Magnetically suspended left ventricular assist devices have only one moving part, the impeller. The impeller has absolutely no contact with any of the fixed parts, thus greatly reducing the regions of stagnant or high shear stress that surround a mechanical or fluid bearing. Measurements of the mean flow patterns as well as viscous and turbulent stresses were made in a shaft-driven prototype of a magnetically suspended centrifugal blood pump at several constant flow rates (3-9 L/min) using particle image velocimetry (PIV). The chosen range of flow rates is representative of the range over which the pump may operate while implanted. Measurements on a three-dimensional measurement grid within several regions of the pump, including the inlet, blade passage, exit volute, and diffuser are reported. The measurements are used to identify regions of potential blood damage due to high shear stress and/or stagnation of the blood, both of which have been associated with blood damage within artificial heart valves and diaphragm-type pumps. Levels of turbulence intensity and Reynolds stresses that are comparable to those in artificial heart valves are reported. At the design flow rate (6 L/min), the flow is generally well behaved (no recirculation or stagnant flow) and stress levels are below levels that would be expected to contribute to hemolysis or thrombosis. The flow at both high (9 L/min) and low (3 L/min) flow rates introduces anomalies into the flow, such as recirculation, stagnation, and high stress regions. Levels of viscous and Reynolds shear stresses everywhere within the pump are below reported threshold values for damage to red cells over the entire range of flow rates investigated; however, at both high and low flow rate conditions, the flow field may promote activation of the clotting cascade due to regions of elevated shear stress adjacent to separated or stagnant flow.  相似文献

A computational fluid dynamics (CFD) model that simulates mechanical mixing for high-solids anaerobic digestion was developed. Numerical simulations of mixing manure slurry which exhibits non-Newtonian pseudo-plastic fluid behavior were performed for six designs: (i) one helical ribbon impeller; (ii) one anchor impeller; (iii) one curtain-type impeller; (iv) three counterflow (CF-2) impellers; (v) two modified high solidity (MHS 3/39°) impellers; and (vi) two pitched blade turbine impellers. The CFD model was validated against measurements for mixing a Herschel-Bulkley fluid by ribbon and anchor impellers. Based on mixing time with respect to mixing energy level, three impeller types (ribbon, CF-2, and MHS 3/39°) stand out when agitating highly viscous fluids, of these mixing with two MHS 3/39° impellers requires the lowest power input to homogenize the manure slurry. A comparison of digestion material demonstrates that the mixing energy varies with manure type and total solids concentration to obtain a given mixing time. Moreover, an in-depth discussion about the CFD strategy, the influences of flow regime and impeller type on mixing characteristics, and the intrinsic relation between mixing and flow field is included.  相似文献

A perfusion system was developed to generate well defined flow conditions within a well of a standard multidish. Human vein endothelial cells were cultured under flow conditions and cell response was analyzed by microscopy. Endothelial cells became elongated and spindle shaped. As demonstrated by computational fluid dynamics (CFD), cells were cultured under well defined but time varying shear stress conditions. A damper system was introduced which reduced pulsatile flow when using volumetric pumps. The flow and the wall shear stress distribution were analyzed by CFD for the steady and unsteady flow field. Usage of the volumetric pump caused variations of the wall shear stresses despite the controlled fluid environment and introduction of a damper system. Therefore the use of CFD analysis and experimental validation is critical in developing flow chambers and studying cell response to shear stress. The system presented gives an effortless flow chamber setup within a 6-well standard multidish.  相似文献

使用CFD模拟软件Fluent对实验室用7L通气搅拌发酵罐内不同桨叶组合的搅拌效果进行气液两相流模拟.首先对发酵罐自带桨叶组合进行模拟,针对模拟结果提出两种改进桨型组合,对比分析3种组合的速度矢量图、速度分布柱形图和速度云图,以此优化出一种搅拌效果较好的桨型组合.结果表明:发酵罐内的流场可受通气的影响而发生改变,通气发酵过程的模拟采用多相流模型更为准确;底层桨离底距离的不同可导致发酵罐内流场的不同,以六直叶圆盘涡轮桨为底桨时,离底距离应不小于T/3;液相死区的大小是由桨叶组合、桨叶安装位置以及气液两相的相互作用共同导致的,不能单纯靠提高转速改善;在800 r/min下,搅拌效果组合3为最佳.  相似文献

