We compared the effects of porcine luteinizing hormone (pLH) versus gonadotropin-releasing hormone (GnRH) on ovulatory response and pregnancy rate after timed artificial insemination (TAI) in 605 lactating dairy cows. Cows (mean ± SEM: 2.4 ± 0.08 lactations, 109.0 ± 2.5 d in milk, and 2.8 ± 0.02 body condition score) at three locations were assigned to receive, in a 2 × 2 factorial design, either 100 μg GnRH or 25 mg pLH im on Day 0, 500 μg cloprostenol (PGF) on Day 7, and GnRH or pLH on Day 9, with TAI 14 to 18 h later. Ultrasonographic examinations were performed in a subset of cows on Days 0, 7, 10, and 11 to determine ovulations, presence of corpus luteum, and follicle diameter and in all cows 32 d after TAI for pregnancy determination. In 35 cows, plasma progesterone concentrations were determined 0, 3, 4, 5, 6, 7, and 12 d after ovulation. The proportion of noncyclic cows and cows with ovarian cysts on Day 0 were 12% and 6%, respectively. Ovulatory response to first treatment was 62% versus 44% for pLH and GnRH and 78% versus 50% for noncyclic and cyclic cows (P < 0.01). Location, ovulatory response to first pLH or GnRH, cyclic status, presence of an ovarian cyst, and preovulatory follicle size did not affect pregnancy rate. Plasma progesterone concentrations after TAI did not differ among treatments. Pregnancy rate to TAI was greater (P < 0.01) in the GnRH/PGF/pLH group (42%) than in the other three groups (28%, 30%, and 26% for GnRH/PGF/GnRH, pLH/PGF/GnRH, and pLH/PGF/pLH, respectively). Although only 3% of cows given pLH in lieu of GnRH on Day 9 lost their embryo versus 7% in those subjected to a conventional TAI using two GnRH treatments, the difference was not statistically significant. In summary, pLH treatment on Day 0 increased ovulatory response but not pregnancy rate. Cows treated with GnRH/PGF/pLH had the highest pregnancy rate to TAI, but progesterone concentrations after TAI were not increased. In addition, preovulatory follicle diameter did not affect pregnancy rate.  相似文献

The aim of this study was to evaluate the effect of presynchronization with or without the detection of estrus on first service pregnancy per artificial insemination (P/AI) and on Ovsynch outcome in lactating dairy cows. A total of 511 cows were divided randomly but unevenly into 3 treatment groups at 44 to 50 days in milk (DIM). Ovsynch was started at the same time (69 to 75 DIM) in all three groups. Cows in the Ovsynch group (CON, N = 126) received no presynchronization before Ovsynch, and all cows were bred by timed AI (TAI). Cows in the presynchronization with estrus detection (PED) and the presynchronization with only TAI (PTAI) groups received two doses of prostaglandin F (PGF) 14 days apart, starting at 44 to 50 DIM. Ovsynch was initiated 11 days after the second PGF treatment. Cows in the PED group (N = 267) received AI if estrus was detected after either PGF injection. Cows that were not determined to be in estrus after PGF injection received Ovsynch and TAI. Cows in the PTAI group (N = 118) were not inseminated to estrus, with all cows receiving TAI after Ovsynch. The ovulatory response to the first GnRH injection administered as part of Ovsynch differed (P = 0.002) among treatment groups (83.1% in PTAI, 72.6% in PED, and 62.7% in CON). However, the ovulatory response to the second injection of GnRH during Ovsynch did not differ among treatment groups. Of the 267 PED cows, a total of 132 (49.4%) exhibited estrus and were inseminated. The P/AI at the 31-day pregnancy diagnosis was similar between the cows in the PED group with AI after estrus detection (37.9%; 50/132) and those bred with TAI (34.1%; 46/135). The P/AI in the CON group (46.8%; 59/126) was greater (P < 0.05) than that in the PED group (36.0%; 96/267). In addition, the P/AI in the CON group was greater (P = 0.04) than that in the PED cows receiving TAI (34.1%; 46/135) but less than that in the PED cows bred to estrus (37.9%; 50/132) (P = 0.16). At the 31-day pregnancy diagnosis, the cows in the PTAI group had greater P/AI (55.9%; 66/118) than both those in the PED group (P < 0.01; either estrus or TAI) and those in the CON group (P = 0.08). Thus, presynchronization with PGF (PTAI) increased the ovulatory response to Ovsynch and improved P/AI in dairy cows. Interestingly, the breeding of cows to estrus during presynchronization reduced fertility to the TAI and overall fertility, including cows bred to estrus and TAI. These results indicate that maximal fertility is obtained when all cows receive TAI after the presynchronization protocol.  相似文献

