评价了燕麦镰刀菌GD-2毒素对野燕麦的除草活性潜力。毒素对野燕麦种子萌发抑制试验表明,当毒素浓度达到5 mg/mL时,对野燕麦种子的萌发抑制效果达77.54%,在相同浓度下,对野燕麦种子胚根的抑制效果高于对胚芽的抑制效果。在温室条件下,毒素处理野燕麦植株后,各个不同处理野燕麦株高和地上部鲜重明显减少,然而,各处理间没有明显差异。用石油醚、氯仿、乙酸乙酯和正丁醇依次萃取燕麦镰刀菌发酵滤液并获得粗提物,其中正丁醇浸提物(5 mg/mL)在离体叶片实验中效果优于其他有机溶剂粗提物。此外,在毒素对10种植物的敏感性实验中,其中稗草和藜表现对燕麦镰刀菌毒素敏感。从燕麦镰刀菌中分离出3个除草活性化合物,且具有相同的紫外吸收峰。当前结果表明燕麦镰刀菌产生的毒素具有开发成为防除野燕麦生物源除草剂的潜力。  相似文献

以一粒小麦-葡萄牙野燕麦杂交后代一粒葡为实验材料,以地高辛标记的葡萄牙野燕麦基因组DNA为探针、一粒小麦基因组DNA为封阻对一粒葡及其衍生系根尖染色体进行基因组原位杂交(GISH)分析,探讨了影响一粒葡GISH效果的主要因素.建立并优化了一粒葡GISH分析的实验体系,即探针DNA与封阻DNA比例为1∶50时可有效分开双方染色体组.优化GISH分析显示,在一粒葡后代衍生系中均检测到燕麦染色质的存在,且不同选系间带有燕麦染色体的数目不同,进一步证明一粒葡是一粒小麦-葡萄牙野燕麦远缘杂交的后代.  相似文献

以目前大面积栽培的甘蓝型油菜(Brassica napus) ‘秦优10号’和其田间常见的8种杂草播娘蒿(Descurainia sophia)、泽漆(Euphorbia helioscopia)、野燕麦(Avena fatua)、棒头草(Polypogon fugax)、婆婆纳(Veronica didyma)、看麦娘(Alopecurus aequalis)、灰绿藜(Chenopodium glaucum)、反枝苋(Amaranthus retroflexus)为试验材料, 利用人工气候培养箱控温, 研究了10 ℃/5 ℃ (昼/夜)、18 ℃/10 ℃、25 ℃/20 ℃、35 ℃/30 ℃、40 ℃/35 ℃不同温度幅度处理对它们的发芽势、发芽率、幼苗形态及生理指标的影响, 旨在为预测气候变化环境下油菜田恶性靶标杂草的演替趋势提供参考依据。试验结果表明: 不同供试杂草对温度变化表现出明显的适应性差异, 看麦娘、灰绿藜和反枝苋的最适萌发温度偏高, 如反枝苋在高温处理(40 ℃/35 ℃)下种子的发芽率达到最高(91%); 而播娘蒿、泽漆、野燕麦、棒头草、婆婆纳种子的最适萌发温度主要集中于10-18 ℃范围内。进一步测试研究表明, 高温处理(40 ℃/35 ℃)下, 反枝苋幼苗生长旺盛, 体内丙二醛的积累也明显低于低温(10 ℃/5 ℃)处理, 可溶性糖和可溶性蛋白含量以及保护酶活性也较高, 表明反枝苋对高温胁迫有较好的适应性, 灰绿藜对高温的响应与反枝苋相似, 在高温气候年份, 要严防这2种草害加重; 而在低温处理(10 ℃/5 ℃)下, 野燕麦的种子萌发率达到100%, 其幼苗生长旺盛, 体内可溶性糖和可溶性蛋白含量及保护酶活性较高, 播娘蒿对低温胁迫的响应类似于野燕麦, 因此在寒冷年份需加强对野燕麦和播娘蒿草害的预防。供试油菜品种相对于所有供试杂草对温度变化的适应性更好, 其种子在不同温度处理下均保持较稳定的高发芽率, 且幼苗长势良好, 表明‘秦优10号’是一个对温度变化适应性很好的油菜品种。  相似文献

