Summary Mature pollen grains from two single cross hybrids, Wf9 × H55 and K64 × K55, were exposed to eleven levels (0 to 6.80 erg/cm2 × 105 at 0.68 intervals) of ultraviolet irradiation and then were used to pollinate their genetic source. The number and weight of the normal and shrunken (partially aborted) kernels on each ear were tabulated. In general, the number of normal kernels decreased and the number and percentage of shrunken kernels increased with increasing exposure. However, significant exposure X hybrid interactions were present indicating that the amount of change depended on the hybrid. No consistent relationship between exposure and either normal or shrunken kernel weight was apparent, but pollen source hybrid was a contributing factor. The embryo weight and coleoptile length after germination were also determined for the normal kernels. Changes in these characters by irradiation were also strongly influenced by the hybrid. These results indicate that the direction and magnitude of the changes in kernel development produced by ultraviolet are modified considerably by the genetic source of the pollen grains. Presumably, genetic variation for ultraviolet response is present and selection would be successful.  相似文献

Summary Correlation, stepwise multiple regression and factor analyses were conducted on grain yield and a number of agronomic characters in the parental, F 1 and F 2 families originating from a 10 X 10 diallel cross in durum wheat. For the F 1 diallel, the correlation analysis indicated that the number of spikes and kernels per plant and 1,000 kernel weight had the highest correlations with grain yield; for the F 2 diallel, the number of spikes and kernels per meter, 1000 kernel weight and plant height showed most striking correlations with same.Stepwise multiple regression analysis indicated that, for the F 1 diallel, number of kernels per plant, 1000 kernel weight and days to maturity were the most potent predictor variables for grain yield, accounting for 78% of its variability. For the F 2 diallel, the number of kernels and number of spikes per meter, 1000 kernel weight and number of kernels per spike were the most potent predictors for grain yield, accounting for 91 % of its variability. Five common factors were extracted which explained 98.8% and 98.1% of the total variance in the F 1 and F 2 diallel, respectively. Summary The free amino acid content of the pollen grains and the style from three single cross hybrids (Wf9 x H55, Ky49 x Ky27, K64 x K55) and two inbred lines (Oh43, H55) was determined. No tyrosine was detected in the pollen grains of any genotype. Significant differences between pollen genotypes were found for aspartic acid, threonine, serine, lysine and histidine with no effect resulting from the vigor of the pollen source. No proline was found in the style of any genotype. Significant differences between style genotypes were obtained for threonine, serine, glutamic acid, leucine, tyrosine, ethanolanine, aminobutyric acid and histidine with a relationship between vigor of the style genotype present for tyrosine, ethanolanine and aminobutyric acid. The relationship between the pollen and style level for each amino acid as influenced by genotype was analyzed. A significant negative correlation was found only for threonine. Apparently, a complementary relationship between the pollen and style exists for some amino acids. Proline and tyrosine are available only from the pollen and style respectively. Threonine levels are balanced with varying contributions from both pollen and style depending on the genotype.  相似文献

Pollen morphology in ten plants of F2 progeny of an interspecific tree hybrid, Chorisia insignis H. B.& K.×C. speciosa St. Hil (Bombacoideae, Malvaceae) has been studied with a view to have an insight in to the effect of hybridization on the pollen morphological features in F2 generation. The study is a sequel to an earlier study on the pollen morphology of Chorisia species and their F1 hybrid, in which case the hybrid pollen uniformly exhibited the apertural features of the male parent and exine features of the female parent. In the F2 progeny the pollen grains display the apertural feature of the male parent in all the plants. However, with regard to exine ornamentation, variability has been observed. Of the ten plants, two plants exhibit the exine features of the male parent showing empty lumina (without bacula), five plants have their pollen exactly like that of the F1 plant (showing prominent columellar heads in the lumina of the apocolpium region) and the remaining three plants showing columellae in lumina of both apocolpium and mesocolpium region being different from the above types. It has been inferred that the variability in the exine pattern in F2 pollen indicates that the pollen exine pattern in hybrids, perhaps, is not unequivocally controlled by sporophytic or gametophytic genomes.  相似文献

