After exposure to thermal stress or a control temperature, the relative abundance of ecdysone (E) and 20-hydroxyecdysone (20E) was measured in a wild-type line of Drosophila virilis (101) that is stress responsive and in a mutant line (147) that is not stress responsive. In line 101, the 20E content was higher and E content lower in females than in males. The abundance of E and 20E in females of line 147 was significantly higher than that in females of line 101. Females of line 101 were found to respond to 60 min of heat stress (38 degrees C) by an increase in the abundance of both E and 20E, while in males of this line the amount of 20E increased and that of E declined. A role of the ecdysteroids in the control of reproduction of D. virilis under stress is discussed.  相似文献

The effect of heat stress (38°C) on the content of octopamine (OA) and 20-hydroxyecdysone (20HE) was studied under normal and stressful conditions in adult flies of Drosophila virilislines contrasting in the level of the juvenile hormone (JH). The wild-type flies (line 101) exhibited a pronounced sex dimorphism for the content of both OA and 20HE, which was substantially lower in this line than in flies of the mutant line 147. The level of both hormones increased in flies of line 101 exposed to heat stress, whereas it remained unchanged in flies of line 147 under the same conditions. The effect of heat stress on the level of JH metabolism and fertility was also studied in D. melanogasterwild-type lines and lines carrying mutations in genes responsible for OA and DA syntheses. In octopamineless females of the Th nM18line and in females of the Steline characterized by a doubled content of DA, JH degradation differed from normal: it was increased in both young and mature Th nM18females, while decreased in young and increased in mature Steflies. Fertility was substantially lower in the Stethan in the wild-type line. Flies of all of the D. melanogasterlines produced a stress response; however, in mutant lines, both fertility and stress reactivity of the systems controlling JH metabolism differed significantly from that of the wild-type lines. The role of JH, 20HE, OA, and DA interaction in regulation of Drosophilareproduction under stressful conditions is discussed.  相似文献

The effect of heat stress (38°C) on the content of DL-β-(3,4-dihydroxyphenyl)alanine (DOPA), dopamine, tyramine, octopamine, and their precursor Tyr was studied in adults of two lines of Drosophila virilis contrasting in their stress response. In individuals of line 101 responding to stress by a hormonal stress reaction, the contents of DOPA, dopamine, octopamine, and Tyr were lower than those of line 147 that did not respond to the stress. However, heat stress caused an increase in the contents of DOPA, dopamine, octopamine, and Tyr in line 101, whereas the equivalent titers in line 147 remain unchanged.  相似文献

The contents of octopamine and its precursors (tyrosine and tyramine) were studied in adults of two lines of Drosophila virilis with contrasting stress responses. It was demonstrated that in individuals responding to stress by a hormonal stress reaction (line 101), the contents of octopamine and tyrosine are lower than in nonresponding flies (line 147). It was found that there is no difference between the lines in the level of tyramine under normal conditions. The dopamine response to stressor was also studied. Genetic analysis of these differences revealed that they are controlled by a single gene and that the gene is not sex-linked. The gene controlling the response was found to be linked to chromosome 6 of D. virilis.  相似文献

The heat-resistant subline 147S was obtained in Drosophila virilis by selecting for viability individuals of heat-sensitive stock 147. It was shown that in the heat-treated 147S pupae the activity of juvenile hormone (JH)-esterase is decreased and, consequently, the titer of juvenile hormone is increased compared with those in the control pupae. These changes are consistent with those observed earlier for resistant stock 101. Heat-resistant stocks 101 and 147S were crossed with heat-sensitive stock 147, whose heat-treated larvae show earlier activation and higher activity of JH-esterase than control larvae. The viability and electrophoretic esterase patterns were analyzed in the F1 and F2 hybrids at different temperatures. It was found that the F1 hybrid is resistant to the effect of high temperature and its activity level of JH-esterase is lower compared with controls. In the F2 hybrid, there was a 3:1 segregation of viability and a 1:2:1 segregation of the activity level of JH-esterase at high temperatures. It is concluded that the activity level of JH-esterase and heat resistance in D. virilis are monogenically controlled at high temperatures.  相似文献