The main aims of the present study were to compare the pregnancy rate (PR), regular returns-to-estrus, and calving interval of a CO-Synch + controlled internal drug release (CIDR) device, commonly used to synchronize ovulations in beef cows, with the classical Ovsynch protocol in high-producing dairy cows. Holstein-Friesian cows (n = 128) from six commercial dairy herds, ≥40 days postpartum and not previously inseminated, were randomly assigned to one of two treatments. Cows submitted to Ovsynch protocol (group OS as control group; n = 66) received 10 μg of a GnRH analogue 7 days before and 48 hours after 25 mg PGF, followed by artificial insemination (AI) 16 hours after the second GnRH administration. Cows submitted to CO-Synch + CIDR (1.38 g of progesterone) inserted for 7 days beginning at the first GnRH administration (group CoS + CD; n = 62) had the second administration of GnRH concurrent with AI, 64 hours after CIDR removal/PGF administration. Nonpregnant cows with return-to-estrus between 18 and 24 days after first AI were reinseminated (second AI). Logistic regressions were used to analyze PR and returns-to-estrus. No effect of group or herd was observed in PR at first timed AI. However, the sum of cows pregnant at first AI and nonpregnant cows with regular returns-to-estrus and the total PR (first + second AI) were influenced by group treatment. Overall, cows of group CoS + CD (total PR = 56.5%) were 2.1 times more likely to became pregnant after AI and until first regular returns-to-estrus than cows of group OS. The calving interval was lower in group CoS + CD (425.9 ± 78.8 days; ±SD) than in group OS (475.3 ± 83.7 days). The CO-Synch + CIDR protocol was reliable to use in dairy herds and provided reproductive advantages when compared with Ovsynch protocol.  相似文献

Two experiments were conducted to test the hypothesis that the 5 d Co-Synch + CIDR (Controlled Internal Drug Release insert containing progesterone) protocol could be applied as an efficient timed AI (TAI) protocol in dairy heifers, and that treatment with flunixin meglumine (FM) during the period of CL maintenance would increase pregnancy per TAI (P/TAI) and late survival of embryos. Objectives were: 1) in Experiment 1, to compare P/TAI with the 5 d Co-Synch + CIDR protocol to a PGF/GnRH protocol; and 2) in Experiment 2, to determine if FM administered 15.5 and 16 d after first TAI would increase P/TAI, using the 5 d Co-Synch + CIDR protocol with a new or previously used (5 d) CIDR insert.In Experiment 1, 248 heifers were assigned randomly to either the PGF/GnRH protocol (n = 120) or the 5 d Co-Synch + CIDR protocol (n = 128). Pregnancy per TAI did not differ between the 5 d Co-Synch + CIDR protocol (53.1%) and the PGF/GnRH protocol (45.8%; P = 0.22). In Experiment 2, 325 heifers synchronized with the 5 d Co-Synch + CIDR protocol were assigned randomly to receive two injections of FM (FM group; n = 158) at 15.5 and 16 d after TAI, or to remain as untreated controls (n = 165). Pregnancy per TAI in Experiment 2 was 59.4 and 59.5% at 45 d for control and FM groups, respectively, with no differences between groups (P = 0.83). The 5 d Co-Synch + CIDR protocol resulted in an acceptable P/TAI in dairy heifers. However, FM did not improve P/TAI in dairy heifers.  相似文献

