A rapid micropropagation protocol through induced multiple shoots from the cotyledonary explant of mulberry (Morus alba L) is described. The highest number of shoots (20.3) was obtained when explants from 14-d-old embryos were cultured on Murashige and Skoog (MS) medium supplemented with 7 μM thidiazuron for 45 d. Of the three cultivars used, cv. S-36 was the best followed by cv. K-2 and S-1. The shoots were transferred to MS medium supplemented with 5 μM 6-benzylaminopurine for elongation. The elongated shoots were rooted on half strength MS medium containing 1 – 7 μM indole 3-butyric acid or 1-naphthalene acetic acid. The rooted plants were transplanted to soil with 90 % success. The emerged shoot primordia probably initiated from the pre-existing meristems since the shoot bud show definite vascular connection to the major vascular tissue. The morphogenetic responses of cotyledonary nodal explants of Vigna mungo (L.) Hepper cv. VBN1 cultured on the same Murashige and Skoog's medium, B5 vitamins, and 13.31 µM N6-benzylaminopurine showed variations in the pattern of multiple shooting and morphology of leaves in dependence on initial explants (presence/absence of cotyledons). The regenerated shoots elongated in the initial medium and most of them rooted in the presence of 2.41 µM indole-3-butyric acid, and flowered in vitro. Rooted plants could be transferred to the field after hardening.  相似文献

A simple and rapid method for multiple shoot formation in vitro from immature embryo axis explants of Carica papaya L. cvs. Honey Dew, Washington and Co2 is described. Multiple shoot regeneration was achieved by culture of the explants on modified Murashige and Skoog (MS) medium supplemented either with thidiazuron (TDZ; 0.45–22.7 μM) or a combination of benzylaminopurine (BAP; 0.2 – 8.84 μM) and naphthalene acetic acid (NAA; 0.5 – 2.64 μM). Highest frequency of shoot regeneration occurred on medium supplemented either with 2.25 μM TDZ or a combination of BAP (4.4 μM) and NAA (0.5 μM). Composition of the basal media influenced the frequency of multiple shoot initiation. Stunted shoots regenerated at 4.5 μM and higher concentrations of TDZ. Such shoots could, however, be elongated by transfer to medium containing 5.7 μM GA3. Rooting of the regenerated shoots was achieved in presence of indolebutyric acid (IBA; 4.92 – 19.68 μM), however, least response was in presence of 14.7 μM IBA. The effects of different plant growth regulators on in vitro adventitious shoot formation in Virginia pine (Pinus virginiana Mill.) zygotic embryo explants were quantitatively evaluated. Using Tang and Ouyang (1999) (TE) basal medium supplemented with 11.4 μM indole-3-acetic acid (IAA) and 2.2 μM N6-benzyladenine (BA), callus was observed after 3–6 weeks of culture. Calluses were transferred to TE basal medium supplemented with 0.49 μM indole-3-butyric acid (IBA) and 8.8 μM BA for 6–9 weeks, where they produced numerous small shoot primordia. They were then transferred to TE basal medium supplemented with 0.49 μM IBA and 4.4 μM BA to promote growth and elongation of adventitious shoots. After elongated shoots were transferred to TE medium containing 0.05 μM α-naphthaleneacetic acid (NAA) for 6 weeks, adventitious roots were formed. A method was developed to initiate multiple shoots from the young shoot of kenaf. Young shoots along with the cotyledons were excised from ten-day old aseptically germinated seeds and pre-cultured for two weeks in Murashige and Skoog (MS) medium supplemented with benzyl adenine (BA) or a combination of BA and kinetin. After two weeks in culture, elongated shoots were excised above the cotyledonary nodes and cultured on fresh medium of the same composition. Multiple shoots were initiated within eight weeks. The number of shoots varied among cultivars. The highest number of shoots (11/explant) occured in cultivar Tainung 2 (T2) cultured in MS medium supplemented with 8.8 μM BA. Concentrations of BA higher than 8.8 μM had a negative effect on the number of shoots. Furthermore, callus growth was initiated from which morphologically abnormal shoots were induced. Kinetin had a significant effect only on cultivar Everglades 41 (E41). Shoot elongation and rooting were obtained simultaneously in half strength MS basal medium with no plant growth regulators. About 98% of the rooted plants were grown to maturity under greenhouse conditions. This method was successful with all four genotypes tested. However, significant genotypic variations were observed among the genotypes.  相似文献

