Throughfall nitrogen of a 15-year-old Picea abies (L.) Karst. (Norway spruce) stand in the Fichtelgebirge, Germany, was labeled with either 15N-ammonium or 15N-nitrate and uptake of these two tracers was followed during two successive growing seasons (1991 and 1992). 15N-labeling (62 mg 15N m-2 under conditions of 1.5 g N m-2 atmospheric nitrogen deposition) did not increase N concentrations in plant tissues. The 15N recovery within the entire stand (including soils) was 94%±6% of the applied 15N-ammonium tracer and 100%±6% of the applied 15N-nitrate tracer during the 1st year of investigation. This decreased to 80%±24% and 83%±20%, respectively, during the 2nd year. After 11 days, the 15N tracer was detectable in 1-year-old spruce needles and leaves of understory species. After 1 month, tracer was detectable in needle litter fall. At the end of the first growing season, more than 50% of the 15N taken up by spruce was assimilated in needles, and more than 20% in twigs. The relative distribution of recovered tracer of both 15N-ammonium and 15N-nitrate was similar within the different foliage age classes (recent to 11-year-old) and other compartments of the trees. 15N enrichment generally decreased with increasing tissue age. Roots accounted for up to 20% of the recovered 15N in spruce; no enrichment could be detected in stem wood. Although 15N-ammonium and 15N-nitrate were applied in the same molar quantities (15NH 4 + : 15NO 3 - =1:1), the tracers were diluted differently in the inorganic soil N pools (15NH 4 + /NH 4 + : 15NO 3 - /NO 3 - =1:9). Therefore the measured 15N amounts retained by the vegetation do not represent the actual fluxes of ammonium and nitrate in the soil solution. Use of the molar ammonium-to-nitrate ratio of 9:1 in the soil water extract to estimate 15N uptake from inorganic N pools resulted in a 2–4 times higher ammonium than nitrate uptake by P. abies.  相似文献

The efficiency of cell-free protein synthesis combined with combinatorial selective 15N-labelling provides a method for the rapid assignment of 15N-HSQC cross-peaks to the 19 different non-proline amino-acid types from five 15N-HSQC spectra. This strategy was explored with two different constructs of the C-terminal domain V of the τ subunit of the Escherichia coli DNA polymerase III holoenzyme, τC16 and τC14. Since each of the five 15N-HSQC spectra contained only about one third of the cross-peaks present in uniformly labelled samples, spectral overlap was much reduced. All 15N-HSQC cross-peaks of the backbone amides could be assigned to the correct amino-acid type. Availability of the residue-type information greatly assisted the evaluation of the changes in chemical shifts observed for corresponding residues in τC16 vs. those in τC14, and the analysis of the structure and mobility of the C-terminal residues present in τC16 but not in τC14.  相似文献

The influence of one DNA region on the stability of an adjoining region (telestability) was examined. Melting curves of three block DNA's, d(C15A15)·d(T15G15), d(C20A15)·d(T15G20), and d(C20A10)·d(T10G20) were analyzed in terms of the nearest neighbor Ising model. Comparisons of predicted and experimental curves were made in 0.01 M and 0.1 M sodium ion solutions. The nearest neighbor formalism was also employed to analyze block DNA transition in the presence of actinomycin, a G·C specific molecule. The results show that nearest neighbor base-pair interaction cannot predict the melting curves of the block DNA's. Adjustments in theoretical parameters to account for phosphate repulsion assuming a B conformation throughout the DNA's do not alter this conclusion. Changes in the theoretical parameters, which provide good overall agreement, are consistent with a substantial stabilization of the A·T region nearest the G·C block. The melting temperature T A·T for the average A·T pari in d(C20A10)·d(T10G20), with 10 A·T pairs, appears to be 4°C greater than TA·T for d(C15A15)·d(T15G15) and d(C20A15)·d(T15G20), both with 15 A·T pairs. Actinomycin bound to the G·C end effectively stabilizes the A·T end by 9°C. These results indicate a long-range contribution to the interactions governing DNA stability. A possible mechanism for these interactions will be discussed.  相似文献

