Mutations in leucine‐rich repeat kinase 2 (LRRK2) represent the most frequent genetic lesions associated with Parkinson's disease (PD). LRRK2 function and the pathogenic mechanisms of LRRK2 genetic mutations remain poorly understood. Cho et al report in this issue of The EMBO Journal a new interaction between LRRK2 and the ER exit site (ERES) protein Sec16A, important for anterograde transport of secretory proteins. Moreover, a disease‐associated mutation disrupts LRRK2–Sec16A interaction and ERES function. 相似文献

The estimated lower limb length (0.761–0.793 m) of the partial skeleton of Australopithecus afarensis from Woranso‐Mille (KSD‐VP‐1/1) is outside the previously known range for Australopithecus and within the range of modern humans. The lower limb length of KSD‐VP‐1/1 is particularly intriguing when juxtaposed against the lower limb length estimate of the other partial skeleton of A. afarensis, AL 288‐1 (0.525 m). A sample of 36 children (age, >7 years, trochanteric height = 0.56–0.765 m) and 16 adults (trochanteric height = 0.77–1.00 m) walked at their self‐selected slow, preferred, and fast walking velocities, while their oxygen consumption was monitored. Lower limb length and velocity were correlated with slow (P < 0.001, r2 = 0.44), preferred (P < 0.001, r2 = 0.55), and fast (P < 0.001, r2 = 0.69) walking velocity. The relationship between optimal velocity and lower limb length was also determined and lower limb length explained 47% of the variability in optimal velocity. The velocity profile for KSD‐VP‐1/1 (slow = 0.73–0.75 m/s, preferred = 1.08–1.11 m/s, and fast = 1.48–1.54 m/s) is 36–44% higher than that of AL 288‐1 (slow = 0.53 m/s, preferred = 0.78 m/s, and fast = 1.07 m/s). The optimal velocity for AL 288‐1 is 1.04 m/s, whereas that for KSD‐VP‐1/1 is 1.29–1.33 m/s. This degree of lower limb length dimorphism suggests that members of a group would have had to compromise their preferences to walk together or to split into subgroups to walk at their optimal velocity. The question raised in the title was answered. (3R, 3'S)-meso-Zeaxanthin was submitted to iodine catalyzed photochemical stereoisomerisation. The enantiomeric (9Z) and (9'Z) geometrical isomers were isolated by semipreparative HPLC and separated as diastereomeric dicarbamates on a chiral column only. Cleavage of the carbamate could not be effected. CD-Spectra of (1"S, 1"S)- and (1"R, 1"R)-dicarbamates of geometrical isomers of (3R, 3'R)- and (3R, 3'S)-meso-zeaxanthin were systematically studied and the contribution from the carbamate moieties revealed. It was concluded that (9Z, 3R, 3'S)-"meso"-zeaxanthin, in spite of having no symmetry elements, is optically inactive. The result has been rationalised in line with the current hypothesis on the origin of carotenoid CD spectra. 相似文献

The rice paddy eel, Pisodonophis boro (P. boro), is of special interest because of its peculiar burrowing habits. P. boro penetrates the substrate tail-first, a technique common for ophichthids, but it is able to burrow head-first as well. P. boro exhibits three feeding modes: inertial feeding, grasping, and spinning. Rotational feeding is a highly specialized feeding mode, adopted by several elongate, aquatic vertebrates and it is likely that some morphological modifications are related to this feeding mode. The detailed morphology of the head and tail of P. boro is examined with the goal to apportion the anatomical specializations among head-first burrowing, tail-first burrowing, and rotational feeding. The reduced eyes, covered with thick corneas may be beneficial for protection during head-first burrowing, but at the same time decreased visual acuity may have an impact on other sensory systems (e.g. cephalic lateral line system). The elongated and pointed shape of the skull is beneficial for substrate penetration. The cranial bones and their joints, which are fortified, are advantageous for resisting high mechanical loads during head-first burrowing. The aponeurotic connection between epaxial and jaw muscles is considered beneficial for transferring these forces from the body to the head during rotational feeding. Hypertrophied jaw muscles facilitate a powerful bite, which is required to hold prey during spinning movements and variability in the fiber angles of subdivisions of jaw muscles may be beneficial for preventing the lower jaw from being dislodged or opened. Furthermore, firm upper (premaxillo-ethmovomerine complex) and lower jaws (with robust coronoid processes) and high neurocranial rigidity are advantageous for a solid grip to hold prey during rotational feeding. The pointed shape of the tail and the consolidated caudal skeleton are beneficial for their tail-first burrowing habits. It is quite likely that the reduction of the caudal musculature is related to the tail-first burrowing behavior because the subtle movements of the caudal fin rays are no longer required. 相似文献

