目的:设计并构建人eya2(eyes absent2)基因小干扰RNA(siRNA)的真核表达载体,并观察其沉默效果。方法:以人eya2为靶基因,以pSliencer2.1-U6 neo质粒为载体,根据人eya2的cDNA序列,设计含有小发卡结构的2条寡核苷酸序列,将其克隆到siRNA表达载体上;转化大肠杆菌DH5Ⅸ菌株,抽提质粒,测序分析;将重组质粒转染人胚肾293T细胞,通过荧光分析、Westemblot和转录活性实验检测其抑制效果。结果:重组体测序结果与目的序列相一致,证明构建了eya2 siRNA真核表达载体;荧光观察表明siRNA能显著减弱细胞中绿色荧光强度,抑制eya2基因表达;Westemblot分析证明构建的siRNA能有效抑制外源性及内源性eya2基因表达;转录活性测定表明,构建的siRNA能有效抑制eya2基因表达。结论:构建了eya2 siRNA真核表达载体,该siRNA能有效地抑制eya2基因表达。  相似文献

Dual siRNA against different regions of gene in hepatitis C virus (HCV) synergistically inhibited replication of HCV RNA. An HCV-infected cell model was established, and HCV RNA and core protein were detected by RT-PCR and Western blot, respectively. Four HCV-specific siRNAs (siCore, siNS3, siNS4B, siNS5B) were designed and transfected into HCV-infected Huh7.5.1 cells. The antiviral efficacies of the siRNAs were compared using real time PCR and agarose gel electrophoresis. HCV replication in infected cells was inhibited by IFNα-2b in a dose-dependent manner. Synergistic inhibition effects were achieved with combination treatment of any two of the siRNAs (siCore, siNS3 and siNS5B) at low doses (0.1 and 10 nM), as compared to single siRNA treatment (P < 0.05). Furthermore, CCK-8 assay showed no toxicity of the siRNAs to Huh7.5.1 cells. These findings indicate a promising new therapeutic approach for treatment of HCV.  相似文献

干扰小RNA(small interference RNA, siRNA)是基因敲减的常用工具,广泛用于基因沉默技术和基因功能研究,在临床疾病治疗等方面也有潜在的应用。一般认为,达到一定长度(比如大于27 bp)的双链RNA可以诱导干扰素反应,降低相关基因的表达。目前,siRNA对基因表达的非特异性作用尚不完全清楚。为研究siRNA干扰的非特异基因表达,本研究以胰腺癌细胞HPAC 和BxPC3 为模型,采用高通量测序技术对6种不同干扰小RNA处理及未作处理的HPAC和BxPC3细胞进行转录组测序分析,筛选出干扰小RNA处理后表达量共同下调的基因进行研究。通过生物信息学方法对表达下调基因的功能进行研究,并利用实时荧光定量PCR(qRT-PCR)技术对部分下调基因进行验证。结果表明,短片段双链小RNA能够显著改变细胞的基因表达,而这些基因表达谱的变化是有规律的,特定功能的基因优先发生变化。在表达下调的基因中,某些特定类型的基因变化非常显著,包括氨基酸代谢相关基因、Hedgehog信号途径基因和多巴胺受体D5基因等。这些结果表明,在使用siRNA时需要考虑其序列非依赖性地基因表达调控作用。  相似文献

Several synthetic siRNAs were designed to target various regions of hepatitis C virus (HCV) replicon RNA. The antiviral efficacies of the siRNAs were compared using real time PCR and western blot assessment. siRNAs targeting either specific coding region of HCV NS3 or NS5B were the most efficacious in terms of gene silencing and inhibitory activity of the HCV replicon replication. There was no activation of genes involved in innate immune response by the HCV-specific siRNA, indicating that HCV replication inhibition was not due to non-specific antiviral response. Moreover, 5′-RACE PCR analysis showed that the silencing effect by the siRNAs was mainly caused by specific cleavage of targeted HCV genomic RNA. These findings suggest that RNAi targeting HCV coding regions could provide a useful approach to anti-HCV treatment.  相似文献

