嗜热拟青霉固体发酵产木聚糖酶条件的优化*   总被引:4,自引:0,他引:4  
从土壤中筛选出一株高产木聚糖酶的嗜热真菌J18,经鉴定为一种新的拟青霉,暂定为嗜热拟青霉。该菌能够利用几种天然纤维质材料固体发酵产木聚糖酶,小麦秸杆为最佳碳源。单因素优化试验表明:小麦秸杆粒度为0.3mm-0.45mm,初始水分含量83%,初始pH7.0,温度为50℃为最佳产酶条件。在优化后的条件下,培养8d产木聚糖酶的水平高达18,580U/g干基碳源。因此,嗜热拟青霉固体发酵产木聚糖酶将具有很大的工业化应用前景。  相似文献

对草酸青霉菌(Penixillium oxalicum)BZH-2002菌株固体发酵果胶酶的主要影响因子温度、初始pH值、含水量及接种量进行了实验探讨,确定了最佳培养条件:温度为30℃,初始pH值为4.8,固体培养基含水量控制在30~35ml/10g甜菜渣,接种量3~4%。同时对该菌株固体发酵提取液中果胶酶的酶学特性进行了初步研究,结果表明,该酶最适反应温度和pH分别为55℃和pH4.8,在40℃温度下和pH3.5~5.5范围内,酶活性较稳定。  相似文献

研究了压力脉动固态发酵反应器内环境参数的周期性变化以及这些周期性的环境刺激对于固态培养的斜卧青霉发酵的影响。研究结果显示:在这种反应器中,在空气压力脉动的带动下,反应器内的温度和空气湿度也会发生较大幅度的周期性变化,变化的周期和空气压力脉动的周期相同,变化的幅度随着压力脉动的幅度增大而增加;在外界周期刺激的条件下,较未加外界周期刺激斜卧青霉的生物量增加了104% ,二氧化碳的产生总量增加了229%和纤维素酶的产量增加了320 % ,数据显示外界周期刺激不仅增加了菌体的生物量同时增加了其代谢活性。  相似文献

The research was conducted to investigate comparative oxidative damage including probable protective roles of antioxidant and glyoxalase systems in rice (Oryza sativa L.) seedlings under salinity stress. Seedlings of two rice genotypes: Pokkali (tolerant) and BRRI dhan28 (sensitive) were subjected to 8 dSm−1 salinity stress for seven days in a hydroponic system. We observed significant variation between Pokkali and BRRI dhan28 in phenotypic, biochemical and molecular level under salinity stress. Carotenoid content, ion homeostasis, antioxidant enzymes, ascorbate and glutathione redox system and proline accumulation may help Pokkali to develop defense system during salinity stress. However, the activity antioxidant enzymes particularly superoxide dismutase (SOD), catalase (CAT) and non-chloroplastic peroxidase (POD) were observed significantly higher in Pokkali compared to salt-sensitive BRRI dhan28. Higher glyoxalase (Gly-I) and glyoxalase (Gly-II) activity might have also accompanied Pokkali genotype to reduce potential cytotoxic MG through non-toxic hydroxy acids conversion. However, the efficient antioxidants and glyoxalase system together increased adaptability in Pokkali during salinity stress.  相似文献

The leather industry needs to switch from the traditional chemically based dehairing process to an environmentally friendly one so that the overall burdens to the environment are reduced. The primary goal of the work was thus to compare the chemical leather dehairing process to an enzymatically based one using the enzymes that are extracted after the application of solid state fermentation (SSF) on hair wastes generated after dehairing. The environmental burdens of the dehairing stage were determined using a life cycle assessment (LCA) approach by comparing the two aforementioned management scenarios. The first scenario was the commonly used technology in which hair is removed via a chemical process and then composted in open piles. This scenario included two subscenarios where hair waste is either incinerated or landfilled. In the second scenario, the proteolytic enzymes extracted during the SSF of the residual hair are used to dehair the new rawhides instead of chemicals. Industrial and laboratory data were combined with international databases using the SimaPro 8.0 LCA software to make comparisons. The environmental impacts associated with the enzymatic dehairing were significantly lower than the ones associated to the conventional chemical dehairing process. This difference is attributed to the impacts associated with the original production of the chemicals and to the electricity consumed in the conventional method. A sensitivity analysis revealed that the results are affected by the amounts of chemicals used during dehairing.  相似文献