Poly(ADP-ribose) polymerase-1 (PARP-1) safeguards genomic integrity by limiting sister chromatid exchanges. Overstimulation of PARP-1 by extensive DNA damage, however, can result in cell death, as prolonged PARP-1 activation depletes NAD(+), a substrate, and elevates nicotinamide, a product. The decline of NAD(+) and the rise of nicotinamide may downregulate the activity of Sir2, the NAD(+)-dependent deacetylases, because deacetylation by Sir2 is dependent on high concentration of NAD(+) and inhibited by physiologic level of nicotinamide. The Sir2 deacetylase family has been implicated in mediating gene silencing, longevity and genome stability. It is conceivable that poly(ADP-ribosyl)ation by PARP-1, which is induced by DNA damage, could modulate protein deacetylation by Sir2 via the NAD(+)/nicotinamide connection. The possible linkage of the two ancient pathways that mediate broad biological activities may spell profound evolutionary roles for the conserved PARP-1 and Sir2 gene families in multicellular eukaryotes. 相似文献

The capacity of local communities to control introduced plants is called biotic resistance. Biotic resistance has been almost exclusively tested for plant competition and above ground herbivores and pathogens, while neglecting root herbivores and soil pathogens. Here, we present biotic resistance by above- and below ground herbivores in concert, and relate the abundance of the plant enemies to the species diversity of the local plant communities. The study was carried out in a 7-year-old biodiversity field experiment. We used creeping thistle (Cirsium arvense) as a model, and quantified sap-sucking herbivores: above ground aphids, their antagonists, and root-feeding nematodes. As plant diversity treatments, we used field plots sown with high (15 plant species, HSD) or low (4 plant species, LSD) diverse seed mixtures in 1996 and that were not weeded. Creeping thistle became established spontaneously at the start of the experiment. In 2002, in HSD plots, 90 % of the plant community was made up by 11 species, compared to seven species in LSD plots. No differences were found for C. arvense abundance or biomass. Above ground, three aphid species were found on C. arvense-Uroleucon cirsii, Aphis fabae, and Macrosiphum euphorbiae, but the latter was found only in low densities. Significantly more aphid species were found on individual plants in HSD plots. Moreover, in HSD plots, on average 10 % of aphids were parasitized, while no parasitism was observed in LSD plots. In the root zone of C. arvense, significantly more nematodes were found in HSD than in LSD plots, and a significantly higher proportion of those nematodes were plant parasites. The dominant plant parasitic nematode in both treatments was Paratylenchus. We conclude that biotic resistance by natural enemies may be enhanced by plant species diversity, but that above- and below ground sap-sucking herbivores do not necessarily have to respond similarly to the diversity of the surrounding plant community. 相似文献

