Euryhaline crustaceans tolerate exposure to a wide range of dilute media, using compensatory, ion regulatory mechanisms. However, data on molecular interactions occurring at cationic sites on the crustacean gill (Na+,K+)-ATPase, a key enzyme in this hyperosmoregulatory process, are unavailable. We report that Na+ binding at the activating site leads to cooperative, heterotropic interactions that are insensitive to K+. The binding of K+ ions to their high affinity sites displaces Na+ ions from their sites. The increase in Na+ ion concentrations increases heterotropic interactions with the K+ ions, with no changes in K0.5 for K+ ion activation at the extracellular sites. Differently from mammalian (Na+,K+)-ATPases, that from C. danae exhibits additional NH4+ ion binding sites that synergistically activate the enzyme at saturating concentrations of Na+ and K+ ions. NH4+ binding is cooperative, and heterotropic NH4+ ion interactions are insensitive to Na+ ions, but Na+ ions displace NH4+ ions from their sites. NH4+ ions also displace Na+ ions from their sites. Mg2+ ions modulate enzyme stimulation by NH4+ ions, displacing NH4+ ion from its sites. These interactions may modulate NH4+ ion excretion and Na+ ion uptake by the gill epithelium in euryhaline crustaceans that confront hyposmotic media.  相似文献

Ammonium (NH4+) and nitrate (NO3) are major inorganic nitrogen (N) sources for plants. When serving as the sole or dominant N supply, NH4+ often causes root inhibition and shoot chlorosis in plants, known as ammonium toxicity. NO3 usually causes no toxicity and can mitigate ammonium toxicity even at low concentrations, referred to as nitrate-dependent alleviation of ammonium toxicity. Our previous studies indicated a NO3 efflux channel SLAH3 is involved in this process. However, whether additional components contribute to NO3-mediated NH4+ detoxification is unknown. Previously, mutations in NO3 transporter NRT1.1 were shown to cause enhanced resistance to high concentrations of NH4+. Whereas, in this study, we found when the high-NH4+ medium was supplemented with low concentrations of NO3, nrt1.1 mutant plants showed hyper-sensitive phenotype instead. Furthermore, mutation in NRT1.1 caused enhanced medium acidification under high-NH4+/low-NO3 condition, suggesting NRT1.1 regulates ammonium toxicity by facilitating H+ uptake. Moreover, NRT1.1 was shown to interact with SLAH3 to form a transporter-channel complex. Interestingly, SLAH3 appeared to affect NO3 influx while NRT1.1 influenced NO3 efflux, suggesting NRT1.1 and SLAH3 regulate each other at protein and/or gene expression levels. Our study thus revealed NRT1.1 and SLAH3 form a functional unit to regulate nitrate-dependent alleviation of ammonium toxicity through regulating NO3 transport and balancing rhizosphere acidification.  相似文献

运用15N稳定性同位素技术,对15N标记的硝酸盐和铵盐在输入小嵩草(Kobresia pygaea C.B.Clarke)草甸11~13个月后的运移规律进行了研究.在经历11~13个月后,进入无机氮库中的15N很少,一般不超过所输入氮素的l%,而较多的1 5N为土壤有机质、土壤微生物和植物所固持.NO3--15N和NH4 -1 5N在小嵩草草甸中的运移规律差异很大.在11、12和13个月后,NO3--15N的总恢复率分别为92.83%、92.64%和79.96%;而NH4 -15N的则分别为49.6%、63.33%和66.22%.两者的差异在土壤有机质、土壤微生物和植物等库之间的分配中更加明显.输入NO3--15N时在11、12个月后植物所固持的15N最多,而土壤微生物和土壤有机质所固持的15N比较接近,而在13个月后,土壤有机质和植物所固持的15N接近,而土壤微生物所固持的15N下降许多;当输入NH4 -15N,土壤有机质所固持的1 5N比植物和土壤微生物所固持的都多,而且植物所固持的15N比较稳定,而土壤微生物所固持的15N则有较大变化.这表明在较长的时间内嵩草草甸对NO3-和NH4 的固持能力是不一样的.  相似文献

