Nitrate and ammonium uptake by plankton in an Amazon River floodplain lake   总被引:1,自引:0,他引:1  
Uptake of ammonium and nitrate by plankton was measured in tropicalLake Calado, Brazil. Nitrate uptake was strongly influencedby light and was light saturated at {small tilde}340 µEm–2 s–1. In contrast, uptake of ammonium was lessinfluenced by light, and saturated at {small tilde}250 µEm–2 s–1. Uptake rates of both forms of nitrogenwere inhibited by up to 80% at light intensities higher thanthose required for saturation. Concentrations of ammonium andnitrate also had a strong influence on uptake rates. Half-saturationconstants (0.3–5 µM) were usually greater than ambientconcentrations (0.1–0.6 µM), indicating that uptakerates at ambient concentrations were less than one-half of thesaturated rates. Ammonium is the more important type of inorganicnitrogen for plankton of Lake Calado because nitrate concentrationsremain low to undetectable except during periodic inputs ofnitrate-rich water from the Amazon River. The uptake rate of carbon and nitrogen (ammonium, nitrate andurea) by the Microcystis predominating among phytoplankton wasinvestigated in the summer of 1984 in Takahamaira Bay of LakeKasumigaura. The Vmax values of Microcystis for nitrate (0.025–0.046h–1) and ammonium (0.15–0.17 h–1) were considerablyhigher than other natural phytoplankton. The ammonium, nitrateand urea uptake by Microcystis was light dependent and was notinhibited with nigh light intensity. The K1 values were farlower than the Ik values. The carbon uptake was not influencedby nitrogen enrichment. Microcystis accelerated the uptake rateby changing Vmax/K s value when nitrogen versus carbon contentin cells declined. Nitrate was scarcely existent in TakahamairiBay during the summer, when Microcystis usually used ammoniumas the nitrogen source. However, the standing stock of ammoniumin the water was far lower than the daily ammonium uptake rates.Therefore, the ammonium in this water had to be supplied becauseof its rapid turn-over time (–0.7–2.6 h).  相似文献

Nitrate Accumulation and its Relation to Leaf Elongation in Spinach Leaves   总被引:6,自引:0,他引:6  
The leaf elongation rate (LER) of spinach leaves during theday was twice that during the night when grown at a photon fluxdensity of 145 µmol m–2 s–1. All leaves showedthe same LER-pattern over 24 h. Due to low turgor, LER was lowin the afternoon and in the first hours of the night until wateruptake restored full turgor. Osmotic potential remained constantdue to increased nitrate uptake and starch degradation in thisperiod. LER increased to high rates in the second part of thenight and in the morning. The lower rate in the dark comparedto the light was not caused by the lower night temperatures,as increased photon flux density during growth resulted in equalrates in the light and the dark. Increased relative humiditydecreased LER and afternoon rates were most sensitive to waterstress. A ‘low light’ night period did not changeLER-pattern during the night or on the following day. We concludethat nitrate is not an obligatory osmoticum during the nightand can be exchanged for organic osmotica without decreasingLER. During the night the turgor is first restored by increasingwater uptake, nitrate uptake and starch degradation. This resultedin increased leaf fresh weight in this period. Thereafter, elongationincreased by simultaneous uptake of nitrate and water. Nitrateconcentration was, therefore, constant in the older leaves.In the younger leaves nitrate concentration increased to replacesoluble carbohydrates. We examined the effect of pretreatments (18 h at 5 µmoldm–3) with abscisic acid, the ethylene-releasing substance‘Ethephon’, gibberellic acid, indoleacetic acid,kinetin and zeatin on nitrate uptake and in vivo nitrate reductaseactivity (NRA) in roots of nitrogen-depleted Phaseolus vulgarisL. Nitrate uptake showed an apparent induction pattern witha steady state after about 6 h, in all treatments. The nitrateuptake rate after 6 h was unaffected or at most 30% lower aftertreatments with the plant growth regulators. Gibberellic acid, kinetin and zeatin induced substantial NRAin roots in the absence of nitrate, whereas Ethephon enhancedNRA only during nitrate nutrition. Kinetin-induced NRA (Ki-NRA)was maximal after a pretreatment at 1 µmol dm–3,and showed a lag phase of 6–8 h. Ki-NRA was additive tonitrate-induced NRA (NO3-NRA) for at least 24 