The 8-methoxycarbonyloctyl glycoside of the tetrasaccharide hapten, O-α-l-rhamnopyranosyl-(1→2)-O-α-l-rhamnopyranosyl-(1→3)-O-α-l-rhamnopyranosyl-(1→ 3)-2-acetamido-2-deoxy-β-d-glucopyranoside and the trisaccharide glycoside 8-methoxycarbonyloctyl O-α-l-rhamnopyranosyl-(1→3)-O-α-l-rhamnopyr-anosyl-(1→3)-2-acetamido-2-deoxy-β-d-glucopyranoside were synthesized by sequential Koenigs-Knorr reactions from monosaccharide units. The tetrasaccharide represents the complete skeletal repeating unit of Shigella flexneri serogroup Y lipopolysaccharide. Both oligosaccharide haptens are functionalized for covalent attachment to proteins, cell surfaces, and solid supports. 1H-N.m.r. evidence for the conformations of these oligosaccharides in solution is presented and shown to be consistent with predictions based on the exo-anomeric effect  相似文献

Benzyl-3-O-benzyl-2-benzyloxycarbonylamino-6-O-[2-benzyloxycarbonyl-amino-2-deoxy-3,4-O-(tetraisopropyldisiloxane-1,3-diyl)- β-d-glucopyranosyl]-2-deoxy-α-d-glucopyranoside was coupled with methyl (4,5,7,8-tetra-O-acetyl-3-deoxy-α-d-manno-2-octulopyranosyl bromide)onate (13) to yield the α-glycosidically linked trisaccharide. After deacetylation and selective introduction of a second 7′,8′-O-tetraisopropyldisiloxane group, a further glycosidation reaction with 13 led regioselectively to the tetrasaccharide benzyl O-[methyl (4,5,7,8-tetra-O-acetyl-3-deoxy-α-d-manno-2-octulopyranosyl)onate]-(2→4)-O-{methyl [3-deoxy-7,8-O-(tetraisopropyldisiloxane-1,3-diyl)-α-d-manno-2-octulopyranosyl]-onate}-(2→6)-O- [2-benzyloxycarbonylamino-2-deoxy-3,4-O-(tetraisopropyldisiloxane-1,3-diyl)-β-d-glucopyranosyl]- (1→6)-3-O-benzyl-2-benzyloxycarbonyl-amino-2-deoxy-α-d-glucopyranoside. A series of deblocking steps gave O-(3-deoxy-α-d-manno-2-octulopyranosylonic acid)-(2→4)-O-(3-deoxy-α-d-manno-2-octulopyranosylonic acid)- (2→6)-O-(2-amino-2-deoxy-β-d-glucopyranosyl)-(1→6)-2-amino-2-deoxy-d-glucopyranose which was identical with a tetrasaccharide that had been isolated by hydrazinolysis of the lipopolysaccharide from Salmonella minnesota R 595. Hence, synthetic proof is provided for the linkages in this part of the inner core region of lipopolysaccharides.  相似文献

O-(2,4-Di-O-chloroacetyl-α-l-rhamnopyranosyl)-(1 → 2)-O-(3,4,6-tri-O-benzoyl-α-d-galactopyranosyl)-(1 → 3)-O-(2-acetamido-4,6-di-O-acetyl-2-deoxy-α-d-glycopyranosyl)-(1 → 3)-2,4-di-O-benzoyl-α-l-rhamnopyranosyl trichloroacetimidate (1) was synthesized in a stepwise manner, using the following monosaccharide units: 2-(trimethylsilyl)ethyl 2,4-di-O-benzoyl-α-l-rhamnopyranoside, 2-azido-4,6-O-benzylidene-3-O-chloroacetyl-2-deoxy-β-d-glycopyranosyl chloride, methyl 3,4,6-tri-O-benzoyl-2-O-(4-methoxybenzyl)-1-thio-β-d-galactopyranoside, and 2,4-di-O-benzoyl-3-O-chloroacetyl-α-l-rhamnopyranosyl chloride. Compound 1 corresponds to a complete tetrasaccharide repeating unit of the O-specific polysaccharide of the lipopolysaccharide of Shigella dysenteriae type 1.  相似文献

