Warfarin, specifically labeled with deuterium in the 7 position, was incubated with liver microsomes from untreated rats or rats which were pretreated with either phenobarbital of β-napthoflavone. The four phenolic metabolites (6-, 7-, 8- and 4′-hydroxywarfarin) were isolated and quantitated by GCMS and the percent deuterium retention calculated. In all induction states the 7-hydroxy metabolite of (7,2H)warfarin retained greater than 77% of the deuterium. These results suggest that hydroxylation at the 7 position (meta hydroxylation) cannot proceed by either a direct insertion or abstraction mechanism.  相似文献

The reduction of the Δ1 double bond in 5α-androstan-1-en-3-one was investigated utilizing a reductase present in fermenting baker's yeast. The stereochemistry of the hydrogen addition was found to be trans diaxial, as determined by suitable deuterium labeling and lanthanide induced shift nmr spectroscopy.  相似文献

Transport of Pr3+ across phosphatidylcholine vesicles, monitored through 31P nmr, is first-order in monensin (1), second-order in etheromycin (2) or in lasalocid A (3). When 1 and 2 (or 2 and 3) are incorporated together in 1:1 ratio into the lipidic phase, transport is faster than with each ionophore alone. For instance, assuming that the complexes 2.Pr3+.2, 3.Pr3+.3, and 2.Pr3+3 are equiprobable, they effect transport at intrinsic relative rates of 1, 2, and 13.5, i.e. a remarkable synergism is set up.  相似文献

2-Hydroxymethyl-4-nitrophenyl trimethylacetate is rapidly converted, by an intramolecular pathway, to its benzyl ester counterpart in aqueous solutions of dilute buffers. Intramolecular acyl migration is favored by a factor of 105 over intermolecular transfer of the trimethylacetyl group to surrounding water molecules. The activation parameters of the reaction demonstrate that the rate acceleration is primarily entropic in origin. At constant pH, the apparent first-order rate constant for intramolecular acyl migration displays a linear dependence on the concentration of the basic component of the buffer. For catalysis by imidazole, a solvent deuterium isotope effect of kHkD = 2.4 is observed, in accord with a general base-catalyzed pathway. Similarities between intramolecular and intracomplex transacylations are discussed with the conclusion that the migration of a trimethylacetyl group from the phenolic oxygen atom of a 2-hydroxymethyl-4-nitrophenol to the adjacent benzylic oxygen atom provides an accurate model for acylation of the serine hydroxyl group at the active site of α-chymotrypsin by nitrophenyl esters.  相似文献

Magic-angle cross-polarization 15N nmr spectra of intact lyophilized mycelia from N.crassa cultured on media containing [15N] nitrate have been obtained at 9.12 MHz. The time development of the uptake and distribution of label into protein and amino-acid metabolites can be observed directly. Nitrate metabolism is delayed about one hour if the cells innoculating the culture are grown on nitrate-free medium.  相似文献

A series of unsaturated and polyunsaturated fatty acids with a sulfur atom substituting for a methylene unit of the chain has been prepared and characterized. The syntheses were accomplished by the Wittig coupling of the ylid derived from the triphenylphosphonium salt of 9-bromononanoic acid with aldehydes containing sulfur. The newly formed double bond had predominately the natural Z geometry even when the starting aldehyde was conjugated with the sulfur atom. The sulfides 13-thia-9(Z)-octadecenoic acid (2), 13-thia-9(Z), 11(E)-octadecadienoic acid (5) and 13-thia-9(E), 11(E)-octadecadienoic acid (6) were readily converted into their sulfoxide derivatives by treatment with an equivalent amount of m-chloroperoxybenzoic acid. The structures of the novel compounds were confirmed by the application of ir, uv, 1H-nmr, 13C-nmr, and (as methyl esters) chemical ionization mass spectrometry. Two members of this new family of fatty acids (5 and 6) were found to inhibit the catalysis of the oxygenation of linoleic acid by soybean type-1 lipoxygenase. The analysis of the kinetic data for compound 5 indicated that the type of inhibition was reversible competitive with an inhibition constant of 30 μM.  相似文献