以带有与不带有Neotyphodium属内生真菌的醉马草Achnatherum inebrians、披碱草Elymus dahuricus和野大麦Hordeum brevisubulatum的草粉浸提液对细交链孢Alternaria alternata、根腐离蠕孢Bipolaris sorokiniana、燕麦镰孢Fusarium avenaceum和绿色木霉Trichoderma viride进行了抑菌活性研究。结果表明:披碱草、醉马草和野大麦草粉浸提液对细交链孢、根腐离蠕孢、燕麦镰孢和绿色木霉的菌落生长、孢子萌发率和芽管长度均有一定的抑制作用。而披碱草中的Neotyphodium可显著增强披碱草草粉浸提液对细交链孢、燕麦镰孢、绿色木霉菌落生长及对细交链孢和根腐离蠕孢孢子萌发及燕麦镰孢芽管长度的抑制作用;醉马草中的Neotyphodium显著增强了醉马草草粉浸提液对燕麦镰孢、绿色木霉菌落生长和芽管长度,以及细交链孢、根腐离蠕孢和燕麦镰孢孢子萌发的抑制作用;野大麦中的Neotyphodium显著增强了野大麦草粉浸提液对绿色木霉菌落生长、孢子萌发和芽管长度的抑制作用。  相似文献

采用室内培养皿生物测定的方法,测试不同质量浓度(0、0.02、0.03、0.05、0.08、0.10 g·mL-1)麦秸、稻秸浸提液对油菜及油菜田2种常见靶标杂草野燕麦(Avena fatua Linn)、播娘蒿 [Descurainia sophia(L.)Schur]种子萌发及幼苗生长的影响。结果表明,在供试质量浓度下,麦秸与稻秸浸提液对3种受体材料种苗生长的影响存在显著差异,二者对油菜种苗生长安全的最高质量浓度分别为0.08 g·mL-1和0.05 g·mL-1,可以用来开发研制油菜田肥料;在对油菜安全的质量浓度下,麦秸浸提液对野燕麦种苗生长无抑制作用;而稻秸浸提液对野燕麦种苗生长的抑制作用明显,可以用来开发研制油菜田防除野燕麦的生物抑草肥;在油菜的安全的质量浓度下,麦秸、稻秸浸提液对播娘蒿种苗生长均有很强的抑制作用,且麦秸的抑制作用强于稻秸,具有开发研制油菜田治理播娘蒿的生物抑草肥料的应用前景。  相似文献

通过普通细胞学和基因组原位杂交(GISH)研究提莫菲维小麦(Triticum timopheevi)与光稃野燕麦(Avena fatua L.var.glabrata Pat)远缘杂交后代花粉母细胞染色体减数分裂行为.结果表明,该R株系花粉母细胞的减数分裂指数为87.46,表现出一定的遗传不稳定性.GISH分析发现该F3株系中期I细胞中有4对红色杂交信号,且游离的单价体上各有1个杂交信号;后期Ⅰ、后期Ⅱ和末期Ⅱ出现落后染色体、染色体桥、微核等现象,分别占观察数的10.58%、1.92%和12.36%,这些异常现象可能是由于光稃野燕麦遗传物质干扰了小麦同源染色体或部分同源染色体的正常配对造成的.  相似文献

To enhance the limited information available about the palaeo-ecological significance of calcareous dinoflagellates, we have studied their lateral distribution in surface sediments of the equatorial and south Atlantic between 13 degrees N and 36 degrees S. Calcareous dinoflagellate cysts appear to be widely distributed throughout the studied area. In the surface sediments, concentrations (cyst per gram dry sediment) of the vegetative stage Thoracosphaera heimii are generally higher than that of the (presumably) calcareous resting cysts. Distribution patterns in surface sediments of Orthopithonella granifera (Fütterer) Keupp and Versteegh, Rhabdothorax spp. Kamptner., Sphaerodinella albatrosiana (Kamptner) Keupp and Versteegh S. albatrosiana praratabulated, Sphaerodinella tuberosa var. 1 (Kamptner) Keupp and Versteegh and S. tuberosa var. 2 and the ratios between these species have been compared with temperature, salinity, density and stratification gradients in the upper water column. Rhabdothorax spp. is characteristically present in sediments of more temperate regions characterized by high seasonality. Dinoflagellates producing these cysts are able to tolerate high nutrient concentrations, and mixing of the water column. S. albatrosiana is abundant in regions characterized by high sea surface temperatures and oligotrophic surface water conditions. In contrast, the distribution of S. tuberosa var. 2 is negatively related to temperature. The other cyst species did not show a characteristic pattern in relation to the studied environmental gradients.The ratio of Sphaerodinella tuberosa var. 2 to Orthopithonella granifera can be used for reconstructing the presence of stratification in the upper 50m of the water column, whereas the ratios of S. tuberosa var. 2 to Sphaerodinella albatrosiana and of O. granifera to Rhabdothorax spp. might be used for palaeotemperature reconstructions. Calcareous dinoflagellate cysts are abundant in oligotrophic areas and may be useful for the reconstruction of palaeoenvironmental conditions.  相似文献