8-Methoxycarbonyloctyl 2-azido-4,6-O-benzylidene-2-deoxy-β-d-mannopyranoside reacted with 2,3,4-tri-O-acetyl-α-l-rhamnopyranosyl bromide to give a disaccharide from the which the glycosyl-acceptor 8-methoxycarbonyloctyl 2-azido-4,6-O-benzylidene-2-deoxy-3-O-(2,4,-di-O-acetyl-α-l-rhamnopyranosyl)-β-d-manno pyranoside (19) was obtained. This glycosyl-acceptor with 2,3,4,6-tetra-O-benzyl-α-d-glucopyranosyl chloride to give trisaccharide derivative 22 and with 2,3,6-tri-O-(α-2H2)benzyl-4-O-(2,3,4,6-tetra-O-(α-2H2)benzyl-α-d-glucopyranosyl)-α-d-glucopyranosyl chloride to give tetrasaccharide derivative 29. Deblocking of 22 yielded 8-methoxycarbonyloctyl O-(α-d-glucopyranosyl)-(1→3)-O-α-l-rhamnopyranosyl-(1→3)-2-acetamido-2-deoxy-β-d-mannopyranoside and deblocking of 29 8-methoxycarbonyloctyle O-α-d-glucopyranosyl-(1→4)-O-α-d-glucopyranosyl-(1→3)-O-α-l-rhamnopyranosyl- (1→3)-2-acetamido-2-deoxy-β-d-mannopyranoside. Both oligosaccharides represent the “repeating unit” of the O-specific chain of the lipopolysaccharide from Aeromonas salmonicida.  相似文献

The structures of four new saponins, polyphyllin C, D, E and F, isolated from the tubers of Paris polyphylla have been elucidated as diosgenin-3-O-α-l-rhamnopyranosyl(1→3)-β-d-glucopyranoside, diosgenin-3-O-α-l-rhamnopyranosyl(1→3)- [α-l-arabinofuranosyl(1→4)]-β-d-glucopyranoside, diosgenin-3-O-α-l-rhamnopyranosyl(1→2)-α-l-rhamnopyranosyl (1→4)[α-l-rhamnopyranosyl(1→3)]-β-d-glucopyranoside and diosgenin-3-O-α-l-rhamnopyranosyl(1→4)[α-l- rhamnopyranosyl(1→3)][β-d-glucopyranosyl(1→2)]-α-l-rhamnopyranoside, respectively, on the basis of chemical and spectral data.  相似文献

Two new saponins beshornin and beshornoside have been isolated from the methanolic extract of Beshorneria yuccoides leaves and their structures elucidated. Beshornin is 3-O-[α-l-rhamnopyranosyl-(1 → 4)-β-d-glucopyranosyl- (1 → 2)-[α-l-rhamnopyranosyl-(1 -+ 4)-P-D-glucopyranosyl-(1 → 3)]-β-d-glucopyranosyl-(1 → 4)-β-d- galactopyranosyl-(25R)-5α-spirostan-3β-ol, whereas beshornoside is 3-O-[α-l-rhamnopyranosyl-(1 → 4)- β-d)-glycopyranosyl-(1 → 2)]-[α-l-rhamnopyranosyl-(1 → 4)-β-d-glucopyranosyl-(1 → 3)]-β-d-glucopyranosyl- (1 → 4)-β-d-galactopyranosyl 26-O-[β-d]-glucopyranosyl-(25R)-5α-furostan-3β,22α,26-triol.  相似文献

Two new saponins, agavasaponin E and agavasaponin H have been isolated from the methanolic extract of Agave americana leaves and their structures elucidated. Agavasaponin E is 3-O-[β-d-xylopyranosyl-(1→2glc1)-α-l-rhamnopyranosyl-(1→4)-α-l-rhamnopyranosyl-(1→3glc 1)-β-d-glucopyranosyl-(1→4)-β-d-glucopyranosyl-(1→4)-α-d-galactopyranosyl]-(25R)-5α-spirostan-12-on-3β-ol, whereas agavasaponin H is 3-O-[β-d-xylopyranosyl-(1→2 glc 1)-α-l-rhamnopyranosyl-(1→4)-α-l-rhamnopyranosyl-(1→3 glc 1)-β-d-glucopyranosyl-(1→4)-β-d-glucopyranosyl-(1→4)-β-d-galactopyranosyl]-26-O-[β-d-glucopyranosyl]-(25R)-5α-furostan-12-on-3β,22α,26-triol.  相似文献

