谷氨酰胺是一种十分有用的氨基酸,它既可作为治疗药物,又可作为其它合成药物的前体。目前国内以谷氨酸为原料,采用化学合成法生产谷氨酰胺,供作试剂用,产量十分有限。日本用发酵法生产谷氨酰胺,年产1000吨,还有增加趋势。谷氨酰胺的销售价格比较高,经济效益可观。近年来国内有好几个研究小组都在进行谷氨酰胺发酵研究。我们是国内第一个谷氨酰胺研究组,已在5m~3发酵罐上取得中试成功。谷氨酰胺发酵液中混有谷氨酸,而这些少量的谷氨酸采用等电点沉淀法并不能将它们去除,影响谷氨酰胺的纯度。我们采用离子交换树脂法,成功地将谷氨酸从谷氨酰胺和谷氨酸的混合物中分离掉,得到单一的谷氨酰胺。在将发酵液上柱交换之前,必须对发酵液进行预处理,我们试验了4种不同方法,结果表明第4种方法更适合工厂使用。从我们使用过的几种树脂的分离效果及树脂的价格看,考虑到工  相似文献

以嗜乙酰乙酸棒杆菌(Corynebacterium acetoacidophilum)LG-3为出发菌株,经紫外线(UV)和硫酸二乙酯(DES)诱变处理,磺胺胍(SG)、高浓度(NH4)2SO4定向筛选,获得1株谷氨酰胺高产菌株LG-65,在未经优化的条件下摇瓶发酵72 h可产谷氨酰胺43.5 g/L,比出发菌株的产量32.4 g/L提高了34.3%.在此基础上,对其发酵条件进行优化,经72 h摇瓶发酵产量可达47~48 g/L,7 L发酵罐补料分批发酵40 h可达55 g/L.  相似文献

吸水链霉菌谷氨酰胺转胺酶分离纯化方法改进及结构研究   总被引:1,自引:0,他引:1  
利用改进的新方法,对吸水链霉菌谷氨酰胺转氨酶(TGase)进行分离纯化.将发酵上清液经过硫酸铵分级盐析后,用Hitrap Q HP阴离子交换柱除掉干扰较大的色素,之后又经Superdex 75 10/300GL凝胶柱和Hitrap Q HP阴离子交换柱的分离,得到高纯度的TGase.纯化后TGase的比活力可达24.5 U/mg,回收率为39.9%.TGase的N-末端前6个氨基酸经测序为DAADER.该研究是对吸水链霉菌TGase的N-末端氨基酸序列的首次报道.将吸水链霉菌TGase的N-末端氨基酸序列与已报道的其它3种链霉菌来源的TGase的N-端氧基酸序列进行比对,序列相似性不高.预测和分析了吸水链霉菌TGase的高级结构,为进一步研究TGase结构和功能的关系,蛋白分子定向改造提供理论依据.  相似文献

L-乳酸的发酵生产和聚L-乳酸的化学加工   总被引:2,自引:0,他引:2  
L-乳酸广泛应用于食品、医药、日化和工业等各个领域。近年来随着石化资源的不断紧缺,众多化学合成的高分子材料的生产受到了限制。以生物质资源为基础的L-乳酸因此被大量用于加工生产成聚L-乳酸等环境友好型生物可降解材料。正是由于L-乳酸需求量的增大,如何高效低成本地生产L-乳酸显得尤为重要。系统综述了L-乳酸生产菌株的选育,用于L-乳酸发酵生产的廉价资源的开发利用,L-乳酸的发酵生产和L-乳酸的分离纯化等方面的研究进展。目前研究的热点和难点正是基于上述四个部分:菌种方面,以可以高效代谢利用廉价底物,且营养需求低的选育目标获得了多个优良的生产菌种,然而具备综合代谢优势的菌种还有待进一步选育;发酵底物方面,已开发利用多种廉价,来源丰富且易于菌种代谢并高效转化成乳酸的底物,但是对这些底物工业规模应用还有待进一步研究;发酵工艺方面,建立了环境友好型,劳动强度低的发酵工艺,然而实际应用中仍然存在成本高的问题;后提取方面,通过选育低营养需求的生产菌种和采用新型发酵工艺有效地简化了后提取过程,但是实际应用方面仍受发酵工艺成本高的制约。最后对聚L-乳酸的化学加工以及聚L-乳酸的生物降解进行了探讨并提出了一些建议。  相似文献

