We investigated the oligosaccharide sequence of glycoconjugates, mainly sialoglycoconjugates, in the horse oviductal ampulla during oestrus by means of lectin and pre-lectin methods such as the KOH-neuraminidase procedure to remove sialic acid residues and incubation with N-glycosidase F to cleave N-linked glycans. Ciliated cells displayed N-linked oligosaccharides throughout the cytoplasm. The cilia glycocalyx expressed both N- and O-linked (mucin-type) oligosaccharides, both showing a high variety of terminal sequences. In the most non-ciliated cells, the whole cytoplasm contained N-linked oligosaccharides with terminal alphaGal as well as mucin-type glycans with terminal Forssman pentasaccharides. In a few scattered non-ciliated cells, the whole cytoplasm displayed sialylated N-linked oligosaccharides with terminal Neu5Ac-GalNAc and O-linked glycans terminating with neutral and/or alphaGalNAc, Neu5Ac alpha2,6Gal/GalNAc, Neu5AcGal beta1,3GalNAc. Supra-nuclear granules, probably Golgi zones, of non-ciliated cells showed mainly O-linked glycans rich in sialic acid residues. The luminal surface of non-ciliated cells showed N-linked oligosaccharides, containing terminal/internal alphaMan/alphaGlc, betaGlcNAc and terminal alphaGal, as well as mucin-type oligosaccharides terminating with a large variety of either neutral saccharides or sialylated sequences. Apical protrusions containing O-linked oligosaccharides with terminal Forssman pentasaccharide, Neu5Ac-Gal beta1,4GlcNAc, Neu5Ac-GalNAc were seen in non-ciliated cells scattered along the epithelium. These findings show the presence of sialoglycoconjugates in the oviductal ampulla epithelium of the mare and the existence of different lectin binding profiles between ciliated and non-ciliated (secretory) cells, as well as the presence of non-ciliated cell sub-types which might determine functional differences along the ampullary epithelium of mare oviduct.  相似文献

The ultrastructure of oviduct epithelium of clinically healthy cows and 15 sows was investigated using scanning and transmission electron microscopy. In all parts of the oviduct, ciliated and non-ciliated epithelial cells are present, but their number varies in both the investigated animals in different regions of the oviduct, depending on the phase of the estrous cycle. In addition to ciliated cells with numerous cilia on their luminal surface, so-called pale ciliary cells were found in all parts of the oviduct of cows and sows. The cytoplasm of these cells is electron-lucent, their luminal surface carries few cilia and short microvilli. The apical cytoplasm contains species specific secretory granules, which means that these cells have features characteristic of both secretory and ciliated cells. It is suggested that the pale ciliated and non-ciliated secretory cells are functional stages of the same tubar epithelium cell, and that the transformation between these two cell types is regulated by functional requirements of the organ in different phases of the estrous cycle.  相似文献

The organization of the stomach in the compound styelid ascidian, Polyandrocarpa misakiensis, is described, and the morphology and cell types of the stomach is discussed from the phylogenetic viewpoint. The stomach is a sac-like organ whose wall is formed into longitudinal folds. The stomach consists of external and internal epithelium. The internal epithelium is simple columnar, except for the bottom of the folds. There are five cell types: absorptive cells, zymogenic cells, endocrine cells, ciliated mucous cells, and undifferentiated cells. The absorptive cells have numerous microvilli. The apical region of these cells is occupied by coated vesicles. The zymogenic cells have a conical outline and a few microvilli on their apical surfaces. There are secretory granules in the apical region of zymogenic cells. The endocrine cells have low cell height and electron-dense granules around the nucleus. Endocrine cells have one or two cilia and a few microvilli on the apical surfaces. The basolateral part of these cells often bulges into the adjoining cells. Immunoelectron microscopy revealed that some endocrine cells have serotonin-like immunoreactivity. The ciliated mucous cells are restricted to a single ventral groove. They have numerous microvilli and a few cilia on their apical surfaces. Moderately electron-dense granules are accumulated in the apical part of the ciliated mucous cells. Undifferentiated cells, filled with free ribosomes, form a pseudostratified epithelium in the base of each fold. The nucleus of undifferentiated cells has a prominent nucleolus. The pseudostratified epithelium of the pyloric caecum consists of electron-dense and electron-light cells.  相似文献