The role of Na+ in Vibrio alginolyticus oxidative phosphorylation has been studied. It has been found that the addition of a respiratory substrate, lactate, to bacterial cells exhausted in endogenous pools of substrates and ATP has a strong stimulating effect on oxygen consumption and ATP synthesis. Phosphorylation is found to be sensitive to anaerobiosis as well as to HQNO, an agent inhibiting the Na+-motive respiratory chain of V. alginolyticus. Na+ loaded cells incubated in a K+ or Li+ medium fail to synthesize ATP in response to lactate addition. The addition of Na+ at a concentration comparable to that inside the cell is shown to abolish the inhibiting effect of the high intracellular Na+ level. Neither lactate oxidation nor Δω generation coupled with this oxidation is increased by external Na+ in the Na+-loaded cells. It is concluded that oxidative ATP synthesis in V. alginolyticus cells is inhibited by the artificially imposed reverse ΔPNa, i.e., [Na+]in > [Na+]out. Oxidative phosphorylation is resistant to a protonophorous uncoupler (0.1 mM CCCP) in the K+-loaded cells incubated in a high Na+ medium, i.e., when ΔpNa of the proper direction ([Na+]in < [Na+]out) is present. The addition of monensin in the presence of CCCP completely arrests the ATP synthesis. Monensin without CCCP is ineffective. Oxidative phosphorylation in the same cells incubated in a high K+ medium (ΔpNa is low) is decreased by CCCP even without monensin. Artificial formation of ΔpNa by adding 0.25 M NaCl to the K+-loaded cells (Na+ pulse) results in a temporary increase in the ATP level which spontaneously decreases again within a few minutes. Na+ pulse-induced ATP synthesis is completely abolished by monensin and is resistant to CCCP, valinomycin and HQNO. 0.05 M NaCl increases the ATP level only slightly. Thus, V. alginolyticus cells at alkaline pH represent the first example of an oxidative phosphorylation system which uses Na+ instead of H+ as the coupling ion.  相似文献

分析了2003年11月-2004年10月广州市龙洞79次降雨的化学组分及其各组分之间的相关关系。得出:(1)观测期间总降雨量1359.8,单次降雨的pH值在3.22-7.29之间,平均4.83;以pH<5.6为酸雨的临界值,则酸雨频率占降雨次数的77.2%或占降雨量的81.5%;(2)广州降水的化学组成以SO42-、Cl-和NO3-为主要阴离子,阳离子中以Ca2+、NH4+、Na+所占比例较大;(3)广州地区酸雨属硫酸型,降雨中的SO42-、NO3-、PO43-、Cl-、NH4+、K+、Na+、Ca2+、Mg2+离子浓度分别为0.150,0.045,0.002,0.163,0.058,0.006,0.013,0.083和0.007mol·L-1,单次降雨的SO42-/NO3-比值在0.42-17.6之间,年平均为3.33,该值比西南地区相应值低许多,而且与同一地点5年前测定的结果相比较,SO42-/NO3-比值有变小的趋势,说明广州地区NOx排放量高逐渐增加;(4)广州地区酸雨有着明显的季节变化规律,春、夏、秋季酸雨比较严重,冬季酸雨较少出现,这与西南地区的重庆(冬季酸雨较春季严重)酸雨季节规律相反;(5)通过SPSS统计分析得出,雨量与各化学组分含量成负相关,说明雨量对大气污染物质的稀释作用;酸雨各化学成分之间存在着一定的相关性,如SO42-与Ca2+,NO3-与NH4+,K+与Na+、Mg2+离子,Na+与Mg2+等都存在显著的正相关,利用这种相关可以建立某些酸雨成分的预测模型。  相似文献