Summary The aim of the experiment was to study the possibility of facilitating the gene transfer and reducing the number of required backcrosses through pollen irradiation and subsequent selection of F1M1 plants containing a very high proportion of sterile pollen as male parent for backcrossing. Anthers of a donor line, C-3-1, were irradiated with 1,500 rad -rays and the pollen used for pollination of a recipient genotype W-8 which posseses a number of recessive marker genes. Five F1M2 plants containing more than 80% sterile pollen grains and one semi-sterile plant were selected and used for backcross to W-8. The segregation pattern of four characters expressed in the first backcross generation [W–8×(W–8×C–3–1)] was assessed and compared with the non-irradiated control. A changed segregation pattern was observed (in some cases even non-transfer of a paternal allele) as well as a shift towards more plants possessing the investigated maternal alleles. A scheme for backcross procedure in combination with pollen irradiation is discussed.  相似文献

We report the thermoluminescence properties of Sr1.96Al2SiO7:Eu0.04 and Sr1.92Al2SiO7:Eu0.04Dy0.04 phosphors. These phosphors were prepared by a high‐temperature solid‐state reaction method. The prepared phosphors were characterized by X‐ray diffraction. A 254 nm source was used for ultraviolet (UV) irradiation and a 60Co source was used for γ‐irradiation. The effect of heating rate and UV‐exposure were examined. The thermoluminescence temperature shifts to higher values with increasing heating rate and thermoluminescence intensity increases with increasing UV exposure time. The trapping parameters such as activation energy (E), order of kinetics and frequency factor (s) were calculated by peak shape method. The effect of γ‐ and UV‐irradiation on thermoluminescence studies was also examined.  相似文献

A series of Ce3+ ion single‐doped Ca2Al2SiO7 phosphors was synthesized by a combustion‐assisted method at an initiating temperature of 600 °C. The samples were annealed at 1100 °C for 3 h and their X‐ray diffraction patterns confirmed a tetragonal structure. The phase structure, particle size, surface morphology and elemental analysis were analyzed using X‐ray diffraction (XRD), transmission electron microscope (TEM), scanning electron microscopy (SEM) and energy dispersive X‐ray (EDX) spectroscopy techniques. Thermoluminescence (TL) intensity increased with increase in ultraviolet (UV) light exposure time up to 15 min. With further increase in the UV irradiation time the TL intensity decreases. The increase in TL intensity indicates that trap concentration increased with UV exposure time. A broad peak at 121 °C suggested the existence of a trapping level. The peak of mechanoluminescence (ML) intensity versus time curve increased linearly with increasing impact velocity of the moving piston. Mechanoluminescence intensity increased with increase in UV irradiation time up to 15 min. Under UV‐irradiation excitation, the TL and ML emission spectra of Ca2Al2SiO7:Ce3+ phosphor showed the characteristic emission of Ce3+ peaking at 400 nm (UV–violet) and originating from the Ce3+ transitions of 5d‐4f (2F5/2 and 2F7/2). The photoluminescence (PL) emission spectra for Ca2Al2SiO7:Ce3+ were similar to the ML/TL emission spectra. The mechanism of ML excitation and the suitability of the Ca2Al2SiO7:Ce3+phosphor for radiation dosimetry are discussed. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

In both Oenothera villaricae Dietrich and O. picensis ssp. picensis, chromosomes are transmitted as two Renner complexes. Reciprocal combinations of the Renner complexes produce eight different F1 hybrids, but only seven are viable. Each species, and each F1 hybrid, produces three sizes of pollen, approximately 50% small sterile grains, 15% medium-sized viable grains and 35% large viable grains. Medium- and large-sized grains were separated manually and subjected to random amplified polymorphic DNAs (RAPDs) analysis. A pattern of RAPD amplifications was obtained which indicates that, for each species and F1 hybrid, one specific Renner complex characterizes the medium- and another the large-sized viable pollen. The results indicate that pollen size is determined in part by the pollen genotype and in part by the genotype of the other pollen grains developing within the same anther.  相似文献

In interspecific pollination of Brassica rapa stigmas with Brassica oleracea pollen grains, pollen tubes cannot penetrate stigma tissues. This trait, called interspecific incompatibility, is similar to self-incompatibility in pollen tube behaviors of rejected pollen grains. Since some B. rapa lines have no interspecific incompatibility, genetic analysis of interspecific incompatibility was performed using two F2 populations. Analysis with an F2 population between an interspecific-incompatible line and a self-compatible cultivar ‘Yellow sarson’ having non-functional alleles of S-locus genes and MLPK, the stigmas of which are compatible with B. oleracea pollen grains, revealed no involvement of the S locus and MLPK in the difference of their interspecific incompatibility phenotypes. In QTL analysis of the strength of interspecific incompatibility, three peaks of LOD scores were found, but their LOD scores were as high as the threshold value, and the variance explained by each QTL was small. QTL analysis using another F2 population derived from selected parents having the highest and lowest levels of interspecific incompatibility revealed five QTLs with high LOD scores, which did not correspond to those found in the former population. The QTL having the highest LOD score was found in linkage group A02. The effect of this QTL on interspecific incompatibility was confirmed by analyzing backcrossed progeny. Based on synteny of this QTL region with Arabidopsis thaliana chromosome 5, a possible candidate gene, which might be involved in interspecific incompatibility, is discussed.  相似文献

