The effect of tris, choline, and ethanolamine chlorides on theactivity of Mg2–dependent ATPase in membrane fractions(cell walls, mitochondria, and microsomes) of Zea mays L. (cv.Neve Yaar 22), Avena saliva L. (cv. Mulga), and Hordeum vulgareL. (cv. Omer) was compared with the effect of KC1 and NaCl.Considerable salt effects on apparent Mg2+ATPase activity werefound only at relatively high pH values (8.2) at which Mg2+.ATPaseactivity was low in the absence of monovalent cation salts.The Mg2+-dependent ATP hydrolysis by ATPases from all the membranefractions increased in the presence of at least one of the organiccations to the same extent as in the presence of KCI or NaCl.The monovalent organic cations are only very slowly absorbedby corn roots in comparison with K+ and Na+. It is concluded that monovalent salt effects on ATPase fromthese plant roots are not cation specific and not related tothe capability of root cells to absorb cations. The pigment changes that occur during transformation of etioplaststo fully developed chloroplasts have been studied in seedlingsof barley (Hordeum vulgare L.) by greening with white lightof low (15–25 µmol m–2 s–1) and medium(150 µmol m–2 s–1) intensity. At least 24h longer was required in the low light regime for the same concentrationof pigment to be accumulated in the seedlings. The increasein pigment content was mainly due to the synthesis of chlorophyllsa and b. Of the carotenoids present, the increases in the levelsof neoxanthin and, especially, ß-carotene were muchgreater than those observed for the other carotenoids. Levelsof lutein also increased but this change was small by comparisonto those observed for neoxanthin and ß-carotene. Inthe long-term the concentration of violaxanthin remained unalteredalthough significant transient changes were recorded. The levelsof antheraxanthin and zeaxanthin were markedly reduced duringgreening. The rate of pigment synthesis decreased with increasingcell age, i.e. from the base to the tip of the primary leaf.Overall, carotenoid levels increased by approximately 100% atthe base of the seedling but hardly at all at the tip. Early seed development was studied in 17 genotypes of barley,Hordeum vulgare L., and 11 genotypes of rye, Secale cerealeL. The numbers of cells and nuclei in the embryos and endospermsof developing seeds were scored daily for 5 days after selfpollination. For embryos, the mean cell doubling times variedfrom 9.2–12.9 h for barley and 15.7–22.7 h for rye.Endosperm mitotic cycle times of both species were shortestover the first 24 h after pollination but then became longer.A non-linear correlation was found between the number of embryocells and the number of endosperm nuclei in barely and rye andis similar to that for other members of the Triticeae. High temperatures (up to 35 °C) were applied to plants ofmalting barley,Hordeum vulgareL. (‘Schooner’) fora period of 5 d during grain-filling. Heat treatment had a profoundeffect on the structure of the mature barley grain. There wasevidence of degradation of endosperm storage products in heat-treatedgrain. Starch granule development was reduced in sub-aleuronecells following heat treatment and alterations to starch granuledistribution and growth were observed in the endosperms of thesegrains. Endosperm cell wall and crushed cell layer (CCL) developmentwere sensitive to high temperatures, with the reduced thicknessof the CCL and generally patchy Calcofluor fluorescence of endospermcell walls indicative of partial hydrolysis of ß-glucans.Increased growth of the embryo took place in heat-treated grainscompared with control grains. Endosperm texture was generallymore friable in heat-treated grains than in control grains,and these grains overmodified during malting, with considerabledegradation of starch in the form of extensive pitting of A-typestarch granules. Excised embryos of Phaseolus vulgaris incubated in a mediumcontaining 10 mg dm–3 farnesol showed enhanced root growthwhereas the leaves remained rudimentary At lower concentrationsof exogenous farnesol normal leaf development occurred and rootgrowth was comparable to untreated cultures. Enhanced root growthalso occurred when excised embryos of Hordeum vulgare were treatedwith farnesol but only at 10 mg dm–3 and this treatmentdid not prevent leaf growth X-ray micro-probe analysis of leavesrevealed an increased phosphorus content in P vulgaris and adecreased sulphur content in H vulgare in comparison to untreatedplants. This electron microscopic