A model‐based approach for optimization and cascade control of dissolved oxygen partial pressure (pO2) and maximization of biomass in fed‐batch cultivations is presented. The procedure is based on the off‐line model‐based optimization of the optimal feeding rate profiles and the subsequent automatic pO2 control using a proposed cascade control technique. During the model‐based optimization of the process, feeding rate profiles are optimized with respect to the imposed technological constraints (initial and maximal cultivation volume, cultivation time, feeding rate range, maximal oxygen transfer rate and pO2 level). The cascade pO2 control is implemented using activation of cascades for agitation, oxygen enrichment, and correction of the preoptimized feeding rate profiles. The proposed approach is investigated in two typical fed‐batch processes with Escherichia coli and Saccharomyces cerevisiae. The obtained results show that it was possible to achieve sufficiently high biomass levels with respect to the given technological constraints and to improve controllability of the investigated processes.  相似文献

A double polarographic device is described which permits regulation of dissolved oxygen concentration at any preset pO2 in suspensions or cultures of photosynthetic algae. It also simultaneously permits measurements of the rate of oxygen evolution during photosynthesis. The apparatus is designed to operate in closed vessels in the absence of a gas phase, and may be used at increased pressures tested up to 500 atmospheres. Regulation of oxygen levels was maintained constant at various preset concentrations equivalent to atmospheric pO2 values ranging between 0.02 and 0.21 at 25°C.  相似文献

When excised root nodules ofCoriaria arborea are assayed for nitrogenase activity at various pO2 they show a broad optimum between 20 and 40 kPa O2, with some evidence for adaptation. Continuous flow assays of nodulated root systems of intact plants indicate that Coriaria shows an acetylene induced decline in nitrogenase activity. When root systems were subject to step changes in pO2 nitrogenase activity responded with a steep decline followed by a slower rise in activity both at lower and higher than ambient pO2. Thus Coriaria nodules are able to adapt rapidly to oxygen levels well above and well below ambient. Measurement of nodule diffusion resistance showed that the adaptation is accompanied by rapid increase in resistance at above ambient pO2 and decrease in resistance at below ambient pO2. Plants grown with root systems at pO2 from 5–40 kPa O2 did not differ in growth or nodulation. The anatomy of Coriaria nodules shows they have a dense periderm which encircles the nodule and also closely invests the infected zone. The periderm is both thicker and more heavily suberised in nodules grown at high pO2 than at low pO2. Vacuum infiltration of India ink indicates that oxygen diffusion is entirely through the lenticel and via a small gap adjacent to the stele.  相似文献

Oxygen level in mammalian cell culture is often controlled by placing culture vessels in humidified incubators with a defined gas phase partial pressure of oxygen (pO2gas). Because the cells are consuming oxygen supplied by diffusion, a difference between pO2gas and that experienced by the cells (pO2cell) arises, which is maximal when cells are cultured in vessels with little or no oxygen permeability. Here, we demonstrate theoretically that highly oxygen‐permeable silicone rubber membranes can be used to control pO2cell during culture of cells in monolayers and aggregates much more accurately and can achieve more rapid transient response following a disturbance than on polystyrene and fluorinated ethylene‐propylene copolymer membranes. Cell attachment on silicone rubber was achieved by physical adsorption of fibronectin or Matrigel. We use these membranes for the differentiation of mouse embryonic stem cells to cardiomyocytes and compare the results with culture on polystyrene or on silicone rubber on top of polystyrene. The fraction of cells that are cardiomyocyte‐like increases with decreasing pO2 only when using oxygen‐permeable silicone membrane‐based dishs, which contract on silicone rubber but not polystyrene. The high permeability of silicone rubber results in pO2cell being equal to pO2gas at the tissue‐membrane interface. This, together with geometric information from histological sections, facilitates development of a model from which the pO2 distribution within the resulting aggregates is computed. Silicone rubber membranes have significant advantages over polystyrene in controlling pO2cell, and these results suggest they are a valuable tool for investigating pO2 effects in many applications, such as stem cell differentiation. © 2009 American Institute of Chemical Engineers Biotechnol. Nodulated cowpea (Vigna unguiculata [L.] Walp. cv Vita 3: Bradyrhizobium strain CB756) and soybean (Glycine max [L.] Merr. cv White Eye: Bradyrhizobium strain CB1809) were grown with their root systems maintained in a flowing gas stream containing a range of pO2 (1-80%, v/v) in N2 for up to 28 days after planting. At the extremes of sub- and supra-ambient pO2, the levels of leghemoglobin (Lb) in nodules were reduced. However, neither the proportional composition of Lb component proteins (eight in soybean, three in cowpea) nor their oxidation state was affected by pO2. Short-term changes in pO2 (transferring plants grown with sub- or supra-ambient pO2 in the rhizosphere to air or vice versa) caused a significant decline in Lb content and, in cowpea but not soybean, where pO2 was increased, a higher percentage of oxidation of Lb. Combining data on changes in Lb level of cowpea nodules grown in sub-ambient pO2 with those for their structural adaptation to an under supply of O2 indicated that, despite the nodules having a lower level of Lb, the amount per infected cell was increased by up to twofold and per bacteroid up to fivefold (in those from 1% O2) compared to those grown in air. Progressive decline in pO2 resulted in a progressive increase on this basis, indicating a close relationship between Lb content and the adaptation of nodule functioning to external O2 level.  相似文献