h, independentof the induction sequence. After full induction, Ki-NRA approximated20% of NO-3-NRA. Abscisic acid counteracted the developmentof Ki-NRA, but not of NO3-NRA. Cycloheximide and tungstatewere equally effective to suppress the development of nitratereductase activity after supply of kinetin or NO3. Our data are consistent with the operation of two independentenzyme fractions (Ki-NRA and NO3-NRA) with apparentlyidentical properties but with separate control mechanisms. Theabsence of major effects of plant growth regulators on the time-courseand rate of nitrate uptake suggests that exogenous regulators,and possibly endogenous phytohormones are of minor importancefor initial nitrate uptake. The differential effect of someregulators on nitrate uptake and root NRA furthermore indicatesthat the processes of uptake and reduction of NO3 arenot obligatory or exclusively coupled to each other.  相似文献

Nitrate provision has been found to regulate the capacity forChara corallina cells to take up nitrate. When nitrate was suppliedto N sufficient cells maximum nitrate uptake was reached after8 h. Prolonged treatment of the cells in the absence of N alsoresulted in the apparent ability of these cells to take up nitrate.Chlorate was found to substitute partially for nitrate in the‘induction’ step. The effects on nitrate reductionwere separated from those on nitrate uptake by experiments usingtungstate. Tungstate pretreatment had no effect on NO3uptake ‘induced’ by N starvation, but inhibitedNO3 uptake associated with NO3 pretreatment. Chloridepretreatment similarly had no effect on NO3 uptake ‘induced’by N deprivation, but inhibited NO3 uptake followingNO3 pretreatment. The data suggest that there are atleast two mechanisms responsible for the ‘induction’of nitrate uptake by Chara cells, one associated with NO3reduction and ‘induced’ by CIO3 or NO3and one associated with N deprivation. Assimilatory nitrate reductase (NR) was solubilized by acetonetreatment from Plectonema boryanum and was purified 7,700-foldby heat treatment, ammonium sulfate fractionation and chromatographyon DEAE-Sephacel and Sephadex G-150. Purified NR had a specificactivity of 85 µmol NO2 formed min–1 mg–1protein. The enzyme retained both ferredoxin (Fd)- and methylviologen (MV)-linked NR activities throughout the purificationprocedure. Molecular weight was 80,000. The pH optimum was 10.5in the MV-assay and 8.5 when assayed with enzymatically reducedFd as the electron donor. Apparent Km values for nitrate andMV were 700 µM and 2,500µM in the MVassay and 55µM and 75 µM for nitrate and Fd in the Fd-assay.The enzyme was inhibited by thiol reagents and metal-chelatingreagents. Phytoplankton biomass, primary production rates and inorganicnutrients were measured in the uppermost layer of the ice-edgeregion and in open water and compared with environmental factorsduring a three-week cruise in September – October 1979.Biomass and production values were low (maximum 2.2 µgchl a l–1, 2.5 mg C m–3 h–1). A post-bloomcommunity of diatoms, consisting mainly of representatives ofChaetoceros, Leptocylindrus, Nitzschia and Thalassiosira, waspredominant. Concentrations of phosphate were quite low (maximum0.55 µM I–1). Nitrate and silicate ranged from nomeasurable quantities to 5.7 µM l–1 and 3.8 µMl–1, respectively. The possibility of light and nutrientlimitation on phytoplankton growth is discussed.  相似文献

Spruce (Picea abies (L.) Karst.) seedlings were asepticallycultivated and the effects of different N-nutrition on net uptakeand reduction of nitrate were investigated. The characteristicsof nitrate uptake were calculated, Ks as 0?2 mol m–3 andVmax as 18 µmol g–1 d–1. Low pH, and Al3+ in the medium caused adecrease in nitrate uptake rate. An in vivo assay was set upwhich allowed the measurement of NRA in both roots and needlesof spruce seedlings. The in vivo nitrate reductase activitywas repressed by ammonium and stimulated by nitrate. Nitratereduction was similar to nitrate uptake, negatively affectedby low pH and ammonium. A comparison was made between two methods of measuring the relationshipbetween the external [K+] and the flux of K+ into whole plantsof Lolium perenne and Raphanus sativus. The values of flux obtainedfrom solutions of 1.2 µM K+ held constant around the rootswere three and six times greater for Lolium and Raphanus respectivelythan the values obtained at the same concentration in a depletionexperiment in which the solutions, initially 100 µM K+,were depleted to below 1.2 µM K+ by plant uptake. In thedepletion experiment with Lolium, the flux was higher into plantsgrown at low [K+] than into plants grown at 100 µM eventhough [K+] within the plant was about the same for all groupsof plants. It is suggested that Lolium grown at low [K+] hasan efficient mechanism for K+ uptake which continues to operatefor some time after the plants have been transferred to a higherconcentration. With both species, Km was 15–20 µMin the depletion experiment and below 1 µM when concentrationswere held constant.  相似文献

