b-Galactosidase production by Kluyveromyces fragilis was optimized by controling dissolved oxygen concentration (DOC) due to the enzyme production rate being strongly dependent on this parameter in the broth. A system of automatic control of substrate inflow which depends on DOC has been devised. This strategy has resulted in a significant and reproducible improvement in the performance of the fermentation. Volumetric enzyme production rates were greater in this system than in batch culture of the microorganism with DOC control.  相似文献

On-line NAD(P)H fluorescence and culture redox potential (CRP) measurements were utilized to investigate the role of Vitreoscilla hemoglobin (VHb) in perturbing oxygen metabolism of microaerobic Escherichia coli Batch cultures of a VHb-synthesizing E. coli strain and the iso-genic control under fully aerated conditions were subject to several high/low oxygen transitions, and the NAD(P)H fluorescence and CRP were monitored during these passages. The presence of VHb decreased the rate of net NAD(P)H generation by 2.4-fold under diminishing oxygen tension. In the absence of aeration, the strain producing VHb maintained a steady NAD(P)H level 1.8-fold less than that of the control, indicating that the presence of VHb keeps E. coli in a more oxidized state under oxygen-limited conditions. Estimated from CRP, the oxygen uptake rates near anoxia were 25% higher for cells with VHb than those without. These results suggest that VHb-expressing cells have a higher microaerobic electron transport chain turnover rate. To examine how NAD(P)H utilization of VHb-expressing cells responds to rapidly changing oxygen tension, which is common in large-scale fermentations, we pulsed air intermittently into a cell suspension and recorded the fluorescence response to the imposed dissolved oxygen (DO) fluctuation. Relative to the control, cells containing VHb had a sluggish fluorescence response to sudden changes of oxygen tension, suggesting that VHb buffers intracellular redox perturbations caused by extracellular DO fluctuations.(c) John Wiley & Sons, Inc.  相似文献

Polyhydroxyalkanoates (PHAs) are a diverse class of bio-polymers synthesized by bacteria, usually during imbalanced growth conditions. Optimizing PHA productivity is highly dependent on the bioreactor oxygen transfer rate (OTR), which is an important consideration for process performance and economics, particularly with increasing scale. Relatively few in-depth studies are available regarding the effect of OTR and dissolved oxygen content (DOC) on PHA formation, synthesis rates, composition, and characteristics. This review examines past research studies on the effect of low DOC environments on production of short-chain length (scl-) PHAs, synthesized by both pure and mixed cultures, in order to identify opportunities and gaps concerning the effect of DOC on production of medium-chain length (mcl-) PHAs, an area that has not been studied in detail. The literature indicates that production of scl-PHA (a reductive process) acts as an electron sink allowing cells to maintain balanced redox state at low DOC. Conversely, production of mcl-PHA via fatty acid de novo synthesis (also a reductive process) does not occur to any significant extent in low DOC environments, while mcl-PHA synthesis from fatty acids (an oxidative process) can be promoted in low DOC environments. The monomer composition, molecular mass, as well as physical and thermal properties of the polymer can change in response to OTR, but further research in this area is required for both scl- and mcl-PHAs. Process design and management of bioreactor OTR in PHA production might therefore be directed by the final application of the polymer rather than cost considerations.  相似文献

We report here on a low-cost, optical oxygen sensor as an attractive alternative to the widely used amperometric Clark-type oxygen electrode for measuring dissolved oxygen tensions in cell cultures and bioreactor. Our sensor is based on the defferential quenching of the fluorescence lifetime of chromophore in response to the partial pressure of oxygen. This is measured as a phase shift in fluorescence emission from the chromophore due to oxygen quenching when excited by an intensity modulated beam of light. In this article we demonstrate the advantages of lifetime-based optical methods over both intensity based optical methods and amperometric electrodes. Our sensor is particularly suitable for measuring dissolved oxygen in bioreactors. It is autoclavable, is free of maintenance requirements, and solvents the problems of long-term stability, calibration drifts, and reliable measurement of low oxygen tensions in dense microbial cultures that limit the utility of Clark-type elcectordes. An effective automatic control algorithm for set-point control of dissolved oxygen concentration in batch culture has been developed. Adaptation of PI controller to the variable state of batch culture is based on analytically obtained functional relations between the controller parameters and the state variables: oxygen uptake rate, stirring speed, and saturation value of dissolved oxygen concentration, which are measured or estimated on-line. Hypolimnetic anoxic water of Lake Onogawa was subjected to aeration experiments. When the samples were agitated by magnetic stirrers for 24 h, dissolved oxygen increased from 0 to more than 7.6 mg l−1, dissolved iron decreased from 98% to about 5% of the initial total iron, and from 32% to 48% of the dissolved organic carbon (DOC) disappeared. On the other hand, when the anoxic waters were left unstirred, dissolved oxygen increased from 0 to 2.2 mg l−1, dissolved iron decreased from 98% to 31%, and 20% of the DOC disappeared within 48 h. Further 24-h incubation had little effect on the DOC loss, although dissolved oxygen increased to 3.9 mg l−1 and dissolved iron decreased to 5%. These rates of DOC disappearance are too large to be explained by bacterial decomposition. It is quite conceivable that a part of the DOC is coprecipitated with iron(III) precipitates. When Fe(II) in the anoxic hypolimnion is oxidized by autumnal water mixing, probably anoxic water is mixed with aerobic water. The anoxic water must receive oxygen from the aerobic water during this mixing and be simultaneously diluted with the aerobic water. Because the present experimental conditions, especially the stirred one, significantly differ from in situ conditions, the present results are thought to be a potential capacity of DOC coprecipitation.  相似文献

