Studies of seedling population dynamics often focus on survival because it provides an integrated measure of seedling performance. However, this approach involves a substantial loss of information because survival is the net result of a wide range of mechanisms. The present study overcomes these shortcomings by investigating spatial and temporal patterns in the causes of plant mortality in a population of Helianthemum squamatum seedlings. We use new point pattern analyses based on K functions combined with a new null model (“independent labeling”). A total of 871 seedlings of H.squamatum were mapped and regularly monitored over an 18‐month period. More than 60% of seedlings died during this period. Causes of mortality were spatially structured, and these structures shifted through time. Small differences in either the time of emergence or the environment surrounding H. squamatum seedlings had profound influences on their fate. Seedlings emerging late in the season under the canopy of adult plants died from drought more often than expected, whereas those emerging earlier in the same microsite survived more than expected. The identity of neighbors also affected the spatio‐temporal dynamics of mortality causes. Our results show that seedling‐adult interactions cannot be easily predicted from simple models, and that the time of seedling emergence, its age and the identity of its neighbors determine the sign and the spatial scale of these interactions. The new methods introduced in this article open an avenue for the detailed analyses of the spatio‐temporal dynamics of plant mortality and can help to disentangle the complexity of biotic interactions along environmental severity gradients.  相似文献

Quebec's struggle between francophones and anglophones has deeply affected other immigrant communities' identitarian practices. Within Montreal's eighties and nineties nationalist environment some Italo-Quebecois worked as a bridge between the two groups through the transcultural magazine Vice Versa. The article presents some findings from a case study that looks at how Vice Versa challenged dominant perceptions of identity and its role in society. It assumes that identity is negotiated between individuals, that language and ethnicity are key factors for social mobility and symbolic power on the marketplace, and that these function through hegemony. Through its use of untranslated French, English and Italian and its articles' contents Vice Versa's discourse on transculture undermines the assumption that identity is necessary for social interaction and negotiates new categories that dissolve ethnic boundaries. Vice Versa is a unique example of the emergence of a counter-hegemonic movement questioning identity at its roots.  相似文献

The influence of spatial variation in the carrying capacity and migration rate of a geographical barrier on the one-dimensional stepping-stone model is studied. The monoecious, diploid population is subdivided into an infinite linear array of panmictic colonies that exchange gametes. In each deme, the rate of self-fertilization is equal to the reciprocal of the number of individuals in that deme. Generations are discrete and nonoverlapping; the analysis is restricted to a single locus in the absence of selection; every allele mutates to new alleles at the same rate. In the diffusion approximation, a partial differential equation that incorporates spatial (and temporal) variation in the carrying capacity and migration rate is derived for the probability of identity. Transition conditions that simultaneously take into account discontinuities in the carrying capacity and migration rate are established: the probability of identity is continuous, but its partial derivatives are not, their ratio being a simple function of the carrying capacities and migrational variance on the two sides of the inhomogeneity. The partial derivatives of the probability of identity are continuous across a geographical barrier, whereas the probability of identity itself has a discontinuity proportional to the partial derivative at the barrier, the constant of proportionality being a measure of the difficulty of crossing the barrier.  相似文献

Here we deal with the influence of heat-transport effects on a high-pressure-induced enzyme inactivation in packed substances. Special attention is given to the influence of the geometrical scale and to the heat-transfer characteristics of the packaging material. The investigation is based on mathematical modeling and numerical simulation. The method accounts both for compression phase and holding phase. The model includes convective and conductive heat transfer, fluid motion as well as an enzyme transport equation with a first-order kinetic source term accounting for the inactivation. Three configurations with a total volume of 0.8 L, 6.3 L, and 50.3 L are considered. The pressure medium is water. The enzyme solution is B. subtilis alpha-amylase dissolved in a TRIS-HCl-buffer. The packaging material is polypropylene. The heat-transfer coefficient for conduction through the packaging material is varied to simulate both changes in the material properties as well as modifications of the packaging material thickness. It is found that the efficiency of the inactivation increases with increasing chamber volume as long as the kinetic inactivation constant is increasing with temperature. In the considered case the activity retention obtained in a 0.8 L volume is about 2.4 times larger than the one obtained for the same process carried out in a 50.3 L volume. Furthermore, it was found that the properties of the packaging material could induce a significant degree of nonuniformity (worst case = 69%). An appropriate choice of the material can lead to maximum inactivation and maximum process uniformity since advantage is taken from the slow heat exchange after the compression phase.  相似文献

