Summary C14 and N15 doubly labelled alfalfa tissue at addition rates of 5 and 1 ton per acre was incubated in the laboratory for 72 days with virgin and cultivated Brookston silt loam. The alfalfa tissue was more extensively decomposed in the virgin soils than in cultivated soil, but retention of tissue carbon was not affected by rate of addition. Percentage decomposition of organic matter in the virgin soil was greater than that in the cultivated soil. Addition of alfalfa tissue reduced decomposition of soil organic matter in proportion to the rate of addition and resulted in a gain of carbon in the incubation mixture. No priming action was noted.An increase in the rate of tissue addition caused an increase in the amount of nitrogen mineralized from the tissue but had little effect on the amount of nitrogen mineralized from the soil. Nitrogen mineralized from the soil organic matter was preferentially immobilized during the latter part of the incubation period.It appears that the organic matter content of the soil as well as the rate of tissue addition may regulate the priming action of green manures.Published with approval of the Director, Ohio Agricultural Experiment Station as Journal Paper No.94-62.  相似文献

The mode of transformation of dehydroepiandrosterone (I, 3-hydroxyandrost-5-en-17-one) and pregnenolone (II, 3-hydroxypregn-5-en-20-one) was studied using Mucor piriformis. Biotransformation products formed from I were 3,17-dihydroxyandrost-5-ene (Ia), 3-hydroxyandrost-5-ene-7,17-dione (Ib), 3,17-dihydroxyandrost-5-en-7-one (Ic), 3,7-dihydroxyandrost-5-en-17-one (Ie). Biotransformation products formed from compound II were 3,7-dihydroxypregn-5-en-20-one (IIa) and 3,7,11-trihydroxypregn-5-en-20-one (IIb). The organism did not carry out isomerization of the 5-en-3-ol to a 4-en-3-one system in the steroid molecules tested. In addition, it failed to carry out 14-hydroxylation possibly because of the lack of a 4-en-3-one system in I and II, and stereospecific hydroxylation at the C-7 position in I and II.  相似文献

Summary Phosphorus supply factors (capacity, kinetic, intensity, and diffusivity) and plant growth were the approaches used to assess P supply of flooded rice soils. Increases in the capacity, intensity, and kinetic factors, as measured by E-value, solution P concentration, and soil P release rate to a distilled water sink respectively, were unpronouced and infrequent upon water-saturation of ten soils. However, increases in the diffusitivity factor, as measured by 32P diffusion coefficients, were at least ten-fold as soil moisture increased. The greatest increases in P diffusion occurred as soil moisture increased beyond one-third bar.Using a P fertilized soil or P treated powdered cellulose as the P source and a minus P nutrient solution to nourish a split root system with water and nutrients, data were obtained which suggested that P uptake and rice-shoot growth (indicators of P availability) increased with increasing moisture level. Phosphorus uptake and rice-shoot growth were greatst when the soil or P treated cellulose were water-saturated. A new synthetic brassinolide analogue, 2,3-dihydroxy-17-(3-methylbutyryloxy)-7-oxa-B-homo-5-androstan-6-one (11), has been shown to exhibit typical brassinolide activity characterised by elongation, swelling, twisting and splitting of the bean second internode. It was prepared from the known lactone 2,3,17-trihydroxy-7-oxa-B-homo-5-androstan-6-one (4) which was transformed to an isopropylidenedioxy derivative. After protection of the 2- and 3-hydroxy groups it yielded the 2,3-isopropylidenedioxy-17-(3-methyl-butyryloxy)-7-oxa-B-homo-5-androstan-6-one (7) on treating with 3-methylbutyryl chloride in pyridine. The analogue with a 2-methylbutyric moiety (10, 2,3-dihydroxy-17-(2-methyl-butyryloxy)-7-oxa-B-homo-5-androstan-6-one) in position 17 stimulated only elongation and swelling of the bean second internode. However, in this bioassay 100 times more 10 or 11 compared to 24-epibrassinolide is required to obtain the same effects. Analogues with -oriented hydroxyl groups at C-2 and C-3 (14,15), a 6-ketone (17,18) or 6-oxa-7-oxo-lactone system (12,13) in ring B lack the typical brassinolide activity. In addition, the active brassinosteroids applied to the second internode stimulated a similar, but 30% lower elongation of the first internode. From data presented here we conclude that the presence of two hydroxy groups in the positions 22 and 23 of the brassinolide side chain, which are considered as a key structural requirement, is not absolutely necessary for a compound to exhibit typical brassinosteroid activity. Nevertheless, these compounds have generally 2–10 times lower activity than that having 22,23-vicinal diol in the side chain.  相似文献