1. The leaching rates of filterable reactive phosphorus (FRP) and dissolved organic carbon (DOC) from five leaf litter types commonly occurring in urban environments in Mediterranean regions of Southern Australia were compared. The relative composition, bioavailability and oxygen demand of this DOC were also assessed. Four tree species were assessed, including the native river red gum (Eucalyptus camaldulensis) and three introduced deciduous species, the English elm (Ulmus procera), London plane (Platanus acerifolia) and white poplar (Populus alba). Grass cuttings (mixed species) were selected as a common garden waste. 2. Except for English elm, the majority of FRP and DOC was released within the first 48 h. Grass cuttings released the highest amount of FRP with white poplar releasing the most DOC. Species that released relatively high amounts of DOC (white poplar, English elm, river red gum) released relatively low amounts of FRP. Conversely, species that released relatively low amounts of DOC (grass cuttings and London plane) tended to release relatively high amounts of FRP. 3. Analysis of DOC composition, combined with the differing oxygen demand and DOC depletion curves, demonstrated that there were substantial differences in the DOC leached from the leaf litter of the different species. Biochemical oxygen demand and the biodegradability of the DOC was positively correlated with the proportion of hydrophilic and hydrophobic acids present in the leachate. 4. These results demonstrate that simple measurements of nutrient release per gram of leaf litter would be insufficient to predict the ecological impact on receiving waters resulting from changes in dominant vegetation. Furthermore, the use of traps to prevent particulate leaf material from entering streams may have limited potential for reducing the load of dissolved nutrients. We conclude that any changes to vegetation type which substantially alter the timing of leaf fall or the composition of leaf leachates should be avoided.  相似文献

1. We determined the rate of release and microbial uptake of dissolved organic carbon (DOC) leached from three components (leaves, bark and twigs) of river red gum ( Eucalyptus camaldulensis ) forest litter originating from different parts of a floodplain and under different oxygen levels.
2. Preliminary experiments showed that substantially more DOC was released from leaves than from bark or twigs; there was relatively little DOC release from coarse particulate matter or soil.
3. Both the amount of DOC released from each litter component and the amount metabolized by the microbial community were independent of position on the flood-plain or amount of oxygen available to microbes.
4. Although the bioavailability of DOC was independent of oxygen concentration, the microbial utilization of DOC under aerobic and anaerobic conditions differed. Under aerobic conditions, leaves were colonized by fungi, while bacteria were dominant under anoxic conditions.
5. Phospholipid fatty acid profiles of the microbial communities growing on leaf extracts showed that different microbial communities developed in each oxygen concentration treatment suggesting that, irrespective of flood conditions, a microbial community will develop to utilize a significant proportion of the DOC leached from litter.  相似文献

Many fermentation products are produced under microaerobic or anaerobic conditions, in which oxygen is undetectable by dissolved oxygen probe, presenting a challenge for process monitoring and control. Extracellular redox potentials that can be detected conveniently affect intracellular redox homeostasis and metabolism, and consequently control profiles of fermentation products, which provide an alternative for monitoring and control of these fermentation processes. This article reviews updated progress in the impact of redox potentials on gene expression, protein biosynthesis and metabolism as well as redox potential control strategies for more efficient production of fermentation products, taking ethanol fermentation by the yeast Saccharomyces under microaerobic conditions and butanol production by the bacterium Clostridium under anaerobic conditions as examples.  相似文献

