[4-14C]Estrone was injected intramuscularly into two mature laying Rhode Island Red hens. Radioactive steroids and steroid conjugates recovered from the urine on Amberlite XAD-2 columns were fractionated on columns (100 cm) of DEAE-Sephadex A-25 by NaCl gradients. The presences of the following were confirmed, the figures in brackets indicating average proportions as per cent of total radioactivity recovered after Sephadex column chromatography: -the 3-β-glucuronides of estrone (10. 9) and of estradiol-17α plus estradiol-17β(9.8); the 17-β-glucuronides of estradiol-17α plus estradiol-17β (2.1); the 3-sulfates of estrone (14. 5) and of estradiol-17α plus estradiol-17β (27. 4); and the disulfates of estradiol-17α plus estradiol-17β (2. 3). The following additional conjugates were identified:-a β-glucuronide of 16-epiestriol (0.2) and a β-glucuronide of 16-ketoestradiol-17β (0. 2); the 3-sulfates of 16-epiestriol (1. 4), of 17-epiestriol (0. 9), of 16, 17-epiestriol (0. 7), of 16-keto-estradiol-17β (1. 1), and of 2-methoxyestrone (0. 7). Some evidence was obtained for the presence of 16, 17-epoxy-estratrienol-3-sulfate (1.9).  相似文献

A procedure for the assay of estrone, estradiol-17β or estradiol-17α in plasma is described. The technique also appears applicable to the assay of estriol in plasma. The procedure uses a semi-automatic extraction of plasma, rapid micro-alumina column chromatography and competitive binding of the estrogens to stable proteins of sheep uterine cytosol. The use of alumina column chromatography results in consistently low blanks. The assay has been evaluated for the measurement of estradiol-17β and estrone in human and sheep plasma, and for estradiol-17α and estrone in goat plasma. The change in binding affinity of estradiol-17α relative to estradiol-17β when incubated in sheep uterine cytosol at two different temperatures (25°C and 4°C), makes it possible to differentiate the two epimers of estradiol. Measurement of estradiol-17β down to 10 pg and of estrone and estradiol-17α to 25 pg are maintained in routine analyses. The specificity of the procedure was thoroughly checked by various methods, including comparison with spectrophotofluorimetric analysis.  相似文献

The 9β isomers of estradiol-17β, estradiol-17α, estrone and 17-ethinylestradiol-17β were synthesized and compared with their 9α-counterparts in the rat uterine cytosol estrogen receptor, utero-tropic, and gonadotropin release inhibition assays. Except for 17-ethinyl-9β-estradiol-17β which was as active as its 9α isomer in the uterotropic assay, none of the 9β estrogens exhibited any biological activity which was equal to or greater than their 9α counterparts. For examples, 9β-estradiol-17β was 110 as active as estradiol-17β, and 9β-estrone was 14 as active as estrone in the uterotropic assay.  相似文献

The effects of eleven different steroid hormones on in vitro development of fertilizing capacity by hamster sperm were examined. Capacitation of epididymal sperm occurred only in the presence of female genital tract secretions. Fertilizing ability of sperm was poor if estradiol-17β, cortisol, chlormadinone acetate, medroxyprogesterone acetate, or megestrol acetate were present in the incubation medium at 10?5M, whereas similar concentrations of estradiol-17α, progesterone, norethisterone acetate, ethynodiol diacetate, or norgestrel had little effect. Testosterone was a weak inhibitor of capacitation. Capacitation activity of female uterus and oviduct washings was higher at estrus than diestrus. This activity was reduced by treating intact animals with progesterone, cortisol, or testosterone, but increased by estradiol-17β or HCG. Estradiol-17α has no effect. Activity was low in pregnant or ovariectomized hamsters. Treatment of ovariectomized animals with estradiol-17β increased capacitation activity, but estradiol-17α, HCG or progesterone treatment was ineffective.  相似文献

