All entomopathogenic fungi infect insects by direct penetration through the cuticle rather than per os through the gut. Genetic transformation can confer fungi with per os virulence. However, unless the recipient isolate is nonpathogenic to the target insect, mortality caused by a transgenic isolate cannot be attributed solely to oral virulence due to the potential for some simultaneous cuticular infection. Here, a Metarhizium anisopliae wild-type isolate (MaWT) nonpathogenic to Spodoptera litura was genetically engineered to provide a transformed isolate (MaVipT31) expressing the insect midgut-specific toxin Vip3Aa1. Toxin expression was confirmed in MaVipT31 hyphae and conidia using Western blotting. Mortality, leaf consumption and body weight of S. litura larvae (instars I–IV) exposed to a range of concentrations of MaWT conidia were not significantly different to controls although the number of conidia ingested by surviving larvae during the bioassay ranged from 2.3 × 105 (instar I) to 8.1 × 106 (instar IV). In contrast, consumption of MaVipT31 conidia caused high mortalities, reduced leaf consumption rates and decreased body weights in all instars evaluated, demonstrating that oral virulence had been acquired by MaVipT31. Larval mortalities were much more dependent on the number of MaVipT31 conidia ingested than the duration of time spent feeding on conidia-treated leaves (r2: 0.83–0.94 for instars I–IV). LC50 and LT50 trends for MaVipT31 estimated by time-concentration-mortality modeling analyses differed greatly amongst the instars. For 50% kill to be achieved, instar I larvae required 3, 4 and 5 days feeding on the leaves bearing 103, 28 and 8 conidia/mm2 respectively; instar IV larvae required 6, 7 and 8 days feeding on leaves bearing 1760, 730 and 410 conidia/mm2 respectively. Our results provide a deeper insight into the high oral virulence acquired by an engineered isolate and highlight its great potential for biological control.  相似文献

Tenebrio molitor L. (Coleoptera: Tenebrionidae), is an international and serious pest of stored products. So far nothing is known about the activity for each growth stage digestive enzyme regarding this insect species. Thus, the aim of the current study was to get in depth analysis of the stage specific digestion and to investigate the effect of cereal (wheat cultivars including MV17, Aflak, Sivand, Saymon, and Zare) and legume (bean) seed extracts on the two main digestive enzymes i.e. α-amylases and proteases. Therefore, gut enzymes were extracted using distilled water and wheat cultivars and bean seed proteinaceous compounds were extracted using 0.1 M NaCl. Results showed that a steady state increase in the number and amount of digestive enzyme activities from first to fourth instar larvae was seen in both enzyme and in gel assays. In the first instar larvae (L1) only one band of α-amylase activity was seen (A1), whereas in the second (L2), third (L3), fourth (L4) and fifth (L5) instar larvae as well as in the adult (A) more than one amylase band (up to 4 isoenzymes) was seen. The same pattern was observed for α and β glucosidases and proteases. Probit analysis showed that bean and MV17 inhibited the amylase activity with an I50 of 9.73 and 7.4 μg, respectively. The same cultivar seed extract inhibited protease activity with I50s of 11.54 and 6.5 μg proteins. It is concluded that proteinaceous extract of cereals and bean seeds have a strong potential to be used in this pest management.  相似文献

The carob moth, Ectomyelois ceratoniae (Lepidoptera: Pyralidae), is the most important pest of pomegranate orchards (in terms of economic damage) within Iran, and hence, several control procedures, including biological methods of control, have been attempted as a means of controlling populations of this insect. This research was carried out in order to study the biology of Apanteles myeloenta (Hymenoptera: Braconidae), a larval parasitoid of the carob moth. Laboratory studies were conducted to determine larval developmental time, adult longevity, sex ratio, parasite progeny production, and host stage preference of A. myeloenta. At 25 ± 1 °C, immature developmental time (egg to pupa; mean ± SE) was 28.33 ± 0.85 days and 27.46 ± 0.37 days for male and females, respectively. Adult females survived on average 17.5 ± 0.14, 11.7 ± 0.22, 3.4 ± 0.18, and 2.8 ± 0.12 days at 25 C when provided with honey and water, honey only, water only or no food source, respectively. The sex ratio (females to males) of A. myeloenta was 1:3.5 from hosts parasitized in the first instar, 1:3 for second instars and 1:2 for third instar carob moth larvae. Female A. myeloenta typically preferred to parasitize second instar over third or first instar. The oviposition activity peaked on the 7th and 8th days following emergence, when provided with honey, and 10% sucrose solution, respectively.  相似文献

