Tomato bacterial wilt caused by Ralstonia solanacearum is one of the most destructive soil-borne diseases. Many strategies have been taken to improve soil suppressiveness against this destructive disease, but limited success has been achieved. In this study, a novel bioorganic fertilizer revealed a higher suppressive ability against bacterial wilt compared with several soil management methods in the field over four growing seasons from March 2011 to July 2013. The application of the bioorganic fertilizer significantly (P<0.05) reduced disease incidence of tomato and increased fruit yields in four independent trials. The association among the level of disease incidence, soil physicochemical and biological properties was investigated. The soil treated with the bioorganic fertilizer increased soil pH value, electric conductivity, organic carbon, NH4 +-N, NO3 --N and available K content, microbial activities and microbial biomass carbon content, which were positively related with soil suppressiveness. Bacterial and actinomycete populations assessed using classical plate counts were highest, whereas R. solanacearum and fungal populations were lowest in soil applied with the bioorganic fertilizer. Microbial community diversity and richness were assessed using denaturing gel gradient electrophoresis profile analysis. The soil treated with the bioorganic fertilizer exhibited higher bacterial community diversity but lower fungal community diversity. Redundancy analysis showed that bacterial community diversity and richness negatively related with bacterial wilt suppressiveness, while fungal community richness positively correlated with R. solanacearum population. We concluded that the alteration of soil physicochemical and biological properties in soil treated with the bioorganic fertilizer induced the soil suppressiveness against tomato bacterial wilt.  相似文献

Soil physicochemical properties, soil microbial biomass and bacterial community structures in a rice-wheat cropping system subjected to different fertilizer regimes were investigated in two seasons (June and October). All fertilizer regimes increased the soil microbial biomass carbon and nitrogen. Both fertilizer regime and time had a significant effect on soil physicochemical properties and bacterial community structure. The combined application of inorganic fertilizer and manure organic-inorganic fertilizer significantly enhanced the bacterial diversity in both seasons. The bacterial communities across all samples were dominated by Proteobacteria, Acidobacteria and Chloroflexi at the phylum level. Permutational multivariate analysis confirmed that both fertilizer treatment and season were significant factors in the variation of the composition of the bacterial community. Hierarchical cluster analysis based on Bray-Curtis distances further revealed that bacterial communities were separated primarily by season. The effect of fertilizer treatment is significant (P = 0.005) and accounts for 7.43% of the total variation in bacterial community. Soil nutrients (e.g., available K, total N, total P and organic matter) rather than pH showed significant correlation with the majority of abundant taxa. In conclusion, both fertilizer treatment and seasonal changes affect soil properties, microbial biomass and bacterial community structure. The application of NPK plus manure organic-inorganic fertilizer may be a sound fertilizer practice for sustainable food production.  相似文献

以青海高原2 300~4 000 m海拔范围的6处西北小檗(Berberis vernae)生境土壤为试材,采用高通量测序方法,分析不同海拔梯度西北小檗生境土壤微生物群落结构及多样性。研究结果表明:(1)在西北小檗生境土壤中,细菌群落组成主要包括10个细菌门21个细菌属,真菌群落由子囊菌门、担子菌门等8个真菌门59个真菌属组成。(2)低海拔位置的海东乐都1号样点(hdld1) 0~20 cm土层的细菌群落丰富性及多样性均最高,黄南泽库样点(hnzk) 0~20 cm土层的真菌群落丰富度最高,西宁大通样点(xndt) 0~20 cm土层的真菌群落多样性最高;随着海拔的升高,0~20 cm、40~60 cm土层的细菌群落丰富度及多样性呈现出先降低再升高再降低的趋势,20~40 cm土层的细菌群落丰富度及多样性则呈现出先升高后降低的趋势,0~20 cm、20~40 cm土层土壤微生物真菌群落丰富度呈现出先升高再降低再升高的趋势,0~20 cm、40~60 cm土层真菌群落多样性呈现先升高再降低的趋势,40~60 cm土层的真菌丰富度及20~40 cm土层的真菌多样性的变化趋势不明显。(3)硝态氮、速效磷和速效钾对土壤微生物群落的影响较明显。综上可知,高寒地区不同海拔梯度西北小檗生境土壤微生物群落结构多样性呈现出一定的海拔差异趋势,其海拔差异主要受到环境条件、土壤理化性质和植被分布的影响。  相似文献

Background

Soil microbial communities are in constant change at many different temporal and spatial scales. However, the importance of these changes to the turnover of the soil microbial communities has been rarely studied simultaneously in space and time.

