To improve our understanding of the changes in bacterial and fungal diversity in natural pine and planted forests in subtropical region of China, we examined bacterial and fungal communities from a native and a nearby planted pine forest of the Mt. Lushan by constructing clone libraries of 16S and 18S rRNA genes. For bacterial communities, Proteobacteria and Acidobacteria were dominant bacterial taxa in both two types of forest soils. The Shannon–Wiener diversity index, rarefaction curve analysis, and LibShuff analysis suggest that these two forests contained similar diversity of bacterial communities. Low soil acidity (pH ≈ 4) of our study forests might be one of the most important selection factors determining growth of acidophilic Acidobacteria and Proteobacteria. However, the natural forest harbored greater level of fungal diversity than the planted forest according to the Shannon–Wiener diversity index and rarefaction curve analysis. Basidiomycota and Ascomycota were dominant fungal taxa in the soils of natural and planted forests, respectively. Our results suggest that fungal community was more sensitive than the bacterial community in characterizing the differences in plant cover impacts on the microbial flora in the natural and planted forests. The natural and planted forests may function differently due to the differences in soil fungal diversity and relative abundance.  相似文献

In acidic forest soils, availability of inorganic nutrients is a tree-growth-limiting factor. A hypothesis to explain sustainable forest development proposes that tree roots select soil microbes involved in central biogeochemical processes, such as mineral weathering, that may contribute to nutrient mobilization and tree nutrition. Here we showed, by combining soil analyses with cultivation-dependent analyses of the culturable bacterial communities associated with the widespread mycorrhizal fungus Scleroderma citrinum, a significant enrichment of bacterial isolates with efficient mineral weathering potentials around the oak and beech mycorrhizal roots compared to bulk soil. Such a difference did not exist in the rhizosphere of Norway spruce. The mineral weathering ability of the bacterial isolates was assessed using a microplaque assay that measures the pH and the amount of iron released from biotite. Using this microplate assay, we demonstrated that the bacterial isolates harboring the most efficient mineral weathering potential belonged to the Burkholderia genus. Notably, previous work revealed that oak and beech harbored very similar pHs in the 5- to 10-cm horizon in both rhizosphere and bulk soil environments. In the spruce rhizosphere, in contrast, the pH was significantly lower than that in bulk soil. Because the production of protons is one of the main mechanisms responsible for mineral weathering, our results suggest that certain tree species have developed indirect strategies for mineral weathering in nutrient-poor soils, which lie in the selection of bacterial communities with efficient mineral weathering potentials.The mobilization of nutrients via the biotic and abiotic weathering of soil minerals is crucial to satisfying plant nutritional needs (2, 17), especially in acidic forest soils, which are mainly nonfertilized and nutrient poor. Besides the physicochemical weathering reactions, evidence is presently accumulating which indicates that certain soil bacterial strains increase mineral weathering and improve tree nutrition (5, 9, 32, 39-41).By way of their root exudates, plants alter the structure and activity of microbial communities (6, 25, 51) and selectively favor certain ones that are potentially beneficial to them (15, 16, 21, 45, 46). A hypothesis for sustainable forest development proposes that tree roots select from the soil efficient mineral weathering bacterial communities that may contribute to nutrient mobilization and tree growth (20). In this manner, recent studies (10, 46) have revealed that the oak-Scleroderma citrinum ectomycorrhizal symbiosis selects bacterial communities that are more efficient in mineral weathering than those of the surrounding soil, suggesting that the mycorrhizal symbiosis has an indirect effect on plant nutrition through its selective pressure on the functional diversity of the mycorrhizosphere bacterial communities.Distinct impacts of the tree species on the soil bacterial community structure have been previously reported (23, 38), suggesting that the composition and activity of soil bacterial communities depend on tree physiology and notably on its impact on the soil physicochemical properties and nutrient cycling (24, 26, 37). However, no study has ever addressed the question of the impact of tree species on the structure of forest soil bacterial communities involved in mineral weathering. This question regarding the impact of tree species on the functional diversity of the bacterial communities remains a major issue in forestry, especially in the context of today''s climate change, which will give rise to a shift in the spatial distribution of forest tree species.To appreciate the effect of tree species on mycorrhizosphere bacterial communities, we focused on a single but ubiquitous mycorrhizal fungus, S. citrinum, which forms mycorrhizae with different tree species. Since no functional genes have been identified to date, a cultivation-dependent analysis was developed in this study. A total of 155 bacterial isolates were randomly chosen among a collection of 400 bacterial isolates from the soil-Scleroderma citrinum mycorrhiza interface (ectomycorrhizosphere), the extramatrical mycelium (hyphosphere), and the surrounding soil (bulk soil) in 28-year-old stands of oak (Quercus sessiliflora Smith), beech (Fagus sylvatica L.), and Norway spruce (Picea abies Karst.). The mineral weathering potential of each bacterial isolate was evaluated by way of an in vitro microplate assay, putting in interaction a calibrated bacterial suspension and the biotite, a mineral widespread in soils (46). The bacterial isolates were genotypically characterized by amplifying and sequencing a portion of the 16S rRNA gene. Their mineral weathering efficiencies and the functional structure of the bacterial communities were compared with the physicochemical characteristics of the surrounding soil.  相似文献

