Identification of superior parents in a potato breeding programme   总被引:6,自引:0,他引:6  
Summary An incomplete diallel cross was used to study components of genetic variation in potatoes for a range of characters after early and late harvest. The progenies were also used to evaluate five predictors of progeny performance, namely the mean seedling performance, the mid-parent value and the means of the selfed progenies, of the diploid progenies and of the test-cross progenies. For almost all characters, the general combining ability effects were predominant, although the specific combining ability effects present were greater at late than at early harvests. The seedling performance for tuber yield, number of tubers and average tuber weight did not show any relevant relationship to the field performance. The midparent value provided, in general, satisfactory predictions of the mean progeny performance obtained in the diallel, except for ware tuber yield. The selfed and the diploid progenies did not improve the prediction of progeny means compared to the mid-parent value. The predictions based on the test-crosses surpassed those of the mid-parent value, particularly for tuber yield at ware potato harvest. Methods to identify superior parents are discussed.  相似文献

Three somaclonal populations of potato (Solanum tuberosum L.), each comprised of at least 1,000 plants, were regenerated from the cultivars Kennebec, Russet Burbank, and Superior. The frequency of formation of adventitious meristems from tuber disc explants varied significantly between these potato genotypes. Only 1.0–1.3% of each somaclonal population exhibited morphological aberrations. Regenerated populations of Kennebec and Superior, when compared to respective control populations over three asexual generations, were similarly enriched with somaclones having more elongated tubers, a higher total tuber number and weight, a higher cull tuber number and weight, and earlier maturity. Somaclones of Russet Burbank also produced more elongated tubers, a higher total tuber number, and a higher cull tuber number and weight but, in contrast, these somaclones were lower in total tuber weight, lower in U.S. 1 tuber number and weight, shorter in stem length, and lower in vigor. Of the three cultivars, Russet Burbank somaclones possessed the greatest variability for most traits. Besides this significant genotype effect, quantitative traits differed amongst each other in respect of relative changes resulting from somaclonal variation. Summary Most previous studies on cross prediction methods have examined relatively few crosses, particularly in relation to the numbers involved in most breeding programmes. In this paper the feasibility of using cross prediction methods was examined in a practical potato (Solanum tuberosum) breeding scheme by the analyses of progeny from 52 crosses. The variate considered was breeder's preference, a visual assessment made of the harvested tubers to estimate their commercial potential. The results showed that it was possible to identify the superior crosses. Cross prediction based simply on the mean preference scores, averaged over scorers and clones within progenies, estimated on seedlings or first clonal year plants, provided the best estimate of a progeny's performance in the third clonal generation. Predictions based on the expected proportion of clones that would transgress a given target value also provided a good indication of a progeny's potential. The poorest prediction was obtained by using the observed frequency of desirable clones in a progeny sample. The implications for potato breeding are discussed.  相似文献

A diallel set of crosses, including selfs and some reciprocal crosses, was made between 15 parents chosen for their male fertility from those included in a tetraploid potato (Solanum tuberosum subsp. tuberosum) breeding programme at the Scottish Crop Research Institute. Seedling progeny tests were used to evaluate the progenies for non-race-specific resistance to late blight (Phytophthora infestans) in both foliage and tubers, quantitative resistance to the white potato cystnematode (PCN) (Globodera pallida) and the commercial worth of their tubers as judged by breeders' visual preference. No reciprocal differences were found. Comparisons of the selfs and crosses revealed inbreeding depression for breeders' preference, which varied among the parents from negligible to severe, whilst there were also statistically significant differences for foliage and tuber blight, but not for PCN. When the selfs were omitted from the combining ability analyses, large differences in general combining ability (GCA) were found for all four traits, and smaller differences in specific combining ability for tuber blight and breeders' preference. The only statistically significant correlation between GCAs for different traits was a favourable one of r = 0.56 between foliage and tuber resistance to late blight. It was concluded that prospects were good for simultaneously improving all four traits by multitrait genotypic recurrent selection.  相似文献