Inhaled particle deposition sites must be identified to effectively treat human airway diseases. We have determined distribution patterns of a selected aeroallergen, mulberry pollen, among human extrathoracic (ET: i.e., oronasopharyngeal) regions and the lung. A predictive model validated by inhalation exposure data from human subjects was utilized. Deposition locations were primarily functions of (1) mulberry particle parameters (geometric size, 11–18 μm; shape, spherical; and density, 1.14 g cm?3), and (2) mode of breathing. In the general population, two styles of inhalation are prevalent, normal augmentors (NAs) and mouth breathers (MBs). Their clinical definitions are based on intra-ET airflow divisions. For a NA-mode breathing sedentary (minute ventilation = VE = 10 L min?1) adult, 93% of inhaled mulberry pollen was removed by the ET compartment and 7% collected within the lung. For a MB, the respective deposition efficiencies were 75% and 25%. To apply the model, we used a daily springtime mulberry pollen concentration of 1748 grains m?3 and an exposure time of 0.5 hour to calculate actual doses for the respiratory system. Under the stipulated conditions, a MB would inhale 524 pollen grains per day and 131 would be deposited in the lung; the value is 37 grains for a NA. Preliminary epidemiological results suggest 15% of the study population are MBs in whom such pollen deposits are likely contributors to airway disease.  相似文献

Background and Aims

Previous studies have reported effects of pollen source on the oil concentration of maize (Zea mays) kernels through modifications to both the embryo/kernel ratio and embryo oil concentration. The present study expands upon previous analyses by addressing pollen source effects on the growth of kernel structures (i.e. pericarp, endosperm and embryo), allocation of embryo chemical constituents (i.e. oil, protein, starch and soluble sugars), and the anatomy and histology of the embryos.

Methods

Maize kernels with different oil concentration were obtained from pollinations with two parental genotypes of contrasting oil concentration. The dynamics of the growth of kernel structures and allocation of embryo chemical constituents were analysed during the post-flowering period. Mature kernels were dissected to study the anatomy (embryonic axis and scutellum) and histology [cell number and cell size of the scutellums, presence of sub-cellular structures in scutellum tissue (starch granules, oil and protein bodies)] of the embryos.

Key Results

Plants of all crosses exhibited a similar kernel number and kernel weight. Pollen source modified neither the growth period of kernel structures, nor pericarp growth rate. By contrast, pollen source determined a trade-off between embryo and endosperm growth rates, which impacted on the embryo/kernel ratio of mature kernels. Modifications to the embryo size were mediated by scutellum cell number. Pollen source also affected (P < 0·01) allocation of embryo chemical compounds. Negative correlations among embryo oil concentration and those of starch (r = 0·98, P < 0·01) and soluble sugars (r = 0·95, P < 0·05) were found. Coincidently, embryos with low oil concentration had an increased (P < 0·05–0·10) scutellum cell area occupied by starch granules and fewer oil bodies.

Conclusions

The effects of pollen source on both embryo/kernel ratio and allocation of embryo chemicals seems to be related to the early established sink strength (i.e. sink size and sink activity) of the embryos.Key words: Zea mays, maize, pollen, kernel, embryo, endosperm, oil, protein, starch, soluble sugars  相似文献

Summary We conducted this study during the early spring to demonstrate direct response between increases in atmospheric pollen concentrations and symptom prevalence in a general population. We examined pollen concentrations indoors, outdoors and regionally in 31 households with similar background vegetation, pollen concentration, TSP and pollulant gas levels. Indoor pollen concentrations were low but persistent (X=16 grains/m3 air); local outdoor concentrations were 3 times greater. Regional daily mean pollen values of grasses, ragweed, mulberry and total pollen were compared with symptom scores using X2 contingency tests. We obtained daily symptom scores and measures of peak expiratory flow from 121 individuals characterized as ?normal?, ?atopic? or ?peak flow responsive?. In atopic individuals, prevalence of nasal symptoms increased with pollen concentration increases for ragweed, mulberry and total pollen exposure. No significant response was found with spring grasses whose atmospheric pollen concentration was limited in the selected cluster. Decrease of lung function in the peak flow responsive population was found associated with mulberry pollen only. The small pollen grain size may result in greater tracheo-bronchial deposition.  相似文献