Bradysia odoriphaga and B. difformis (Diptera: Sciaridae) are devastating pests of vegetables, ornamentals and edible mushrooms. In Chinese chive fields, the two Bradysia species occur with similar regularities: outbreaks in spring and autumn, and population decreases in summer. Temperature may be an important factor restricting their population abundance in summer. Here, we performed a life-table study under constant high temperatures and assessed the tolerance of two Bradysia species to heat shock. Life parameters of the Bradysia species indicated slow developmental rates, and low survival rates and fecundity, when the temperature was higher than 30 °C. At 34 °C, individuals were unable to reach the adult stages from eggs. Moreover, temperatures above 36 °C showed lethal effects, decreasing their survival rates. The median lethal time (LT50) values of 4th instar B. odoriphaga and B. difformis larvae were 46.82 and 32.97 h, respectively, while the values at 38 °C were 2.12 and 1.51 h, respectively. The 4th instar larvae and pupae possessed higher thermotolerance levels than adults and eggs, indicating sensitivities to heat stress. Moreover, B. odoriphaga was more thermotolerant than B. difformis. Thus, weak thermotolerance levels may restrict their occurrences during the period of summer heat, and the difference in thermotolerance levels between the two species may be related to their regional distributions.  相似文献

This study was performed to investigate the repellent effect of 5 μl doses of ten essential oils (bergamot, chamomile, clary sage, fennel, lavender, lemongrass, majoram, peanut, pennyroyal, and peppermint) against Lycorma delicatula 4th nymphs using an olfactometer. Only lavender oil exhibited significant repellency. We then tested 10, 5, 2.5, and 1 μl doses of lavender oil against the nymphs and females of L. delicatula. The oil showed significant repellency at 10 and 5 μl, although the latter is less potent to 1st instar nymphs. At the lavender oil dose of 2.5 μl, only 3rd and 4th instar nymphs and females were significantly affected. None of the stages tested were affected by 1 μl. Chromatographic and mass spectrometric analyses of lavender oil detected linalool (42.2%), linalyl acetate (49.4%), terpinen-4-ol (5.0%), and caryophyllene oxide (3.4%). Among the four main components, only linalool showed repellency to all instar nymphs and females. No synergism was detected. Antennae of all instar nymphs and females showed electrophysiological responses only to linalool. In field studies using linalool, 4th nymphs and adults were highly repelled at a dose of 30 μl of lavender oil. The effect differed according to test plot and treatment dose.  相似文献

The seven age-classes of Trichoplusia ni (Hübner) larvae evaluated in this study as hosts of Campoletis sonorensis indicates that early 2nd larval instar (3–5 day-old larvae) of T. ni represents the most suitable host stage for the development of the larval endoparasitoid C. sonorensis. The higher suitability of early 2nd larval instar of T. ni resulted in more parasitised larvae, a higher rate of successful parasitoid emergence, a higher rate of female progeny, and a lower rate of immature parasitoid mortality. The fitness gain of C. sonorensis on late 1st larval instar (2 day-old larvae) and late 2nd larval instar –early 3rd instars (6–8 day-old larvae) stages of T. ni is negatively affected by the trade-offs between the different physiological and behavioral characteristics influencing their suitability as hosts of C. sonorensis.  相似文献

Effect of sublethal concentration of B. thuringiensis on the first, third, fourth and fifth instar larvae of the American bollworm, H. armigera was investigated to study their response to food consumption, digestion, utilization, and their development till adult formation. The young larvae surviving B. thuringiensis treatment in their first instar and third instar delayed larval period by two to three days, but did not consume more food as compared to control. However, they showed higher digestibility of food as compared to control, which was compensated by their reduced ability to utilize the digested food for body substance. Contrary to the effect on first and third instar larvae, the fifth instar larvae surviving B. thuringiensis treatment in its fourth instar consumed less food, showed less absorption efficiency in digesting food, but compensated by increase in the utilization of ingested and digested food into body substance. Insects surviving B. thuringiensis HD-1 sublethal toxicity adapted to normal larval growth when fed on untreated food, depending upon insect growth prior to treatment. The moths emerging from B.thuringiensis treated larvae had sex ratio favouring females, and adult pairs laid less fertile eggs than those from the untreated ones. The response of B. thuringiensis treated larvae to their food and developmental needs is discussed.  相似文献

