The green peach aphid, Myzus persicae (Sulz.), is polyphagous on over 400 plant species in more than 50 families. Phenotypic plasticity of individuals and genetic variability in the population presumably contribute to this polyphagy. The genetic variability in field populations of M. persicae was assessed with respect to their adaptation to sugar beets and potatoes. An analysis of more than 1 000 clones, sampled during 1980, 1981 and 1982 from different host plants in the field, revealed a wide genetic variability in host plant adaptation to sugar beets as well as to potatoes. Both traits seem to be inherited independently from each other and do not correlate with clone-specific host plant preference of apterous adults. The aphid M. persicae can be characterized as a polyphagous insect species with a wide, continuously distributed variability and a broad phenotypic plasticity. A general differentiation of herbivorous species into generalists and specialists tends to ignore the genetic component in the complex of insect-plant relationships.
Zusammenfassung Die Grüne Pfirsichblattlaus Myzus persicae (Sulz.) lebt polyphag an über 400 Pflanzenarten in mehr als 50 Pflanzenfamilien. Ein breites Nahrungsspektrum einer Art ergibt sich jedoch aus der phänotypischen Plastizität des Individuums oder einer Klonlinie und der genetischen Variabilität der Population. Felpopulationen der Grünen Pfirsichblattlaus wurden auf ihre genetische Variabilität bezüglich der Wirtspflanzenanpassung an Rübe und Kartoffel untersucht. Eine Analyse von mehr als 1 000 Klonen, die über die Jahre 1980, 1981 und 1982 im Rheinland gesammelt wurden, lassen eine breite Variabilität in der Wirtspflanzenanpassung der Population erkennen. Beide Merkmale scheinen unabhängig voneinander vererbt zu werden und zeigen keine Beziehung zum Wirtswahlverhalten adult apterer Läuse der entsprechenden Klone. Die Art M. persicae kann daher charakterisiert werden als eine polyphage Insektenart mit einer breiten genetischen Variabilität und einer grossen phänotypischen Plastizität. Eine generelle Differenzierung von Herbivoren in Generalisten und Spezialisten vernachlässigt die genetische Komponente in der komplexen Beziehung zwischen Insekten un ihren Wirtspflanzen.

The development of alatae of the green peach aphid, Myzus persicae, as gynoparae rather than as virginoparae was investigated with regard to the number of exposures to a long-night (LN) regime of 15 h darkness per diem which the aphids experienced before and/or after their birth. The minimum number of exposures to LN that resulted in all of the alatae developing into gynoparae was two prenatal plus one postnatal or one prenatal plus two postnatal, provided the scotophases in these treatments were at least 12 h long. A cumulative effect of several successive exposures to LN was also evident when the presumptive alatae were exposed to LN either from birth or not until several days after birth. Fewer exposures to LN were needed in the former case.
Zusammenfassung Die Entwicklung von Alatae der grünen Pfirsichblattlaus, Myzus persicae, hauptsächlich zu Gynoparae, eher als zu Virginoparae, wurde im Hinblick auf den Einfluss der Anzahl an Langnächten (LN: 15 Studen Dunkelheit pro Tag), denen die Aphiden vor und/oder nach der Geburt ausgesetzt waren, untersucht. Zur ausschliesslichen Entwicklung aller Alatae zu Gynoparae waren mindestens 2 prenatale und eine postnatale LN-Exposition oder eine prenatale und 2 postnatale LN-Expositionen notwendig, vorausgesetzt die Dunkelphasen betrugen mindestens 12 Stunden. Ausserdem zeigte sich ein kumulativer Effect durch mehrere, aufeinanderfolgende LN-Expositionen, wenn die Alatae diesen von Geburt an, oder einige Tage nach der Geburt, ausgesetzt waren. Im ersten Fall waren weniger LN-Expositionen notwendig.