In the experimental and clinical support of the failing heart, the impeller-type centrifugal pumps continue to be of interest because of their inherent advantages; however, the blood compatibility of these pumps still remains to be improved. From the viewpoint of haemodynamics, thrombosis and haemolysis could be reduced by eliminating the stagnation and turbulence of blood flow within the pump, which frequently takes place near the blood contracting surfaces of the pump, when the impeller contours do not coincide with the stream surfaces of the blood. It is suggested that it could be advantageous to design impeller contours according to the stream surfaces, by solving the partial differential equations of continuity, motion and energy. An impeller shroud and vane based on this approach would be fully rinsed by non-turbulent flow and there would then be neither stagnation nor turbulence within the pump, with the result that thrombosis and haemolysis could be reduced. A new impeller pump, developed according to this method, was evaluated as a left ventricular device in four dogs. The bypass flow was controlled at 40–50% of the total flow, each test lasting 6 h. All of the haematological parameters, measured every 2 h, remained within normal range. There was no thrombosis, and coagulation in the pump was avoided by a small dose of heparin to maintain the activated coagulation time (ACT) under 200″ in the experiments.  相似文献

A pulsatile fully implantable impeller pump with low haemolysis has been produced by developing a pulsatile impeller for a nonpulsatile pump also developed in this laboratory. The impeller was designed according to the 3-dimensional theory of fluid dynamics. The impeller shroud retains the same parabolic form and the vane has a form compacted by a radial logarithmic spiral and an axial helical spiral so that the absolute vibration velocity of the blood in a peripheral direction is a minimum as the impellar changes its speed periodically to generate a physiological pulsatile blood flow. Thus the Reynolds shear and the Newton shear are a minimum for the required pulse pressure. The mean volume and mean pressure are controlled by adjusting the voltage. The shape of the pressure pulse is determined by a square was of vollage and the systole/diastole ratio. In order to abolish regurgitation of the pump, a 40 per cent systole period and a 5 V voltage pulse are desirable for 40 mmHg pulse pressure (80–120 mmHg mean pressure). The pulse frequency has almost no effect on pump output. The pump can delivery 4 l/min mean volume and 100 mmHg mean pressure (40 mmHg pulse pressure), and these conditions result in an index of heamolysis (IH) for porcine blood of 0.020—only slighly more than the nonpulsatile pump (0.016). When the pulsatile impeller was used under nonpulsatile conditions its IH was almost doubled, but when the nonpulsatile impeller was used under pulsatile conditions the IH reached 0.13. The power consumption is approximately equal to that for the nonpulsatile pump: 3 W for 4 l/min and 100 mmHg output. The pump weights 240 g (including 190 g for the motor) and, because a commercial motor has been used, costs only a few hundred US dollars.  相似文献

Aortic dissecting aneurysm is one of the most catastrophic cardiovascular emergencies that carries high mortality. It was pointed out from clinical observations that the aneurysm development is likely to be related to the hemodynamics condition of the dissected aorta. In order to gain more insight on the formation and progression of dissecting aneurysm, hemodynamic parameters including flow pattern, velocity distribution, aortic wall pressure and shear stress, which are difficult to measure in vivo, are evaluated using numerical simulations. Pulsatile blood flow in patient-specific dissecting aneurismal aortas before and after the formation of lumenal aneurysm (pre-aneurysm and post-aneurysm) is investigated by computational fluid dynamics (CFD) simulations. Realistic time-dependent boundary conditions are prescribed at various arteries of the complete aorta models. This study suggests the helical development of false lumen around true lumen may be related to the helical nature of hemodynamic flow in aorta. Narrowing of the aorta is responsible for the massive recirculation in the poststenosis region in the lumenal aneurysm development. High pressure difference of 0.21 kPa between true and false lumens in the pre-aneurismal aorta infers the possible lumenal aneurysm site in the descending aorta. It is also found that relatively high time-averaged wall shear stress (in the range of 4-8 kPa) may be associated with tear initiation and propagation. CFD modeling assists in medical planning by providing blood flow patterns, wall pressure and wall shear stress. This helps to understand various phenomena in the development of dissecting aneurysm.  相似文献

