The design and performance of a fully-climatized environmental chamber for the study of the physiology of conventional or germ-free research animals are described. The chamber temperature can be regulated between 14°–36°C ± 1°C, the humidity between 45–95 ± 2% at 14°C and between 20–95 ± 2% at 36°C. Bilaterally symmetrical lighting is variable over 15 stages between 50–2,400 lux. The performance of the chamber was evaluated in a trial study in which the effect of different chamber temperatures on the O2-consumption of 24 conventional Sprague-Dawley rats held in groups of six to a cage with sexes separate. This showed a mean O2-consumption at 14° to 16°C of 743 ml/(kg·hr), at 18° to 20°C of 653 ml/(kg·hr) at 26° to 28°C of 629 ml/(kg·hr) and at 30° to 32°C of 627 ml/(kg·hr). The differences between O2-consumption at 14° to 16°C and 26° to 28°C and higher were significant (0.05 > p > 0.01).
Zusammenfassung Konstruktion und Leistung einer vollklimatisierten Kammer zur Untersuchung physiologischer Reaktionen konventioneller sowie keimfreier Versuchstiere auf Umgebungseinflüsse werden beschrieben. Die Kammertemperatur kann zwischen 16° bis 36°C ± 1°C, die Feuchtigkeit zwischen 45–95 ± 2% bei 14°C und zwischen 20–95 ± 2% bei 36°C eingestellt werden. Die symmetrische Beleuchtung ist über 15 Stufen zwischen 0–2.400 Lux veränderlich. Die Eignung der Kammer wurde in einer Arbeit über den Effekt verschiedener Kammertemperaturen auf den O2-Verbrauch bei 24 konventionellen Ratten Sprague-Dawley in Gruppen von sechs in einem Käfig nach Geschlechtern getrennt, geprüft. Der mittlere O2-Verbrauch bei 14° bis 16°C war 743 ml/(kg·Std), bei 18° bis 20°C 653 ml/(kg·Std), bei 26° bis 28°C 629 ml/(kg·Std) und bei 30° bis 32°C 627 ml/(kg·Std). Die Unterschiede sind bei 14° bis 16°C und 26° bis 28°C und höher waren signifikant (0.05> p > 0.01).

Resume On décrit ici l'agencement et le fonctionnement d'une chambre entièrement climatisée pour l'étude de la physiologie d'animaux conventionnels ou libres de germes. La température peut y être réglée à ± 1°C de 14° à 36°C et l'humidité peut y varier à ± 2% pris entre 45 et 95% à 14°C et entre 20 et 95% à 36°C. Des échairages symétriques placés de part et d'autre de la chambre peuvent êtres allumés selon 15 graduations différentes entre 50 et 2.400 lux. Le fonctionnement de la dite chambre a été mis à l'épreuve lors d'un essai de consommation d'oxygène par différentes températures. L'essai portait sur 24 rats conventionnels de la race Sprague-Dawley placés par groupes de 6 par cage en séparant les sexes. L'essai a montré une consommation de 743 ml/(kg·h) entre 14° et 16°C, de 653 ml/(kg·h) entre 18 et 20°C, de 629 ml/(kg·h) entre 26° et 28°C et de 627 ml/(kg·h) entre 30 et 32°C. La différence de consommation en O2 est signifacative (0.05> p> 0.01) entre 14° à 16°C et 26° à 28°C ou davantage.