The stereoselective glycosylation of a model alcohol (cyclohexanol) by derivatives of 2-azido-2-deoxy-d-galactopyranose was studied under various conditions. 2-Azido-3,4,6-tri-O-benzyl-2-deoxy-β-d-galactopyranosyl chloride (9) was found to be the most efficient glycosylating agent for the synthesis of oligosaccharides containing 2-acetamido-2-deoxy-α-d-galactopyranose residues, and gave a tetrasaccharide, which is a determinant of the blood-group A (Type 1), i.e., O-α-l-fucopyranosyl-(1→2)-[O-2-acetamido-2-deoxy-α-d- galactopyranosyl-(1→3)]-O-β-d-galactopyranosyl-(1→3)-2-acetamido-2-deoxy-d-glucose, and its trisaccharide fragment, O-2-acetamido-2-deoxy-α-d-galactopyranosyl-(1→3)-O-β-d-galactopyranosyl-(1→3)-2-acetamido-2-deoxy-d-glucose. In the course of this synthesis, the determinant trisaccharide related to the H blood-group, i.e., O-α-l-fucopyranosyl-(1→2)-O-β-d-galactopyranosyl-(1→3)-2-acetamido-2- deoxy-d-glucose, was also obtained.  相似文献

The structure of microphyllic acid, the main glycosidic acid from the ether soluble resin of Convolvulus microphyllus has been elucidated as the O-α-l-rhamnopyranosyl-(1 → 6)-[O-α-l-rhamopyranosyl -(1 → 4)]-O-β-d-glucopyranosyl-(1 → 3)-O-α-l5-rhamnopyranosyl-(1 → 3)-O-β-d-fucopyranoside of 11-hydroxypalmitic acid, using mainly GLC and mass spectrometry of the derivatized sugars.  相似文献

Partial hydrolysis of a larch arabino(4-O-methylglucurono)xylan afforded two series of oligouronides composed of 4-O-methyl- d-glucuronic acid and d-xylose residues. The first series included aldouronic acids up to the aldopentaouronic acid. Methylation analysis indicated that the aldopentao- and aldotetrao-uronic acids were mixtures of isomers. One aldotetraouronic acid was isolated and identified as O-β-d-Xylp-(1 → 4)-O-β-d-Xylp-(1 → 4)-O-(4-O-Me-α-d-GlcAp)-(1 → 2)-d-Xyl. The two isomeric aldotriouronic acids were separated from each other. The acids of the second series, which were composed of two uronic acids and 2-4 d-xylose residues, were identified as follows: O-β-d-Xylp-(1 → 4)-O-(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-O(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-d-Xyl, O-(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-O-(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-O-β-d -Xylp-(1 → 4)-D-Xyl, O-(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-O-(4-O-Mec-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-D-Xyl, and O-(4-O-Me-α-d-GlcAp)-(1 → 2)-O-β-d-Xylp-(1 → 4)-O-(4-O-Me-α-d-GlcAp)-(1 → 2)-D-Xyl. The first three compounds were new acidic oligosaccharides. The 4-O-methyl-d-glucuronic acid in the second series was present in a larger proportion than in the first series, indicating that a large proportion of the uronic acid side-chains were located on two contiguous D-xylose residues in the backbone of the softwood xylan.  相似文献

S-2:3-O-[α-l-arabinopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→2)-α-l-arabinopyranosyl]-hederagenin; S-3:3-O-[α-d-xylopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→2)-α-l-arabino-pyranosyl ]-hederagenin; S-4:3-O- [α-l-arabinopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→ 2)-α-l-arabinopyranosyl]-hederagenin; S-5:3-O- [α-l-rhamnopyranosyl-(1→2)-α-l-arabinopyranosyl ]-hederagenin. A new α-D-glucan, designated elsinan, has been isolated from the culture filtrate of Elsinoe leucospila grown in potato extract-sucrose medium. Acid hydrolysis of the methylated polysaccharide gave 2,3,6- and 2,4,6-tri-O-methyl-D-glucose, in the ratio of 2.5:1.0, together with small proportions of 2,3,4,6-tetra- (0.7%) and 2,4-di-O-methyl-D-glucose (0.5%), indicating that the glucan is an essentially linear polymer containing (1→4)- and (1→3)-α-D-glucosidic linkages. Periodate oxidation, followed by borohydride reduction and mild hydrolysis with acid (mild Smith degradation) yielded 2-O-α-D-glucosyl-D-erythritol and erythritol, in the molar ratio of 1.0:1.4, and a trace of glycerol. Partial acid hydrolysis, and also acetolysis, of elsinan gave nigerose, maltose, O-α-D-glucopyranosyl-(1→3)-O-α-D-glucopyranosyl (1→4)-D-glucopyranose, O-α-D-glucopyranosyl-(1→4)-O-α-D-glucopyranosyl-(1→3)-D-glucopyranose, maltotriose, and a small proportion of maltotetraose. It is concluded that elsinan is composed mainly of maltotriose residues joined by α-(1→3)-linkages, in the sequence →3)-α-D-Glcp-(1→4)-α-D-Glcp-(1→.The unique structural features of elsinan are discussed in comparison with other glucans.  相似文献