The content of JH-esterase was assayed by radial immunodiffusion in Drosophila virilis pupae under normal conditions and under the effects of extreme factors. It was found that JH-esterase content is the same (not different from the control) in pupae showing a high activity of the enzyme and in those not showing it. These data are evidence for a gene controlling JH-esterase activity. It was also shown that a regulatory factor converts inactive into active JH-esterase when homogenates of pupae, with active and inactive forms, were mixed and incubated together. It was demonstrated that the source of the activating factor is the larval brain. Sublines 147-R and 147-I were produced by introducing the second chromosome pair of stocks 103 and 101, which are heat resistant, into the genome of individuals of stock 147, which is heat sensitive. Sublines 160-III, 160-IV, 160-V, and 160-VI were produced by introducing the third, fourth, fifth, and sixth chromosome pairs of stock 147 into the genome of stock 160S, which is heat-resistant. The results of analysis of JH-esterase activity and the viability of individuals of these sublines at high temperatures indicated that the gene regulating the activity of JH-esterase is located in the sixth chromosome of D. virilis.  相似文献

20-hydroxyecdysone (20E) and the juvenile hormone (JH) have an age-specific effect on total dopamine (DA) content in Drosophila (Gruntenko and Rauschenbach 2008). Earlier we studied the mechanism of influence of 20E and JH on DA metabolism in young females (Rauschenbach et al. in J Insect Physiol 53:587–591, 2007a: Arch Insect Biochem Physiol 65:95–102, 2008a; Gruntenko et al. in Arch Insect Biochem Physiol 72:263–269, 2009). Here we investigate the effects of 20E and JH on the activities of the alkaline phosphatase (ALP), tyrosine hydroxylase (TH) and DA-dependent arylalkylamine N-acetyltransferase (AANAT) in mature females of wild type D. virilis under normal conditions and under heat stress (38°C). 20E feeding of the flies led to a substantial decrease in ALP and TH activities and to an increase in AANAT activity in mature females. JH application resulted in an increasing of ALP and TH activities, but did not influence AANAT activity in mature females. A rise in JH and 20E levels was found to change ALP and TH stress reactivities. Mechanisms of age-specific regulation of DA level by 20E and JH in Drosophila females are discussed.  相似文献

The effect of exogenous 20‐hydroxyecdysone (20E) and juvenile hormone (JH) on the activities of the tyrosine decarboxylase (TDC), the first enzyme in octopamine (OA) synthesis, has been studied in young females of wild type D. virilis and D. melanogaster under normal and heat stress (38°C) conditions. Flies fed 20E expressed increased TDC activity in both species. JH application decreased TDC activity in both species. A rise in JH and 20E levels did not prevent a TDC response to heat stress, but changed the response intensity. A long‐term increase in JH titre had no effect on the activity of main OA catabolyzing enzyme, arylalkylamine N‐acetyltransferase, in females of both species. A possible mechanism of regulation of OA levels by 20E and JH in Drosophila females is discussed. © 2009 Wiley Periodicals, Inc.  相似文献

Dopamine (DA) content, tyrosine decarboxylase (TDC) activity and survival were studied under normal and environmental stress conditions in the ste and e strains carrying ebony mutation increasing DA level and the octopamineless strain Tbetah(nM18) of Drosophila melanogaster. Wild-type strains Canton S and Oregon R, and strain p845 from which Tbetah(nM18) strain was derived were used as controls. Sexual dimorphism of TDC activity, DA content, and survival in flies of all D. melanogaster strains under study was found. Tbetah(nM18) mutation sharply reduced TDC activity in females, while ebony had no such effect. DA content and survival under heat stress in Tbetah(nM18) flies did not differ from those in the wild type. ste and e flies had drastically increased DA content under normal conditions, dramatically decreased survival under heat stress, but increased survival under starvation. DA content and survival under heat stress were also studied in the reciprocal hybrids (males) F(1) of the cross D. virilis strains 101 (wild type) and 147 with X-linked mutation, which significantly increases DA content. 147x101 males had a considerably higher DA content and lower survival than 101x147 ones. Individuals of all D. melanogaster strains under study developed the stress reaction, as judged by changes in TDC activity and DA levels. Adult males of Drosophila species (Drosophila melanogaster L. and Drosophila virilis) show a lower tolerance to heat stress compared with females. The present study investigates the effects of RNA interference (RNAi) knockdown of the insulin‐like receptor in the corpus allatum of D. melanogaster males on dopamine metabolism and content, heat stress resistance and juvenile hormone metabolism. In male flies, the knockdown of insulin‐like receptor in the corpus allatum is shown to change metabolism of juvenile hormone but not dopamine. It is also shown that knockdown of the insulin‐like receptor in the corpus allatum results in a decrease of heat stress resistance in females but not males, and that juvenile hormone rescues this decrease. The results suggest that at least one of the ways in which insulin‐like growth factors affect heat stress resistance is by successive mediation through juvenile hormone and dopamine, which could lead to sexual dimorphism in the trait.  相似文献