Acute hyperammonemia, induced by two consecutive injections of ammonium acetate (550 and 450 mg per kg b.wt.), decreased the activity of γ-glutamyl transpeptidase (GGT) in most brain regions of 18- and 30-day-old rats. This decrease in the brain GGT activity was more pronounced in younger than in older rats. After the addition or NH4Cl to the incubation medium, the inhibitory action of NH4+ on this enzyme activity was also demonstrated in crude synaptosomal membranes at pH 7.4, but in a range of NH4+ concentrations many-times higher then those found in the plasma or brains of young hyperammonemic rats. Because similar concentrations of NH4+ stimulated the activity of the purified enzyme from rat kidney (mainly at pH 9.0), the inhibition of GGT activity in the young rat brain is probably mediated indirectly and not by a direct interaction of ammonia with the enzyme molecules.  相似文献

A large number of studies indicate that K+-induced contractions of smooth muscle depend on extracellular calcium. If these contractions depend exclusively on extracellular calcium then contractile responses to 140 mM K+, which are larger than the response to 35 mM K+, should be associated with a larger influx of 45Ca. This is not the case in the vas deferens from reserpine pretreated rats. During a 2 min interval, 45Ca influx induced by 140 mMK+ was identical to that produced by 35 mM K+. This suggests that a second mechanism may be involved in responses to high K+. Indeed, 140 mM K+ caused an approximately 300% increase above control in the formation of inositol trisphosphate (IP3) in tissues prelabelled with 3H-myoionositol whereas 35 mM K+ did not increase IP3. IP3 is thought to cause the release of calcium from internal stores which is consistent with our finding of an increase in 45Ca efflux into calcium-free medium from tissues prelabelled with 45Ca and stimulated with 140 mM K+. Stimulation with 35 mM K+ did not influence 45Ca efflux. We conclude that in the rat vas deferens high K+ promotes tension development by smooth muscle by a dual mechanism: influx of extracellular calcium and release of calcium from internal stores via a IP3 mechanism.  相似文献

硝酸铵、硫酸铵和硫酸氢铵等铵盐作为PM2.5中主要的二次无机气溶胶污染物,在灰霾形成过程中发挥了重要作用.关于其来源及转化过程的研究备受关注.本文在前人研究基础上,基于稳定同位素分析技术改进了样品进样量,增加了调节浸提液pH的步骤,给出了一种能够快速、准确测量大气样品中铵态氮同位素的化学转化方法.即向含有0.25 μg·mL-1铵态氮的4 mL大气气溶胶滤膜样品浸提液中加入碱性次溴酸盐溶液,使铵盐(NH4+)氧化为亚硝酸盐(NO2-);然后通过调节pH值,在 pH<0.3 的条件下,使用盐酸羟胺将亚硝酸盐(NO2-)还原为氧化亚氮气体(N2O),最终通过Precon-GasBench-IRMS系统的连用实现大气气溶胶样品中的铵态氮同位素的测定.该方法所需进样含量低,避免了剧毒易制爆试剂的使用,重复测试精确度可达到0.2‰(n=10).通过调节大气气溶胶滤膜样品浸提液的pH可提高还原效率(约100%).测量精确度和准确度满足了大气气溶胶中颗粒态铵的测定,有助于进一步开展大气铵盐的来源、化学转化过程、传输沉降等研究.  相似文献

以‘中农26’黄瓜为试材,研究亚适宜温光(18 ℃/10 ℃,180±20 μmol·m-2·s-1)下不同硝铵比(26:2、21:7和14:14)营养液对黄瓜幼苗生长和生理特性的影响.结果表明:硝铵比为21:7处理的黄瓜总根长最长、根体积和根表面积最大、根尖数最多;根中H+-ATPase活性最高;硝态氮转运蛋白(NRT)和铵转运蛋白(AMT)基因表达量也升高,提高了根系的氮吸收能力;叶中硝酸还原酶(NR)、谷氨酰胺合成酶(GS)和谷氨酸合酶(GOGAT)活性较高,提高了体内氮代谢能力,使黄瓜氮含量和干物质积累显著增加.亚适宜温光下,营养液中硝铵比为21:7时,黄瓜幼苗干质量分别比26:2和14:14处理增加14.0%和19.3%.亚适宜温光下,可通过调整营养液中硝铵比,提高黄瓜氮吸收与代谢能力,缓解亚适宜温光对黄瓜幼苗的不利影响,促进黄瓜生长.  相似文献