Chemically synthesized small interfering RNAs (siRNAs) can specifically knock-down expression of target genes via RNA interference (RNAi) pathway. To date, the length of synthetic siRNA duplex has been strictly maintained less than 30 bp, because an early study suggested that double-stranded RNAs (dsRNAs) longer than 30 bp could not trigger specific gene silencing due to the induction of nonspecific antiviral interferon responses. Contrary to the current belief, here we show that synthetic dsRNA as long as 38 bp can result in specific target gene silencing without nonspecific antiviral responses. Using this longer duplex structure, we have generated dsRNAs, which can simultaneously knock-down expression of two target genes (termed as dual-target siRNAs or dsiRNAs). Our results thus demonstrate the structural flexibility of gene silencing siRNAs, and provide a starting point to construct multifunctional RNA structures. The dsiRNAs could be utilized to develop a novel therapeutic gene silencing strategy against diseases with multiple gene alternations such as viral infection and cancer.  相似文献

 建立一种利用绿色荧光蛋白(GFP)作为报告分子筛选能有效抑制目的基因表达的siRNA的方法.以巨噬细胞移动抑制因子(MIF)基因为研究对象,筛选能有效沉默MIF表达的质粒载体介导的siRNA.构建拥有同一Kozak共有翻译启始序列、翻译启始密码子ATG的MIF-GFP融合表达载体pEGFP-MIF.分别将3个靶向MIF的siRNA表达质粒与pEGFP-MIF共转化HEK293细胞,在荧光显微镜下观察HEK293细胞中GFP的表达,并用荧光定量PCR检测HEK293细胞中MIF mRNA的表达水平.同时,将MIF siRNA表达质粒分别与MIF表达载体共转化HEK293细胞,用荧光定量PCR检测HEK293细胞中MIF mRNA的表达水平.定量PCR结果显示,GFP表达低的细胞中,MIF mRNA的表达也明显降低;利用pEGFP-MIF和MIF表达载体筛选到的有效MIF siRNA的结果一致.因此,建立了目的基因与GFP融合表达,以GFP作为报告分子来筛选抑制目的基因表达siRNA的方法,并为进行多个基因的有效siRNA的筛选提供解决方案.  相似文献

To screen for effective small interference RNA (siRNA), a simple and visualized method was developed using the green fluorescence protein (GFP) as a reporter. Candidate siRNAs targeting macrophage migration inhibition factor genes (MIF) were identified. By using the pEGFP-N3 vector, the MIF-GFP expression plasmid, pEGFP-MIF, was constructed with the same Kozak consensus translation initiation site and start code ATG for the MIF-EGFP coding sequence. Based on the siRNA expression vector pSilencer-4.1, 3 candidate MIF siRNA expression plasmids were constructed and co-transfected with the pEGFP-MIF into the HEK293 cells, respectively. The GFP expression in HEK293 cells could be viewed by fluorescence microscopy and the MIF mRNA expressions were determined by real-time quantitative PCR. The 3 candidate MIF siRNA expression plasmids were also co-transfected with the MIF expression plasmid into the HEK293 cells, respectively, and the MIF mRNA expressions were determined by real-time quantitative PCR. The results show that the down-regulated expression of the MIF mRNA was consistent with the GFP expression and the same effective MIF siRNAs were screened by using the pEGFP-MIF or MIF expression plasmid with the candidate MIF siRNAs expression plasmids. Therefore, by using the GFP as a reporter, a useful method was provided to screen for effective siRNAs targeting specific genes co-expressed with the GFP. RNA沉默(RNA silencing)是真核生物中的一种抵抗外源遗传因子(病毒、转座子或转基因)及调控基凶表达的防御机制。参与植物RNA沉默的酶及蛋白质主要包括6种RNA依赖的RNA聚合酶、4种Dicer-like(DCL)核酸内切酶和10种Argonautes蛋白。植物中4条RNA沉默途径分别由微小RNA(miRNAs)和3种小干扰RNA(siRNAs)介导,包括反式作用siRNAs(ta-siRNAs)、天然反义siRNAs(natsiRNAs)和异染色质siRNAs(hc-siRNAs)。在植物RNA沉默的系统性传播中,由DCL4或DCL2将dsRNAs裁剪为次级SiRNAS,以放大RNA沉默信号和增强沉默效应。  相似文献