The ultrastructural changes occurring in the fully functional oviduct of Isa Brown laying hens were studied during various stages of the laying cycle. Hens were killed at different positions of the egg in the oviduct. The oviduct was lined by ciliated and non-ciliated cells (also referred to as granular cells). The granular cells in the infundibulum contributed to secretion during egg formation, whereas ciliated cells showed little evidence of secretion. Ultrastructural changes were recorded in the granular and glandular cells of the distal infundibulum. In the magnum, the surface ultrastructure revealed glandular openings associated with the ciliated and granular cells. Cyclic changes were recorded in the glandular cells of the magnum. With respect to the three observed types of glands, the structure of gland type A and C cells varied at different egg positions in the oviduct, whereas type B cells represented a different type of gland cell containing amorphous secretory granules. The surface epithelium of the isthmus was also lined by mitochondrial cells. Two types of glandular cell (types 1 and 2) were recorded in the isthmus during the laying cycle. Intracisternal granules were found in type 2 cells of the isthmus. A predominance of glycogen particles occurred in the tubular shell gland. The granular cells in the shell gland contain many vacuoles. During egg formation, these vacuoles regressed following the formation of extensive rough endoplasmic reticulum; the reverse also occurred. The disintegrated material found in the vacuoles may have been derived from the disintegrating granules. The Physiology Teaching Unit, University of New England, provided financial support to K. Isthmic and ampullary oviductal epithelia sampled from Merino ewes at days -1, 1, 3, and 10 of the estrous cycle (estrus = day 0) were studied by scanning and transmission electron microscopy after fixation by vascular perfusion. Secretory cells, ciliated cells, and lymphocytelike basal cells were observed in both isthmic and ampullary epithelium at all stages of the estrous cycle studied and their ultrastructural features were analyzed. Synthesis of lamellated secretory granules occurred in the ampullary secretory cells during the follicular and early luteal phases, and their contents were released by exocytosis into the oviductal lumen during the luteal phase. Granule release was associated with nucleated apical protrusion of these cells into the oviductal lumen. No such secretory activity was displayed by isthmic secretory cells even though a few cells contained nonlamellated granules. Apocrine release of apical vesicles and accompanying cytoplasmic material from apical protrusions of ciliated cells occurred in the isthmus around estrus but not in the ampulla. This unexpected feature has not previously been reported in any other mammal. Dendritic basal cells were distinguished in the lower part of the epithelium by their heterochromatic nuclei, electron-lucent cytoplasm, and lack of attachment zones. No migration of basal cells was observed, and their ultrastructural features were similar in the ampulla and isthmus and at all stages of the estrous cycle examined. The function of these lymphocytelike cells in the epithelium is uncertain, but the presence of phagocytic bodies and lysosomes in 20% of them may indicate a phagocytic role.  相似文献

Ciliated vacuoles and intraepithelial cysts have been observed in oviductal and endocervical epithelia of rabbits. In this study, rabbits under various hormonal conditions were studied by light and transmission electron microscopy and tissue culture in an attempt to determine their distribution and origin. Ciliated vacuoles most frequently lay in the basal cytoplasm, below or beside the nucleus, and very close to the basal lamina. A few were apically located. Their average diameter was 8.8 by 5.1 microns. Cilia and microvilli projected into the vacuolar lumen. These vacuoles were located intracellularly as evidenced first by the degeneration of both their cilia and microvilli and the moderately dense matrix that often filled the vacuolar lumen, as observed by electron microscopy. Secondly, phase microscopy of the living endocervical epithelium allowed us to observe the beating of the cilia within the vacuoles, not on the surface of such cells. Thirdly, ruthenium red stained the surface glycocalyx of ciliated and secretory cells, but not that of the cilia and microvilli within the vacuoles. The intraepithelial cysts were not observed in all tissue blocks. The largest numbers were found in ovariectomized animals treated for 3 and 5 days with estradiol. More were seen in the isthmus and cervix than in the fimbria and ampulla. The cysts were located most often within the epithelium along the sides of, and at the bases of, the mucosal folds. They were lined by flattened epithelium of various combinations of secretory and ciliated cells. An unusual cell type was associated with some of the cysts and ciliated vacuoles. Its cytoplasm contained aggregates of mitochondria and vesicles whose contents varied in density. Although the genesis of the ciliated vacuoles is not certain, our results indicate that they may arise from aberrant positioning of proliferating procentrioles or from a defect in targeting or transporting the centrioles to the apical plasma membrane to serve as basal bodies. Fusion of adjacent ciliated vacuoles with lumina lined by secretory cells having deep apical invaginations appeared to contribute to the formation of cysts.  相似文献