Nitric oxide (NO) is a highly reactive, membrane-permeable free radical, which has recently emerged as an important antioxidant. Here we investigated the protective effect of NO against the toxicity and NH4+ accumulation in rice leaves caused by excess CuSO4 (10 mmol L−1). It was found that free radical scavengers (sodium benzoate, thiourea, and reduced glutathione) reduced the toxicity and NH4+ accumulation in rice leaves caused by excess CuSO4. NO donor sodium nitroprusside (SNP) was also effective in reducing CuSO4-induced toxicity and NH4+ accumulation in rice leaves. The protective effect of SNP on the toxicity and NH4+ accumulation can be reversed by 2-(4-carboxy-2-phenyl)-4,4,5,5-tetramethyl- imidazoline-1-oxyl-3-oxide, a NO scavenger, suggesting that the protective effect of SNP is attributable to NO released. Results obtained in the present study suggest that reduction of CuSO4-induced toxicity and NH4+ accumulation by SNP is most likely mediated through its ability to scavenge active oxygen species.  相似文献

Electron self-exchange in solutions of the ‘blue’ copper protein plastocyanin is catalysed by the redox-inert multivalent cations Mg2+ or Co(NH3)3+6. Measurements of specific 1H-NMR line broadening with 50% reduced solutions in the presence of these cations show that electron exchange proceeds through encounters of cation-protein complexes which dissociate at high ionic strength. In the presence of 8mM (5 equivalents/total protein) Co(NH3)3+6, with 10 mM cacodylate (pH*6.0) as background electrolyte, the bimolecular rate constant at 25°C is 7 × 104 M−1·s−1. For comparison, the ‘electrostatically screened’ rate constant measured in 0.1 M KCl in the absence of added multivalent cations is ˜ 4 × 103 M1·s−1.

At pH 7, addition of glucose under anaerobic conditions to a suspension of the yeast Saccharomyces cerevisiae causes both a transient hyperpolarization and a transient net efflux of K+ from the cells. Hyperpolarization shows a peak at about 3 min and a net K+ efflux at 4–5 min. An additional transient hyperpolarization and net K+ efflux are found after 60–80 and 100 min, respectively. Addition of 2-deoxyglucose instead of glucose does not lead to hyperpolarization of the cells or K+ efflux. At low pH, neither transient hyperpolarization nor a transient K+ efflux are found. With ethanol as substrate and applying aerobic conditions, both a transient hyperpolarization and a transient K+ efflux are found at pH 7. The fluorescent probe 2-(dimethylaminostyryl)-1-ethylpyridinium appears to be useful for probing changes in the membrane potential of S. cerevisiae. It is hypothesized that the hyperpolarization of the cells is due to opening of K+ channels in the plasma membrane. Accordingly, the hyperpolarization of the cells at pH 7 is almost completely abolished by 1.25 mM K+, whereas the same amount of Na+ does not reduce the hyperpolarization  相似文献

2012年雨季(4—9月),收集广州市城市区、近郊区和远郊区森林公园的PM2.5样品,测定PM2.5质量浓度,分析了其中SO42-、NO3-、NO2-、Cl-、F-、Na+、NH4+、Ca2+、K+、Mg2+ 共10种水溶性无机离子含量.结果表明:帽峰山(远郊)、大夫山(近郊)、火炉山(城区)PM2.5质量浓度的日变化分别为17.2~66.5、19.4~156.3、21.8~161.7 μg·m-3,平均值分别为44.4、49.8、55.9 μg·m-3.SO42-、Na+和NH4+为水溶性无机离子主要组分,其中,SO42-含量最大,并从城区至郊区呈递减趋势.固定源对3个森林公园空气中SO2和NOx的贡献大于移动源,从城区至远郊呈递减趋势,说明机动车对城区空气中SO2和NOx的贡献大于近郊和远郊森林公园.采样期间,海盐对大夫山空气PM2.5中水溶性组分的贡献最大,其中K+受海盐的影响超过其他元素.NH4+当量浓度远小于SO42-和NO3-的当量浓度,中和度远小于1,反映PM2.5酸性较强,且从远郊至城区PM2.5粒子酸性呈增强趋势.  相似文献