Two new flavonol tetraglycosides, quercetin 3-O-(4-O-trans-p-coumaroyl)-α-l-rhamnopyranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (krathummuoside A) and quercetin 3-O-(4-O-cis-p-coumaroyl)-α-l-rhamnopyranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside (krathummuoside B) were isolated from the leaves of Mitragyna rotundifolia in addition to eight known compounds, quercetin 3-O-α-l-rhamnopuranosyl (1→2) [α-l-rhamnopyranosyl (1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside, rutin, (−)-epi-catechin, 3,4,5-trimethoxyphenyl β-d-glucopyranoside, (6S, 9R)-roseoside, 3-O-β-d-glucopyranosyl quinovic acid 28-O-β-d-glucopyranosyl ester, (+)-lyoniresinol 3α-O-β-d-glucopyranoside, and (+)-syringaresinol-4-O-β-d-glucopyranoside. The structure elucidation of these compounds was based on analyses of spectroscopic data including 1D- and 2D-NMR.  相似文献

Obtusilobinin and obtusilobin, two new saponins, have been isolated from the ethanolic extract of Anemone obtusiloba (Ranunculaceae). The structural elucidation of obtusilobinin and obtusilobin have showed them to be olean-12-ene-28-oic-3-O-(α-l-arabinofuranosyl 1→2) (α-l-rhamnopyranosyl 1→4)-β-d-glucopyranoside and olean-12-ene-28-oic-3-O-α-l-rhamnopyranosyl 2→1-O-α-l-arabinofuranoside, respectively.  相似文献

Two new saponins, yuccoside C and protoyuccoside C, have been isolated from the methanolic extract of Yucca filamentosa root and their structures elucidated. Partial, acid hydrolysis of the extracellular polysaccharide from Xanthomonas campestris gave products that were identified as cellobiose, 2-O-(β-d-glucopyranosyluronic acid)-d-mannose, O(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-d-glucose, O-(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-[O-β-d-glucopyranosyl-(1→4)]-d-glucose, and O-(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-[O-β-d-glucopyranosyl-(1→4)-O-β-d-glucopyranosyl-(1→4)-d-glucose. This and other evidence supports the following polysaccharide structure (1) which has been proposed independently by Jansson, Kenne, and Lindberg:

Three new phenylpropanoid glycosides, mussatioside I, mussatioside II and mussatioside III were isolated from the methanolic extract of the bark of a new Mussatia species. On the basis of the chemical and spectral evidence their structures were determined as [β-(4′-hydroxyphenyl)ethyl-O-β-D-glucopyranosyl(6 → 1)]-O-β-D-xylopyranosyl(1 → 3)-α-L-(4-O-t-cinnamoyl)rhamnopyranoside, [β-(4′-hydroxyphenyl)-ethyl-O-β-D-glucopyranosyl(6 → 1)]-O-β-D-xylopyranosyl(1 → 3)-α-L-(4-O-dimethylcaffeoyl)rhamnopyranoside and [β-(4′-hydroxyphenyl)ethyl-O-β-D-glucopyranosyl(6 → 1)]-O-β-D-xylopyranosyl(1 → 3)-α-L-(4-O-p-methylcoumaroyl)rhamnopyranoside, respectively. Two new furostanol glycosides, trigofoenosides F and G, have been isolated as their methyl ethers from the methanolic extract of Trigonella foenum-graecum seeds (Leguminosae). The structures of the original glycosides have been determined as (25R)-furost-5-en-3β,22,26-triol, 3-O-α-l-rhamnopyranosyl (1 → 2)β-d-glucopyranosyl (1 → 6)β-d-glucopyranoside; 26-O-β-d-glucopyranoside and (25R)-furost-5en-3β,22,26-triol, 3-O-α-L-rhamnopyranosyl (1 → 2) [β-d-xylopyranosyl (1 → 4)]β-d-glucopyranosyl (1 → 6)β-d-glucopyranoside; 26-O-β-d-glucopyranoside, respectively.  相似文献