All three races of Neotricula aperta, an epilithic, schistosometransmitting, snail of the Mekong and Mul rivers of NortheastThailand and southern Laos, were found to take up acetate froma dilute solution. After 48 h incubation the mean specific netuptake rates (µmol–1 g–1 h–1), from750 µM acetate, were: 1.86, -race; 1.39, ß-raceand 3.25, y-race. Over 48 h the snails were able to achievereductions in the ambient acetate concentration of up to 60%.Incubations under bacteriostatis suggested that bacteria arenot involved in the uptake of acetate by N. aperta. The uptakecharacteristics conform to the Michaelis-Menten model. The transportconstants, Jmax (µmol–1 g–1 h–1) andKt (µM) were 1.10 and 102 respectively (-race). Racialdifferences in uptake characteristics are discussed in relationto micro-habitat differences. HPLC indicated concentrations of acetate in y-N. aperta microhabitatsof around 325 µM. This suggests a pool size sufficientto satisfy only 6% of the snail's basal metabolic rate (BMR).Levels within the epilithic aufwuchs, however, are probablyhigh enough to provide for more than 50% of the BMR. The possible role of acetate in the energy metabolism of N.aperta is discussed. Short-chain carboxylic acids (such as acetate),arising from the decomposition of the aufwuchs, could representsources of fermentable organics that may be taken up by N. apertasnails and used to supplement their nutrition during times offood shortage. However, further investigations involving 14C-labellingtechniques are required. The findings of this investigationhave implications for the chemical ecology and life-cycle ofN. aperta. The uptake of [3H]-abscisic acid in barley (Hordeum vulgareL. cv. Heartland) cell cultures was found to be mediated throughboth non-saturable and saturable components. The kinetic parametersof the saturable component, determined at pH 4.5 and 21 °C,showed a Km for natural or (+ )-ABA of 1.3±0.7µMand an apparent Vmex of 7.0 ± 2.8 nmol g–1 cellsh–1. The carrier showed a strong preference for the naturalenantiomer of ABA as compared to the unnatural one. Other substancestested, e.g. amino acids, organic acids, and other growth regulators,did not appear to interfere with the carrier-mediated uptakeof ABA. At low external concentrations of ABA (below 2.0 µM),the saturable component was greater than the diffusion component.Similarly, between pH 4.0 and 6.0, the saturable uptake wasresponsible for more than 50% of the total uptake. The carriermay be important in vivo for mediating uptake when endogenouslevels of ABA are low (c. 1 µM). The carrier specificity was evident in inhibition experimentsdone with ABA analogues. Our data showed that the carrier couldaccommodate small modifications in the ABA structure. Four analogueswere able to compete efficiently with ( + )-ABA for the bindingsite of the carrier. Three of these competitors were of the(+)-series. Only one ( –)-analogue, (–)-ABA, wasable to inhibit markedly the saturable uptake of ( + )-ABA.The induction of the ABA-respons-ive gene WCS120 (Houde et al.,1992) presented stricter requirements for the ABA molecule thanthe carrier, although with a similar preference for the ( +)-analogues. Besides ( + )-ABA itself, only two of the analoguestested, both ( + )-series, were able to induce the WCS120 geneafter a 24 h incubation period. The absence of correlation betweenthe activity of the analogues as ABA inhibitors in the carriersystem, and their capacity to induce the WCS120 gene tend tosuggest that the carrier is not directly involved in the signaltransduction pathway leading to the induction of this specificgene. Dark uptake of inorganic 14C by offshore plankton was measuredat two depths at 36 stations in the Atlantic Ocean from 52°Sto 26°N, mainly along 30°W. The samples were incubatedfor 2 h with and without