Oxidative stress to B-cells is thought to be of relevance in declining B-cell function and in the process of B-cell destruction. In other tissues including heart, brain and liver, oxidative stress has been shown to elevate the intracellular free calcium concentration and to provoke potassium efflux. We studied the effect of oxidative stress on Ca2+ and K+ (Rb+) outflow from pancreatic islets using the thiol oxidants DIP and BuOOH. Both compounds reversibly increased 86Rb+ efflux in the presence of 3 and 16.7 mmol/l glucose. Stimulation of 86Rb+ efflux was also evident in the absence of calcium. DIP evoked release of 45Ca2+ from the pancreatic islets both in the presence or absence of extracellular calcium. Employing inhibitors of the calcium-activated potassium channel (KCa) and the high conductance K+-channel (BKCa), the effect of DIP on 86Rb+ efflux was slightly diminished. Tolbutamide had no effect on 86Rb+ efflux in the presence of DIP. On the other hand thapsigargin, a blocker of the Ca2+-ATPase of the endoplasmic reticulum, completely suppressed the DIP-mediated 86Rb+ outflow. The data suggest that thiol oxidant-induced potassium efflux from pancreatic islets is mainly mediated through liberation of intracellular calcium and subsequent stimulation of calcium-activated potassium efflux.  相似文献

综述了过去几十年来铵盐和硝酸盐稳定同位素丰度测定方法的历史发展变化,分析了各种方法的优缺点,并对新方法作了介绍和推荐.目前铵盐稳定同位素丰度的最新测定方法为次溴酸盐氧化结合羟胺还原法,硝酸盐氮氧同位素丰度主流的测定方法为反硝化细菌法和镉粉叠氮酸还原化学法.这些方法的主要共同特点是以N2O为分析物,分析精度高,对样品的含氮量需求小,一般只需要10~60 nmol N,适用于低浓度样品.新方法的建立对于国内外开展氮素循环研究将起到极大的推动作用.  相似文献

Aims Although acquisition of soil organic nitrogen (N)(mainly amino acids) by plants is a widespread ecological phenomenon in many terrestrial ecosystems, the rate of organic N uptake and their contributions to plant nutrient supply are poorly understood. Our objective was to determine the relative contributions of inorganic N (NO3-N and NH4+-N) and organic N (amino acids) to plant N uptake in a high-frigid forest ecosystem.Methods The differences in the uptake rate of three different forms of N (NO3-N, NH4+-N and glycine) were quantified by exposing seedlings of two dominant tree species (Picea asperata and Betula albo-sinensis) in subalpine coniferous forests of western Sichuan, China, to trace quantities of K15NO3,15NH4Cl and (U-13C2/15N) glycine.Important findings Both 13C and 15N were significantly enriched in fine roots 2 h after tracer application, indicating the occurrence of glycine uptake in P. asperata and B. albo-sinensis seedlings. The seedlings of two tree species had a significant preference for NO3-N compared with glycine and NH4+-N, and the uptake rate of NO3-N was 5 to 10 times greater than that of glycine and NH4+-N. The roots of seedlings in the two species took up glycine more rapidly than NH4+-N, implying that soil organic N (i.e., amino acids) could be an important N source for the two species in subalpine coniferous forests. The results of this study are of great theoretical significance for understanding N utilization strategies and nutrient regulation processes in plants of the high-frigid forest ecosystems.  相似文献