What is the relation between things and theories, the material world and its scientific representations? This is a staple philosophical problem that rarely counts as historically legitimate or fruitful. In the following dialogue, the interlocutors do not argue for or against realism. Instead, they explore changing relations between theories and things, between contested objects of knowledge (like the cell) and less contested, more everyday things (like frog eggs scooped from a pond). Widely seen as the life sciences' first general theory, the cell theory underwent dramatic changes during the nineteenth century. The dialogue established that each successive version of the cell theory was formulated - each identity of the object cell was formed - around a different material: cork, cartilage, eggs in cleavage, muscle. Such things thus serve as exemplary materials, in ways not described by standard concepts like induction, theory-testing, theory-laden observation, and construction. Still, how can theories and perspective possibly be honed on things if these are apprehended differently by different observers according to their interests, training, culture, or indeed theories? The second part of the dialogue addresses this problem, partly through the verbal and visual schemata that were used by nineteenth-century microscopists and that are comparable to schemata in the visual arts. The third part of the dialogue considers the exemplary materials as a historical sequence, itself needing explanation. Theoretical change devolved partly from wider histories and geographies of the prevalence, availability, or scientific and cultural status of materials such as plants, animals, and muscle.  相似文献

The successive expression of neuronal transients is related to dynamic correlations and, as shown in this paper, to dynamic instability. Dynamic instability is a form of complexity, typical of neuronal systems, which may be crucial for adaptive brain function from two perspectives. The first is from the point of view of neuronal selection and self-organizing systems: if selective mechanisms underpin the emergence of adaptive neuronal responses then dynamic instability is, itself, necessarily adaptive. This is because dynamic instability is the source of diversity on which selection acts and is therefore subject to selective pressure. In short, the emergence of order, through selection, depends almost paradoxically on the instabilities that characterize the diversity of brain dynamics. In this communication, we present a unifying framework to understand the emergence and maintenance of diversity in ecological systems. We do this by developing a deterministic population model including density-dependent limitation in resources and available space. Our model shows that competitive exclusion and neutral coexistence represent different regimes of the same adaptive dynamics suggesting that neutrality is the general result of an adaptive process in a finite habitat with limited energetic resources. Our model explains the emergence of biodiversity through mutation and its maintenance through neutrality. We show that this framework provides the theoretical foundations to understand the emergence and maintenance of diversity in microbial ecosystems.  相似文献

Here, we present a protocol to estimate material and surface optical properties using the photoacoustic effect combined with total internal reflection. Optical property evaluation of thin films and the surfaces of bulk materials is an important step in understanding new optical material systems and their applications. The method presented can estimate thickness, refractive index, and use absorptive properties of materials for detection. This metrology system uses evanescent field-based photoacoustics (EFPA), a field of research based upon the interaction of an evanescent field with the photoacoustic effect. This interaction and its resulting family of techniques allow the technique to probe optical properties within a few hundred nanometers of the sample surface. This optical near field allows for the highly accurate estimation of material properties on the same scale as the field itself such as refractive index and film thickness. With the use of EFPA and its sub techniques such as total internal reflection photoacoustic spectroscopy (TIRPAS) and optical tunneling photoacoustic spectroscopy (OTPAS), it is possible to evaluate a material at the nanoscale in a consolidated instrument without the need for many instruments and experiments that may be cost prohibitive.  相似文献

Human languages are rule governed, but almost invariably these rules have exceptions in the form of irregularities. Since rules in language are efficient and productive, the persistence of irregularity is an anomaly. How does irregularity linger in the face of internal (endogenous) and external (exogenous) pressures to conform to a rule? Here we address this problem by taking a detailed look at simple past tense verbs in the Corpus of Historical American English. The data show that the language is open, with many new verbs entering. At the same time, existing verbs might tend to regularize or irregularize as a consequence of internal dynamics, but overall, the amount of irregularity sustained by the language stays roughly constant over time. Despite continuous vocabulary growth, and presumably, an attendant increase in expressive power, there is no corresponding growth in irregularity. We analyze the set of irregulars, showing they may adhere to a set of minority rules, allowing for increased stability of irregularity over time. These findings contribute to the debate on how language systems become rule governed, and how and why they sustain exceptions to rules, providing insight into the interplay between the emergence and maintenance of rules and exceptions in language.  相似文献

