The knobbed acrosome defect (KAD) has only once been reported in dog sperm. This report describes the breeding history, clinical presentation and semen evaluation of four closely related miniature Schnauzer dogs that had between 8 and 44% sperm with KAD. Two of the dogs had no substantial reproductive or sperm defects other than the KAD and were of normal fertility.  相似文献

Differences in background and outcome of three behavior problems of dogs   总被引:1,自引:0,他引:1  
In order to characterize the three major behavior problems, aggression toward owners, aggression toward strangers and separation anxiety, backgrounds of dogs and general outcomes of the behavioral treatments were analyzed retrospectively. There were 169 cases of aggression toward owners, 84 cases of aggression toward strangers and 78 cases of separation anxiety which did not overlap each other during the 5 years from 1993 to 1997 at Cornell University Animal Behavior Clinic. Based on the case records, including discharge instructions, follow-up information, and pre-presentation questionnaires, several variables were compared among these three groups. The sexual status of these groups was not statistically different, although dogs with aggression toward owners had the highest proportion of males and there were more males in all behavior groups than in the hospital population. Also, breed types were different among three groups with a significantly higher proportion of mixed breed dogs among dogs with separation anxiety and aggression to strangers as compared to dogs with aggression to owners and to the hospital population. A higher percentage of dogs in the separation anxiety group tended to live in apartments and to be disciplined only verbally by the owner than in the other two groups. Age differences were apparent among the three groups in relation to when the dogs were obtained, and the separation anxiety group was different from at least one of the other groups in the age when first obtained, the age the owners first noticed the problem, and the age of behavioral examination. Regarding the general outcome of the behavioral treatment, there were no significant differences among the behavioral groups with regards to the proportion of dogs reported improved. These results provide new characterizations of these three major behavior problems.  相似文献

Male sterilization by chemical agents is a nonsurgical contraceptive approach designed to induce azoospermia and, therefore, infertility. Intratesticular injection of zinc gluconate for sterilization of dogs has been described, but its use in cats remains limited. The objective of the present study was to evaluate, by light and transmission electron microscopy, the efficacy of a single intratesticular injection of a zinc gluconate solution (Testoblock) as a sterilant for male cats. Twelve sexually mature mixed breed cats were allocated at random into two groups (control = 6; treated = 6) and given a single injection into each testis of either isotonic saline or zinc gluconate, respectively. Histopathologic and ultrastructural evaluation was assessed at 120 days postinjection. Histopathologic changes were not detected in the testes from the control group. However, histologic evaluation of the treated group revealed atrophic and dilated seminiferous tubules, a decrease in the number of germ cells, and incomplete spermatogenesis. Sertoli cells had various degrees of cytoplasmic vacuolization. Intertubular tissue revealed active fibroblasts, collagen deposition, and inflammatory cells. The diameter of seminiferous tubules, epithelial height and tubular area were reduced (P < 0.05) in the treated group compared with controls. Azoospermia occurred in 8 of the 11 treated cats (73%). Ultrastructural evaluation of Leydig cells revealed loss of nuclear chromatin, increased smooth endoplasmatic reticulum, and mitochondria degeneration. Intratesticular injection of zinc gluconate solution impaired spermatogenesis in cats and has great potential as a permanent sterilant in this species.  相似文献

Behavioral evaluations of 2017 shelter dogs were used for identifying dogs with aggressive tendencies and for predicting post-adoption behavior problems. Associations between failure of the behavioral evaluation and demographic factors (age, breed, and sex) and the dog's behavioral history, evaluated by logistic regression, were highly significant (P < 0.0005), except for “old” dogs (age > 72 months). Dogs that failed the behavior evaluation were not placed for adoption; therefore it was not possible to study prospectively the capability of behavioral evaluation to predict future aggressiveness in these dogs. Instead we developed tests for classifying dogs as aggressive or not aggressive based on their demographic factors and behavior evaluation outcomes. The results were compared retrospectively to the dogs’ known behavioral histories, which were obtained at intake to the shelter. This allowed estimation of the sensitivity, specificity, and accuracy of the classification tests. The most significant postdictor (i.e., “retrospective predictor”) of aggressiveness was failure of the behavioral evaluation (odds ratio 11.83, P < 0.0005). Among dogs that passed the behavioral evaluation, classification as “unsocial” by the behavioral evaluation was associated with nonaggressive behavior problems reported at 6-month follow-up (P = 0.005), and classification as “borderline” was associated with return to the shelter for aggressive behavior (P = 0.028). Behavioral evaluations reduced the rate of returns of adopted dogs from 19% in the year prior to their institution to 14% (P = 0.001) during the 2-year study period. Returns for aggression were reduced from 5% to 3.5% (P = 0.05), with many fewer incidents of serious aggression. The majority of dogs (86%) that failed the behavioral evaluation failed multiple component tests, indicating general aggressive tendencies in these dogs. Together with demographic information and behavioral history, behavioral evaluations can help to improve decisions regarding the disposition of shelter dogs.  相似文献