Cotton seed (Gossypium hirsutum L. cv. Stoneville 825), treated with 0, 0.2, 1.0, and 2.0 g active ingredient (a.i.) mepiquat chloride (MC) kg–1, was evaluated for the effect of MC on early plant growth. Emergence rate and total emergence of MC-treated seed and control were similar regardless of germination temperature. However, the number of leaves and squares and the dry weight of leaves, stems, and roots for hydroponically grown cotton plants were significantly lower at lower germination temperatures (15°C for 3 day/30°C for 1 day and 15°C for 4 days) than at higher germination temperatures (30°C for 4 days and 30°C for 3 days/15°C for 1 day). All MC treatments significantly decreased the number of nodes, leaves, and squares, as well as dry weight of leaves, stems, and roots, as compared to control plants at 28 days after emergence. MC seed treatments also significantly reduced plant height and total leaf area compared to controls. Water-use efficiency (WUE) was significantly lower for the 1.0 g a.i. MC treatment than for control plants. In general, the highest rate of MC seed treatment resulted in greater concentrations of calcium, phosphorus, and nitrogen in plant leaves and stems and also in greater concentrations of magnesium, phosphorus, and nitrogen in roots than in controls.  相似文献

An extracellular, alkali-tolerant, thermostable lipase was from a Pseudomonas sp. It had optimal activity at 65 °C and retained 75% of its activity at 65 °C for 90 min. The pH optimum was 9.6 and it retained more than 70% activity between pH 5 and 9 for 2 h. The culture broth was free of protease and, at 30 °C, the culture filtrate retained all the activity for at least 7 days, without any stabilizer. In shake flask culture, addition of groundnut oil (3 g l–1) towards the end of growth phase increased the activity from 4 U ml–1 to 8 ml–1.  相似文献

Oviposition and fecundity in the grain miteAcarus siro were studied at 5–30°C and 62.5–90% RH. At and above 20°C, 80% RH, mating and oviposition occurred soon after emergence, but at lower temperatures and humidities egg laying was progressively delayed from one to several days. Females needed to mate repeatedly in order to achieve maximum egg production, optimum conditions for which were 15°C, 90% RH, where total output per female averaged 435 with a maximum of 858. Oviposition rates were highest at higher temperatures, the mean daily rate at 20 and 25°C, 90% RH, rising to maximum levels of 28/29 eggs per female per day on day six.Oviposition followed clearly defined patterns, favourable conditions producing rapid increases in the mean daily oviposition rate to high peak levels reached at an early stage in the oviposition period. Less favourable conditions resulted in reduced outputs and lower, more uniform rates of egg laying. The mean oviposition period, varying with humidity, fell from 72–122 days at 5°C to 9–13 days at 30°C and the mean incubation period from 42–70 days at 5°C to 3–4 days at 30°C. Egg viability increased with increasing humidity but was little affected by temperature and unaffected by age of the female at time of oviposition.Males tended to live longer than females at most conditions; longevity—depending on humidity—averaging 13–15 days at 30°C and 129–175 days at 5°C. Adult life for females averaged 12–19 days at 30°C and 88–169 days at 5°C. An index of suitability, calculated from egg number, viability and duration of the egg stage and oviposition period, indicated that the most favourable conditions for oviposition and hatching were 20–25°C and 80–90% RH.  相似文献

The thermal preferences in a grain mass and respiration at various temperatures in mites (Acari: Acarididae) of medical and economical importance [Acarus siro (L. 1758), Dermatophagoides farinae Hughes 1961, Lepidoglyphus destructor (Schrank 1871), and Tyrophagus putrescentiae (Schrank 1781)] were studied under laboratory conditions. Based on the distribution of mites in wheat, Triticum aestivum L., grain along a thermal gradient from 10 to 40 degrees C, L. destructor, D. farinae, and A. siro were classified as eurythermic and T. putrescentiae as stenothermic. The lowest preferred temperature was found for D. farinae (28 degrees C), followed by A. siro (28.5 degrees C), L. destructor (29.5 degrees C), and T. putrescentiae (31.5 degrees C). The relationship between the respiration rate and the temperature was similar for all four mite species. The highest respiration was found in the range from 31 to 33 degrees C. This is approximately 2 degrees C higher than the preferred temperature of these species. The lower temperature threshold of respiration ranged from 1 to 5 degrees C and the upper threshold ranged from 45 to 48 degrees C. Acclimatization of A. siro to temperature regimes of 5, 15, and 35 degrees C resulted in thermal preferences between 9 and 12 degrees C, 9 and 20 degrees C, and 28 and 35 degrees C, respectively. The respiration rate of acclimatized specimens increased with the temperature, reaching a maximum at 29.0 degrees C for mites acclimatized at 5 and 15 degrees C and a maximum at 33.7 degrees C for those acclimatized at 30 degrees C.  相似文献