Glycosphingolipids (GSLs) contain many carbohydrate epitopes or crypto-glycotopes for Gal and GalNAc reactive lectins. Many of them are in the nervous system and function as important receptors in various life processes. During the past two decades, 11 mammalian structural units have been used to express the binding domain of applied lectins. They are: F, GalNAc1 3GalNAc; A, GalNAc1 3Gal; T, Gal1 3GalNAc; I, Gal1 3GlcNAc; II, Gal1 4GlcNAc; B, Gal1 3Gal; E, Gal1 4Gal; L, Gal1 4Glc; P, GalNAc1 3Gal; S, GalNAc1 4Gal, and Tn, GalNAc1 Ser(Thr). Although 10 of them occur in GSLs, only 3 (L , S , and T ) are found in human brain, and 2 (L and II ) are present in the inner structures of human blood group active GSLs. In the families of gangliosides, L and II represent 55% of the total structural units, while the other three units (T , P , and S ) constitute the rest. To facilitate the selection of lectins that could serve as structural probes, the carbohydrate binding specificities of Gal/GalNAc reactive lectins have been classified according to their highest affinity for the structural units and their binding properties expressed by decreasing order of reactivity. Hence, the binding relation between GSLs and Gal/GalNAc specific lectins can be established.  相似文献

The interaction of short nucleotide duplexes with bis-netropsins, in which netropsin fragments are linked tail-to-tail via cis-diammineplatinum group (Nt–Pt(NH 3 )–Nt) or aliphatic pentamethylene chain (Nt–(CH 2 ) 5 –Nt), has been studied. Both bis-netropsins have been shown to bind to DNA oligomer 5"-CCTATATCC-3" (I) as a hairpin with parallel orientation of netropsin fragments in 1:1 stoichiometry. Monodentate binding has been detected upon binding of bis-netropsins to other duplexes of sequences 5"-CCXCC-3" [where X = TTATT (II), TTAT (III), TTTTT (IV), and AATTT (V)] along with the binding of bis-netropsins as a hairpin. The formation of dimeric antiparallel motif between the halves of two bound bis-netropsin molecules has been observed in the complexes of Nt–(CH 2 ) 5 –Nt with DNA oligomers IV and V. The ratio of binding constant of bis-netropsin as a hairpin ( 2) to monodentate binding constant ( 1) has been shown to correlate with the width and/or conformational lability of DNA in the binding site. The share of bis-netropsin bound as a hairpin decreases in the order: TATAT > TTATT > TTAAT > TTTTT > AATTT, whereas the contribution of monodentate binding rises. The minimal strong binding site for Nt–Pt(NH 3 )–Nt and Nt–(CH 2 ) 5 –Nt binding as a hairpin has been found to be DNA duplex 5"-CGTATACG-3".  相似文献