The courtship rituals of Drosophila include an exchange of several signals with different modalities, chemical, visual, acoustic and tactile stimuli, between sexes. Using a video recording method, we studied the role of acoustic communication in courtship behavior in three species of the Drosophila virilis group, D. virilis, D. lummei and two populations of D. littoralis. Five series of experiments were performed: tests with intact flies (control), tests with mute flies (wingless males or females), and tests with deaf flies (aristaless males or females). We distinguished the two situations: either a female did not hear a male or vice versa, males did not hear females. When females did not hear males, the reduction in the copulation number was found in D. virilis and both populations of D. littoralis, but not in D. lummei. When males did not hear females, the reduction in the copulation number was only found in D. littoralis. The ablation of the male aristae in D. virilis and D. lummei even increased the mating success as compared to the control, which may be explained by the ‘sensory overload’ hypothesis. The changes in courtship temporal structure usually included the delayed onset of the main courtship elements (tapping, licking, and singing), and the variation in their duration and the total time of courtship. These effects were, however, more substantial in D. virilis and both populations of D. littoralis than in D. lummei. Thus, the effect of blocking the acoustic channel was different in the three species regardless of their phylogenetic relationship, and the role of acoustic communication in courtship behavior seemed to increase in the order D. lummei – D. virilis – D. littoralis.

Summary When crossing Drosophila virilis females with D. littoralis males, the elimination of D. littoralis sixth chromosome (microchromosomes) was often observed. The absence of the sixth chromosome of D. littoralis was revealed when studying F1 hybrids, because of the mosaic expression of the recessive gene gl, located in the sixth chromosome of D. virilis. In the reciprocal cross the elimination of the sixth chromosome of D. littoralis did not take place (Sokolov 1959).Genetic analysis enabled the authors to conclude that the observed maternal effect on mitosis is controlled by recessive genes located on the second and fourth chromosome of D. virilis. The genes located on the second chromosome, differ from those on the fourth chromosome both in temperature sensitivity and in the time and/ or the mechanism controlling the mitotic behaviour of the chromosomes.By means of back-crosses a new stock was established where all chromosomes except the sixth belonged to D. virilis. The sixth pair (microchromosomes) in this line was represented by one D. virilis and one D. littoralis chromosome. It was shown that the sixth chromosome of D. littoralis might be eliminated or undergo non-disjunction in D. virilis germline but the frequency of such atypical behaviour was very low (about 2 %). Low temperature treatment was not effective for increasing the frequency of either elimination or non-disjunction of the D. littoralis sixth chromosome in D. virilis germ-line.  相似文献

The effect of 20-hydroxyecdysone (20E) and the juvenile hormone (JH) on the activity of the arylalkylamine N-acetyltransferase (AANAT) was studied in young females of wild-type D. virilis and D. melanogaster. 20E feeding of the flies led to a decrease in AANAT activity in both species when dopamine (DA) was used as substrate, but did not affect the enzyme activity when octopamine (OA) was used as substrate. JH application increased AANAT activity with DA as substrate in both species, but did not change it with OA as substrate. AANAT activity was also measured in young females of a JH-deficient strain of D. melanogaster, apterous 56f . A decrease in the enzyme activity was observed in the mutant females as compared to wild-type. Mechanisms of regulation of DA level by gonadotropins in Drosophila are discussed.  相似文献