The presence of algae within the cases ofAgapetus fuscipes was investigated. Cases recognised as dirty or clean with the naked eye had more and less algal growth, respectively. Larvae in the former survived significantly longer when starved in the laboratory. It is suggested that the presence of algae within the cases would be of ecological advantage during periods of flood.  相似文献

A radioimmunoassay, combined with high-performance liquid chromatography, has been used to analyse the zeatin-type cytokinins of potato (Solanum tuberosum L. cv. Majestic) tubers and tuber buds throughout growth and storage. During tuber growth, zeatin riboside was the predominant cytokinin detected in all tissues. Immediately after harvest, the total cytokinin concentration fell dramatically in the storage tissue, largely as a consequence of the disappearance of zeatin riboside. During storage, levels of cytokinins in the storage tissue remained relatively constant, but increased in the tuber buds. In the buds of tubers stored at 2°C there was a 20-to 50-fold increase in total cytokinin over six weeks, coinciding with the natural break of innate dormancy. At 10°C the rise in the level of bud cytokinins was slower, correlating with the longer duration of innate dormancy. Injecting unlabelled cytokinins into tubers in amounts known to induce sprouting gave rise to increases in cytokinin concentrations in the buds of the same order as the increase associated with the natural break of dormancy. Metabolism of injected cytokinins was greater in non-dormant than in dormant tubers. The restriction landmark genomic scanning (RLGS) method was applied to rice, using two Japanese cultivars, Nipponbare and Koshihikari, and a Chinese landrace, LiuZhou'Bao'Ya'Zao'. More than 3000 landmarks were detected as spots on the individual autoradiograms of each cultivar. Nipponbare and LiuZhou' Bao'Ya'Zao' showed apparently different RLGS profiles, from which the genetic similarity (GS) between them was estimated as 0.344. Although the two Japanese cultivars, Nipponbare and Koshihikari showed quite similar RLGS profiles, they were easily distinguished on the basis of the presence or absence of specific spots; the GS value between them was calculated as 0.980. The RLGS method is shown to be a powerful fingerprinting technique, especially for the classification and identification of cultivars in rice and probably in other crops as well.  相似文献

Summary The molecular basis of a lectin gene mutation in the Phaseolus vulgaris cultivar Pinto was investigated. Rocket immunoelectrophoresis studies showed that seed lectin is reduced approximately 40-fold in comparison to the normal Phaseolus vulgaris cultivar Contender. DNA gel blot studies using a lectin cDNA probe suggested that the Pinto and Contender varieties contain similar numbers of lectin genes, although qualitative differences were observed in the gel blot banding patterns. Hybridization of the lectin cDNA probe to gel blots containing normal and mutant embryo mRNAs produced a 1 kb mRNA band in both mRNA populations. However, the amount of lectin mRNA was reduced approximately tenfold in the mutant Pinto cultivar. Progenies from first-generation self, half-sib, full-sib, and cross fertilizations were generated to evaluate the magnitude of inbreeding depression for vegetative and production traits in strawberry. Tests were conducted to determine the linearity of trait mean depression with inbreeding rate (F) over this range of inbreeding values, as an indication of the presence of non-additive epistasis. A control population, for which a similar range of coancestry had accumulated over several cycles of breeding and selection, was also generated to compare the consequences of ancestral and current-generation inbreeding. Trait means for crosses among current-generation half-sibs, full-sibs, and selfs were 2–17%, 3–12%, and 14–45% lower than for unrelated crosses among the same set of parents, respectively. Linear regression of progeny means on current generation F was significantly negative for all traits and explained 17–44% of the variance among progeny means. Mean depression was largely linear over the range of inbreeding rates tested in this population, indicating the absence of epistasis for the traits evaluated. Conversely, (F) regressions of progeny means on pedigree inbreeding coefficients, where coancestry had accumulated over several cycles of breeding and selection, were uniformly non-significant and explained 0–10% of the variance among cross means. Further, multiple regression of progeny means for current-generation relatives on pedigree F failed to improve fit significantly over regression on current-generation F alone for all traits. Together, these results suggest that pedigree inbreeding coefficients are poor predictors of changes in homozygosity when populations are developed through multiple cycles of breeding and selection. They also imply that inbreeding depression will be of minor importance for strawberry breeding populations managed with adequate population sizes and strong directional selection.  相似文献