An ectomycorrhiza is a multitrophic association between a tree root, an ectomycorrhizal fungus, free-living fungi and the associated bacterial communities. Enzymatic activities of ectomycorrhizal root tips are therefore result of the contribution from different partners of the symbiotic organ. However, the functional potential of the fungus-associated bacterial communities remains unknown. In this study, a collection of 80 bacterial strains randomly selected and isolated from a soil–ectomycorrhiza continuum (oak–Scleroderma citrinum ectomycorrhizas, the ectomycorrhizosphere and the surrounding bulk soil) were characterized. All the bacterial isolates were identified by partial 16S rRNA gene sequences as members of the genera Burkholderia, Collimonas, Dyella, Mesorhizobium, Pseudomonas, Rhizobium and Sphingomonas. The bacterial strains were then assayed for β-xylosidase, β-glucosidase, N-acetyl-hexosaminidase, β-glucuronidase, cellobiohydrolase, phosphomonoesterase, leucine-aminopeptidase and laccase activities, chitin solubilization and auxin production. Using these bioassays, we demonstrated significant differences in the functional distribution of the bacterial communities living in the different compartments of the soil–ectomycorrhiza continuum. The surrounding bulk soil was significantly enriched in bacterial isolates capable of hydrolysing cellobiose and N-acetylglucosamine. In contrast, the ectomycorrhizosphere appeared significantly enriched in bacterial isolates capable of hydrolysing glucopyranoside and chitin. Notably, chitinase and laccase activities were found only in bacterial isolates belonging to the Collimonas and Pseudomonas genera. Overall, the results suggest that the ectomycorrhizal fungi favour specific bacterial communities with contrasting functional characteristics from the surrounding soil.  相似文献

开展川西亚高山相似土壤母质背景下天然次生林土壤微生物群落结构及其多样性探究,可加深次生林更新过程中土壤微生物群落结构变化的认知。选取川西米亚罗林区20世纪60年代采伐后经自然更新恢复形成的3种天然次生林(槭-桦阔叶林,ABB;桦-槭-冷杉针阔混交林,BAA;岷江冷杉林,AFF),分析林下表层(0-20 cm)土壤微生物群落结构变化及其影响因素,结果显示:(1)3种林型土壤细菌Chao1和Shannon指数均极显著高于真菌,但仅真菌群落的Shannon指数差异显著,表现为BAA > ABB > AFF;(2)细菌群落优势门主要为变形杆菌门、酸杆菌门、疣微菌门、拟杆菌门、绿弯菌门,相对丰度占比超过82%;真菌群落则为子囊菌门和担子菌门,占比超过85%,AFF担子菌门相对丰度最高而子囊菌门最低。(3) RDA分析显示,土壤pH和乔木物种多样性(Shannon指数)是影响微生物群落结构变化的主导因子;土壤养分元素对细菌群落影响不显著,真菌群落主要受TN、TP含量显著影响。总体上,林型间乔木层物种多样性、土壤酸碱度及其氮磷含量是导致微生物群落结构变化的关键因素。  相似文献