Microwave exposure (2450 MHz, 60 mW/g, CW) of rabbit erythrocytes increases Na passive transport only at membrane phase transition temperatures (Tc) of 17–19°C. This permeability effect is enhanced for relative hypoxia which is characteristic of intracellular oxygen tension (pO2 ? 5 mm Hg). Neither the permeability nor the pO2 effects are observed in temperature-matched (± 0.05°C), sham-exposed controls. In addition, at Tc, microwave exposure is observed to induce the shedding or release of two erythrocyte proteins not seen in sham-exposed controls. Moreover, the enhanced shedding of at least seven other proteins all of molecular weight ? 28,000 D was detected in the microwave-treated samples. Using sensitive silver staining we estimate that approximately 450 fg of protein were shed per erythrocyte. These results demonstrate that temperature and pO2 are important influences on both functional and structural responses of cell membranes to microwave radiation.  相似文献

Summary The respiration rate of individual soybean (Glycine max Merr.) nodules was measured as a function of pO2 and temperature. At 23°, as the pO2 was increased from 0.1 to 0.9 atm, there was a linear increase in respiration rate. At 13°, similar results were obtained, except that there was an abrupt saturation of respiration at approximately 0.5 atm pO2. When measurements were made on the same nodule, the rate of increase in respiration with pO2 was the same at 13° and 23°. Additional results were that 5% CO in the gas phase had no effect on respiration, except for a small decrease in the pO2 at which respiration became saturated. Also, nodules still attached to the soybean root displayed the same respiratory behavior as detached nodules. A model for oxygen transport in the nodule is presented which explains these results quantitatively. The essence of the model is that the respiration rate of the central tissue of the nodule is almost entirely determined by the rate of oxygen diffusion to the respiratory enzymes. Evidence is given that the nodule cortex is the site of almost all of the resistance to oxygen diffusion within the nodule.  相似文献