Our objective was to determine the magnitude of, and factors affecting, pregnancy loss for lactating Holstein cows on a commercial dairy farm when diagnosed with twin (n = 98) or single (n = 518) pregnancies using transrectal ultrasonography. Pregnancy losses were assessed with records of non-viable embryos at first pregnancy examination and embryo losses between the first (25-40 d after AI) and second (48 and 82 d after AI) post-breeding pregnancy examinations. Among cows diagnosed with single pregnancies, 3.7% were diagnosed with a non-viable embryo at first pregnancy examination, and 4.6% of those diagnosed with a viable embryo underwent pregnancy loss by the second examination. A total of 11.2% of cows diagnosed with twins experienced a single embryo reduction, whereas 13.3% lost both embryos. Overall, the total proportion of cows experiencing pregnancy loss or experiencing embryo reduction was greater for cows diagnosed with twin than single pregnancies (odds ratio; OR = 3.6), resulting in an embryo survival rate of 91.9% for cows diagnosed with single compared to 75.5% for cows diagnosed with twin pregnancies. Season of breeding and milk production were associated with pregnancy loss for single pregnancies, whereas CL number was associated negatively with embryo reduction and pregnancy loss for twin pregnancies. The risk of twinning and double ovulation among pregnant cows increased with days in milk (DIM), and the risk of double ovulation was greater for cows diagnosed with ovarian cysts and lacking a CL at initiation of an Ovsynch protocol. We concluded that in this herd, embryo reduction and pregnancy loss during early gestation was greater for lactating Holstein cows diagnosed with twin compared to single pregnancies. In addition, cows diagnosed with ovarian cysts and lacking a CL had an increased risk for double ovulation.  相似文献

Reproductive efficiency is not optimal in high-producing dairy cows. Although many aspects of ovarian follicular growth in cows are similar to those observed in heifers, there are numerous specific differences in follicular development that may be linked with changes in reproductive physiology in high-producing lactating dairy cows. These include: 1) reduced circulating estradiol (E2) concentrations near estrus, 2) ovulation of follicles that are larger than the optimal size, 3) increased double ovulation and twinning, and 4) increased incidence of anovulation with a distinctive pattern of follicle growth in anovular dairy cows. The first three changes become more dramatic as milk production increases, although anovulation has not generally been associated with level of milk production. To overcome reproductive inefficiencies in dairy cows, reproductive management programs have been developed to synchronize ovulation and enable the use of timed AI in lactating dairy cows. Effective regulation of the CL, follicles, and hormonal environment during each part of the protocol is critical for optimizing these programs. This review discusses the distinct aspects of follicular development in lactating dairy cows and the methodologies that have been utilized in the past two decades in order to manage the dominant follicle during synchronization of ovulation and timed AI programs.  相似文献

After 80 years of the commercial application of artificial insemination (AI) in the cow, the method still has numerous benefits over natural insemination including worldwide gene improvement. The efficiency of insemination depends, among many other factors, on the delivery of an appropriate number of normal spermatozoa to the appropriate reproductive tract site at the appropriate time of estrus. The metabolic clearance of steroid hormones and pregnancy associated glycoproteins and the negative effects of different types of stress related to high milk production makes the high-producing dairy cow a good animal model for addressing factors affecting fertility. Nevertheless, extensive studies have shown a positive link between high milk production in an individual cow and high fertility. When a cow becomes pregnant, the effect of pregnancy loss on its reproductive cycle is also a topic of interest. This paper reviews the factors of a noninfectious nature that affect the fertility of lactating dairy cows following AI. Special attention is paid to factors related to the cow and its environment and to estrus confirmation at insemination. Pregnancy maintenance during the late embryonic/early fetal period is discussed as a critical step. Finally, the use of Doppler ultrasonography is described as an available research tool for improving our current understanding of the health of the genital structures and conceptus.  相似文献