土壤微生物群落对麻栎-刺槐混交林凋落物分解的影响   总被引:1,自引:0,他引:1  
以麻栎-刺槐混交林和麻栎纯林为研究对象,采用野外定点采样、室内分析与高通量测序的方法,对凋落物分解过程中土壤微生物菌群多样性特征及其对凋落物分解速率的影响进行了研究。结果表明:(1)麻栎-刺槐混交林凋落物的分解速率高于麻栎纯林。两种林分凋落物有机碳(TOC)、全氮(TN)发生释放,全磷(TP)发生积累-释放的过程。(2)两种林分土壤细菌优势类群为放线菌门(Acidobacteria)、变形菌门(Proteobacteria)、酸杆菌门(Actinobacteria)和疣微菌门(Verrucomicrobia),土壤真菌优势类群为担子菌门(Basidiomycota)、子囊菌门(Ascomycota)和被孢霉门(Moritierellomycota)。(3)凋落物分解过程中,麻栎-刺槐混交林土壤微生物菌群丰富度指数和菌群多样性指数变化范围小于麻栎纯林。(4)凋落物分解速率与土壤细菌菌群丰富度指数和菌群多样性指数呈显著正相关,与土壤真菌菌群丰富度指数呈显著正相关。土壤微生物群落对麻栎-刺槐混交林和麻栎纯林凋落物分解速率具有重要影响,研究结果为深入开展混交林土壤微生物多样性对凋落物分解的影响研究提供理论依据。  相似文献

Phosphate-mobilizing bacteria (PMB) play a critical role in the regulation of phosphorus availability in the soil. The microbial genes pqqC and phoD encode pyrroloquinoline quinone synthase and bacterial alkaline phosphatase, respectively, which regulate inorganic and organic phosphorus mobilization, and are therefore used as PMB markers. We examined the effects of soil properties in three Moso bamboo forest sites on the PMB communities that were profiled using high-throughput sequencing. We observed differentiated responses of pqqC- and phoD-harbouring PMB communities to various soil conditions. There was significant variation among the sites in the diversity and structure of the phoD-harbouring community, which correlated with variation in phosphorus levels and non-capillary porosity; soil organic carbon and soil water content also affected the structure of the phoD-harbouring community. However, no significant difference in the diversity of pqqC-harbouring community was observed among different sites, while the structure of the pqqC-harbouring bacteria community was affected by soil organic carbon and soil total nitrogen, but not soil phosphorus levels. Overall, changes in soil conditions affected the phoD-harbouring community more than the pqqC-harbouring community. These findings provide a new insight to explore the effects of soil conditions on microbial communities that solubilize inorganic phosphate and mineralize organic phosphate.  相似文献

Increases in the magnitude and variability of precipitation events have been predicted for the Chihuahuan Desert region of West Texas. As patterns of moisture inputs and amounts change, soil microbial communities will respond to these alterations in soil moisture windows. In this study, we examined the soil microbial community structure within three vegetation zones along the Pine Canyon Watershed, an elevation and vegetation gradient in Big Bend National Park, Chihuahuan Desert. Soil samples at each site were obtained in mid-winter (January) and in mid-summer (August) for 2 years to capture a component of the variability in soil temperature and moisture that can occur seasonally and between years along this watershed. Precipitation patterns and amounts differed substantially between years with a drought characterizing most of the second year. Soils were collected during the drought period and following a large rainfall event and compared to soil samples collected during a relatively average season. Structural changes within microbial community in response to site, season, and precipitation patterns were evaluated using fatty acid methyl ester (FAME) and polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analyses. Fungal FAME amounts differed significantly across seasons and sites and greatly outweighed the quantity of bacterial and actinomycete FAME levels for all sites and seasons. The highest fungal FAME levels were obtained in the low desert scrub site and not from the high elevation oak–pine forests. Total bacterial and actinomycete FAME levels did not differ significantly across season and year within any of the three locations along the watershed. Total bacterial and actinomycete FAME levels in the low elevation desert-shrub and grassland sites were slightly higher in the winter than in the summer. Microbial community structure at the high elevation oak–pine forest site was strongly correlated with levels of NH4 +–N, % soil moisture, and amounts of soil organic matter irrespective of season. Microbial community structure at the low elevation desert scrub and sotol grasslands sites was most strongly related to soil pH with bacterial and actinobacterial FAME levels accounting for site differences along the gradient. DGGE band counts of amplified soil bacterial DNA were found to differ significantly across sites and season with the highest band counts found in the mid-elevation grassland site. The least number of bands was observed in the high elevation oak–pine forest following the large summer-rain event that occurred after a prolonged drought. Microbial responses to changes in precipitation frequency and amount due to climate change will differ among vegetation zones along this Chihuahuan Desert watershed gradient. Soil bacterial communities at the mid-elevation grasslands site are the most vulnerable to changes in precipitation frequency and timing, while fungal community structure is most vulnerable in the low desert scrub site. The differential susceptibility of the microbial communities to changes in precipitation amounts along the elevation gradient reflects the interactive effects of the soil moisture window duration following a precipitation event and differences in soil heat loads. Amounts and types of carbon inputs may not be as important in regulating microbial structure among vegetation zones within in an arid environment as is the seasonal pattern of soil moisture and the soil heat load profile that characterizes the location.  相似文献

