Cells respond to the accumulation of unfolded proteins by activating signal transduction cascades that improve protein folding. One example of such a cascade is the unfolded protein response (UPR), which senses protein folding stress in the endoplasmic reticulum (ER) and leads to improvement in the protein folding and processing capacity of the organelle. A central paradox of the UPR, and indeed of all such stress pathways, is that the response is designed to facilitate both adaptation to stress and apoptosis, depending upon the nature and severity of the stressor. Understanding how the UPR can allow for adaptation, instead of apoptosis, is of tremendous physiological importance. Recent advances have improved our understanding of ER stress and the vertebrate UPR, which suggest possible mechanisms by which cells adapt to chronic stress.  相似文献

ObjectiveTo determine the effectiveness of a brief cognitive behavioural intervention in reducing the incidence of sexually transmitted infections among gay men.DesignRandomised controlled trial with 12 months'' follow up.SettingSexual health clinic in London.Participants343 gay men with an acute sexually transmitted infection or who reported having had unprotected anal intercourse in the past year.Results72% (361/499) of men invited to enter the study did so. 90% (308/343) of participants returned at least one follow up questionnaire or re-attended the clinic and requested a check up for sexually transmitted infections during follow up. At baseline, 37% (63/172) of the intervention group and 30% (50/166) of the control group reported having had unprotected anal intercourse in the past month. At 12 months, the proportions were 27% (31/114) and 32% ( 39/124) respectively (P=0.56). However, 31% (38/123) of the intervention group and 21% (35/168) of controls had had at least one new infection diagnosed at the clinic (adjusted odds ratio 1.66, 95% confidence interval 1.00 to 2.74). Considering only men who requested a check up for sexually transmitted infections, the proportion diagnosed with a new infection was 58% (53/91) for men in the intervention group and 43% (35/81) for men in the control group (adjusted odds ratio 1.84, 0.99 to 3.40). Using a regional database that includes information from 23 sexual health clinics in London, we determined that few participants had attended other sexual health clinics.ConclusionsThis behavioural intervention was acceptable and feasible to deliver, but it did not reduce the risk of acquiring a new sexually transmitted infection among these gay men at high risk. Even carefully designed interventions should not be assumed to bring benefit. It is important to evaluate their effects in randomised trials with objective clinical end points.

Two gorillas (Gorilla gorilla) were trained on a series of two-dimensional problems, each problem involving an extradimentsional shift after original learning, then an intradimensional shift, followed by another extradimensional shift. The results indicated that during the early problems the subjects appeared to solve the problems as independent subproblems, i.e., using non-conceptual behavior. However during the last two or three problems the subjects appeared to shift to a conceptual mode of behavior. These data are interpreted as further evidence of the cognitive capacity of the gorilla. Annual draft systems are the principal method used by teams in major sporting leagues to recruit amateur players. These draft systems frequently take one of three forms: a lottery style draft, a weighted draft, or a reverse-order draft. Reverse-order drafts can create incentives for teams to deliberately under-perform, or tank, due to the perceived gain from obtaining quality players at higher draft picks. This paper uses a dynamic simulation model that captures the key components of a win-maximising sporting league, including the amateur player draft, draft choice error, player productivity, and between-team competition, to explore how competitive balance and incentives to under-perform vary according to league characteristics. We find reverse-order drafts can lead to some teams cycling between success and failure and to other teams being stuck in mid-ranking positions for extended periods of time. We also find that an incentive for teams to tank exists, but that this incentive decreases (i) as uncertainty in the ability to determine quality players in the draft increases, (ii) as the number of teams in the league reduces, (iii) as team size decreases, and (iv) as the number of teams adopting a tanking strategy increases. Simulation models can be used to explore complex stochastic dynamic systems such as sports leagues, where managers face difficult decisions regarding the structure of their league and the desire to maintain competitive balance.  相似文献

Animals often select one item from a set of candidates, as when choosing a foraging site or mate, and are expected to possess accurate and efficient rules for acquiring information and making decisions. Little is known, however, about the decision rules animals use. We compare patterns of information sampling by western scrub-jays (Aphelocoma californica) when choosing a nut with three decision rules: best of n (BN), flexible threshold (FT), and comparative Bayes (CB). First, we use a null hypothesis testing approach and find that the CB decision rule, in which individuals use past experiences to make nonrandom assessment and choice decisions, produces patterns of behavior that more closely correspond to observed patterns of nut sampling in scrub-jays than the other two rules. This approach does not allow us to quantify how much better CB is at predicting scrub-jay behavior than the other decision rules. Second, we use a model selection approach that uses Akaike Information Criteria to quantify how well alternative models approximate observed data. We find that the CB rule is much more likely to produce the observed patterns of scrub-jay behavior than the other rules. This result provides some of the best empirical evidence of the use of Bayesian information updating by a nonhuman animal.  相似文献

Understanding the neural mechanisms responsible for human social interactions is difficult, since the brain activities of two or more individuals have to be examined simultaneously and correlated with the observed social patterns. We introduce the concept of hyper-brain network, a connectivity pattern representing at once the information flow among the cortical regions of a single brain as well as the relations among the areas of two distinct brains. Graph analysis of hyper-brain networks constructed from the EEG scanning of 26 couples of individuals playing the Iterated Prisoner's Dilemma reveals the possibility to predict non-cooperative interactions during the decision-making phase. The hyper-brain networks of two-defector couples have significantly less inter-brain links and overall higher modularity--i.e., the tendency to form two separate subgraphs--than couples playing cooperative or tit-for-tat strategies. The decision to defect can be "read" in advance by evaluating the changes of connectivity pattern in the hyper-brain network.  相似文献