Frequency of safflower (Carthamus tinctorius L.) somatic embryogenesis, number of somatic embryos per responding explant and somatic embryo maturation and germination were affected by genotype, explant age, carbon source, and ethylene. Among 8 cultivars tested, 7 were embryogenic with varying frequencies. The best response was obtained with cv. Girna. Whole cotyledonary explant from 10-d-old plants was best responding compared to 5- or 15-d-old ones. Among different carbon sources, sucrose at 87.6 mM concentration was most suitable for embryo induction, maturation and germination. Of the different ethylene inhibitors, silver nitrate at 50 [micro ]M concentration significantly increased the embryogenic frequency and also the number of embryos per responding explant. Silver nitrate has pronounced effect on embryo maturation but had no effect on germination.  相似文献

In vitro propagation protocols were established for two leguminous trees, Bauhinia variegata and Parkinsonia aculeata. In each case axillary shoot proliferation was achieved from nodal explants from mature (6-2-8 years) trees using Murashige & Skoog's medium supplemented with 2.22–31.1 M of 6-benzyladenine. Subsequent rooting of the regenerated shoots was achieved on medium containing 2.46–14.8 M of indole-3-butyric acid. Successful transfer of the regenerants to soil has been accomplished.  相似文献

A method for micropropagation ofDalbergia sissoo has been developed. Single node segments obtained from coppice shoots of a mature tree (20 – 25 year old) produced 3–4 shoots per explant on Murashige and Skoog (MS) medium containing 4.4 x 10−6 M benzylaminopurine (BAP) and 4.4 × 10−7 M of Β-naphthoxy acetic acid (NOA) (shoot multiplication medium) within 4 weeks. Thein vitro regenerated shoots were 3 – 4 cm in length and provided 2 to 3 culturable nodal segments which on shoot multiplication medium again produced 3–4 shoots. Following this procedure 18–24 shoots were produced from single nodal segment within 60 d. 80 % of the shoots directly produced five roots when they were firstly treated with MS medium supplemented with 10−5 M indole-3-butyric acid (IBA) and subsequently transferred to half strength liquid MS medium containing 1 % activated charcoal followed by half strength liquid MS free hormones, vitamins and activated charcoal. Thein vitro raised plants were hardened for survival after transplantation to soil by exposing them to various humidity conditions, gradually from higher to low, with nearly 100 % transplant success. Eryngium foetidum L. plants were regenerated from mature leaf and petiole explants through direct organogenesis without intervening callus phase. From leaf explants, adventitious multiple shoots raised on Murashige and Skoog (MS) medium supplemented with 4.43 M benzylaminopurine (BAP) and 0.57 M indole-3-acetic acid (IAA), whereas in petiole explants shoot regeneration occurred at 8.86 M BAP and 0.57 M IAAA. 80% of the leaf explants and 44% of petiole explants produced shoots after four weeks of culture. The regenerated plants were rooted on MS medium supplemented with 2.46 M indole-3-butyric acid and 2.88 M gibberellic acid. The plants were successfully established in the soil and showed 70.9% survival in the field.  相似文献

We developed an efficient method for sugar beet multiplication in vitro from excised immature inflorescence tips. On Murashige & Skoog medium supplemented with 4.4 μM 6-benzylamino- purine and 1.3 μM naphthaleneacetic acid, multiple bud clumps were induced from segments of inflorescence tips. The clumps proliferated rapidly. By radiation of small bud clumps at an appropriate dose and by directional selection for NaCl tolerance, we obtained salt-tolerant bud clumps and regenerated plantlets. The plants were vernalized and self-pollinated. The seeds of the regenerated plants were sown in pots of sand and irrigated every day with a solution of 342 mM NaCl. Some of the seeds germinated and grew normally in the 342 mM NaCl solution, exhibiting higher salt-tolerance than the control ones; such seedlings after the saline selection were transplanted to soil and the plants grew normally and produced plump root tuber similar to controls. The seeds from two selected lines germinated and grew for a few weeks in 513 mM NaCl solution before the seedlings withered. In saline soil where the salt concentration was about 154 mM, the yields of tuber from the plants of three salt-tolerant lines were about 45–50 tons ha−1, approximately 2.6–2.9 times of the controls. It is concluded that we have got salt-tolerant materials with good agronomic traits for sugar beet breeding via selection in vitro.  相似文献