Grey alder (Alnus incana) and black alder (Alnus glutinosa) stands on forest land, abandoned agricultural, and reclaimed oil-shale mining areas were investigated with the aim of analysing the functional diversity and activity of microbial communities in the soil–root interface and in the bulk soil in relation to fine-root parameters, alder species, and soil type. Biolog Ecoplates were used to determine community-level physiological profiles (CLPP) of culturable bacteria in soil–root interface and bulk soil samples. CLPP were summarized as AWCD (average well color development, OD 48 h−1) and by Shannon diversity index, which varied between 4.3 and 4.6 for soil–root interface. The soil–root interface/bulk soil ratio of AWCD was estimated. Substrate-induced respiration (SIR) and basal respiration (BAS) of bulk soil samples were measured and metabolic quotient (Q = BAS/SIR) was calculated. SIR and Q varied from 0.24 to 2.89 mg C g−1 and from 0.12 to 0.51, respectively. Short-root morphological studies were carried out by WinRHIZOTM Pro 2003b; mean specific root area (SRA) varied for grey alder and black alder from 69 to 103 and from 54 to 155 m2 kg−1, respectively. The greatest differences between AWCD values of culturable bacterial communities in soil–root interface and bulk soil were found for the young alder stands on oil-shale mining spoil and on abandoned agricultural land. Soil–root interface/bulk soil AWCD ratio, ratio for Shannon diversity indices, and SRA were positively correlated. Foliar assimilation efficiency (FOE) was negatively correlated with soil–root interface/bulk soil AWCD ratio. The impact of soil and alder species on short-root morphology was significant; short-root tip volume and mass were greater for black alder than grey alder. For the investigated microbiological characteristics, no alder-species-related differences were revealed.  相似文献

Truffles are edible hypogeous ectomycorrhizal fungi which have great economic importance for their organoleptic properties and have significant ecological interests for forestry. Although some new precious Chinese white truffle have been described constantly, the molecular mechanisms that control truffle body formation are largely unknown. It has been hypothesized that ectomycorrhizosphere soil communities may have influences on truffle production. Thus, isolation and molecular characterisation of culturable bacteria were carried out to investigate the bacteria diversity in mycorrhizosphere soil of Tuber panzhihuanense Pinus armandii in this work. Sequencing results showed a significant presence mostly affiliated with Burkholderia was β Proteobacteria (3098%). The second culturable fraction which dominated by Pseudomonas was γ Proteobacteria (288%) other isolates were mostly Phyllobacterium and Rhizobium, members of α Proteobacteria (1467%), actinobacteria (125%) and Firmicutes (76%) represented by Arthrobacter and Bacillus, respectively. Chryseobacterium ureilyticum was the only bacterial strain belonging to Bacteroidetes. Similarities and differences of culturable bacterial community of ascocarps and ectomycorrhizosphere soil associated with Tuber were discussed.  相似文献