Summary Changes occurring in the epithelium covering bronchus-associated lymphoid tissue (BALT) in the rat after several intratracheal administrations of horseradish peroxidase (HRP) were studied using morphological and ultrastructural methods. The epithelium is invaded by W3/ 25-positive (T-helper) lymphocytes, the BALT epithelial cells become Ia-positive and develop microvilli; there is an apparent loss of cilia. The number of non-ciliated cells in stimulated BALT increases. The non-ciliated cells can be subdivided into two cell types, one with electron-dense cytoplasm and cytoplasmic granules and the other without granules. The electron-density of the latter cell type is intermediate between that of the ciliated cells and that of the granulecontaining non-ciliated cells. The granule-containing cell types may be responsible for the uptake of antigens, while the other non-ciliated cell may be involved in the production of the secretory component and the passage of secretory IgA.Supported by a research grant from the Nederlands Astma Fonds  相似文献

The present study deals with an immunohistochemical localization of S-100 protein in the bovine oviduct. The epithelium of the infundibulum, ampulla and isthmus showed a positive staining for S-100 protein. The immunoreactivity for S-100 was observed both in the ciliated and nonciliated (secretory) cells of the oviductal epithelium at any stages of the estrous cycle. The immunoreactivity was also found in nervous elements and endothelial cells of blood vessels. No cell outside these cells showed any immunoreactivity for S-100. Although the functional significance of S-100 protein in the oviductal epithelium remains to be elucidated, the present results introduce new perspectives into the investigation of function and localization of S-100 protein.  相似文献

The mouse oviduct is covered by dense tracts of ciliated cells interspersed at random with occasional non-ciliated cells. Correlation between scanning electron microscopy and thin section images indicates that in the isolated fimbria most cilia are short (5 µm) and inactive, resting at the end of a uterad-directed effective stroke. These cilia terminate in a 9S−2 tip, the microtubules ending in an electron-dense plaque underneath the cell membrane. At the tip of the cilium a crown of fine extracellular hairs is attached to the ciliary membrane. In the ampulla and isthmus the ciliated cells decrease progressively in number and appear to lie in crypts.  相似文献

The oviduct is a very thin organ with a very tortuous appearance. It is divided into three segments: the infundibulum, the ampulla and the isthmus. Particularly, the oviduct of the viscacha lacks the intramural portion described in other species. The mucosa shows longitudinal pleats. The free edge of the infundibulum ends as small fimbriae that are of variable length and do not completely cover the ovary. The proportion of ciliated and secretory epithelial cells varied both in relation to the segments of the oviduct analysed and to the physiological state (anoestrus, follicular phase, early pregnancy and late pregnancy). The glycocalix and the apical region of the superficial epithelial cells are PAS and alcian‐blue positive. The muscular layers vary in thickness in different regions. Some lectins such as UEA‐1 and DBA showed variations in the binding pattern during the different physiological stages analysed whereas RCA‐1and WGA had a very stable pattern.  相似文献

Studies on histomorphometrical changes in different segments (infundibulum, magnum, isthmus, shell gland and vagina) of oviduct of mallard, Anas platyrhynchos during active and quiescent phases of the reproductive cycle have been made. The absolute and per cent length and width of each segment showed a marked change. The magnum showed an increase of 280 per cent. Of all the histological parameters studied the number and height of mucosal folds and mucosal epithelium showed more marked increase in all segments of oviduct. The size of tubular glands and frequency of ciliated and secretory cells were studied in relation to oviductal activity.  相似文献

The porcine oviduct synthesizes de novo and secretes a number of proteins into culture medium, many of which are unidentified. The objectives of the present study were to 1) semipurify and identify a M(r) 45 000 secreted protein of the oviduct, 2) examine its synthesis within the three functional segments (infundibulum, ampulla, and isthmus), and 3) evaluate its distribution throughout the oviduct. Oviductal tissue was collected during early pregnancy, divided into functional segments, and subsequently cultured. Medium was collected, and the M(r) 45 000 protein was concentrated by gel-filtration chromatography. The semipurified protein was transferred onto a polyvinylidene fluoride membrane and subjected to N-terminal amino acid analysis. The 26-amino acid sequence was 96% identical to that of pig plasminogen activator inhibitor (PAI)-1. Analysis by 1-dimensional SDS-PAGE and fluorography of rabbit anti-human PAI-1-immunoprecipitated product confirmed PAI-1. Subsequent 2-dimensional SDS-PAGE and fluorographic analyses of media revealed greater PAI-1 synthesis by the isthmus than by the ampulla or infundibulum. PAI-1 was immunolocalized throughout the oviduct and was heavily concentrated in the apical region of epithelial cells. Immunogold electron microscopy localized PAI-1 within putative secretory granules in the epithelial apical region and also associated with cilia in the isthmus. Isthmic PAI expression suggests a crucial role in protecting the preimplantation embryo from proteolytic degradation as well as in regulation of extracellular matrix turnover and remodeling.  相似文献

