中国部分黄牛品种mtDNA遗传多态性研究   总被引:49,自引:4,他引:49  
对我国8个黄牛品种22个个体的mtDNA D-loop区910bp全序列进行了分析。结果表明:8个黄牛品种D-loop区序列中,A T平均含量为61.65%;经比对,共检测到66个核苷酸多态位点,约占核苷酸总数的7.25%;D-loop全序列突变类型有5种,即转换、颠换、插入、缺失及转换与颠换共存,它们分别占核苷酸多态位点的81.82%、6.06%、7.57%、3.03%及1.52%。以欧洲牛mtDNA D-loop全序列为标准,8个黄牛群体D-loop的平均核苷酸变异率分3个层次:西镇牛、蒙古牛、黑白花牛及秦川牛的核苷酸变异率最低,分别为0.37%、0.44%、0.52%和0.66%;南阳牛与郏县红牛的核苷酸变异率居中,分别为1.91%和2.02%;晋南牛与岳阳牛的核苷酸变异率最高,分别为4.47%和4.73%。中国黄牛品种内D-loop区序列歧异度为0.55%~5.39%,品种间序列歧异度为1.21%~6.59%。在所测黄牛个体中,mtDNA D-loop序列由19种单倍型组成,单倍型比例为86.36%,说明中国黄牛mtDNA遗传多态性很丰富。由此构建了中国8个黄牛品种的NJ分子系统树,聚类分析表明:所测黄牛的mtDNA D-loop序列表现为3个单倍型组,从而揭示中国黄牛可能有3个母系起源,以普通牛起源和瘤牛起源为主。  相似文献

对贵州4个地方黄牛品种共计82个个体的线粒体DNA D-loop区全序列910 bp进行分析,检测到31种单倍型,其核苷酸多态位点65个,约占所测核苷酸总长的7.14%,其中有62个转换,2个颠换,1个转换/颠换共存。贵州4个黄牛品种mtDNA D-loop区核苷酸多样度(π值)为2.16%~2.61%,单倍型多样度(H)为0.695~0.909,表明贵州黄牛mtDNA遗传多样性比较丰富。根据单倍型构建了贵州4个黄牛品种的NJ分子系统树。聚类表明,贵州黄牛有普通牛和瘤牛2大母系起源,其影响较为均一。并探讨了用核苷酸多样度π值的大小来衡量黄牛群体遗传分化程度的可行性。   相似文献

Next generation sequencing of mitochondrial DNA (mtDNA) facilitates studies into the metabolic characteristics of production animals and their relation to production traits. Sequence analysis of mtDNA from pure-bred swine with highly disparate production characteristics (Mangalica Blonde, Mangalica Swallow-bellied, Meishan, Turopolje, and Yorkshire) was initiated to evaluate the influence of mtDNA polymorphisms on mitochondrial function. Herein, we report the complete mtDNA sequences of five Sus scrofa breeds and evaluate their position within the phylogeny of domestic swine. Phenotypic traits of Yorkshire, Mangalica Blonde, and Swallow-belly swine are presented to demonstrate their metabolic characteristics. Our data support the division of European and Asian breeds noted previously and confirm European ancestry of Mangalica and Turopolje breeds. Furthermore, mtDNA differences between breeds suggest function-altering changes in proteins involved in oxidative phosphorylation such as ATP synthase 6 (MT-ATP6), cytochrome oxidase I (MT-CO1), cytochrome oxidase III (MT-CO3), and cytochrome b (MT-CYB), supporting the hypothesis that mtDNA polymorphisms contribute to differences in metabolic traits between swine breeds. Our sequence data form the basis for future research into the roles of mtDNA in determining production traits in domestic animals. Additionally, such studies should provide insight into how mtDNA haplotype influences the extreme adiposity observed in Mangalica breeds.  相似文献

中国部分牦牛品种线粒体DNA遗传多态性研究   总被引:29,自引:1,他引:29  
采用DNA测序技术首次测定了中国5个牦牛品种(类群)35个个体线粒体DNA控制区(D loop)全序列,结果表明:牦牛线粒体DNA控制区全序列长度为891~895bp,T、C、A、G4种核苷酸的含量分别为28.5%、25.3%、32.5%、13.7%。检测到55个变异位点,约占分析位点总数的6.16%,核甘酸变异类型有转换、颠换、插入/缺失4种。确定了24种单倍型,单倍型H4和H6为中国牦牛的主体单倍型,各单倍型在品种间分布不平衡,所测定的5个牦牛品种(类群)单倍型多样度平均为0.9697±0.0180,说明我国牦牛D loop单倍型类型丰富。牦牛品种内各序列平均核苷酸差异数为10.936,核苷酸多样度为1.231%;牦牛品种间核苷酸分歧度(Dxy)约为0.760%~2.155%,品种间双参数距离范围为0.000~0.029,表明我国牦牛遗传多态性丰富。对D环序列变异的分子方差分析和单倍型网络关系图的结果表明:我国牦牛品种间出现显著的遗传分化,牦牛单倍型网络关系图聚为2个聚类簇,表明我国牦牛有2个母系来源,或者有2个主要的驯化地点。  相似文献