Bacteria and fungi naturally coexist in various environments including forest ecosystems. While the role of saprotrophic basidiomycetes in wood decomposition is well established, the influence of these fungi on the functional diversity of the wood-associated bacterial communities has received much less attention. Based on a microcosm experiment, we tested the hypothesis that both the presence of the white-rot fungus Phanerochaete chrysosporium and the wood, as a growth substrate, impacted the functional diversity of these bacterial communities. Microcosms containing sterile sawdust were inoculated with a microbial inoculum extracted from a forest soil, in presence or in absence of P. chrysosporium and subsequently, three enrichment steps were performed. First, bacterial strains were isolated from different microcosms previously analyzed by 16S rRNA gene-based pyrosequencing. Strains isolated from P. chrysosporium mycosphere showed less antagonism against this fungus compared to the strains isolated from the initial forest soil inoculum, suggesting a selection by the fungus of less inhibitory bacterial communities. Moreover, the presence of the fungus in wood resulted in a selection of cellulolytic and xylanolytic bacterial strains, highlighting the role of mycospheric bacteria in wood decomposition. Additionally, the proportion of siderophore-producing bacteria increased along the enrichment steps, suggesting an important role of bacteria in iron mobilization in decaying-wood. Finally, taxonomic identification of 311 bacterial isolates revealed, at the family level, strong similarities with the high-throughput sequencing data as well as with other studies in terms of taxonomic composition of the wood-associated bacterial community, highlighting that the isolated strains are representative of the wood-associated bacterial communities.  相似文献

为研究黄河三角洲地区混交人工林土壤细菌群落特征,应用高通量测序技术,比较分析了刺槐臭椿混交林以及臭椿和刺槐纯林土壤细菌结构及多样性,并结合土壤理化性质进行分析。试验结果表明:在细菌门分类水平上,臭椿纯林、刺槐纯林、刺槐臭椿混交林土壤中分别检测出27、25、31门细菌,3种不同林分土壤中酸杆菌门、变形菌门、放线菌门、硝化螺旋菌门、绿弯菌门、浮霉菌门、芽单胞菌门、疣微菌门8种细菌是土壤中的主要细菌群落,其中酸杆菌门、变形菌门和放线菌门为优势细菌群落。不同类型人工林土壤中各门细菌相对丰度差异显著。混交林土壤细菌物种数和Shannon指数值分别为1910和9.1高于两种纯林。通过对土壤主要细菌群落与土壤理化性质进行主成分分析发现,3种不同林分之间在土壤细菌群落结构上有较高程度的分离,差异显著(P < 0.05),有效磷含量与混交林土壤细菌群落有较强的正相关关系。因此可以得出结论,不同林分类型、土壤理化性质和细菌群落结构三者相互影响,刺槐臭椿混交增加了土壤细菌群落多样性,土壤理化性质在一定程度上影响土壤细菌结构和多样性。  相似文献

不同林型土壤的酸化缓冲能力不同,真菌在土壤系统中扮演着重要的角色,而对土壤真菌群落结构和组成与土壤酸化的关系缺乏深入研究。以重庆铁山坪林场的马尾松纯林(Pi)和经马尾松纯林改造后的香樟纯林(Ci)、木荷纯林(Sc)、马尾松-香樟混交林(Pi_Ci)以及马尾松-木荷混交林(Pi_Sc)为研究对象,每个林型分别设置4个20 m×20 m的样地,分别采集腐殖质层(O层)和淋溶层(A层)土壤进行土壤性质及真菌群落分析,以探讨酸雨区森林土壤真菌群落与缓解土壤酸化的关系。研究表明:(1)与Pi相比,Ci土壤酸化明显缓解(高pH低NH4 : NO3),且能有效提高土壤全磷(TP)含量;而Sc虽然土壤pH值与Pi没有显著差异,但显著(P<0.05)提高了NH4 : NO3,且显著降低土壤TP和全钾(TK)含量(P<0.05);(2)不同林型土壤真菌群落多样性以Ci最为丰富,且表征土壤酸化的指标pH值、阳离子交换量(CEC)与真菌多样性显著正相关(P<0.05),NH4 : NO3与多样性显著负相关(P<0.05);(3)林型和土层都对真菌群落结构有显著影响(P<0.001),且林型的影响大于土层的影响;而土壤酸化程度将五个林型的土壤真菌群落区分成两个大类:Ci和Pi_Ci;Pi,Sc以及Pi_Sc。(4) Ci中有益菌(如Mortierella)更多,Pi以外生菌根真菌占优势(Russulaceae、Russula、Tomentella以及Sebacina);Sc以及Pi_Sc则含有更多的植物病原菌(Cladophialophora,Paecilomyces,Venturiales)、嗜酸菌及产酸菌(Paecilomyces,Penicillium)。在酸雨区受损马尾松林地种植香樟促进土壤真菌多样性提高,且产酸真菌、嗜酸菌丰度降低,而有益真菌丰度增加,可有效缓解土壤酸化;而种植木荷后土壤中的病原菌、嗜酸菌和产酸菌相对丰度增加,导致土壤进一步酸化。因此,通过将受酸雨损害严重的马尾松纯林改造成香樟纯林或马尾松-香樟混交林,有助于缓解土壤的酸化,实现酸雨区森林生态系统的可持续发展。  相似文献