study revealed that solutes enterthe apoplasm of root mcristems and move from there to the steleof the root and to the shoots of intact plants. Lanthanum wasused as a plasma membrane-impermeable electron-dense markerof apoplastic solute flux in Hordeum vulgare L., Saiicorniavirgimca L., Spartina alternflora Loisel. and Zea mays L. Thepresence of lanthanum in EM specimens was confirmed by X-raymicroanalysis. Lanthanum that entered the root apoplasm wasalso localized in membrane-bound compartments within cells ofeach plant Lanthanum was localized in vesicles, ER, and vacuolesof root and leaf cells. Following root application, lanthanumwas evident in the leaves of the three grass species studied.Lanthanum was rarely observed in S. virginica leaves. Only plantsexposed to 23 mol m–3 lanthanum for 24 h or more showedlanthanum in root cell cytosol and this was concluded to bea toxic response. Techniques are described for the culture of developing barley(Hordeum vulgare L.) caryopses. Over a 7 d period in culturethe dry weights and the amounts of starch and protein increasedby at least twofold. Growth was sustained for at least 20 d.The effects of glutamine and cysteine on the amount and compositionof the hordein storage proteins were also studied. Glutaminestimulated total hordein accumulation but caused a disproportionateincrease in the amount of the S-poor ‘C’ hordeinswhen supplied at 100 mol m–3. Addition of cysteine at1·0 mol m–3 did not increase the amount of Srich‘B’ hordeins. The results suggest that althoughisolated caryopses are able to take up sucrose and glutamineand convert them to starch and protein there is some limitationin their ability to convert externally supplied cysteine intoproteins. We wished to determine the nature of differences in epidermalcell numbers and dimensions between leaves of different lengthin mutants of barley (Hordeum vulgare L. ‘Himalaya’).Three comparisons were made: leaf one (L1)vs. leaf four (L4);wild typevs. nine dwarf mutants and wild typevs. a slender mutant.L1 was shorter than L4, and for most lines this was associatedwith a change in epidermal cell number for the blade, and inboth cell number and length for the sheath. Compared to wildtype, the smaller leaves of dwarf plants generally had shorterand fewer cells in both blade and sheath. The blade of slenderplants was the same length (L1) or longer (L4) than wild type,while the sheath was longer than that of wild type for bothL1 and L4. Slender plants had longer but fewer cells than thewild type along the blade of L1, and shorter but more cellsfor the blade of L4. In the sheath, slender plants had longerand more (L1) or fewer (L4) cells than did the wild type. ForL1, variation in blade width amongst the barley lines was associatedwith a change in file width and file number. For L4, blade widthvaried only with file number, except for slender plants wherenarrow blades were associated with reduced file width. Hencethere was no consistent correlation between changes in cellsize or cell (or file) number with changes in leaf length orwidth. Differences depended on the leaf (L1vs. L4), leaf part(bladevs. sheath), and the nature of the mutation (dwarfvs.slender). Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. Putrescine, spermidine, and spermine content were analysed inzygotic embryos of barley (Hordeum vulgare L.). Changes in polyaminecontent were observed during zygotic embryo growth. In two cultivars,‘Bomi’ and ‘Golden Promise’, the totalpolyamine content in the embryos was 2.6–2.9 nmol mg–1fresh weight 10 d after anthesis, the highest content observed.It dropped to 1.3 nmol mg–1 fresh weight 14 d after anthesis.This drop was caused by decreases in all three polyamine concentrations.From 14 to 35 d after anthesis the putrescine content continuedto decrease while the spermidine and spermine content increased,thus the total polyamine content remained constant until 35d after anthesis. The mutant ‘Ris? 1508’ showeda constant polyamine content around 1.3 nmol mg–1 freshweight from 14 to 35 d after anthesis. The polyamine patternwas conserved in all three lines throughout the period of investigationshowing a spermidine content higher than putrescine contentwhich was, in turn, higher or equal to the spermine content.The polyamine content measured as nmol µg–1 proteindecreased from 14 to 21 d post anthesis in all three lines,because the protein content (µg mg–1 fresh weight)increased during the period. In dedifferentiating zygotic embryoscultured