Adult apterousMyzus persicae, which had fed for 26 h on diet containing azadirachtin at concentrations of 25–100 ppm, produced nymphs at less than half the rate of aphids on control diets. After 50 h on azadirachtin-treated diets nymph production had virtually ceased. After this period, any nymphs produced by adults that had fed on azadirachtin-treated diets were born dead with undeveloped appendages. Honeydew production on azadirachtin-treated diets was unaffected during the first 26 h period but was approximately three times less than on control diets during the subsequent 24 h period.  相似文献

Electrical penetration graphs of Myzus persicae (Sulzer) (Homoptera: Aphididae) feeding behaviour on four resistant and two susceptible genotypes of peach (Prunus persica L. Batsch) and related species showed that resistance was mainly linked to (i) reduced duration of phloem sap uptake, (ii) reduced percentage of pattern E1 (salivary secretion into sieve elements) followed by pattern E2 (sap ingestion) and (iii) increased number of shifts from E1 to E2 and back. These results suggest the unsuitability of phloem sap, and thus repetitive failures to initiate sustained ingestion. Extensive comparisons of the EPGs also revealed more specific trends. Aphids on the most susceptible cultivar GF305 produced significantly longer potential drops than on other peach genotypes. On the resistant Rubira, aphids generated more penetrations before the first E occurred, indicating the possible presence of a resistance factor before the phloem was reached. The clone P1908 of the wild species Prunus davidiana displayed traits of both susceptibility (less but longer probes) and resistance. In particular, aphids produced more E1, suggesting difficulties in preparing sieve elements before feeding. The aphid probing process could be correlated with aphid settling behaviour and bionomics, as previously reported, and gave evidence for the existence of different mechanisms underlying resistance in the tested genotypes against M. persicae.  相似文献

Preimaginal development, mortality, aphid consumption rate, and size and weight upon reaching the adult stage of the aphidophagous coccinellids Hippodamia convergens Guérin-Méneville and Coccinella septempunctata L. collected from Karditsa, central Greece, were examined at four constant temperatures (14, 17, 20 and 23 °C) and L16:D8. The coccinellids fed on the tobacco aphid, Myzus persicae nicotianae Blackman. Egg, larval and pupal mortality was highest at 14 °C reaching 85.0, 73.8 and 29.4% in H. convergens and 49.3, 75.4 and 58.8% in C. septempunctata, respectively. Total preimaginal development ranged from 57.2 to 70.4 days at 14 °C, and to 16.9 and 22.1 days at 23 °C in H. convergens and C. septempunctata, respectively. Heavier and larger adults of H. convergens were obtained at 17 and 20 ° C. In C. septempunctata temperature did not affect adult weight while the lowest size was observed at 14 and 17 °C. Day-degrees requirements for preimaginal development in H. corvengens were 212.9 above a developmental threshold of 11.0 °C. The corresponding values for C. septempunctata were 281.5 and 10.7 °C. In H. convergens total and daily aphid consumption ranged from 46.8 aphids at 14 °C to 85.0 aphids at 23 °C and from 1.5 aphids at 14 °C to 9.2 aphids at 23 °C, respectively. The corresponding values for C. septempunctata were 112.0 and 2.7 at 14 °C and 157.7 and 12.4 at 23 °C. The results show the high potential of both predators as biological control agents against the tobacco aphid. The knowledge obtained could be essential for their appropriate use and for the improvement of mass rearing systems.  相似文献

Adults of the lady beetle Hippodamia convergens Guérin-Méneville requiring aphids for egg production, were fed suboptimal amounts of apteriform virginoparous larvae or alatiform gynoparous larvae of the green peach aphid, Myzus persicae (Sulz.), and artificial diet ad lib. The beetles initiated oviposition sooner and deposited significantly more eggs when supplied with the same weight of apterous virginoparous morph. The possible nutritional, hormonal, and ecological implications of the findings are discussed.
Zusammenfassung Die Eiproduktion des Marienkäfers Hippodamia convergens, welcher hierzu Blattläuse als Nahrung benötigt, wurde bei Fütterung mit zwei verschiedenen Morphen der Blattlaus Myzus persicae untersucht. Der Verzehr derselben Menge flügelloser Virginoparen führte im Vergleich zu geflügelten Gynoparen zu früherer und signifikant erhöhter Eiabgabe. Der hormonale und ernährungsphysiologische Hintergrund und die ökologische Bedeutung dieser Ergebnisse werden diskutiert.