树种选择是林下山参护育成败的关键,研究树叶凋落物对人参土壤养分、微生物群落结构组成的影响,旨在为林下山参护育选择适宜林地及农田栽参土壤改良提供科学依据和理论指导。通过盆栽试验,研究添加5.0 g色木槭Acer mono.Maxim.var.mono(A)、赤松Pinus densiflora Sieb.et Zucc.(B)、胡桃楸Juglans mandshurica Maxim.(C)、紫椴Tilia amurensis Rupr.(D)、蒙古栎Quercus mongolica Fisch.ex Ledeb.(E)树叶凋落物到土壤中,种植人参(Panax ginseng C.A.meyer)后研究土壤理化性质以及微生物群落结构的变化。结果表明:添加不同树叶处理后人参土壤性质发生改变,土壤p H值显著高于对照土壤5.91(P0.05),土壤全氮、速效氮磷、微生物碳氮在所有树叶处理中显著增加(P0.05),而土壤容重、速效钾和C/N在添加树叶处理中降低。18个土壤样品基因组,经16S和ITS1测序分别得到6064和1900个OUTs。其中细菌涵盖了42门、117纲、170目、213科、225属,真菌涵盖了24门、98纲、196目、330科、435属。不同树叶处理人参土壤细菌和真菌地位发生改变,细菌Proteobacteria是树叶分解的关键微生物,添加树叶后其多样性显著高于对照(P0.05)。而细菌Bacteroidetes和真菌Basidiomycota可能是区别阔叶林和针叶林树种的关键微生物,针叶林中含量显著低于阔叶林(P0.05),而真菌Ascomycota是针叶林分解的关键微生物。进一步从不同分类水平上得到特定树叶凋落物的特异细菌和真菌。典型相关分析(CDA)表明细菌Bacteroidetes、Chloroflexi、Actinobacteria及真菌Basidiomycota、Zygomycota、Chytridiomycota及Ascomycota的位置及多样性的改变均与土壤因子SMBN、TN、AP、SOC、AK、C/N、p H有关。综上所述,添加不同树叶后不仅提高土壤微生物量碳氮、改善土壤理化性质,同时改变微生物群落结构组成,不同树叶处理土壤理化性质不同导致人参土壤微生物组成的差异,本结果对于林下参选地和农田栽参土壤微生物改良具有理论指导作用。  相似文献

【背景】三七根际微生物群落特征与其土传根腐病害密切相关,而针对自然林下根腐三七的相关研究鲜见报道。【目的】比较分析自然林下与田间根腐三七根际土壤微生物群落的组成特征,结合土壤理化性质与酶活性分析,为三七根腐病害防治与仿野生栽培提供科学依据。【方法】采集自然林下与田间根腐三七根际土壤,利用高通量测序技术,分析土壤细菌与真菌群落的物种组成与多样性,并测定土壤理化性质和酶活性。【结果】自然林下与田间根腐三七根际土壤中细菌和真菌群落组成具有明显差异,自然林下根腐三七根际土壤中担子菌门(Basidiomycota)、酸杆菌门(Acidobacteria)和疣微菌门(Verrucomicrobia)的相对丰度较高,而田间根腐三七根际土壤中子囊菌门(Ascomycota)、变形菌门(Proteobacteria)和绿弯菌门(Chloroflexi)的相对丰度较高。在属分类水平,镰刀菌属(Fusarium)是自然林下根腐三七根际土壤中的优势菌群,相对丰度为17.30%,而癣囊腔菌属(Plectosphaerella)是田间根腐三七根际土壤中的优势菌群,相对丰度为22.55%;Candidatus Ba...  相似文献