in vitro the putrescine content (nmol mg–1 freshweight) rose by a factor of nine and the spermidine contentdoubled within the first week of cultivation, whereas sperminecontent did not change. Seed survival data for eight diverse species, namely the cerealbarley (Hordeum vulgare L.), the grain legumes chickpea (Cicerarietinum L.), cowpea [Vigna unguiculata (L.) Walp.] and soyabean [Glycine max (L.) Merr.], the timber trees elm (Ulmus carpinifoliaGleditsch.), mahogany (Swietenia humilis Zucc.), and terb (Terminaliabrassii Exell.), and the leaf vegetable lettuce (Lactuca sativaL.) were compared over a wide range of storage environments(temperatures from –13 °C to 90 °C, seed moisturecontents from 1.8 to 25% f. wt) using a viability equation developedpreviously. In accordance with that equation, the effect oftemperature on seed longevity was dependent upon the temperaturerange. The temperature coefficients of the viability equationdid not differ significantly (P > 0.05) among the eight speciesdespite their contrasting taxonomy. Thus the quantitative relationbetween seed longevity and temperature does not vary among diversespecies. The same conclusion was obtained for the coefficientsof a proposed alternative model of the relation between seedlongevity and temperature. The implications of the two temperaturemodels in the viability equation for extrapolations to low andvery low temperatures are discussed. The fine structure of barley aleurone cells was studied in theenzyme secretion phase. An ultrastructural feature of this phaseis the formation of stacked rough endoplasmic reticulum (rER),for such a structure was never found in cells during the enzymesynthesis phase. The anatomical structure of the second leaf blade of barley{Hordeum vulgare L. cv. Koral) was studied in plants exposedto a photosynthetic photon flux density (PPFD) of 200 µmolm–2 s–1 compared with those grown under 25µmolm–2–11. Design-based stereological methods wereused for the estimation of various leaf anatomical characteristicssuch as mesophyll volume, proportion of intercellular spaces,number of mesophyll cells, mean mesophyll cell volume, and internalleaf surface area. The structure of the mesophyll was more affectedby different levels of PPFD than were the stomatal characteristics.Increased PPFD produced thicker leaves with a larger mesophyllvolume having a higher number of less elongated mesophyll cellsand a larger internal leaf surface area. Using data from a well-known, published experiment [Drew (1975)TheNew Phytologist75: 479–490], the potential exploitationof locally available nutrients by barley roots is calculated.Local proliferation of lateral roots does not necessarily achievesignificantly greater exploitation of mobile soil resourceslike nitrate, but it does for less mobile ones such as phosphate.Despite this, the magnitude of the proliferative response isas great to locally available nitrate as it is to phosphate.This implies an ‘over-production’ of roots in responseto localized nitrate availability, prompting a re-evaluationof the nature and implications of the response mechanism(s)of roots to soil heterogeneity. Seeds of barley (Hordeum vulgare L.) and mung bean (Vigna radiata(L.) Wilczek), with orthodox seed storage behaviour, were imbibedfor between 8 h and 96 h at 15 °C and 25 °C, respectively,while barley seeds were also maintained in moist aerated storageat 15 °C for 14 d. These seeds and seedlings, together withcontrols, were then dried to various moisture contents between3% and 16% (wet basis) and hermetically stored for six monthsat —20°C, 0°C or 15°C. In both species, neitherdesiccation nor subsequent hermetic storage of the control lotsresulted in loss in viability. The results for barley seedsimbibed for 24 h were similar to the control, but desiccationsensitivity increased progressively with duration of imbibitionbeyond 24 h in barley or 8 h in mung bean; these treatmentsalso reduced the longevity of the surviving seeds in air-drystorage. Loss in viability in barley imbibed for 48 h was mostrapid at the two extreme seed storage moisture contents of 3·6%and 14·3%, and in both these cases was more rapid at15 °C than at cooler temperatures. Similarly, for mung beanimbibed for 8 h, loss in viability was most rapid at the lowest(4·3%) moisture content, but in this case it was morerapid at –20 °C than at warmer temperatures. Carbon exchange rate (CER) and transpiration were measured inflag leaves, whole ears, glumes (referring to the total