Inhibition of microsomal prostaglandin E synthase-1 (mPGES-1) results in decreased production of proinflammatory PGE2 and can lead to shunting of PGH2 into the prostaglandin D2 (PGD2)/15-deoxy-Δ12,14-prostaglandin J2 (15dPGJ2) pathway. 15dPGJ2 forms Michael adducts with thiol-containing biomolecules such as GSH or cysteine residues on target proteins and is thought to promote resolution of inflammation. We aimed to elucidate the biosynthesis and metabolism of 15dPGJ2 via conjugation with GSH, to form 15dPGJ2-glutathione (15dPGJ2-GS) and 15dPGJ2-cysteine (15dPGJ2-Cys) conjugates and to characterize the effects of mPGES-1 inhibition on the PGD2/15dPGJ2 pathway in mouse and human immune cells. Our results demonstrate the formation of PGD2, 15dPGJ2, 15dPGJ2-GS, and 15dPGJ2-Cys in RAW264.7 cells after lipopolysaccharide stimulation. Moreover, 15dPGJ2-Cys was found in lipopolysaccharide-activated primary murine macrophages as well as in human mast cells following stimulation of the IgE-receptor. Our results also suggest that the microsomal glutathione S-transferase 3 is essential for the formation of 15dPGJ2 conjugates. In contrast to inhibition of cyclooxygenase, which leads to blockage of the PGD2/15dPGJ2 pathway, we found that inhibition of mPGES-1 preserves PGD2 and its metabolites. Collectively, this study highlights the formation of 15dPGJ2-GS and 15dPGJ2-Cys in mouse and human immune cells, the involvement of microsomal glutathione S-transferase 3 in their biosynthesis, and their unchanged formation following inhibition of mPGES-1. The results encourage further research on their roles as bioactive lipid mediators.  相似文献

Following an invasion of exotic annual grasses into California oak woodlands, grass species dominance shifted from native perennials to exotic annuals. In combination with other ecosystem and species characteristics, species-specific N preferences may influence species coexistence and dominance. If species N preferences follow dominance patterns in California oak woodlands, then the more dominant exotic grasses should prefer the most abundant inorganic soil N form (NH4+), while the subordinate native grasses should prefer the less available inorganic (NO3?) or organic (glycine) soil N forms. To investigate this prediction, we applied 15N-labeled NH4+, NO3?, and glycine to soil and measured % 15N recovery by two dominant annual grasses (Bromus diandrus and Bromus hordeaceus) and two subordinate perennial grasses (Elymus glaucus and Nassella pulchra). As expected, shoots of B. diandrus recovered more 15N-NH4+ (74%) than either 15N-NO3? (51%) or 15N-glycine (39%). B. diandrus also captured at least 3.2 times more 15N-NH4+ than subordinate grasses. Dominant B. hordeaceus, however, demonstrated no N form preferences. As hypothesized, shoots of subordinate E. glaucus and N. pulchra recovered 2.1–2.3 times more 15N-NO3? than 15N-NH4+ and increased %N by 4.8–5.7% in response to the application of 15N-NO3?. Both subordinate grasses did not prefer 15N-glycine over 15N-NH4+, suggesting that the importance of this N form in structuring species coexistence in California oak woodlands is minimal. These results support our theory that species N preferences follow dominance patterns in California oak woodlands. To further understand the role of these species-specific N preferences in structuring dominance, the importance of N form versus such characteristics as rooting distribution and species phenologies needs to be explored in the presence of interspecific competition.  相似文献

The semisynthesis of homologues of aprotinin, the bovine pancreatic trypsin inhibitor, is described. The P1 lysine15 residue was replaced by two methods. The first procedure, which consisted of two enzymatic steps for the incorporation of other amino acids has previously been described. The second approach consisted of six steps of both enzymatic and chemical nature. The modified inhibitor, in which the lysine15-alanine16 peptide bond is hydrolyzed, was used as the starting material. All carboxyl groups of the modified inhibitor were esterified with methanol; the lysine15 methylester group was then selectively hydrolyzed. Afterward, lysine15 itself was split off. Arginine, glutamic acid, methionine, andl-2-aminohexanoic acid (norleucine, Nle) were incorporated using water-soluble carbodiimide combined with an acylation catalyst. The methylester group was used to prevent polymerization. The reactive-site peptide bonds were resynthesized using either chymotrypsin or trypsin.  相似文献