Previous research has demonstrated that legume proteins have insecticidal activity against stored-product pests, but activity against stored-product mites has not been tested. A study was therefore conducted to explore the potential of bean, Phaseolus vulgaris L., flour as novel botanical acaricide against five species of storage and dust mites: Acarus siro L., Aleuroglyphus ovatus (Troupeau), Caloglyphus redickorzevi (Zachvatkin), Lepidoglyphus destructor (Schrank), and Tyrophagus putrescentiae (Schrank). The effect of wheat, Triticum aestivum L., grain enriched with bean flour to eight concentrations (0, 0.01, 0.1, 0.5, 1, 2.5, 5, and 10%) on population growth initiating from the density of 50 mites per 100 g of wheat was recorded for 21 d under laboratory conditions (grain moisture 14.6% moisture content and 25 degree C in darkness). The enrichment of grain with bean flour suppressed the population growth of all tested species: 0.01% concentration reduced population growth of all tested species to >50% in comparison with the control population. The most sensitive species were A. siro and L. destructor, followed by T. putrescentiae and C. redickorzevi. The least sensitive species was A. ovatus. The terminal (i.e., after 21 d) density of mites positively correlated with bean flour concentration. The suppressive effect of bean flour was not linear but rather asymptotic. The results of this study are discussed in the context of the application of bean flour in integrated control of stored-product mites and the elimination of stored-product mite allergens.  相似文献

Low average temperatures and temperature fluctuations in temperate soils challenge the efficacy of microbial strains used for clean up of pollutants. In this study, we investigated the cold tolerance of Arthrobacter chlorophenolicus A6, a microorganism previously shown to degrade high concentrations of 4-chlorophenol at 28°C. Luciferase activity from a luc-tagged derivative of the strain (A6L) was used to monitor the metabolic status of the population during 4-chlorophenol degradation. The A6L strain could degrade 200–300 g mL–1 4-chlorophenol in pure cultures incubated at 5°C, although rates of degradation, growth and the metabolic status of the cells were lower at 5°C compared to 28°C. When subjected to temperature fluctuations between 5 and 28°C, A6L continued to degrade 4-chlorophenol and remained active. In soil microcosm experiments, the degradation rates were significantly faster the first week at 28°C, compared to 5°C. However, this difference was no longer seen after 7 days, and equally low 4-chlorophenol concentrations were reached after 17 days at both temperatures. During 4-chlorophenol degradation in soil, CFU and luciferase activity values remained constant at both 5 and 28°C. However, once most of the 4-chlorophenol was degraded, both values decreased by 1–1.5 logarithmic values at 28°C, whereas they remained constant at 5°C, indicating a high survival of the cells at low temperatures. Because of the ability of A. chlorophenolicus A6 to degrade high concentrations of 4-chlorophenol at 5°C, together with its tolerance to temperature fluctuations and stress conditions found in soil, this strain is a promising candidate for bioaugmentation of chlorophenol-contaminated soil in temperate climates.This revised version was published online in November 2004 with corrections to Volume 48.  相似文献