The ultrastructural changes that occur in the ciliated cells of the tubal epithelium of the adult rabbit during ovum transport were previously reported. In this communication, the changes that occur in the secretory cells of the tube under identical conditions are described. Estrous and pregnant rabbits were used. 14-70 hours postcoitum the size and shape of the microvilli of the ampullar cells differed from the isthmic ones. The prominence of the microvilli was roughly correlated with the possible arrival of the ovum in any part of the tube. The secretory granules in the cells of the ampulla were more electron dense than those in the isthmus. With the progress of the post ovulatory period, the secretory granules changed in nature and location. During the postovulatory period, the secretory process in both parts of the tube indicate a "merocrine" type of secretion. A cilium was occasionally present in the luminal epithelium of a secretory cell. Hybrid cells may arise when a cell malfunctions while receiving a set of messages from the nucleus at the time appropriate for cell differentiation.  相似文献

Summary An electron microscopical study of the epithelium of the uterine tube was carried out in the newborn. Among the epithelial cells at least two morphologically well defined types can be distinguished: ciliated and non-ciliated cells.The ultrastructure of the cilia and related structures corresponds to what has been described by other authors in ciliated cells of various organs and of different species. Near the basal bodies of the cilia there is a concentration of vesicular mitochondria, which is thought to be evidence of a high metabolic activity in this region of the cell. Large opaque granules in the supranuclear zone of the ciliated cells are, it is suggested, paraplasmatic inclusions, perhaps supporting material for the ciliokinetic processes. There was no evidence of a secretory function of the ciliated cells.Among the non-ciliated cells, which in general show a straight lined luminal border with few microvilli, there are some cells containing dense granules, which are distributed throughout the cytoplasm and concentrated in the luminar side of the cell. The apical parts of these cells are protruding and sometimes digitated or branched; they contain accumulated granular materials and are separated from the rest of the cell after the formation of an intracellular plasmalemma. A similar detachment was found in an other cell type, but here the protruded apical parts of the cells are edematous and do not contain any visible secretory materials. It is uncertain if the detached cytoplasmic substances form a part of a specific secretory product; there are no secretory granules within the cytoplasm. On the contrary, the detachment of cytoplasmic parts may only accompany the excessive proliferation of cells which takes place during this period of growth.  相似文献

The secretory processes in the shell gland of laying chickens were the subject of this study. Three cell types contribute secretory material to the forming egg: ciliated and non-ciliated columnar cells of the uterine surface epithelium, and cells of tubular glands in the mucosa. The ciliated cells as well as the non-ciliated cells have microvilli, which undergo changes in form and extent during the secretory cycle. At the final stages of shell formation they resemble stereocilia. It is postulated that the microvilli of both cells are active in the production of the cuticle of the shell. The ciliated cell which has both cilia and microvilli manufactures secretory granules which arise from the Golgi complex in varying amounts throughout the egg laying cycle. Granule production reaches its greatest intensity during the early stages of shell deposition. The ciliated cell probably supplies proteinaceous material to the matrix of the forming egg shell. The non-ciliated cell has only microvilli. Secretory granules, containing an acid mucopolysaccharide, arise from the Golgi complex. Some granules are extruded into the uterine lumen where they supply the egg shell with organic matrix. Others migrate towards the supranuclear zone. Here a number of them disintegrate. This is accompanied by the formation of a large membraneless space, which is termed “vacuoloid.” Subsequently the vacuoloid regresses and during regression an extensive rough endoplasmic reticulum with numerous polyribosomes of spiral configuration appears. It is suggested that material in the vacuoloid originating from the disintegrating granules is resynthesized and utilized for the formation of secretory product. The uterine tubular gland cells have irregular, frondlike microvilli. During egg shell deposition, these microvilli form large blebs and are probably related to the elaboration of a watery, calcium-containing fluid.  相似文献

The oviduct of non-pregnant females of the ovoviviparous salamander, Salamandra salamandra, was examined using SEM-techniques. In the luminal epithelium polygonal ciliated cells were found along the entire surface of the oviduct, except the uterus, and non-ciliated cells with a varying number of short or long microvilli. The ciliated cells occur in the most anterior portion of the oviduct, the pars recta; they are sparsely distributed in the p. convoluta I, but abundant in the p. convoluta II and III. Non-ciliated cells comprise several small gland cells, restricted to the p. convoluta I, II, III, and undifferentiated cells both provided with microvilli, but difficult to be discerned from their surface appearance. The p. convoluta I, II, III is characterized by three types of secretory cells forming tubular glands, each type confined to a given zone. The secretory cells have slender microvilli at their surfaces. In freeze-cracked glands details of their secretory products can be visualized. The findings are compared to previously published TEM-investigations and discussed with regard to some functions of the oviduct during reproduction.  相似文献