During 2003–2005, the diversity of culturable filamentous soil microfungi in saline and acidic soils of the Soos National Natural Reserve (Czech Republic) was studied. Altogether, 28 soil samples were collected from four sampling sites and were processed by various approaches. In total, 92 fungal taxa were identified using classical and molecular markers. Several detected species were known from similar substrata worldwide; however, the overall fungal spectrum was distinct, as shown by comparison to similar studies. All methodological approaches increased the observed fungal diversity. The different fungal communities observed on the four sampling sites were influenced by the complex effects of environmental factors. The growth response of selected strains to different salinities and pH values was determined. The results of the growth tests showed high adaptability of all tested species to the extreme conditions of the studied substrate. Two acidophilic species (Acidomyces acidophilus, Sporothrix sp.) were isolated.  相似文献

We investigated ground beetle communities (Coleoptera, Carabidae) in ancient woodland remnants in north-western Lower Saxony, Germany. A total of 90 pitfall traps was exposed in a stratified design in 10 stands of mature oak–beech and oak–hornbeam forests in the year 2003. Overall, 47 species (10,676 individuals) were recorded. Among these were the two relict species Carabus glabratus and Abax parallelus, and 14 further eurytopic forest species. Eleven species exhibited a high frequency and were found in all of the ten stands. Multiple linear regressions showed several significant relationships at two scales for species richness of different groups of carabids and for several of the measured environmental factors. Forest area, litter depth, amount of dead wood, distance to forest edge, and soil moisture were found to be key factors determining species richness. Furthermore, recent disturbance by logging reduced the number of forest species. According to direct gradient analyses soil moisture and litter depth have greatest influence on species communities of both, forest species and widespread species. Habitat suitability models for the two recorded relict species, A. paralellus and C. glabratus, were developed using logistic regression. The presence of A. parallelus in the mature ancient woodland remnants depends mainly on higher values of soil moisture, whereas for C. glabratus none of the measured environmental variables appeared to be key factors. Implications for the conservation of carabid assemblages in mature ancient woodlands include the advice to spread out logging over long periods of time and over various woodlands in order to keep the stand disturbance at a long-term low level. Variation in logging practices may help to conserve diverse structures. Afforestation with non-native tree species should be avoided in the managed ancient woodlands. Finally, especially the preservation of a high soil moisture seems to be important to conserve typical carabid communities.  相似文献

Sponges are well documented to harbor large amounts of microbes. Both culture-dependent and molecular approaches have revealed remarkable bacterial diversity in marine sponges. Fungi are commonly isolated from marine sponges, yet no reports on phylogenetic diversity of sponge-inhabiting fungi exist. In this report, we investigated the phylogenetic diversity of culturable fungi from the Hawaiian alien marine sponges Suberites zeteki and Gelliodes fibrosa. A total of 44 independent isolates were recovered from these two sponge species, representing 7 orders and 22 genera of Ascomycota. The majority (58%) of fungal isolates from S. zeteki resided in the Pleosporales group, while the predominant isolates (52%) from G. fibrosa were members of the Hypocreales group. Though differing in fungal species composition and structure, culturable communities of these two sponges displayed similar phylogenetic diversity. At the genus level, only two genera Penicillium and Trichoderma in the Eurotiales and Hypocreales orders, respectively, were present in both sponge species. The other genera of the fungal isolates were associated with either S. zeteki or G. fibrosa. Some of these fungal genera had been isolated from sponges collected in other marine habitats, but more than half of these genera were identified for the first time in these two marine sponges. Overall, the diversity of culturable fungal communities from these two sponge species is much higher than that observed in studies of marine sponges from other areas. This is the first report of phylogenetic diversity of marine sponge-associated fungi and adds one more dimension to our current understanding of the phylogenetic diversity of sponge-symbiotic microbes.  相似文献