The interconversion of estradiol-17β and estrone in the rat uterus is due to the action of 17β-hydroxysteroid dehydrogenase. Whole uteri or 800 × g supernatant fractions of the uteri were incubated in the presence of [3H] estradiol-17β and NAD at 37°C for 3 h or 1 h, respectively. In the mature rat uterus the oxidation of estradiol-17β and estrone was dependent on the stage of the estrous cycle, suggesting hormonal control. The 17β-hydroxysteroid dehydrogenase activity was highest at estrus (200 fmol estrone) and lowest at diestrus (80 fmol estrone). An enhancement of activity occurred when adult rats at each stage of the estrous cycle were administered estradiol-17β, while progesterone administration at each stage resulted in decreased enzyme activity. The uterine 17β-hydroxysteroid dehydrogenase activity of estradiol-17β treated ovariectomized rats was time and dose dependent but decreased when progesterone was administered with or without estradiol-17β administration. These results suggest that estradiol-17β caused an increase in enzyme activity that was inhibitable by progesterone in the rat uterus. The increased 17β -hydroxysteroid dehydrogenase activity may reflect a specific response of the rat uterus to estradiol-17β.  相似文献

Synthesis, biochemical and biological testing of the first carborane derivatives of estrogens are described. Estrone 3-carboranylmethyl ether was synthesized in two steps from estrone. Reduction of estrone 3-carboranylmethyl ether with sodium borohydride provided estradiol-17β 3-carboranylmethyl ether. Enzyme kinetic measurements showed that estrone 3-carboranylmethyl ether is a substrate for human placental 17β-hydroxy-steroid dehydrogenase with Km = 5×10?6M, and Vmax = 0.016 μmol min?1 μg?1. The relative affinity constant of estradiol-17β 3-carboranylmethyl ether for rat uterine estrogen receptor was 0.5 (compared with a value of 100 for estradiol-17β). Consistent with its low affinity for estrogen receptor, the dose-dependent uterotropic response to estradiol-17β 3-carboranylmethyl ether in castrated female rats was one sixtieth that of estradiol-17β. None of the tested rats had a toxic reaction to estradiol-17β 3-carboranylmethyl ether. These results demonstrate that exceptionally stable carborane derivatives of estrogens can be synthesized with preservation of their biochemical and biological properties. Boron-containing estrogens may be useful for thermal neutron capture therapy of cancers with estrogen receptors to concentrate boron in the cell nucleus.  相似文献

The ability of isolated large or small bovine luteal cells to synthesize estradiol-17β was tested by incubations in the absence or in the presence of exogenous testosterone.Using a specific radioimmunoassay, no synthesis of estradiol-17β could be detected in the small or large luteal cells after incubation in the absence of testosterone. However, after incubation in the presence of exogenous unlabelled testosterone, radioimmunoassay data suggested the existence in the large but not the small luteal cells of synthesis of estradiol-17β. However, the results obtained by measuring the conversion of 3H-testosterone to 3H-estradiol-17β by cocrystallization with unlabelled estradiol-17β failed to confirm the presence of aromatase in the large cells. These data indicate that aromatization in large and small bovine luteal cells is probably negligible. Moreover, they cast serious doubt on studies of aromatization in luteal tissue based on radioimmunoassay data only.  相似文献

The metabolism of estradiol-17β by the guinea pig uterus late in pregnancy was studied in vivo and in vitro.Whole uteri were examined for estrogen metabolites one hour following an intravenous injection of [3H]-estradiol-17β or uterine sections were examined after incubation for one hour at 37°C in medium containing [3H]-estradiol-17β.In both instances uterine tissue metabolized estradiol-17g to five products: estrone, estrone-3-sulfate, 17β-estradiol-3-sulfate, estrone-3-glucuronide and 17β-estradiol-3-glucuronide. Of the total radioactive products 11 – 43% were glucuronides, 17 – 26% were sulfates and 4 – 17% was estrone. These results indicate that the guinea pig uterus actively transforms estradiol-17β into glucuronides and sulfates late in pregnancy.  相似文献