Male mate guarding can take many forms but often involves aggression toward male conspecifics and continued proximity with a female. This study describes a previously undocumented behavior in a promiscuous primate, the ring‐tailed lemur: post‐ejaculatory (PE) mounting. PE mounting was documented across eight mating seasons in a ring‐tailed lemur colony on St. Catherines Island (SCI), USA. During PE mounting, a male remounts a female following ejaculation and clasps her midsection as if to mate again, but copulation does not occur; males showing this behavior typically lack erections, and their mounts show an absence of penile intromission and rhythmic thrusting. Male PE mounting was more common among males mating earlier in the queue, and when PE mounting occurred, it accompanied mate guarding. Four non‐mutually exclusive hypotheses to explain PE mounting were evaluated as follows: (1) gaining additional copulations, (2) prevention of re‐mating, (3) lengthening sperm residence time, and (4) re‐mounting as a function of female proceptivity. Male PE mounting did not aid males in gaining additional copulations nor did PE mounting prevent females from mating with new males. Equivocal support was found for Hypothesis 3: although there was much overlap in copulatory plug residence times for males who did and did not show PE mounting, one‐third of males who practiced PE mounting had plug residence times of 2 h or more, much longer than that of males who did not show PE mounting. PE mounting may therefore be related to increased plug residence time, which may provide an advantage to males in sperm competition. Strong evidence was found in support of Hypothesis 4: males overwhelmingly performed PE mounts in response to continued female sexual presentations, suggesting that females can solicit this male behavior. Females consequently exercise an even greater degree of control over males than was previously realized in this ‘female dominant’ primate. 相似文献

Data from National Health and Nutrition Examination Surveys (NHANES) 1999-2004 have recently shown the percent body fat of American adults. Average American men and women have ~28 and 40% body fat. When categorized by BMI and age, the data also show high percent body fat values, particularly in lower BMI categories. These data should make one reflect on the health status of Americans in all BMI categories and the use of these data for public health recommendations. 相似文献

Structural (UV) and carotenoid‐based plumage coloration – signals for parental investment? 下载免费PDF全文
Parental care increases parental fitness through improved offspring condition and survival but comes at a cost for the caretaker(s). To increase life‐time fitness, caring parents are, therefore, expected to adjust their reproductive investment to current environmental conditions and parental capacities. The latter is thought to be signaled via ornamental traits of the bearer. We here investigated whether pre‐ and/or posthatching investment of blue tit (Cyanistes caeruleus) parents was related to ornamental plumage traits (UV crown coloration and carotenoid‐based plumage coloration) expressed by either the individual itself (i.e. “good parent hypothesis”) or its partner (i.e. “differential allocation hypothesis”). Our results show that neither prehatching (that is clutch size and offspring begging intensity) nor posthatching parental investment (provisioning rate, offspring body condition at fledging) was related to an individual's UV crown coloration or to that of its partner. Similar observations were made for carotenoid‐based plumage coloration, except for a consistent positive relationship between offspring begging intensity and maternal carotenoid‐based plumage coloration. This sex‐specific pattern likely reflects a maternal effect mediated via maternally derived egg substances, given that the relationship persisted when offspring were cross‐fostered. This suggests that females adjust their offspring's phenotype toward own phenotype, which may facilitate in particular mother‐offspring co‐adaptation. Overall, our results contribute to the current state of evidence that structural or pigment‐based plumage coloration of blue tits are inconsistently correlated with central life‐history traits. 相似文献

Paradox of Bcl‐2 (and p53): why may apoptosis‐regulating proteins be irrelevant to cell death? 总被引:2,自引:0,他引:2
Blagosklonny MV 《BioEssays : news and reviews in molecular, cellular and developmental biology》2001,23(10):947-953
Although the Bcl-2 family members and p53 are involved in the regulation of apoptosis, the status of apoptotic machinery (eg caspases) plays a major role in determining the mode and timing of cell death. If the apoptotic machinery is lost, inhibited, or intrinsically inactivated, the "death stars", Bcl-2 and p53, may become irrelevant to cell death. In this light, high levels of Bcl-2 may indicate that downstream apoptotic pathways are still functional. This explains why Bcl-2 overexpression can be a marker of chemosensitivity and favorable prognosis in certain cancers and why retention of wild-type p53 may manifest inactivation of caspases in aggressive cancers. 相似文献