Methodology/Principal Findings

In this study, we explored the temporal and spatial responses of soil bacterial, archaeal and fungal β-diversities to abiotic parameters. Taking into account data from a 3-year sampling period, we analyzed the abundances and community structures of Archaea, Bacteria and Fungi along with key soil chemical parameters. We questioned how these abiotic variables influence the turnover of bacterial, archaeal and fungal communities and how they impact the long-term patterns of changes of the aforementioned soil communities. Interestingly, we found that the bacterial and fungal β-diversities are quite stable over time, whereas archaeal diversity showed significantly higher fluctuations. These fluctuations were reflected in temporal turnover caused by soil management through addition of N-fertilizers.

Conclusions

Our study showed that management practices applied to agricultural soils might not significantly affect the bacterial and fungal communities, but cause slow and long-term changes in the abundance and structure of the archaeal community. Moreover, the results suggest that, to different extents, abiotic and biotic factors determine the community assembly of archaeal, bacterial and fungal communities.  相似文献

Lacking systematic evaluations in soil quality and microbial community recovery after different amendments addition limits optimization of amendments combination in coal mine soils. We performed a short‐term incubation experiment with a varying temperature over 12 weeks to assess the effects of three amendments (biochar: C; nitrogen fertilizer at three levels: N‐N1~N3; microbial agent at two levels: M‐M1~M2) based on C/N ratio (regulated by biochar and N level: 35:1, 25:1, 12.5:1) on mine soil properties and microbial community in the Qilian Mountains, China. Over the incubation period, soil pH and MBC/MBN were significantly lower than unamended treatment in N addition and C + M + N treatments, respectively. Soil organic carbon (SOC), total nitrogen (TN), available nitrogen (AN), available phosphorus (AP), available potassium (AK), microbial biomass carbon (MBC), and nitrogen (MBN) contents increased significantly in all amended treatments (p < .001). Higher AP, AK, MBC, MBN, and lower MBC/MBN were observed in N2‐treated soil (corresponding to C/N ratio of 25:1). Meanwhile, N2‐treated soil significantly increased species richness and diversity of soil bacterial community (p < .05). Principal coordinate analysis further showed that soil bacterial community compositions were significantly separated by N level. C‐M‐N treatments significantly increased the relative abundance (>1%) of the bacterial phyla Bacteroidetes and Firmicutes, and decreased the relative abundance of fungal phyla Chytridiomycota (p < .05). Redundancy analysis illustrated the importance of soil nutrients in explaining variability in bacterial community composition (74.73%) than fungal composition (35.0%). Our results indicated that N addition based on biochar and M can improve soil quality by neutralizing soil pH and increasing soil nutrient contents in short‐term, and the appropriate C/N ratio (25:1) can better promote microbial mass, richness, and diversity of soil bacterial community. Our study provided a new insight for achieving restoration of damaged habitats by changing microbial structure, diversity, and mass by regulating C/N ratio of amendments.  相似文献