To determine the efficacy of reducing the dosage of GnRH used in a protocol for synchronization of ovulation and timed AI, primiparous and multiparous lactating Holstein cows (n=237) were randomly assigned to 1 of 2 treatment groups. Ovulation was synchronized for cows in the first group using intramuscular injections of GnRH and PGF2 as follows: Day 0, 100 μg GnRH; Day 7, 25 mg PGF2; Day 9, 100 μg GnRH. Ovulation was synchronized in the second group of cows using the same injection schedule and dosage of PGF2 but only 50 μg GnRH per injection. All cows underwent a timed AI at 12 to 18 h after the second GnRH injection. The proportion of cows ovulating in response to the second GnRH injection (synchronization rate) and pregnancy status at 28 and 56 d post AI were determined using transrectal ultrasonography. The synchronization rate, double-ovulation rate, conception rate at 28 and 56 d post AI, and pregnancy loss from 28 to 56 d post AI did not differ statistically between treatment groups. For all cows, synchronization rate was 84.0%, and double-ovulation rate was 14.1%. Conception rates calculated using all cows receiving synchronization of ovulation were 41.1% at 28 d and 34.4% at 56 d post AI. Conception rates calculated for only synchronized cows were 47.6% at 28 d and 40.1% at 56 d post AI. For all cows, pregnancy loss from 28 to 56 d post AI was 13.5%, with an attrition rate of 0.5% per day. Estimated savings in hormone costs using 50 rather than 100 μg GnRH per injection for synchronizing ovulation were $6.40 per cow and $20.27 per pregnancy. Thus, decreasing the dosage of GnRH used for synchronization of ovulation and timed AI in lactating dairy cows reduces synchronization costs per cow and per pregnancy without compromising the efficacy of the synchronization protocol.  相似文献

Two modifications of the Ovsynch protocol, GnRH + TAI after PGF 48 h (CO-48) or 72 h (CO-72), were compared with the original protocol (OVS: GnRH–7 d–PGF–2 d–GnRH–16 h–TAI) to study their effects on reproductive performance in 785 lactating dairy cows (Holstein Friesian, Bos Taurus). Results showed that more cows (P < 0.001) returned to estrus within a week after TAI with CO-48 treatment compared with that in OVS and CO-72 treatments. Pregnancy rates were greater (P < 0.001) for the CO-72 cows than those for both OVS and CO-48 treatments and for primiparous cows compared with multiparous cows. Moreover, pregnancy rates were lower in summer compared with those in winter. Pregnancy losses for cows in both CO-48 and CO-72 were greater (P < 0.05) than that for cows in OVS treatment. Pregnancy losses were greater in summer (P < 0.001) than in winter and for multiparous cows (P < 0.001) than for primiparous cows. In conclusion, primiparous and winter-bred cows had greater pregnancy rates and fewer pregnancy losses than those of multiparous cows and summer-bred cows, respectively. Because of the presence of significant treatment, parity, and season interactions, TAI with ovulation synchronization protocols should be tailored according to the season and parity. CO-72 is recommended for primiparous cows but not for multiparous cows, and CO-48 is not recommended for synchronization. Furthermore, cows that exhibited estrus at any time were inseminated to improve pregnancy rates in ovulation synchronization protocols.  相似文献

Previous studies have demonstrated the facilitating effect of a single cloprostenol treatment postinsemination on ovulation and pregnancy rate in dairy cattle and Italian buffalos. This study was conducted to test the effects of 500 μg cloprostenol given intramuscularly immediately postinsemination to 55 primiparous and 60 multiparous Holstein cows (Bos taurus; TR) of a typical dairy operation in East Germany and to compare them with 57 primiparous and 48 multiparous saline-treated cows (CON). Animals of the TR and CON groups did not differ or only differed marginally for age at treatment, interval calving–treatment, lactation number, milk production on the day of treatment, body condition score, and their peri- and postparturient case histories. All animals were clinically and reproductively healthy on the day of treatment. They were inseminated once 12 h after the onset of estrus, scanned 24 h after insemination to confirm ovulation, and tested for pregnancy by transrectal palpation between Days 42 and 48 postinsemination. Treatment did not affect the number of cows ovulating. The overall combined pregnancy rate (PR) was 47.3%. Pregnancy rate did not differ statistically between TR and CON (46.1% vs. 48.6%; P > 0.05). In conclusion, this study failed to demonstrate beneficial effects of a postinsemination treatment of primiparous and multiparous cows with cloprostenol on ovulation and PR in a typical dairy cattle operation in East Germany.  相似文献