Spatial and temporal variations of Cocconeis placentula var. euglypta in drift and periphyton were studied in mountain streams of the Córdoba Province (Argentina). The sampling program was conducted in study sites located on a confluence between different order streams during an annual cycle. Samples were also taken every two hours during the daylight period in high and low water conditions. The relationship between drift and cellular reproduction was evaluated by valve length biometrics analysis. C. placentula var. euglypta drift was continuous; its density was not always dependent on periphyton density in each locality. C. placentula var. euglypta drift could be related to abiotic factors such as temperature and flow during the annual cycle. There were significant differences between periphyton and drift valve lengths. Moreover, drift can be associated with cellular reproduction because density was higher when valve lengths were shorter at different hours of the day. C. placentula var. euglypta epiphytims on Cladophora glomerata also influenced drift density and size distribution, modifying the relationship between periphyton and drift during the late spring when C. placentula var. euglypta was detached from senescent mats.  相似文献

GroEL/S is a molecular chaperone system in Escherichia coli which not only assists the folding of intracellular proteins but also affects the cellular activity against the change of environmental condition. Here we show that the growth rate of E. coli DH5alpha can be improved at low temperature by expressing a GroEL/S variant achieved through irrational protein engineering approach. The GroELS variant (GroELS(var)) accelerating the growth of E. coli DH5alpha was screened through enrichment culture of the mutant libraries obtained by random mutagenesis. E. coli DH5alpha harboring the groELS(var) gene exhibited approximately 1.5-2 times higher growth rate compared to the strain with wild-type GroELS at 15-30 degrees C. At 10 degrees C, a temperature that the growth of E. coli DH5alpha almost stops, the GroELS(var) triggered the growth of E. coli DH5alpha. We identified that seven nucleotides of groELS gene and six amino acids of the GroELS were changed through the mutagenesis and screening. Site directed mutagenic analysis revealed that H360 in GroEL(var) is the most crucial residue determining the activity of GroELS(var) and more than one of the other residues in GroEL(var) may be additionally involved in the activity of GroELS(var). The improvement of growth rate induced by the GroELS(var) was observed only in the strain DH5alpha and not detected in other E. coli strains, such as BL21, BW25113, codon+, JM110, Top10, and XL1-blue.  相似文献

Pichia amethionina varieties have different sensitivity to the acidity of the medium: P. amethionina var. amethionina has the optimum pH 3 for agglutination and 5 for conjugation; P. amethionina var. pachycereana has the optimum pH 4-6 for agglutination and 6 for conjugation. The optimum temperature for the both organisms is 24 degrees C. Under the optimum conditions, P. amethionina var. amethionina has a 98% agglutination and a 28% conjugation; for P. amethionina var. pachycereana, these values are 83 and 55%, respectively. The agglutination of the two varieties does not depend on the presence of glucose. The conjugation of P. amethionina var. pachycereana does not occur in a medium without glucose while the conjugation of P. amethionina var. amethionina is reduced twofold under these conditions. Inositol, biotin and folic acid produce positive effect on the conjugation of P. amethionina var. pachycereana, but only folic acid favours the conjugation of P. amethionina var. amethionina. No differences have been found between the varieties by staining their cells and zygotes with FITC-ConA.  相似文献

The var gene family of Plasmodium falciparum encodes the immunodominant variant surface antigens PfEMP1. These highly polymorphic proteins are important virulence factors that mediate cytoadhesion to a variety of host tissues, causing sequestration of parasitized red blood cells in vital organs, including the brain or placenta. Acquisition of variant-specific antibodies correlates with protection against severe malarial infections; however, understanding the relationship between gene expression and infection outcome is complicated by the modular genetic architectures of var genes that encode varying numbers of antigenic domains with differential binding specificities. By analyzing the domain architectures of fully sequenced var gene repertoires we reveal a significant, non-random association between the number of domains comprising a var gene and their sequence conservation. As such, var genes can be grouped into those that are short and diverse and genes that are long and conserved, suggesting gene length as an important characteristic in the classification of var genes. We then use an evolutionary framework to demonstrate how the same evolutionary forces acting on the level of an individual gene may have also shaped the parasite's gene repertoire. The observed associations between sequence conservation, gene architecture and repertoire structure can thus be explained by a trade-off between optimizing within-host fitness and minimizing between-host immune selection pressure. Our results demonstrate how simple evolutionary mechanisms can explain var gene structuring on multiple levels and have important implications for understanding the multifaceted epidemiology of P. falciparum malaria.  相似文献