The aim of this study was to describe the presence of estrogen receptor-α (ERα) in several vaginal histological compartments in healthy adult bitches throughout three estrous cycle stages (proestrus, estrus, and early diestrus) and to relate ERα presence with serum progesterone and estradiol-17β concentrations. For this purpose, serial blood samples and vaginal biopsies were taken from five bitches every 48 hours, starting at the clinical onset of proestrus, marked by the beginning of serosanguineous vaginal secretion. Serum progesterone and estradiol-17β concentrations were determined by RIA, whereas detection of steroid receptors was carried out through immunohistochemistry. Subjective image analysis was conducted by two independent observers in the following histological compartments: superficial, intermediate, and deep epithelia and superficial (loose) and deep (dense) stroma (connective tissue). Nuclear ERα immunoreactivity was detected in every histological compartment and estrous cycle stage studied. ERα expression varied among histological compartments and during stages of the cycle. Receptor expression was associated with estradiol-17β and progesterone serum profiles. Most relevant cyclic changes were detected in the superficial and deep epithelia and in the dense connective tissue. The highest ERα expression was detected during diestrus, although each compartment had a different pattern throughout the other cycle stages. Thus, vaginal ERα expression in the bitch varied throughout proestrus, estrus, and early diestrus according to the histological compartment involved.  相似文献

The effect of the two C-17 isomers of estradiol on the shape of the action potential of rat atrial tissue was studied by means of classical glass electrodes for different concentrations of estradiol. Resting potential and upstroke were not affected by estradiol, but the duration of the action potential was reduced. Only estradiol-17β exhibits an effect in a concentration dependent way, while estradiol-17α has no effect at all. The ionic mechanism was studied by adding specific ionic blockers to the perfusate. Since the effect was much less pronounced when a slow inward current blocker was added, it was concluded that estradiol-17β acts mainly via the slow inward current channel. Only a small part of the interaction takes place via the potassium outward channel.  相似文献

The target tissues (e.g., hypothalamus, pituitary, uterus and vagina) of mature female ovariectomized rats show selective uptake of radioactivity in one hour after the injection of 6,7, 3H-estradiol-17β in a dose of 0.1 μg per 100 g body weight. Injection of 100 μg norethindrone or norgestrel per 100 g body weight 15 min before or 15 min after the administration of tritiated estradiol reduced the radioactivity in most target tissues, and also in the non-target tissues to a lesser extent. The uptake of radioactivity in the pituitary and uterus is reduced more by norethindrone than by norgestrel treatment when these Steroids were injected 15 min after estradiol-17β injection. It appears that there exists a competitive inhibition of estradiol-17β by these contraceptive Steroids in the rat. It is speculated that such competition with estradiol-17β may be an inherent property of the 17-substituted 19-nortestosterone group of Steroids.  相似文献

The metabolism of estradiol-17β is primarily an oxidative process at either carbon-2 or carbon-16 in the human. The objective of this study was to determine the relative importance of these two oxygenation pathways in the chimpanzee. The rate of oxidation of estradiol-17β at each position was determined by measuring the release of tritium into body water from carbon-2 or carbon-16. [2-3H]-Estradiol-17β or [16-3H]-estradiol-17β was injected intravenously into three adult male chimpanzees, and blood samples were obtained at several time intervals between 1 and 48 hr. The blood was lyophilized, and the release of tritium from the specifically labeled estrogens into the body fluid pool was determined. The release of tritium from the 16α-position was very low and did not exceed 3% in any animal. The release of tritium from the carbon-2 was much faster, amounting to 29%, 34%, and 35%, respectively, by 24 hr. The ratio of tritium released from carbon-2:carbon-16 was 5.0, 13.2 and 16.9, respectively, at 24 hr after injection of the specifically labeled estradiol-17β. These results demonstrate clearly that the major pathway for oxidative metabolism of estradiol-17β in the chimpanzee is via oxygenation at carbon-2, with the formation of catechol estrogens, as in the human.  相似文献