In this study we analyse several aspects of cytoplasmic RNA silencing by agroinfiltration of DNA constructs encoding single- and double-stranded RNAs derived from a GFP transgene and from the endogenous Virp1 gene. Both types of inductors resulted after 2–4 days in much higher concentration of siRNAs in the agroinfiltrated zone than normally seen during systemic silencing. More specifically, infiltration of two transgene hairpin constructs resulted in elevated levels of siRNAs. However, differences between the two constructs were observed: the antisense–sense arrangement was more effective than the sense–antisense order. For both double-stranded forms, we observed a relative increase of the 24-mer size class of siRNAs. When a comparable hairpin construct of the endogenous Virp1 gene was assayed, the portion of the 24-mer siRNA class remained low as observed for all kinds of single-stranded inducers. The lack of increase of Virp1-derived 24-mers was independent of the expression level, as demonstrated by agroinfiltration into a transgenic plant that overexpressed Virp1 and showed the same pattern. Using transducer constructs, we could detect within a week transitive silencing from GFP to GUS sequences in the infiltrated zone and in either direction 5′–3′ and 3′–5′. Conversely, for the endogenous Virp1 gene neither transitive silencing nor the induction of systemic silencing could be observed. These results are discussed in view of the current models of RNA silencing.  相似文献

利用pSIREN-RetroQ载体构建了3个沉默多药耐药相关蛋白(MRP1)基因表达质粒pSI REN-siRNAs.并通过限制性内切酶酶切鉴定和DNA测序鉴定,将截断MRP和全长MRP1 cDNA分别克隆到真核表达载体pEGFP-N2和pcDNA3.1中,产生了pEGFP-MRP1T和pcDNA-MRP1表达质粒.质粒pEGFP-MRP1T分别与3个pSIREN-siRNAs共转染HEK293细胞沉默MRP1T-GFP靶基因,pSIREN-siRNA1作为阴性对照.荧光显微镜下显示结果表明,与pSIREN-siRNA1相比,pSIREN-siRNA2和pSIREN-siRNA3产生的siRNA能够有效沉默MRP1T-GFP融合蛋白的表达.为了沉默全长MRP1基因的表达,pcDNA-MRP1分别与3个pSIREN-siRNAs共转染HEK293细胞.Western印迹和MTT分析表明,pSIREN-siRNA2和pSIREN- siRNA3能有效抑制190 kD MRP1在HEK293细胞中的表达,而pSIREN-siRNA1则不能.pSIREN-siRNA2和pSIREN-siRNA3能逆转MRP1转染HEK293细胞产生的多药耐药性.RNA二级结构预测结果分析表明,siRNA1靶序列mRNA局部自由能热动力参数ΔG低于siRNA2和siRNA3靶序列mRNA局部自由能热动力参数,siRNA1的GC含量和Tm值高于siRNA2和siRNA3.这些数据提示,siRNA和局部靶结构可能影响siRNA对MRP1 mRNA表达的沉默作用.  相似文献

Specific gene silencing using small interfering RNAs in fish embryos   总被引:7,自引:0,他引:7  
Recently, small interfering RNAs (siRNAs) have been used for gene knockdown in mammalian cultured cells, but their utility in fish has remained unexplored. Here we demonstrate a siRNA-mediated gene silencing technique in rainbow trout embryos. We found that siRNAs effectively suppressed the transient expression of episomally located foreign GFP genes at an early developmental stage and inhibited the expression of GFP genes in stable transgenic trout embryos. Similar gene silencing was observed with an siRNA against the endogenous tyrosinase A gene. siRNAs interfered with the expression of maternally inherited mRNA. siRNAs did not affect non-relevant gene expression and siRNAs with a 4 base mismatch did not affect target gene expression. siRNA gene silencing is therefore highly sequence-specific. Our findings are the first evidence that siRNA-mediated gene silencing is effective in fish. This technique could be a powerful tool for studying gene function during embryonic development in aquacultural fish species, zebrafish, and medaka.  相似文献