The development and application of an algorithm to assess the ability of an infrared thermography (IRT) device to predict cows in estrus and about to ovulate was investigated. Twenty cows were synchronized using a controlled internal drug release and PGF2α. Vulval and muzzle temperatures were measured every 12 hours from controlled internal drug release insertion to 32 hours after PGF2α treatment and then every 4 hours until ovulation occurred or until 128 hours after PGF2α treatment (whichever occurred first). Thermal images obtained with a FLIR T620 series infrared camera were analyzed using ThermaCAM Researcher Professional 2.9 software. Cows were also monitored for behavioral signs of estrus and color changes of an Estrotect applied to the tail head of each cow 36 hours after PGF2α treatment. Algorithms were developed by adjusting body surface temperature of individual animals for ambient temperature and humidity during each observation period, and were expressed as a deviation from the baseline temperature. Of the 20 cows enrolled in this study, 12 (60%) ovulated. An IRT estrus alert was defined using different thresholds (D = 1 °C, 1.25 °C, and 1.5 °C). Sensitivity and specificity to predict estrus depended upon the chosen threshold level. At a threshold D = 1 °C, the highest sensitivity (92%; n = 11) and the lowest specificity (29%) and positive predictive value (64%) were observed. Conversely, D = 1.5 °C resulted in sensitivity of 75%, specificity of 57%, and positive predictive value of 69%. The mean ± standard deviation intervals between onset and the end of IRT estrus alert to ovulation were 30.7 ± 8.2 and 13.3 ± 7.7 hours, respectively. Ovulation occurred 24 to 47 hours after the onset of the IRT estrus alert for eight out of the 11 ovulated cows (73%). Although the sensitivity of the IRT alert was greater than visual observation (67%) and Estrotect activation (67%), the specificity and positive predictive value were lower than these two aids (i.e., the IRT overpredicted the incidence of ovulation). Results presented indicate that IRT shows some potential as an estrus detection aid; however, further studies investigating the potential to improve the specificity and capturing data throughout entire 21-day reproductive cycles would be worthwhile.  相似文献

The uterine condition of clinically normal postpartum Holstein-Friesian dairy cows (n = 45) was evaluated once weekly (Weeks 3 to 7) by endometrial cytology, vaginal mucus collection device (VMCD), vaginoscopy, and ultrasonography to establish a relationship with postpartum resumption of ovulatory cycles. The time of first detection of the corpus luteum (CL) by ultrasonography and plasma progesterone concentration ≥1 ng/mL was recorded. By 49 d postpartum, 78% of the cows (n = 35) had resumed ovarian function (CL group), whereas the remainder (n = 10) had no CL (NCL group). There was a positive correlation between VMCD score and presence of fluid in the uterus in cows with a CL (P < 0.01) during Week 3 postpartum but no significant correlation in cows without a CL. Percentage of polymorphonuclear leukocytes (PMN%) was higher in the NCL group (mean ± SEM, 24.6 ± 9.4%) than in the CL group (11.7 ± 2.2%) during Week 5 postpartum (P < 0.05). The PMN% (4.5 ± 6.5%) and VMCD (0.5 ± 0.5) scores during Week 5 in cows ovulating by Day 28 were lower (P < 0.01) than the PMN% (15.0 ± 14.3%) and VMCD (1.1 ± 0.9) scores in those ovulating by Day 49. In conclusion, higher PMN% at 5 wk postpartum was associated with delayed resumption of ovarian cyclicity in high-producing dairy cows.  相似文献