目的分析五指山小型猪、巴马小型猪和贵州香猪线粒体DNA控制区碱基序列,比较研究不同猪种的遗传标志。方法应用PCR技术分别对这三种小型猪的血液总DNA样品中线粒体DNA D-loop区进行扩增,测序比对。结果猪的线粒体DNA D-loop区分三个区域。I区(靠近5’端区域)704bp,五指山小型猪在此区共有6个变异位点,通过6个变异位点中归纳出3个单倍体,而巴马小型猪在此区有9个变异位点,通过9个变异位点归纳出4个单倍体,贵州香猪在此区共有6个变异位点,通过6个变异位点归纳出3个单倍体。Ⅱ区(串联重复序列区),五指山小型猪、巴马小型猪和贵州香猪序列相同。Ⅲ区(靠近3’端区域)三种小型猪的序列几乎相同。结论五指山小型猪、巴马小型猪和贵州香猪三种小型猪之间线粒体DNA碱基序列变异位点较少,五指山小型猪和巴马小型猪亲缘关系较近。  相似文献

Structural DNA Nanotechnology uses unusual DNA motifs to build target shapes and arrangements. These unusual motifs are generated by reciprocal exchange of DNA backbones, leading to branched systems with many strands and multiple helical domains. The motifs may be combined by sticky ended cohesion, involving hydrogen bonding or covalent interactions. Other forms of cohesion involve edge-sharing or paranemic interactions of double helices. A large number of individual species have been developed by this approach, including polyhedral catenanes, a variety of single-stranded knots, and Borromean rings. In addition to these static species, DNA-based nanomechanical devices have been produced that are ultimately targeted to lead to nanorobotics. Many of the key goals of structural DNA nanotechnology entail the use of periodic arrays. A variety of 2D DNA arrays have been produced with tunable features, such as patterns and cavities. DNA molecules have be used successfully in DNA-based computation as molecular representations of Wang tiles, whose self-assembly can be programmed to perform a calculation. About 4 years ago, on the fiftieth anniversary of the double helix, the area appeared to be at the cusp of a truly exciting explosion of applications; this was a correct assessment, and much progress has been made in the intervening period.  相似文献

运用19种限制性内切酶对中国11个家鹅品种138个样本进行了mtDNA的限制性片段长度多态性(RFLP)分析。在使用的19种内切酶中,有7个酶检测出多态。综合27种限制性态型(restrictionmorph),可得到6种mtDNA单倍型(hopotype)。伊犁鹅与另外10个鹅品种没有共享的单倍型,遗传距离和UPGMA聚类分析也表明,伊犁鹅与这些品种具有不同的起源。EcoRV、HaeⅡ、HincⅡ和KpnⅠ4种酶的限制性态型可作为鉴别两种起源家鹅的母系遗传标记。起源于鸿雁的10个鹅品种群体内出现一定的遗传差异,其群体多态度(π)、单倍型间平均遗传距离(P)、品种间平均净遗传距离(δnet)分别为0.025%、0.266%和0.029%。白羽鹅品种在形成过程中经历过创立者效应(foUndereffect)。这10个鹅品种可能起源于两个不同地理区的鸿雁类群。  相似文献