【目的】通过研究林地转型耕地对土壤编码碱性磷酸酶基因的细菌群落丰度、多样性和结构的影响,为丘陵区耕地长期施肥下农田土壤微生物多样性丧失的影响机制以及未来的退耕还林过程中土壤微生物多样性的提升和土地可持续利用研究提供一些基础数据和技术支撑。【方法】采用实时荧光定量PCR (real-time quantitative PCR,qPCR)和高通量测序技术解析土壤编码碱性磷酸酶基因的细菌群落的丰度、多样性和结构变化,并耦合土壤化学性质分析,明确土壤编码碱性磷酸酶基因的细菌群落丰度和多样性与土壤化学性质的关系以及关键的驱动因子。【结果】林地垦殖为农田后,长期施肥导致土壤酸化,pH从5.58降至4.72,而土壤速效磷则从2.49 mg/kg增至49.3 mg/kg。相应地,耕地土壤编码碱性磷酸酶基因的细菌群落的丰度和Shannon指数均显著低于林地。基于编码碱性磷酸酶的phoD基因(alkaline phosphatase-encoding gene)序列的物种分类表明,丘陵区土壤编码碱性磷酸酶基因的细菌群落的优势门为变形菌门(Proteobacteria)、蓝藻门(Cyanobacteria)、浮霉菌门(Planctomycetes)、放线菌门(Actinobacteria)、厚壁菌门(Firmicutes)和疣微菌门(Verrucomicrobia),其中林地土壤的蓝藻门的相对丰度显著高于耕地。耕地土壤的慢生根瘤菌属(Bradyrhizobium)和芽孢杆菌属(Bacillus)的相对丰度显著高于林地,而中慢生根瘤菌属(Mesorhizobium)、假单胞菌属(Pseudomonas)、Chlorogloea属、Gemmata属、Phormidesmis属和Pseudolabrys属的相对丰度显著低于林地。土壤编码碱性磷酸酶基因的细菌群落结构因林地转型耕地而发生显著改变。phoD基因丰度和Shannon指数与pH显著正相关,而与总磷、速效磷、硝态氮和铵态氮均显著负相关,其中土壤速效磷是这些影响因素中影响最强烈的,长期施用无机磷肥导致含碱性磷酸酶的土壤细菌群落对有机磷分解的能力退化。【结论】林地转型耕地加之长期施肥改变了土壤pH和速效磷,并在其他理化因子的协同驱动下,导致土壤编码碱性磷酸酶基因的细菌群落丰度、多样性和结构的显著变化。  相似文献

为了揭示寒温带针叶林不同树种倒木分解中真菌群落的分布格局,探讨影响倒木真菌群落分布的养分驱动因子。采用Illumina MiSeq高通量测序技术和R语言等分析方法对微生物多样性数据进行信息挖掘,解析白桦(Betula platyphylla Suk.)、兴安落叶松(Larix gmelinii(Rupr.)Kuzen)、樟子松(Pinus sylvestris var. mongolica Litv.)倒木分解初期真菌群落的多样性及功能差异。研究发现,寒温带针叶林中白桦、樟子松倒木优势菌群为Basidiomycota(担子菌门)、Trichaptum(附毛菌属),兴安落叶松倒木优势菌群为Ascomycota(子囊菌门)、Acidea菌属。多样性分析表明,不同树种倒木真菌群落的Alpha多样性由高到低依次为兴安落叶松、白桦、樟子松,树种的差异极显著的改变了操作分类单元(OTU)、属水平上倒木真菌群落的Beta多样性。菌群生态分析表明,腐生型是3种倒木上优势真菌的主要生态类型,木质腐生为白桦、樟子松倒木上真菌的主要生活方式,土壤腐生为兴安落叶松真菌类群的主要生活方式。养分关联分析表明,pH、全碳(TC)、全氮(TN)、碳氮比(C/N)、半纤维素、木质素等养分指标在3种倒木间存在显著性差异,其中含水率(MC)、TC、TN、纤维素、半纤维素是影响倒木优势真菌群落分布的主要养分因素。不同树种对倒木真菌群落的富集具有差异性,这种差异带来的微生物多样性及功能变化对寒温带森林生态系统的物质循环具有重要的科学意义。  相似文献