We investigated the flow pattern and mixing behavior of a poly(γ‐glutamic acid) (γ‐PGA) solution in a bioreactor equipped with two Rushton turbines by simulation and experiment. Computational fluid dynamics (CFD) is used to solve the three‐dimensional hydrodynamics in the bioreactor and to obtain the flow patterns and tracer concentration at every point. The flow circulation patterns by inter‐impeller clearance and viscosity and their effects on overall mixing time were studied. Based on the results we can conclude that the impeller clearance should not be larger than 0.2 D for the efficient mixing under non‐aerated condition when the liquid viscosity is above 20 cp, which corresponds to concentrations of 20 g/L or above for γ‐PGA.  相似文献

A variety of methods by which mechanical circulatory support (MCS) can be provided have been described. However, the haemodynamic benefits of the different methods have not been adequately quantified. The aim of this paper is to compare the haemodynamic effects of six forms of MCS by numerical simulation. Three types of ventricular assist device (VAD) are studied: positive displacement; impeller and a novel reciprocating-valve design. Similarly, three pumping modes are modelled: constant flow; counterpulsation and copulsation. The cardiovascular system is modelled using an approach developed previously, using the concentrated parameter method by considering flow resistance, vessel elasticity and inertial effects of blood in individual conduit segments. The dynamic modelling of displacement and impeller pumps is represented by VAD inlet/outlet flow-rate changes. The dynamics of the reciprocating-valve pump is modelled with a specified displacement profile. Results show that in each simulation, the physiological variables of mean arterial pressure and systemic flow are adequately maintained. Modulation of the impeller pump flow profile produces a small (5 mmHg) oscillatory component to arterial pressure, whereas the displacement and reciprocating-valve pumps generate substantial arterial pressure and flow pulsatility. The impeller pump requires the least power input, the reciprocating valve pump slightly more, and the displacement pump the most. The in parallel configuration of the impeller and displacement pump designs with respect to the left ventricle provides near complete unloading and can cause the aortic valve to remain closed throughout the entire cardiac cycle with the attendant risk of aortic valve leaflet fusion following prolonged support. The in series configuration of the reciprocating-valve pump avoids this shortcoming but activation must be carefully synchronized to the cardiac cycle to allow adequate coronary perfusion. The reciprocating-valve pump is associated with haemodynamic advantages and a favourable power consumption.  相似文献

The local geometry of a bifurcation has been hypothesized to be a potential geometrical risk factor for the development of atherosclerosis. While flow division and branch area ratios clearly affect the flow field, the importance of the flow divider shape is not as clear. A fast spectral element computational fluid mechanics (CFD) solver was used to simulate flow through 90 ° T-bifurcations with three different flow divider shapes. Other factors, such as flow partition, area ratio, and bifurcation angle, were kept constant. A Reynolds number range of 15 to 350 was studied to bracket experimental results in the literature. The variation in the sharpness of the corners had a dramatic effect on both the flow field and wall shear stress distribution in the side branch, but little effect on the flow in the main tube. The magnitude of reverse velocities and wall shear stress in the side branch increased linearly over a physiological range of Reynolds number and corner shape. This paper verifies the accuracy and usefulness of spectral element CFD in studying three-dimensional hemodynamics.  相似文献

The concept of "design space" plays an integral part in implementation of quality by design for pharmaceutical products. ICH Q8 defines design space as "the multidimensional combination and interaction of input variables (e.g., material attributes) and process parameters that have been demonstrated to provide assurance of quality. Working within the design space is not considered as a change. Movement out of the design space is considered to be a change and would normally initiate a regulatory post-approval change process. Design space is proposed by the applicant and is subject to regulatory assessment and approval." Computational fluid dynamics (CFD) is increasingly being used as a tool for modeling of hydrodynamics and mass transfer. In this study, a laboratory-scale aerated bioreactor is modeled using CFD. Eulerian-Eulerian multiphase model is used along with dispersed k-ε turbulent model. Population balance model is incorporated to account for bubble breakage and coalescence. Multiple reference frame model is used for the rotating region. We demonstrate the usefulness of CFD modeling for evaluating the effects of typical process parameters like impeller speed, gas flow rate, and liquid height on the mass transfer coefficient (k(L)a). Design of experiments is utilized to establish a design space for the above mentioned parameters for a given permissible range of k(L)a.  相似文献