Nodulated soybean (Glycine max L. Merr. cv White Eye inoculated with Bradyrhizobium japonicum strain CB 1809) plants were cultured in the absence of combined N from 8 to 28 days with their root systems maintained continuously in 1, 2.5, 5, 10, 20, 40, 60, or 80% O2 (volume/volume) in N2. Plant dry matter yield was unaffected by partial pressure of oxygen (pO2) and N2 fixation showed a broad plateau of maximum activity from 2.5 to 40 or 60% O2. Slight inhibition of nitrogenase activity occurred at 1% O2 and as much as 50% inhibition occurred at 80% O2. Low pO2 (less than 10%) decreased nodule mass on plants, but this was compensated for by those nodules having higher specific nitrogenase activities. Synthesis and export of ureides in xylem was maintained at a high level (70-95% of total soluble N in exudate) over the range of pO2 used. Measurements of nitrogenase (EC 1.7.99.2) activity by acetylene reduction indicated that adaptation of nodules to low pO2 was largely due to changes in ventilation characteristics and involved increased permeability to gases in those grown in subambient pO2 and decreased permeability in those from plants cultured with their roots in pO2 greater than ambient. A range of structural alterations in nodules resulting from low pO2 were identified. These included increased frequency of lenticels, decreased nodule size, increased volume of cortex relative to the infected central tissue of the nodule, as well as changes in the size and frequency of extracellular voids in all tissues. In nodules grown in air, the inner cortex differentiated a layer of four or five cells which formed a band, 40 to 50 micrometers thick, lacking extracellular voids. This was reduced in nodules grown in low pO2 comprising one or two cell layers and being 10 to 20 micrometers thick in those from 1% O2. Long-term adaptation to different external pO2 involved changes which modify diffusive resistance and are additional to adjustments in the variable diffusion barrier.  相似文献

Dysfunctions in tissue metabolism can be detected at early stages by oxygen partial pressure (pO2) measurement. The measurement of emission lifetimes offers very promising and non‐invasive approach to estimate pO2 in vivo. This study compares two extensively used oxygen sensors and assesses their in vivo oxygen sensitivity and phototoxic effect. Luminescence lifetime of Ru‐polypyridyl complex and of Pd‐porphyrin is measured in the Chick's Chorioallantoic Membrane (CAM) model with a dedicated optical fiber‐based, time‐resolved spectrometer. The Pd‐porphyrin luminescence lifetimes measured in the CAM model exposed to different pO2 levels are longer and have a broader dynamic range (10–100 μs) than those of Ru‐polypyridyl complex (0.6–1 μs). The combined statistical analysis based on an estimate of the kurtosis and skewness, bootstrapping method and routine normality tests is performed. The indicators of the averages and signal to noise ratio stability are also calculated. The combination of several data processing allows selection of the better sensor for a given application. In particular, it is found that the advantage of Ru‐polypyridyl complex over Pd‐porphyrin is two‐fold: i) Ru‐polypyridyl complex datasets have consistently better statistical characteristics, ii) Ru‐polypyridyl exhibits lower cytotoxicity.

Hypoxia is considered one of the microenvironmental factors associated with the malignant nature of glioblastoma. Thus, evaluating intratumoural distribution of hypoxia would be useful for therapeutic planning as well as assessment of its effectiveness during the therapy. Electron paramagnetic resonance imaging (EPRI) is an imaging technique which can generate quantitative maps of oxygen in vivo using the exogenous paramagnetic compound, triarylmethyl and monitoring its line broadening caused by oxygen. In this study, the feasibility of EPRI for assessment of oxygen distribution in the glioblastoma using orthotopic U87 and U251 xenograft model is examined. Heterogeneous distribution of pO2 between 0 and 50?mmHg was observed throughout the tumours except for the normal brain tissue. U251 glioblastoma was more likely to exhibit hypoxia than U87 for comparable tumour size (median pO2; 29.7 and 18.2?mmHg, p?=?.028, in U87 and U251, respectively). The area with pO2 under 10?mmHg on the EPR oximetry (HF10) showed a good correlation with pimonidazole staining among tumours with evaluated size. In subcutaneous xenograft model, irradiation was relatively less effective for U251 compared with U87. In conclusion, EPRI is a feasible method to evaluate oxygen distribution in the brain tumour.  相似文献