Conversion of native broadleaf forest (NF) and introduction of broadleaf species into monospecific Cunninghamia lanceolata plantations are silvicultural practices driven by the increasing demand for timber production. This study was conducted to evaluate the impacts of successive planting of C. lanceolata and mixed plantations (C. lanceolata-Michelia macclurei, CFM; C. lanceolata-Alnus cremastogyne, CFA; C. lanceolata-Kalopanax septemlobus, CFK) on microbial community diversity. Microbial biomass (MBC) was assessed using chloroform fumigation-extraction. Using denaturing gradient gel electrophoresis (DGGE), we examined the biodiversity within eubacterial (16S rRNA gene) and fungal (28S rRNA gene) species and two genes involved in N cycling: nifH and amoA. Microbial community diversities and microbial biomass decreased as NF was substituted by successive plantings of C. lanceolata plantations, whereas the trend reversed after introducing the broadleaf, M. macclurei, into pure C. lanceolata plantations. A strong positive correlation between MBC changes and total organic C (TOC), total organic N (TON), available N and extractable C (Cext) were seen, which suggests that MBC was tightly coupled with the content of soil organic matter. The Shannon index showed that bacterial diversity and two functional genes (nifH and amoA) diversities associated with monospecific C. lanceolata plantations were lower than that of NF or mixed C. lanceolata plantations, such as CFM and CFA, whereas the opposite was seen for fungal diversity. Bacterial diversity was positively correlated with pH, TOC, TON, Cext and NH 4 + -N; while fungal diversity was positively correlated with C/N ratio and negatively correlated with pH. Both nitrogen fixing and ammonia oxidizing bacterial diversities were positively correlated with pH. Thus, soil pH was not only significantly positively correlated with bacterial diversity (r?=?0.502, P?<?0.05), nifH gene diversity (r?=?0.564, P?<?0.01) and amoA gene diversity (r?=?0.659, P?<?0.001), but also negatively correlated with fungal diversity (r?=?? 0.505, P?<?0.05), which seemed to be responsible for the discrimination of the soil microbial communities among these plantations. These findings suggest that different silvicultural practices have significant impacts on the soil microbial community through influences on soil chemical properties.  相似文献

Biofumigation (BIOF) is carried out mainly by the incorporation of brassica plant parts into the soil, and this fumigation activity has been linked to their high glucosinolate (GSL) content. GSLs are hydrolyzed by the endogenous enzyme myrosinase to release isothiocyanates (ITCs). A microcosm study was conducted to investigate the effects induced on the soil microbial community by the incorporation of broccoli residues into soil either with (BM) or without (B) added myrosinase and of chemical fumigation, either as soil application of 2-phenylethyl ITC (PITC) or metham sodium (MS). Soil microbial activity was evaluated by measuring fluorescein diacetate hydrolysis and soil respiration. Effects on the structure of the total microbial community were assessed by phospholipid fatty acid analysis, while the impact on important fungal (ascomycetes (ASC)) and bacterial (ammonia-oxidizing bacteria (AOB)) guilds was evaluated by denaturating gradient gel electrophoresis (DGGE). Overall, B, and to a lesser extent BM, stimulated microbial activity and biomass. The diminished effect of BM compared to B was particularly evident in fungi and Gram-negative bacteria and was attributed to rapid ITC release following the myrosinase treatment. PITC did not have a significant effect, whereas an inhibitory effect was observed in the MS-treated soil. DGGE analysis showed that the ASC community was temporarily altered by BIOF treatments and more persistently by the MS treatment, while the structure of the AOB community was not affected by the treatments. Cloning of the ASC community showed that MS application had a deleterious effect on potential plant pathogens like Fusarium, Nectria, and Cladosporium compared to BIOF treatments which did not appear to inhibit them. Our findings indicate that BIOF induces changes on the structure and function of the soil microbial community that are mostly related to microbial substrate availability changes derived from the soil amendment with fresh organic materials.  相似文献