1. Temperature, organic carbon and oxygen consumption were measured over a year at 13 sites in four lowlands streams within the same region in North Zealand, Denmark with the objectives of determining: (i) spatial and seasonal differences between open streams, forest streams and streams with or without lakes, (ii) factors influencing the temperature dependence of oxygen consumption rate, (iii) consequences of higher temperature and organic content in lake outlets on oxygen consumption rate, and (iv) possible consequences of forecasted global warming on degradation of organic matter. 2. High concentrations of easily degradable dissolved (DOC) and particulate organic carbon (POC) were found in open streams downstream of plankton‐rich lakes, while high concentrations of recalcitrant DOC were found in a forest brook draining a forest swamp. Concentrations of predominantly recalcitrant POC and DOC were low in a groundwater‐fed forest spring. Overall, DOC concentration was two to 18 times higher than POC concentrations. 3. Oxygen consumption rate at 20 °C was higher during summer than winter, higher in open than shaded streams and higher in lake outlets than inlets. Rate was closely related to concentrations of chlorophyll and POC but not to DOC. The ratio of oxygen consumption rate to total organic concentrations (DOC + POC), serving as a measure of organic degradability, was highest downstream of lakes, intermediate in open streams and lowest in forest streams. 4. Temperature coefficients describing the exponential increase of oxygen consumption rate between 4 and 20 °C averaged 0.121 °C?1 (Q10 of 3.35) in 70 measurements and showed no significant variations between seasons and stream sites or correlations with ambient temperature and organic content. 5. Oxygen consumption rate was enhanced downstream of lakes during summer because of higher temperature and, more significantly, greater concentrations of degradable organic carbon. Oxygen consumption rates were up to seven times higher in the stream with three impoundments than in a neighbouring unshaded stream and 21 times higher than in the groundwater‐fed forest spring. 6. A regional climate model has calculated a dramatic 4–5 °C rise in air temperature over Denmark by 2070–2100. If this is realised, unshaded streams are estimated to become 2–3 °C warmer in summer and winter and 5–7 °C warmer in spring and, thereby, increase oxygen consumption rates at ambient temperature by 30–40% and 80–130%, respectively. Faster consumption of organic matter and dissolved oxygen downstream of point sources should increase the likelihood of oxygen stress of the stream biota and lead to the export of less organic matter but more mineralised nutrients to the coastal waters.  相似文献

Formation of nitrate reductase (NaR) and nitrous oxide reductase (N2OR) by a Pseudomonas sp. G59 did not occur in aerobic or anaerobic conditions, but was observed in a microaerobic incubation in which an anaerobically grown culture was agitated in a sealed vessel initially containing 20 kPa oxygen in the headspace. During the microaerobic incubation, the oxygen concentration in the headspace decreased and dissolved oxygen reached 0.1-0.2 kPa. NaR activity was detected immediately and N2OR activity after 3 h of incubation irrespective of the presence or absence of NO3- or N2O. In the presence of NO3-, NO2- was accumulated as a major product, but N2O was observed in low concentrations only after N2OR appeared. After microaerobic incubation for 3 h, N2OR formation continued even anaerobically in an atmosphere of N2O. In contrast, Escherichia coli formed NaR not only microaerobically but also anaerobically. However, NaR formation by E. coli was inhibited by sodium fluoride under anaerobic, but not under microaerobic conditions. The Pseudomonas culture did not possess fermentative activity. It is suggested that the dependence on microaerobiosis for the formation of these reductases by the Pseudomonas culture was due to an inability to produce energy anaerobically until these anaerobic respiratory enzymes were formed.  相似文献