目的:采用响应面法对L-谷氨酰胺发酵培养基成分进行优化,以提高L-谷氨酰胺发酵产量。方法:首先利用Plackett-Burman试验设计筛选出影响L-谷氨酰胺产量的主要因素葡萄糖、玉米浆和(NH4)2SO4,在此基础上采用最陡爬坡实验逼近最大响应区域,并利用中心组合设计及响应面分析法进行回归分析。结果:通过求解回归方程得到L-谷氨酰胺发酵培养基最佳浓度为葡萄糖100 g/L、玉米浆4.5 ml/L、(NH4)2SO437.2 g/L,L-谷氨酰胺产量理论最大值达41.0 g/L。结论:经模型验证,预测值与验证试验平均值接近,在优化条件下谷氨酰胺产量提高了37.6%。  相似文献

The aim of the study was to evaluate haematological responses in Red Sokoto goats (RSGs) administered with L-glutamine during the hot-dry season. Experimental animals included 28 clinically healthy RSGs divided into treated group (n = 14); each administered L-glutamine at 0.2 g/kg body weight, dissolved in 10 mL distilled water, and control group (n = 14); each administered 10 mL distilled water, per os once daily for 21 days. The ambient temperature and relative humidity recorded daily for 4 weeks were used to calculate the temperature-humidity index. Three millilitres of blood sample was collected from each goat by jugular venipuncture for haematology, while rectal temperature (RT), heart rate (HR) and respiratory rate (RR) were also measured once weekly at weeks 0 (before), 1, 2, 3 (during) and 4 (after L-glutamine administration). The haematological, RT, HR and RR data obtained weekly were analysed using repeated-measures one-way ANOVA, followed by Tukey's post-hoc test to evaluate differences between periods, and between treated and control groups. The PCV, haemoglobin concentration and RBC count were higher (P < 0.05) in the treated group compared to the control group during the period of L-glutamine administration. These differences were sustained till week 4. Beginning from week 1 of the study, the total leucocyte count in treated group (10.10 ± 0.25 × 103/μL) was higher (P < 0.05) than the count in control group (7.23 ± 0.41 × 103/μL), this trend was also maintained throughout the study. The neutrophil:lymphocyte ratio during weeks 3 and 4 of the experiment was lower (P < 0.05) in the treated compared to the control group. RT was lower (P < 0.05) in treated group than the control group. In conclusion, L-glutamine administration ameliorated the adverse effects of heat stress on the haematological parameters in RSGs during the hot-dry season.  相似文献

Abstract

Protonation equilibria of L-glutamine and speciation of its complexes with Co(II), Ni(II) and Cu(II) have been studied in 0–2.5% w/v SLS–water media using pH-metric method. The protonation constants and binary stability constants have been calculated with the computer program MINIQUAD75. Selection of the best fit chemical models is based on standard deviation in stability constants and residual analysis using crystallographic R-factor and sum of squares of residuals in all mass-balance equations. The trend in the variation of stability constants of the complexes with mole fraction of the medium is attributed to the compartmentalisation of complexation equilibria. Distribution of species and effect of influential parameters on chemical speciation have also been presented.  相似文献