Six plants of a self-incompatible population ofLeavenworthia çrassa were grown from seed collected in nature and cross-pollinated in all combinations. The incompatibility relationships between sibs were determined in eleven of the F1 families. A one-locus sporophytic incompatibility system was established. None of the parents was homozygous at the S locus. At least five, and possibly all six, of the parents did not share an S allele. Only one pair of alleles was shown to have different interactions in the pollen and stigmata. The identity and expression of the S alleles were determined in six families. Eight pairs of alleles were independently expressed in both the pollen grains and the stigmata. Sixteen pairs of alleles showed dominance of one allele over the other in the pollen grains or the stigmata or both.F1 plants of two crosses between different self-incompatible races were self-incompatible. F1 plants of six crosses between self-incompatible and self-compatible races were self-incompatible; in five of the families, the frequency of pseudo-compatibility was higher than in the self-incompatible parent. Self-incompatible hybrids from a cross between a self-incompatible and a self-compatible population provide a method for rapidly determining allelic interactions in plants with a sporophytic incompatibility system.The research was carried out at the Biological Laboratories, Harvard University, Cambridge, Massachusetts, U.S.A.  相似文献

Two mutations were obtained in a two-rowed, spring-type barley variety, Asahi No. 5. KM 114 was obtained in the X2 population of X-irradiated dormant seeds (15 Kr) and KM 218 was found in X2 of gamma-irradiated seeds (15 Kr). KM 114 had very short awns, short plant height, almost normal spike size, and narrow and thinner seeds with pointed tip. The mutant gene is Mendelian recessive. KM 218 plants were normal or slightly taller, had somewhat larger spike, with somewhat longer rachis internodes, medium-long awn, and slightly longer, narrower and thinner kernel than those of the parental variety. At maturity, grains are exposed in many seeds (semi-naked kernels). This mutant also has a conspicuous diagnostic trait; the abnormal development of rachilla into extra florets, some of which set seeds. This mutant gene is also Mendelian recessive. Genes for these two mutants are not allelic and are inherited independently as indicated in F2 and F3 analyses. Genes of these two mutations will serve as good marker genes in genetic and linkage studies in barley, because their character expression is consistent and the recessive homozygotes are viable and fertile.  相似文献

In interspecific crosses in Solanum using certain introductions of Solanum verrucosum as female partners, a certain type of male sterility is observed referred to as tetrad sterility. This type is characterized by clumping of the sterile pollen grains in tetrads. In the same F1 hybrids, the pollen grains show a three-lobed appearance. Both tetrad sterility and lobed appearance of pollen grains are inherited through the interaction of dominant genes (Tr and Ld respectively) with the corresponding sensitive [Trs] and [Lds] plasmons, both plasmons occurring in S. verrucosum. The study of one of the interspecific hybrids led to the detection of duplicate linkage groups including the loci Tr and Ld with a recombination percentage for these loci of 13.7%. The possibility of a basic chromosome number less than 12 in Solanum is discussed in connexion with the occurrence of duplicate loci.  相似文献

Analyzing Rotorod® pollen samples can be time consuming when one uses the standard method of evaluating an entire collector rod. This present investigation explored an abbreviated analysis method which is indicated by Poisson statistical theory. The authors systematically analyzed 18 Rotorod pollen samples from Spring 1994 which contained 408–7394 pollen grains per rod. The atmospheric pollen concentration (pollen grains/m3) was calculated from the number of pollen grains contained on the entire rod surface (Ptotal) and a sub-area containing at least 400 grains (P>400). The estimate of the atmospheric pollen concentration resulting from Ptotal and P>400 for each rod did not vary by more than ±9% (mean, 2.0%±3.7). These data indicate that pollen grains populated the sample rods rather uniformly, suggesting a mode of random recovery from the atmosphere. This study's results are consistent with the expectations concerning a Poisson process and support analyzing collector rods until a threshold number of pollen grains is counted.  相似文献