Twenty four anestrous ewes were evenly assigned to one of six groups and administered either sesame oil, estradiol-17β, estradiol-17α, estrone, estradiol benzoate or estradiol valerate. All estrogen treated ewes received 50 μg of the respective estrogen. Blood plasma was collected for 28 hours post-treatment and quantified for luteinizing hormone (LH) by radioimmunoassay. An estrogen induced LH surge was detected in at least three of the four ewes administered either estradiol-17β, estrone, estradiol benzoate or estradiol valerate whereas only one of the four estradiol-17α treated ewes and none of the ewes administered sesame oil had an LH surge. The interval from treatment to peak LH was similar for estradiol-17β (17.3±2.7 hours), estrone (18.5±1.0 hours) and estradiol benzoate (19.0±0.6 hours) treated ewes but delayed 7 to 9 hours for ewes administered estradiol valerate (26.0±1.2 hours).  相似文献

The concentrations of total estrogens in fetal calf plasma were determined during a 6–10 day period immediately before delivery. Comparison was made between levels found in untreated calves and calves infused with dexamethasone at the rate of 0.1, 1.0 and 10 mg/24 hours. In untreated calves the plasma estrone, estradiol-17β and estradiol-17α levels remained relatively constant at 38 ± 7 ng ml?1 (mean ± SEM n = 3), 46 ± 6 ng ml?1 and 29 ± 5 ng ml?1 respectively. Infusion with dexamethasone at 0.1 mg/24 hr (3 calves) and 1.0 mg/24 hr (3 calves) was without dramatic effect on plasma estrogen levels. However, in one fetus infused with 10.0 mg/24 hr the dexamethasone treatment may have caused a transitory rise in the levels of all estrogens examined.  相似文献

The patterns of serum luteinizing hormone (LH), follicle stimulating hormone (FSH), progesterone and estradiol-17β during the estrous cycle of six crossbred (Alpine × Nubian × Native) and six native goats showing a 21 day estrous cycle in a semiarid zone of Venezuela are presented. In the crossbred goats, FSH had two significant peaks on Days 19 and 0 (33 ± 8.6 ng ml−1 and 25 ± 6 ng ml−1, respectively); in contrast, native goats only had one significant peak on the day of estrus (22 ± 2 ng ml−1), with the increase beginning on Day 17. During the follicular phase of crossbred goats, estradiol-17β and LH increased to 28 ± 6 pg ml−1 and 23 ± 6.9 ng ml−1, respectively, on Day 0. Prior to Day 0, LH increased to 10.0 ± 4.9 ng ml−1 on Day 18, decreasing to 1.5 ng ml−1 on Day 19, while estradiol-17β was increasing. This relationship between estradiol-17β and LH was not found to exist in native does, which presented a LH peak on Day 0 (30 ± 8 ng ml−1 and 35 ± 10 ng ml−1 in first and second estrus, respectively). LH basal levels were notably higher in native does. The highest concentrations of progesterone (10 and 12 ng ml−1) were detected on Days 12 and 15 in crossbred and native females, respectively. In conclusion, the relationship between estradiol-17β and gonadotropins during the follicular phase in crossbred goats suggests negative and positive feedback effects on both LH and FSH. Serum concentrations of LH were higher in native than in crossbred goats, whereas concentrations of FSH were higher in crossbred does. Thus, genetic factors need to be taken into account when comparing blood levels of gonadotropins in goats raised in tropical semiarid zones.  相似文献

N-Acetylcysteine derivatives of catechol estrogens have been isolated from the urine of adult male hooded rats with ligated bile ducts, following injection of [4-14C]2-hydroxyestradiol-17β and of [4-14C]estradiol-17β-By application of double isotope methods previously described, it was shown that 2-hydroxyestradiol-17β was converted into mercapturic acids in a yield of 6–8%, confirming two previous experiments with bile duct cannulated rats, and that estradiol-17β was converted into mercapturic acids to the extent of 3–6%. Since these figures are small, and since it has been shown that in two women estrogen mercapturic acids were not formed, it appears that this class of compound will not provide an answer to the problem of unidentified water-soluble metabolites of the estrogens.  相似文献