Summary A detailed conformational analysis was performed for the synthetic branched trisaccharide -d-Man-NAc-(14)-[-d-Glc-(13)]-l-Rha 1 which represents the repeating unit of the O-antigenic polysaccharide of Aeromonas salmonicida. The study was based on 26 experimental NOE curves from 1D transient NOE experiments, employing Gaussian-shaped inversion pulses at 600 MHz. Eight of the NOE curves were interglycosidic and thus useful for an analysis of glycosidic linkage orientations. Metropolis Monte Carlo (MMC) simulations and minimum-energy calculations with the program GEGOP were used to obtain theoretical NOE curves which were compared to the experimental ones. MMC simulations with different temperature parameters of 310, 600, 900 and 2000 K allowed identification of NOEs which are sensitive towards different conformation distributions-not only different conformations-at both glycosidic linkages in 1. A comparison of trisaccharide 1 with the constituent disaccharides -d-ManNAc-(14)-l-Rha 2 and -d-Glc-(13)-l-Rha 3 revealed effects of branching on glycosidic linkage flexibility. A quantitative evaluation was facilitated by the introduction of entropy-related flexibility parameters. Our study indicates a notable restriction of flexibility, especially at the (13) linkage in 1. Although overall flexibility in 1 is reduced as compared to the constituent disaccharides 2 and 3, it cannot be neglected altogether. In summary, combined transient NOE experiments and MMC simulations provide a simple approach to analyse glycosidic linkage flexibility.  相似文献

Summary Aluminum toxicity resulted in abnormal root development with many short thick roots and was found at pH 4.3 with or without added Al. The toxicity of Mn was found with no added Mn, and with 50 and 100 ppm added Mn at pH values ranging from 4.0 to 4.7 and appeared as dark spots on the leaves. At pH values ranging from 5.8 to 6.0, no toxicity symptoms were recorded in the absence of added Mn. Al was more detrimental to seedling development than was Mn.No kernels developed at 25 and 50 ppm added Al at pH4.1. Such toxicity was associated with Al contents of 9.6 to 28.5 g/ml of saturated extract of soil. The highest kernel yields were recorded at pH 5.8 to 6.0 and were associated with 116 to 296 ppm Mn in tissue and of less than 0.1 g/ml of Al in the saturated extract. Increased rates of Mn and Al resulted in increased concentrations of Ca, Mg, and K in the saturated extract of soil. The results indicated that Al toxicity can be eliminated by liming to soil pH values of greater than 5.5; however, Mn toxicity may occur at pH values as high as 5.8 in the presence of large quantities of Mn.Contribution no. 274, Research Branch, Research Station, P. The natural abundance of 15N was examined in soil profiles from forests and pastures of the Brazilian Amazon Basin to compare tropical forests on a variety of soil types and to investigate changes in the sources of nitrogen to soils following deforestation for cattle ranching. Six sites in the state of Rondônia, two sites in Pará and one in Amazonas were studied. All sites except one were chronosequences and contained native forest and one or more pastures ranging from 2 to 27 years old. Forest soil 15N values to a depth of 1 m ranged from 8 to 23 and were higher than values typically found in temperate forests. A general pattern of increasing 15N values with depth near the soil surface was broadly similar to patterns in other forests but a decrease in 15N values in many forest profiles between 20 and 40 cm suggests that illuviation of 15N-depleted nitrate may influence total soil 15N values in deeper soil where total N concentrations are low. In four chronosequences in Rondônia, the 15N values of surface soil from pastures were lower than in the original forest and 15N values were increasingly depleted in older pastures. Inputs of atmospheric N by dinitrogen fixation could be an important N source in these pastures. Other pastures in Amazonas and Pará and Rondônia showed no consistent change from forest values. The extent of fractionation that leads to 15N enrichment in soils was broadly similar over a wide range of soil textures and indicated that similar processes control N fractionation and loss under tropical forest over a broad geographic region. Forest 15N profiles were consistent with conceptual models that explain enrichment of soil 15N values by selective loss of 14N during nitrification and denitrification.  相似文献