Current concerns over the potential impacts of climate change and the increased movement between countries of people and companion animals on the distribution of ectoparasites, highlight the need for accurate understanding of existing prevalence patterns. Without these future changes will not be detected. Here, the distribution and prevalence of tick infestations of domestic dogs in Great Britain were examined. A total of 173 veterinary practices were recruited to monitor tick attachment to dogs in their local areas between March and October 2009. Practices selected five dogs at random each week from those brought to the surgery and undertook a thorough, standardized examination for ticks. Each veterinary practice participated for 3 months before being replaced. Any ticks identified were collected and a sample sent to the investigators for identification, along with a clinical history of the dog. A total of 3534 dogs were examined; 810 dogs were found to be carrying at least one tick. Ixodes ricinus (Linnaeus) (Acari: Ixodidae) was identified in 72.1% of cases, Ixodes hexagonus Leach in 21.7% and Ixodes canisuga Johnston in 5.6% of cases. Five samples of Dermacentor reticulatus (Fabricius) (Acari: Ixodidae) were also found, adding to the growing evidence that an established population of D. reticulatus now exists in south-eastern England. Almost all the ticks found were adults. Overall, 19.2% of the veterinary practices reported no tick detections, 50% reported that ≥14.9% of the dogs seen were infested and 14.6% reported that >50% of the dogs inspected carried ticks. The estimated incidence of tick attachment was 0.013 per day in March (lowest) and 0.096 per day in June (highest). A number of risk factors affected the likelihood of tick attachment on dogs. Gundog, terrier and pastoral breed groups were more likely to carry ticks, as were non-neutered dogs. Dogs with shorter hair were less likely to have ticks, and dogs were most likely to carry a tick in June. This study is of value because, unusually, it presents the results of a randomized sample of dogs and gives a prevalence which is higher than those previously recorded in Great Britain.  相似文献

The assisted reproductive techniques (ARTs) such as in vitro fertilization, embryo transfer, and cryopreservation of gametes have contributed considerably to the development of biomedical sciences in addition to improving infertility treatments in humans as well as the breeding of domestic animals. However, ARTs used in canine species have strictly limited utility when compared with other mammalian species, including humans. Although successful somatic cell cloning has been reported, artificial insemination by frozen semen to date is only available for the improved breeding and reproduction for companion and working dogs as well as guide dogs for the blind. We describe here the successful cryopreservation of embryos and subsequent embryo transfer in dogs. Canine embryos were collected from excised reproductive organs after artificial insemination and subsequently cryopreserved by a vitrification method. When the 4-cell to morula stage of cryopreserved embryos were nonsurgically transferred into the uteri of nine recipient bitches using a cystoscope, five recipients became pregnant and four of them delivered a total of seven pups. The cryopreservation of embryos in canine species will facilitate the transportation and storage of genetic materials and will aid in the elimination of vertically transmitted diseases in dogs. In addition, this technique will contribute to the improved breeding of companion and working dogs such as guide dogs, drug-detecting dogs, and quarantine dogs.  相似文献