It is generally assumed that the complex map of metabolism is a result of natural selection working at the molecular level. However, natural selection can only work on entities that have three basic features: information, metabolism and membrane. Metabolism must include the capability of producing all cellular structures, as well as energy (ATP), from external sources; information must be established on a material that allows its perpetuity, in order to safeguard the goals achieved; and membranes must be able to preserve the internal material, determining a selective exchange with external material in order to ensure that both metabolism and information can be individualized. It is not difficult to understand that protocellular entities that boast these three qualities can evolve through natural selection. The problem is rather to explain the origin of such features under conditions where natural selection could not work. In the present work we propose that these protocells could be built by chemical evolution, starting from the prebiotic primordial soup, by means of chemical selection. This consists of selective increases of the rates of certain specific reactions because of the kinetic or thermodynamic features of the process, such as stoichiometric catalysis or autocatalysis, cooperativity and others, thereby promoting their prevalence among the whole set of chemical possibilities. Our results show that all chemical processes necessary for yielding the basic materials that natural selection needs to work may be achieved through chemical selection, thus suggesting a way for life to begin.  相似文献

Centromeres are differentiated chromatin domains, present once per chromosome, that direct segregation of the genome in mitosis and meiosis by specifying assembly of the kinetochore. They are distinct genetic loci in that their identity in most organisms is determined not by the DNA sequences they are associated with, but through specific chromatin composition and context. The core nucleosomal protein CENP-A/cenH3 plays a primary role in centromere determination in all species and directs assembly of a large complex of associated proteins in vertebrates. While CENP-A itself is stably transmitted from one generation to the next, the nature of the template for centromere replication and its relationship to kinetochore function are as yet poorly understood. Here, we investigate the assembly and inheritance of a histone fold complex of the centromere, the CENP-T/W complex, which is integrated with centromeric chromatin in association with canonical histone H3 nucleosomes. We have investigated the cell cycle regulation, timing of assembly, generational persistence, and requirement for function of CENPs -T and -W in the cell cycle in human cells. The CENP-T/W complex assembles through a dynamic exchange mechanism in late S-phase and G2, is required for mitosis in each cell cycle and does not persist across cell generations, properties reciprocal to those measured for CENP-A. We propose that the CENP-A and H3-CENP-T/W nucleosome components of the centromere are specialized for centromeric and kinetochore activities, respectively. Segregation of the assembly mechanisms for the two allows the cell to switch between chromatin configurations that reciprocally support the replication of the centromere and its conversion to a mitotic state on postreplicative chromatin.  相似文献

This article explores the material practices through which lower-caste and poor villagers engage with bureaucracy in contemporary India. We take documents and paperwork – such as ration cards and community certificates – as a ‘lens’ through which to explore how paper materiality is infused with the politics of power, patronage, and identity. The article brings ethnography from rural Tamil Nadu, South India, in conversation with two bodies of literature: one on the materiality of bureaucracy and one on the nature of political mediation in contemporary India. We demonstrate how everyday engagements with paperwork as well as processes of applying, form filling, and securing recommendations are constitutive of social and political relationships and, ultimately, of citizenship itself. Political mediation around paperwork and bureaucracy generates a hierarchy of citizens rather than equal citizenship for all, yet ordinary villagers transpire as anything but passive. Drawing on patronage networks, engaging in affective performances, and navigating a politics of identity, they actively negotiate access to the state in an attempt to claim their rights as citizens.  相似文献

Purpose

In the transportation sector, reducing vehicle weight is a cornerstone strategy to improve the fuel economy and energy efficiency of road vehicles. This study investigated the environmental implications of lightweighting two automotive parts (Ford Taurus front end bolster, Chevrolet Trailblazer/GMC Envoy assist step) using glass-fiber reinforced polymers (GFRP) instead of steel alloys.

Methods

The cradle-to-grave life cycle assessments (LCAs) for these studies consider a total service life of 150,000 miles for two applications: a 46 % lighter GFRP bolster on the 2010 Ford Taurus that replaced the 2008 steel and GFRP bolster, and a 51 % lighter GFRP running board for the 2007 Chevrolet Trailblazer/GMC Envoy that replaced the previous steel running board including its polymer fasteners. The life cycle stages in these critically reviewed and ISO-compliant LCA studies include the production of upstream materials and energy, product manufacturing, use, and the end-of-life treatment for all materials throughout the life cycle.

Results and discussion

The results show that the lighter GFRP products performed better than the steel products for global warming potential and primary energy demand for both case studies. In addition, the GFRP bolster performed better for acidification potential. The savings of fuel combustion and production during the use stage of a vehicle far outweigh the environmental impacts of manufacturing or end-of-life. An even greater benefit would be possible if the total weight reduction in the vehicle would be high enough to allow for the reduction of engine displacement or an elongation of gear ratio while maintaining constant vehicle dynamics. These so-called secondary measures allow the fuel savings per unit of mass to be more than doubled and are able to offset the slightly higher acidification potential of the GFRP running board which occurs when only the mass-induced fuel savings are considered.