Soil microbes play an essential role in the forest ecosystem as an active component. This study examined the hypothesis that soil microbial community structure and metabolic activity would vary with the increasing stand ages in long-term pure plantations of Pinus elliottii. The phospholipid fatty acids (PLFA) combined with community level physiological profiles (CLPP) method was used to assess these characteristics in the rhizospheric soils of P. elliottii. We found that the soil microbial communities were significantly different among different stand ages of P. elliottii plantations. The PLFA analysis indicated that the bacterial biomass was higher than the actinomycic and fungal biomass in all stand ages. However, the bacterial biomass decreased with the increasing stand ages, while the fungal biomass increased. The four maximum biomarker concentrations in rhizospheric soils of P. elliottii for all stand ages were 18:1ω9c, 16:1ω7c, 18:3ω6c (6,9,12) and cy19:0, representing measures of fungal and gram negative bacterial biomass. In addition, CLPP analysis revealed that the utilization rate of amino acids, polymers, phenolic acids, and carbohydrates of soil microbial community gradually decreased with increasing stand ages, though this pattern was not observed for carboxylic acids and amines. Microbial community diversity, as determined by the Simpson index, Shannon-Wiener index, Richness index and McIntosh index, significantly decreased as stand age increased. Overall, both the PLFA and CLPP illustrated that the long-term pure plantation pattern exacerbated the microecological imbalance previously described in the rhizospheric soils of P. elliottii, and markedly decreased the soil microbial community diversity and metabolic activity. Based on the correlation analysis, we concluded that the soil nutrient and C/N ratio most significantly contributed to the variation of soil microbial community structure and metabolic activity in different stand ages of P. elliottii plantations.  相似文献

The aim of the study was to determine effects of a wildfire on soil nutrients and soil microbial functional diversity in short-term time scales. Burned and unburned control soil samples were collected 1 day, and 2, 4, 8, 10, 12 and 15 months after a shrubbery fire in Yumin county of Xinjiang, Northwest China. Nutrients of soil in each sampling time were detected and soil microbial functional diversity was measured by Biolog Eco plates. Results of the study showed that soil nutrients were significantly affected by fire. Soil pH increased immediately after the wildfire and was higher than that of unburned soil during 15 months post fire. Soil organic matter and total N significantly decreased immediately after the fire and was even lower than control soil at the 15th month post fire. Soil available P level increased sharply during the 4th month after the fire, and later reached to the maximum value with eight times higher than that of unburned soil. Soil available N and available K were more than the control site in 2 months after the fire, then decreased, but available N began to increase, when vegetations restored 1 year after the fire. Soil microbial activity and functional diversity recovered gradually after fire. The average well color development (AWCD) and functional diversity indices (Shannon index, Simpson index, and McIntosh index) decreased significantly 1 day after the fire, but then increased and were similar to that of undisturbed soil 15 months after the fire, when plant started to regenerate in burned area. The changes in soil nutrients after the fire affected soil microbial activity and functional diversity. Correlation analysis revealed that AWCD was negatively correlated with soil pH and positively correlated with soil total N and available N, Shannon and Simpson index had positive significantly correlation with soil total N and McIntosh index had positive significantly correlation with available N. Result of principal component analysis based on the data of carbons metabolism showed that microbial catabolic profiles of burned soils of each sampling time after the wildfire were different and all were distinct from those of unburned soils, which might suggest that microbial community structure of fire-impacted area changed dynamically on monthly scale and was distinct from that of the control site in 15 months after fire, although microbial activity or richness showed similar to pre-fire level at the 15th month post-fire.  相似文献

为探究根肿病对十字花科作物根际土壤微生物多样性的影响,以罹病大白菜和健康株根际土壤为研究对象,采用高通量测序技术对2组样本的细菌16S rDNA和真菌ITS基因进行序列测定,分析了样本间的微生物群落结构和组成差异,同时测定根际土壤理化性质,探讨根肿病、土壤微生物群落、土壤环境因子三者的相关性。研究表明:1)患病植株根际土壤pH和总磷、总钾、碱解氮、速效钾含量显著低于正常植株根际土,而交换性钙含量明显增加。2)根肿病的发生降低了根际土壤中细菌种群的丰富度和多样性程度,但对根际土壤中的真菌α-多样性无明显影响。3)变形菌门、拟杆菌门、放线菌门等是所测土壤样本的主要优势细菌种群,其中患病植株根际土壤中拟杆菌门丰度显著高于健康植株根际土壤,放线菌门丰度则显著降低(P<0.05)。优势细菌纲为γ-变形菌纲、拟杆菌纲、α-变形菌纲、放线菌纲、酸杆菌纲等,2组土壤样本间多种优势细菌纲相对丰度差异显著。4)根际土壤优势真菌类群为子囊菌门、被孢霉门、担子菌门和壶菌门,其相对丰度在患病和健康株根际土壤样本中均有明显差异。主要真菌纲为散囊菌纲、被孢霉纲、锤舌菌纲等,并且土壤样本间的多种优势真菌纲相对丰...  相似文献