The metabolism of tritium-labeled estrone and estradiol-17β in slices of lung tissue obtained from an adult human was studied: estrone was identified as the only metabolite of estradiol-17β and estradiol-17β as the exclusive product of estrone metabolism. Product formation remained linear as a function of time of incubation up to 3 h and of wet lung tissue mass up to 300 mg/ml. At equimolar substrate concentrations, the rates of estrone formation were at least 2-fold greater than those of estradiol-17β. The apparent KM of 17β-hydroxysteroid oxidoreductase for estrone was 11 μM and that for estradiol-17β was 10 μM. These results are suggestive that the human lung enzyme binds estrone and estradiol-17β with similar affinities; however, the oxidative pathway is favored as indicated by the greater Vmax attained in the formation of estrone. It is possible that, in vivo, the human lung constitutes a site for estradiol-17β inactivation to estrone as well as a site for the conversion of estrone to estradiol-17β. This last process may become particularly important in instances in which the ovaries have ceased to function and secrete estradiol-17β, e.g. the postmenopausal women.  相似文献

Anordrin, administered in a single s.c. dose of 62.5 μg in sesame oil, stimulated sustained uterine growth (wet weight) when measured at 24 and 72 hr, but total soluble protein and total DNA per uterus was not increased. By comparison, 3 μg of estradiol-17β under the same conditions significantly increased all three parameters of uterine growth. Both of the above steroid treatments significantly increased nuclear estrogen receptor content of the uterus, but only the estradi-ol-17β treatment resulted in significantly elevated cytosol receptor content per uterus. Anordrin binds to the 8S estrogen receptor with an affinity of about 2 × 105 M-1 as determined by competition with [3H]estradiol-17β. The abortifacient activity of Anordrin when given orally (8 mg/kg b.w.) to mice on the 7th day of pregnancy was almost completely blocked by simultaneous oral administration of estradiol-17β (0.8 mg/kg b.w.). It is concluded that the actions of Anordrin on the uterus can be attributed to its antiestrogenic activities.  相似文献

Plasma hormone levels were determined at frequent intervals in 12 two-year-old Hereford heifers after treatment with prostaglandin F2α on day 267 of gestation to induce parturition. Of the 12 animals, six also received a daily injection of estradiol-17β until calving occurred. An additional six heifers received no treatment and therefore served as a control group. The treatment with PGF2α was effective since the treated heifers calved a mean of 12 days sooner than the untreated heifers (P < .01). A metabolite of PGF2α, 13, 14-dihydro-15-keto PGF2α, rose to a peak level in the plasma within one hour after injection (im) of PGF2α then declined to a baseline level within four hours. The plasma progesterone level had fallen significantly within two hours after injection, but no change in plasma estradiol-17β or estrone was observed. In the eight heifers that responded, plasma estrogen levels were greater at the time of treatment than in the four heifers that did not respond.  相似文献

The metabolism, in vitro, of [3H]-estrone, [3H]-estradiol-17β and [3h]-estrone-3-sulfate by the livers of pregnant, young virgin female and female fetus guinea-pigs has been compared using 900 g supernatants and microsomes. The ability of the guinea-pig livers to synthesize polyhydroxylated estrogens has been found to be small. The major metabolites isolated were unconjugated estrone and estradiol-17β or their glucuronides. The percentage of sulfates was lower after incubations with [3H]-estrone than with [3H]-estradiol-17β. A kinetic study with microsomes has shown a direct conversion of estrone-sulfate to estradiol sulfate. Fetal microsomes have been found to exhibit a more active hydrogenation of estrone to estradiol-17β than microsomes from young female or pregnant animals.  相似文献