The effects of sub-lethal imidacloprid concentrations on acquisition and inoculation of potato leafroll virus (PLRV) by Myzus persicae (Sulzer) (Hemiptera: Aphididae) were investigated. In experiments using two aphid clones to acquire PLRV from infected potatoes, virus transmission declined significantly with increasing concentrations of imidacloprid. The same was true in experiments using imidacloprid-treated Physalis floridana Rydb. as acquisition sources. When viruliferous M. persicae were placed on uninfected, imidacloprid-treated P. floridana, there were significant declines in PLRV transmission. Sub-lethal concentrations of imidacloprid clearly inhibited both acquisition and inoculation of PLRV by M. persicae, either through poisoning, temporary intoxication, and/or antifeedant effects.  相似文献

Discovery in the late seventies of resistance to the green peach aphid Myzus persicae (Sulzer) (Homoptera: Aphididae) in Prunus species was based on screening in the field or in greenhouses with natural aphid populations. Here, we assess the impact of these wild and domesticated peach trees on the behaviour, development, reproductive performance and demography of cloned aphids under controlled light and temperature. Four peach varieties, i.e., Rubira, Weeping Flower Peach, Summergrand and Malo konare and the clone P1908 of the related species Prunus davidiana were tested against the highly susceptible cultivar GF305. Besides a variability in the performance of aphids among experiments, our results showed that (i) distinct mechanisms were involved in the sources of resistance studied and (ii) the ranking of the genotypes on their resistance/susceptibility status remained roughly stable throughout the experiments. Observations on the settling behaviour of first instar nymphs demonstrated antixenosis components in the resistance conferred by Rubira and Weeping Flower Peach. Nymphs began to leave the plants after a short exposure (19–21 h) and no aphid was left after 4 days. Nymphal mortality remained rather low (16%) compared to the repellent effect on aphids of both genotypes. Nymphs disappeared from Weeping Flower Peach significantly earlier than from Rubira. Summergrand, Malo konare and P. davidiana clone P1908 were accepted as host plants by aphids. On P. davidiana, decreased fecundity and intrinsic rate of natural increase (rm=0.20, averaged on all experiments) were clear expression of antibiosis. In addition, the mean length of the mature embryos within the gonads of the females on the day of adult moult was negatively correlated with the total number of embryos, providing evidence that aphids on this genotype lacked sufficient ressources to be directed both towards production and growth of embryos. Compared to the most susceptible cultivar GF305 (rm=0.36), Summergrand (rm=0.26) and Malo konare (rm=0.28) had, to a lesser extent, a negative impact on nymph production and rate of increase.  相似文献

The effect of the entomopathogenic fungus Beauveria bassiana on the biological characteristics and life table of Aphidius matricariae, a parasitoid of the green peach aphid, Myzus persicae, was studied under laboratory conditions. Aphids were first infected with twice the LC95 of B. bassiana for third-instar M. persicae (2 × 108 conidia/ml). Subsequently, at different intervals they were exposed to 1-day-old mated parasitoid females for 24 h. The number of mummies produced per female and the percentage emergence of the F1 generation differed significantly as a function of the time interval between application of the fungus and exposure to the parasitoid. The interference of B. bassiana on parasitoid development was also studied by first exposing the aphid hosts to the parasitoid for 24 h and subsequently applying B. bassiana. The number of mummies produced by a female A. matricariae varied from 11.8 to 24.8 and was significantly different when the aphids were first exposed to the parasitoids and then treated with B. bassiana 24, 48, 72, and 96 h after exposure. There were no significantly different effects of B. bassiana on net reproductive rate (R0), mean generation time (T), intrinsic rate (rm) and the finite rate of increase (λ) of A. matricariae as a result of development in hosts exposed to low or high conidial concentrations (1 × 102, 2 × 108 conidia/ml). The parasitoids developed in infected hosts had lower rm, λ, T and DT (doubling time) values compared with those that developed in uninfected hosts but no differences were observed in R0 values. With proper timing, A. matricariae and B. bassiana can be used in combination in the successful biological control of M. persicae.  相似文献

