) and long-stay, full copulatory responses. In both species, young, unfed males were significantly less likely to attempt to copulate with a decoy after encounter than were fed males. Among fed males the proportion of interactions that proceeded to full copulatory attempts did not change with increasing age, but decreased consistently with increasing hunger. At all ages and hunger levels, G.pallidipes were more active than G.m.morsitans. However, after encountering a decoy, G.pallidipes were less likely to attempt to copulate than G.m.morsitans. In both species the duration of copulatory attempts did not change with age, but declined with increasing hunger. Copulatory attempts by G.pallidipes were significantly shorter than those of G.m.morsitans. The results are discussed in relation to the behaviour of tsetse in response to control devices such as traps and targets.  相似文献

ABSTRACT. The uterine gland of the tsetse fly Glossina morsitans morsitans Westw. synthesizes a secretion which nourishes the developing larva in the uterus. Aqueous extracts of the brain have been shown to stimulate the synthesis of the protein and amino acid components of this secretion from L- [U-14C]leucine by uterine gland tubules in vivo and in vitro. A linear dose response relationship was demonstrated in vitro with extract concentrations ranging from 1 × 10-4 to 1 × 10-2 brains μl-1. The maximum response, a > 300% increase in the rate of protein and amino acid synthesis, was achieved with as little as 1 × 10-2 brains μl-1 The concentration of active factor(s) in the brain declined during a single interlarval period coincident with the period of release of secretion associated with larval growth. The stimulatory activity in brain extracts was destroyed by proteolytic enzymes indicating that it is probably a protein or peptide. Results suggest that the active factor(s) is a hormone responsible for the stimulation of uterine gland protein synthesis essential for larval nutrition.  相似文献

The roles of visual and/or olfactory stimuli in eliciting mating responses from male Glossina morsitans morsitans Westwood were examined, using a system for automatically recording the number and duration of mating strikes made towards decoys, under controlled conditions. The results confirm that there is no olfactory component of the female sex recognition pheromone sensed by the male antennae, and the attraction of males to females appears to be visual. The absence of male-male mating strikes was the result of the absence of female sex-pheromone, rather than the presence of a repellent mating deterrent in the male cuticle. Experiments with coloured, artificial, sex-pheromone-dosed, cotton decoys showed that colour had only weak effects on attractiveness and number of encounters with decoys, and that no colour caused significant enhancement of mating responses over those shown to decoy females.  相似文献

To study the population structure of Glossina morsitans morsitans Westwood (Diptera: Glossinidae), polymerase chain reaction (PCR) and singlestrand conformational polymorphism (SSCP) methods were used to estimate mitochondrial DNA diversity at four loci in six natural populations from Zambia, Zimbabwe and Mozambique, and in two laboratory cultures. The Zambian and Zimbabwean samples were from a single fly belt. Four alleles were recorded at 12S and 16S1, and five alleles at 16S2 and COI. Nucleotide sequencing confirmed their singularities. Chi-square contingency tests showed that allele frequencies differed significantly among populations. Mean allele diversities in populations averaged over loci varied from 0.14 to 0.61. Little loss in haplotype diversity was detected in the laboratory cultures thereby indicating little inbreeding. Wright's fixation index F(ST) in the natural populations was 0.088+/-0.016, the correlation of haplotypes within populations relative to correlations in the total. A function of its inverse allows an estimate of the mean equivalent number of females exchanged per population per generation, 5.2. No correlation was detected between pairwise genetic distance measures and geographical distances. Drift explains the high degree of differentiation.  相似文献

Tsetse flies occupy discontinuous habitats and gene flow among them needs to be investigated in anticipation of area-wide control programs. Genetic diversities were estimated at six microsatellite loci in seven Glossina morsitans submorsitans Newstead (Diptera: Glossinidae) populations and five microsatellite loci in six G. m. morsitans Westwood populations. Nei's unbiased diversities were 0.808 and 76 alleles in G. m. submorsitans and 0.727 and 55 alleles in G. m. morsitans. Diversities were less in three laboratory cultures. Matings were random within populations. Populations were highly differentiated genetically. Populations were strongly subdivided, as indicated by fixation indices (F(ST)) of 0.18 in G. m. morsitans and 0.17 in G. m. submorsitans. 35% of the genetic variance in G. m. submorsitans was attributed to differences between populations from The Gambia and Ethiopia. All available genetic evidence suggests that genetic drift is much greater than gene flow among G. morsitans s.l. populations.  相似文献