Outward and inward currents, mainly carried by K+, were detected in protoplasts of pollen grains (PG) and pollen tubes (PT) of Lilium longiflorum Thunb. by using the whole-cell configuration of the patch-clamp technique. The outward K+ current (IK+ out) was similar in both protoplast types, while the inward K+ current (IK+ in) was higher in pollen tube protoplasts. In PT but not in PG protoplasts, inward K+ currents were already detectable at negative membrane voltages usually monitored in lily pollen. IK+ in consisted of a slow and a fast current component, as revealed by fitting a sum of two exponential functions to the time-dependent current. The contribution of the fast component to the total inward current was higher in PT than in PG protoplasts, which was even more evident at acidic pH of the external medium. Therefore, based on the measured characteristics, the IK+ in of PT protoplasts may contribute to the endogenous K+ currents surrounding a growing pollen tube. A genetic linkage map of peach [Prunus persica (L.) Batch] was constructed in order to identify molecular markers linked to economically important agronomic traits that would be particularly useful for long-lived perennial species. An intraspecific F2 population was generated from self-pollinating a single F1 plant from a cross between a flat non-acid peach, ‘Ferjalou Jalousia®’ and an acid round nectarine ‘Fantasia’. Mendelian segregations were observed for 270 markers including four agronomic characters (peach/nectarine, flat/round fruit, acid/non-acid fruit, and pollen sterility) and 1 isoenzyme, 50 RFLP, 92 RAPD, 8 inter-microsatellite amplification (IMA), and 115 amplified fragment length polymorphism (AFLP) markers. Two hundred and forty-nine markers were mapped to 11 linkage groups covering 712 centiMorgans (cM). The average density between pairs of markers is 4.5?cM. For the four agronomic characters studied, molecular markers were identified. This map will be used for the detection of QTL controlling fruit quality in peach and, particularly, the acid and sugar content.  相似文献

The effect of a wide range of temperatures (?15 and 60°C) in darkness or under strong irradiation [1,600 μmol(photon) m?2 s?1] on quantum yield of photosystem II photochemistry and xanthophyll cycle pigments was investigated in a tropical fruit crop (Musa sp.) and a temperate spring flowering plant (Allium ursinum L.). In darkness within the nonlethal thermal window of A. ursinum (from ?6.7 to 47.7°C; 54.5 K) and of Musa sp. (from ?2.2°C to 49.5°C; 51.7 K) maximal quantum yield of PSII photochemistry (Fv/Fm) was fairly unaffected by temperature over more than 40 K. At low temperature Fv/Fm started to drop with ice nucleation but significantly only with initial frost injuries (temperature at 10% frost damage; LT10). The critical high temperature threshold for PSII (Tc) was 43.8°C in A. ursinum and 44.7°C in Musa sp. Under strong irradiation, exposure to temperatures exceeding the growth ones but being still nonlethal caused photoinhibition in both species. Severity of photoinhibition increased with increasing distance to the growth temperature range. ΔF/Fm′ revealed distinctly different optimum temperature ranges: 27–36°C for Musa sp. and 18–27°C for A. ursinum exceeding maximum growth temperature by 2–7 K. In both species only at temperatures > 30°C zeaxanthin increased and violaxanthin decreased significantly. At nonlethal low temperature relative amounts of xanthophylls remained unchanged. At temperatures > 40°C β-carotene increased significantly in both species. In Musa sp. lutein and neoxanthin were significantly increased at 45°C, in A. ursinum lutein remained unchanged, neoxanthin levels decreased in the supraoptimal temperature range. In darkness, Fv/Fm was highly temperature-insensitive in both species. Under strong irradiation, whenever growth temperature was exceeded, photoinhibition occurred with xanthophylls being changed only under supraoptimal temperature conditions as an antiradical defence mechanism.  相似文献

Summary Heritability estimates of five characters of the wheat plant were studied in five crosses involving six cultivars of bread wheat. Parents, F1, F2 and backcrosses to both parents were used in the estimation of the genetic parameters.Heritability was low for number of fertile spikes/plant, moderate for number of spikelets/spike, number of kernels/spike, 1000-kernel weight and moderately high for number of kernels/spike. Evidence for mainly nonadditive gene effects were observed in the expression of number of fertile spikes and 1000-kernel weight. Although nonadditives contributed to a lesser degree to the gene action, additives seemed to be the most important genetic expression regulating number of spikelets/spike, number of kernels/spike, and number of kernels/spikelet. Except for number of fertile spikes/plant, selection in F2-populations seems to be promising.  相似文献