A split-stream flow-injection analysis system is described for simultaneous determination of glucose and L-glutamine in serum-free hybridoma bioprocesses media. Amperometric measurement of glucose is based on anodic oxidation of hydrogen peroxide produced by immobilized glucose oxidase within a triple layer membrane of an integrated flow-through glucose-selective biosensor. Determination of L-glutamine is based on quantitating ammonium ions produced in a flow-through enzymes reactor containing immobilized glutaminase enzyme, and subsequent downstream potentiometric detection of these ions by a nonacting-based ion-selective polymer membrane electrode. Endogenous potassium and ammonium ion interference in the L-glutamine determination are eliminated by using a novel in-line tubular cation-exchange membrane unit to exchange these interferent species for cations undetectable by the membrane electrode. The first generation split-steam flow-injections system can assay 12 samples/h using direct injections of 50 muL of media samples, with linear responses to glucose in the range of 0.03 to 30mM, and log-linear response to L-glutamine from 0.1 to 10 mM. The catabolism of D-glucose was recently found to be impaired in pancreatic islets from rats depleted in long-chain polyunsaturated omega3 fatty acids. The specificity of this alteration was now investigated by characterizing the oxidative fate of endogenous nutrients in islets preincubated with either L-[U-14C]glutamine or [U-14C]palmitate and then incubated variously in the absence of D-glucose, presence of the hexose or presence of metabolic poisons. Relative to their radioactive content after preincubation, the production of 14CO2 by islets prelabelled with [U-14C]glutamine was higher in omega3-depleted rats than control animals. The enhancing action of D-glucose upon such production was impaired, however, in the omega3-depleted rats. The net uptake of 14C-palmitate and absolute value for 14CO2 output were both increased in omega3-depleted rats, whilst the ratio between 14CO2 output and islet radioactive content was decreased in the same animals. The inhibition of 14CO2 production by metabolic poisons was comparable in all cases. These results are consistent with recent findings on such items as the availability of endogenous amino acids and uptake of unesterified fatty acids in extrapancreatic sites of the omega3-depleted rats. They also support the view that the alteration of D-glucose metabolism in the islets of the latter animals may be attributable, in part at least, to alteration of glucokinase kinetics by high intracellular acyl-CoA levels.  相似文献

维生素是维持人体生命活动必需的一类有机物质,机体本身一般不能合成或合成量不足,因此需经食物或其他强化产品获取。目前,维生素产品已广泛应用于医药、食品添加剂、饲料添加剂、化妆品等领域,而且全球对维生素的需求也是呈逐年增长态势。维生素的生产方法主要包括化学合成法和生物合成法。化学合成法通常安全隐患大、反应条件严苛、废物污染严重,相比之下,代谢工程生产维生素绿色环保安全、能耗低,因此建立微生物细胞工厂具有重大的科学意义和应用需求。文中回顾了近30年来代谢工程在维生素生产领域的研究进展,详细阐述了水溶性维生素(维生素B1、B2、B3、B5、B6、B7、B9、B12和维生素C的前体)和脂溶性维生素(维生素A、维生素D的前体、维生素E和维生素K)的生物合成研究现状,并对其发酵生产的瓶颈进行了探讨,最后对合成生物技术创建维生素生产菌种进行了展望。  相似文献

The review is focused on several aspects of butyric acid production: butyric acid-producing bacterial strains, the characteristics of the genus Clostridium (the bacterium most used for butyrate production), and alternative methods of obtaining butyric acid by alcohol biotransformation. Further, the main metabolic pathways of butyrate production, and possibilities for their control are outlined. Batch, fed-batch or continuous fermentation combined with cell recycle or immobilization are applicable for anaerobic fermentations using Clostridium as the production strain. The best process comprises a combination of high cell concentration and slowly growing biomass, in addition to high production selectivity and low inhibitory effects of the end-product. Inhibitory effects may be avoided by on-line removal of the end-product. Extraction alone or extraction combined with simultaneous stripping of the organic phase (liquid membrane) into the second aqueous phase (pertraction) seem to be the most suitable methods for on-line butyrate removal. The biocompatibility and the distribution coefficient of the organic phase under fermentation conditions should be considered before designing a fermentation apparatus.