ABSTRACT. The sexual behaviour of male Glossina morsitans morsitans Westwood (Diptera: Glossinidae) towards decoys dosed with sex phero-mone (15,19,23-trimethylheptatriacontane) is described quantitatively from field observations. The flies responded with a standard behavioural pattern after landing. This involved: first, the perception of pheromone; second, orientation on the decoy and genitalia engagement; and third, a lengthy quiescence corresponding to natural copulation. The probability of a fly leaving a decoy decreased during the sequence and was least in the final 'copulatory' phase, when it remained constant under constant conditions. The initial, rapid rates at which flies left decoys were affected mostly by pheromone dose: the lower the dose, the higher the rate. The final, slow rate of leaving was unaffected by pheromone dose, being most affected by environmental stimuli, especially interference from other flies: at high fly densities, final rates of leaving were high. The results are discussed in the context of improving the potential for using pheromone and chemo-sterilant-dosed decoys for tsetse autosterilization.  相似文献

ABSTRACT. The responses of male Glossina morsitans morsitans West-wood and Glossina pallidipes Austen to freeze-killed females were examined in the laboratory. Analyses were performed using a specially designed, automated, computer-based, recording system. G. pallidipes were more active than G. m. morsitans , interacting with the female decoys twice as often. Interactions with the decoys divided broadly into short-stay (<60 s) and long-stay, full copulatory attempts. For G. m. morsitans 90% of interactions resulted in full copulatory attempts, the mean duration of which was >1 h. For G. pallidipes only 40% of interactions resulted in full copulatory attempts, the mean duration of which was 35 min. The initiation of interactions showed a clear V-shaped activity pattern in G. m. morsitans but in G. pallidipes only a morning peak was observed. In neither species was there a tendency for full copulatory responses to be initiated in any specific period of the diurnal activity pattern. The results indicate that the two species have very different mating systems, and represent an initial step in the quantification of these differences.  相似文献

ABSTRACT. Experiments were performed on male Glossina morsitans morsitans Westw. and female Musca domestica L. to investigate their phototactic responses at long wavelengths. In both species, green light was less attractive than red (relative to a 400 nm standard). In M. domestica this can be adequately explained by an intensity-dependent phototactic reversal at wavelengths above 500 nm; red light could be matched in attractiveness by a low intensity green light, implying that no true red-green discriminating ability exists, but that red is perceived as a dim green. Although a partial phototactic reversal was also observed in G. m. morsitans at 525–575 nm, no tested intensity of green matched red in attractiveness. It appears therefore that G. m. morsitans may be an unusual dipteran in possessing a method of discriminating red and green light independently of intensity. ABSTRACT. Glossina morsitans morsitans Westw. and G. pallidipes Aust. flying around and landing on coloured screens and traps were sampled using electrocuting nets. External colour affected both attractiveness and efficiency of traps, such that yellow and green traps were unattractive and inefficient; black and red, attractive and inefficient; white, moderately attractive, and very efficient; and blue traps, attractive and efficient. The order of attractiveness of coloured screens was similar to that for traps. Landing responses were generally strongest on black surfaces, and weakest on white, but the results for blue were variable. Carbon dioxide and acetone odours improved trap catches and also eliminated most catch differences between traps differently coloured on their outer surfaces. The relative performances of traps coloured differently on inside surfaces only were not affected significantly by these odours, and in all cases black or red target areas inside the trap were required for optimum trap performance. When acetone and 1-octen-3-ol odours were used, catches were improved but the relative performance of differently coloured traps and screens was not changed. There were no obvious species differences in colour responses although numbers of G. morsitans were too low for statistical comparisons.  相似文献