Farnesyl diphosphate synthase (FPPS) catalyzes the formation of FPP, providing the precursor for the biosynthesis of (E)‐β‐farnesene (EβF) in plants, but it is unknown if FPPS supplies the precursor for the biosynthesis of EβF, the major component of aphid alarm pheromone, though our previous studies support the hypothesis that EβF is synthesized by the aphid itself. Here, we used two cohorts of the green peach aphid Myzus persicae separately, reared on pepper plant and artificial diet to test the correlations among droplet emission, EβF quantity, and FPPS gene expression. It was found that the proportion of aphids emitting cornicle droplets and the quantity of EβF per milligram of aphid were both significantly different between the two cohorts, which were positively correlated with the expression of the two FPPS genes ( MpFPPS1/ 2) in M. persicae. These results were further confirmed by RNAi‐mediated knockdown of MpFPPS1/ 2. Specifically, knockdown of MpFPPS1/ 2 imposed no significant cost on the survival of aphid but remarkably increased the number of offspring per aphid; most importantly, knockdown of MpFPPS1/ 2 significantly reduced the proportion of aphids emitting droplets and the quantity of EβF calculated as per the weight of aphid. Our results suggest that both FPPS genes are involved in the production of EβF in M. persicae and cornicle droplet emission is closely associated with the EβF release in the aphid.  相似文献

The ecological consequences of genetic variability in host plant exploitation of Myzus persicae (Hemiptera, Aphididae) were studied in several agro-ecosystems, differing in crop plant heterogeneity. An analysis of aphid populations sampled from sugar beet, potatoes, and lettuce revealed changes in the frequency distribution of aphid clones according to their respective host plant adaptations. The spatial unit of differentiation was below an average field size of 3–5 hectares and a differentiation could be found in the centre of potato and sugar beet fields but not at the edge of the fields. On a temporal scale, the differentiation of populations is transitory even on a large-scale basis of highly specialized cropping areas. The ecological parameters of differentiation are discussed with special reference to agro-ecosystems and integrated pest management. Increasing the genetic heterogeneity of crop fields may help to reduce population outbreaks.
Zusammenfassung Die ökologischen Konsequenzen genetischer Variabilität in der Wirtspflanzennutzung von Myzus persicae (Hemiptera, Aphididae) wurden in mehreren Agro-Ökosystemen untersucht, die sich in der Kulturpflanzenheterogenität unterscheiden. Eine Analyse von auf Rüben, Kartoffeln und Salat siedelnden Populationen zeigte eine genetische Differenzierung in Wirtspflanzen-angepaßte Feld-populationen. Die räumliche Einheit solcher Differenzierungen liegt unterhalb einer Feldgröße von 3–5 ha. Eine Differenzierung konnte hier in der Mitte der Felder aber nicht am Rand festgestellt werden. Aus zeitlicher Sicht ist die Differenzierung selbst in großräumigen, spezialisierten Anbaugebieten nur vorübergehend feststellbar. Die ökologischen Parameter der Differenzierung werden besonders in Hinsicht auf die integrierte Schädlingsbekämpfung in Agro-ökosystemen diskutiert. Eine Erhöhung der genetischen Heterogenität von Kulturpflanzenbeständen kann möglicherweise zu einer Verringerung von Blattlaus-Massenvermehrungen beitragen.