Recent experimental animal studies suggested that the circadian locomotor output cycles kaput protein gene (CLOCK) has been reported to play a critical role in sperm function and male fertility. The aim of this study was to determine whether variants of the CLOCK gene are involved in idiopathic male infertility. The study included 478 idiopathic infertile men and 194 fertile controls who completed physical examinations. Each subject donated 5?ml of peripheral blood and a sample of semen in the ejaculate. An aliquot of each blood sample was used to separate the serum for the measurement of testosterone as well as follicular stimulating hormone (FSH) using the standard radioimmunoassay. The rest of the blood samples was used to extract the DNA for the assay of three tagging single-nucleotide polymorphisms of CLOCK gene, viz., rs1801260, rs3817444 and rs3749474, using the real-time fluorescence quantitative PCR. The ejaculate of each subject was used for semen analysis by computer-assisted semen analysis system. The results indicated: (a) the variant rs1801260 associated with normal semen parameters was linked to a significant increase in the risk of idiopathic infertility, (b) the variant rs3817444 associated with both normal and abnormal semen parameters also indicated an increased risk of idiopathic infertility, and (c) the variants rs3749474 associated with both normal and abnormal semen parameters, on the other hand, conferred no significant risk for male infertility. Furthermore, elevated serum testosterone and FSH levels were correlated with the three variants of CLOCK gene in idiopathic infertility. The findings demonstrate that the human subjects with variants of the CLOCK gene are associated with idiopathic male infertility and therefore may be applied as a risk factor of male infertility.  相似文献

A hysterosalpingography method was developed to examine the uterine cavity and oviducts of potential additions to our squirrel monkey breeding colony and those animals within the colony with a history of infertility. Females to be examined were anesthetized and placed in dorsal recumbency. A needle was inserted transabdominally into the body of the uterus, the cervix was sealed with a clamp applied vaginally, and water soluble contrast media was infused into the uterine cavity with sufficient pressure to fill the oviducts. Radiographs were obtained during infusion resulting in visualization of the uterine cavity and the lumina of the oviducts. The ovaries were visualized as they were surrounded by contrast media draining from the fimbriae of the oviducts. Hysterosalpingography proved to be a useful method for clinical evaluation of reproductive potential of female squirrel monkeys. It also has provided a means to diagnose causes of infertility associated with dysfunction of the oviducts.  相似文献

Cystic fibrosis (CF) is an autosomal recessive disease caused by mutations in the CFTR gene. Among males with CF, 95% are infertile due to congenital absence of the vas deferens. We investigated the role of family history of infertility among CF subjects and characterized mutations in them. Among 50 CF subjects, four had a family history of infertility. A homozygous c.1521_1523delCTT mutation was detected in one, two had a compound heterozygous genotype (c.1521_1523delCTT/c.3717 + 10 kbC>T), and c.1521_1523delCTT mutation was identified on one allele of fourth CF subject. Genetic analysis of each infertile family members of CF subjects revealed the c.1521_1523delCTT mutation on one allele; however, no mutation could be identified on other allele. Haplotype analysis of the infertile family members showed that at least one of the alleles shared the same haplotype as that of the index case. It is suggested that the CFTR gene is implicated in the infertile members of the CF families. Failure to detect mutations on the other allele by SSCP analysis demands direct gene sequencing to detect mutations in the intronic or promoter region.  相似文献

Canine infertility has many causes that must be considered during evaluation of abnormal reproductive function. An important infectious agent is Brucella canis. Classically deemed a major reason of abortion, this organism also produces infertility in stud dogs and poses a potential health hazard to dogs and humans. The State of Georgia has, out of necessity, instigated regulations to manage outbreaks and seek compliance by educating the pet owner population about this disease. A review of its etiology, methods of transmission, pathophysiology, clinical signs, diagnosis, serology and culture, pathology, treatment options, and regulated prevention featured by Georgia, are presented.  相似文献

Bovine infertility is a major cause of loss in the livestock industry. In the present study bovine oviduct cell cultures were infected with a Chlamydophila abortus strain. A direct evaluation of infection was performed by means of May Grünwald-Giemsa and immunocytochemistry for chlamydial LPS, which revealed inclusion bodies and vacuolisation. SEM and TEM analysis of infected cells showed various degrees of cell damage and conglutination of microvilli. This finding suggests that cattle infertility may result from an alteration of oviduct environment caused by multiplication of C. abortus. This microorganism, among other infectious agents, could be considered a potential causative agent of bovine infertility.  相似文献