The activity of tyrosine decarboxylase (TDC) and dopa decarboxylase (DDC) was studied in adults of two lines of Drosophila virilis,contrasting in their reaction to stress conditions. Differences were found in the activity of both enzymes between individuals of the examined lines. Genetic analysis of these differences was made. Each of the two enzymes was found to be controlled by a single gene or, possibly, by a block of closely linked genes. The gene responsible for TDC activity is located on one of the autosomes (excluding chromosome II). DDC activity in D. virilisis regulated by a gene located, apparently, on chromosome II. Adults of the line responding to stress by a stress reaction (r-line) were shown to react to a short-term heat stress (38°C, 60 min) by a decrease in TDC activity. TDC activity in flies of the line incapable of the stress reaction (nr-line) did not alter in such conditions. DDC activity of adults of both lines was found to be unchangeable under stress conditions.  相似文献

Chronic restraint stress (CRS) induces a variety of changes in brain function, some of which are mediated by glucocorticoids. The response to stress occurs in a sex-specific way, and may include mitochondrial and synaptic alterations. The synapse is highly dependent on mitochondrial energy supply, and when mitochondria become dysfunctional, they orchestrate cell death. This study aimed to investigate the CRS effects on mitochondrial respiratory chain activity, as well as mitochondrial potential and mass in cell body and synapses using hippocampus, cortex and striatum of male and female rats. Rats were divided into non-stressed (control) and stressed group (CRS during 40 days). Results showed that CRS increased complex I–III activity in hippocampus. We also observed an interaction between CRS and sex in the striatal complex II activity, since CRS induced a reduction in complex II activity in males, while in females this activity was increased. Also an interaction was observed between stress and sex in cortical complex IV activity, since CRS induced increased activity in females, while it was reduced in males. Glucocorticoid receptor (GR) content in cortex and hippocampus was sexually dimorphic, with female rats presenting higher levels compared to males. No changes were observed in GR content, mitochondrial potential or mass of animals submitted to CRS. It was concluded that CRS induced changes in respiratory chain complex activities, and some of these changes are sex-dependent: these activities are increased in the striatal mitochondria by CRS protocol mainly in females, while in males it is decreased.

The genetic basis of the difference in cold shock tolerance between the southern temperate Drosophila virilis and its boreal relative D. lummei is studied. After adult eclosion, the parental stocks, reciprocal F1 and backcross hybrids were pretreated for eight days at 18°C or at 6°C. The cold shock used consisted of fast cooling to-10°C and exposure to this temperature for varying lengths of time. D. lummei tolerated such exposure for 40–50% longer than did D. virilis (100–135% after acclimation). Reciprocal F1 females, differing only in their maternal cytoplasm deviated significantly from each other, and the reciprocal F1 males even more so, the contribution of the X chromosome being three to four times that of the cytoplasm. The cold resistance scores of the hybrid males were more extreme than those of the parental stocks. Autosomally heterozygous males with the X chromosome and cytoplasm of virilis were the weakest flies studied. The reciprocal males (X chromosome and cytoplasm of lummei) survived better than the parental lummei stock. The reciprocal differences decreased after cold temperature acclimation. The roles of the four major autosomes were analyzed by backcrossing the reciprocal F1 males with females of the virilis marker stock. The third chromosome of lummei as heterozygous contributed most to cold tolerance, while the other autosomes had a rather weak effect in the opposite direction (virilis homozygotes survived better), which disappeared after acclimation at 6°C. Some of the cold susceptibility of F1 hybrids disappeared in chromosomally identical backcross flies, indicating complex cytoplasmchromosomal interactions.  相似文献

The effect of various duration of heat stress (38°C) on the activity of ecdysone 20-monooxygenase converting ecdysone into 20-hydroxyecdysone has been studied in D. virilis of wild type and mutant strain females, which differ by the mode of heat stress response of ecdysone and 20-hydroxyecdysone. We are the first to show that heat stress induces activity of ecdysone 20-monooxygenase in Drosophila females and enzyme activity correlates with the level of 20-hydroxyecdysone.  相似文献