The corpus luteum inhibiting properties of eighteen 15-methyl prostaglandin analogs were determined in the rhesus monkey during concomitant stimulation of the corpus luteum with chorionic gonadotropin. The methyl ester of (15S)-15-methyl PGF2α (15M-PGF2α, 12.5 mg/monkey) lowered serum progesterone to 12% of pretreatment values within 24 hours, however progesterone returned to normal limits within 48 hours. Elongation of the top side-chain by two carbons (2a,2b-dihomo-15M-PGF2α methyl ester, 13 mg/monkey), substitution of a hydroxymethyl group at carbon 1 (2-decarboxy-2-hydroxymethyl-15M-PGF2α, 12 mg/monkey), or the formation of the carbon 1 amide (15M-PGF2α amide, 12.5 mg/monkey) improved the inhibitory activity of 15M-PGF2α; serum progesterone for these 3 analogs was depressed to 15–30% of pretreatment levels within 24 hours, and did not return to control values. Luteal function was not inhibited (12 or more mg/monkey) when the 15-methyl group was placed in the R configuration, the top side chain was shortened by two carbons, an amino group was substituted for carbon 1, the 5-oxa modification was added, or the 1,9-lactone was formed. Some other modifications of 15M-PGF2α were also inactive, although not all were tested at equivalent doses: 2,2-difluoro; 4,5-cis-didehydro; 9,11-dideoxy-9α,11α-dichloro; 11-deoxy; 17-phenyl; 1,15-lactone; and the p-benzamidophenyl ester of 2a,2b-dihomo-15M-PGF2α. (15S)-15-Methyl PGE2 methyl ester (1 mg/monkey) depressed serum progesterone concentrations to 42% of pretreatment values within 24 hours; 2a,2b-dihomo-11-deoxy-(15S)-15-methyl PGE2 methyl ester was inactive (5 mg/monkey). A corpus luteum inhibiting action of certain 15-methyl prostaglandins can be demonstrated in the rhesus monkey.  相似文献

The 15N paramagnetic shifts of iron-bound C15N? were studied for myoglobin, hemoglobin, cytochrome c and other modified hemoproteins. Two characteristic 15N resonances at 977 and 1045 ppm (with respect to 15NO3? as an internal standard) were found for human adult hemoglobin cyanide, while only single resonances were observed for other cyano hemoproteins. These two resonances are assigned to iron-bound C15N of α and β subunits of hemoglobin. The substantial difference in the C15N isotropic shifts in various hemoproteins is discussed in relation to iron-proximal histidine binding and heme-apoprotein interactions.  相似文献

Abstract

15N uniformly labeled lac repressor and lac repressor headpiece were prepared. 15N NMR spectra of lac repressor were shown resolution inadequate for detailed study while the data showed that the 15N labeled N-terminal part of the protein is quite suitable for this type of study allowing future investigation of the specific interaction of the lac repressor headpiece with the lac operator. We report here the total assignment of proton 1H and nitrogen 15NH backbone resonances of this headpiece in the free state. Assignments of the 15N resonances of the protein were obtained in a sequential manner using heteronuclear multiple quantum coherence (HMQC), relayed HMQC nuclear Overhauser and relayed HMQC-HOHAHA spectroscopy. More than 80 per cent of residues were assigned by their 15NH(i)-N1H(i+1) and 15NH(i)-N1(i-1) connectivities. Values of the 3JNHα splitting for 39 of the 51 residues of the headpiece were extracted from HMQC and HMQC-J. The observed 15NH(i)-CβH cross peaks and the 3JNHα coupling constants values are in agreement with the three α-helices previously described [Zuiderweg, E.R.P., Scheek, R.M., Boelens, R., van Gunsteren, W.F. and Kaptein, R., Biochimie 67, 707 (1985)]. The 3JNHα coupling constants can be now used for a more confident determination of the lac repressor headpiece. From these values it is shown that the geometry of the ends of the second and third α-helices exhibit deviation from the canonical α-helix structure. On the basis of NOEs and 3JNHα values, the geometry of the turn of the helix-turn-helix motif is discussed.  相似文献

The elucidation of the molecular recognition of adenosine deaminase (ADA), the interpretation of the catalytic mechanism, and the design of novel inhibitors are based mostly on data obtained for the crystalline state of the enzyme. To obtain evidence for molecular recognition of the physiologically relevant soluble enzyme, we studied its interactions with the in situ formed inhibitor, 6-OH-purine riboside (HDPR), by 1D-15N- and 2D-(1H-15N)- NMR using the labeled primary inhibitor [15N4]-PR. We synthesized both [15N4]-PR and an [15N4]-HDPR model, from relatively inexpensive 15N sources. The [15N4]-HDPR model was used to simulate H-bonding and possible Zn2+-coordination of HDPR with ADA. We also explored possible ionic interactions between PR and ADA by 15N-NMR monitored pH-titrations of [15N4]-PR. Finally, we investigated the [15N4]-PR-ADA 1:1 complex by 2D-(1H-15N) NMR. We found that HDPR recognition determinants in ADA do not include any ionic-interactions. HDPR N1 H is an H-bond acceptor, and not an H-bond donor. Despite the proximity of N7 to the Zn2+-ion, no coordination occurs; instead, N7 is an H-bond acceptor. We found an overall agreement between the crystallographic data for the crystallized ADA:HDPR complex and the 15N-NMR signals for the corresponding soluble complex. This finding justifies the use of ADA's crystallographic data for the design of novel inhibitors.  相似文献