In the present study, 3 replanted black pepper orchards with continuously cropping histories for 10, 21, and 55 years in tropical China, were selected for investigating the effect of monoculture on soil physiochemical properties, enzyme activities, bacterial abundance, and bacterial community structures. Results showed long-term continuous cropping led to a significant decline in soil pH, organic matter contents, enzymatic activities, and resulted in a decrease in soil bacterial abundance. 454 pyrosequencing analysis of 16S rRNA genes revealed that the Acidobacteria and Proteobacteria were the main phyla in the replanted black pepper orchard soils, comprising up to 73.82% of the total sequences; the relative abundances of Bacteroidetes and Firmicutes phyla decreased with long-term continuous cropping; and at genus level, the Pseudomonas abundance significantly depleted after 21 years continuous cropping. In addition, bacterial diversity significantly decreased after 55 years black pepper continuous cropping; obvious variations for community structures across the 3 time-scale replanted black pepper orchards were observed, suggesting monoculture duration was the major determinant for bacterial community structure. Overall, continuous cropping during black pepper cultivation led to a significant decline in soil pH, organic matter contents, enzymatic activities, resulted a decrease in soil bacterial abundance, and altered soil microbial community membership and structure, which in turn resulted in black pepper poor growth in the continuous cropping system.  相似文献

Soil degradation has been found in urban forests in Shanghai, especially in the pure plantations. Mixed plantations are considered to improve soil quality because they can stimulate organic matter cycling and increase soil carbon and nutrient content. Although soil microbes play crucial roles in regulating soil biogeochemical processes, little is known about how mixed plantations affect soil microbial communities, including bacteria, archaea, and fungi. Here, we evaluated soil chemical properties, abundances and compositions of soil bacterial, archaeal, and fungal communities, and enzyme activities in pure and mixed Metasequoia glyptostroboides and Bischofia polycarpa plantations, located in Shanghai, China. The results showed that soil available phosphorus content in the mixed plantation of M. glyptostroboides and B. polycarpa was significantly higher than that in pure plantations, while no significant difference was observed in the content of soil organic carbon, total and available nitrogen, total and available potassium among the three studied plantations. We found higher fungal abundance in the mixed plantation, when compared to both pure plantations. Moreover, fungal abundance was positively correlated with the content of soil available phosphorus. No significant difference was found in the abundance and diversity of bacterial and archaeal community among the three studied plantations. A similarity analysis (ANOSIM) showed that mixed plantation significantly altered the community composition of archaea and fungi, accompanied with an increase of alkaline phosphatase activity. However, ANOSIM analysis of bacterial communities showed that there was no significant group separation among different plantations. Overall, results from this study indicated that fungal and archaeal communities were more sensitive to aboveground tree species than bacterial community. Moreover, mixed plantations significantly increased the activity of alkaline phosphatase and the content of soil available phosphorus, suggesting that afforestation with M. glyptostroboides and B. polycarpa is an effective way to alleviate phosphorus deficiency in urban forests in Shanghai, China.  相似文献