inhibition of biological activity withHgCl2. In addition, six time course experiments were performed.The mean dark uptake rate varied from 0.68 to 4.82 (µmolC m–3 h–1 over the transect and showed a significantpositive relationship with chlorophyll a. The dark uptake wasusually >5% of the maximum photosynthetic capacity (Pm),and higher values relative to Pm were associated with low valuesof Pm and not with high absolute dark values. A linear relationshipbetween dark uptake and Pm was found with a background value(y-axis intercept) of 0.51 (µmol C m–3 h–1and a slope of 0.77% of Pm. A major fraction of the dark signal,66–80% of the total signal, persisted in bottles treatedwith HgCl2, indicating that most of the dark signal was independentof biological activity. Time course experiments showed a lineardark uptake with time for the first hours, whereafter the uptakeceased. At stations with low concentrations of inorganic nitrogen[>1 (µmol (NH4++NO3)], a second stage was observedafter 3–8 h, probably due to an increase in bacterialactivity. The results suggest three mechanisms for the darkvalue in short-term incubations in oligotrophic waters. A backgroundvalue independent of biomass and incubation time which was thedominant part of the dark signal in samples with very low phytoplanktonbiomass (>0.3 p-g Chi a 1"). Another important part was residualsof 14C associated with plankton, probably adsorbed to compoundsinside the cells. This fraction was dominant in short-term incubationsat chlorophyll concentrations >0.3 p.g Chi a H. Active uptakeby living cells (total minus ‘HgCl2 uptake‘) wasonly a minor part of the dark signal in short-term incubations,but dominated at longer incubation time (>3–9 h), probablydriven by an increase in bacterial activity. A significant enhancementof the non-photosynthetic uptake of 14C was observed in light,probably associated with a carbon-concentrating mechanism inphytoplankton or light stimulation of ß-carboxylationactivity. The results strongly suggest that dark values shouldbe subtracted from the light uptake. This correction is particularlyimportant when photosynthetic rates are low, e.g. at low lightor in short-term incubations where a time-zero background becomesa significant part of the total uptake in light. Exponentially growing cultures of Nannochloropsis sp. were transferredfrom high light (650 µ.mol quanta m–2 s–1)to low light (30 (µrnol quanta m–2 s–1). Thekinetics of changes in the various parameters of the photoacclimationprocess were followed until steady state was reached. In thisorganism, the observed 4.25 increase in cellular chlorophylla in the low-light-acclimated cells was linearly correlatedwith an increase in the number of photosynthetic units (PSU),while the PSU size remained constant. The new steady state wasreached after a transition period of 87 h and following an initialovershoot in the chlorophyll a concentration. During the processof photoacclimation, harvested and utilized light was increaseddue to a combination of increased cellular chlorophyll and anincrease of the quantum yield. These improvements in efficiencywere coupled with the reduction of respiration. The overalleffect of photoacclimation, therefore, amounted to reducingthe impact of low light on the growth rate of the algae. Ourstudy demonstrates the importance of the photoacclimation processin the context of the planktonic way of life. An index of the'effectiveness of photoacclimation' is proposed.  相似文献

Spinach plants were grown in bowls of aerated nutrient solutionin a controlled environment chamber for 24 h, and harvestedevery 3·5-5 h to record their growth, nitrate and wateruptake, and plant nitrate concentration. Twelve such experimentsare described, either with a 14/10 h dark/light regime, or continuouslight or darkness. The irradiance was either 110, 320, or 510µmol m-2 s-1 (PPFD). All these regimes began at the endof the light period of a 14/10 h dark/light regime (510 µmolm-2 s-1) lasting approximately 2 weeks. Nitrate uptake rate per g of dry weight of plant continued almostunabated at about 17 µmol h-1 through the initial 14-hdark period, and then fell away sharply if the light was notrestored, but increased slightly when it was. With continuouslight at 510 µmol m-2 s-1, uptake rate rose steadily forthe first 24 h of light, and then fell sharply for about 6 h.Shoot nitrate concentration increased about three-fold in thedark