Populations of the ant Camponotus punctulatusundergo demographic explosions after agricultural activities, buildingconspicuous, vegetation-covered soil mounds. We investigated the effects ofC. punctulatus on floristic composition and soilpropertiesalong a gradient of agricultural disturbance in Northeastern Argentina. Wesampled vegetation and soil on and off anthills in,at least, three replicate plots of each of the following situations thatrepresent an increasing gradient of soil disturbance: natural grasslands, sownpastures of Digitaria decumbens, sown pastures ofSetaria sphacelata, and recently abandoned rice fields.Sets of characteristic plant species for each of the land use histories, foron and off anthills as well as for anthills ofdifferent sizes were identified through Indicator Species Analysis. 64% of thevariation in plant community composition was mainly explained by land-usehistory which was associated to the first 2 axes of a Correspondence Analysisbased on the frequency of 126 species across all sites. At the replicate scale,Correspondence Analyses revealed patterns of plant species composition relatedto the presence and size of anthills. Larger mounds became enriched in species,especially herb weeds, in comparison to smaller mounds or samples gatheredoutside the anthills. A Principal Component Analysis of soil data revealed that71% of the variation in soil properties was explained by the presence ofanthills. Soils from on anthills were more fertile than soilsfromoff anthills, independent of land-use history. The fertilityeffect of C. punctulatus mounds in addition with thevegetation patterns observed along the gradient of anthill-sizes highlights theimportance of these ants at the landscape and local scales.  相似文献

To study the influence of soil moisture on phosphorus (P) depletion in the rhizosphere, maize (Zea mays cv. Trak) was pre-grown in vermiculite filled-PVC tubes for 9 days and then the plants with the tubes were transplanted into soil columns maintained at two soil moisture levels () of 0.14 and 0.20 cm3 cm–3 for 10 days. The soil columns were separated at 1 cm depth by a nylon screen of 53 m inner mesh size, into 1 cm soil layer above and 3 cm soil column below screen. A root mat developed over the screen, but root hairs only could penetrate it. Regardless of the soil moisture level in the columns, and adequate and equal water and nutrients supply was maintained via wicks from an external nutrient solution to the plant roots in vermiculite. After 10 days, the soil columns were separated from the root mats, quickly frozen in liquid nitrogen and sliced into thin layers (0.2mm) using a refrigerated microtome to give soil samples at defined distances from the root mats for analyses. Lower soil moisture (=0.14) resulted in narrower and steeper depletion profile of 0.5 M NaHCO3 extractable P (NaHCO3-Pi) as compared to higher soil moisture (=0.20). Depletion of P in soil solution in the immediate vicinity of root mats did not differ much but the extension of the depletion zones was 0.10 cm at =0.14 and 0.20 cm at =0.20. The depletion up to 0.05cm with =0.14 and up to 0.07 cm with =0.20 was uniform, and may be attributed to the depletion in the root hair zone. Beyond the root hair zones, the theory of diffusion and mass flow was able to explain the observed differences in shape and extent of the P depletion profiles at the two soil moisture levels.  相似文献

The effects of trees and contamination on microbial metabolic activity, especially that of hydrocarbon degrading bacteria, were compared during phytoremediation to find which conditions increase diesel fuel removal. Diesel fuel utilisation, microbial extracellular enzyme activities and utilisation of Biolog ECO plate carbon sources by soil bacteria were determined during phytoremediation experiments consisting of two separate diesel applications. Diesel fuel removal after 28 days of second diesel application was 20–30% more than after the first application 1 year earlier. Soil microbiota utilised 26–31 of the 31 Biolog ECO plate carbon sources. Carbon source utilisation profiles indicated minor differences in microbiota in soil vegetated with pine compared to microbiota in soil vegetated with poplar. The potential maximum rates of aminopeptidase activity were 10–102 M AMC/h/g dry soil prior to and after second diesel application, except 14days after the second diesel addition, where the rates were at the scale of 103M AMC/h/g dry soil. The potential maximum rates of esterase activity were 103–104M MUF/h/g dry soil. The presence of plants did not influence the activity of esterases. The utilisation of diesel by soil bacteria in Biolog MT2 plate assay was higher in contaminated soil, especially when vegetated, than in uncontaminated soil, measured both as lag times and maximum specific utilisation rates. MT2 plate assay detected the biological response after diesel fuel addition better than general activity methods.  相似文献