The electrical penetration graph (EPG) method was used to analyse the feeding behaviour of apterous, adultMyzus persicae (Sulzer) (Homoptera: Aphididae) onNicotiana clevelandii (Gray) seedlings, treated systemically with azadirachtin. A preliminary experiment showed that the effects of tethering aphids for EPG recording were minimal. The percentage of the 9 h recording period devoted to non-penetration activities, and to stylet pathway patterns increased as the azadirachtin concentration in the root treatment increased. The number of probes initiated, and the numbers of sieve tube penetrations also increased with increased azadirachtin concentration. The mean time elapsing between the initiation of the first probe to reach a sieve element and contact with this tissue was not significantly altered by azadirachtin treatment. However, azadirachtin treatment significantly reduced the percentage of probes that reached sieve elements and increased non-penetration activity before and after the first perid of ingestion from the sieve elements. The percentage of the recording period spent in the EPG pattern associated with sieve tube penetration was significantly reduced by an azadirachtin concentration of 300 ppm, and the duration of each individual penetration was significantly reduced by an azadirachtin concentration of 100 ppm. When the total EPG was split into 3 h periods, significant interactions were seen between time period and azadirachtin concentration for the duration of non-penetration, pathway, and sieve tube penetration patterns.  相似文献

The suitability of Aphis fabae Scopoli, Myzus persicae Sulzer and Aleyrodes proletella L. as food sources for Coccinella undecimpunctata L. was evaluated by studying the impact of prey consumption on the predator’s population growth parameters and feeding parameters. Unlike A. proletella, A. fabae and M. persicae supported the development and reproduction of C. undecimpunctata. A. fabae and M. persicae were considered to be essential prey, whereas A. proletella was considered to be an alternative prey. Aphid species showed different degrees of suitability: M. persicae significantly decreased the pre-oviposition period and increased adult longevity, fecundity and fertility compared with A. fabae. Moreover, A. fabae represents a suitable diet for larval development, but is not a suitable food source for adult reproduction. The predator’s population growth parameters, R0, rm and λ were increased with M. persicae, whereas T decreased. We found that the 4th instar larvae were the most voracious, particularly when fed on M. persicae; nevertheless, with this prey daily weight gain and feeding efficiency of 4th instar larvae were similar to that of individuals fed with A. fabae.  相似文献

The life table and biological characteristics of the predatory bug Macrolophus pygmaeus Rambur (Hemiptera: Miridae) were studied when the bugs were fed with Myzus persicae (Sulzer) (Homoptera: Aphididae) feeding on eggplant and with Trialeurodes vaporariorum Westwood (Homoptera: Aleyrodidae) feeding on tomato plants. The tests were done at five temperatures between 15 and 30 °C, using a L16:D8 photoperiod and 65 ± 5% r.h. Most eggs (range 85 to 90%) hatched at 15 and 20 °C. Incubation period was shortest at 27.5 °C (8.45 and 8.38 days on eggplant and tomato, respectively). Preoviposition was also shortest at 27.5 °C (5.10 and 4.75 days on eggplant and tomato, respectively) whereas fecundity was highest at 20 °C (213.90 and 228.25 eggs on eggplant and tomato, respectively). Maximum longevity of females was at 15 °C (122.40 and 129.35 days on eggplant and tomato, respectively). Mean generation time was longest at 15 °C on both host plants (122.75 and 124.64 days, respectively). The intrinsic rate of increase of M. pygmaeus was highest at 27.5 °C with similar values on eggplant (0.0981 day–1) and tomato (0.1040 day–1). Doubling time was shortest at 27.5 °C (7.06 and 6.67 days on eggplant and tomato, respectively) and, also, finite rate of increase was highest at 27.5 °C (1.1031 and 1.1096 on eggplant and tomato, respectively). The results show that the predator M. pygmaeus develops well on the aphid M. persicae or on the whitefly T. vaporariorum, both of which are important pests of vegetable crops. This predator is also well adapted to the temperatures that occur both in greenhouses and in the open field in the Mediterranean region. Compared to other natural enemies of whiteflies, such as Encarsia formosa Gahan (Hymenoptera: Aphelinidae), Macrolophus pygmaeus can increase at relatively low temperatures.  相似文献