Conclusions

The lightweight GFRP components are shown to outperform their steel counterparts over the full life cycle mainly due to the reduced fuel consumption of the vehicle in the use phase. To harvest the benefits of light weighting to their full extent, it is recommended that the sum of all mass reductions in the design process be monitored and, whenever feasible, invested into fuel economy by adapting the drive train while maintaining constant vehicle performance rather than leveraging the weight reduction to improve vehicle dynamics.  相似文献

The bacterial transferrin ferric binding protein A (FbpA) requires an exogenous anion to facilitate iron sequestration, and subsequently to shuttle the metal across the periplasm to the cytoplasmic membrane. In the diverse conditions of the periplasm, numerous anions are known to be present. Prior in vitro experiments have demonstrated the ability of multiple anions to fulfill the synergistic iron-binding requirement, and the identity of the bound anion has been shown to modulate important physicochemical properties of iron-bound FbpA (FeFbpA). Here we address the kinetics and mechanism of anion exchange for the FeFbpA–nitrilotriacetate (NTA) assembly with several biologically relevant anions (citrate, oxalate, phosphate, and pyrophosphate), with nonphysiologic NTA serving as a representative synergistic anion/chelator. The kinetic data are consistent with an anion-exchange process that occurs in multiple steps, dependent on the identity of both the entering anion and the leaving anion. The exchange mechanism may proceed either as a direct substitution or through an intermediate FeFbpA–X* assembly based on anion (X) identity. Our kinetic results further develop an understanding of exogenous anion lability in the periplasm, as well as address the final step of the iron-free FbpA (apo-FbpA)/Fe3+ sequestration mechanism. Our results highlight the kinetic significance of the FbpA anion binding site, demonstrating a correlation between apo-FbpA/anion affinity and the FeFbpA rate of anion exchange, further supporting the requirement of an exogenous anion to complete tight sequestration of iron by FbpA, and developing a mechanism for anion exchange within FeFbpA that is dependent on the identity of both the entering anion and the leaving anion.  相似文献

Cyclase-associated proteins (CAPs) are among the most highly conserved regulators of actin dynamics, being present in organisms from mammals to apicomplexan parasites. Yeast, plant, and mammalian CAPs are large multidomain proteins, which catalyze nucleotide exchange on actin monomers from ADP to ATP and recycle actin monomers from actin-depolymerizing factor (ADF)/cofilin for new rounds of filament assembly. However, the mechanism by which CAPs promote nucleotide exchange is not known. Furthermore, how apicomplexan CAPs, which lack many domains present in yeast and mammalian CAPs, contribute to actin dynamics is not understood. We show that, like yeast Srv2/CAP, mouse CAP1 interacts with ADF/cofilin and ADP-G-actin through its N-terminal α-helical and C-terminal β-strand domains, respectively. However, in the variation to yeast Srv2/CAP, mouse CAP1 has two adjacent profilin-binding sites, and it interacts with ATP-actin monomers with high affinity through its WH2 domain. Importantly, we revealed that the C-terminal β-sheet domain of mouse CAP1 is essential and sufficient for catalyzing nucleotide exchange on actin monomers, although the adjacent WH2 domain is not required for this function. Supporting these data, we show that the malaria parasite Plasmodium falciparum CAP, which is entirely composed of the β-sheet domain, efficiently promotes nucleotide exchange on actin monomers. Collectively, this study provides evidence that catalyzing nucleotide exchange on actin monomers via the β-sheet domain is the most highly conserved function of CAPs from mammals to apicomplexan parasites. Other functions, including interactions with profilin and ADF/cofilin, evolved in more complex organisms to adjust the specific role of CAPs in actin dynamics.  相似文献

For many bacterial infections, drug resistant mutants are likely present by the time antibiotic treatment starts. Nevertheless, such infections are often successfully cleared. It is commonly assumed that this is due to the combined action of drug and immune response, the latter facilitating clearance of the resistant population. However, most studies of drug resistance emergence during antibiotic treatment focus almost exclusively on the dynamics of bacteria and the drug and neglect the contribution of immune defenses. Here, we develop and analyze several mathematical models that explicitly include an immune response. We consider different types of immune responses and investigate how each impacts the emergence of resistance. We show that an immune response that retains its strength despite a strong drug-induced decline of bacteria numbers considerably reduces the emergence of resistance, narrows the mutant selection window, and mitigates the effects of non-adherence to treatment. Additionally, we show that compared to an immune response that kills bacteria at a constant rate, one that trades reduced killing at high bacterial load for increased killing at low bacterial load is sometimes preferable. We discuss the predictions and hypotheses derived from this study and how they can be tested experimentally.  相似文献