areaof glumes and lemmas) and awns, in six hexaploid spring wheats(Triticum aestivum L.), three cultivated tetraploid spring wheats(T. turgidum L.), four wild tetraploid wheats (T. dicoccoides),eight six-rowed barleys (Hordeum vulgare L.) and five two-rowedbarleys (H. vulgare L.). Differences between varieties and between species in total earCER and transpiration were associated largely with differencesin ear surface area rather than with rates per unit area. Ratesof CER and transpiration per unit area of ears were 40–80%of those of flag leaves, depending on the species. However, since ear surface area was greater than flag leaf areaby a factor of 1.1, 3.9, 5.5 and 4.4, in hexaploid wheat, tetraploidwheat, six-rowed barley, and two-rowed barley, respectively,total ear CER reached up to 90% of that of the flag leaf. The contribution of awns to total ear CER depended largely ontotal awn surface area per ear, rather than on CER per unitawn area. Awns contributed about 40–80% of total spikeCER, depending on the species, but only 10–20% of spiketranspiration. The disproportionately small contribution ofawns to ear transpiration was caused by the very low rate oftranspiration per unit area of awns. Thus, while transpirationratio (CER/transpiration) was about the same in flag leavesand glumes, it was higher by several orders of magnitude inthe awns. A large amount of awns in the ear is therefore a drought adaptiveattribute in these cereals, for which tetraploid wheat exceededhexaploid wheat and six-rowed barley exceeded two-rowed barley. A study was made of the possible involvement of calcium channelsand calmodulin during the calcium-dependent inhibition of mitochondrialrespiration by abscisic acid (ABA) in meristematic protoplastsobtained from light-grown barley (Hordeum vulgare L. cv. Patty)seedlings. The calcium channel blockers lanthanum, verapamiland nifedipine were all found to reduce the Ca2+-dependent inhibitionof protoplast respiration by ABA. The ionophore A23187 [GenBank] itselfcaused an inhibition of protoplast respiration, possibly becauseit mimicked the action of ABA by increasing plasmalemma permeabilityto extracellular calcium. By contrast, calmodulin antagoniststrifluoperazine and compound 48/80 both caused a partial decreasein the Ca2+-dependent inhibition of protoplast respiration byABA. In contrast to the action of ABA, gibberellic acid markedlyincreased the rates of protoplast respiration but this did notappear to require the presence of extracellular calcium ions.These results support the hypothesis that ABA increases plasmalemmapermeability to extracellular calcium which might then directlyor indirectly act as a second messenger, possibly in conjunctionwith calmodulin, to regulate mitochondrial dark respirationwhich is an important part of early meristematic cell development. The experiments described in this paper concern the nature ofthe K+-influx mechanism in Lemna minor L. It is establishedthat K+ influx is ‘site restricted’ and that theaffinity of the system involved is in close agreement with thatof the well-documented System I mechanisms that are found inmany plants (Epstein, 1972). Experiments with ATP and CCCP suggestthat K+ influx is an active process, that the influx machineryresides at the plasmalemma and that this machinery containsan ATPase activity. The method for assay of in vivo nitrate reductase (NR) activitywas standardized for barley (Hordeum vulgare L.). NR activitywas determined in the various organs of the main shoot of field-grownJyoti barley at 40 kg N ha–1. Total nitrate reductaseactivity (TNRA) of each organ for the period it was metabolicallyactive was calculated. The NR activity was highest in the laminae,followed by the sheaths, reproductive organs; and internodes.The NR activity was high in the first-formed laminae and itshowed a decline in the ones formed subsequently. The valuesvaried from 43.2 ± 4.33 to 7.2 ± 1.49 µmolNO3 reduced g–1 dry wt. h–1. Maximum TNRAin the laminae, sheath, and internodes was at 49, 84, and 84–93d after sowing, respectively. The TNRA of the main shoot asa whole showed three peaks, one around 49 d, a second around63 d, and a third around 84 d after sowing. Correlation coefficient(r) between NR and NO3 concentration was highly significantin the laminae and sheath viz. 0.76*** and 0.62***, respectively.The results are discussed in relation to alteration in managementpractices to maximize nitrate assimilatory activity and theamount of reduced N harvested.  相似文献