Chaetomium thermophilum CT2 produced endocellulases at 50 °C, when grown on 2% microcrystalline cellulose, 1% soluble starch, and 0.4% yeast extract medium. A major endocellulase component was purified to homogeneity by fractional ammonium sulphate precipitation, ion-exchange chromatography on DEAE-Sepharose, Phenyl-Sepharose hydrophobic interaction chromatography and gel filtration on Sephacryl S-100. The molecular weight of the enzyme was estimated to be 67.8 kDa and the enzyme was found to be a glycoprotein containing 18.9% carbohydrate. The Km of the purified enzyme for carboxymethyl cellulose, sodium salt (CMC), was 4.6 mg ml−1. The enzyme displayed highest activity towards CMC and significantly lower activities towards phosphoric acid swollen cellulose and filter paper. The activity was enhanced in the presence of Na+, K+ and Ca2+ but inhibited by Hg2+, Zn2+, Ag+, Mn2+, Ba2+, Fe2+, Cu2+, Mg2+ and NH4+. Optimum activity was at 60 °C and pH 4.0. The enzyme was stable over 60 min incubation at 60 °C and half-life at 70, 80 and 90 °C was approximately 45, 24 and 7 min, respectively.  相似文献

研究森林可燃物燃烧释放的细颗粒物(PM2.5)的排放因子对于揭示森林火灾对大气和生态系统的影响至关重要,而水溶性离子是细颗粒物的重要化学成分,对颗粒物的形成具有重要意义。利用自主设计的生物质燃烧系统,模拟内蒙古大兴安岭5种典型乔木(蒙古栎、白桦、兴安落叶松、黑桦、山杨)的3种组成部分(树干、树枝、树皮)及其地表死可燃物(凋落物层、半腐殖质层、腐殖质层)以及3种典型灌木(平榛、二色胡枝子、兴安杜鹃)树枝燃烧,采用ISC1100离子色谱分析仪测定2种燃烧状态(阴燃和明燃)下PM2.5中水溶性离子(Na+、NH4+、K+、Mg2+、Ca2+、F-、Cl-、NO3-、NO2-、SO42-)的排放因子。结果表明:乔木所有组成部分及其地表死可燃物和灌木树枝燃烧所排放PM2.5中检测到的水溶性离子,阴燃以K+、Cl-和Na+为主要组分,明燃以K+、Cl-和SO42-为主要组分。不同燃烧状态下相同乔木树种及其地表死可燃物和相同灌木排放PM2.5中检测到的水溶性离子总量均存在显著差异。灌木树枝在阴燃期间PM2.5中水溶性无机离子的排放因子比明燃更高。乔木释放的PM2.5中阳离子与阴离子的比率为1.26,地表死可燃物为1.12,灌木为2.0,表明颗粒物呈碱性。内蒙古大兴安岭的森林大火不会通过释放水溶性离子导致生态系统的酸化。  相似文献

1. During incubation at 1° in saline medium buffered either with phosphate or bicarbonate, slices of Morris hepatoma 3924A, and of a chemically induced tumour of rat mammary gland, lost K+ and gained Na+, Ca2+ and water.

2. Upon subsequent incubation at 38° in oxygenated medium, these changes were partially reversed. In the hepatoma, the reaccumulation of K+ was equally efficient in phosphate or bicarbonate medium, and in the presence and absence of glucose. Ca2+ was extruded in bicarbonate, but not in phosphate medium, and its extrusion was reduced in the presence of glucose.

3. When respiration was inhibited in the presence of glucose, K+ transport by the hepatoma continued to an extent which varied with the glycolytic activity of the slices, suggesting that the rate of ATP synthesis was a limiting factor under these conditions.

4. In the absence of glucose, the transport of Na+ and K+ was completely stopped by respiratory inhibition. However, more than 50% of the O2 uptake had to be inhibited before any effect on transport was observed, suggesting that the rate of synthesis of ATP from endogenous respiration is in excess of that required to maintain transport.

5. Inhibition of transport by ouabain was accompanied by a 30% fall in the rate of endogenous respiration, and by a fall of 33% in the rate of glycolysis in the presence of cyanide plus glucose.

6. Comparison of the minimum rates of respiration and of glycolysis (in the presence of glucose plus cyanide) required to maintain the maximal extent of K+ transport in the hepatoma slices, suggests that ATP derived from oxidative phosphorylation or from anaerobic glycolysis is equally efficient as a source of energy for ion transport.  相似文献