Lactating Holstein cows (n = 135) were randomly assigned to one of the two sources of supplemental selenium (Se), sodium selenite (SS) or Se yeast (SY), fed at 0.3 mg/kg diet dry matter from 25 d before calving to 70 d in milk (DIM), in diets not suboptimal in basal Se concentrations. Cows were evaluated for health daily in the first 10 DIM, and uterine cytology of the previously gravid uterine horn was assessed at 30 DIM. The Ovsynch protocol was initiated at 42 DIM; ovarian responses to hormonal treatments were evaluated by ultrasonography. The uteri of cows were flushed 6 d after timed AI for collection of embryos and oocytes. Plasma concentrations of Se and progesterone were measured throughout the postpartum period and during the reproductive protocol, respectively, and plasma glutathione peroxidase activity was determined 6 d after AI. Concentrations of Se in pre- and postpartum diets ranged from 0.43 to 0.56 mg/kg of dry matter. Incidence of retained placenta, fever, ketosis, mastitis, acute puerperal metritis, clinical endometritis, and subclinical endometritis were not significantly different between treatments. There were no differences between groups in concentrations of Se and progesterone or glutathione peroxidase activity in plasma. Treatment did not influence ovarian responses to the synchronization protocol, fertilization rate, number of blastomeres and live blastomeres, or proportions of grades 1 and 2, degenerated, and degenerated-unfertilized embryos/oocytes. Odds of subclinical endometritis on Day 30 postpartum more than doubled in cows with fever of unknown origin or acute puerperal metritis in the first 10 DIM. Fertilization rate tended to be reduced in cows with subclinical endometritis. In summary, replacing SS with an organic source of Se in diets not suboptimal in basal Se concentrations did not improve Se status, uterine health, fertilization, or embryo quality in early lactation dairy cows.  相似文献

Most dairy cows experience negative energy balance (NEB) in early lactation because energy demand for milk synthesis is not met by energy intake. Excessive NEB may lead to metabolic disorders and impaired fertility. To optimize herd management, it is useful to detect cows in NEB in early lactation, but direct calculation of NEB is not feasible in commercial herds. Alternative methods rely on fat-to-protein ratio in milk or on concentrations of non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHB) in blood. Here, we considered methods to assess energy balance (EB) of dairy cows based on the fatty acid (FA) composition in milk. Short- and medium-chain FAs (primarily, C14:0) are typically synthesized de novo in the mammary gland and their proportions in milk fat decrease during NEB. Long-chain FAs C18:0 and C18:1 cis-9 are typically released from body fat depots during NEB, and their proportions increase. In this study, these FAs were routinely determined by Fourier-transform infrared spectroscopy (FTIR) of individual milk samples. We performed an experiment on 85 dairy cows in early lactation, fed the same concentrate ration of up to 5 kg per day and forage ad libitum. Daily milk yield and feed intake were automatically recorded. During lactation weeks 2, 4, and 6 after calving, two milk samples were collected for FTIR spectroscopy, Tuesday evening and Wednesday morning, blood plasma samples were collected Thursday morning. Net energy content in feed and net energy required for maintenance and lactation were estimated to derive EB, which was used to compare alternative indicators of severe NEB. Linear univariate models for EB based on NEFA concentration (deviance explained = 0.13) and other metabolites in blood plasma were outperformed by models based on concentrations of metabolites in milk: fat (0.27), fat-to-protein ratio (0.18), BHB (0.20), and especially C18:0 (0.28) and C18:1 cis-9 (0.39). Analysis of generalized additive models (GAM) revealed that models based on milk variables performed better than those based on blood plasma (deviance explained 0.46 vs. 0.21). C18:0 and C18:1 cis-9 also performed better in severe NEB prediction for EB cut-off values ranging from −50 to 0 MJ NEL/d. Overall, concentrations of C18:0 and C18:1 cis-9 in milk, milk fat, and milk BHB were the best variables for early detection of cows in severe NEB. Thus, milk FA concentrations in whole milk can be useful to identify NEB in early-lactation cows.  相似文献