基于Cyt b基因全长序列,对我国10个地方黄牛品种的遗传多态性和系统发育关系进行了分析.中国黄牛Cyt b基因序列富含碱基A和T,A+T平均含量为56.7%,密码子第3位点核苷酸表现出较强的碱基组成偏倚,该位点上G的含量最低,为 3.5%,而A的含量最高,为44.6%,核苷酸组成偏差指数为0.45.在47个个体1 140 bp的序列中检测到30个变异位点,约占核苷酸总数的2.63%,密码子第3位点的突变率(76.7%)远高于第1位点(16.7%)和第2位点(6.6%),第2位点的非同义替代率(100%)远高于第1位点(40%)和第3位点(4.3%).定义了15种单倍型.将10个品种作为一个大的种群分析时,发现中国黄牛Cyt b基因具有较高的单倍型多样性(Hd=0.783±0046)和一定程度的核苷酸多样性(Pi=0.00828±0.00030),这与中国黄牛地理分布广、种群庞大和起源复杂有关系.基于Kimura 2 parameter距离采用UPGMA法构建的10个黄牛品种分子系统树中将47个个体分别与瘤牛和普通牛聚在一起,说明中国黄牛主要起源于瘤牛和普通牛.北方牛受普通牛的影响较大,南方牛受瘤牛的影响较大,但是对于亲缘关系复杂的中原地区黄牛而言,瘤牛和普通牛对它们的影响力大小因品种而异,因而它们分别和北方牛和南方牛之间的亲缘关系远近程度也存在差异.不同品种之间的亲缘关系差异可能与它们的地理分布差异具有一定的关系.  相似文献

目的:了解中国新疆吉木萨尔野马繁殖中心的普氏野马(Equus przewalskii)遗传多样性及其遗传背景.方法:采用PCR产物直接测序法,对15匹普氏野马线粒体DNA D-loop高变区进行测序分析.结果:测定15个个体的线粒体DNA D-loop高变区15464~15866片段序列402bp.检测到12种单倍型,包括37个多态位点,占全部序列的9.2%,其中转换位点24个、颠换位点20个、转换位点和颠换并存位点8个、缺失位点3个.A%+T%含量(56.1%)高于G%+C%含量(43.9%),平均A含量为28.4%,T含量为27.7%,C含量为29%,G含为14.9%.单倍型间平均遗传距离为0.030,单倍型多态性(h)为1±0.00116,核苷酸多态性(π)为2.90%.15匹普氏野马线粒体DNA D-loop高变区之间平均核苷酸变异率为2.48%.结论:研究表明我国新疆吉木萨尔野马繁殖中心的普氏野马线粒体DNA D-loop区序列存在丰富的多态性.  相似文献

Summary Mitochondrial (mt) DNA from a Southern Italian family with Leber hereditary optic neuropathy was analyzed for the presence of the reported mutation at position 11778 of the ND4 subunit gene. The point mutation was found in mt DNA extracted from peripheral blood in all members of the family with the exclusion of the father, and was present in a homoplasmic fashion, despite the phenotypic heterogeneity of disease presentation among family members.  相似文献

用24种限制性内切酶分析了我国21个具代表性地方猪品种、1个引进品种和2个来自中国和越南的野生近缘种mtDNA的RFLP。结果表明:在74个个体中检出的32种限制性态型可归结成7种单倍型,其间的差异主要来源于少数几个限制性位点的点突变;地方猪种4种单倍型间的平均遗传距离为0.143%,遗传多态程度(π值)仅为0.007%,说明遗传多样性非常贫乏,提示中国地方猪种可能起源于一个野猪亚种。  相似文献

Mitochondrial-DNA (mtDNA) restriction patterns were studied in 22 wolves (Canis lupus) sampled in central-northern Italy. A total of 60 restriction sites were detected, encompassing about 2 % of the mitochondrial genome of canids. All wolves showed the same restriction pattern. Therefore, a single mtDNA haplotype was detected in the Italian wolf population. Historical information on peninsular isolation and demographic decline suggest that low genetically effective population size and random drift may have strongly reduced the mtDNA variability of wolves in Italy over the last 100–200 years. A different mtDNA restriction pattern in feral dogs sampled from a wolf range in central Italy was detected. These findings suggest that the hybridization and introgression of female dog genomes into the Italian wolf population may be rare or absent.  相似文献

线粒体DNA遗传多样性用于评价6个中国地方猪种和3个瑞典家猪系统发育关系。采用PCR和序列分析方法得到了来自9个品种140头猪的线粒体中控制区440bp和细胞色素b基因798bp核苷酸序列。系统发育分析结果表明:6个中国地方猪种起源于亚洲野猪。中国地方猪种和欧洲野猪的线粒体DNA核苷酸序列变异发生在413000-875000年以前,而亚洲紧猪的变异仅发生在7000-156000上以前,由于2000年以前或18世纪初中国猪种导入欧洲家猪,因此瑞典家猪既属于欧洲类也属于亚洲类。  相似文献