The dynamic environment within a bioreactor and in the purification equipment is known to affect the activity and yield of enzyme production. The present research focuses on the effect of hydrodynamic flow parameters (average energy dissipation rate, maximum energy dissipation rate, average shear rate, and average normal stress) and the interfacial flow parameters (specific interfacial area and mass transfer coefficient) on the activity of lysozyme. Flow parameters were estimated using CFD simulation based on the k-epsilon approach. Enzyme deactivation was investigated in 0.1, 0.3, 0.57, and 1 m i.d. vessels. Enzyme solution was subjected to hydrodynamic stress using various types of impellers and impeller combinations over a wide range of power consumption (0.03 < P(G)/V < 7, kW/m3). The effects of tank diameter, impeller diameter, blade width, blade angle, and the number of blades on the extent of deactivation were investigated. At equal value of P(G)/V, epsilon(max), and gamma(avg), the extent of deactivation was dramatically different for different impeller types. The extent of deactivation was found to correlate well with the average turbulent normal stress and the mass transfer coefficient.  相似文献

Measurements of the time-varying flow in a centrifugal blood pump operating as a left ventricular assist device (LVAD) are presented. This includes changes in both the pump flow rate as a function of the left ventricle contraction and the interaction of the rotating impeller and fixed exit volute. When operating with a pulsing ventricle, the flow rate through the LVAD varies from 0-11 L/min during each cycle of the heartbeat. Phase-averaged measurements of mean velocity and some turbulence statistics within several regions of the pump, including the inlet, blade passage, exit volute, and diffuser, are reported at 20 phases of the cardiac cycle. The transient flow fields are compared to the constant flow rate condition that was reported previously in order to investigate the transient effects within the pump. It is shown that the quasi-steady assumption is a fair treatment of the time varying flow field in all regions of this representative pump, which greatly simplifies the comprehension and modeling of this flow field. The measurements are further interpreted to identify the effects that the transient nature of the flow field will have on blood damage. Although regions of recirculation and stagnant flow exist at some phases of the cardiac cycle, there is no location where flow is stagnant during the entire heartbeat.  相似文献

The structure and function of the renal artery ostium flow diverter on the caudal side of the renal branch point were previously reported; in this study, we further evaluate the diverter?s possible functions. The protrusion of this structure into the abdominal aorta suggests that the diverter may preferentially direct blood flow to the renal arteries, and that it may also influence flow patterns and recirculation known to be involved in atherogenesis. Three-dimensional computational fluid dynamics (CFD) simulations of steady and pulsatile blood flow are performed to investigate the influence of diverter size and position, and vascular geometry, on the flow patterns and fluid mechanical forces in the neighborhood of the diverter. CFD results show that the flow diverter does affect the blood distribution; depending on the diverter?s position, the flow to the renal arteries may be increased or reduced. Calculated results also demonstrate the diverter?s effect on the wall shear stress (WSS) distribution, and suggest that the diverter contributes to an atherogenic environment in the abdominal aorta, while being atheroprotective in the renal arteries themselves. These results support previous clinical findings, and suggest directions for further clinical study. The results of this work have direct implications in understanding the physiological significance of the diverter, and its potential role in the pathophysiological development of atherosclerosis.  相似文献

Introduction  

The success of vascular stents in the restoration of blood flow is limited by restenosis. Recent data generated from computational fluid dynamics (CFD) models suggest that the vascular geometry created by an implanted stent causes local alterations in wall shear stress (WSS) that are associated with neointimal hyperplasia (NH). Foreshortening is a potential limitation of stent design that may affect stent performance and the rate of restenosis. The angle created between axially aligned stent struts and the principal direction of blood flow varies with the degree to which the stent foreshortens after implantation.  相似文献