Rooting of Eucalyptus globulus shoots was influenced by the concentration of the indole butyric acid (IBA) and NH4 + in the root-induction medium. Optimum plantlet vigor and survival were achieved using low concentrations (1 – 2.5 μM) of IBA and when NH4NO3 was removed. Removal of NH4 + also had a significant effect on medium pH, its presence caused a decrease in pH as the culture period proceeded. When different nitrate compounds (excluding NH4NO3) were used as the nitrogen source, the medium pH was more stable and this was associated with higher root production. The higher root production, in association with appropriate IBA concentrations, produced plantlets with higher survival and better growth on transfer to soil. Radioprotective ability of the aqueous extract of the fruit of Terminalia chebula (TCE) was evaluated for its antioxidant and radioprotective abilities. TCE (50 μg) was able to neutralise 1,1-diphenyl-2-picrylhydrazyl, a stable free radical by 92.9%. The free radical neutralizing ability of TCE was comparable to that of ascorbate (100 μM) 93.5% and gallic acid (100 μM) 91.5% and was higher than that of the diethyldithiocarbamate (200 μM) 55.4%, suggesting the free radical activity of TCE. TCE protected the plasmid DNA pBR322 from undergoing the radiation-induced strand breaks. Radiation damage converts the supercoiled form (ccc) of plasmid to open circular form (oc); the presence of TCE during radiation exposure protected the plasmid from undergoing these damages. The administration of TCE (80 mg/kg body weight, i.p.) prior to whole body irradiation of mice (4 Gy) resulted in a reduction of peroxidation of membrane lipids in the mice liver as well as a decrease in radiation-induced damage to DNA, as assayed by single-cell gel electrophoresis (comet assay). TCE also protected the human lymphocytes from undergoing the gamma radiation-induced damage to DNA exposed in vitro to 2 Gy gamma-radiation. Shoot regeneration was achieved from immature cotyledons of five chickpea (Cicer arietinum L.) genotypes: C235, ICC4971, ICC11531, ICC12257 and ICC12873. The cotyledons cultured on Murashige and Skoog (MS) medium supplemented with 3 or 5 mg dm–3 zeatin with or without 0.04 mg dm–3 indole acetic acid (IAA) showed formation of cotyledon like structures (CLS) at their proximal ends. Subsequently, shoot regeneration took place in some of the CLS forming explants. CLS were also formed in cotyledons cultured on MS + 0.2 – 1 mg dm–3 thidiazuron (TDZ); direct shoot regeneration was observed in cotyledons cultured on 1 mg dm–3 TDZ. The shoot buds elongated on media containing indole butyric acid (IBA), benzylaminopurine (BAP) and gibberellic acid (GA3). Complete plantlets were obtained by rooting of shoots following pulse treatment with 200 mg dm–3 IBA for 5 min and culture on growth regulator free half-strength MS medium.  相似文献

This study was carried out to determine the effect of temporary submersion of hypocotyl segments in water on in vitro explant growth and shoot regeneration on MS (Murashige and Skoog, 1962) medium supplemented with 1 mg l−1 BAP (6-benzylaminopurine) and 0.02 mg l−1 NAA (naphthaleneacetic acid) in three flax cultivars. It was observed that water-treated hypocotyl explants gave rise to the highest values with respect to shoot regeneration percentage, shoot number per hypocotyl, shoot length and total shoot number per Petri dish, successful rooting and plantlet establishment. This procedure may be applicable for other species cultured in vitro.  相似文献

Multiple shoot regeneration from the cut plumular ends of embryo axes of chickpea (Cicer arietinum L.) was evaluated on Murashige and Skoog medium having different concentrations of thidiazuron (TDZ) (0.1 to 10.0 mg dm–3) 6-benzylaminopurine (BAP) (0.5 and 1.0 mg dm–3), kinetin (0.5 and 1.0 mg dm–3) or zeatin (2.0 and 4.0 mg dm–3). TDZ (0.2 mg dm–3) was found to be the most effective cytokinin as it produced multiple shoots in 100 % of the explants from genotypes C235, ICC5166, ICC12269, ICC4951, ICC11531, BG256 and a local cultivar. Shoots were elongated on growth regulator-free medium, and rooted on growth regulator-free medium containing 1/4 MS salts + full vitamins + 3 % sucrose. Plantlets formed were acclimatized for 12 – 15 d in MS medium with a gradual reduction in sucrose concentration and transferred into pots filled with soil and kept in the field; this resulted in more than 70 % survival. The plants developed normally and produced fertile flowers and set seeds. Low temperatures, maximum 19.0 °C, and minimum 8.2 °C, during the first 15 d of transfer favoured survival on transfer to pots.  相似文献