A related group of parvoviruses infects members of many different carnivore families. Some of those viruses differ in host range or antigenic properties, but the true relationships are poorly understood. We examined 24 VP1/VP2 and 8 NS1 gene sequences from various parvovirus isolates to determine the phylogenetic relationships between viruses isolated from cats, dogs, Asiatic raccoon dogs, mink, raccoons, and foxes. There were about 1.3% pairwise sequence differences between the VP1/VP2 genes of viruses collected up to four decades apart. Viruses from cats, mink, foxes, and raccoons were not distinguished from each other phylogenetically, but the canine or Asiatic raccoon dog isolates formed a distinct clade. Characteristic antigenic, tissue culture host range, and other properties of the canine isolates have previously been shown to be determined by differences in the VP1/VP2 gene, and we show here that there are at least 10 nucleotide sequence differences which distinguish all canine isolates from any other virus. The VP1/VP2 gene sequences grouped roughly according to the time of virus isolation, and there were similar rates of sequence divergence among the canine isolates and those from the other species. A smaller number of differences were present in the NS1 gene sequences, but a similar phylogeny was revealed. Inoculation of mutants of a feline virus isolate into dogs showed that three or four CPV-specific differences in the VP1/VP2 gene controlled the in vivo canine host range.  相似文献

Abstract

In April 1986, a five-year-old boy was fatally mauled by a pit bull terrier in Multnomah County, (Portland) Oregon. This tragic event sparked concern over how effectively the public was being protected from the hazards posed by aggressive dogs. In June 1986, a new ordinance was passed that identifies and regulates potentially dangerous dogs. In the first three years of the program, 1,652 dogs were classified as potentially dangerous, and restrictions were placed on the ownership of these animals. A pretest-posttest evaluation methodology examining the rate of recidivism was used to measure the program's effectiveness in limiting the opportunity for identified potentially dangerous dogs to repeat their behavior. In the five years prior to the implementation of the program, 25% of those dogs that had caused injury to people or other animals through attacking or biting repeated that same behavior within one year. After the implementation of the program, that rate of repeat incidence has been reduced to 7%.  相似文献

Only one mutation in the prothrombin gene (Factor II), 20210G>A, has been definitively associated with an increased risk for venous thrombosis. Using hybridization probe analysis for mutation detection on the LightCycler (Roche Molecular Biochemicals), we identified seven patient samples with atypical melt curve patterns. Sequence analyses of each of these samples revealed heterozygosity for a C to T transition at position 20209. As in other reported cases, each of the apparently unrelated patients was of African-American descent, suggesting that the variant is population specific. Two patients were referred for testing due to a history of stroke, one with a right major coronary artery embolic stroke and the other with a right cerebellar stroke. A third patient had chronic renal failure secondary to hypertension with a reported family history of renal failure. Three patients had a history of multiple pregnancy losses. The last patient had a kidney transplant for end stage renal disease secondary to glomerulonephritis. She was being evaluated for a second transplant, but to our knowledge, had a negative history of venous thrombosis. The prevalence and the clinical significance of the prothrombin 20209C>T mutation is unknown. Recently reported functional studies revealed conflicting results. The clinical utility of testing for and reporting this variant remains unresolved.  相似文献

Combining the traditional morphologic evaluation of spermatozoa with computer assisted image analysis adds randomness, objectivity, repeatability and accuracy to morphometric measurements. We collected semen from 10 fertile, normospermic dogs aged 1 to 7 yr and from 3 teratozoospermic breed-matched dogs. Sperm head morphology was examined in Giemsa-stained smears by light microscopy, using a computer-assisted image analyzer and by transmission electron microscopy. We found significant variation in sperm head area, length, width and degree of roundness among normospermic individual dogs, indicating that it would be necessary to examine many more dogs before the size and shape of normal dog spermatozoa could be determined. The normospermic dogs were used as controls for the teratozoospermic cases. Case 1: A 2-yr-old subfertile Cavalier King Charles Spaniel had semen with small and narrow-based sperm heads and a proximal cytoplasmic droplet in most of the spermatozoa. With the image analysis system, sperm heads were shown to be smaller and more oval than in normospermic dogs. The variatons in size and shape were similar in magnitude to those of control dogs. An examined infertile half-brother had similar semen quality. Case 2: A 3-yr-old Petit Basset Griffon Vendeen with 2 unsuccesfull matings exhibited spermatozoa with severe abnormalities. Measured by image analyzer, sperm heads were irregular in shape and very small in area. One of the two littermates examined had semen of the same quality as the case dog. Case 3: A 3-yr-old fertile Golden Retriever had semen with giant sperm heads in about 50% of spermatozoa. Image analyzing results revealed 2 populations of different sized sperm heads. Giant heads consisted of 52.2% of all spermatozoa. The results of the study reported here suggest that the image analysis technique may be useful in evaluating structural changes in sperm morphology, supplementing visual assessment that is used in conventional methods.  相似文献

Obiective

To describe a case of seminoma occurring in a patient with a past history of cryptorchidism, infertility and clomiphene citrate (CC) treatment. To review the literature of seminoma in association with these risk factors.