The metabolism of estradiol-17β by the guinea pig uterus late in pregnancy was studied in vivo and in vitro.Whole uteri were examined for estrogen metabolites one hour following an intravenous injection of [3H]-estradiol-17β or uterine sections were examined after incubation for one hour at 37°C in medium containing [3H]-estradiol-17β.In both instances uterine tissue metabolized estradiol-17g to five products: estrone, estrone-3-sulfate, 17β-estradiol-3-sulfate, estrone-3-glucuronide and 17β-estradiol-3-glucuronide. Of the total radioactive products 11 – 43% were glucuronides, 17 – 26% were sulfates and 4 – 17% was estrone. These results indicate that the guinea pig uterus actively transforms estradiol-17β into glucuronides and sulfates late in pregnancy.  相似文献

13C nmr spectra of Streptococcus faecium dihydrofolate reductase containing [13C-guanidino] arginine and ligand complexes with the labeled enzyme are reported. The spectrum of the native enzyme shows 5 well-resolved resonances (the enzyme contains 8 Arg) with a chemical shift range of 1.2 ppm. Addition of ligands to the enzyme produces distinct changes in the enzyme spectrum, and demonstrates that 13C nmr of labeled protein can be used in studies of protein-ligand interactions. An example of the use of 13C-depleted material is also presented.  相似文献

81Br nmr measurements have been made on high (bovine; BCA) and low (human-B; HCAB) specific activity forms of carbonic anhydrase and on a chemically modified form of the human enzyme (carboxyamidomethyl; CAM-HCAB). The high specific activity form of the enzyme, BCA, exhibits a 81Br line broadening which is determined by the lifetime of Br? bound to the zinc ion of the enzyme. The low specific activity form of the enzyme, HCAB, under similar conditions of concentration, pH, etc., does not exhibit a 81Br nmr line broadening. Cl?Br? competitive binding studies, using 35Cl nmr, suggests that the failure to observe 81Br broadening is due to an increase in the lifetime of a zinc bound Br?. An increase in this lifetime by a factor of 10–100 over that exhibited by BCA is sufficient to abolish the line broadening. A modified form of HCAB, CAM-HCAB, does, however, exhibit a 81Br nmr line broadening. Estimates of the lifetime of zinc bound Br?, τM, are 4 × 10?7 sec. for CAM-HCAB at pH 8 and 1 × 10?7 sec. for BCA at pH 7. The lifetime for Br? bound to HCAB is estimated to be ≥10?6 sec.  相似文献

The kinetic α-secondary deuterium isotope effect, kHkD, for the pH-independent hydrolysis of nicotinamide riboside, yielding nicotinamide and ribose, in water at 25 ° is 1.14, establishing that this reaction proceeds with unimolecular substrate decomposition to yield a carboxonium ion, or related species, in the rate-determining step. Surprisingly, the corresponding isotope effect for the base-catalyzed decomposition of the same substrate is 1.12, a value indicating considerable sp2 character at the Cl′ position in the transition state for this reaction. A similar result, kHkD = 1.15, was obtained for base-catalyzed hydrolysis of NAD+. The kinetic alpha deuterium isotope effect for the pig brain NAD glycohydrolasecatalyzed hydrolysis of nicotinamide riboside is 1.08. This value suggests that CN bond cleavage to form an intermediate carboxonium ion, or structurally related species, is at least partially rate-determining. In contrast, the corresponding value for the hydrolysis of this substrate catalyzed by Escherichia coli nicotinamide ribonucleotide glycohydrolase is very near unity, a result consistent with several interpretations including a rate-determining enzyme isomerization reaction.  相似文献

Dipalmitoyl-3-sn-phosphatidylcholine is specifically deuterated at the C-2 position of the fatty acyl chains. Using deuterium magnetic resonance it is then possible to probe the chain conformation in the vicinity the polar head groups. Three separate quadrupole splittings are observed for bilayers of 1,2[2′-2H2] palmitoyl-3-sn-phosphatidylcholine, indicating that the two chains behave differently. The synthesis of phosphatidylcholines each deuterated in only one chain allows the assignment of the three resonances. It is concluded that the beginnings of the two chains have orientations parallel and perpendicular to the bilayer normal. The data further suggest the possibility of two long-lived conformations of the glycerol constituent.  相似文献