土壤微生物多样性在生态系统功能的维持方面发挥着至关重要的作用,但是土壤生物多样性与生态系统功能(Biodiversity-ecosystem function,BEF)关系仍存在争议。以往的研究多基于简单多样性指标(如物种数、香浓多样性指数等)对BEF关系进行探究,忽略了物种间复杂的相互作用在BEF关系中的重要性。以青藏高原米拉山高寒草甸为研究对象,使用Illumina MiSeq高通量测序技术测定了6个海拔高度(3755 m、3994 m、4225 m、4534 m、4900 m、5120 m)土壤细菌和真菌群落特征,分析了简单微生物多样性指标(物种数)和共现网络复杂性与生态系统多功能性(Ecosystem multifunctionality,EMF)的关系,以期进一步揭示微生物多样性与EMF的关系。共现网络分析表明,表征土壤细菌和真菌网络复杂性的节点(Node)和边(Link)沿海拔高度的升高显著下降(P<0.05)。土壤细菌和真菌的多样性和网络复杂性均沿海拔的升高显著下降(P<0.05),而且网络复杂性比相应的多样性下降明显。在未控制环境因素时,真菌和细菌的多样性和网络复杂性均与EMF显著正相关(P<0.05);其中真菌和细菌网络复杂性对EMF的解释度高于相应多样性对EMF的解释度。通过偏回归分析(Partial least squares regression,PLSR)控制年降水、年均温、黏粒含量、盐基离子和酸性离子等气候及土壤环境因子影响后,土壤细菌和真菌物种多样性与EMF的显著正相关关系变为不相关(P>0.05),而网络复杂性与EMF的显著正相关关系(P<0.05)仍然存在。利用方差分解分析(Variance partition analysis,VPA)将环境因子纳入对EMF的影响后发现,土壤微生物网络复杂性和环境因子对EMF变化的解释度可达80%,高于土壤微生物多样性与环境因子对EMF变化的解释度。结构方程模型(Structural equation model,SEM)分析进一步显示,土壤细菌多样性和真菌多样性通过促进对应共现网络的复杂性,间接对EMF产生正向影响。综上所述,相较于简单的多样性指标,土壤微生物网络复杂性对EMF具有更好的解释度和预测性,微生物多样性主要通过促进网络复杂性间接正向影响EMF。研究结果扩展了BEF关系的研究,证明微生物物种多样性主要通过促进对应的网络复杂性维持EMF。  相似文献

Nitrogen (N) deposition poses a serious threat to terrestrial biodiversity and alters plant and soil microbial community composition. Species turnover and nestedness reflect the underlying mechanisms of variations in community composition. However, it remains unclear how species turnover and nestedness contribute to different responses of taxonomic groups (plants and soil microbes) to N enrichment. Here, based on a 13‐year consecutive multi‐level N addition experiment in a semiarid steppe, we partitioned community β‐diversity into species turnover and nestedness components and explored how and why plant and microbial communities reorganize via these two processes following N enrichment. We found that plant, soil bacterial, and fungal β‐diversity increased, but their two components showed different patterns with increasing N input. Plant β‐diversity was mainly driven by species turnover under lower N input but by nestedness under higher N input, which may be due to a reduction in forb species, with low tolerance to soil Mn2+, with increasing N input. However, turnover was the main contributor to differences in soil bacterial and fungal communities with increasing N input, indicating the phenomenon of microbial taxa replacement. The turnover of bacteria increased greatly whereas that of fungi remained within a narrow range with increasing N input. We further found that the increased soil Mn2+ concentration was the best predictor for increasing nestedness of plant communities under higher N input, whereas increasing N availability and acidification together contributed to the turnover of bacterial communities. However, environmental factors could explain neither fungal turnover nor nestedness. Our findings reflect two different pathways of community changes in plants, soil bacteria, and fungi, as well as their distinct community assembly in response to N enrichment. Disentangling the turnover and nestedness of plant and microbial β‐diversity would have important implications for understanding plant–soil microbe interactions and seeking conservation strategies for maintaining regional diversity.  相似文献

Background

Previous studies have focused on linking soil community structure, diversity, or specific taxa to disturbances. Relatively little attention has been directed to crop monoculture soils, particularly potato monoculture. Information about microbial community changes over time between monoculture and non-monoculture treatments is lacking. Furthermore, few studies have examined microbial communities in potato monoculture soils using a high throughput pyrosequencing approach.