The biological microenvironment is interrupted when tumour masses are introduced because of the strong competition for oxygen. During the period of avascular growth of tumours, capillaries that existed play a crucial role in supplying oxygen to both tumourous and healthy cells. Due to limitations of oxygen supply from capillaries, healthy cells have to compete for oxygen with tumourous cells. In this study, an improved Krogh's cylinder model which is more realistic than the previously reported assumption that oxygen is homogeneously distributed in a microenvironment, is proposed to describe the process of the oxygen diffusion from a capillary to its surrounding environment. The capillary wall permeability is also taken into account. The simulation study is conducted and the results show that when tumour masses are implanted at the upstream part of a capillary and followed by normal tissues, the whole normal tissues suffer from hypoxia. In contrast, when normal tissues are ahead of tumour masses, their pO2 is sufficient. In both situations, the pO2 in the whole normal tissues drops significantly due to the axial diffusion at the interface of normal tissues and tumourous cells. As the existence of the axial oxygen diffusion cannot supply the whole tumour masses, only these tumourous cells that are near the interface can be partially supplied, and have a small chance to survive.  相似文献

The N2-fixing legume nodule requires O2 for ATP production; however, the O2 sensitivity of nitrogenase dictates a requirement for a low pO2 inside the nodule. The effects of long term exposures to various pO2s on N2[C2H2] fixation were evaluated with intact soybean (Glycine max [L.] Merr., var. Wye) plants. Continuous exposure of their rhizosphere to a pO2 of 0.06 atmospheres initially reduced nitrogenase activity by 37 to 45% with restoration of original activity in 4 to 24 hours and with no further change in tests up to 95 hours; continuous exposure to 0.02 atmosphere of O2 initially reduced nitrogenase activity 72%, with only partial recovery by 95 hours. Similar exposures to a pO2 of 0.32 atmospheres had little effect on N2[C2H2] fixation; a pO2 of 0.89 atmospheres initially reduced nitrogenase activity by 98% with restoration to only 14 to 24% of that of the ambient O2 controls by 95 hours. Re-exposure to ambient pO2 of plants adapted to nonambient pO2s reduced N2[C2H2] fixation to similar magnitudes as the reductions which occurred upon initial exposure to variant pO2 conditions, and a time period was required to readapt to ambient O2. It is concluded that the N2[C2H2]-fixing system of intact soybean plants is able to adapt to a wide range of external pO2s as probably occur in soil. We postulate that this occurs through an undefined mechanism which enables the nodule to maintain an internal pO2 optimal for nitrogenase activity.  相似文献

  When Aureobasidium pullulans was grown at a number of agitation rates under batch conditions, exopolysaccharide yields were dramatically reduced at high rates i.e. at least 750 rpm. Investigations with gas blending, which allowed pO2 manipulation and control independently of the agitation rate, showed that this yield reduction was due solely to the high pO2 levels that occurred at these agitation rates. Thus, polysaccharide production at 1000 rpm could be elevated by maintaining the pO2 at a low level during the initial phase of the fermentation. However, both the timing of the pO2 decrease and the level at which it was maintained were crucial for obtaining yields at 1000 rpm, similar to those observed at low agitation rates. The enormous versatility of plants has continued to provide the impetus for the development of plant tissue culture as a commercial production strategy for secondary metabolites. Unfortunately problems with slow growth rates and low products yields, which are generally non-growth associated and intracellular, have made plant cell culture-based processes, with a few exceptions, economically unrealistic. Recent developments in reactor design and control, elicitor technology, molecular biology, and consumer demand for natural products, are fuelling a renaissance in plant cell culture as a production strategy. Dry matter accumulation, nitrogen content and N2 fixation rates of soybean (Glycine max [L.] Merr. cv. Wye) plants grown in chambers in which the aerial portion was exposed to a pO2 of 5, 10, 21, or 30% and a pCO2 of 300 μl CO2/l or a pO2 of 21% and a pCO2 of 1200 μl CO2/l during the complete growth cycle were measured. Total N2[C2H2] fixed was increased by CO2/O2 ratios greater than those in air and was decreased by ratios smaller than those in air; the effects on N2 fixation of decreased pO2 or elevated pCO2 were quantitatively similar during the period of vegetative growth. Decreased pO2 produced a smaller increase then elevated pCO2 during the reproductive period, presumably because of the decreased sink activity of the arrested reproductive growth under subambient pO2. At a pO2 of 5% and a pCO2 of 300 μl CO2/l total N2 fixed was increased 125% and per cent nitrogen content in the vegetative parts was increased relative to air while that in the seed was decreased. Dry matter production was increased and reproductive growth was arrested as previously reported for plants receiving only fertilizer nitrogen. At a pO2 of 30% and a pCO2 of 300 μl CO2/l total N2 fixed was decreased 50% and per cent nitrogen content in the vegetative part was increased relative to air while that in the reproductive structures was unaffected. Dry matter production was similarly decreased in both vegetative and reproductive structures. These effects of altered pO2 in the aerial part on N2 fixation are consistent with the hypothesis that the amount of photosynthate available to the nodule may be the most significant primary factor limiting N2 fixation while sink activity of the reproductive structures may be a secondary factor.  相似文献