An experimental method for studying microaerobic fermentation, called oxygen programmed fermentation, is introduced. The method if based on a chemostat. The mathematical equations governing the dynamics of the system are derived and simulations are made for two principally different cases: a purely respirative organism, and an organism capable of fermentation during oxygen limitation. It is shown that at a suitably chosen ramp rate, the dissolved oxygen concentration in the broth can be made to decrease almost linearly. It is suggested that the greatest use of oxygen programmed fermentation will be in initial experiments. Compared with chemostat studies, a scan of different oxygenation rates will provide a time-saving method of finding the interacting regions for metabolic transitions. The morphology of Mucor racemosus in cultures continuously sparged with nitrogen gas was investigated. When appropriate precautions were taken to prevent oxygen from entering the cultures, the morphology of the cells was uniformly yeastlike irrespective of the N2 flow rate. When small amounts of oxygen entered the cultures the resulting microaerobic conditions evoked mycelial development. Polypeptides synthesized by aerobic mycelia, microaerobic mycelia, anaerobic yeasts, and yeasts grown in a CO2 atmosphere were compared by two-dimensional gel electrophoresis. The results indicated that a large number of differences in polypeptide expression exist when microaerobic mycelia or anaerobic yeasts are compared with aerobic mycelia and that these alterations correlate with a change from an oxidative to a fermentative metabolic mode. Relatively few differences in polypeptide composition exist when microaerobic cells are compared with anaerobic cells, but these changes correlate with a change from the mycelial to the yeast morphology. We hypothesize that oxygen regulates the expression of polypeptides involved in both the metabolic mode and in morphogenesis.  相似文献

The physiological activity of microorganisms in environments with low dissolved oxygen concentrations often differs from the metabolic activity of the same cells growing under fully aerobic or anaerobic conditions. This article describes a laboratory-scale system for the control of dissolved oxygen at low levels while maintaining other parameters, such as agitator speed, gas flowrate, position of sparger outlet, and temperature at fixed values. Thus, it is possible to attribute in dilute nonviscous fermentations all physiologic changes solely to changes in dissolved oxygen. Experiments were conducted with Azotobacter vinelandii and Escherichia coli. Critical oxygen concentrations for growth (that value of oxygen allowing growth at 97% of mu max) were measured as 0.35 +/- 0.03 mg/L for A. vinelandii and 0.12 +/- 0.03 mg/L for E. coli. These values are significantly different from the commonly quoted values for critical oxygen concentrations based on respiration rates. Because of the superior dissolved oxygen control system and an improved experimental protocol preventing CO2 limitation, we believe that the values reported in this work more closely represent reality.  相似文献

In aerobic bioprocesses, oxygen is a key substrate; due to its low solubility in broths (aqueous solutions), a continuous supply is needed. The oxygen transfer rate (OTR) must be known, and if possible predicted to achieve an optimum design operation and scale-up of bioreactors. Many studies have been conducted to enhance the efficiency of oxygen transfer. The dissolved oxygen concentration in a suspension of aerobic microorganisms depends on the rate of oxygen transfer from the gas phase to the liquid, on the rate at which oxygen is transported into the cells (where it is consumed), and on the oxygen uptake rate (OUR) by the microorganism for growth, maintenance and production.  相似文献

A system of automatic control of substrate inflow into an aerated culture of microorganisms which depend on oxygen-absorption rate (OAR) has been devised and tested. As the control variable, dissolved oxygen concentration (DOC), which shows the equilibrium between OAR and oxygen-uptake rate in the microbial culture, was chosen. If the equilibrium is disturbed by changes in OAR, then the oxygen-uptake rate is changed by substrate limitation. The DOC is measured by means of a Clark-type polarographic electrode, and the signal is used to actuate the substrate inflow valve or pump. The oxygen-uptake rate changes of microorganisms, after the addition or exhaustion of substrate in the medium, are so rapid that they can be used for this type of control. Fundamental equations were derived and graphical solutions for the control system parameters were suggested for the steady-state relations between DOC, oxygen-uptake rate, specific growth rate, substrate concentration, KLa, and concentration of microorganisms. The system is stable in the entire range of the uptake rates up to nearly the maximum attainable in unlimited substrate conditions, and can be operated in batch feed or continuous flow modifications. It was experimentally tested with the yeast Saccharomyces cerevisiae. The complete utilization of aeration-system capacity of the fermentor was achieved with high yeast yield and no alcohol formation. The quality of the product was excellent.  相似文献

Dissolved oxygen (DO) concentrations have often been shown to be important to decomposition rates of plant litter and thus may be a key factor in determining the supply of dissolved organic carbon (DOC) and carbon-dependent denitrification in wetlands. During the 2 months operation, DOC accumulation in anaerobic condition was superior to aerobic condition due to higher activities of hydrolase enzymes and lower hydrolysates converted to gaseous C. Also, much higher denitrification rates were observed in wetland when using anaerobic litter leachate as the carbon source, and the available carbon source (ACS) could be used as a good predictor of denitrification rate in wetland. According to the results of this study, extracellular enzymes activities (EEAs) in wetland would change as a short-term consequence of DO. This may alter balance of litter carbon flux and the characteristics of DOC, which may, in turn, have multiple effects on denitrification in wetlands.  相似文献