In order to establish and optimize an in vitro micropropagation method of Drosera peltata (a tuberous sundew), a carnivorous plant, the effects of medium type, MS medium concentration, pH and cytokinin type on shoot proliferation and tuber formation were investigated, using one month-old shoot tips. The shoot proliferation and tuber formation were most effective on 1/2 MS medium without cytokinins. The optimum pH of the media was pH 5.7. Tubers were planted in plastic pots filled with 1:1 peat moss and sand. The survival rate of the plantlets was almost 100%, and they exhibited normal development. With subculture every 12 weeks, hundreds of the plants were propagated from a single plant within a year.  相似文献

Phylloclade explants of Schlumbergera and Rhipsalidopsis were cultured in vitro to produce axillary and adventitious shoots. The explants of both species, taken from greenhouse-grown plants, produced only axillary shoots. There was a pronounced improvement in adventitious shoot formation in phylloclade explants of cultivar CB4 of Rhipsalidopsis by increasing numbers of subcultures of axillary shoots used as donor plants. The axillary shoots generated from the explants were either subcultured to produce successive generations of axillary shoot cultures or made into phylloclade explants and tested for adventitious shoot formation at each subculture. The duration of each subculture varied from 6 to 12 weeks. After the first subculture, sporadic adventitious shoot formation began, and after the third subculture 87% explants of cultivar CB4 produced adventitious shoots at a frequency of about 12 shoots per explant. In contrast, there was no improvement in regenerative ability in explants of cultivar Thor-Olga of Schlumbergera up to third subculture. Adventitious shoots could be produced by callus culture too. Cultivar CB4 was highly regenerative, producing as many as 10 adventitious shoots per square cm of callus. In vitro grown plantlets, when transferred to pots continued to show prolific growth.  相似文献

Conditions affecting rhizogenesis in vitro and ex vitro and subsequent acclimatisation of Telopea speciosissima (waratah) were investigated. Clonal selections were successfully rooted in vitro in agar, on filter paper bridges or using crushed quartz-sand, the last substrate resulting in superior growth of roots. The in vitro substrates were impregnated with half-strength MS, 7.5 gl-1 sucrose and various concentrations of IBA. For the quartz-sand, an IBA concentration of 50 M was optimal, 70% of microcuttings were rooted. No plantlets rooted in vitro were acclimatised to ex vitro conditions (using mist, fog or humidity tent regimes). Microcuttings (25–45 mm in length) were rooted ex vitro in a fog humidity regime (droplet size <10 m) using an IBA powder dip (3 g IBA kg-1). Neither a mist nor a humidity-tent regime was suitable for rooting of waratah microshoots ex vitro. A peat and perlite mixture was superior to crushed quartz-sand or potting mix for the rooting of microshoots; this appeared to be related to the air-filled porosity (>20%) of the mixture, measured after the medium was saturated and then drained for 24h. Plantlets must be left under the high humidity regime until shoot growth resumes (four to eight weeks) otherwise plant mortality increase significantly. Shoot regeneration was achieved from leaves of in vitro cultures of Prunus avium L. cv. 'Lapins' and 'Sweetheart' using woody plant medium (WPM) supplemented with 1-naphthalene-acetic acid (NAA) and thidiazuron (TDZ) or benzyladenine (BA). Percent regeneration was influenced by plant growth regulators and by explant type, orientation and wounding. Optimal regeneration was observed with whole-leaf explants wounded by transverse cuts along the midrib and incubated abaxial surfaces uppermost, on media supplemented with 2.27 or 4.54 µM TDZ plus 0.27 µM NAA. The percent regeneration of the two cultivars was not significantly different. Optimum conditions for regeneration resulted in 71.4% of 'Lapins' and 54% of 'Sweetheart' explants producing one or more shoots per explant.  相似文献

Axillary buds from adult field-grown plants of Lavandula dentata L. were used to evaluate the effect of growth regulators and culture media on the in vitro shoot proliferation and growth. The highest multiplication rate was obtained using Murashige and Skoog (MS) medium supplemented with a combination of 2.2 μM of benzyladenine and 2.5 μM indole-3-butyric acid. The best condition for rooting was MS medium plus 2.5 μM naphthaleneacetic acid. Rooted plantlets were successfully transferred to soil. Short-term culture derived plants (6 month) exhibited a normal development, but a low frequency of not heritable morphological changes were detected in long term culture derived plants (more than 1 year).This work was supported by a grant from the University of Caxias do Sul and CNPq, Brazil.  相似文献