Multiparasitism of the green peach aphid, Myzus persicae (Sulzer) by the solitary aphidiid parasitoids Aphidius matricariae Haliday and Ephedrus cerasicola Starý was studied in the laboratory at 22°C. Single aphids first stung by an A. matricariae female were then offered to a female of E. cerasicola 1, 2 or 3 days later. E. cerasicola oviposited without restraint in such aphids. The multiparasitized aphids were dissected 3 or 4 days after the Ephedrus parasitization or allowed to mummify.Only a single parasitoid, either egg or larva, was found in a dissected host. The relative frequency of the two species was roughly the same in dissection series as in corresponding mummification series. When E. cerasicola oviposited 1 or 2 days after A. matricariae, E. cerasicola was the intrinsically superior species. Dissection showed that the competition occurred at least one day before the Ephedrus egg hatched, and that very few eggs of A. matricariae hatched in such aphids. Apparently, the Aphidius eggs must have been killed by substances injected by the ovipositing Ephedrus female or by substances or structures connected with the Ephedrus egg itself. When E. cerasicola oviposited 3 days after A. matricariae, A. matricariae survived. Probably its 4th instar larvae devoured the eggs of E. cerasicola together with the other host contents.Interspecific competition without larvae involved has not previously been demonstrated in Aphidiidae.
Résumé L'étude du multiparasitisme de Myzus persicae (Sulzer) par les parasitoïdes Aphidiidae solitaires, Aphidius matricariae Haliday et Ephedrus cerasicola Stary a été réalisée à 22°C. Des pucerons isolés, piqués par une femelle de A. matricariae avaient été proposés ensuite à une femelle de E. cerasicola, 1, 2 ou 3 jours plus tard. E. cerasicola a pondu sans restriction dans de tels pucerons.Des témoins constitués de pucerons piqués par A. matricariae ont permis de déterminer le pourcentage des pucerons piqués, réellement parasités par A. matricariae. Les pucerons parasités une ou plusieurs fois ont pu se mommifier ou ont été disséqués 3 ou 4 jours après avoir été parasités par Ephedrus.Les fréquences relatives de développement des deux espèces, obtenues par dissection ou par étude des mommies, ont été rigoureusement les mêmes. Quand A. matricariae avait pondu 1 ou 2 jours avant E. cerasicola, A. matricariae s'est développé dans au moins 15% des pucerons apparemment multiparasités, contre 60 à 70% dans les témoins. Chez ces pucerons multiparasités, E. cerasicola était un compétiteur intrinsèquement supérieur. Quand A. matricariae avait pondu 3 jours avant E. cerasicola, A. matricariae survivait et se développait dans environ 70% des pucerons multiparasités. Chez ces pucerons, les oeufs d'E. cerasicola avaient probablement été dévorés avec le contenu de l'hôte par les larves âgées d'A. matricariae.Les oeufs des parasitoïdes sont difficiles à observer dans l'hôte les premiers jours après la ponte. Un seul parasitoïde, oeuf ou larve, était découvert lors de la dissection du puceron. La conclusion en a été que la compétition en faveur de E. cerasicola s'est produite quand les deux espèces étaient sous forme d'oeuf et au moins un jour avant l'éclosion d'Ephedrus. On peut en déduire que les oeufs d'A. matricariae sont tués par des substances injectées par la femelle de E. cerasicola lors de la ponte.Une compétition interspécifique n'impliquant pas les larves n'avait jamais été démontrée chez les Aphidiidae.