Methodology/Principal Findings

Soils along a seven-year gradient of potato monoculture were collected and microbial communities were characterized using high throughput pyrosequencing approach. Principal findings are as follows. First, diversity (H Shannon) and richness (S Chao1) indices of bacterial community, but not of fungal community, were linearly decreased over time and corresponded to a decline of soil sustainability represented by yield decline and disease incidence increase. Second, Fusarium, the only soilborne pathogen-associated fungal genus substantially detected, was linearly increased over time in abundance and was closely associated with yield decline. Third, Fusarium abundance was negatively correlated with soil organic matter (OM) and total nitrogen (TN) but positively with electrical conductivity (EC). Fourth, Fusarium was correlated in abundances with 6 bacterial taxa over time.

Conclusions

Soil bacterial and fungal communities exhibited differential responses to the potato monoculture. The overall soil bacterial communities were shaped by potato monoculture. Fusarium was the only soilborne pathogen-associated genus associated with disease incidence increase and yield decline. The changes of soil OM, TN and EC were responsible for Fusarium enrichment, in addition to selections by the monoculture crop. Acidobacteria and Nitrospirae were linearly decreased over time in abundance, corresponding to the decrease of OM, suggesting their similar ecophysiologial trait. Soil salinization is detrimental to plant growth and yield in agroecosystems worldwide. Epichloë endophytes, a class of clavicipitaceous fungi, enhance the resistance of host plants to saline-alkali stress. This study explored the effects of the systemic fungal endophyte Epichloë coenophiala on the root microbial community and growth performance of tall fescue (Lolium arundinaceum) growing under different saline-alkali stress conditions. Structural equation modeling (SEM) was conducted to analyze the direct and indirect effects (mediated by root microbial community diversity and soil properties) of the endophyte on the growth of tall fescue under saline-alkali stress. The endophyte-infected plants produced higher shoot and root biomass compared to endophyte-free plants under saline-alkali stress (200 and 400 mM). Endophyte infection increased the fungal community diversity and altered its composition in the roots, decreasing the relative abundance of Ascomycota and increasing that of Glomeromycota. Furthermore, endophyte infection decreased the bacterial community diversity and the relative abundance of dominant Proteobacteria. SEM showed that endophyte infection increased the shoot and root biomass under saline-alkali stress (200 and 400 mM) by increasing the arbuscular mycorrhizal fungal diversity in the roots, and soil total nitrogen and phosphorus concentrations. Therefore, it is important to examine aboveground microbes as factors influencing plant growth in saline-alkali stress by affecting belowground microbes and soil chemical properties.  相似文献

The Mediterranean basin has been identified as a biodiversity hotspot, about whose soil microbial diversity little is known. Intensive land use and aggressive management practices are degrading the soil, with a consequent loss of fertility. The use of organic amendments such as dry olive residue (DOR), a waste produced by a two-phase olive-oil extraction system, has been proposed as an effective way to improve soil properties. However, before its application to soil, DOR needs a pre-treatment, such as by a ligninolytic fungal transformation, e.g. Coriolopsis floccosa. The present study aimed to describe the bacterial and fungal diversity in a Mediterranean soil and to assess the impact of raw DOR (DOR) and C. floccosa-transformed DOR (CORDOR) on function and phylogeny of soil microbial communities after 0, 30 and 60 days. Pyrosequencing of the 16S rRNA gene demonstrated that bacterial diversity was dominated by the phyla Proteobacteria, Acidobacteria, and Actinobacteria, while 28S-rRNA gene data revealed that Ascomycota and Basidiomycota accounted for the majority of phyla in the fungal community. A Biolog EcoPlate experiment showed that DOR and CORDOR amendments decreased functional diversity and altered microbial functional structures. These changes in soil functionality occurred in parallel with those in phylogenetic bacterial and fungal community structures. Some bacterial and fungal groups increased while others decreased depending on the relative abundance of beneficial and toxic substances incorporated with each amendment. In general, DOR was observed to be more disruptive than CORDOR.  相似文献