Our objective was to determine whether rates of luteolysis or pregnancy differed in lactating dairy cows of known progesterone status and either known or unknown luteal status after either cloprostenol or dinoprost was injected as part of a timed-insemination program. In Experiment 1, 2358 lactating dairy cows in six herds were given two injections of PGF 14 d apart (Presynch), with the second injection given 12 to 14 d before the onset of a timed AI protocol (Ovsynch). Cows (n = 1094) were inseminated when detected in estrus after the Presynch PGF injections. Cows not inseminated (n = 1264) were enrolled in the Ovsynch protocol and assigned randomly to be treated with either cloprostenol or dinoprost as part of the timed-AI protocol. In cows having pretreatment concentrations of progesterone ≥ 1 ng/mL and potentially having a functional corpus luteum (CL) responsive to cloprostenol (n = 558) or dinoprost (n = 519), dinoprost increased (P < 0.05) luteal regression from 86.6 to 91.3%. Despite a significant increase in luteolysis, pregnancies per AI did not differ between luteolytic agents (dinoprost = 37.8% and cloprostenol = 36.7%). Fertility was improved in cows of both treatments having reduced concentrations of progesterone at 72 h and in cows showing signs of estrus. In Experiment 2, an ovulation-resynchronization program was initiated with GnRH or saline in 427 previously inseminated lactating dairy cows of unknown pregnancy status in one herd. Seven days later, pregnancy was diagnosed and nonpregnant cows were blocked by number of CL and assigned randomly to be treated with cloprostenol or dinoprost. Compared with cloprostenol, dinoprost increased (P < 0.05) luteal regression from 69.1 to 78.5%, regardless of the number of CL present or the total luteal volume per cow. Pregnancies per AI did not differ between dinoprost (32.8%) and cloprostenol (31.3%). Although dinoprost was more effective than cloprostenol at inducing luteolysis in lactating dairy cows exposed to an Ovsynch or ovulation-resynchronization protocol, resulting fertility did not differ between products.  相似文献

The objectives were to compare: (1) preovulatory serum LH concentrations, and (2) synchronization of ovulation, after im or iu administration of the second GnRH treatment of Ovsynch in lactating dairy cows. Lactating cows (N = 23) were presynchronized with two injections of PGF given 14 days apart (starting at 34 ± 3 days in milk), followed by Ovsynch (GnRH-7 d-PGF-56 h-GnRH) 12 days later. At the time of the second GnRH of Ovsynch (Hour 0), cows were blocked by parity and randomly assigned to 1 of 3 groups: (1) control group (CON; N = 7) were given 2 mL sterile water im; (2) intramuscular group (IM; N = 8) received 100 μg of GnRH im; and (3) intrauterine group (IU; N = 8) had 100 μg GnRH infused in the uterus (2 mL). Blood samples for serum LH concentrations were collected at Hours 0, 0.5, 1, 1.5, 2, 3, and 4. Furthermore, ultrasonography was performed twice daily (12-h intervals) from Hours 0 to 60 to confirm ovulation. The LH concentrations were greater (P < 0.05) in the IM than IU and CON groups at Hours 0, 0.5, 1, 1.5, 2, 3, and 4. Although LH concentrations were numerically higher in the IU group, LH concentrations within the IU and CON groups did not change over time. More cows ovulated in the IM (8/8) and IU (7/8) groups within 60 h after the second GnRH administration compared with the CON (2/7) group. In summary, serum LH concentrations were lower in the IU versus IM group, but the proportion of cows that ovulated within 60 h was similar between these two groups. Therefore, iu administration of GnRH may be an alternative route of delivery to synchronize ovulation in beef and dairy cattle.  相似文献