It is generally believed that the glowing behaviour of lampyrid larvae may be an aposematic display. Moreover, larvae of the common glow-worm ( Lampyris noctiluca ) show at least two other features, which can be used in aposematic strategies. The first is their suggestive colour pattern of yellow-pinkish lateral dots on a jet-black background and the second a possible warning odour. We performed experiments with starlings ( Sturnus vulgaris ) to test, in particular, for the significance of the colour pattern as a warning signal. Learning experiments showed that glow-worm larvae were distasteful and that starlings showed increasing avoidance of the distasteful prey through a learning process. Experiments with mimics and glow-worm larvae with obscured colours showed that starlings recognized glow-worm larvae by their colour pattern. However, there was an important effect of experience as one group of starlings that had previous contact with edible glow-worm mimics, showed delayed avoidance learning and was able to discriminate mimics from glow-worms thereafter. 相似文献

Understanding species‐specific habitat selection is essential to identify how natural systems are assembled and maintained, and how emerging natural and anthropogenic disturbances will affect ecosystem function. In the Neotropics, Peter's tent‐roosting bat (Uroderma bilobatum), known to roost in forests, has become abundant in human‐modified areas. To understand how habitat characteristics in both intact forest and human‐modified areas influence the presence and density of U. bilobatum, we characterized habitat use at two scales (macrohabitat and microhabitat) and used logistic and poisson regressions to determine which habitat characteristics best predicted the presence and density of U. bilobatum within each scale. Moreover, we performed a redundancy analysis to determine which habitat scale explained more variation. As these bats are obligate tent roosters, we used tent as a surrogate for bat presence and density. We found that both macrohabitat and microhabitat scales explained variation in presence and density. Characteristics of the microhabitat scale, however, had higher predictive power, revealing that U. bilobatum preferentially inhabits areas with high density of coconut palms. Coconut palms were introduced recently in the Neotropics and are found only in human‐modified areas. Therefore, we hypothesize that U. bilobatum is expanding its range into these areas following the expanded distribution of this exotic plant species. 相似文献

Is applied nucleation a straightforward, cost‐effective forest restoration approach? Counter‐response to Holl and Zahawi (2018) 下载免费PDF全文

Holl and Zahawi (2018) agree on a number of approaches that we describe in a local case study of tropical montane cloud forest restoration in Mexico. However, they contend our argument that most applied nucleation projects have taken place in tropical lowlands as a mistake. They also provide data on the per‐hectare cost of restoration projects in Costa Rica and compare it to a higher cost figure of plantation‐style reforestation, a view contrary to ours. Last, Holl and Zahawi recognize that applied nucleation requires specialized personnel, but imply that the amount of training needed for this endeavor is not very different than what is required to implement other forest recovery projects. In this counter‐response, we provide some refinements to our original opinion and offer further information in support to our perspective. 相似文献

The α-sheet has been speculated to play a role as a toxic conformer in amyloid diseases. However, except for relatively short fragments, its detection has remained elusive. Here, we present molecular dynamics simulations that support the existence of the α-sheet as a stable, metastable, or long-lived secondary structure in polyglutamine and, to a lesser extent, in polyasparagine aggregates. 相似文献

Do some deep‐sea, sediment‐dwelling species of harpacticoid copepods have 1000‐km‐scale range sizes? 下载免费PDF全文
The range sizes of sediment‐dwelling deep‐sea species are largely unknown. Such knowledge is important because a deep sea composed in large part of species with 100‐km‐scale ranges would be very different from one composed predominantly of species with 1000‐km‐scale ranges. For example, the total species richness would be much greater in the first case than in the second. As a step towards the determination of the distribution of species’ range sizes in the deep sea, we asked whether harpacticoid copepods (Crustacea) on the continental rise in the northeastern Pacific had 1000‐km‐scale range sizes. We chose harpacticoids because they occur widely in deep‐sea sediments and thus are a typical deep‐sea taxon. In addition, they have no pelagic stage in their life history, so they allow a conservative test of hypotheses about species’ range sizes. We used morphology and gene‐sequence data to assign individuals to species. At least 13.3% of the species we studied had 1000‐km‐scale ranges, raising the question of how these species maintain genetic continuity. 相似文献