The predatory multicolored Asian lady beetle, Harmonia axyridis, was attracted to volatiles released from Chinese cabbage infested by the green peach aphid, Myzus persicae, in T-tube olfactometer choice tests. However, lady beetle adults and larvae did not respond to clean air, Chinese cabbage alone or green peach aphid alone. Of different prey densities, H. axyridis adults were most attracted to Chinese cabbage infested by 60 M. persicae adults after 24 h. However, H. axyridis larvae were not attracted to Chinese cabbage infested by M. persicae. Mechanically damaged Chinese cabbage attracted neither lady beetle adults nor larvae. Predatory adults were attracted to 60 M. persicae adults after 24 and 48 h, and to 90 M. persicae adults after 12 h, suggesting that the predatory response depends on the prey density. Lady adult beetles did not prefer the volatiles induced by Diamondback moth, Plutella xylostella, indicating that specific host insect specificity attracts respective natural enemies. It can be explained that the volatile compounds emitted from the host plant as a result of herbivore attack preferred by the specific insect species.  相似文献

Food utilization was measured in female fifth instar larvae and adults of the migratory locust by following the weight of food ingested, the weight of faeces produced, and the increase in body weight. These parameters were measured in normally developing locusts, in locusts that had been implanted with a pair of active corpora allata (CA) in the beginning of the fifth instar period, and in allatectomized locusts, operated on the first day after adult ecdysis.A high titre of corpus allatum hormone results in a considerably higher water content of the insects; allatectomy reduces this content. The dry weight of the locusts is not essentially influenced by excess or absence of juvenile hormone.Food consumption in corpora-allata-implanted locusts does not differ from that in normally developing insects. Within each developmental period the digestive capacity remains constant, but the absolute value of this capacity may differ between the two developmental periods. The approximate digestibility is somewhat higher after CA-implantation and lower after allatectomy. The efficiency of conversion of digested food into body substance is greater in normally developing larvae than in adults. CA-implantation lowers this efficiency in developing larvae. Allatectomy slightly raises the efficiency of conversion in adult locusts.In the second half period of larval development, CA-implantation raises the respiratory rate, as estimated by measuring oxygen consumption. During adult development no significant influence of CA-implantation on respiration was established. Relations between the amount of food digested, the efficiency of conversion of digested food into body substance, and the respiratory activity are discussed.  相似文献

The solitary parasitoid Microplitis tuberculifer (Wesmael) is an important biological control agent of various lepidopteran pests in Asia. We examined the preference of M. tuberculifer for different instars of its common host, Mythimna separata (Walker), host instar effects on parasitoid development, and the consequences of parasitism in different stages for growth and consumption of host larvae. The wasp successfully parasitized the first four larval instars of M. separata, but not the fifth, which appeared to be behaviorally resistant. First and second instars were parasitized at higher rates compared to thirds and fourths in no-choice situations, ostensibly due to longer handling times for the latter, but second instars were most preferred in a choice test that presented all stages simultaneously. Although later instar hosts yielded heavier cocoons, the fastest parasitoid development was obtained in second instars. Lower sex ratios were obtained from first instars as females appeared to lay a smaller proportion of fertilized eggs in small hosts. Both weight gain and food consumption of parasitized larvae were reduced significantly within 24 h of parasitism, regardless of the stage parasitized, and final body weights were less than 10% those of unparasitized larvae. Thus, M. tuberculifer has good potential as a biological control agent of M. separata, successfully parasitizing the first four larval instars and dramatically reducing plant consumption by the host in all cases.  相似文献

Food and feeding habits of mud crab Scylla olivacea (Herbst, 1896) in Pichavaram mangroves was investigated quantitatively and qualitatively for a period of two years from June 2010 to May 2012. Gut contents from 1737 specimens comprising 843 males and 894 females in the size range between 45 mm and 148 mm were examined. Crustaceans form the predominant food item in a majority of size groups in terms of percentage wet weight and frequency of occurrence, while molluscs showed a preference in few size groups. The other dietary items includes fishes, detritus, mud and sand and miscellaneous. Gut content analysis revealed no significant variation between the quantities of food consumed by both sexes. Feeding intensity was higher in juveniles and subadults of both sexes than that of adults, revealing a greater preference to feed on fast moving prey such as crustaceans and fishes. The results of the present study indicate that S. olivacea in Pichavaram mangroves exhibited a clear preference for crustaceans.  相似文献