(22S)-[22-3H1]-Cholesterol was incubated with an adrenocortical preparation and the isolated (22R)-[22-3H1]-22-hydroxycholesterol had a small loss of radioactivity, proving that direct replacement of the hydrogen from the now hydroxylated position occurred.In addition [1-3H1]-4-methylpentanol was isolated, which also had incurred a relatively small loss of its specific activity, thereby excluding (20R)-3β,20-dihydroxycholest-5-en-22-one as an important metabolite in the degradation of cholesterol to pregnenolone by adrenal tissue.  相似文献

Proton inventory investigations of the hydrolysis N-acetylbenzotriazole at pH 3.0 (or the equivalent point on the pD rate profile) have been conducted at two different temperatures and at ionic strengths ranging from 0 to 3.0 M. The solvent deuterium isotope effects and proton inventories are remarkably similar over this wide range of conditions. The proton inventories suggest a cyclic transition state involving four protons contributing to the solvent deuterium isotope effect for the water-catalyzed hydrolysis. The hydrolysis data are described by the equation kn = ko (1 ? n + nπa1)4 with πa1 ~ 0.74, where ko is the observed first-order rate constant in protium oxide, n is the atom fraction of deuterium in the solvent, kn is the rate constant in a protium oxide-deuterium oxide mixture, and πa1 is the isotopic fractionation factor.  相似文献

We wish to report here the syntheses of (5S, 6R)-5-hydroxy-, (5R, 6R)-5-hydroxy-, (5R, 6S)-5-hydroxy-, and (5S, 6S)-5-hydroxy-PGI1 and their methyl ester derivatives. Treatment of (5R, 6S)-5-epoxy- and (5S, 6R)-5-epoxy-PFG methyl esters with acid washed silica gel afforded (5R, 6R)-5-hydroxy- and (5S, 6S)-5-hydroxy-PGI1 methyl esters; correspondingly, silica promoted cyclization of (5S, 6S)-epoxy- and (5S, 6R)-5-epoxy- PGF1 methyl esters yielded (5S, 6R)-5-hydroxy- and (5R, 6S)-5-hydroxy- PGI1 methyl esters. Alternatively, the 5-hydroxyl group was introduced into the PGI1 skeleton via reaction of the 5-mercuric halides with sodium borohydride in the presence of oxygen. Stereochemical assignments were based on their mode of synthesis and 1H nmr shift differences.  相似文献

Previous proton nuclear magnetic resonance (nmr) studies have indicated that inositol hexaphosphate (IHP) can stabilize hemoglobin (Hb) Kansas in a deoxy-like quaternary structure even when fully liganded with carbon monoxide (CO) (S. Ogawa, A. Mayer, and R. G. Shulman, 1972, Biochem. Biophys. Res. Commun., 49, 1485–1491). In the present report we have investigated both CO binding at equilibrium and the CO binding and release kinetics to determine if Hb Kansas + IHP is devoid of cooperativity, as would be suggested by the nmr studies just quoted. The equilibrium measurements show that Hb Kansas + IHP has a very low affinity for CO (P12 = 1.2 mm Hg and Keq = 5.4 × 105M?1) and almost no cooperativity (n = 1.1) at pH 7, 25 °C. The CO “on” and “off” kinetics also show no evidence for cooperativity. In addition, the equilibrium constant estimated from the kinetic rate constants (Keq = 5.2 × 105M?1 with kon = 1.03 × 105M?1 · S? and koff = 0.198 S?1) is in excellent agreement with the equilibrium constant determined directly. Thus, both kinetic and equilibrium measurements allow us to conclude that CO binding to Hb Kansas + IHP occurs without significant cooperativity.  相似文献