An implantable artificial kidney using a hemofilter constructed from an array of silicon membranes to provide ultrafiltration requires a suitable blood flow path to ensure stable operation in vivo. Two types of flow paths distributing blood to the array of membranes were evaluated: parallel and serpentine. Computational fluid dynamics (CFD) simulations were used to guide the development of the blood flow paths. Pressure data from animal tests were used to obtain pulsatile flow conditions imposed in the transient simulations. A key consideration for stable operation in vivo is limiting platelet stress accumulation to avoid platelet activation and thrombus formation. Platelet stress exposure was evaluated by CFD particle tracking methods through the devices to provide distributions of platelet stress accumulation. The distributions of stress accumulation over the duration of a platelet lifetime for each device revealed that stress accumulation for the serpentine flow path exceeded levels expected to cause platelet activation while the accumulated stress for the parallel flow path was below expected activation levels.  相似文献

Haemodynamic factors, in particular wall shear stresses (WSSs) may have significant impact on growth and rupture of cerebral aneurysms. Without a means to measure WSS reliably in vivo, computational fluid dynamic (CFD) simulations are frequently employed to visualise and quantify blood flow from patient-specific computational models. With increasing interest in integrating these CFD simulations into pretreatment planning, a better understanding of the validity of the calculations in respect to computation parameters such as volume element type, mesh size and mesh composition is needed. In this study, CFD results for the two most common aneurysm types (saccular and terminal) are compared for polyhedral- vs. tetrahedral-based meshes and discussed regarding future clinical applications. For this purpose, a set of models were constructed for each aneurysm with spatially varying surface and volume mesh configurations (mesh size range: 5119-258, 481 volume elements). WSS distribution on the model wall and point-based velocity measurements were compared for each configuration model. Our results indicate a benefit of polyhedral meshes in respect to convergence speed and more homogeneous WSS patterns. Computational variations of WSS values and blood velocities are between 0.84 and 6.3% from the most simple mesh (tetrahedral elements only) and the most advanced mesh design investigated (polyhedral mesh with boundary layer).  相似文献

This paper presents Computational fluid dynamic (CFD) analysis of blood flow in three different 3-D models of left coronary artery (LCA). A comparative study of flow parameters (pressure distribution, velocity distribution and wall shear stress) in each of the models is done for a non-Newtonian (Carreau) as well as the Newtonian nature of blood viscosity over a complete cardiac cycle. The difference between these two types of behavior of blood is studied for both transient and steady states of flow. Additionally, flow parameters are compared for steady and transient boundary conditions considering blood as non-Newtonian fluid. The study shows that the highest wall shear stress (WSS), velocity and pressure are found in artery having stenosis in all the three branches of LCA. The use of Newtonian blood model is a good approximation for steady as well as transient blood flow boundary conditions if shear rate is above 100 s-1. However, the assumption of steady blood flow results in underestimating the values of flow parameters such as wall shear stress, pressure and velocity.  相似文献

Computational fluid dynamics (CFD) models to quantify momentum and mass transport under conditions of tissue growth will aid bioreactor design for development of tissue-engineered cartilage constructs. Fluent CFD models are used to calculate flow fields, shear stresses, and oxygen profiles around nonporous constructs simulating cartilage development in our concentric cylinder bioreactor. The shear stress distribution ranges from 1.5 to 12 dyn/cm(2) across the construct surfaces exposed to fluid flow and varies little with the relative number or placement of constructs in the bioreactor. Approximately 80% of the construct surface exposed to flow experiences shear stresses between 1.5 and 4 dyn/cm(2), validating the assumption that the concentric cylinder bioreactor provides a relatively homogeneous hydrodynamic environment for construct growth. Species mass transport modeling for oxygen demonstrates that fluid-phase oxygen transport to constructs is uniform. Some O(2) depletion near the down stream edge of constructs is noted with minimum pO(2) values near the constructs of 35 mmHg (23% O(2) saturation). These values are above oxygen concentrations in cartilage in vivo, suggesting that bioreactor oxygen concentrations likely do not affect chondrocyte growth. Scale-up studies demonstrate the utility and flexibility of CFD models to design and characterize bioreactors for growth of tissue-engineered cartilage.  相似文献