A triple resonance pulse scheme is presented for recording 13C-1H one-bond dipolar couplings in 15N, 13C labeled proteins. HNCO correlation maps are generated where the carbonyl chemical shift is modulated by the 13C-1H coupling, with the two doublet components separated into individual data sets. The experiment makes use of recently described methodology whereby the protein of interest is dissolved in a dilute solution of bicelles which orient above a critical temperature, thus permitting measurement of significant couplings (Tjandra and Bax, 1997a). Summary The distributive properties of a single population or of a population resulting from a cross between two populations are reproduced when inbreds randomly extracted from the population itself or from the two parental populations are randomly paired. Hence, population parameters that are usually obtained during a breeding programme can be used to predict the performance of the F1 hybrids that can be derived from them at that stage. Multiple allelism, epistasis and deviations from Hardy-Weinberg equilibria should not cause biasis to the predictions. While in theory genotype x environment interaction and linkage disequilibrium may disturb the predictions, in practice they are unlikely to create problems that cannot be accommodated. Genotypic and phenotypic predictions of the proportions of the F1 hybrid distribution scoring above or below a given standard are made and analysed for three characters, weight of the ears, plant height and height of the ear, in two populations of maize per se and their interpopulational cross. Because no random inbred lines from the experimental populations are presently available we cannot check our predictions. However, genotypic and phenotypic predictions and observations of F1 hybrids obtained from populations created by computer simulation are provided to illustrate our procedures.J. F. Summary The transmission of variation from protoplast-derived plants of tetraploid potato cultivar Bintje to tuber progeny was examined. The morphological alterations of a majority of the variant protoclones were transmitted to corresponding tuber progeny. Some of the normal and variant protoclones gave new phenotypes, or segregated into parental and new phenotypes after vegetative propagation. The ploidy levels of almost all these clones remained unchanged after propagation. It was concluded that the occurrence of variation after vegetative propagation was due to somatic segregation of chimeras resulting from gene mutations or chromosome structural rearrangements in only part of the regenerated plant. The origin of variation is discussed in the light of these results.  相似文献

Summary Mitochondrial DNAs (mtDNA) from four stable revertant strains generated from high frequency petite forming strains of Saccharomyces cerevisiae have been shown to contain deletions which have eliminated intergenic sequences encompassing ori1, ori2 and ori7. The deleted sequences are dispensable for expression of the respiratory phenotype and mutant strains exhibit the same relative amount of mtDNA per cell as the wild-type (wt) parental strain. These deletion mutants were also used to study the influence of particular intergenic sequences on the transmission of closely linked mitochondrial loci. When the mutant strains were crossed with the parental wt strains, there was a strong bias towards the transmission into the progeny of mitochondrial genomes lacking the intergenic deletions. The deficiency in the transmission of the mutant regions was not a simple function of deletion length and varied between different loci. In crosses between mutant strains which had non-overlapping deletions, wt mtDNA molecules were formed by recombination. The wt recombinants were present at high frequencies among the progeny of such crosses, but recombinants containing both deletions were not detected at all. The results indicate that mitochondrial genomes can be selectively transmitted to progeny and that two particular intergenic regions positively influence transmission. Summary The objective of this study was to separate and determine effects on the field performance of transgenic potatoes that originate from the tissue culture process of transformation and from the genes inserted. The constructs introduced contained the reporter gene for betaglucuronidase (GUS) under the control of the patatin promoter (four different constructs) and the neomycin phosphotransferase gene under the control of the nopaline synthase promoter. Both genes might be expected to have a neutral effect on plant phenotype. The field performance of transgenic plants (70 independent transformants) was compared with non-transgenic plants regenerated from tuber discs by adventitious shoot formation and from shoot cultures established from tuber nodal cuttings. Plants from all three treatments were grown in a field trial from previously field-grown tubers, and plant performance was measured in terms of plant height at flowering, weight of tubers, number of tubers, weight of large tubers and number of large tubers. There was evidence of somaclonal variation among the transgenic plants; mean values for all characters were significantly lower and variances generally higher than from plants derived from nodal shoot cultures. A similar change in means and variances was observed for the non-transgenic tuber-disc regenerants when compared with shoot culture plants. Plant height, tuber weight and tuber number were, however, significantly lower in transgenic plants than in tuber-disc regenerants, suggesting an effect on plant performance either of the tissue culture process used for transformation or of the genes inserted. There were significant differences between constructs for all five plant characters. The construct with the smallest segment of patatin promoter and the lowest level of tuber specificity for GUS expression had the lowest values for all five characters. It is proposed that the nature of GUS expression is influencing plant performance. There was no indication that the NPTII gene, used widely in plant transformation, has any substantial effect on plant performance in the field.  相似文献