Entomotoxic plant lectins have been extensively studied in the past two decades, yet the exact mechanisms underlying their toxic effects remain unknown. This study investigated the effects of Dioclea violacea lectin (DVL) on larval development in Anagasta kuehniella. Chronic exposure of larvae (from neonates to the fourth instar) demonstrated that DVL interfered with larval growth, retarding development and decreasing larval mass without affecting survival. DVL decreased trypsin-like, chymotrypsin-like, and α-amylase activities and proved resistant to proteolysis by midgut proteases up to 24 h. Shorter exposures to dietary DVL had no effect on midgut enzyme activity. Feeding fourth-instar larvae with fluorescently-labeled DVL revealed lectin binding to the peritrophic membrane.  相似文献

Cicadulina bipunctata was originally distributed in tropical and subtropical regions of the Old World. This leafhopper recently expanded its distribution area to southern parts of temperate Japan. In this study, factors affecting the overwintering ability of C. bipunctata were examined. A series of laboratory experiments revealed that cold acclimation at 15 °C for 7 days enhanced the cold tolerance of C. bipunctata to the same level as an overwintering population, adult females were more tolerant of cold temperature than adult males, and survival of acclimated adult females was highly dependent on temperature from −5 to 5 °C and exposure duration to the temperature. The temperature of crystallization of adult females was approximately −19 °C but temperatures in southern temperate Japan rarely dropped below −10 °C in the winter, indicating that overwintering C. bipunctata adults in temperate Japan are not killed by freezing injury but by indirect chilling injury caused by long-term exposure to moderately low temperatures. An overwintering generation of C. bipunctata had extremely low overwinter survival (<1%) in temperate Japan; however, based on winter temperature ranges, there are additional areas amenable to expansion of C. bipunctata in temperate Japan.  相似文献

The effect of electron beam irradiation on each developmental stage of diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae), was examined. Eggs, larvae, pupae, and adults were irradiated at target doses of 30, 50, or 100 Gy or they were left untreated as controls in replicated experiments. When eggs and pupae were irradiated with 100 Gy, emergence rates greatly decreased and, although some adults laid eggs, they did not hatch. Egg hatching of irradiated larvae and adult decreased with increasing irradiation doses. However, electron beam irradiation did not kill P. xylostella directly. Adult longevity was not affected. Reciprocal crosses between irradiated and unirradiated moths demonstrated that females were more radiosensitive than males in hatchability; however, the difference was not significant. In addition, electron beam-irradiated larvae showed typical DNA fragmentation in a dose-dependent manner compared with cells from unirradiated larvae. Our findings suggest that electron beam irradiation induces abnormal development and reproduction of P. xylostella; therefore, it may contribute to effective disinfestation and quarantine treatments of P. xylostella.  相似文献

Although the pupation behavior of blowflies has been widely studied, this preliminary study was done on the vertical dispersal behavior (both ascending and descending) and fly emergence rate of third instar larvae of Chrysomya megacephala (Fabricius) to evaluate weather immature stages of blowflies survive burial and emerge out as adult. Third instar larvae of Chrysomya megacephala were placed at three different depths (5 cm, 25 cm and 45 cm) of soil under laboratory conditions to determine the impact of soil depth on the ascending and descending vertical dispersal behavior and the subsequent emergence of adults. The results of this study, although preliminary, but valuable to the field of forensic entomology because they provide new information about both ascending and descending vertical dispersal behavior of Chrysomya megacephala forensically important species of blowfly. In all the cases, maximum number of pupae recovered at the depth of 0 to 5 cm are 35.5 ± 4.5, 34 ± 1, 25 ± 5, when food was located at 5 cm, 25 cm and 45 cm depth respectively. This means that maximum no of larvae reached to the depth of 0 to 5 cm by ascending dispersal irrespective of at which depth they are placed. Paramount pupae were recovered from shallow burial depth of 0–5 cm in ascending dispersal and showed highest eclosion success i.e. 90.1% followed by 25 cm and 45 cm i.e. 71.7% and 55% respectively. While the number of pupae recovered as well as eclosion success was less in descending dispersal with an average of 62.8%, 39.25% and 33.9% at depths of 5, 25 and 45 cm respectively. This manifest if larvae disperse ascendingly, it increases their chance of survival.  相似文献