Dusts containing etrimfos, methacrifos and pirimiphos-methyl controlledGlycyphagus destructor within 3 weeks when applied to the surface layers of a large bulk of barley stored at 1.5–8.0°C. Only etrimfos controlledAcarus siro.Laboratory tests on wheat at 17.5°C and 3°C suggested that low temperatures had retarded the action of the acaricides in the field. After induction, seven strains ofBotrytis cinerea released into the culture broth considerable amounts of laccase in a brief production time. The set-up of a suitable production process was studied with a selected strain in a 10-L fermenter. The optimum fermentation conditions were a 3% inoculum with a high degree of sporulation, a simple medium containing 20 g L–1 of glucose and 2 g L–1 of yeast extract at pH 3.5, 2 g L–1 gallic acid as inducer, added after 2 days of growth, an agitation speed of 300 rpm, an aeration rate of 1.2 vvm and a temperature of 24°C. By optimizing the culture conditions, the enzyme activity reached 28 U ml–1 in 5 days with a specific activity of 560 U mg–1 protein. The best procedure to obtain a suitable crude enzyme preparation was concentration of the supernatant medium to 10% of the initial volume by ultrafiltration, followed by a fractional precipitation with ethanol. The optimum pH and temperature for laccase activity were 5.5 and 40°C, respectively, with syringaldazine as the substrate.  相似文献

Synopsis Growth and survival of Colorado squawfish, Ptychocheilus lucius, larvae under fluctuating 18, 22, and 26° C (5° C diel fluctuations) and constant 18, 22, 26° C, and 30° C temperature conditions and ration size corresponding to 12.5, 28,64,142, 320 brine shrimp nauplii fish–1 day–1 determined from laboratory experiments. Growth was optimal at 31° C and high at temperatures of 26° C to 30° C, at the highest food abundance. Lowest growth was under lowest food rations and highest temperatures. Growth of Colorado squawfish larvae declined substantially at temperatures < 22° C. Neither growth nor survival was significantly different between fluctuating or constant regimes. Survival of Colorado squawfish larvae was highest (95%) at 26.2° C and 235 nauplii fish–1 day–1 and high at temperatures of 20 to 30° C with food abundance > 180 nauplii fish–1 day–1. Survival was lowest when food abundance was low and temperature was high. Highest mortality occurred more than 20 days after experiments began and mortalities occurred sooner in higher than lower temperatures. Colorado squawfish larvae denied food for 5, 10, or 15 d after first feeding could have begun (6 d), had survival greater than 87 % which was equivalent to continuously fed controls. Survival of fish denied food for 17.5 d after feeding could have begun declined from 84% before feeding to 57% after feeding. Point of no return was estimated between 17.5 and 20 d. Colorado squawfish have relatively high starvation resistance. Low, stable flows that simulate natural hydrographs may enhance growth, survival, and recruitment of early life stages of Colorado squawfish by increasing water temperature and food abundance in regulated rivers of the Colorado River basin.  相似文献

The effect of LAB 173 711, a synthetic analogue of abscisic acid, has been evaluated on chilling-sensitive mung bean (Vigna radiata L. cv. Local V.) seedlings. Electrical conductivity was used for assessing the degree of chilling injury. Exposure of 8-day-old mung bean seedlings to 4°C for 35 h resulted in a 50% electrolyte leakage and induced irreversible chilling injury. The seedlings gained the best protection against chilling injury by pretreatment with LAB 173 711 (5 × 10–4 M) for 3 days. The protection effect could be sustained for 4 days. The LAB 173 711 pretreatment at 28°C did not cause a significant difference in the electrolyte leakage over the ambient temperature (28°C) control. Application of LAB 173 711 at 28°C reduced visible injury and the treated seedlings had higher ethylene production and respiration rate over the untreated control. LAB 173 711 helped maintain the integrity of the cell membrane and thus reduced the leakage of soluble sugar and amino acids. These combined effects led to a higher chilling tolerance in the mung bean seedlings.  相似文献