Summary The characteristics of Bifidobacterium bifidum grown in solid state fermentation (SSF) system (water content of media 54.5 and 68.8%) was compared with the submerged fermentation (SmF) system (water content of medium: 89.8%). Besides lactic acid (lactate) and acetic acid (acetate), the bacterium was able to secrete propionic acid (propionate) and butyric acid (butyrate) under SSF conditions. However, it only produced lactate and acetate under SmF conditions. The ratio of lactate to acetate was 1.26–1.62:1 in SSF but it was 1:2 in SmF. A higher content of C16:0 and C18:1 as well as a lower content of C18:0 cell membrane fatty acids were observed in SSF than in SmF. There was a lower growth rate, a lower viable count and a longer logarithmic growth phase for B. bifidum cultivated in SSF than in SmF.  相似文献

Summary In order to obtain an industrial strain with higher xylanase production, the original strain of Aspergillus niger was mutated by N+ implantation (10 keV, 1.04 · 1015–2.08 · 1015 ions/cm2), and a mutant P602 was obtained. The mutant showed xylanase production increased from 2989 to 6320 IU/g and the fermentation time was decreased from 72 h to 64 h. The fermentation temperature range was 28–32 °C which was a little wider than the original strain.  相似文献

从土壤中筛选出一株产木聚糖酶的真菌CQ31, 经鉴定后命名为毛壳霉CQ31。该菌能够利用几种农业废弃物固体发酵高产木聚糖酶, 玉米杆为最佳碳源。单因素优化试验表明: 以玉米杆为碳源, 胰蛋白胨为氮源, 初始水分含量80%, 初始pH值9.0为最佳产酶条件。在优化后的条件下培养7 d产木聚糖酶水平高达4897 U/g干基碳源, 此时甘露聚糖酶酶活达803 U/g干基碳源。因此, 毛壳霉CQ31固体发酵产木聚糖酶和甘露聚糖酶具有一定的工业化应用前景。  相似文献

The present work was aimed to investigate the impact of the solid substrates mixture on Fructosyltransferases (FTase) and Fructo-oligosaccharides (FOS) production. An augmented simplex lattice design was used to optimize a three component mixture for FTase production. Among selected substrates corn cobs has highest impact on FTase production followed by wheat bran and rice bran. All two substrates and three substrate combinations showed the highest enzyme production than their individual levels. Among the tested various models quadratic model was found to be the best suitable model to explain mixture design. Corncobs, wheat bran and rice bran in a ratio of approximately 45:29:26 is best suitable for the FTase production by isolated Aspergillus awamori GHRTS. This study signifies mixture design could be effective utilize for selection of best combination of multi substrate for improved production of high value products under solid state fermentation.  相似文献

以黄瓜热敏感品种‘长春密刺'和耐热品种‘津春4号'为试材,在人工气候箱内采用营养液栽培法研究了外源亚精胺(Spd)预处理对短期高温胁迫(42℃)下黄瓜幼苗叶片抗氧化系统的影响.结果显示:(1)随高温胁迫时间的延长,2个黄瓜品种幼苗叶片相对电导率升高、MDA含量增加,热敏感品种的膜脂过氧化程度大于耐热品种;高温胁迫4h,SOD活性降低,POD、CAT、APX活性升高,抗坏血酸(AsA)含量升高,类胡萝卜素(Car)含量降低,且随胁迫时间反映出耐热品种抗氧化系统的自我调节能力大于热敏感品种.(2)外源Spd预处理能有效抑制高温胁迫引起的膜脂过氧化伤害,增强SOD、POD、APX活性以及AsA和Car含量,增强植株抗氧化能力,且在热敏感品种上的应用效果优于耐热品种.研究表明,外源Spd预处理能有效提高黄瓜幼苗叶片处于高温胁迫时的抗氧化能力,对缓解高温胁迫有重要作用.  相似文献

本文对蜂王浆中抗生素残留物质氯霉素(Chloramphenicol)的检测方法进行了探讨,建立了氯霉素残留的HPLC定量方法。样品采用液液分配和固相萃取法(SPE)进行纯化。氯霉素在0.1~10μg/mL间呈良好的线形关系,加样回收率为80.6~90.3%,RSD为3.48~5.63%。该方法稳定可靠,为蜂王浆产品中氯霉素残留的检测提供了依据。  相似文献

钙离子参与一氧化氮促进盐胁迫下的玉米种子萌发   总被引:8,自引:0,他引:8  
0.001~1.0 mmol·L~(-1)一氧化氮(NO)供体硝普钠(SNP)均能提高玉米种子的发芽率,缓解盐胁迫下种子萌发的抑制作用,其中0.1 mmol·L~(-1)SNP的效果最佳。用胞外游离Ca~(2 )螯合剂EGTA、质膜Ca~(2 )通道阻断剂LaCl_3和液泡Ca~(2 )释放抑制剂钌红与0.1 mmol·L~(-1) SNP共处理,均能减弱或抵消SNP促进种子萌发的作用。据此推测,钙离子参与SNP促进盐胁迫下玉米种子萌发的信号转导过程。  相似文献