Case report

A 33 year old man developed typical seminoma in the right testis. A bilateral cryptorchidism was lately diagnosed and treated by orchidopexy at the age of 17. Primary infertility due to oligoasthenospermia was diagnosed at the age of 29. Treatment was initiated with CC at the dose of 25 mG/day. Semen quality improved gradually. After failure of various assisted reproductive techniques, a spontaneous pregnancy was achieved and a healthy baby delivered. Shortly after delivery the seminoma was diagnosed, after a totla of three years of treatment. Six cases of testicular tumors have been reported in the literature in which patients had been pretreated with fertility drugs for various priods of time before tumors was diagnosed. Cryptorchidism is a well-known risk factor for testicular malignancy, as is the infertil state. Clomiphene citrate may promote malignant transformation by activation of estrogen receptors, by raising gonadotropin and/or testosterone concentrations, or by other mechanisms.

Conclusion

Clomiphene citrate may have a previously unappreciated malignancy-promoting effect in infertile men, especially those with other risk factors. Clinicians treating similar patients should take this into account and the literature monitored for other associated cases.  相似文献

Morphological aspects of the myocardium in dogs with experimentally induced jaundice were assessed ultrastructurally. Obstructive jaundice was produced by chronic bile-duct ligation and by choledochocaval anastomosis. The left ventricular myocardium and the papillary muscle were used. Statistical analysis of sarcomere length and of mitochondrial density showed no significant differences between jaundiced and sham-operated dogs. Qualitative evaluation of the mitochondria, the intercalated disc and other sarcoplasmic constituents revealed no damage to the jaundiced dogs.  相似文献

Aggression towards owners in a dominance context is a common behavioral problem in dogs. A review of 24 cases (21 males and 3 females) presented to the Veterinary Hospital of the University of Pennsylvania for dominance aggression revealed that it is a predominantly male trait, and pure-bred dogs (n=21) are presented more frequently than are mixed breeds (n=3). Each dog diagnosed as dominant aggressive manifested aggression in three or more of 17 circumstances involving physical manipulation, discipline, or guarding of food, objects or resting places. A combination of techniques including castration, use of synthetic progestins, and behavioral techniques were used to treat the dogs. Three to 15 months after the behavioral consultation, 19 owners were contacted by telephone and interviewed by a person identified as a neutral party unassociated with the service. The owners were asked a standardized set of questions. Twenty-one percent of the owners reported that their dogs were 90% or greater improved, 53% reported a 70% or greater improvement, and 79% reported a 50% or greater improvement. Although the frequency and intensity of aggressive behavior was reduced and the majority of persons were highly satisfied, the dominant aggressive tendencies of the majority of the dogs were not completely suppressed.  相似文献

By the use of isoelectric focusing in polyacrylamide gels serum samples from 146 Alaskan wolves were studied with regard to transferrin (Tf) and esterase (ArE) polymorphism, comparing the phenotypic band patterns with those of selected Norwegian dogs. The study revealed Tf and ArE polymorphisms in the wolf with phenotypic band patterns being indistinguishable from the corresponding ones in dogs. This suggests the occurrence of the same two common Tf alleles in the wolf as in the dog. In the ArE system the results are consistent with the occurrence of three alleles which also occur in dogs whereas a fourth allele, so far not seen in dogs, is seen in Alaskan wolves.  相似文献

Domestic dogs (Canis familiaris) were administered a progressive elimination task in which they had to visit and deplete three baited sites. Dogs were brought back to the starting point after each visit to any site whether they had made a correct or an incorrect choice. In Experiment 1 (n=10) the results revealed that the dogs randomly selected among the sites when they were equidistant from the starting point whereas they relied on the least distance rule when one of sites was closer to the starting point than were the other sites. In Experiment 2 (n=12), the dogs first chose the left target when angular deviation between adjacent targets varied whether the least angular deviation was on the right of the left. Results are interpreted in terms of Gibson's hypothesis about cooperative hunters. The discussion also emphasizes comparisons with cats (i.e., solitary hunters).  相似文献