phase, and declined in the light at a rate which was positivelyrelated to the irradiance. Root nitrate concentration was severaltimes higher than that of the shoot: its diurnal change wassmaller (relative to the mean) than that of the shoot. Nitratereduction occurred to a small extent in the dark, and increasedrapidly as soon as the lights came on, to remain at a roughlyconstant rate (related to the irradiance) throughout the lightphase. Dry matter increase in the light was related to irradiance,but with little increase above 320 µmol m-2 s-1. Respiratoryweight loss in the dark was not detectable. Rate of fresh weightincrease was approximately constant throughout light and darkperiods. Plants of Italian ryegrass (Lolium multiflorum Lam.) grown insoil with two rates of added 15N-labelled nitrate were exposed,in chambers, for 40 d to NH3 in the air at concentrations of16, 118 and 520 µg m–3. At the highest concentrationof NH3, this source provided 47?3% of the total nitrogen inplants grown with the lower rate of nitrate addition (100mgN kg–1 dry soil) and 35?2% with the higher rate (200mgN kg–1 dry soil) At the intermediate concentration ofNH3, the contributions to total plant N were 19?6% and 10?8%,respectively, at low and high nitrate while, at the lowest concentrationof NH3, they were 5?1% and 32%. Most of the N derived from theNH3 remained in the leaves, but some was transported to theroots. The amount of N derived from the NH3 that was presentin the leaves was not reduced by washing the leaves in waterat pH 5?0 before harvesting, indicating that the N was assimilatedby the plant and not adsorbed superficially. Rates of uptakeof NH3 per unit leaf area ranged from 1?7 µg dm–2h–1 at a concentration of 16 µg m–3 to 29?0µg dm–2 h–1 at a concentration of 520 µgm–3 and with the lower rate of nitrate addition. Increasingthe supply of nitrate to the roots slightly reduced the rateof uptake of NH3 per unit leaf area. The bloom-forming marine dinoflagellate Gyrodinium cf. aureolumwas grown in batch cultures over a range of irradiances (35–380µmolm–2 s–1 and growth, photosynthesis and respirationrates determined. Saturation of growth occurred at irradiancesof 100µmol m–2 s–1 Below this light level,decreases in growth rates and cell size, and a relative increasein carbon specific respiration rates, were observed. On theother hand, photosynthesis-irradiance relationships determinedfrom dissolved oxygen incubations showed that on a cellularand carbon basis, cultures grown at low irradiances had higherrates of light-limited and light-saturated photosynthesis, mainlyas a result of large increases in cell chlorophyll content.This adaptation strategy enables low-light-grown organisms toexploit available high irradiance through a relatively highphotosynthetic capacity. In cells grown at higher light levels(>100µmol m–2 s–1), excess photosynthatemay be diverted to storage rather than used for growth.  相似文献

The growth of four heathland species, two grasses (D. flexuosa,M. caerulea) and two dwarf shrubs (C. vulgaris, E. tetralix),was tested in solution culture at pH 4.0 with 2 mol m–3N, varying the N03/NH4+ ratio up to 40% nitrate. In addition,measurements of NRA, plant chemical composition, and biomassallocation were carried out on a complete N03/NH4+ replacementseries up to 100% nitrate. With the exception of M. caerulea, the partial replacement ofNH4+ by NO3 tended to enhance the plant's growth ratewhen compared to NH4+ only. In contrast to the other species,D. flexuosa showed a very flexible response in biomass allocation:a gradual increase in the root weight ratio (RWR) with NO3increasing from 0 to 100%. In the presence of NH4+, grassesreduced nitrate in the shoot only; roots did not become involvedin the reduction of nitrate until zero ambient NH4+. The dwarfshrubs, being species that assimilate N exclusively in theirroots, displayed an enhanced root NRA in the presence of nitrate;in contrast to the steady increase with increasing NO3in Calluna roots, enzyme activity in Erica roots followed arather irregular pattern. Free nitrate accumulated in the tissuesof grasses only, and particularly in D. flexuosa. The relative uptake ratio for NO3 [(proportion of nitratein N uptake)/(proportion of nitrate in N supply)] was lowestin M. caerulea and highest in D. flexuosa. Whereas M. caeruleaand the dwarf shrubs always absorbed ammonium highly preferentially(relative uptake ratio for