In order to evaluate the efficacy and acceptability of 15(S)-15-methyl-prostaglandin F (15-me-PGF) for pregnancy termination, we induced 30 abortions with single intra-amniotic injections of 2,5 mg of 15-me-PGF and 25 abortions with intra-muscular 15-me-PGF administered 200 μg initially and 300 μg every third hour until 30 hrs or abortion. Abortion occurred within 30 hrs in 97 % of cases in the intra-amniotic group, with a mean abortion time of 17,6 hrs and in 80 % in the intramuscular group, with a mean abortion time of 15.0 hrs. Neither parity nor gestational age was significantly related to the abortifacient efficacy of 15-me-PGF. No serious complications occurred. Vomiting (83–84 %) and diarrhoea (23–92 %) were the most common complaints. Uterine contractions were more painful if induction was effected with intra-amniotic rather than intramuscular injections. 15-me-PGF appears to be an effective and practicable abortifacient which can be used intra-amniotically or intramuscularly according to the ease of amniocentesis.  相似文献

Summary The simultaneous acquisition of a 4D gradient-enhanced and sensitivity-enhanced [13C,15N]/[15N,15N]-separated NOESY is presented for the 74-residue [13C,15N]-labeled N-terminal SH3 domain of mGrb2 complexed with a peptide gragment from mSOS-2 in 90% H2O. The method readily accommodates different 13C and 15N spectral widths, but requires that the same number of increments be collected for both 13C and 15N in the simultaneous dimension (F2). For purposes of display and analysis, the two 4D spectra can be deconvolved during the processing stage by the appropriate linear combination of separately stored FIDs. Compared to collecting each of these two 4D data sets separately, the presented method is a factor (2)1/2 more efficient in sensitivity per unit acquisition time. The interleaved nature of this method may also lead to improved peak registration between the two 4D spectra.  相似文献

Li15Si4, the only crystalline phase that forms during lithiation of the Si anode in lithium‐ion batteries, is found to undergo a structural transition to a new phase at 7 GPa. Despite the large unit cell of Li15Si4 (152 atoms in the unit cell), ab initio evolutionary metadynamics (using the USPEX code) successfully predicts the atomic structure of this new phase (β‐Li15Si4), which has an orthorhombic structure with an Fdd2 space group. In the new β‐Li15Si4 phase Si atoms are isolated by Li atoms analogous to the original cubic phase (α‐Li15Si4), whereas the atomic packing is more efficient owing to the higher Si? Li coordination number and shorter Si? Li, Li? Li bonds. β‐Li15Si4 has substantially larger elastic moduli compared with α‐Li15Si4, and has a good electrical conductivity. As a result, β‐Li15Si4 has superior resistance to deformation and fracture under stress. The theoretical volume expansion of Si would decrease 25% if it transforms to β‐Li15Si4, instead of α‐Li15Si4, during lithiation. Moreover, β‐Li15Si4 can be recovered back to ambient pressure, providing opportunities to further investigate its properties and potential applications.  相似文献

Solid-state 15N NMR spectroscopy was used to determine the chemical nature of nitrogen in 15N-enriched material from the roots and stems of wheat (Triticum aesitivum), field pea (Pisum sativum) and kikuyu grass (Pennisetum clandestinum) and from the roots, stems and leaves of a eucalyptus species (Eucalyptus globulus). Nitrogen-15 cross polarization (CP) spectra of the materials were all very similar, with 64–75% of total signal assigned to amide N. Spin counting analysis indicated that 37–80% of potential signal was accounted for in the CP spectra, and that NMR observability using the CP technique (N obs -CP) was higher for stems and leaves than for roots, and higher for wheat and eucalyptus than for peas and kikuyu. The 15N direct polarization (DP) spectra contained higher proportions of signal assigned to amine (up to 22%) and nitrate (up to 17%), and less assigned to amide N (50–72%) than the corresponding CP spectra. Spin counting analysis indicated that 68–93% of potential signal was accounted for in the DP spectra, confirming the DP technique to be more quantitatively reliable than CP.  相似文献

The systematic difference between T 2 values obtained from CPMG and T 1 experiments was observed for backbone 15N nuclei of bacterial ribonuclease barnase. Theoretical consideration suggests that the observed difference is caused by off-resonance effects of 180° pulses of the CPMG pulse train. Namely, at off-resonance conditions T 1-dependent secondary echo coherence pathways considerably contribute to the signal decay in the CPMG experiment and result in systematic (up to 10%) offset-dependent overestimation of 15N T 2 measured by the CPMG technique. Under certain circumstances off-resonance effects result in dependence of 15N T 2 on CPMG frequency, which might be erroneously interpreted as conformational exchange on the millisecond time-scale. A procedure for numerical correction of 15N T 2 (CPMG) data is proposed.  相似文献