Intrafollicular insemination (IFI) is based on direct introduction of a sperm suspension into a preovulatory follicle. To our knowledge, the first cases of IFI resulting in pregnancy of the cow are reported here. The experiment was performed on a dairy herd with low fertility during the warm season of the year. Following the sequence of estrus detection, one in three inseminations was intrafollicular (n = 17), whereas deposition of semen was performed into the uterine body (IUI) in the remaining cows (n = 33). Approximately 0.06 ml of a seminal dose containing five million spermatozoa (one quarter of a commercial seminal dose in a 0.25 ml French straw) was injected trans-vaginally into the preovulatory follicle in the IFI group. Four (23.5%) and 3 (9%) cows of the IFI and IUI groups, respectively, became pregnant. On days 8-11 post-insemination, ovulation failure was registered in 3 (18%) and in 4 (12%) cows of the IFI and IUI groups, respectively. Our results suggest that IFI could be used as an alternative procedure to the usual deposition of semen into the uterus in cows of low fertility.  相似文献

It has been established that the immunologic and endocrine status of the peripartum dairy cow determines the animal's subsequent productive and reproductive performance. Thus, at parturition reduced immune functions of peripheral blood polymorphonuclear cells (PMN) has been observed after a peak in pregnancy-associated glycoproteins (PAGs), and, more recently, the inseminating bull was linked to plasma levels of bovine PAGs in pregnant Holstein-Friesian dairy cows. The present study sought to determine whether changes in leukocyte counts during the peripartum period, indicative of the animal's immune status, could be related to the inseminating bull and to PAG levels. Ninety-six clinically healthy, single pregnant cows in a commercial dairy herd were selected. Four samples were collected before parturition (on gestation Days 220-226, 234-240, 248-254, and 262-268) and two samples after parturition (on Days 14-21, and 28-34 postpartum) to analyze total and differential blood cell counts. Based on GLM analysis procedures of variance for repeated measures, the inseminating bull was found to affect counts of total leukocytes and lymphocytes (P < 0.001; between-subject effects) throughout the peripartum period. In addition, cows with high plasma PAG levels (> 900 ng/ml) on Day 262-268 of gestation had higher numbers of total leukocytes and neutrophils throughout the peripartum (P < 0.001; between-subject effects). Young animals (≤ 1 lactation) had higher total leukocyte and lymphocyte counts than older cows (2 or more lactations) throughout the study period. These results reveal a clear relationship between the inseminating bull or plasma PAG levels and peripheral leukocyte counts during the peripartum period in dairy cows.  相似文献

The objective of this study was to examine the effect of a chronic stressor, lameness, on reproductive parameters. Seventy cows 30-80 days post-partum were scored for lameness and follicular phases synchronized with GnRH followed seven days later by prostaglandin (PG). Fifteen Lame animals did not respond to GnRH ovarian stimulation. Milk progesterone for 5 days prior to PG was lower in the remaining Lame cows than Healthy herdmates. Fewer Lame cows ovulated (26/37 versus 17/18; P = 0.04) and the interval from PG to ovulation was shorter in Lame cows. In Subset 1 (20 animals), the LH pulse frequency was similar in ovulating animals (Lame and Healthy) but lower in Lame non-ovulators. An LH surge always preceded ovulation but lameness did not affect the interval from PG to LH surge onset or LH surge concentrations. Before the LH surge, estradiol was lower in non-ovulating cows compared to those that ovulated and estradiol concentrations were positively correlated with LH pulse frequency. In Subset 2 (45 cows), Lame ovulating cows had a less intense estrus than Healthy cows, although Lame cows began estrus and stood-to-be-mounted earlier than Healthy cows. In conclusion, we have identified several parameters to explain poor fertility in some chronically stressed animals. From 30 to 80 days post-partum, there was a graded effect that ranged from 29% Lame cows with absence of ovarian activity, whereas another 21% Lame cows failed to express estrus or ovulate a low estrogenic follicle; in 50% cows, many reproductive parameters were unaffected by lameness.  相似文献