Nodulated cowpea (Vigna unguiculata L. Walp. cv Vita 3:Bradyrhizobium CB 756) plants were cultured with their whole root system or crown root nodulation zone maintained for periods from 5 to 69 days after planting in atmospheres containing a range of pO2 (1-80%, v/v) while the rest of the plant grew in normal air. Growth (dry matter yield) and N2 fixation were largely unaffected by pO2 from 10 to 40%. Decrease in fixation at pO2 below 5% was due to lower nodulation and nodule mass and, at pO2 above 60%, to a fall in specific N2-fixing activity of nodules. Root:shoot ratios were significantly lower at pO2 below 2.5%. The effect of pO2 on nitrogenase activity (acetylene reduction), both of whole nodulated root systems and crown root nodulation zones, varied with plant age but was generally lower at supra- and subambient extremes of O2. H2 evolution showed a sharp optimum at 20% O2 but was at most 4% of total nitrogenase activity. The ratio of CO2 evolved to substrate (C2H2+H+) reduced by crown root nodulation zones was constant (6 moles CO2 per mole substrate reduced) from 2.5 to 60% O2 but at levels below 2.5 and above 80% O2 reached values between 20 and 30 moles CO2 per mole substrate reduced. Effects of long-term growth with nonambient pO2 on adaptation and efficiency of functioning of nodules are discussed.  相似文献

The effects of time after exposure to acetylene and of nodule excision were examined using a flow-through system. After a transient depression in the rate of acetylene reduction that began about 1.5 min after exposure to acetylene, the rate recovered to 98% of the initial maximum value after 40 min. After nodule excision the rate stabilized to 90% of the initial maximum value observed in the intact plant.Excised nodules, measured at 6-min intervals in a closed system, with frequent changes of the gas mixture, were used for the remaining experiments. Acetylene reduction by the nodules increased rapidly as temperature was increased between 6 and 26°C. Between 26 and 36°C there was relatively little effect of temperature on acetylene reduction.Nodules and cultures ofFrankia were compared with respect to the effect of temperature and pO2 (partial pressure of oxygen) on oxygen uptake. Cultures ofFrankia were grown on a nitrogen-free medium at either 0.3 kPa O2 (vesicles absent) or 20 kPa O2 (vesicles present). Oxygen uptake by nodules (vesicles absent) and by vesicle-containing cultures was strongly dependent on pO2 at values below 20 kPa. This suggests the presence of a barrier to oxygen diffusion. Oxygen uptake was dependent on temperature as well as on pO2, but the Q10 was much larger for the cultures than for the nodules. This suggests that vesicles or related structures are not the source of the diffusion barrier in Casuarina nodules. Respiration by cultures ofFrankia lacking vesicles became O2-saturated at low pO2 values. Thus these cultures did not have a significant diffusion barrier. From these results it is concluded that nodules ofCasuarina cunninghamiana have a barrier to oxygen diffusion supplied by the host tissue and not byFrankia.  相似文献

Human lymphocytes stimulated with phytohaemagglutinin in vitro show maximal 14C-thymidine incorporation when the gas phase of the cultures contains 20% O2. The rate of DNA synthesis is more affected by hyperoxic conditions than by hypoxia. Tumor hypoxia can be identified by [18F]FAZA positron emission tomography, or invasively using oxygen probes. The impact of anesthetics on tumor hypoxia remains controversial. The aim of this comprehensive study was to investigate the impact of isoflurane and ketamine/xylazine anesthesia on [18F]FAZA uptake and partial oxygen pressure (pO2) in carcinoma and muscle tissue of air- and oxygen-breathing mice.