The presence of the mu-opioid receptor and the type of glycosylation in the third extra-cellular loop of this receptor was investigated in the isthmus of mare oviduct during oestrus by means of immunoblotting and immunohistochemistry combined with enzymatic (N-glycosidase F and O-glycosidase) and chemical (beta-elimination) treatments. Immunoblotting analysis showed that the mu-opioid receptor consists of two peptides with molecular weights of around 65 and 50 kDa. After N-deglycosylation with N-glycosidase F an additional immunoreactive peptide was observed at around 30 KDa. The cleavage of O-glycans by O-glycosidase failed in immunoblotting as well as in immunohistochemistry investigations, revealing that the third extra-cellular loop of the mu-opioid receptor expressed in mare isthmus oviduct contains some modifications of the Galbeta(1-3)GalNAc core binding to serine or threonine. Immunohistochemistry revealed the mu-opioid receptor in the mucosal epithelium, some stromal cells, muscle cells and blood vessels. In ciliated cells the mu-opioid receptor showed N-linked glycans, since the immunoreactivity was abolished after N-glycosidase F treatment, whereas it was preserved in the apical region after beta-elimination. Most non-ciliated cells expressed the mu-opioid receptor with both N- and O-linked oligosaccharides, as revealed by the abolition of immunostaining after N-glycosidase F and beta-elimination. Stromal cells, endothelial and muscle cells of blood vessels expressed the mu-opioid receptor containing both N- and O-linked oligosaccharides. Myosalpinx myocytes expressed the mu-opioid receptor with O-linked oligosaccharides. The immunopositive myocytes formed a circular coat in the intrinsic musculature, whereas they were arranged in some isolated, oblique bundles in the extrinsic musculature. In conclusion, the mu-opioid receptor could have a role in the production and the movement of isthmus lumen content that contributes to ensuring the effective condition of the sperm in the mare oviduct.  相似文献

Summary Two-month-old female Swiss mice that had come into estrus were injected intravenously with L-3H-fucose and killed at 5, 15, 40 min, and 4 h after injection. Pieces of the isthmus and of the ampulla of the uterine tubes were processed for light-and electron-microscopic radioautography. Incorporation of 3H-fucose was more intense in the isthmian secretory cells than in the ciliated cells of the ampulla. Electron-microscopic radioautography of the isthmian secretory cells demonstrated that 3H-fucose was incorporated into newly synthesized glycoproteins in the Golgi apparatus from where labelled glycoproteins migrated mainly to secretory granules and apical microvilli. The histochemical technique using ruthenium red confirmed the presence of glycoproteins in the contents of the secretory granules released to the lumen of the uterine tubes as demonstrated by radioautography. Other glycoproteins are transported inside small vesicles and most likely are related to the renewal of the plasma membrane. The role of the secretory glycoproteins in various events of mammalian reproduction is discussed.  相似文献

Thirty parous ewes were divided into six groups and sacrificed on day 0 (first day of estrus), 1, 2, 10, 15 or 16 of the estrous cycle. The cervices were removed immediately and processed for examination with the scanning electron microscope. Observation of the tissues reveals that the surface of the cervix is highly convoluted, which results in the formation of numerous folds or crypts. Two forms of columnar epithelial cells, a ciliated and a non-ciliated cell with microvilli, line the luminal surface of the cerix in the day 10, luteal-phase ewes. However, on day 15, 2 days before estrus, the non-ciliated cells differentiate into two morphologically distinct types of secretory cells. One type forms when the apex of the non-ciliated cell dilates outward into the lumen of the cervix. Concurrent with apical enlargement, the microvilli are lost and the limiting cell membrane becomes smooth. The other type of cell is characterized by only a slight apical swelling. Consequently, remnants of microvilli along with secretory granules can be observed on the limiting membrane of this cell. Both cells release a particulate component, which is believed to be a precursor of mucus, into the lumen of the cerix. These particles undergo a series of morphological transformations to form a fibrillar layer, generally referred to as 'cervical mucus', that covers the epithelial surface at estrus. One to 2 days following the onset of estrus, the fibers become more closely assoicated with amorphous material that begins to coagulate, thereby revealing the underlying ciliated and non-ciliated cells that characterize the cervix of the luteal-phage ewe. The cyclical variation in secretory cells and factors that may influence that structural transformations which occur in mucus are discussed.  相似文献