NO3 <0.20), D. flexuosashowed a strong preference for NO3 at low external nitrate(the relative uptake ratio for N03 reaching a value of2.0 at 10% NO3). The ecological significance of thisprominent high preference for NO3 at low NO3/NH4+ratio by D. flexuosa and its consequences for soil acidificationare briefly discussed. The contribution of nanoplankton (< 10 µm fraction)to winter – spring (1977 – 78) and summer (1978,1979) phytoplankton nitrogen dynamics in lower NarragansettBay was estimated from ammonium, nitrate and urea uptake ratesmeasured by 15N tracer methods. During the winter – spring,an average of 80% of chlorophyll a and nitrogen uptake was associatedwith phytoplankton retained by a 10 µm screen. In contrast,means of 51 – 58% of the summer chlorophyll a standingcrops and 64 – 70% of nitrogen uptake were associatedwith cells passing a 10 µm screen. Specific uptake ratesof winter – spring nanoplankton populations were consistentlylower than those of the total population. Specific uptake ratesof fractionated and unfractionated summer populations were notsignificantly different. Ammonium uptake averaged between 50and 67% of the total nitrogen uptake for both the total populationand the < 10µm fraction. The total population and the10 µm fraction displayed similar preferences for individualnitrogen species. Nutrient-saturated cultures of Skeletonema costatum were grownat 15C and 42 combinations of photon flux density (PFD) anddaylength. The growth rate increased with the daylength andPFD up to 460–630 µE m–2 s–1 (maximum2.5 doublings day At 2000 µE m–2 s–1 the growthrate was reduced by 45%. The chlorophyll (chl) content of thecells and the rate of production of carbon per unit chlorophylland ambient light increased for declining light regimes as didcellular nitrogen and carbon. The N/C ratio, cellular phosphorusand ratios between in vivo fluorescence, with and without DCMU,and chlorophyll varied negligibly. The ATP/C ratio was linearlyrelated to the growth rate. The results were described mathematically.The chl/C ratio was low both in strong light and in marginallylow light, corresponding to low cellular chlorophyll and highcellular carbon, respectively. The observed increase in cellularnitrogen and carbon at shade adaptation probably represent anincrease in the size of internal stores of organic nitrogenand may imply that Skeletonema cells become enriched with organicnitrogen when staying in nitrate-rich subsurface layers, e.g.in or below a nutricline. However, close to zero growth in marginallight the cells become greatly enriched with respect to everymeasured factor. Such cells may be physiologically resting stageswhich may ensure survival during dark periods and promote rapiddevelopment during the initial phase of blooms. The response of phytoplankton to variations in the light regimewas studied during the VULCAN and ACDA cruises in the Antarctic.Unenriched batch cultures of 12–19 days' duration reachedchl concentrations of 10–50 µg–1 and exhibitedexponential growth rates, with the maximal rate being 0.41 doubl,day–1. Ice edge algae exhibited maximum growth rates atphoton flux densities (PFD) of 30–100 µE m–2S–1and the growth rate was reduced by about 30% at 500–1000µE m–2S–1 The chl/C ratio ranged between 0.004and 0.018, with the lowest ratios at PFDs above 500 µEm–2S–1 chl/C ratios were also below maximum at PFDsbelow 40–50 µE m–2S–1 The C:N:P ratioswere close to the Redfield ratios; the Si/C ratio averaged 0.16(atoms), and the ATP/C ratio averaged from 0.0024 to 0.0050in different culture senes. When thawed after having been frozenfor 10 days, shade-adapted cultures were in a much better conditionthan sun-adapted ones. P versus I data showed that the maximumassimilation number varied from 0.75 to 4.4 µg C (µgchl)–1h–1. It varied inversely with the chl/C ratio;therefore the maximum carbon turnover rate varied little betweensamples (0.024/0.035 h–1). Low biomass communities exhibitedrelatively high values for (the initial slope of P versus Icurves), low values for 1sat (160–330 µE m–2S–1),and they were susceptible to photoinhibition. In contrast, communitiesdominated by Odontella weissflogii exhibited low values for, a high value for Isat (560 µE m–2S–1 andthey tolerated high PFDs. The photo-adaptational status of thephytoplankton in natural water samples is discussed relativeto the profile of water column stability and mixing processes.  相似文献