Methods

CT26 colon carcinoma-bearing mice were anesthetized with isoflurane (IF) or ketamine/xylazine (KX) while breathing air or oxygen (O2). We performed 10 min static PET scans 1 h, 2 h and 3 h after [18F]FAZA injection and calculated the [18F]FAZA-uptake and tumor-to-muscle ratios (T/M). In another experimental group, we placed a pO2 probe in the tumor as well as in the gastrocnemius muscle to measure the pO2 and perfusion.

Results

Ketamine/xylazine-anesthetized mice yielded up to 3.5-fold higher T/M-ratios compared to their isoflurane-anesthetized littermates 1 h, 2 h and 3 h after [18F]FAZA injection regardless of whether the mice breathed air or oxygen (3 h, KX-air: 7.1 vs. IF-air: 1.8, p = 0.0001, KX-O2: 4.4 vs. IF-O2: 1.4, p < 0.0001). The enhanced T/M-ratios in ketamine/xylazine-anesthetized mice were mainly caused by an increased [18F]FAZA uptake in the carcinomas. Invasive pO2 probe measurements yielded enhanced intra-tumoral pO2 values in air- and oxygen-breathing ketamine/xylazine-anesthetized mice compared to isoflurane-anesthetized mice (KX-air: 1.01 mmHg, IF-air: 0.45 mmHg; KX-O2 9.73 mmHg, IF-O2: 6.25 mmHg). Muscle oxygenation was significantly higher in air-breathing isoflurane-anesthetized (56.9 mmHg) than in ketamine/xylazine-anesthetized mice (33.8 mmHg, p = 0.0003).

Conclusion

[18F]FAZA tumor uptake was highest in ketamine/xylazine-anesthetized mice regardless of whether the mice breathed air or oxygen. The generally lower [18F]FAZA whole-body uptake in isoflurane-anesthetized mice could be due to the higher muscle pO2-values in these mice compared to ketamine/xylazine-anesthetized mice. When performing preclinical in vivo hypoxia PET studies, oxygen should be avoided, and ketamine/xylazine-anesthesia might alleviate the identification of tumor hypoxia areals.  相似文献

The effect of oxygen tension above atmospheric pO2 on the development of respiratory capacity in potato disks has been examined. Raising the oxygen tension of the aqueous environment to 40% during the aging of 2.0 mm or 3.0 mm thick disks at 25° progressively increased the respiration rate of the tissue as shown by subsequent assay in 100% oxygen. Disks 3.0 mm thick showed a greater response to increased pO2 than did 2.0 mm disks. A comparison of center 1.0 mm sections excised from 3.0 mm disks after aging, showed that the respiration rate of internal tissue from disks aged in high pO2 was approximately 40% greater than such tissue aged with atmospheric pO2. The characteristic inverse relationship between respiration rate and thickness in aged disks can be modified from a concave-downwards curve to a convex-downwards curve by pretreating the tissue with increased pO2, thus indicating that raising the pO2 during aging can increase the thickness threshold at which the transition from tissue manifesting the respiratory characteristics of thin disks to that manifesting the characteristics of thick disks, occurs. Similarly increased pO2 during aging can modify the hyperbolic relationship obtaining between pretreatment temperature in the range 10° to 25° and respiratory capacity of aged 3.0 mm disks, to approximate to the linear relationship observed with 0.75 mm disks. It is concluded that the development of respiratory capacity in disks between 0.75 mm and 3.0 mm thick is restricted by oxygen dificiency and that the characteristic inverse relationship between respiration rate and thickness in aged disks is largely attributable to this factor, the influence of which is discernible both on the development of respiratory capacity and on its subsequent assay.  相似文献