The green bottle blow fly Lucilia sericata is recognized as being among the first wave of the faunal succession on human cadavers. Thus, it is used to estimate the postmortem interval (PMI). The nutritional intake of larvae is likely to vary subject to the part of a corpse on which they are feeding. A study was therefore conducted to investigate the effect for the type of food substrate on larval growth in this species. Larvae were reared on cow liver, brain, heart, lung, kidney, intestine or minced meat. Results showed significant differences in the duration of the feeding, postfeeding larval, and pupal stages from different substrata (F = 42.43; 74.71 and 655.71; P < 0.0001, respectively). Larval growth measured as length varied significantly between different tissues (F = 3.56; P < 0.05). Larvae that were reared on heart were smaller than those reared on other tissues. Also, there was a significant effect for the type of tissue on the adult size of males (F = 2.41, P < 0.05) and females (F = 2.85, P < 0.05). These results may have important implications for forensic entomologists, since initial infestations commonly occur in wounds or in the cranial area. Therefore, the position at which larvae have been feeding on a body will be a crucial observation at a crime scene and is very important in PMI estimation.  相似文献

The toxicity of the nitric oxide donor S-nitrosoglutathione (GSNO) was tested on the Drosophila melanogaster model system. Fly larvae were raised on food supplemented with GSNO at concentrations of 1.0, 1.5 or 4.0 mM. Food supplementation with GSNO caused a developmental delay in the flies. Biochemical analyses of oxidative stress markers and activities of antioxidant and associated enzymes were carried out on 2-day-old flies that emerged from control larvae and larvae fed on food supplemented with GSNO. Larval exposure to GSNO resulted in lower activities of aconitase in both sexes and also lower activities of catalase and isocitrate dehydrogenase in adult males relative to the control cohort. Larval treatment with GSNO resulted in higher carbonyl protein content and higher activities of glucose-6-phosphate dehydrogenase in males and higher activities of superoxide dismutase and glutathione-S-transferase in both sexes. Among the parameters tested, aconitase activity and developmental end points may be useful early indicators of toxicity caused by GSNO.  相似文献

The lepidopteran insect pests have significant importance in vegetable production. The present study was performed to investigate the baseline studies about the assessment of feeding and consumption potential, utilization indices and losses promises of leafworm, Spodoptera litura (Fab.) on Okra. The data regarding feeding potential, food utilization and consumption indices as well as losses of different larval instars were recorded and subjected to appropriate statistical analysis. The results showed that, in the beginning, the approximate digestibility of various instars was increase, e.g. third instar (51.36%–64.03%), fourth instar (63.42%–69.45%) and fifth instar (70.25%–76.10%). However, after a certain period, the digestibility was decreased and efficiency to convert the ingested food into biomass varied significantly. The consumption index values increased with an increase in time but the consumption and growth rate was declined of fourth instar larvae. The ingestion and digestion increased of third (10.01–13.06, 8.32–11.91 mg), fourth (11.27–17.28, 10.96–14.03 mg) and fifth (12.60–19.40, 11.93–15.28 mg) larval instars. The corrected weight of consumed leaves increased with a gain in body weight. However, in the third instar, a decline was observed on the last day of feeding. Maximum leaf area was consumed by fifth instar larvae (44.66 cm2) followed by fourth (35.41 cm2) and third (27.98 cm2) instars. In conclusion, all the dependent parameters, including food utilization potential, consumption indices and losses were higher for fifth instar larvae than others. These results emphasized the re-establishment of fundamental (economic threshold level: ETL, economic injury level: EIL) integrated pest management concepts.  相似文献

The isolation and purification of fractions with juvenile hormone activity from whole animal extracts of larvae, and from extracts of haemolymph from larvae and adults is described. Using the Galleria bioassay three such fractions were isolated from both third and fourth instar hoppers and from adult females. The chromatographic behaviour of these fractions indicates that one contains JH III, but the other two contain unknown juvenilizing compounds.  相似文献