In the 10 years since we published our first full analysis of mitochondrial DNA (mtDNA) variation in Rattus exulans as a means for tracking human migration in Polynesia, we have extended the commensal approach through time and space with the use of ancient DNA (aDNA) and by analysing samples from across the Pacific. Not only can mtDNA phylogenies provide information regarding population origins and paths of migration, they have also provided information regarding degrees of contact and interaction between islands. An important extension of the R. exulans project is the creation and on-going development of a genetic database for the identification of Rattus species based on mtDNA sequences. The phylogenetic analysis of sequences from 18 species and 1 subspecies of Rattus thus far have raised some questions regarding species identification and species distributions in the Pacific.  相似文献

To assess the genetic diversity of Japanese native horse populations, we examined seven such populations using mitochondrial DNA (mtDNA) and microsatellite analyses. Four reference populations of Mongolian horses and European breeds were employed as other equids. In the mtDNA analysis, the control region (D-loop) of 411 bp was sequenced, and 12 haplotypes with 33 variable sites were identified in the Japanese native horses. The phylogenetic tree constructed by haplogrouping and using worldwide geographic references indicated that the haplotypes of the Japanese native horses were derived from six equid clusters. Compared with the foreign populations, the Japanese native populations showed lower within-population diversity and higher between-population differentiation. Microsatellite analysis, using 27 markers, found an average number of alleles per locus of 9.6 in 318 native and foreign horses. In most native populations, the within-population diversity was lower than that observed in foreign populations. The genetic distance matrix based on allelic frequency indicated that several native populations had notably high between-population differentiation. The molecular coancestry-based genetic distance matrix revealed that the European populations were differentiated from the Japanese and Mongolian populations, and no clear groups could be identified among the Japanese native horse populations. The genetic distance matrices had few correlations with the geographic distribution of the Japanese native populations. Based on the results of both mtDNA and microsatellite analyses, it could be speculated that each native population was formed by the founder populations derived from Mongolian horses. The genetic construction of each population appears to have been derived from independent breeding in each local area since the time of population fission, and this was accompanied by drastic genetic drift in recent times. In spite of the common belief of Europe as reasonably homogeneous at genetic level, advances in high-throughput genotyping technology have resolved several gradients which define different geographical areas with good precision. When Northern and Southern European groups were considered separately, there were clear genetic distinctions. Intra-country genetic differences were also evident, especially in Finland and, to a lesser extent, within other European populations. Here, we present the first analysis using the 125,799 genome-wide Single Nucleotide Polymorphisms (SNPs) data of 1,014 Italians with wide geographical coverage. We showed by using Principal Component analysis and model-based individual ancestry analysis, that the current population of Sardinia can be clearly differentiated genetically from mainland Italy and Sicily, and that a certain degree of genetic differentiation is detectable within the current Italian peninsula population. Pair-wise FST statistics Northern and Southern Italy amounts approximately to 0.001 between, and around 0.002 between Northern Italy and Utah residents with Northern and Western European ancestry (CEU). The Italian population also revealed a fine genetic substructure underscoring by the genomic inflation (Sardinia vs. Northern Italy = 3.040 and Northern Italy vs. CEU = 1.427), warning against confounding effects of hidden relatedness and population substructure in association studies.  相似文献

The first hypervariable (HV1) region of mitochondrial DNA (mtDNA) of two popular Nigerian goat breeds: West African Dwarf (WAD) (n = 35) and Red Sokoto (RS) (n = 37) and one exotic breed: Kalahari Red (KR) (n = 38) imported from South Africa were sequenced to investigate sequence diversity, genetic structure, origin, and demographic history of the populations. A total of 68 polymorphic sites were found in 110 sequences that grouped into 68 haplotypes. Average haplotype and nucleotide diversities for all breeds were 0.982 ± 0.005 and 0.02350 ± 0.00213, respectively. Phylogenetic analysis revealed two mtDNA lineages (A and B). Lineage A was predominant and included all haplotypes from WAD and RS and 5 out of 11 haplotypes of KR goats. The remaining haplotypes (6 Amills M, Ramírez O, Tomàs A, et al. Mitochondrial DNA diversity and origins of South and Central American goats. Animal Genetics 2009; 40(3): 315322.[Crossref], [PubMed], [Web of Science ®] [Google Scholar]) of KR belong to lineage B. The analysis of molecular variance revealed a high-within breed genetic variance of 82.4% and a low-between breed genetic variance of 17.6%. The three breeds clustered with Capra aegagrus as their wild ancestor. Mismatch distribution analysis showed that WAD, RS and haplogroup A have experienced population expansion events. The study has revealed very high diversity within the three breeds which are not strongly separated from each other based on mtDNA analysis. The information obtained on the genetic structure of the breeds will be useful in planning improvement and conservation programs for the local populations.  相似文献