A study was carried out to determine the effect on the reproductive performance of female Glossina morsitans morsitans Westwood when allowed to feed, in vitro, for 63 days on fresh defibrinated blood of buffalo, bushbuck, cattle, eland, oryx, warthog, waterbuck or wildebeest. There were marginal differences in the survival and reproductive performance between eight different groups of tsetse, 200 per group, when fed on the blood of these mammalian species. When allowed to feed for 14 consecutive days on the blood of buffalo, wildebeest or warthog, the mean number of feeds were 6.2 +/- 0.3, 6.5 +/- 0.3 and 6.3 +/- 0.3, respectively. The mean weight of the bloodmeal taken also did not differ significantly between these three groups. Whereas the protein patterns of the blood plasma of the above eight host animals were different, the protein patterns of the haemolymph from tsetse fed on the blood of these hosts were identical. It is thus concluded that the preference shown by tsetse for some mammalian species investigated here may not be based on any aspect of the nutritional value of their blood.  相似文献

Abstract. . Midgut infection rates of Trypanosoma congolense in Glossina palpalis palpalis and of Trypanosoma brucei rhodesiense in Glossina pallidipes are potentiated by the addition of D+ glucosamine to the infective feed, but not to the levels of super-infection reported for G. m. morsitans. G. p. palpalis and G.pallidipes are shown to possess two trypanocidal molecules: a glucosyl lectin which can be inhibited by D+ glucosamine and a galactosyl molecule inhibited by D+ galactose. Addition of both D+ glucosamine and D+ galactose to the teneral infective feed promotes super-infection of the midguts of G.p.palpalis. The glucosyl lectin is specific for rabbit erythrocytes and is present in guts of fed G.m.morsitans and G.p.palpalis , titres of lectin activity do not increase substantially after the second bloodmeal. The galactosyl specific molecule does not show any erythrocyte specificity, although haemolytic activity is observed only in G.p.palpalis and not in G.m.morsitans. The presence of two trypanocidal molecules in some species of tsetse may account for the innate refractoriness of these flies to trypanosome infection.
As D+ glucosamine also inhibits the killing of procyclic trypanosomes taken as an infective feed, it is suggested that the midgut lectin is normally responsible for the agglutination of trypanosomes in the fly midgut by binding to the pro-cyclic surface coat, prior to establishment in the ecto-peritrophic space.  相似文献

ABSTRACT. Actograph measurements of the spontaneous flight activity of isolated female Glossina morsitans morsitans Westw. were made through their first two pregnancy cycles in constant conditions under LD 12:12. Activity fell from a mean of c. 20 units/day to near zero during the 24 h preceding the day on which larvipositionoccurred. Activity on the day of larviposition rose significantly, to c. 35 units/day, then fell back to the baseline 20 units on subsequent days. The circadian pattern of activity was changed markedly by larviposition: 48 h before it, activity in the morning peak of the usual V-shaped diel pattern was twice normal; over 70% of the flies then fed, and the evening arm of their V disappeared; 24 h before larviposition, most flies were totally inactive, the few that did fly performing only 1 or 2 units/day, and only in the first and last hours of the photophase; on the day of larviposition, the morning peak of the V was reduced by c. 70%, and activity peaked instead during the post-noon 2–3 h, before and during the act of parturition. Almost all larvipositions occurred between noon and lights-out. It is suggested that the c. 48 h of pre-larviposition quiescence may play a role in the fact that pregnant G. morsitans females often survive non-residual aerosol insecticide campaigns.  相似文献

Abstract. Action potentials from olfactory cells in antennae (funiculi) of living tsetse flies, Glossina morsitans morsitans Westwood, were recorded using a surface-contact technique. Stimuli were the vapours of the seven alkylphenols identified in cattle urine: phenol, 3-methyl-, 3-ethyl-, 3- n -propyl-, 4-methyl-, 4-ethyl-, and 4- n -propylphenol. In addition, responses to the vapours of 1-octen-3-ol, acetone and dichloromethane were recorded. The phenol-sensitive cells could be grouped into four subclasses. Subclass, 1, 2 and 3 cells responded to the phenols only, cells of subclass 1 and 2 being activated by these substances, those of subclass 3 inhibited. Cells of subclass 4 were activated to a similar degree by all phenols and by one or more of the other chemicals tested. Subclass 1 cells were strongly activated by the 3-alkylphenols, whereas subclass 2 cells were most sensitive to 4-methylphenol. Subclass 3 cells were most strongly inhibited by phenol, and 3- and 4-methylphenol. The results suggest that though individual phenols may be attractive to G. m. morsitans , preference for certain blends of phenols may exist; for example, blends composed of moderate doses of 4-methylphenol and 3-methyl-, 3-ethyl- or 3- n -propylphenol.  相似文献