Summary The effect of incorporating tea prunings with soil on some edaphic characteristics was examined. During the first two months of incorporation tea prunings decomposed rapidly and brought about an increase in the availability of N and K and the organic matter content both in the surface (0–3) and sub-surface (3-6) soil. Incorporation of prunings reduced leaching of added N and K and the application of fertilizer N tend to reduce K availability in soil.  相似文献

The biometric analysis of bacterial cells in soil by light, fluorescence, and scanning electron microscopy showed that their average size is 0.8 m in diameter, 1.4 m in length, and 0.7 m3 in volume. In soil loci with enhanced microbiological activity (the rhizoplane of plants and the intestinal tract of soil invertebrates), the average size of bacterial cells was found to be 40% smaller than that of cells occurring in other parts of soil. This is the first experimental evidence showing that the metabolic activity of soil bacteria, their concentration, and allometric parameters are related.  相似文献

Molecular recognition between 4aS/R-galanthamine diastereoisomers (1: 4aS-galanthamine; 2: 4aR-galanthamine) and -cyclodextrin (-CD) were studied by use of docking and molecular dynamics (MD) simulation approaches. The binding energy of constructed 2···-CD complexes is ~17 kcal mol–1 lower than that of 1···-CD, implying a stronger binding ability of 2 with -CD than that of 1. The theoretical modeling result is consistent with our previous CZE result, which demonstrated that -CD is an efficient chiral additive for separating 1 and 2. The modeling result also indicates that both hydrophobic interaction and H-bond force may work as major factors for molecular recognition between the galanthamine diastereoisomers and -CD. Relationships of vegetation, climate and soils in Shanxi plateau wereanalyzed by use of Canonical Correspondence Analysis (CCA). Shanxi province,located at 34°35–40°43 N, 110°15–114°33 E, was divided into a series of rectangular districts of30 latitude by 20 longitude. Areas of vegetation formations and soil types ineach district were measured carefully using fine grain on the vegetation andsoil maps of Shanxi. Climatic data were mean values of 25 years records in eachdistrict. Three data matrices of climate, vegetation and soil were combined byCCA. The results showed that the distribution of vegetation is closely relatedto the variety of climates and to soils distribution.  相似文献

Changes in microbial populations were evaluated following inoculation of contaminated soil with a 3-chlorobenzoate degrader. Madera sandy loam was amended with 0, 500, or 1000 g 3-chlorobenzoate g-1 dry soil. Selected microcosms were inoculated with the degrader Comamonas testosteroni BR60. Culturable bacterial degraderswere enumerated on minimal salts media containing 3-chlorobenzoate. Culturableheterotrophic bacteria were enumerated on R2A. Isolated degraders were grouped by enterobacterial repetitive intergenic consensus sequence-polymerase chain reaction fingerprints and identified based on 16S ribosomal-DNA sequences. Bioaugmentation increased the rate of degradation at both levels of 3-chlorobenzoate. In both the 500 and 1000 g 3-chlorobenzoate g-1 dry soil inoculated microcosms, degradersincreased from the initial inoculum and decreased following degradation of 3-CB.Inoculation delayed the development of indigenous 3-chlorobenzoate degrading populations. It is unclear if inoculation altered the composition of indigenous degrader populations. In the uninoculated soil, degraders increased from undetectable levels to 6.6 × 107 colony-forming-units g-1 dry soil in the 500 g 3-chlorobenzoate g-1 dry soil microcosms, but none were detected in the 1000 g 3-chlorobenzoate g-1 dry soil microcosms. Degraders isolated from uninoculated soil were identified as one of two distinct Burkholderia species.In the uninoculated soil, numbers of culturable heterotrophic bacteria initially decreased following addition of 1000 g 3-chlorobenzoate g-1 dry soil. Inoculation with C. testosteroni reduced this negative impact on culturable bacterial numbers. The results indicate that bioaugmentation may not only increase the rate of 3-chlorobenzoate degradation but also reduce the deleterious effects of 3-chlorbenzoate on indigenous soil microbial populations.  相似文献