Two viviparous parthenogenetic clones of the green peach aphid, Myzus persicae (Sulzer), one collected from Rydalmere, New South Wales (NSW), and the other from South Perth (SP), Western Australia, were reared on radish, Raphanus sativus L. cv. Scarlet Globe, under controlled conditions. The NSW clone was fed on simple artificial diets containing alkaloid extracted from narrow-leafed lupin, Lupinus angustifolius L. cv. Fest., and its reproductive performance monitored over 112 h. Forty (40) h into the experiment and thereafter, aphids on the control diet (sucrose solution) produced significantly more offspring (P<0.05) than those on diets containing alkaloid. In a separate experiment, apterae of each clone were caged on three lines (cv. Yorrel, cv. Danja and 84L:441) of narrow-leafed lupin, and allowed to reproduce. The first three offspring were retained, and all developed to 3rd or 4th instar stage. Two nymphs were removed, and the remaining nymph reared through. All three lines produced adults. The number of young produced were counted over 11 days. Fecundity of the SP clone was lower on line 84L:441, but there was no difference in the fecundity of the NSW clone. Phloem exudate and green tissue was concomitantly collected from all lines, and analysed by GC-MS for the alkaloids lupanine and 13-hydroxylupanine. Line 84L:441 contained the highest level of total alkaloids in both phloem and tissue. All experiments indicate that alkaloid level may suppress fecundity of green peach aphids.  相似文献

Light reflectance in five wavebands of the spectrum was measured from broccoli (Brassicae oleracea var.botrytis [L].) interplanted with leguminous cover crops (cover crop background) or broccoli grown as monoculture (bare soil background), and fertilized with compost or synthetic fertilizer. AlateBrevicoryne brassicae (L.) andMyzus persicae (Sulzer) (Homoptera: Aphididae) were monitored in yellow pan water traps and on broccoli leaves. Reflectance intensity was higher with a background of bare soil at all wavebands except blue (400–455 nm) in the early-season. Intensity decreased as broccoli canopy cover increased at all wavebands except blue and green (515–550 nm), declining-most dramatically in the yellow (550–590 nm). Highest late-season intensities were in plots with bare soil background and fertilized with compost (those stressed for nitrogen). Few differences in spectral composition, expressed for each waveband as a percentage of total intensity, were recorded. Numbers of alatae were lowest in cover crop background plots in the early season, reached equivalency with bare soil background by mid-season, and showed highest positive correlations with intensity in the yellow (550–590 nm). Results correspond to laboratory findings that aphids are attracted to higher intensity light, especially in the yellow waveband, and support a phototactic explanation for aphid orientation in the field.  相似文献

Resistance to cyclodiene insecticides is associated with replacements of a single amino acid (alanine 302) in a γ-aminobutyric acid (GABA) receptor subunit encoded by the single-copy gene Resistance to dieldrin (Rdl). Alanine 302 is predicted to reside within the second membrane-spanning region of the Rdl receptor, a region that is thought to line the integral chloride ion channel pore. In all cyclodiene-resistant insects studied to date, this same alanine residue is replaced either by a serine, or, in some resistant strains of Drosophila simulans, a glycine residue. Therefore, individuals can carry only two different Rdl alleles. In contrast, here we report the presence of up to four different Rdl-like alleles in individual clones of the green peach aphid, Myzus persicae. In addition to the wild-type copy of Rdl gene (encoding A302 or allele A), M. persicae carries three other alleles with the following amino acid replacements: A302 → Glycine (allele G), A302 → SerineTCG (allele S) and A302 → SerineAGT (allele S′). Evidence from direct nucleotide sequencing and Single Stranded Conformational Polymorphism (SSCP) analysis shows that at least three of these different Rdl alleles (i.e. A, G and S) are commonly present in individual aphids or aphid clones. Southern analysis using allele-specific probes and analysis of sequences downstream of the exon containing the resistance-associated mutation confirm the presence of two independent Rdl-like loci in M. persicae. One locus carries the susceptible alanine (A) and/or resistant glycine (G) allele while the other carries the two serine alleles (S or S′). Whereas resistance levels are correlated with the glycine replacement, the S allele was present in all aphid clones, regardless of their resistance status. These results suggest that target site insensitivity is associated with replacements at the first (A/G) but not the second (S/S′) locus. Phylogenetic analysis of nucleotide sequences indicates that both putative aphid Rdl loci are monophyletic with respect to other insect Rdl genes and may have arisen through a recent gene duplication event.