通过对科尔沁沙地东南缘樟子松人工林气温、地温及樟子松人工林根系分布的观测,从温度对外生菌根菌存活与生长影响的角度分析了沙地樟子松人工林衰退的现象,结果表明:由于沙地表层土壤(0~5cm)高温环境,外生菌根菌无法存活;在沙地20~40cm土层内,温度条件有利于外生菌根菌的存活、生长,而13~42年生沙地樟子松人工林树木平均78%的根系分布于该层,说明外生菌根菌的存在阻止或延缓了樟子松人工林的生长衰退,不是导致沙地樟子松人工林枯叶、枯梢、枯枝、死亡的原因.表层土壤外生菌根菌的缺乏使萌发的幼苗不能形成有效根系是导致沙地樟子松人工林不能天然更新的主要原因之一.  相似文献

选择青藏高原东南林线地区优势植物薄毛海绵杜鹃为对象,研究薄毛海绵杜鹃结实特性和萌发特性随海拔(4183—4673 m)、坡度、坡向等环境梯度的变化规律。对比了不同环境条件下薄毛海绵杜鹃果实的长、宽、重量、单果种子数、种子千粒重等结实特性;并测试了不同环境所产种子的萌发对温度和光照的响应。结果表明:(1)在高海拔低温胁迫环境下,薄毛海绵杜鹃的生殖投入加大,果实大且质量重,种子数量多但质量轻;(2)坡向是影响单果种子数量的主要环境因子,阴坡单果种子数显著大于阳坡,生活在阴坡的薄毛海绵杜鹃的繁衍能力更强;(3)较高和较低土壤湿度都会影响薄毛海绵杜鹃的种子质量,中等土壤湿度(28.3%—32.5%)薄毛海绵杜鹃种子萌发能力最强;(4)薄毛海绵杜鹃的种子是需光性种子,在黑暗条件下几乎不萌发;(5)在光照条件下,温度对种子萌发率和萌发速率有显著影响,高温(25/5℃)有助于提高萌发率并促使萌发进程显著提前。研究揭示了不同海拔薄毛海绵杜鹃结实和萌发特性与环境之间的关系,为薄毛海绵杜鹃的种质资源保护和气候变化背景下藏东南林线动态预测提供了基础资料。  相似文献

The button mushroom Agaricus bisporus commercially cultivated requires 16-19 °C during the fruiting period. Wild strains are also present in natural habitat, and in light of their wide range of geographic distribution reported, from boreal region to tropical region, questions on the development adaptation to temperature arose. Isolates from various geographic areas were screened for their ability to fruit at higher temperature (FHT ability) than commercial cultivars. The FHT trait discriminated at the varietal rank. Agaricus bisporus var. eurotetrasporus was unable to develop any sporophores whilst A. bisporus var. burnettii adapted perfectly to 25 °C for fruiting, suggesting that the FHT ability is a fixed trait in these varieties. In contrast, FHT ability of A. bisporus var. bisporus appeared variable and correlated neither with climate/microclimate nor with habitat. However, FHT ability taken as a whole appeared higher in North American populations than in European ones. Some A. bisporus var. bisporus isolates revealed a good potential for cultivation at 25 °C.  相似文献

An investigation of the role of the var1 protein in the assembly of the yeast mitochondrial ribosomes was carried out in a temperature conditional mutant, strain h56, which contains a mutation (tsv1) just upstream of the structural gene for the var1 protein. The mutation results in a marked decrease in the synthesis of the var1 protein at the permissive temperature of 28 degrees C and an apparently complete absence of var1 synthesis at the restrictive temperature of 36 degrees C. Long-term growth of strain h56 at the non-permissive temperature was found to result in the loss of the small (37 S) ribosomal subunit and the appearance of a novel 30 S ribonucleoparticle. Both the small (37 S) and the large (54 S) mitochondrial ribosomal subunits were found to be assembled in strain h56 for at least 3 h after transfer to the non-permissive temperature.  相似文献

Polyploidy is a major mechanism of adaptation and speciation in plants. Two varieties of Hedychium villosum, var. villosum and var. tenuiflorum, primarily differ in plant and flower size. Chromosome number suggests that var. tenuiflorum is diploid (2n = 34) and var. villosum is tetraploid (2n = 68). Although the flowers of the two varieties do not have any difference in floral shape, each can be easily distinguished morphologically because the shoots, leaves, and flowers of the tetraploid var. villosum are consistently larger than the diploid var. tenuiflorum. The two varieties each possess distinct geographic ranges and habitats, and no sympatric distribution has been found.The tetraploid var. villosum has a broader geographic distribution range and more diverse ecological habitats than the diploid var. tenuiflorum. The two varieties are also completely reproductively isolated due to the non-overlap of their flowering times. The two varieties should be recognized as two distinct species as they fulfill the requirements of various species concepts. Thus, we suggested that the tetraploid var. villosum should be kept as Hedychium villosum and the diploid var. tenuiflorum should be renewed to Hedychium tenuiflorum.  相似文献