The results of a laboratory investigation into the effects of modified atmospheres (MA) on the eggs of mite pests of grain and cheese are presented. Four species of astigmatid mite were tested; Acarus farris (Oudemans). A. siro L., Lepidoglyphus destructor (Schrank) and Tyrophagus longior (Gervais). All are found in many habitats including grain and cheese stores. Three low oxygen (O2) MA mixtures were used, based on carbon dioxide (CO2), nitrogen (N2) or simulated burner gas (0.5 or 2% O2, 10% CO2, balance N2) plus 60% CO2 in air (8% O2). The mites were exposed at 15 degrees C and 80% r.h., a combination of conditions that occurs at the surface of stored grain during the autumn which promotes mite population growth. The exposure periods required to prevent egg hatch for each species in every mixture are given. Tyrophagus longior was the most tolerant species, followed by A. siro and A. farris, with L. destructor the most susceptible. Burner gas was the most effective mixture overall with 0.5% O2 but with an increase in the O2 level to 2% for all the mixtures, CO2 became the more effective control agent. With 60% CO2 in air some loss of efficacy was observed against the three most tolerant species and even more so for L. destructor. Sublethal exposures to MAs for at least 4 days in L. destructor, 6 days in A. farris and A. siro and 8 days for T. longior caused a delay in egg hatch.  相似文献

Mycelial growth and extracellular polysaccharide production of Phellinus linteus were optimal at pH 5 and 25 °C. Maximum biomass production (14.2 g l–1) was after 15 d of cultivation, whereas, extracellular polysaccharide was maximal (3.5 g l–1) after 21 d. The hypoglycemic effect of the polysaccharide, investigated in streptozotocin-induced diabetic rats, decreased plasma glucose, total cholesterol and triacylglycerol concentrations by 49%, 32%, and 28%, respectively, and aspartate aminotransferase activity by 20%. The results indicate the potential of this polysaccharide to prevent hyperglycemia in diabetic patients.  相似文献

All life-stages ofSarcoptes scabiei var.canis survive in the hosts' environment for several days to several weeks depending on r.h. and temperature. Survival of larvae was comparable to males; survival of nymphs was comparable to females. Females and nymphs generally survived longer than larvae and males.Low temperature (10–15°) and high r.h. prolonged survival of all life stages. At 10–15°C, females and nymphs survived 1–3 weeks at 97% r.h., 1–2 weeks at 75% r.h. and 5–8 days at 45% r.h. At 20–25°C, survival was significantly reduced but all life-stages survived at least 2 days at 25% r.h. and 5–6 days at 75–100% r.h. Long survival off the host coupled with host-seeking behavior of these mites make it likely that environmental contamination is a source of scabies in domestic and wild mammals, and in humans.  相似文献

Pichia anomala, isolated from dried flower buds of Woodfordia fruticosa, produced a high activity of an intracellular phytase, at 68 U per g dry biomass, when grown at 20 °C for 24 h in a medium containing glucose (40 g l–1) and beef extract (10 g l–1) supplemented with Fe2+ (0.15 mM). Partially purified phytase was optimally active at 60 °C and pH 4 with a half life of 7 days at 60 °C. It retained 85% of its activity at 80 °C for 15 min. The enzyme is suitable for supplementing animal feeds to improve the availability of phosphate from phytate.  相似文献

Nicotinic acid, pyridoxine, and picloram were stable in a liquid MS culture medium (pH 5.5–5.6) during autoclaving and during cell-free incubation in the dark at 5°C or 25°C for up to 6 weeks. Thiamine loss under the same conditions was 16% at 5°C and 18% at 25°C. Five percent of the sucrose in the liquid medium was hydrolyzed during autoclaving. During cell-free incubation in the light (100 E m–2 s–1) at 25°C, pyridoxine was not detected after 6 days, while 78% of the picloram and 56% of the thiamine were degraded after 6 weeks. All of the niacin and pyridoxine, 13% of the picloram and 42% of the thiamine in a liquid MS culture medium were utilized in 4 days by potato (cv. The optimal growth of Cistanche deserticola callus and formation of phenylethanoid glycosides (PeG) was at 25°C with light irradiation intensity of 24 mol m–2 s–1 on solidified B5 media supplemented with 0.5 mg 6-benzylaminopurine l–1, 10 mg gibberellin l–1, 800 mg casein hydrolysate l–1 and 20 g sucrose l–1. After 30 d culture, the biomass reached 15.5 g dry wt callus l–1 medium and its PEG content was 10.7% (w/w). The PeG content was 42%–127% higher than those in explants.  相似文献