Measurements of root-length density (RLD) in a range of 31 apple, kiwifruit, peach, Asian pear and grape orchards were used to derive indices to describe the exploration and exploitation of rooting volumes. Orchards were of various ages and located on a range of soil types, geographic regions, management systems etc. Data were obtained from core samples of volume 1.66×10-4 m3 randomly taken within a standard volume, determined by average planting grids, of 2 m radius centred on tree stems, and 1 m depth. Root systems were described using an exploitation index, E(), and an exploration index, E(0). E() is defined as the proportion of the soil volume which contains roots at RLD greater than or equal to some specified value, . E(0) is defined as the proportion of the soil volume which contains roots at any RLD greater than zero. These indices are dependent on sample size, as are all volumetric or soil-coring data.Estimates of E(0) for each orchard were obtained as the proportions of cores containing any RLD>O and assessed for dependence on species. Peach trees had a significantly higher value of E(0), equal to almost 1.0, compared to the other four species where E(0) was approximately 0.8 (p0.01) or less. There was also some variation with age. E(0) was lower for very young plants which had not fully occupied the sampled soil volume. Exploration indices for woody roots increased with rootstock age but otherwise did not explain large differences in E() between species for given values.For example at =0.05×104 m.m-3, E() was approximately 0.45 for peach and kiwifruit, and 0.05 for apple, Asian pear and grape, whereas at =0.5×104 m.m-3 the corresponding values were 0.1 and almost zero. Negative exponential curves relating E(), scaled by dividing by E(0), to were fitted for each of the 31 orchards. Exponents for these curves, k, were significantly smaller for kiwifruit and peaches than apples, grapes and Asian pears (p0.05), and smaller for apples than grapes and Asian pears (p0.05). A larger k implies a rapid fall-off in E() as increases. Although all five species contained zero and low RLD samples, only kiwifruit and peaches contained higher RLD values and consequently have higher mean RLD. This trend was consistent across all soils, regions, sampling dates, and plant ages.The analyses demonstrate that core sampling can give useful insights into macro-scale root-system distribution, such as the proportion of a soil volume explored and how it is exploited. If positions of core samples are noted during sampling using angular direction, depth and radial distance as spatial coordinates the method can be used to describe root-system structures.  相似文献

Vertical stratification of plant-available K in vermiculitic soil profiles contributes to a late-season K deficiency that limits cotton (Gossypium hirsutum L.) yields on affected soils. Split-root solution culture and split-pot soil experiments were conducted to determine whether root distribution and cultivar differences in root extension in these stratified profiles result from a compensatory response to localized enrichment with NO3-N, PO4-P, and/or K in the root zone. Compensatory root growth was greatest in response to localized NO3-N enrichment. For two cultivars examined in solution culture, 74% of new root development occurred in the half-pot providing 90% of the total NO3-N supply. Only 60% of cultivar root development occurred in the half-pot providing 90% of the PO4-P. No compensatory root growth was observed in response to localized K enrichment. In the split-pot system, the proportion of total root surface area developing in a half-pot was highly correlated with localized soil NO3-N levels (r2=0.81), while increased K availability in one half of the root zone did not affect root distribution. Mean soil NO3-N supply to the whole root system determined shoot N accumulation (r2=0.97). Shoot K accumulation was not related to soil K availability but was strongly correlated with mean root surface area density (r2=0.86). Cultivar Acala GC510, known to be less sensitive to K deficiency than Acala SJ-2, had significantly larger root diameter in all nutrient-supply environments. Under conditions of K stress, Acala GC510 had increased root branching and allocated greater dry matter to roots relative to shoots than Acala SJ-2. The results demonstrate that K acquisition by cotton is strongly influenced by the quantity and distribution of NO3-N in the root zone through its effects on root proliferation, and that distinct cultivar differences associated with crop performance on low K soils can be detected in short-term, solution culture growth systems.  相似文献