The aim of this work was to discover whether genetic manipulation of 6-phosphofructokinase [EC 2.7.1.11; PFK(ATP)] influenced the rate of respiration of tuber tissue of Solanum tuberosum L. Transgenic plants were produced that contained the coding sequence of the Escherichia coli pfkA gene linked to a patatin promoter. Expression of this chimaeric gene in tubers resulted in a 14to 21-fold increase in the maximum catalytic activity of PFK(ATP) without affecting the activities of the other glycolytic enzymes. Tubers, and aged disks of tuber tissue, from transformed plants showed no more than a 30% fall in the content of hexose 6-monophosphates; the other intermediates of glycolysis increased threeto eightfold. Fructose-2,6-bisphosphate was barely detectable in aged disks of transformed tubers. The relative rates of 14CO2 production from [1-14C]-and [6-14C]-glucose supplied to disks of transformed and control tubers were similar. Oxygen uptake and CO2 production by aged disks of transformed tubers did not differ significantly from those from control tubers. The same was true of CO2 production, in air, and in nitrogen, for tuber tissue. Summary Genetic control of tiller number, grain number, grain weight, harvest index and grain yield in six generations, along with the biparentals, F3s, F2xparental progeny, and F2xF1 progeny were investigated in an intervarietal cross of bread wheat involving two highly competitive varieties, WL711 and HD 2009. The performance of F1, B1, B2, F2, × p1, F2 × P2 and F2 × F1 progeny was midway between the parents involved with respect to all the evaluated characters. The biparental progeny excelled the mean performance of their corresponding F2 and F3 progeny in tiller number, seed weight and grain yield. The estimates of variance components obtained from the two models deployed were almost similar. Considerable additive genetic variance was observed for grains per spike, seed weight and grain yield while dominance variance was more pronounced for harvest index. The additive-dominance model was adequate for grains per spike and harvest index. Epistatic effects of additive × additive and additive × dominance type for tiller number and grain yield, and of additive × dominance type for seed weight were observed. The digenic epistatic model was inadequate for explaining the nature of gene action for tiller number, seed weight and grain yield. The studies indicated that non-allelic interactions should not be ignored in formulating wheat breeding programmes and that a biparental approach could be adopted as an extremely useful tool for enhancing genetic variability and the creation of transgressive segregants. The usefulness of breeding methodologies utilising a biparental approach is discussed.  相似文献