Soil microbial communities undergo rapid shifts following modifications in environmental conditions. Although microbial diversity changes may alter soil functioning, the in situ temporal dynamics of microbial diversity is poorly documented. Here, we investigated the response of fungal and bacterial diversity to wheat straw input in a 12-months field experiment and explored whether this response depended on the soil management history (grassland vs. cropland). Seasonal climatic fluctuations had no effect on the diversity of soil communities. Contrastingly fungi and bacteria responded strongly to wheat regardless of the soil history. After straw incorporation, diversity decreased due to the temporary dominance of a subset of copiotrophic populations. While fungi responded as quickly as bacteria, the resilience of fungal diversity lasted much longer, indicating that the relative involvement of each community might change as decomposition progressed. Soil history did not affect the response patterns, but determined the identity of some of the populations stimulated. Most strikingly, the bacteria Burkholderia, Lysobacter and fungi Rhizopus, Fusarium were selectively stimulated. Given the ecological importance of these microbial groups as decomposers and/or plant pathogens, such regulation of the composition of microbial successions by soil history may have important consequences in terms of soil carbon turnover and crop health.  相似文献

比较新疆叶城两种主栽红枣—骏枣与灰枣的土壤养分、微生物多样性和枣营养品质之间的差异,分析土壤养分、微生物与枣营养品质之间的相关性,从土壤养分、微生物角度研究枣营养品质的影响因素。通过化学分析法测定叶城骏枣与灰枣土壤养分、枣营养品质指标,高通量测序分析土壤微生物多样性,将土壤养分和微生物优势门属微生物进行冗余分析,研究枣营养品质指标与土壤养分、微生物功能类群的相关性。结果显示,新疆叶城两种主栽红枣土壤的全氮、速效氮、有机质含量显著不同;细菌群落变化的主要原因是土壤TN(全氮)值,真菌群落变化的主要原因是土壤AN(速效氮)值。两种主栽红枣土壤微生物多样性显示,骏枣土壤微生物在细菌水平上具有更高的物种丰富性和多样性,薄壁杆菌属(Gracilibacillus)、芽胞杆菌目(Bacillales)等溶磷微生物、固氮微生物等功能土壤微生物相对丰度和占比较高,差异显著;枣营养品质,尤其是不可溶膳食纤维、Fe、K、Zn的含量差异明显。而不可溶膳食纤维、K、蔗糖含量与TP(全磷)、OM(有机质)存在显著负相关;Fe、Zn含量与薄壁杆菌属、芽胞杆菌目等九类功能微生物存在显著正相关;Mg、可溶性糖、可溶性...  相似文献

We aimed to evaluate the capability of bio-organic fertilizer suppressing watermelon Fusarium wilt disease, compare the variations of the rhizosphere bacterial and fungal community compositions after treatment with different fertilizers, and explore mechanisms causing disease suppression in rhizosphere microbial community. A rhizobacterium (Bacillus amyloliquefaciens JDF35) was identified to control watermelon Fusarium wilt disease. Bio-organic fertilizer JDF35 (BOF) was generated by inoculating JDF35 into the organic fertilizer (OF) composed of cow and chicken manure compost (1:50 v/w). A three successive growing season pot experiment was designed to evaluate the effects of BOF compared with OF and chemical fertilizer (CF). Next-generation sequencing using the Illumina MiSeq platform was used to investigate the variations in rhizosphere microbial community composition. The growth of the watermelon plants, soil pH, and available N, P and K concentrations were the highest in the BOF treatment. The Fusarium wilt incidence in the BOF treatment was lower than that in the CF and OF treatment, and the differences for disease incidence were significant (P < 0.001). The diversity of the rhizosphere bacterial community was higher, and that of the fungal was lower in the BOF treatment. Most importantly, the BOF treatment had lowest abundances of Fusarium. The application of the BOF altered the composition of rhizosphere microbial community, suppressing Fusarium wilt disease and promoting plant growth.  相似文献