Women's mating adaptations may vary between fertile and luteal phases, given different costs and benefits of sexual activity during each phase. Women's non-conceptive (“extended”) sexuality might function in the context of pair-bonding. The current studies examined associations between women's loyalty and faithfulness to their relationships and frequency of sexual intercourse in women using hormonal contraception. As predicted, in study 1 estimated levels (adjusted for potency) of both synthetic estrogen and progestin delivered to women moderated the association between women's loyalty/faithfulness to their partner and frequency of intercourse: as estradiol levels diminished, and progestin levels increased, women's loyalty/faithfulness became more positively associated with frequency of intercourse. Study 2 replicated these findings in a sample of women studied over a 12 week period. Results further support claims for a possible function of extended sexuality, and speak to hormonal mechanisms affecting it. They also have important methodological and applied implications.  相似文献

Pulsatile insulin output, paralleled by oscillations in intracellular Ca(2+), reflect oscillating metabolism within beta-cells in response to secretory fuels. Here we question whether oscillatory periodicity is conserved or varied from stimulation to stimulation, whether glycolysis is essential for the manifestation of an oscillatory response, and if an environment of nutrient oversupply affects oscillatory regularity. We have determined that a beta-cell oscillatory Ca(2+) pattern is independent of the type of applied secretory fuel (glucose, methyl-pyruvate, or alpha-ketoisocaproate). In addition, single cells respond with the same pattern when repeatedly stimulated, regardless of the type of stimulatory fuel. Presence of substimulatory glucose is not necessary to obtain an oscillatory responses to methyl-pyruvate or alpha-ketoisocaproate. Glucose-6-phosphate, as a measure of glycolytic flux, is not detectable under these conditions. These data suggest that multiple systems, rather than a single enzyme component, can contribute to the beta-cell oscillatory behavior. Prolonged exposure to high levels of palmitate impaired oscillatory regularity in the individual beta-cells. This supports the hypothesis that a high-fat environment might contribute to loss of regular oscillatory pattern in diabetic subjects, acting, at least in part, at the level of the single beta-cell.  相似文献

We propose antimicrobial photodynamic therapy (aPDT) as an alternative strategy to reduce the use of antibiotics in shrimp larviculture systems. The growth of a multiple antibiotic resistant Vibrio harveyi strain was effectively controlled by treating the cells with Rose Bengal and photosensitizing for 30 min using a halogen lamp. This resulted in the death of > 50% of the cells within the first 10 min of exposure and the 50% reduction in the cell wall integrity after 30 min could be attributed to the destruction of outer membrane protein of V. harveyi by reactive oxygen intermediates produced during the photosensitization. Further, mesocosm experiments with V. harveyi and Artemia nauplii demonstrated that in 30 min, the aPDT could kill 78.9% and 91.2% of heterotrophic bacterial and Vibrio population respectively. In conclusion, the study demonstrated that aPDT with its rapid action and as yet unreported resistance development possibilities could be a propitious strategy to reduce the use of antibiotics in shrimp larviculture systems and thereby, avoid their hazardous effects on human health and the ecosystem at large.  相似文献

While lupins suffer severely from Fe deficiency when grown on calcareous soils, field peas under the same conditions grow normally. This paper aimed to identify whether these differences were related to differences in either the pattern or capacity for rhizosphere acidification or Fe3+ reduction between these species. Two lupin species (Lupinus angustifolius, L. cosentinii) and field peas (Pisum sativum) were grown in solution culture for 5 weeks with both an adequate and a low supply of Fe. Plants were reliant on symbiotically fixed N. The extent of iron reduction was determined using the chelates TPTZ and BPDS. The pattern of reactions around roots was determined by placing roots in agar containing either bromocresol purple or TPTZ. The low supply of Fe decreased the growth of lupins by over 30% and induced severe chlorosis and necrosis. Growth of the peas was reduced by less than 15% and no symptoms appeared. All species acidified the solutions by about 1 pH unit regardless of the Fe treatment. The level of Fe3+ reduction was higher for all species grown with low Fe than with adequate Fe. Capacity for Fe3+ reduction was higher for all species grown with low Fe than with adequate Fe. Capacity for Fe3+ reduction was similar for all species. The pattern of acidification and reduction around roots was also similar between species. Thus it appears that the capacity of lupins to reduce Fe3+ in the rhizosphere is not the primary cause of Fe deficiency in lupins.  相似文献

Genetic control of the rover/sitter behavioral polymorphism in Drosophila melanogasterlarvae was localized to the left arm of chromosome 2.Ten independent left and right compound second chromosomes were generated in isogenic rover and sitter strains by gamma irradiation and substituted into 25 different lines. This paper proposes new models of diseased joints and evaluates the effectiveness of walking aids such as a cane and a brace for compensating for lost functions due to joint disorders. The ZMJ concept described in the previous work (Yamashita and Tagawa, 1988. In: Radharaman (Ed.), Robotics and Factories of the Future'87. Springer, New York, pp. 670-677) is modified into three joint models as follows: a passive element joint (PEJ) which has a spring at the diseased joint; a constrained range joint (CRJ), the motion of which stays within some constrained relative angle; a partial moment joint (PMJ) which can produce a partial amount of the moment produced about the joint in normal walking. A cane can enlarge a supporting area and adjust the posture of the upper torso to be upright. An ischial weight-bearing brace is effective for conservative management of hip disorders by reducing a load to the joint (Shiba et al., 1998. Clinical Orthopaedics and Related Research 351, 149-157). Walking aids like a cane or brace have been conveniently used by the handicapped. Abnormal walking was simulated for each joint model. Dynamic effects of a cane and a brace on abnormal walking were examined by the multi-body walking model.  相似文献