The CO2 evolution of intact potato tubers (Solanum tuberosum, L., var. Bintje) was analyzed during a 10-day period of their warm (25 ± 2°C) or cold (5 ± 1°C) storage, to evaluate cold-stress effects on expression and activities of plant uncoupling mitochondrial protein (PUMP) and alternative oxidase (AOX). CO2 evolution rates were analyzed at 20°C, to reflect their possible capacities. The 20°C CO2 production declined from 13 to 8 mg kg–1 h–1 after 2 days of warm storage and then (after 3 to 7 days) decreased from 8 to 6.5 mg kg–1 h–1. In contrast, 20°C CO2 evolution did not change after the first day of cold storage, increased up to 14.5 mg kg–1 h–1 after 2 days, and decreased to about 12 mg kg–1 h–1 after 3 to 7 days of cold storage. Cold storage increased PUMP expression as detected by Western blots and led to elevated capacities of both PUMP (44%) and CN-resistant AOX (10 times), but not the cytochrome pathway. Since we found that cold storage led to about the same mitochondrial respiration of 40 nmol O2 min–1 mg–1 attributable to each of the respective proteins, we conclude that both AOX and PUMP equally contribute to adaptation of potato tubers to cold.  相似文献

The development times and survival of immature stages in rockwool and the fecundity and longevity of adult Scatella stagnalis were determined and stage-specific life-tables constructed for the species at constant 20 and 25 °C and at a fluctuating temperature (23–34 °C, mean 28.5 °C). Development time from egg to adult decreased with temperature, being 15.9±0.1 days at 20 °C, 11.4±0.1 days at 25 °C and 10.1±0.2 days at fluctuating temperature with mean of 28.5 °C. The lower threshold for egg-to-adult development was 6.4±2.7 °C and the total quantity of thermal energy required to complete development was 212.8±.0 °C. The proportion of females in two populations studied was 0.521. High temperature increased the mortality of pupae from 7% (20 °C) and 10% (25 °C) to 29% at 28.5 °C. At 25 °C, female longevity was 15.5±0.7 days and fecundity 315±19 eggs/female (20.4 eggs/female/day). Males lived for 22.0±1.1 days. At constant 25 °C, the net reproductive rate was 126.1 female eggs/female, generation time was 18.4 days, the doubling time of the population 5.3 days, and the intrinsic rate of increase (r m) 0.263 day–1.  相似文献

The parameters which govern infection of rubber-vine weed by the rustMaravalia cryptostegiae were investigated. The infection process, from appressorial formation to sporulation, is described and illustrated. Uredinioid teliospores have an optimum temperature range for germination at 22–27 °C, both in vitro and in vivo. However, germination on the rubber-vine leaf was more than double (81–92%) that in the absence of the host, and appressoria were formed only in vivo. An optimum temperature of 20–22°C and a dew period of 12 hours or more gave the highest level of infection as measured by sporulation density. The latent period from inoculation to pustule formation decreased with increasing temperature; the shortest period (8–11 days) being recorded at 25–27°C. At the lower temperatures (18°C), this was significantly extended (19–21 days). Four successive inoculations significantly reduced plant height and dry weight, although a compensatory growth flush occurred after the third inoculation. The addition of cryoprotectants had a negative affect on spore viability and subsequent infectivity. Cooling dry spores to –196°C at the rate of 10°C min–1 gave the best results, with high germination (93–65%) up to 8 days after thawing.  相似文献