Frozen aqueous suspensions of partially purified membrane-bound renal (Na+ + K+)-ATPase have been irradiated at –135°C with high-energy electrons. (Na+ + K+)-ATPase and K+-phosphatase activities are inactivated exponentially with apparent target sizes of 184 ± 4 kDa and 125 ± 3 kDa, respectively. These values are significantly lower then found previously from irradiation of lyophilized membranes. After reconstitution of irradiated (Na+ + K+)-ATPase into phospholipid vesicles the following transport functions have been measured and target sizes calculated from the exponential inactivation curves: ATP-dependent Na+?K+ exchange, 201 ± 4 kDa; (ATP + Pi)-activated Rb+?Rb+ exchange, 206 ± 7 kDa and ATP-independent Rb+?Rb+ exchange, 117 ± 4 kDa. The apparent size of the α-chain, judged by disappearance of Coomassie stain on SDS-gels, lies between 115 and 141 kDa. That for the β-glycoprotein, though clearly smaller, could not be estimated. We draw the following conclusions: (1) The simplest interpretation of the results is that the minimal functional unit for (Na+ + K+)-ATPase is αβ. (2) The inactivation target size for (Na+ + K+)-dependent ATP hydrolysis is the same as for ATP-dependent pumping of Na+ and K+. (3) The target sizes, for K+-phosphatase (125 kDa) and ATP-independent Rb+?Rb+ exchange (117 kDa) are indistinguishable from that of the α-chain itself, suggesting that cation binding sites and transport pathways, and the p-nitrophenyl phosphate binding site are located exclusively on the α-chain. (4) ATP-dependent activities appear to depend on the integrity of an αβ complex.  相似文献

Oxidative cleavage of aromatic compounds is often part of a degradative process and is widely observed in nature. The immediate catabolic products can sometimes cyclize or rearrange to new secondary metabolites. The enzymatic contraction of a dehydroisocoumarin to yield cyclopentenoid metabolites in Cryptosporiopsis sp. is reported. The label distribution of (+) cryptosporiopsin, a chlorinated cyclopentenone, was determined by analysis of the [13C]nmr of [1-13C] and [2-13C]acetate enriched-cryptosporiopsin. The putative aromatic precursor of cyclopentenoid metabolites, 2,3-dihydro-6,8-dihydroxy-2-methylisocoumarin (6), was isolated from Aspergillus terreus. This metabolite (6) was prepared doubly labeled (T14C). The aromatic origin of the Cryptosporiopsis chlorinated cyclopentenoid metabolites was rigorously proven from feeding experiments with doubly labeled compound 6. A related but nonchlorinated metabolite, terrein, was isolated from A. terreus and was also shown to be derived from [T14C]-2,3-dihydro-6,8-dihydroxy-2-methylisocoumarin.  相似文献

The (Ca2+ + Mg2+)-ATPase of rabbit sarcoplasmic reticulum, when labelled at two Ca2+-protected sites with N-cyclohexyl-N′-(4-dimethylamino-α-naphthyl)carbodiimide (NCD-4) retains Ca2+ binding capacity at the sites with Kd values of approx. 3 μM and 0.12 mM as assessed by fluorescence titration. The sites correspond to the two high-affinity Ca2+ binding sites present in the native ATPase. The NCD-4 labelled ATPase exhibits slow conformational changes at each site on addition of Ca2+. It retains the ability to form phosphoenzyme, and can most likely translocate Ca2+.  相似文献

Growth rates of two lines of tobacco (Nicotianatabacum) cell suspension cultures were measured in the presence or absence of G418, a new 2-deoxystreptamine antibiotic related to Gentamycin. Cell growth rates of N. tabacum cv. Burley were inhibited at drug concentrations as low as 1.65 × 10?7 M. At 4 × 10?7 M, the doubling time was increased from 1.5 days (control) to 2.3 days (treatment). The drug was lethal to cells at 4 × 10?6 M, and inhibition was irreversible. Cells of N. tabacum cv. Wisconsin 38 also were inhibited by the drug, although at slightly higher concentrations (ca. 2–5 fold).In view of our findings, G418 and its associated resistance factors could be of great value in plant genetic engineering.  相似文献