Summary Three crosses and descendant generations were used in a field study of the inheritance of tolerance to Verticillium wilt, caused by Verticillium dahliae Kleb., in upland cotton (Gossypium hirsutum L.). The tolerant cultivar Acala SJC-1 was crossed to more susceptible parents, breeding line S5971 and cultivars Acala 4-42 and Deltapine 70. Seven generations were evaluated for each cross: the two parents (P1 and P2), F1; F2, F3, and reciprocal backcrosses (B1 and B2). The genetic control of tolerance in these crosses appears to involve more than one gene, based on an unsatisfactory fit to expected phenotypic distributions for the generations under a single-locus model. An analysis of generation means indicated that pooled additive and pooled dominance effects over loci were adequate to explain the variation among generations for crosses of SJC-1 X S5971 and SJC-1 X DPL70. Tolerance in these crosses appeared to be controlled by recessive factors. For the SJC-1 X 4-42 cross, an adequate fit to a digenic epistatic model was not possible, and none of the genetic parameters except the F2 mean were significant. Heritabilities for tolerance to Verticillium wilt, determined from regressions of F3 progeny on F2 parents for the crosses of SJC-1 X S5971 and SJC-1 X DPL70, ranged from 0.12 to 0.28. Summary The genetics of resistance to bacterial blight, Xanthomonas oryzae (Uyeda and Ishiyama) Dowson, for 74 cultivars of rice, Oryza sativa L., was studied. The PX061 isolate of bacterial blight from the Philippines was used for inoculation of parental and hybrid populations. Single dominant genes at the Xa 4 locus convey resistance in 38 cultivars. Of these, 18 are resistant at all stages of plant growth and thus have the Xa 4 aallele for resistance. However, 20 are susceptible up to maximum tillering stage but are resistant at booting and flowering stages. These cultivars have the Xa 4 ballele for resistance. Thirty-two cultivars have single recessive genes for resistance which are allelic to xa 5.The resistance in DV85, DV86 and DZ78 is conditioned by two genes. At maximum tillering stage xa 5 conveys resistance. However, at later growth stages an additional dominant gene, designated Xa 7 in DZ78, also gives resistance. The dominant genes of DV85 and DV86 are probably allelic to Xa 7. Xa 7 segregates independently of Xa 4, xa 5 and Xa 6, however like Xa 6, it conveys resistance at booting and post-booting stages only.The resistance in PI 231129 is conditioned by a single recessive gene, designated xa 8. Two l-threonine (l-serine) dehydratases (EC 4.2.1.16) of the thermophilic phototrophic bacterium Chloroflexus aurantiacus Ok-70-fl were purified to electrophoretic homogeneity by procedures involving anion exchange and hydrophobic interaction chromatography. Only one of the two enzymes was sensitive to inhibition by l-isoleucine (K i=2 M) and activation by l-valine. The isoleucine-insensitive dehydratase was active with l-threonine (K m=20 mM) as well as with l-serine (K m=10 mM) whereas the other enzyme, which displayed much higher affinity to l-threonine (K m=1.3 mM), was inactivated when acting on l-serine. Both dehydratases contained pyridoxal-5-phosphate as cofactor. When assayed by gel filtration techniques at 20 to 25° C, the molecular weights of both enzymes were found to be 106,000±6,000. In sodium dodecylsulfate-polyacrylamide gel electrophoresis, the two dehydratases yielded only one type of subunit with a molecular weight of 55,000±3,000. Theory suggests that parental relatedness is a continuous variable with a fitness optimum that we heretoforth will refer to as optimal outbreeding. In the present paper, we test this proposition from a conservation (translocation) perspective. Amphibians are facing a global decline and many amphibian populations are today small and threatened by extinction. Because genetic differentiation is often high between amphibian populations, they could be particularly sensitive to outbreeding depression, e.g. due to breakdown of locally adapted gene complexes. We tested if outbreeding would reduce fitness in common frogs, Rana temporaria, crossed from a large and an isolated, small population, separated by 130km, using artificial fertilization. For females from the large population, tadpoles were significantly smaller and more malformed in crosses with males from the small population, than with males from the large population. For the small population, however, no significant paternal genetic effects could be found. The difference in response to outbreeding between populations was accompanied with significant differences in the importance of maternal effects. We conclude that care should be taken when translocating frogs between distantly related populations to avoid outbreeding depression.  相似文献
